Abstract:In this study the geographical differentiation of dried sclerotia of the medicinal mushroom Wolfiporia extensa, obtained from different regions in Yunnan Province, China, was explored using Fourier-transform infrared (FT-IR) spectroscopy coupled with multivariate data analysis. The FT-IR spectra of 97 samples were obtained for wave numbers ranging from 4000 to 400 cm-1. Then, the fingerprint region of 1800-600 cm-1 of the FT-IR spectrum, rather than the full spectrum, was analyzed. Different pretreatments were applied on the spectra, and a discriminant analysis model based on the Mahalanobis distance was developed to select an optimal pretreatment combination. Two unsupervised pattern recognition procedures- principal component analysis and hierarchical cluster analysis-were applied to enhance the authenticity of discrimination of the specimens. The results showed that excellent classification could be obtained after optimizing spectral pretreatment. The tested samples were successfully discriminated according to their geographical locations. The chemical properties of dried sclerotia of W. extensa were clearly dependent on the mushroom's geographical origins. Furthermore, an interesting finding implied that the elevations of collection areas may have effects on the chemical components of wild W. extensa sclerotia. Overall, this study highlights the feasibility of FT-IR spectroscopy combined with multivariate data analysis in particular for exploring the distinction of different regional W. extensa sclerotia samples. This research could also serve as a basis for the exploitation and utilization of medicinal mushrooms.

Abstract:In this study, a calibration model based on Near-infrared spectroscopy (NIR) technique and chemometrics method was developed for rapid and non-destructive detecting the polysaccharide contents of lentinula edodes samples collected from different regions. The polysaccharide contents of these samples were firstly determined by standard phenol-sulphruic acid method. Then, NIR spectra of these samples were collected by using Fourier transform infrared spectrometry. Based on these experimental data, a random forest method was further used to distinguish the regions of these samples, with a classification accuracy of 96.6%. After that, a rapid, accurate, and quantitative model was established for predicting the polysaccharide contents of these samples. In the model establishing process, some signal pre-treatment methods were optimized, and the validation results with highest determination coefficient (R2) and low root mean square errors of prediction (RMSEP) were, 0.925 and 0.720, respectively. These results showed that combined NIR technique with chemometrics was an effective and green method for lentinula edodes quality control.
Geographical traceability and multielement analysis of edible and medicinal fungi: Taking Wolfiporia cocos (F.A. Wolf) Ryvarden and Gilb. as an example

Abstract:Different geographical environment has a certain influence on the accumulation of fungi elements and chemical components. However, our knowledge is limited to elucidate the fungi elements in response to heterogeneous environmental and the quality differences among different habitats. Here, multielement analysis, FTIR spectrum, and feature-level fusion technique combined with chemometrics were used to study Wolfiporia cocos from different geographical areas, different sampling sites and different altitude sources. From the results, (1) there is significant difference in element content of samples from different sampling sites and no positive correlation with geographical ranges. (2) There is a correlation between elevation and elements, and relatively low elevation (<1,800 m) is conducive to the enrichment of elements. (3) From the perspective of elements, the W. cocos in Yuxi have relatively better quality. (4) FTIR and feature-level models can well realize origin identification. The SVM models are better than the PLS-DA models, and the feature-level model is better than the single FTIR models. In summary, this study demonstrated that the developed method was reliable and could realize the genuineness evaluation and origin identification of W. cocos. The results have implications for the establishment of the technology system of geographical traceability and the development of high-quality geographical indication products of W. cocos.

Abstract:Wolfiporia cocos is a well-known traditional East-Asian medicinal fungus. In this study, four Wolfiporia cocos samples (FC-DM, RC-DM, FC-YN and SC-YN) of three cultivation methods harvest from Dabie Mountain area and Yunnan province were discriminated and identified by a multi-step IR macro-fingerprinting technique. The correlation coefficient of the three cultivation methods of Wolfiporia cocos are higher than 0.98 in the range of 450–2600 cm−1. According to retrieving the spectrum in Sadtler database, the main compositions of Wolfiporia cocos are laminarin polysaccharide. While Correlation coefficients of synchronous 2D-IR spectra of FC-DM and other Wolfiporia cocos samples were furtherly calculated, acquiring a quantitative discrimination result. The thermal sensitivity of polysaccharides of SC-YN is different from other samples in the range of 850–1370 cm−1. The difference in thermal sensitivity of CH (1402 and 1474 cm−1) and amino acid (1666 or 1651 cm−1) could also be considered as the key factors to discriminate them. The result proved that the 2DCOS-IR could discriminate Wolfiporia cocos of different cultivation methods.
Determination of Cultivation Regions and Quality Parameters of Poria cocos by Near-Infrared Spectroscopy and Chemometrics

Abstract:Poria cocos (PC) is an important fungus with high medicinal and nutritional values. However, the quality of PC is heavily dependent on multiple factors in the cultivation regions. Traditional methods are not able to perform quality evaluation for this fungus in a short time, and a new method is needed for rapid quality assessment. Here, we used near-infrared (NIR) spectroscopy combined with chemometric method to identify the cultivation regions and determine PC chemical compositions. In our study, 138 batches of samples were collected and their cultivation regions were distinguished by combining NIR spectroscopy and random forest method (RFM) with an accuracy as high as 92.59%. In the meantime, we used partial least square regression (PLSR) to build quantitative models and measure the content of water-soluble extract (WSE), ethanol-soluble extract (ASE), polysaccharides (PSC) and the sum of five triterpenoids (SFT). The performance of these models were verified with correlation coefficients (R2cal and R2pre) above 0.9 for the four quality parameters and the relative errors (RE) of PSC, WSE, ASE and SFT at 4.055%, 3.821%, 4.344% and 3.744%, respectively. Overall, a new approach was developed and validated which is able to distinguish PC production regions, quantify its chemical contents, and effectively evaluate PC quality.

Abstract:Nowadays, wild-grown edible mushrooms which are natural, nutritious, and healthy get more and more popular by large consumers. In this paper, UV spectra of different Boletaceae mushrooms with the aid of partial least squares discriminant analysis (PLS-DA) and hierarchical cluster analysis (HCA) were shown to be a practical and rapid method for discrimination purpose. The specimens of Boletus edulis , Boletus ferrugineus , Boletus tomentipes , Leccinum rugosiceps, and Xerocomus sp. were described based on the UV spectra. From the results, all the specimens were characterized by strong absorption at the wavelengths of 274 and 284 nm and showed the shoulder at 296 nm. However, changes could be seen in the peak heights at the same wavelength for different samples. After analyzing by chemometrics, visual discrimination among samples was presented and the relationships among them were also obtained. This study showed that UV spectroscopy combined with chemometrics methods could be used successfully as a simple and effective approach for characterization of these five wild-grown edible mushrooms at species and genus levels. Meanwhile, this rapid and simple methodology could also provide reference for the discrimination of edible mushrooms.

Abstract:There are a wide variety of latex examination gloves now available for use by health care providers. A prospective randomized trial was completed to quantify the forces required to don a sample of seven cornstarch-lubricated gloves and 13 powder-free latex examination gloves. The data collected was analyzed by a 20 x 2 general factorial ANOVA, as well as two 1-way ANOVAs using a least significance difference post hoc test. Some powder-free gloves can be easily donned with dry or wet hands without tearing with forces comparable to those encountered with powdered gloves. With the advent of these powder-free examination gloves, powdered gloves can now be abandoned, protecting health professionals and patients from the dangers of absorbable dusting powders. Despite the dangers of the absorbable dusting powders and the Food and Drug Administration's requirement for labeling examination glove boxes, some manufacturers of powdered examination gloves do not appropriately label their boxes with a warning to the health professional and patient of the presence of powder.

Abstract:Origin traceability is an important step to control the nutritional and pharmacological quality of food products. Boletus edulis mushroom is a well-known food resource in the world. Its nutritional and medicinal properties are drastically varied depending on geographical origins. In this study, three sensor systems (inductively coupled plasma atomic emission spectrophotometer (ICP-AES), ultraviolet-visible (UV-Vis) and Fourier transform mid-infrared spectroscopy (FT-MIR)) were applied for the origin traceability of 192 mushroom samples (caps and stipes) in combination with chemometrics. The difference between cap and stipe was clearly illustrated based on a single sensor technique, respectively. Feature variables from three instruments were used for origin traceability. Two supervised classification methods, partial least square discriminant analysis (FLS-DA) and grid search support vector machine (GS-SVM), were applied to develop mathematical models. Two steps (internal cross-validation and external prediction for unknown samples) were used to evaluate the performance of a classification model. The result is satisfactory with high accuracies ranging from 90.625% to 100%. These models also have an excellent generalization ability with the optimal parameters. Based on the combination of three sensory systems, our study provides a multi-sensory and comprehensive origin traceability of B. edulis mushrooms.

Abstract:Changes in immunoglobulin class and subclass levels and the development of antitumor antibodies were assessed in normal and tumor-bearing mice challenged with Corynebacterium parvum. C. parvum administration resulted in a marked increase in certain immunoglobulin levels, especially Ig G2b, and in the development of antibodies reacting with syngeneic and allogeneic tumor cells. The serologic changes induced by C. parvum were dependent on the dose and route of administration; preliminary studies suggested that they may have been largly independent of T-cell function. These changes were suppressed by the administration of gold salts, which also inhibited the antitumor effect of C. parvum.

Abstract:The purpose of our study was to determine the effect of vascular occlusion on neuromuscular activation and/or the energy metabolic characteristics of the quadriceps femoris (QF) muscles during muscle contractions. Seven men participated in the study. An occlusion cuff was attached to the proximal end of the right thigh, so that blood flow in the anterior medial malleolar artery was reduced to approximately 88 % of the non-occluded flow. Muscle functional magnetic resonance imaging and maximal voluntary contraction (MVC) were carried out before and immediately after 5 sets of 10 repetitions of knee extension exercises at 50 % of the 10 repetitions maximum, from which transverse relaxation times (T2) and maximal force were measured, respectively. Integrated electromyography (iEMG) activity was recorded from the belly of the rectus femoris, vastus lateralis, and vastus medialis muscles during MVC and repetitive exercises. The percentage change in T2 was significantly increased for individual QF muscles, and there was a significant increase in iEMG activity over the 5 sets of repetitive exercises under conditions of vascular occlusion, but there was no significant effect on isometric force and iEMG activity during MVC. These results are consistent with the idea that there is greater osmolite accumulation during exercise with occlusion, although increased neural activation cannot be ruled out.

Abstract:Polyporus umbellatus(PU) belongs to the family Polyporaceae in the phylum of Basidiomycota. The sclerotia is its main medicinal parts. It is reputedly used in traditional Chinese Medicine as anti-cancer agent and consequently is in high demand. A major problem on the medicinal use of this mushroom is the menace of the counterfeit product. Therefore, the detection of counterfeit PU and the confirmation of genuine PU are important. This study was done by collecting wild PU and cultivated PU and compared with a counterfeit which is commonly found in the market. Fourier Transform Infrared (FTIR) and Two-Dimension Infrared (2DIR) for the qualitative analysis are often regarded as green methods because the original resources of the test sample are preserved. The different spectral patterns of the counterfeit PU compared with the genuine PU were clearly indicated in 1D FTIR spectrum. The 2DIR spectrum of counterfeit PU showed more than 70% dissimilarity compared to genuine PU. This rapid and simple sample preparation enhances the speed of qualitative analysis on PU and rapidly determines their differences in term of their raw material content and the active site of certain compounds. Characterization of volatile profiles and markers prediction of eleven popular edible boletes using SDE-GC-MS and FT-NIR combined with chemometric analysis

Abstract:Wild edible boletes mushrooms are regarded as a delicacy in many countries and regions due to their rich nutritional contents and strong aromatic compounds. This study aimed to identify 445 samples of 11 boletes species collected from Yunnan and Sichuan provinces through molecular analysis. Using simultaneous distillation-extraction (SDE) combined with gas chromatography-mass spectrometry (GC-MS), 97 volatile compounds were identified. Chemometric methods were then applied to analyze the heterogeneity of these volatile compounds among the different species. The results showed that, 22 and 21 volatile compounds were selected using variable importance in projection (VIP > 1) and relative odor activity values (ROAV > 0.1), respectively. Partial least squares discrimnatint analysis (PLS-DA) was then employed to develop pattern recognition models for 11 species, which demonstrated strong identification performance. Furthermore, correlation heat maps, volcano plots, and Fisher linear discriminant analysis identified five volatile organic compounds, including methyl (9E)-9-octadecenoate, 2, 6-dimethylpyrazine, 1-decen-3-one, furfural, and methional as markers for distinguishing 11 boletes species. Ultimately, the rapid content prediction models of partial least squares regression (PLSR) were established by combining Fourier Transform Near-Infrared Spectroscopy (FT-NIR) with the concentrations of these five marker compounds. These findings provide a methodological strategy for the effective species identification of wild edible mushrooms and the rapid prediction of their characteristic aroma compounds.

Abstract:A high performance liquid chromatography-ultraviolet (HPLC-UV) fingerprint method for the overall chemical analysis of edible mushrooms was established based on Auricularia heimuer for the first time, and then applied to analyze 60 batches of A. heimuer, Auricularia cornea, Auricularia cornea Modified Letter Turned CommaYu Muer' and Tremella fuciformis. A total of 9 characteristic peaks of A. heimuer, 11 characteristic peaks of A. cornea, 6 characteristic peaks of A. cornea Modified Letter Turned CommaYu Muer', and 9 characteristic peaks of T. fuciformis were designated. Then, a combinatory analysis, including similarity evaluation, hierarchical cluster analysis and principal component analysis, revealed the chemical consistency and difference between samples from the same and different species. The HPLC fingerprint method established in this paper could be used to characterize the components of A. heimuer, A. cornea, A. cornea Modified Letter Turned CommaYu Muer', and T. fuciformis and discriminate the 4 edible mushrooms effectively in combination with pattern recognition analysis.
Correlation between Quality and Geographical Origins of Poria cocos Revealed by Qualitative Fingerprint Profiling and Quantitative Determination of Triterpenoid Acids

Abstract:Poria cocos (Schw.) Wolf (PC) is a well-known saprophytic fungus, and its sclerotium without the epidermis (PCS) is widely used in traditional Chinese medicine and as a functional food in many countries. PCS is normally collected from multiple geographical regions, but whether and how the quality of PCS correlates with where it grows have not been determined. This correlation could be significant both for quality control and optimum utilization of PCS as a natural resource. In this study, a qualitative fingerprint profiling method performed by ultra-performance liquid chromatography (UHPLC) with diode array detection (DAD) combining quadrupole time-of-flight-mass spectrometry (QTOF-MS/MS) and a quantitative UHPLC coupled with triple quadrupole mass spectrometry (QqQ-MS/MS) approach were established to investigate whether and how the quality of PCS correlates with its collection location. A standard fingerprint of PCS was generated by median simulation of 25 tested samples collected from four main producing areas of China, and similarity analysis was applied to evaluate the similarities between the fingerprints of samples and the standard fingerprint. Twenty three common peaks occurring in the fingerprint were unequivocally or tentatively identified by UHPLC-QTOF-MS/MS. Meanwhile, principal component analysis (PCA), supervised orthogonal partial least squares-discriminate analysis (OPLS-DA) and hierarchical cluster analysis (HCA) were employed to classify 25 batches of PCS samples into four groups, which were highly consistent with the four geographical regions. Ten compounds were screened out as potential markers to distinguish the quality of PCS. Nine triterpene acids, including five compounds that played important roles in the clusters between different samples collected from the four collection locations, were simultaneously quantified by using the multiple reaction monitoring (MRM) mode of UHPLC-QqQ-MS/MS. The current strategy not only clearly expounded the correlation between quality and geographical origins of PCS, but also provided a fast, accurate and comprehensive qualitative and quantitative method for assessing the quality of PCS.

Abstract:Wolfiporia cocos is a widely used traditional Chinese medicine and dietary supplement. Artificial intelligence algorithms use different types of data based on the different strategies to complete multiple tasks such as search and discrimination, which has become a trend to be suitable for solving massive data analysis problems faced in network pharmacology research. In this study, we attempted to screen the potential biomarkers in different parts of W. cocos from the perspective of measurability and effectiveness based on fingerprint, machine learning, and network pharmacology. Based on the conclusions drawn from the results, we noted the following: (1) exploratory analysis results showed that differences between different parts were greater than those between different regions, and the partial least squares discriminant analysis and residual network models were excellent to identify Poria and Poriae cutis based on Fourier transform near-infrared spectroscopy spectra; (2) from the perspective of effectiveness, the results of network pharmacology showed that 11 components such as dehydropachymic acid and 16α-hydroxydehydrotrametenolic acid, and so on had high connectivity in the "component-target-pathway" network and were the main active components. (3) From a measurability perspective, through orthogonal partial least squares discriminant analysis and the variable importance projection > 1, it was confirmed that three components, namely, dehydrotrametenolic acid, poricoic acid A, and pachymic acid, were the main potential biomarkers based on high-performance liquid chromatography. (4) The content of the three components in Poria was significantly higher than that in Poriae cutis. (5) The integrated analysis showed that dehydrotrametenolic acid, poricoic acid A, and pachymic acid were the potential biomarkers for Poria and Poriae cutis. Overall, this approach provided a novel strategy to explore potential biomarkers with an explanation for the clinical application and reasonable development and utilization in Poria and Poriae cutis.

Abstract:Streptococcus mutans has been identified as the principal etiological agent in human dental caries. Therefore, the specificity and sensitivity of 3 potential gene probes derived from S. mutans GS5 have been examined as potential reagents for detecting the cariogenic bacteria. The gene probes derived from the cloned gtfB, gtfD and ftf genes were examined in Southern blots with a panel of representative oral bacteria. The gtfB and ftf genes were apparently specific for S. mutans under high-stringency hybridization conditions. However, the gtfD gene fragment did cross-hybridize with the DNA from other mutans streptococci. These results suggest that gtfB and ftf gene fragments may be used as specific probes for S. mutans.

Abstract:Different geographical regions of traditional Chinese medicine (TCM ) ,its chemical composition is different ,the accu-mulation of drug and medicinal properties is different .The accurate identification and analysis of different production area of me-dicinal herbs is critical for the quality control and pharmacological research of TCM .In this paper ,a tri-step infrared spectrosco-py (Fourier transform infrared spectroscopy (FTIR) combined with second derivative spectra and two-dimensional correlation in-frared spectroscopy (2D-COS) were employed to identify and analyze the main components of Hubei(HB) ,Anhui(AH) ,Yun-nan(YN) genuine Poria Cocos peels .The emergence of several characteristic absorption peaks of carbohydrates including 1 149 , 1 079 1 036 cm -1 ,peaks around 1 619 ,1 315 ,780 cm -1 belonged to calcium oxalate suggested that HB and AH Poria Cocos peels contained calcium oxalate ,but peaks around 797 ,779 ,537 ,470 cm -1 belonged to kaoline suggested that YN Poria Cocos peels contained kaoline .Their carbohydrates were different by comparing the second derivative infrared spectra in the range of 1 640~450 cm -1 and Yongping come from YN contains both calcium oxalate and kaoline .Furthermore ,the above differences were visually validated by two-dimensional correlation spectroscopy (2D-COS) .It was demonstrated that the Tri-step infrared spectroscopy were successfully applied to fast analyze and identify Poria Cocos peels from different geographical regions and sub-sequently would be applicable to explain the relevance of geographical regions and medicinal properties for the TCM .
Quantitative analysis of nutrients for nucleosides, nucleobases, and amino acids hidden behind five distinct regions-derived Poria cocos using ultra-performance liquid chromatography coupled with triple-quadrupole linear ion-trap tandem mass spectrometry

Abstract:Poria cocos is an edible fungus used as a health product and traditional Chinese medicinal preparation. Nevertheless, little is known about its nutrients. In this study, ultra-high performance liquid chromatography coupled with triple-quadrupole linear ion-trap tandem mass spectrometry was conducted to quantify nucleosides, nucleobases, and amino acids in 32 batches of Poria cocos samples collected from Anhui, Sichuan, Hubei, Hunan, and Guizhou. Subsequently, the linearity, precision, repeatability, stability, and recovery of our methods were validated. Samples from different regions were clearly separated by partial least squares discriminant analysis and cluster analysis. Our results suggested that Poria cocos samples from different geographical environments differed in nucleosides, nucleobases, and amino acids. The plot of variable importance for projection disclosed differential compositions of L-Leucine, Uridine, L-Asparagine, L-Glutamine, L-phenylalanine, L-Ornithine monohydrochloride, L-Hydroxyproline, Taurine, and Inosine in Poria cocos from five regions. We found the highest content of total analytes, total amino acids, and total non-essential amino acids in Poria cocos from Anhui, total essential amino acids in the Sichuan samples, and total nucleosides in the Hunan samples. Overall, we determined the content of Poria cocos-derived nucleosides, nucleobases, and amino acids, providing the foothold for further chemical mining and use of Poria cocos.
Attenuated Total Reflection-Fourier Transform Infrared Spectroscopy (ATR-FTIR) Combined with Chemometrics Methods for the Classification of Lingzhi Species

Abstract:Due to the existence of Lingzhi adulteration, there is a growing demand for species classification of medicinal mushrooms by various techniques. The objective of this study was to explore a rapid and reliable way to distinguish between different Lingzhi species and compare the influence of data pretreatment methods on the recognition results. To this end, 120 fresh fruiting bodies of Lingzhi were collected, and all of them were analyzed by attenuated total reflection-Fourier transform infrared spectroscopy (ATR-FTIR). Random forest (RF), support vector machine (SVM) and partial least squares discriminant analysis (PLS-DA) classification models were established for raw and pretreated second derivative (SD) spectral matrices to authenticate different Lingzhi species. The results of multivariate statistical analysis indicated that the SD preprocessing method displayed a higher classification ability, which may be attributed to the analysis of powder samples that requires removal of overlapping peaks and baseline shifts. Compared with RF, the results of the SVM and PLS-DA methods were more satisfying, and their accuracies for the test set were both 100%. Among SVM and PLS-DA, the training set and test set accuracy of PLS-DA were both 100%. In conclusion, ATR-FTIR spectroscopy data pretreated by SD combined with PLS-DA is a simple, rapid, non-destructive and relatively inexpensive method to discriminate between mushroom species and provide a good reference to quality assessment.
