Abstract:Studies in adult patients suggest that copeptin (C-terminal fragment of antidiuretin propeptide) is related to kidney and cardiovascular diseases. AIM The aim was to assess copeptin concentration in children with chronic kidney disease (CKD). MATERIALS AND METHODS In a group of 38 children with CKD (age: from 4.70 to 18.00 mean 12.23±4.19 years) we evaluated: serum copeptin concentration [ng/mL], age, sex, etiology of CKD, presence of arterial hypertension (AH), medications, glomerular filtration rate (GFR), hemoglobin, calcium-phosphorus metabolism parameters, and lipids. Control group consisted of 38 healthy children aged from 5.51 to 18.0 mean 11.79±3.29 years. RESULTS Serum copeptin concentration did not differ between children with CKD and healthy children (0.72±0.34 vs. 0.84±0.33 [ng/mL], p=0.088). In children with CKD there were no differences in copeptin concentration depending on sex, presence of AH, and CKD grade. In children with CKD only positive correlation between copeptin and hemoglobin concentrations was found (r=0.35, p=0.031); no other significant correlations between copeptin and clinical and biochemical parameters including GFR were revealed. Also no significant correlations were found between copeptin and evaluated parameters in the control group. CONCLUSIONS In children copeptin concentration does not seem to be related to kidney function. Copeptin may be a marker of hydration status in children with chronic kidney disease. There is a need for further studies evaluating clinical significance of copeptin in children with chronic kidney disease.

Abstract:A method based on the measurement at room temperature of tryptophan phosphorescence (RTTP) gives the unique possibility to investigate the dynamic structure of membrane proteins without their isolation from cells. This method was used to study the influence of tert‐ butyl hydroperoxide (t ‐BHP) on Chinese hamster fibroblasts. The treatment of fibroblasts with t ‐BHP in a concentration range of 0.5–2m m for 60min caused an increase of frequency and amplitude of membrane protein motions with lifetimes of hundreds miliseconds (a decrease of RTTP τ2). In parallel, cell viability was studied by trypan blue exclusion test and the content of thiobarbituric acid reactive substances was measured in cells. The dependences of the RTTP τ2and cell viability on t ‐BHP concentration were similar. Contrary to this, t ‐BHP did not induce the activation of lipid peroxidation processes in cells. This indicates that cell death is connected with the excessive increase of intramolecular dynamics of membrane proteins during t ‐BHP action.

Abstract:Summary The purified 12000 dalton (p12) phosphoprotein of Rauscher (R) and wild mouse (WM) strains of murine leukaemia virus (MuLV) was analysed for the distribution patterns of its variously charged molecular species by urea-poly-acrylamide gradient gel electrophoresis. The distribution patterns of the p12 of two different field isolates of WM viruses, 292 and 1504, and the mouse-tropic and amphotropic clonal sub-populations of 15-4 field isolate were very similar but different from that of MuLV-R. A unique characteristic of the p12 of the WM isolates is the presence of two major apparently non-phosphorylated species in approximately constant proportions relative to the phosphorylated species. Similar studies on the p12 of the same virus (MuLV-R or WM viruses) grown in different host cells showed that the patterns of phosphorylated and non-phosphorylated species are virus-specific and independent of the cell lines of propagation. These analyses and their comparison with urea-gel patterns of the phosphoproteins of other mammalian type C viruses indicated that the number and relative proportion of the variously phosphorylated and non-phosphorylated species are predetermined for a virus. Therefore, the virus must have the genetic information for the phosphoprotein as well as other necessary genetic information which functions, perhaps in conjunction with appropriate cellular factors, in regulating the specific proportions of these multiple species. Possible biological significance of the variously charged molecular species in the phosphoprotein of RNA tumour viruses is discussed.

Effect of different solutions in reversing the damage caused by radiotherapy in dentin structure

Abstract:Background: Previous studies have shown that radiotherapy of the head and neck region can cause direct changes in dental structure. This study evaluated the effect of different solutions on the dentin chemical composition and collagen structure of irradiated dentin. Material and methods: Sixty maxillary canines were distributed in 2 groups (n=30): non-irradiated and irradiated (radiotherapy: X-rays of 6 MV in 30 cycles of 2 Gy to 60 Gy). The teeth were sectioned, sanded, and polished to obtain 3x3x2 mm fragments, which were redistributed in 3 subgroups (n=10) according to the treatment employed: chlorhexidine 2% (CL), chitosan 0.2% (QT), and 0.5 M carbodiimide (EDC). The samples were analyzed in FTIR at time zero (T0-control) and after 1 (T1), 3 (T3), and 5 (T5) minutes of immersion in the tested solutions. The data for the areas of the carbonate (C), amide I (AI) bands, and the ratio between the areas of the amide III/proline and hydroxyproline (AIII/PH) bands were analyzed using ANOVA and Tukey test (α=5%). Results: QT showed lower C values at T1, T3, and T5 (P<0.0001), presenting lower values when compared to CL and EDC subgroups (P<0.05). AI values at T3 and T5 were higher than T0-control and T1, independently of the radiotherapy and dentin treatment factors (P<0.05). At T0-control, the AIII/PH ratio was lower in the irradiated group (P<0.05), whereas the EDC treatment at T1, T3, and T5 and QT at T3 and T5 increased these values (P<0.05), making them similar to non-irradiated subgroups (P>0.05). Conclusions: Radiotherapy changes the secondary structure of collagen, and EDC was able to restore collagen integrity after 1 minute of immersion, without changing dentin inorganic composition.
Allogeneic Bone Marrow Cocultured With Human Islets Significantly Improves Islet Survival and Function In Vivo

Abstract:Background A significant barrier to islet transplantation is the rapid loss of human islet function in vivo. The present study evaluates whether bone marrow (BM) could be used to support human islet survival and function in vivo. Methods We cocultured human islets and BM for 3 weeks before transplantation into the left subrenal capsule of diabetic severe combined immunodeficient mice. Results The cocultured human islets before transplantation demonstrated improved viability, increased size, and migration capacity in vitro. After 4 months, animals transplanted with precultured BM/islets exhibited euglycemia and detectable human insulin levels (157 &mgr;U/mL), whereas no human insulin was detected in the islet-only transplantation group. Furthermore, the removal of the transplants on day 126 resulted in hyperglycemia, indicating that the reduction of blood glucose was dependent on the transplants. Diabetic mice transplanted with BM/islets demonstrated the longest survival period (130 vs. 40 days for those with islet-only transplants). The transplanted BM/islets showed signs of vascularization and migration from the renal capsule into medulla. Conclusions Our results suggest that BM precultured with human islets may enhance the survival and function of transplanted islets, thus significantly improving the therapeutic efficacy of islet transplantation for type 1 diabetes.
Clinical efficacy of endodontic protocols on reducing cultivable bacteria and endotoxin in infected root canal in patients submitted to head and neck radiotherapy: a randomised clinical trial

Abstract:Objectives: Assess the efficacy of biomechanical preparation using a reciprocating system followed by final irrigation protocols, then intracanal medication, on reducing endotoxins and cultivable bacteria of infected teeth in irradiated patients. Materials and methods: Twenty-two infected single-rooted canals in patients submitted to head and neck radiotherapy were prepared by reciprocating motion and 2.5% NaOCl. Patients were randomly divided into two groups of 11 patients before the final irrigation protocol: apical positive pressure (APP) or passive ultrasonic activation (PUA). Both groups were treated in two sessions, using Ca(OH)2 as intracanal medication for 14 days. Root canal content sampling was performed after canal access (S1), after biomechanical preparation plus the irrigation protocol (S2), and after intracanal medication (S3). Chromogenic limulus amoebocyte lysate assay measured endotoxin levels (EU/mL), and bacterial load was determined by culture techniques (CFU/mL). Results: Treatment protocols reduced bacterial counts after S2 in both groups (p = 0.01). S3 differed from S1 (p = 0.01), but not from S2 (p = 0.4). Endotoxin levels were reduced in both groups after S2 (P = 0.03) and were lower in S3 than in S2, with significant differences in the APP group (p = 0.03). Conclusions: Biomechanical preparation using a reciprocating system and 2.5% NaOCl in irradiated teeth, followed by the irrigation protocol (APP or PUA), demonstrated efficacy in reducing endodontic contaminants. Ca(OH)2 as intracanal medication should be performed in irradiated patients with infected root canals. Clinical relevance: This clinical study demonstrated that endodontic treatment in irradiated patients is efficacious at reducing bacterial load and endotoxin levels.

Abstract:Hunterian proximal artery occlusion was used in the treatment of 160 of 335 patients harboring giant aneurysms of the anterior circulation. One hundred and thirty-three of these aneurysms arose from the internal carotid arteries, 20 from the middle cerebral arteries, and seven from the anterior cerebral arteries. Ninety percent of the patients had satisfactory outcomes. The safety of internal carotid artery occlusion has been greatly enhanced by preoperative flow studies and by test occlusion with an intracarotid balloon to identify those patients who require preliminary extracranial-to-intracranial bypass, which was used in all of the middle cerebral occlusions. The anterior cerebral artery had magnificent leptomeningeal collateral flow that prevented infarction even without cross flow. Obliteration of the aneurysm by thrombosis was complete, or nearly so, in all but four patients whose treatment was completed. Analysis of poor outcome in 16 patients revealed that hemodynamic ischemic infarction was known to occur after only two of the carotid occlusions.

Abstract:To investigate the effects of Epigallocatechin gallate (EGCG) and curcumin, as a final irrigant on the fracture resistance of irradiated root that obturated with an epoxy resin sealer. Eighty mandibular premolars were randomly divided into non-irradiated (NIR) and irradiated (IR) groups. The teeth were irradiated at 2 Gy per fraction, 5 times a week for a total dose of 60 Gy over 6 weeks. All specimens were decoronated, remaining 13±1 mm root length. Two groups were subdivided into four groups (n = 10): 1) non-instrumented; the intact root served as control. The other roots were instrumented with a pro-taper NiTi rotary system. The final irrigation used was 17% EDTA, followed by three irrigation solution groups; 2) 2.5% NaOCl, 3) 0.02% EGCG, and 4) 0.1% curcumin. Root canals were filled with gutta-percha and AH plus. All specimens were embedded in self-curing acrylic resin and loaded vertically at 1 mm/min until fracture occurred. Also, sealer penetration was assessed by confocal laser scanning microscopy (CLSM). The data were evaluated statistically using two-way ANOVA and Tukey test (α = 0.05). In irradiated roots, fracture resistance of EGCG and curcumin groups did not differ from non-instrumented roots, but they were higher than the NaOCl group (P = 0.006). However, NaOCl, EGCG, and curcumin in irradiated roots had comparable strength that was higher than in the non-instrumented group (p<0.001). Difference between irradiated and non-irradiated roots was observed only for NaOCl and non-instrumented groups (P≤0.004). In irradiated roots, a higher sealer penetration was observed in EGCG and curcumin groups compared to NaOCl. EGCG and curcumin could be promising final irrigants to reverse the adverse effect of radiotherapy on the strength of irradiated roots obturated with AH Plus sealer.

Abstract:This study aimed to evaluate the effects of radiotherapy (RT) and chemoradiotherapy (CRT) on the wear and surface roughness of in vitro irradiated human enamel and dentin subjected to abrasive challenge. Enamel and dentin specimens (n = 42) were prepared from teeth donated by healthy patients and those with head and neck cancer who had received radiotherapy (RT) or chemoradiotherapy (CRT). The specimens were categorized into three groups: control, RT, and CRT (n = 14 per group for both enamel and dentin). These samples were subjected to an in vitro abrasive experiment using a brushing machine, followed by wear and surface roughness assessments with a confocal laser scanning microscope conducted before and after the abrasive challenge, considering both exposed and non-exposed areas. Statistical analysis used Shapiro–Wilk tests for normality, Wilcoxon tests for comparing two means, and Kruskal–Wallis tests. A significance level of 5% was adopted. In enamel specimens, wear profile values of CRT and RT groups were not different from the control ( p > 0.05). The RT group presents lower step values than the CRT and control groups ( p < 0.001). No significant difference in final surface roughness was observed in all groups ( p > 0.05). In dentin specimens, no significant difference in wear profile and step was observed in all groups ( p > 0.05). However, CRT and RT groups present higher values in final surface roughness ( p < 0.001). The exposure to ionizing radiation (associated or not to chemotherapy) influenced the surface roughness of dentin and the wear (step) of enamel after the in vitro abrasive challenge.

The effects of re-irradiation on the chemical and morphological properties of permanent teeth

Abstract:This study aimed to assess the in vitro effects of re-irradiation on enamel and dentin properties, simulating head and neck cancer radiotherapy retreatment. Forty-five human permanent molars were classified into five groups: non-irradiated; irradiated 60 Gy, and re-irradiated with doses of 30, 40, and 50 Gy. Raman spectroscopy, scanning electron microscopy (SEM), and energy dispersive x-ray spectroscopy (EDS) were employed for analysis. Raman spectroscopy assessed intensity, spectral area, and specific peaks comparatively. Statistical analysis involved Kolmogorov–Smirnov and One-Way ANOVA tests, with Tukey's post-test (significance level set at 5%). Significant changes in irradiated, non-irradiated, and re-irradiated enamel peaks were observed, including phosphate (438 nm), hydroxyapatite (582 nm), phosphate (960 nm), and carbonate (1070 nm) ( p < 0.05). Re-irradiation affected the entire tooth ( p > 0.05), leading to interprismatic region degradation, enamel prism destruction, and hydroxyapatite crystal damage. Dentin exhibited tubule obliteration, crack formation, and progressive collagen fiber fragmentation. EDX revealed increased oxygen percentage and decreased phosphorus and calcium post-reirradiation. It is concluded that chemical and morphological changes in irradiated permanent teeth were dose-dependent, exacerbated by re-irradiation, causing substantial damage in enamel and dentin.

Oral cancer radiotherapy affects enamel microhardness and associated indentation pattern morphology

Abstract:Objectives: The aim of this study is to determine the effects of in vitro and in vivo high-dose radiotherapy on microhardness and associated indentation pattern morphology of enamel. Materials and methods: The inner, middle, and outer microhardness of enamel was evaluated using three experimental groups: control (non-radiated); in vitro irradiated; in vivo irradiated. In vitro specimens were exposed to simulated radiotherapy, and in vivo specimens were extracted teeth from oral cancer patients previously treated with radiotherapy. Indentations were measured via SEM images to calculate microhardness values and to assess the mechanomorphological properties of enamel before and after radiotherapy. Results: Middle and outer regions of enamel demonstrated a significant decrease in microhardness after in vitro and in vivo irradiation compared to the control group (p < 0.05). Two indentation patterns were observed: pattern A-presence of microcracks around indent periphery, which represents local dissipation of deformation energy; pattern B-clean, sharp indents. The percentage of clean microindentation patterns, compared to controls, was significantly higher following in vitro and in vivo irradiation in all enamel regions. The highest percentage of clean microindentations (65%) was observed in the in vivo irradiated group in the inner region of enamel near the dentin-enamel junction. Conclusions: For the first time, this study shows that in vitro and in vivo irradiation alters enamel microhardness. Likewise, the indentation pattern differences suggest that enamel may become more brittle following in vitro and in vivo irradiation. Clinical relevance: The mechanomorphological property changes of enamel following radiation may be a contributory component of pathologic enamel delamination following oral cancer radiotherapy.

Abstract:Aim: This study aims to investigate the effects of exposure to different doses delivered by radiographic diagnostic tools on cell death pathways in isolated dental pulp stem cells on a molecular level using qRT-PCR data. Methodology: Seventy fully impacted mandibular third molars were used for the study. Extracted third molars were divided into seven groups (n = 10 each): A control group receiving no radiation, periapical (PER), panoramic (PAN), and dental tomography (CBCT) groups receiving one dose of radiation each, and periapical, panoramic, and CBCT groups receiving three consecutive doses of radiation each. Then, gene expressions in dental pulp stem cells were evaluated using the qRT-PCR this method. Results: A significant difference was found in the expression of the ULK1 gene in the dental pulp tissue samples from the PAN1 group; GABARAP, LAMP3, APAF1, BCL2, CASPASE2, and CASPASE7 genes from the PER1 group; the ULK1 gene from the PER3 group; and ATG12, ATG16L1, ATG4A ATG5, BECN1L1, GABARAP, LAMP1, LAMP3, MAP1LC3B, ULK1, APAF1, BCL2, CASPASE-2, CASPASE-3, CASPASE-7, and CASPASE-8 genes from the CBCT3 group. Conclusion: While the radiation doses we applied to the DPSC cell culture are considered low levels, there was an increase in cell death markers with increased exposure to radiation through multiple radiography procedures. Taking measures to prevent errors during radiography and avoiding repeated, unnecessary patient exposure to radiation would help minimize the damaging effects of radiation. Effect of synthetic and natural-derived novel endodontic irrigant solutions on mechanical properties of human dentin

Abstract:This study aimed to evaluate the effects of different synthetic and natural-derived root canal irrigants (6% sodium hypochlorite [NaOCl], 6% calcium hypochlorite [Ca(OCl)2] and 6.5% grape seed extract [GSE]) on dentin mechanical properties (flexural strength, ultimate tensile strength [UTS] and fracture resistance). Rectangular-shaped beams and hourglass-shaped sections obtained from mid-coronal and root dentin were treated with 6% NaOCl, 6% Ca(OCl)2 or 6.5% GSE for 30 min. The irrigant solutions were replaced every 5 min. Then, the dentin specimens were rinsed with distilled water (DW) followed by incubation with 17% EDTA for 1 min, and thoroughly rinsed with DW again. Specimens from the control group were tested without prior irrigation. After treatment with the irrigants, dentin beams were used to assess the flexural strength (n = 10) while UTS was evaluated using the root dentin hourglass-shaped sections (n = 10). Similarly, roots with 1 mm of dentinal wall thickness were obtained from human teeth and treated with the same irrigant solutions (n = 10). A compressive loading was applied to the coronal surfaces of roots until fracture. The values of each mechanical test were statistically analyzed individually by one-way ANOVA followed by Tukey HSD test (P < 0.05). NaOCl significantly reduced the mechanical properties of dentin in all mechanical tests (P < 0.05) and no statistical difference was found among Ca(OCl)2, GSE and control group (P > 0.05). It can be concluded that Ca(OCl)2 and GSE may be alternative irrigant solutions, since they do not negatively affect the dentin mechanical properties.

Abstract:OBJECTIVE To provide macroscopic and microscopic identification basis for Ligularia przewalskii. METHODS Macroscopic and microscopic identification of roots, stems and leaves of Ligularia przewalskii were carried out with the methods of paraffin section, leaves epidermal section and powder transdermal section. RESULTS The microscopic characteristics included: Open collateral vascular bundles in stem were not in the same size and arranged in two rings; Lots of fiber bundles scattered in the column parts; There were two vascular bundles in principal vein of leaf; Anticlinal wall of upper epidermis cells was thickened like moniliform, lower epidermis were like waves with irregular; The type of stoma was anomocytic; Calcium oxalate acicular crystal could be seen in the powder. CONCLUSION These features can provide references for identification of Ligularia przewalskii.
Effect of Endodontic Irrigation Protocols on Crown Fracture Resistance

Abstract:Aim: This study aimed to evaluate the fracture resistance of tooth crowns endodontically irrigated using different protocols. Materials and methods: A total of 76 bovine incisors were divided into four groups (n = 19): irrigation with distilled water (control; CON); conventional irrigation with positive apical pressure (PAP); passive ultrasonic irrigation using continuous flushing (PUI); and irrigation with PAP and heated sodium hypo-chlorite solution (PHS). The force required to fracture the crown was measured on a universal testing machine at an angle of 45°. Results: The CON group had higher fracture resistance (351.71 ± 58.66 N) than the PAP and PUI groups (140.96 ± 37.26 N; 167.49 ± 40.08 N respectively). The PHS group had the lowest fracture resistance value (115.15 ± 41.07 N). Irrigation protocols had a significant effect on crown resistance (p < 0.01). Conclusion: Teeth subjected to irrigation with heated sodium hypochlorite showed decreased resistance to crown fracture. Clinical significance: The contact of the irrigating solution with the root canal walls potentially can cause their weakness, determining crown fracture.

Abstract:Objectives To investigate the effect of cumulative doses of radiation on the pushout bond strength (BS) of a universal resin cement used in the self‐etch (SE) and self‐adhesive (SA) modes to the intraradicular dentin. Materials and Methods Forty‐eight human teeth were distributed into three groups (n = 16) according to the radiation therapy dose (RT): NoRT (no‐radiotherapy), 70RT (70 Gy), and 70 + 70RT (70 Gy + 70 Gy). The teeth were redistributed into two subgroups (n = 8), according to the adhesive mode: SE (NoRT‐SE, 70RT‐SE, and 70 + 70RT‐SE) and SA (NoRT‐SA, 70RT‐SA, and 70 + 70RT‐SA).
