Abstract:Particulate platforms capable of delivering multiple actives as well as providing diagnostic features have gained considerable interest over the last few years. In this study, magnetic polymer yolk-shell particles (YSPs) were engineered using a tri-needle coaxial electrospraying technique enabling dual-mode (ultrasonic and magnetic resonance) imaging capability with specific multidrug compartments via an advanced single-step encapsulation process. YSPs comprised magnetic Fe3O4 nanoparticles (MNPs) embedded in the polymeric shell, an interfacing oil layer, and a polymeric core (i.e., composite shell-oil interface-polymeric core). The frequency of the ultrasound backscatter signal was modulated through YSP loading dosage, and both T-1- and T-2-weighted magnetic resonance imaging signal intensities were shown to decrease with increasing MNP content (YSP outer shell). Three fluorescent dyes (selected as model probes with varying hydrophobicities) were coencapsulated separately to confirm the YSP structure. Probe release profiles were tuned by varying power or frequency of an external auxiliary magnetic field (AMF, 0.7 mT (LAMF) or 1.4 mT (HAMF)). In addition, an inversion phenomenon for the AMF-enhanced drug release process was studied and is reported. A low YSP cytotoxicity (5 mg/mL) and biocompatibility (murine, L929) was confirmed. In summary, magnetic YSPs demonstrate timely potential as multifunctional theranostic agents for dual-imaging modality and magnetically controlled coactive delivery.

Abstract:Magnetic and temperature-responsive luminescent composites based on poly (N-isopropylacrylamide), magnetic nanoparticles and Na-9[EuW10O36] were fabricated. The composites were investigated by IR, UV, XRD, TGA, SEM and TEM. The prepared nanocomposites exhibit good superparamagnetic property and thermo-responsive switchable luminescence properties. The multifunctional nanocomposites can be separated by external magnetic held. Moreover, the nanocomposites exhibit an appreciable temperature response, and the red luminescence of the nanocomposites in solution can be controlled by the temperature stimuli. The hydrogel emits strong red luminescence in solution at 25 degrees C, which can be seen by naked eyes. When the external temperature is 40 degrees C, the red luminescence almost disappears in solution. Abstract:Multifunctional colloidal nanoparticles which exhibit fluorescence, superparamagnetism, and thermosensitivity are produced by two step seed emulsifier-free emulsion polymerization in the presence of oleic acid (OA) and sodium undecylenate (NaUA) modified Fe3O4 nanoparticles. In the first step, St and NIPAM polymerize the NaUA on the surface of Fe3O4 nanoparticles to form Fe3O4/poly(St-NIPAM) nanoparticles which act as seeds for the polymerization of Eu(AA)3Phen with the remaining St and NIPAM in the second step to form an outer fluorescent layer. The core–shell composite nanoparticles show reversible dimensional changes in response to external temperature stimuli. Fluorescence spectra acquired from the composites exhibit characteristic emission peaks of Eu3+ at 594 and 619 nm and vivid red luminescence can be observed by 2-photon confocal scanning laser microscopy (CLSM). In vitro cytotoxicity tests based on the MTT assay demonstrate good cytocompatibility and the composites also possess paramagnetic properties with a maximum saturation magnetization of 6.45 emu/g and high transverse relaxivity rates (r2) of 411.78 mM−1 s−1. In vivo magnetic resonance imaging (MRI) studies show significant liver and spleen contrast with relative signal intensity reduction of about 86% 10 min after intravenous injection of the composites. These intriguing properties suggest that these nanocarriers have large clinical potential as multimodal optical/MRI probes.

Abstract:Monodisperse organic/inorganic composite microspheres with well-defined structure were prepared through the encapsulation of silica coated superparamagnetic magnetite colloidal nanoparticle clusters (CNCs) with cross-linked poly(N-isopropylacrylamide) (PNIPAM) shell. At first, the sub-micrometer-sized CNCs were fabricated by the solvothermal process, and then a silica layer was coated on the surface of CNCs through a sol-gel process, and finally, a thermoresponsive shell of PNIPAM was deposited onto the surface of the core/shell magnetic microspheres by a precipitation polymerization. The experimental results showed that the size of Fe(3)O(4) core, the thickness of SiO(2) shell, as well as volume phase transition temperature (VPTT) of PNIPAM shell could be well controlled, and this structured modulation could satisfy different requirements. The superparamagnetic behavior, high magnetization (the saturation magnetization of Fe(3)O(4)/SiO(2)/PNIPAM microspheres with a 10% cross-linking density is 41.6 emu/g), and good thermosensitivity make these composite microspheres an ideal candidate for various important applications such as in controlled drug delivery, bioseparation, and catalysis.

Abstract:Gold nanoclusters (AuNCs) with fluorescence in the Near Infrared (NIR) by both one- and two-photon electronic excitation were incorporated in mesoporous silica nanoparticles (MSNs) using a novel one-pot synthesis procedure where the condensation polymerization of alkoxysilane monomers in the presence of the AuNCs and a surfactant produced hybrid MSNs of 49 nm diameter. This method was further developed to prepare 30 nm diameter nanocomposite particles with simultaneous NIR fluorescence and superparamagnetic properties, with a core composed of superparamagnetic manganese (II) ferrite nanoparticles (MnFe2O4) coated with a thin silica layer, and a shell of mesoporous silica decorated with AuNCs. The nanocomposite particles feature NIR-photoluminescence with 0.6% quantum yield and large Stokes shift (290 nm), and superparamagnetic response at 300 K, with a saturation magnetization of 13.4 emu g-1. The conjugation of NIR photoluminescence and superparamagnetic properties in the biocompatible nanocomposite has high potential for application in multimodal bioimaging.

Abstract:Uniform magnetic nanoparticle-loaded polymer nanospheres with different loading contents of manganese ferrite nanoparticles were successfully synthesized using a flexible emulsion process. The MnFe2O4-loaded polymer nanospheres displayed an excellent dispersibility in both water and phosphate buffer saline. The effect of loading ratio and size of MnFe2O4 nanoparticles within the nanospheres on the specific absorption rate (SAR) under an alternating magnetic field was investigated. Our results indicate that a large size (here 18 nm) and a low loading ratio are preferable for a high SAR. For a smaller particle size (6 nm), the low loading ratio did not result in an enhancement of the SAR value, while a very low SAR value is expected for 6 nm. In addition, the SAR of low-content MnFe2O4 (18 nm)-loaded polymer nanospheres in the agarose gel which is simulated for in vivo environment is the highest among the samples and does not change substantially in physiological environments. This differs largely from the behaviour of singly dispersed nanoparticles. Our results have paved the way for the design of MnFe2O4-loaded polymer nanospheres as magnetic hyperthermia agents for in vivo bio-applications.

Abstract:Multifunctional hybrid nanoparticles, Fe3O4@poly[(2-dimethylamino)ethyl methacrylate]-block-poly(2-hydroxyethyl methacrylate)-graft-carbazole, with pH-responsivity, superparamagnetism and fluorescence for targeted drug delivery and release have been synthesized. The nanoparticles have a core-shell structure as determined from transmission electron microscopy, pH-responsivity as determined from hydrodynamic radius analysis, superparamagnetism as determined from vibrating samplemagnetometry and fluorescence as determined from fluorescence spectroscopy and fluorescence microscopy. The release behavior of model drug progesterone indicates that the release rate can be effectively controlled by altering the pH of the environment. The multifunctional nanoparticles could be applied extensively in targeted drug delivery and release, and with fluorescence they can serve as efficient tracers to record magnetic targeting routes. Abstract:Magnetic nanoparticles are prominent in biomedical applications due to their adjustable properties through surface functionalization and inherent magnetism. In the present manuscript, multifunctional nanoparticles were synthesized by combining polymer with a fluorophore and magnetic nanoparticles, resulting in a magnetic and fluorescent nanomaterial. A miniemulsion polymerization technique was employed, using the comonomers methyl methacrylate (MMA) and acrylic acid (AA) in the presence of superparamagnetic iron oxide nanoparticles (SPIONs) and natural turmeric extract (curcumin). Physicochemical characterizations revealed efficient encapsulation of SPIONs within the polymer matrices while maintaining superparamagnetic behavior. The nanosystems exhibited ILP values consistent with those commonly used for magnetic hyperthermia applications, ranging from 0.13 – 0.32 nHm 2 kg −1 . Additionally, a high fluorescence intensity in the blue/green region was observed, alongside no cell toxicity and substantial internalization in human osteosarcoma cells (OSAS2). Given their intrinsic magnetic and fluorescent properties exhibited, the nanosystems can be considered potential candidates for cell tracking and as a platform in nanotheranostics (MRI, magnetic hyperthermia, and photodynamic therapy).

Abstract:Background/Objectives: Magnetic Fe3O4 nanoparticles (MNPs) are becoming more important every day. We prepared MNPs in a simple one-step reaction by following the solvothermal method, assisted by azide and alkyne functionalized polyethylene glycol (PEG400) polymers, as well as by PEG6000 and the polyol β-cyclodextrin (βCD), which played a crucial role as electrostatic stabilizers, providing polymeric/polyol coatings around the magnetic cores. Methods: The composition, morphology, and magnetic properties of the nanospheres were analyzed using Transmission Electron and Atomic Force Microscopies (TEM, AFM), Nuclear Magnetic Resonance (NMR), X-ray Diffraction Diffractometry (XRD), Fourier-Transform Infrared Spectroscopy (FT-IR), Matrix-Assisted Laser Desorption/Ionization (MALDI) and Vibrating Sample Magnetometry (VSM). Results: The obtained nanoparticles (@Fe3O4-PEGs and @Fe3O4-βCD) showed diameters between 90 and 250 nm, depending on the polymer used and the Fe3O4·6H2O precursor concentration, typically, 0.13 M at 200 °C and 24 h of reaction. MNPs exhibited superparamagnetism with high saturation mass magnetization at room temperature, reaching values of 59.9 emu/g (@Fe3O4-PEG6000), and no ferromagnetism. Likewise, they showed temperature elevation after applying an alternating magnetic field (AMF), obtaining Specific Absorption Rate (SAR) values of up to 51.87 ± 2.23 W/g for @Fe3O4-PEG6000. Additionally, the formed systems are susceptible to click chemistry, as was demonstrated in the case of the cannabidiol-propargyl derivative (CBD-Pro), which was synthesized and covalently attached to the azide functionalized surface of @Fe3O4-PEG400-N3. Prepared MNPs are highly dispersible in water, PBS, and citrate buffer, remaining in suspension for over 2 weeks, and non-toxic in the T84 human colon cancer cell line, Conclusions: indicating that they are ideal candidates for biomedical applications.

Abstract:Multifunctional magnet–fluorescent nanocomposites are widely applied in biomedical applications. Incorporating biocompatible quantum dots with highly ferrimagnetic magnetic nanoparticles into one nanoplatform for achieving efficient magnetic hyperthermia therapy (MHT) is very important. Herein, we reported an amphiphilic block copolymer with a flowable hydrophobic chain to encapsulate highly ferrimagnetic magnetic nanoparticles and ZnS/InP quantum dots via a facile self-assembly method. The obtained ferrimagnetic fluorescent micelle (FMFM) exhibited a uniform diameter of about 180 nm. In stark contrast, larger aggregation (400 nm in diameter) inevitably occurred using common poly( D , L -lactide) (PLA)-based amphiphilic block copolymer with a rigid hydrophobic chain, which was readily cleared by the reticuloendothelial system (RES). The flowable FMFM exhibited long-term colloidal stability within one month and desired fluorescent stability within 84 h. Benefiting from the high ferrimagnetism, the FMFM revealed excellent magnetic heating effect and magnetic resonance imaging capability. With accurate manipulation under an external magnetic field, FMFM realized in vitro enhanced fluorescence imaging sensitivity and accumulation efficiency at the tumor region, achieving in vitro and vivo improved MHT efficacy.

Abstract:An anionic grafted conjugated polyelectrolyte was synthesized, and then magnetic nanoparticles stabilized with this material were successfully prepared by a convenient method and used for bioimaging and drug delivery. Grafted conjugated polymer (PFPAA) containing abundant carboxyl groups was attached to the surface of Fe3O4 nanoparticles through ligand exchange with oleic acid and anionic grafted conjugated polyelectrolyte-stabilized magnetic nanoparticles (MNPs@PFPANa) were then obtained by ionization with sodium carbonate. These as-synthesized nanoparticles showed good water solubility and stability, with no precipitation observed in 8 months, and had a narrow size distribution with a mean hydrodynamic diameter of 26 +/- 2.4 nm. In addition, these nanoparticles exhibited superparamagnetic properties with a saturation magnetization (M-s) of 20 emu g(-1), which sufficient for bioapplications. Upon 48 h incubation with macrophage cells, the obtained nanoparticles showed good biocompatibility of 2 pg Fe per cell as measured by ICP-OES. Furthermore, MNPs@PFPANa were low toxicity as confirmed by an MTT assay using NIH-3T3 fibroblasts. Confocal microscopy results revealed that MNPs@PFPANa can be retained in cytoplasm with high fluorescence. MNPs@PFPANa exhibited good DOX drug loading efficiency of about 10 wt% and showed good therapeutic efficiency for BGC-823 cancer cells. These results indicated such multifunctional nanoparticles would be useful in bioimaging and as drug carriers for cancer treatment.

Abstract:To overcome traditional barriers in optical imaging and microscopy, optoacoustic-imaging has been changed to combine the accuracy of spectroscopy with the depth resolution of ultrasound, achieving a novel modality with powerful in vivo imaging. However, magnetic resonance imaging provides better spatial and anatomical resolution. Thus, a single hybrid nanoprobe that allows for simultaneous multimodal imaging is significant not only for cutting edge research in imaging science, but also for accurate clinical diagnosis. A core-shell-structured coordination polymer composite microsphere has been designed for in vivo multimodality imaging. It consists of a Fe3 O4 nanocluster core, a carbon sandwiched layer, and a carbocyanine-Gd(III) (Cy-Gd(III) ) coordination polymer outer shell (Fe3 O4 @C@Cy-Gd(III) ). Folic acid-conjugated poly(ethylene glycol) chains are embedded within the coordination polymer shell to achieve extended circulation and targeted delivery of probe particles in vivo. Control of Fe3 O4 core grain sizes results in optimal r2 relaxivity (224.5 × 10(-3) m(-1) s(-1) ) for T2 -weighted magnetic resonance imaging. Cy-Gd(III) coordination polymers are also regulated to obtain a maximum 25.1% of Cy ligands and 5.2% of Gd(III) ions for near-infrared fluorescence and T1 -weighted magnetic resonance imaging, respectively. The results demonstrate their impressive abilities for targeted, multimodal, and reliable imaging.
Multifunctional NaYF4:Yb, Er@mSiO2@Fe3O4-PEG nanoparticles for UCL/MR bioimaging and magnetically targeted drug delivery

Abstract:A low toxic multifunctional nanoplatform, integrating both mutimodal diagnosis methods and antitumor therapy, is highly desirable to assure its antitumor efficiency. In this work, we show a convenient and adjustable synthesis of multifunctional nanoparticles NaYF4:Yb, Er@mSiO2@Fe3O4-PEG (MFNPs) based on different sizes of up-conversion nanoparticles (UCNPs). With strong up-conversion fluorescence offered by UCNPs, superparamagnetism properties attributed to Fe3O4 nanoparticles and porous structure coming from the mesoporous SiO2 shell, the as-obtained MFNPs can be utilized not only as a contrast agent for dual modal up-conversion luminescence (UCL)/magnetic resonance (MR) bio-imaging, but can also achieve an effective magnetically targeted antitumor chemotherapy both in vitro and in vivo. Furthermore, the UCL intensity of UCNPs and the magnetic properties of Fe3O4 in the MFNPs were carefully balanced. Silica coating and further PEG modifying can improve the hydrophilicity and biocompatibility of the as-synthesized MFNPs, which was confirmed by the in vitro/in vivo biocompatibility and in vivo long-time bio-distributions tests. Those results revealed that the UCNPs based magnetically targeted drug carrier system we synthesized has great promise in the future for multimodal bio-imaging and targeted cancer therapy.

Abstract:Folate receptor-targeted multifunctional fluorescent magnetic nanoparticles (FMNPs) composed of cores containing iron oxide nanocrystals and amphiphilic oligo(p-phenyleneethynylene) shells with multimodal imaging capability were successfully prepared through a convenient hydrophobic encapsulation approach. The iron oxide nanoparticles in the core provided T2-weighted magnetic resonance imaging (MRI), whereas the amphiphilic oligomers on the surface of the nanoparticles introduced good water-solubility, biocompatibility, excellent fluorescent properties and cancer-targeting. These nanoparticles exhibited superparamagnetic properties with saturation magnetization (Ms) of 23 emu g(-1) and a transverse relaxivity rate of 140.89 mM(-1) s(-1). In vitro studies indicated that the dual-modal FMNPs can serve as an effective two-photon fluorescent and a magnetic probe to achieve the targeted imaging of Hela cells without obvious cytotoxicity. In vivo two-photon fluorescence and MRI results demonstrated that the FMNPs were able to preferentially accumulate in tumor tissues to allow dual-modal detection of tumors in a living body. These studies provided insight in developing novel multifunctional probes for multimodal imaging, which would play an important role for theranostics in biomedical science.

Abstract:Multifunctional hybrid nanoparticles (NPs, ∼100 nm) that combine magnetic Fe3O4 nanocrystals and fluorescent carbon dots (CDs) in porous carbon (C) were successfully synthesized using a one-pot solvothermal method by simply increasing the H2O2 concentration. The resultant Fe3O4@C-CDs hybrid NPs not only demonstrate excellent magnetic responsive properties (Ms = 32.5 emu g-1) and magnetic resonance imaging ability (r = 674.4 mM-1 s-1) from the Fe3O4 nanocrystal core, but also exhibit intriguing photoluminescent (quantum yield ∼6.8%) properties including upconversion fluorescence and excellent photostability from the CDs produced in the porous carbon. The hybrid NPs can enter the intracellular region and illuminate mouse melanoma B16F10 cells under different excitation wavelengths. Meanwhile, the mesoporous carbon shell and hydrophilic surface functional groups endow the hybrid NPs with high loading capacity (835 mg g-1) for the anti-cancer drug doxorubicin and excellent stability in aqueous solutions. More importantly, the hybrid NPs can absorb and convert near-infrared (NIR) light to heat due to the existence of CDs, and thus, can realise NIR-controlled drug release and combined photothermo/chemotherapy for high therapeutic efficacy. Such nanostructured Fe3O4@C-CDs hybrid NPs demonstrate great promise towards advanced nanoplatforms for simultaneous imaging diagnostics and high efficacy therapy.
Multifunctional Magnetic Gd3+-Based Coordination Polymer Nanoparticles: Combination of Magnetic Resonance and Multispectral Optoacoustic Detections for Tumor-Targeted Imaging in Vivo

Abstract:To overcome traditional barriers in optical imaging and microscopy, optoacoustic-imaging has been changed to combine the accuracy of spectroscopy with the depth resolution of ultrasound, achieving a novel modality with powerful in vivo imaging. However, magnetic resonance imaging provides better spatial and anatomical resolution. Thus, a single hybrid nanoprobe that allows for simultaneous multimodal imaging is significant not only for cutting edge research in imaging science, but also for accurate clinical diagnosis. A core-shell-structured coordination polymer composite microsphere has been designed for in vivo multimodality imaging. It consists of a Fe3 O4 nanocluster core, a carbon sandwiched layer, and a carbocyanine-Gd(III) (Cy-Gd(III) ) coordination polymer outer shell (Fe3 O4 @C@Cy-Gd(III) ). Folic acid-conjugated poly(ethylene glycol) chains are embedded within the coordination polymer shell to achieve extended circulation and targeted delivery of probe particles in vivo. Control of Fe3 O4 core grain sizes results in optimal r2 relaxivity (224.5 × 10(-3) m(-1) s(-1) ) for T2 -weighted magnetic resonance imaging. Cy-Gd(III) coordination polymers are also regulated to obtain a maximum 25.1% of Cy ligands and 5.2% of Gd(III) ions for near-infrared fluorescence and T1 -weighted magnetic resonance imaging, respectively. The results demonstrate their impressive abilities for targeted, multimodal, and reliable imaging.

Abstract:The development of nanomaterials has drawn considerable attention in nanomedicine to advance cancer diagnosis and treatment over the last decades. Gold nanorods (GNRs) and magnetic nanoparticles (MNPs) have been known as commonly used nanostructures in biomedical applications due to their attractive optical properties and superparamagnetic (SP) behaviors, respectively. In this study, we proposed a simple combination of plasmonic and SP properties into hybrid NPs of citrate-coated manganese ferrite (Ci-MnFe2O4) and cetyltrimethylammonium bromide-coated GNRs (CTAB-GNRs). In this regard, two different samples were prepared: the first was composed of Ci-MnFe2O4 (0.4 wt%), and the second contained hybrid NPs of Ci-MnFe2O4 (0.4 wt%) and CTAB-GNRs (0.04 wt%). Characterization measurements such as UV-Visible spectroscopy and transmission electron microscopy (TEM) revealed electrostatic interactions caused by the opposing surface charges of hybrid NPs, which resulted in the formation of small nanoclusters. The performance of the two samples was investigated using magneto-motive ultrasound imaging (MMUS). The sample containing Ci-MnFe2O4_CTAB-GNRs demonstrated a displacement nearly two-fold greater than just using Ci-MnFe2O4; therefore, enhancing MMUS image contrast. Furthermore, the preliminary potential of these hybrid NPs was also examined in magnetic hyperthermia (MH) and photoacoustic imaging (PAI) modalities. Lastly, these hybrid NPs demonstrated high stability and an absence of aggregation in water and phosphate buffer solution (PBS) medium. Thus, Ci-MnFe2O4_CTAB-GNRs hybrid NPs can be considered as a potential contrast agent in MMUS and PAI and a heat generator in MH.

Abstract:Multifunctional PNIPAM/Fe3O4–ZnS hybrid hollow spheres integrating magnetism, luminescence, and temperature responses into one single entity have been successfully fabricated via a facile method. First, the cross-linked Poly (N-isopropylacrylamide) (PNIPAM) hollow spheres were synthesized via a classical method, named one-pot “self-removing” process. Then, Fe3O4 and ZnS nanoparticles were in situ precipitated on the surface of the PNIPAM hollow spheres. The PNIPAM/Fe3O4–ZnS hybrid hollow spheres exhibit magnetic property, which indicates the potential application in drug targeting. In addition, ZnS nanoparticles in the shell play an important role as a luminescent label for tracking the drug delivery. Meanwhile, the existence of PNIPAM provides a thermosensitive property. All these results indicate that the obtained composite can be expected to be used as a smart drug delivery material.
