Abstract:The application of immobilized lipase in the enzymatic production of biodiesel has shown numerous advantages. In this study, surface of Polyacrylonitrile (PAN) hollow membrane was first modified using nitrile-click chemistry in order to fit for interaction with enzyme proteins. Then sodium alginate (SA) was introduced and the membrane was post-treated by CaCl2. When the prepared PAN-PEI-SA-CaCl2 was used for lipase immobilization, the protein loading was 36.90 mg/g, and the enzyme activity reached up to 54.47 U/g, which was 2.5 times as much as that of Novozym® 435. As a result, the constructed immobilized lipase obtained a maximum biodiesel yield of 78.5%, which was 2.4 times that of the Novozym® 435 in transesterification reactions. Moreover, the biodiesel yield decreased by only 11% after the immobilized enzyme was continuously used for 20 times. This study exhibits that this technic has broad application prospects in the field of conversion of biomass resources.

Abstract:The immobilized lipase-catalyzed transesterification of Jatropha curcas L. seed oil and methanol for biodiesel production in tert-butanol was investigated The effects of different tert-butanol volume, methanol molar ratio, reaction temperature, reaction time and immobilized lipase amount on the total conversion were systematically analyzed by response surface methodology (RSM. RSM analysis showed good correspondence between experimental and predicted values. The optimal conditions for the transesterification were a reaction time of 17.355 h, a reaction temperature of 34.868 degrees C, an immobilized lipase amount of 12.435 %, a methanol molar ratio of 5.282:1, a tert-butanol volume ratio of 0.577:1. The optimal predicted yield of fatty acid methyl esters (FAME) was 88.5 % and the actual value was 88.1 %. The predicted yield of fatly acid esters and the real one was very close, indicating that the RSM based on central composite design (CCD) was adaptable for a FAME study for the present transesterification system. Moreover, the infrared spectum of biodiesel showed the characteric bands of C=O, O-C-O, C=C and (CH2)n. Furthermore, GC-linked mass spectrometry showed that biodiesel was mainly composed of the methyl esters of hexadecanoic, 9,12-octadecadienoic and 9-octadecadienoic acid

Abstract:Biodiesel as alkyl esters derived from vegetable oils has considerable advantages in terms of environmental protection. In the present work, methyl esters were produced from sunflower oil by lipase-catalyzed reactions. The effect of organic solvent on alcoholysis rate was investigated and highest conversion (80%) was found in n-hexane and petroleum ether. Among several microbial lipases tested for alcoholysis activity in a solvent-free system, the best conversion (>90%) was found with lipase from Pseudomonas fluorescens (Amano AK) at the highest molar equivalent of oil:methanol (1:4.5). To reduce inactivation of commercial immobilized lipases by methanol, a three-step protocol consisting of the stepwise addition of 1M equivalent of methanol at 5h intervals, was developed. Also with immobilized lipase from Rhizomucor miehei (Lipozyme RM IM) high conversion (>80%) was possible. Moreover, the transesterification could be conducted for at least 120h during five batch runs without significant loss of activity.

Abstract:BACKGROUND: Biodiesel is increasingly perceived as an important component of solutions to the important current issues of fossil fuel shortages and environmental pollution. Biocatalysis of soybean oils using soluble lipase offers an alternative approach to lipase-catalyzed biodiesel production using immobilized enzyme or whole-cell catalysis. The central composite design (CCD) of response surface methodology (RSM) was used here to evaluate the effects of enzyme concentration, temperature, molar ratio of methanol to oil and stirring rate on the yield of fatty methyl ester. RESULTS: Lipase NS81006 from a genetically modified Aspergillus oryzae was utilized as the catalyst for the transesterification of soybean oil for biodiesel production. The experimental data showed that enzyme concentration, molar ratio of methanol to oil and stirring rate had the most significant impact on the yield of fatty methyl ester; a quadratic polynomial equation was obtained for methyl ester yield by multiple regression analysis. The predicted biodiesel yield was 0.928 (w/w) under the optimal conditions and the subsequent verification experiments with biodiesel yield of 0.936 +/- 0.014 (w/w) confirmed the validity of the predicted model. CONCLUSION: RSM and CCD were suitable techniques to optimize the transesterification of soybean oil for biodiesel production by soluble lipase NS81006. The related lipase NS81006 reuse stability, chemical or genetic modification, and transesterification mechanism should be taken into consideration. (c) 2007 Society of Chemical Industry.

Utilization of soluble lipase offers an alternative approach to lipase-catalyzed biodiesel production using immobilized enzyme or whole-cell catalysis. Soluble lipase NS81020, produced by submerged fermentation of genetically modified Aspergillus oryzae microorganism, was first proposed here as the catalyst of biodiesel preparation with oleic acid in the biphasic aqueous-oil systems. The effect factors such as enzyme concentration, water content, temperature, molar ratio of methanol to oil, stirring rate and pH of buffer solution on the esterification rate were investigated systematically. The reaction time could be shortened with the increasing of enzyme concentration as long as the maximum enzyme absorptive capacity on the interface in the biphasic aqueous-oil systems was not achieved. The optimal water content in the biphasic aqueous-oil systems was 10 wt% by oleic acid weight. The reaction rate was enhanced with the increasing molar ratio of methanol to oil, the increasing stirring rate or the decreasing temperature. Although soluble lipase NS81020 had lower activity at pH 10.55, hydroxyl ion conduced to restrain hydrolysis of methyl ester and facilitated the reaction toward the methyl ester formation.

Abstract:The increasing global demand for biodiesel is due to the urgent need to replace fossil diesel with a fuel based on renewable energy sources. Although chemical catalysis is widely used to produce biodiesel, it uses harsh operating conditions, has high energy consumption, and generates unwanted byproducts. In this scenario, biocatalysis stands out as an efficient and environmentally friendly alternative to chemical catalysis. In biocatalysis, the use of immobilized enzymes plays an important role in the reduction in costs. In this sense, we investigated the use of the lipase produced by an Amazonian endophytic fungus in an immobilized form in the transesterification of waste cooking oil for biodiesel production. The fungus Endomelanconiopsis endophytica QAT_7AC demonstrated a high production of lipase. The lipolytic extract was precipitated in ethanol, which increased the specific enzyme activity. The lipolytic extract and the precipitated lipolytic extract were immobilized in calcium alginate beads. Immobilization efficiency was over 89%. The immobilized biocatalysts showed thermal stability and were used in the production of biodiesel using waste cooking oil and ethanol. It was possible to reuse them for up to four reaction cycles, with yields greater than 70%. These results prove the efficiency of immobilized biocatalysts in the production of biodiesel from waste oils.

Abstract:A theoretical and experimental study was carried out on the biocatalytic production of babassu biodiesel through enzymatic hydroesterification. The complete hydrolysis of babassu oil was carried out using a 1:1 mass solution at 40 °C for 4 h using 0.4% of lipase from Thermomyces lanuginosus (TLL). Then, with the use of Eversa® Transform 2.0 lipase in the esterification step, a statistical design was used, varying the temperature (25–55 °C), the molar ratio between free fatty acids (FFAs) and methanol (1:1 to 1:9), the percentage of biocatalyst (0.1% to 0.9%), and the reaction time (1–5 h) using the Taguchi method. The ideal reaction levels obtained after the statistical treatment were 5 h of reaction at 40 °C at a molar ratio of 1:5 (FFAs/methanol) using 0.9% of the biocatalyst. These optimal conditions were validated by chromatographic analysis; following the EN 14103 standard, the sample showed an ester concentration of 95.76%. A theoretical study was carried out to evaluate the stability of Eversa with FFAs. It was observed in the molecular docking results that the ligands interacted directly with the catalytic site. Through molecular dynamics studies, it was verified that there were no significant conformational changes in the studied complexes. Theoretical and experimental results show the feasibility of this process.

Abstract:The use of new materials in the field of biofuel production has been represented as a step in the development of remarkable catalysts. The use of lipases in the production of biodiesel is often seen as a cost-limiting step, as the operating expenses in recovering such catalysts can lead to unfeasible market expectations. In this study, hydroxyapatite (HAp) particles were evaluated as a support to immobilize commercial lipase, following application in ethyl ester synthesis. First, hydroxyapatite was synthesized through the co-precipitation method at constant pH and selected as a support to be used in enzyme immobilization. The characterization of the biocatalyst support materials produced was carried out using DRX, BET, FTIR, TGA, and SEM analysis. The lipase from Thermomyces lanuginosus was then immobilized in the matrices, and, subsequently, there was transesterification of the vegetable oil deodorization distillate (VODD). The biodiesel samples generated showed that they were within commercial standards, achieving ester conversion greater than 96.5%. Other properties such as density (0.87 g.cm −3 ) and viscosity (4.36 mm 2 .s −1 ) meet the specifications required by ASTM to be used as a biofuel. In the experiment planning technique, the results revealed an experimental trend and a defined behavior: a higher lipase loading in the immobilization and the use of temperatures in the range of 40–50 °C favor high conversions of ethyl esters. Thus, this confirms that the enzymatic chemical catalyst was able to form the main fatty acid esters even using a residual lipid raw material.

Abstract:Development of green, clean, and sustainable processes presents new challenges in today’s science. Production of fuel is no exception. Considering the utilisation of various renewable sources, the synthesis of biodiesel, characterised as more environmentally-friendly then fossil fuel, has drawn significant attention. Even though the process based on chemical transesterification in a batch reactor still presents the most used method for its production, enzyme catalysed synthesis of biodiesel in a microreactor could be a new approach for going green. In this research, edible sunflower oil and methanol were used as substrates and lipase from Thermomyces lanuginosus (Lipolase L100) was used as catalyst for biodiesel synthesis. Experiments were performed in a polytetrafluoroethylene (PTFE) microreactor with three inlets and in glass microreactors with two and three inlets. For a residence time of 32 min, the fatty acids methyl esters (FAME) yield was 30% higher than the yield obtained for the glass microreactor with three inlets. In comparison, when the reaction was performed in a batch reactor (V = 500 mL), the same FAME yield was achieved after 1.5 h. In order to enhance the productivity of the process, we used proposed reaction kinetics, estimated kinetic parameters, and a mathematical model we developed. After validation using independent experimental data, a proposed model was used for process optimization in order to obtain the highest FAME yield for the shortest residence time.

Application of a chitosan-immobilized Talaromyces thermophilus lipase to a batch biodiesel production from waste frying oils

Abstract:Waste frying oil, which not only harms people's health but also causes environmental pollution, can be a good alternative to partially substitute petroleum diesel through transesterification reaction. This oil contained 8.8 % of free fatty acids, which cause a problem in a base-catalyzed process. In this study, synthesis of biodiesel was efficiently catalyzed by the covalently immobilized Talaromyces thermophilus lipase and allowed bioconversion yield up to 92 % after 24 h of reaction time. The optimal molar ratio was four to six parts of methanol to one part of oil with a biocatalyst loaded of 25 wt.% of oil. Further, experiments revealed that T. thermophilus lipase, immobilized by a multipoint covalent liaison onto activated chitosan via a short spacer (glutaraldehyde), was sufficiently tolerant to methanol. In fact, using the stepwise addition of methanol, no significant difference was observed from the one-step whole addition at the start of reaction. The batch biodiesel synthesis was performed in a fixed bed reactor with a lipase loaded of 10 g. The bioconversion yield of 98 % was attained after a 5-h reaction time. The bioreactor was operated successfully for almost 150 h without any changes in the initial conversion yield. Most of the chemical and physical properties of the produced biodiesel meet the European and USA standard specifications of biodiesel fuels.
Lipase-catalyzed Transesterification of Rapeseed Oils for Biodiesel Production with a Novel Organic Solvent As the Reaction Medium

Abstract:tert-Butanol, as a novel reaction medium, has been adopted for lipase-catalyzed transesterification of rapeseed oil for biodiesel production, with which both the negative effects caused by excessive methanol and by-product glycerol could be eliminated. Combined use of Lipozyme TL IM and Novozym 435 was proposed further to catalyze the methanolysis and the highest biodiesel yield of 95% could be achieved under the optimum conditions (tert-butanol/oil volume ratio 1:1; methanol/oil molar ratio 4:1; 3% Lipozyme TL IM and 1% Novozym 435 based on the oil weight; temperature 35°C; 130rpm, 12h). There was no obvious loss in lipase activity even after being repeatedly used for 200 cycles with tert-butanol as the reaction medium. Furthermore, waste oil was also explored for biodiesel production and it has been found that lipase also showed good stability in this novel system.

Abstract:This work describes the use of an ultrasound system for the enzymatic transesterification of oils using combi-lipases as biocatalyst. The reactions were carried out evaluating the individual use of waste oil and fresh soybean oil, and the immobilized lipases CALB, TLL, and RML were used as biocatalysts. It was performed in a mixture design of three factors to obtain the ideal mixture of lipases according to the composition of fatty acids present in each oil, and the main reaction variables were optimized. After 18 h of reaction, ultrasound provided a biodiesel yield of about 90% when using soybean oil and 70% using the waste oil. The results showed that ultrasound technology, in combination with the application of enzyme mixtures, known as combi-lipases, and the use of waste oil, could be a promising route to reduce the overall process costs of enzymatic production of biodiesel.

Ultrasound affects the selectivity and activity of immobilized lipases applied to fatty acid ethyl ester synthesis

Abstract:Hydrophobic carriers can be used to improve the activity, stability and other properties of enzymes. Physical agents, like ultrasound, may also contribute to improving the dispersion and collision of the reagent molecules, decreasing the reaction time and intensifying the catalytic process. However, its effect on the enzyme activity and reaction selectivity is still not entirely understood. Here, enzyme modulation of immobilized lipases was studied under pulsed ultrasound irradiation in fatty acid ethyl ester (FAEE) synthesis for biodiesel production. Novozym 435® and two commercial lipases from Thermomyces lanuginosus and Rhizomucor miehei, immobilized on Octadecyl-Sepabeads were used as a biocatalyst in the transesterification reaction of vegetable oils and ethanol. The use of ultrasound associated with catalysis by the Novozym 435 increased the production of FAEE by about three times (from 8.9 to 26.4%) using soybean oil and changes were observed in the profile of the products. From the sonicated reaction, ethyl-palmitate production decreased from 23.4 to 11.7%, while the ethyl-linoleate content rose from 47.5 to 59.2%. On the other hand, the T. lanuginosus lipase was less affected by sonication with the overall production of FAEE increasing from 17.2 to 24.1%, with ethyl-palmitate and ethyl-linoleate content changing from 16.2 to 17.5% and 55.0 to 47.8%, respectively. Although the changes in the production yield are not too high, the main idea here was to show that ultrasound modulates the lipase activity as well as its respective selectivity. Thus, ultrasound, is responsible for changing the ethyl ester production, which can be applied to many other biochemical processes to improve or modulate their synthesis yield.

Abstract:Abstract To produce biodiesel cost-effective, low-cost, high free fatty acid (FFA) oil feedstock is desirable. But high FFA creates difficulty during the base-catalyzed transesterification process by yield loss due to the formation of soap. However, these problems are overcome by the use of an acid catalyst. The acid catalysts can directly convert both triglycerides and FFAs into biodiesel without the formation of soaps or emulsions. The shortcomings of mostly used inorganic acids are that they work well for esterification of FFA present in low-cost oil, but their kinetics for transesterification of triglycerides present in oils is considerably slower. Corrosion of equipment is another major problem associated with an inorganic acid catalyst. The usage of an organic acid catalyst of the alkyl benzene sulfonic type, like 4-dodecyl benzene sulfonic acid (DBSA) minimizes these disadvantages of inorganic acid-catalyzed transesterification. The aim of the present investigation was to reduce the reaction time of transesterification of triglycerides further by using microwaves as a heating source in the presence of DBSA catalyst to achieve higher conversions under mild operating conditions. To optimize the transesterification variables for the higher conversion of biodiesel, the response surface methodology was employed to design the experiment. By using the DBSA catalyst under microwave heating at a temperature of 76 °C, conversion close to 100% in only 30 min of reaction time was obtained using a 0.09 molar ratio of catalyst to oil and 9.0 molar ratio of methanol to oil. A modified polynomial model was developed and was adequately fitted with the experimental data and could be used for understanding the effect of various process parameters. The catalyst to oil molar ratio and reaction temperature created a stronger effect on the biodiesel production than that exhibited by the methanol to oil molar ratio. It was observed that the microwave heating process outperformed conventional heating, providing a rapid, easy method for biodiesel synthesis from triglycerides in the presence of DBSA, an organic acid catalyst. The produced biodiesel was of good quality, as all the properties were within the prescribed limits of the ASTM D6751 standard.

Abstract:Using free lipase as the catalyst for biodiesel production has drawn increasing attention in recent years due to its advantages of lower cost and faster reaction rate compared to immobilized lipase. Our previous study showed that free lipase NS81006 could effectively catalyze the methanolysis of renewable oil. Ethanol, derived from renewable biomass, has a greater potential for biodiesel production. In this paper, ethanol was explored for the first time as the acyl acceptor for free lipase-mediated biodiesel preparation. The effect of stirring rate, water content, molar ratio of ethanol to oil and ethanol adding strategy was investigated systematically during the process of free lipase NS81006-catalyzed ethanolysis. An ethyl ester yield of 90% was obtained under the optimized conditions. Further study showed that the free lipase could be repeatedly used by simple separation of the water phase from the oil phase and there was no obvious loss in lipase activity after five repeated uses.
Optimization of immobilized lipase catalyzed transesterification of cottonseed oil for biodiesel production using response surface methodology

Abstract:The process of immobilized lipase catalyzed transesterification of cottonseed oil for biodiesel production was optimized by response surface methodology.With fatty acid methyl ester conversion rate as index,the effects of reaction time,reaction temperature,immobilized lipase amount(on the basis of cottonseed oil mass),molar ratio of methanol to cottonseed oil and volume ratio of tert-butanol to cottonseed oil on the conversion rate were studied.The optimal reaction conditions were determined as follows: reaction time 19.128 h,reaction temperature 37.801℃,immobilized lipase amount 12.070%,molar ratio of methanol to cottonseed oil 4.949,volume ratio of tert-butanol to cottonseed oil 0.323.The verification test showed that the conversion rate reached 92.9%,which had a high degree of agreement with the response suface methodology predicted value(94.3%).
Lipase-catalyzed Production of Biodiesel from Several Oils in a Novel Reaction Medium

Abstract:Tert-butanol was adopted as a reaction medium for lipase-catalyzed methanolysis of oils for biodiesel production,in which both the negative effects caused by excessive methanol and by-product glycerol could be eliminated.The optimum conditions of methanolysis of rapeseed oil were as follow:tert-butanol/oil(volumetric ratio)1:1,methanol/oil(molar ratio)4:1,3% Lipozyme TLIM and 1% Novozym 435 based on oil weight,35℃,130 r/min and 12 h.The highest biodiesel yield was 95% and the lipases could be reused 200 cycles without obvious loss of their activity.The technology was further tested in a pilot plant and the product was up to the biodiesel standards of US(ASTM PS121-99)and Germany(DINE51606).Furthermore,soybean oil,tung oil,cottonseed oil,sapium fat and acidified waste oil were also explored for biodiesel production,and lipases also showed good stability in tert-butanol reaction medium.

Abstract:The purpose of this study was to find the optimum conditions in synthesis of Biodiesel from Jatropha Curcas with heterogeneous catalyst of CaO and MgO by transesterification reaction using microwave. The research was conducted in five stages, that is determining acid number of Jatropha oil; esterification process; preparation of CaO and MgO catalysts; synthesis of biodiesel via transesterification reaction in various catalyst concentration and various times; identification and characterization of biodiesel involves the synthesis, testing TLC, GC-MS analysis, the index test bias, the density test, test the viscosity and acid number test. The results showed that optimum conditions the synthesis of biodiesel from Jatropha oil through transesterification reaction using microwaves produce yield 92.39% ± 0.04, with a catalyst concentration of 7.5% CaO-MgO. Furthermore, identification of compounds contained in biodiesel using GC-MS showed that biodiesel contains palmitoleic methyl compound (2.40%), methyl palmitate (21.20%), methyl oleate (62.40%), and methyl stearate (14.00%). We found also that the refractive index of the biodiesel is 1.465, the density of 0.888 g/mL, viscosity 4.23 cSt, and acid number is 0.58 mg KOH/g.
