Abstract:Longer and more intense heat and drought stresses will occur in terrestrial ecosystems in the future. Although the effects of individual heat or drought stress on wheat plants have been largely explored, the regulatory effect of nitrogen (N) on winter wheat under heat, drought, and combined stresses and whether N alleviates damage to wheat plants caused by these stresses remain unclear. Therefore, the objective of the present study was to investigate the growth, photosynthesis, antioxidant enzyme and N metabolism-related enzyme activity, cell membrane system, osmoregulatory substance, and yield responses to heat, drought, and combined stresses in wheat plants and to clarify the regulatory effects of N on the growth, physiological and biochemical characteristics, and yield of wheat plants under stress conditions. The results showed that wheat plant exposure to individual heat or drought stress reduced photosynthesis and N metabolism-related enzyme activities and increased antioxidant enzyme activities, electrolyte leakage (EL), and the contents of MDA (malondialdehyde) and O2− (superoxide anion). The above parameters showed typical superposition effects under combined stress. Under individual heat or drought stress, wheat plants treated with a medium (N2) or high (N3) N supply maintained higher photosynthesis and N metabolism-related enzyme activities than did those treated with a low N supply (N1). Enhanced heat and drought tolerance in wheat plants under an appropriate N supply may be attributed to improved antioxidant capacity, as exemplified by increased activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), glutathione reductase (GR) and ascorbate peroxidase (APX), and to enhanced osmoregulation capacity, as signified by increased contents of soluble sugar (SS), soluble protein (SP), and proline (Pro). Variable importance in projection (VIP) analysis indicated that efficient SOD, POD, CAT, and GR activities and an increased Pro content had superior potential to alleviate heat, drought, and combined stress stresses in wheat plants, and the improvements in growth and grain yield in wheat plants further confirmed the oxidative stress alleviation and stress tolerance enhancement. However, positive effects of N on wheat growth and grain yield under combined stress were usually observed under a low N supply. These results may facilitate future research on the effects of N fertilizer on the stress resistance of winter wheat.Graphical Abstract

Abstract:Two strains of spring wheat (Triticum aestivum L-high productive Gaoyuan 602 and drought-tolerating Dingxi 24) were grown at two different CO2 concentrations (350 and 700 μmol·mol-1) and three soil water levels (80%, 60%, 40%FWC) in open-top-chambers from sowing until final harvest. We examined the response of photosynthesis and stomatal resistance to increasing CO2 concentration and drought and found that the photosynthesis rate and stomatal resistance and transpiration efficiency in the two spring wheats had increased. However, the positive effect of high CO2 on Dingxi 24 was bigger than that on Gaoyuan 602. Dingxi 24 showed higher water saving effect. Based on the analysis of the results, we predict that drought-tolerating spring wheat cultivars will have selective advantages in future high CO2 environment.

Abstract:Stomatal density (SD) and stomatal index (SI) on the abaxial flag leaf epidermis, gas exchange of flag leaves and yield of six wheat ( Triticum aestivum L.) cultivars differing in drought resistance were studied in plant growth chamber. There were significant differences among cultivars in SD, SI, epidermal cell density (ED), photosynthetic rate (P n ), transpiration rate (E), instantaneous transpiration efficiency (ITE), shoot dry weight yield (Ys), grain yield (Yd) and whole growth season water use efficiency (WUE), but not in stomatal conductance (g s ). Wheat cultivars with moderate drought resistance had higher P n and Yd than cultivars with strong or weak drought resistance. SD was negatively correlated with ITE (r=−0.85) and WUE (r=−0.84), but not with P n , E and Yd. There were significant relationships between ED and Yd (r=−0.92), between SI and P n (r=0.84). These results revealed that breeding wheat cultivar only to raise drought resistance might cause yield decrease when water stress was not too severe. Wheat cultivars with lower SD might improve ITE and WUE, but P n and yield (Ys and Yd) were not influenced significantly. Greater ED was not beneficial to Yd. SI might be a criterion to use in selecting parents for producing high-yielding wheat cultivars.
Effects of Low Nitrogen Supply on Relationships Between Photosynthesis and Nitrogen Status at Different Leaf Position in Wheat Seedlings

Abstract:Hydroponic experiments were conducted to investigate the effects of low nitrogen (N) nutrition on photosynthesis and its relationships with N status in wheat (Triticum aestivum L.). Two wheat cultivars, Zaoyangmai and Yangmai158, differing in low N nutrition tolerances, were used. The results show that under low N nutrition the area of the first top leaf was significantly reduced, while there was no significant difference in the top second and third leaf areas compared with the control for either cultivar. The net photosynthetic rate and chlorophyll content were significantly reduced in the top three leaves of Zaoyangmai, while no significant difference in these factors was observed in the top first and second leaves of Yangmai158 compared with control under N-limited conditions. The effective quantum yield of photosystem II (PSII) photochemistry and the maximal quantum yield of PSII photochemistry were only slightly altered in both cultivars, indicating that PSII was not damaged by low N nutrition. In addition, the non-photochemical quenching coefficient increased significantly in the top three leaves of Zaoyangmai, and only in the top third leaf of Yangmai158 under N-limited conditions. Furthermore, the ribulose 1,5-bisphosphate carboxylase/oxygenase (Rubisco) and soluble protein contents decreased significantly in the top second and third leaves of Zaoyangmai, while no significant difference was observed in the top first and second leaves of Yangmai158 between low N nutrition and control. We concluded that in Yangmai158, N status changed less, and it maintained almost normal photosynthesis in young leaves, thus Yangmai158 could be more tolerance to low N nutrition.
[Effects of Nitrogen Supply on Flag Leaf Photosynthesis and Grain Starch Accumulation of Wheat from Its Anthesis to Maturity under Drought or Waterlogging].

Abstract:In this paper, a cement pool culture experiment with three water treatments (waterlogging, drought, and moderate water supply) and two nitrogen levels (120 and 240 kg x hm(-2)) was conducted to study the effects of nitrogen supply on the flag leaf photosynthesis and grain starch accumulation of two wheat varieties from anthesis to maturity under soil drought and waterlogging. In comparing with moderate water supply, soil drought and waterlogging reduced the photosynthesis rate (Pn) and SPAD of flag leaf and dry matter accumulation. Nitrogen supply under drought increased Pn and SPAD, while that under waterlogging was in adverse. The total soluble sugar content in grain was reduced under both drought and waterlogging, while that in leaf was decreased under waterlogging but increased under drought. Under waterlogging, increasing nitrogen application rate reduced starch accumulation. The unit grain weight and the yields of grain and starch were reduced under both drought and waterlogging, but nitrogen application favored them under drought while in adverse under waterlogging. It was indicated that both leaf photosynthesis and grain starch accumulation could be regulated by nitrogen supply under stress of soil drought or waterlogging from anthesis to maturity of wheat.

Abstract:Extreme drought stress is often accompanied by heat stress after anthesis in winter wheat. Whether nitrogen (N) can mitigate the damage caused by combined stress on wheat plants by regulating root physiological characteristics is still unclear. Thus, this study aimed to study the effects of combined heat and drought stress on photosynthesis, leaf water relations, root antioxidant system, osmoregulatory, and yield in wheat to reveal the physiological mechanism of N regulating the adverse impacts of combined stress on wheat. Heat and drought stress markedly reduced photosynthesis, leaf water content, root vitality, and bleeding sap. The combination of heat and drought strengthens these changes. Within a certain stress range, the increase in soluble sugar and proline contents and the activities of superoxide dismutase, peroxidase, catalase, and ascorbate peroxidase under combined stress effectively alleviated the oxidative damage. Compared with those under high N application (N3), wheat plants under low N application (N1) maintained higher yield and yield components under combined stress; the number of grains per spike, 1000‐grain weight, and yield increased by 13.65%, 9.07%, and 15.33%, respectively, under N1 compared with those under N3 treatment, which may be attributed to the greater maintenance of photosynthesis, leaf water status, root vitality, and antioxidant and osmoregulation capacities. In summary, reduced N application mitigated the damage caused by combined heat and drought stress in wheat by improving root physiological characteristics and enhanced adaptability to combined stress, which is an appropriate strategy to compensate for yield losses.

Low Nitrogen Priming Enhances Photosynthesis Adaptation to Water-Deficit Stress in Winter Wheat (Triticum aestivum L.) Seedlings

Abstract:Drought is among the main environmental stressors that reduces wheat production. Nitrogen (N) availability affects plant adaptation to abiotic stress, but the effect of low N (LN) on drought tolerance is unclear. To identify the effect of LN priming on water-deficit stress tolerance in wheat seedlings, we primed cultivar Yangmai158 with 0.25 mM N for 7 days, and then added 20% polyethylene glycol 6000 as a water-deficit treatment for 5 days. The net photosynthetic rate (Pn), plant biomass, and plant growth rate (GR) were significantly reduced under water-deficit conditions; such decreases were less severe in LN-primed (LND) plants than non-primed (CKD) plants. The leaf relative water content (LRWC) decreased under water-deficit conditions, which in turn led to a reduced transpiration rate, stomatal conductance, and intercellular CO2 concentration (C i), causing a stomatal limitation on photosynthesis. LN priming also enhanced root growth, resulting in a higher LRWC and less stomatal limitation in LND plants than CKD plants. PSII quantum efficiency, photochemical quenching, and maximum PSII quantum efficiency were reduced under water-deficit conditions, indicating photoinhibition. However, LN priming increased the electron flux to photorespiration and the Mehler pathway, reducing photoinhibition. In conclusion, LN priming improved the leaf water status and increased alternative electron flux to attenuate photoinhibition, thus alleviating the inhibition of photosynthesis, and growth due to water deficiency.
Enhanced Rubisco Activation Associated with Maintenance of Electron Transport Alleviates Inhibition of Photosynthesis under Low Nitrogen Conditions in Winter Wheat Seedlings.

Abstract:Studying the response of photosynthesis to low nitrogen (N) and the underlying physiological mechanism can provide a theoretical basis for breeding N-efficient cultivars and optimizing N management. We conducted hydroponic experiments using two wheat (Triticum aestivum) cultivars, Zaoyangmai (low N sensitive) and Yangmai158 (low N tolerant), with either 0.25 mM N as a low N (LN) treatment or 5 mM N as a control. Under LN, a decrease in net photosynthetic rate (Pn) was attributed to reduction in the maximum Rubisco carboxylation rate, which then accelerated a reduction in the maximum ribulose-1,5-bisphosphate regeneration rate, and the reduction in Pn was 5-35% less in Yangmai158 than in Zaoyangmai. Yangmai158 maintained a 10-25% higher Rubisco concentration, especially in the upper leaves, and up-regulated Rubisco activase activity compared with Zaoyangmai to increase the Rubisco activation to sustain Rubisco carboxylation under LN conditions. In addition, Yangmai158 increased electron flux to the photorespiratory carbon oxidation cycle and alternative electron flux to maintain a faster electron transport rate and avoid photodamage. In conclusion, the LN-tolerant cultivar showed enhanced Rubisco activation and sustained electron transport to maintain a greater photosynthetic capacity under LN conditions.
Efficient Physiological and Nutrient Use Efficiency Responses of Maize Leaves to Drought Stress under Different Field Nitrogen Conditions

Abstract:Inadequate water and nitrogen (N) supplies can limit the productivity of maize. Climate change will likely increase drought in many regions on a global scale. The determination of N fertilizer rates under field drought conditions will be critical toward the reduction of agricultural risk. For this study, drought-resistant/sensitive cultivars were selected as experimental samples. Our results revealed that drought stress reduced the relative water content (RWC) of leaves, which resulted in leaf curling, while decreasing photosynthesis levels and N accumulation. In contrast to those without N treatments, the application of N significantly increased grain yields by 26.8% during the wet year but increased only by 5.4% during the dry year. Under the same N levels, the reduction in yield caused by drought increased with the increased application of N. This was because the application of the N fertilizer translated to increase the leaf area and transpiration, exacerbated the soil water loss and induced a leaf curling state in maize, which had deleterious effects on photosynthesis and N absorption. During the dry year, the yields of drought-sensitive cultivars were even less than those without the application of N. Compared with those of drought-sensitive cultivars, the RWCs of drought-resistant cultivars decreased more rapidly, and they entered the state of leaf curling earlier. Thus, N fertilizer inputs should be reduced, and the extent of N fertilization for drought-sensitive cultivars should be reduced even further.

Abstract:Elevated CO2 concentrations and persistent drought are predicted to inhibit the photosynthetic performance and yield of crops. Such effects vary considerably between species groups. Modern cultivated (Triticum aestivum L., AABBDD) and wild (Triticum dicoccoides Korn L., AABB and Triticum monococcum L., AA) species of wheat were subjected to elevated CO2 concentration (ambient concentration +200 μmol mol-1) and drought (well watered: 75-85% of the field water capacity; drought: 50-60% of the field water capacity) at open-top chamber experimental facilities. Elevated CO2 concentration decreased the limitation of stomatal morphology traits on stomatal conductance regulation. This could compensate the disadvantage of plants who has low stomatal density and large single stomatal area as well as low leaf water use efficiency such as modern wheat Z9023 in drought acclimation in the future CO2 rising world. Moreover, elevated CO2 concentration largely increased the dependence of light harvesting and electron transportation performance per photosynthesis system II reaction center, maximum rubisco carboxylation rate, and maximum Ribulose-1,5-bisphosphate regeneration rate on leaf nitrogen concentration across the selected wheat species and water regimes. Modern cultivated cultivars Z9023 and CH58 have higher photosynthetic performance per unit of leaf nitrogen than wild species under elevated CO2 concentrations. The increasing CO2 may present opportunities to breeders and possibly allow them to select for cultivars with better photosynthetic nitrogen use efficiency response to future CO2 rising climate.

Effects of nitrogen supply on nitrogen metabolism and grain protein accumulation of wheat under different water treatments

Abstract:Soil water stress is the major ecological factor limiting grain quality formation of wheat, while elucidating the physiological mechanism and regulation principles of this formation is of important significance for understanding grain quality physiology and guiding wheat cultural management. In this paper, a cement pool culture experiment was conducted to study the effects of nitrogen supply rate on nitrogen metabolism and grain protein accumulation of two wheat varieties differed in grain protein content under drought and waterlogging. Three water treatments were established from anthesis to maturity, i.e., waterlogging, drought, and moderate water supply (CK). In each treatment, two nitrogen levels of 120 and 240 kg.hm-2 were implemented. Compared with CK, both drought and waterlogging significantly reduced the nitrate reductase (NR) activity in flag leaf, and the contents of total nitrogen and free amino acid in leaf, stem and sheath. Drought increased, while waterlogging reduced the contents of total nitrogen and free amino acid in stem and sheath, and that of protein in grain. Under drought and waterlogging, nitrogen supply increased the NR activity in flag leaf, the total nitrogen and free amino acid contents in leaf, stem and sheath, and the free amino acid and protein contents in grain. Both drought and waterlogging stress reduced grain yield and protein yield. Nitrogen supply increased grain yield under CK and drought, but reduced it under waterlogging, illustrating that nitrogen supply could obviously regulate the nitrogen metabolism and grain protein accumulation of wheat from anthesis to maturity under drought or waterlogging.

Abstract:More frequent and more intense heat waves and greater drought stress will occur in the future climate environment. Short-term extreme heat and drought stress often occur simultaneously after winter wheat anthesis, which has become the major constraint threatening future wheat yield. In this study, short-term heat, drought and their combination stress were applied to wheat plants after anthesis, and all wheat plants were restored to the outdoor normal temperature and full watering after stress treatment. The aim of the current study was to evaluate the role of nitrogen (N) in modulating the effects of post-anthesis short-term heat, drought and their combination stress on photosynthesis, N metabolism-related enzymes, the accumulation of N and protein and growth, as well as on the yield and water (WUE) and N use efficiency (NUE) of wheat after stress treatment. The results showed that compared with low N application (N1), medium application (N2) enhanced the activities of nitrate reductase (NR) and glutamine synthase (GS) in grains under post-anthesis heat and drought stress alone, which provided a basis for the accumulation of N and protein in grains at the later stage of growth. Under post-anthesis individual stresses, N2 or high application (N3) increased the leaf photosynthetic rate (An), PSII photochemical efficiency and instantaneous WUE compared with N1, whereas these parameters were usually significantly improved by N1 application under post-anthesis combined stress. The positive effect of increased An by N application on growth was well represented in a higher green leaf area, aboveground dry mass and plant height, and the variation in An can be explained more accurately by the N content per unit leaf area. Short-term heat, drought and combined stress after anthesis resulted in a pronounced decrease in yield by reducing grain number per spike and thousand kernel weight. The reduction in NUE under combined stress was higher than that under individual heat and drought stress. Compared with N1, N2 or N3 application significantly prevented the decrease in yield and NUE caused by post-anthesis heat and drought stress alone. However, N1 application was conducive to improving the productivity, WUE and NUE of wheat when exposed to post-anthesis combined stress. The current data indicated that under short-term individual heat and drought stress after anthesis, appropriately increasing N application effectively improved the growth and physiological activity of wheat compared with N1, alleviating the reduction in yield, WUE and NUE. However, under combined stress conditions, reducing N application (N1) may be a suitable strategy to compensate for the decrease in yield, WUE and NUE.

Abstract:Taking drought-sensitive wheat cultivar Wangshuibai and drought-tolerance cultivar Luohan 7 as test materials, a hydroponic experiment was conducted to study the effects of drought stress on root system morphology, physiological characteristics and leaf photosynthesis of wheat seedlings, aimed to elucidate the adaptation mechanisms to drought stress. Under drought stress, the root vitality of the cultivars increased markedly, but the root number and root surface area decreased. Drought stress decreased relative water content and increased the ratio of bound water to free water in leaves of Wangshuibai, but had less effects on Luohan 7. Drought stress decreased, the leaf chlorophyll content, Pn g(s), Ci, and transpiration rate of the two cultivars, but had no significant effects on leaf chlorophyll content and Pn of Luohan 7. Drought stress decreased the leaf area of the two cultivars and the root biomass, shoot biomass, and plant biomass of Wangshuibai, but had no significant effects on Luohan 7. The results indicated that under drought stress, drought-tolerant wheat cultivar was able to compensate decreased root absorption area and retain higher root water uptake capability via enhancing root vitality and maintaining higher root biomass, and further, to keep higher leaf photosynthetic area and Pn to mitigate the inhibition of drought on wheat seedlings growth.

Abstract:In semi-arid areas of north-west China, grain yields of wheat (Triticum aestivum L.) are higher in recently bred cultivars than those released six decades earlier. The gas exchange, chloroplast activity and yield of six spring wheat cultivars grown in the 1950s, Hst and Gs96 (early), in the 1970s, Gy602 and Dx24 (intermediate), and in the 1990s, Gc20 and Lc8275 (modern) were compared with adequate water (WET) and drought stress (DRY) to determine the effects of drought stress among the cultivars. The results showed that in the WET treatment, the modern cultivars had significantly higher rates of leaf gas exchange, photosystem 2 (PS2) maximal photochemical efficiency, actual quantum yield of PS2 (?PS2), photochemical quenching of chlorophyll ((q)p) and lower non-photochemical quenching (NPQ) than early cultivars, but had significantly lower gas exchange rates, intercellular CO2 concentration, ?PS2, (q)p and NPQ in the DRY treatment. In the WET treatment, the grain yield of early cultivars was significantly lower (10%) than intermediate cultivars, but was significantly higher (17%) than intermediate cultivars in the DRY treatment. The modern and intermediate cultivars had more sensitive stomata to water shortage, but the decreased activity of the PS2 reaction centre helped avoid damage from photoinhibition in these cultivars.

Abstract:This study aims to understand the influence of chlorophyll fluorescence parameters on the yield of winter wheat in some areas of China. Nitrogen (N) application is believed to improve photosynthesis in flag leaf, which ultimately increases the final yield. The experiment was conducted in the wheat experimental base of Shanxi Agricultural University in Taigu, Shanxi Province, China; before sowing, four N application rates were set—N0, N120, N150, and N210 kg ha–1 of the Yunhan-20410 variety from 2019 to 2022. The results from different parameters of research showed that the organic manure partial substitution for chemical fertilizer increased post-anthesis N uptake by 16.4 and 81.4%, thus increasing the post-anthesis photosynthetic capacity and delaying leaf senescence. N150 treatment can improve dry matter (DM) accumulation, thus promoting the increase of the yield. The maximum net photosynthesis PN value of the booting stage and flowering stage indicated that nitrogen application could significantly improve the photosynthetic rate of wheat leaves, among which medium nitrogen treatment had the most significant promoting effect. The single-photon avalanche diode (SPAD) value of the leaf of wheat in each treatment increased rapidly in a small range from the jointing stage to the booting stage, respectively. The grain yield under N fertilizer partial substitution for N fertilizer treatment increased by 23%. According to the different significance test, the effects of nitrogen application rate on net photosynthesis PN of winter wheat were extremely significant at all growth stages, indicating that changing the population distribution mode and nitrogen level could effectively improve leaf photosynthetic performance and that N150 level was the best.

Abstract:This study aimed to analyze the combined impact of CO2 and drought stress at the flowering stage on carbon (C), nitrogen (N), and CN ratios in leaves, stem, and grains of bread wheat. Six diverse bread wheat genotypes, comprised of two commercial checks, two landraces, and two synthetics derivatives, were grown at two levels of CO2, i.e., 400 ppm and 800 ppm, and drought stress was imposed at the flowering stage through progressive soil drying. Stem, leaf, and grain samples were taken at maturity and concentrations of C and N were determined. Our results indicate that the threshold value of fraction of transpirable soil water (CFTSW) at which it diverges towards closure of stomata was different among genotypes and a higher range of values was estimated under elevated CO2. Drought significantly increased C levels in leaves and N levels in grains but decreased N levels in leaves, which increased CN ratios in leaves. In contrast, drought significantly reduced CN ratios in grains. Genotypes differed significantly in N content in grains, where the landrace derivative L2 maintained the highest N content. Moreover, pronounced changes in leaf N and CN ratios were induced by the combination of elevated CO2 and drought stress. Additionally, combined correlation and biplot analyses indicate a strong positive association of grain CN (GCN) with grain number, weight, and grain yield. These effects possibly interact with drought to strongly interfere with the impact of elevated CO2. The differential performance of the tested genotypes shows that selection of appropriate germplasm is essential to maintain agricultural production.

Abstract:Root respiration was measured in vivo by infrared gas analysis of complete root systems in drought-sensitive (Longchun 8139-2) and drought-tolerant (Dingxi 24) spring wheat (Triticum aestivum) cultivars. Plants were grown in sand culture for 3 weeks under the following regimes: (a) water sufficiency, (b) moderate drought stress or (c) severe drought stress. The aim was to study the acclimation to drought stress in terms of changes in root growth, root respiration and energy requirement for water uptake. Drought stress increased the root:shoot ratio of both varieties. Roots of water-sufficient treatments respired 4.03 and 2.87mg glucoseh−1g−1 in Longchun 8139-2 and Dingxi 24, respectively. However, severe drought stress enhanced root respiration of Longchun 8139-2 and Dingxi 24 to 6.52 and 3.01mg glucoseh−1g−1, respectively. Compared with water-sufficient plants, drought-sensitive spring wheat (Longchun 8139-2) used a relatively higher amount of glucose to absorb water in drought stress treatment, especially in severe drought stress. The drought-resistant spring wheat Dingxi 24 used lower amount of glucose to absorb water in drought stress conditions, relative to drought-sensitive plants. In the arid and semi-arid region of the Losses Plateau, drought-resistant wheat species have many advantages over drought-sensitive species.

Abstract:The effects of nitric oxide (NO) on the photosynthetic adaptation mechanisms of wheat plants in waterlogging during the flowering stage are poorly understood. Field and pot experiments using two cultivars were conducted with three treatments: waterlogging (WL), waterlogging plus NO donor sodium nitroprusside (WLsnp), and adequate water (CK). The results indicated that the WLsnp and CK treatments exhibited significantly higher 1000‐kernel weight and yield than the WL treatment because of high photosynthetic potential(P<0.05). We found that the photosynthetic performance, including photosynthetic rate (Pn), stomatal conductance (gs), mesophyll conductance (gm), carbon dioxide concentration at the carboxylation site (Cc), maximum carboxylation rate (Vcmax), maximum electron transfer rate (Jmax), actual electron transfer rate (J), actual PSII efficiency (ΦPSII) and potential maximum efficiency in PSII (Fv/Fm), was significantly improved in the WLsnp treatment compared to the WL treatment, both during waterlogging and after de‐waterlogging. Little difference was observed in the photosynthetic performance between the WLsnp and CK treatments during waterlogging for cultivar YM15 and after de‐waterlogging for cultivar YM24. Further analysis indicated that gs, gm and J were identified as key physiological indicators that synergistically regulate Pn of waterlogged wheat plants. Overall, the improvement of wheat's waterlogging resistance capacity after spraying NO is mainly related to high gs, great gm, and high J.

Abstract:Characterizing the responses of plant growth and nitrogen (N) uptake to low-nitrogen conditions will provide information towards elucidating the acclimation mechanism of N-efficient wheat seedlings. Two winter wheat cultivars, Yangmai158 (low-nitrate tolerant) and Zaoyangmai (low-nitrate sensitive), were supplied either 0.25 mmol L-1 nitrate (low nitrate nutrition; LN) or 5 mmol L-1 nitrate (control; CK), to investigate the responses of root morphology and nitrate transporter expression in a hydroponic experiment. Decreases in plant dry weight and N content were greater in Zaoyangmai than in Yangmai158, suggesting that Yangmai158 has a better growth status and N-storage ability in low-nitrate treatment. Additionally, root dry weight and uptake area increased to a greater extent in Yangmai158, especially in nodal roots. The decrement in nitrate concentration stimulated simultaneous upregulation of TaNRT1.1, TaNRT2.1 and TaNRT2.2 in Yangmai158, followed by upregulation of TaNAR2.1 in low-nitrate conditions. Plasma membrane H+-ATPase activity. (PM H+-ATPase), which is involved in N-uptake energy supply, increased significantly in nodal roots of Yangmai158, consistent with increasing TaHA1 transcript abundance. Furthermore, the activities of nitrate reductase (NR) and glutamine synthetase (GS) were much higher in Yangmai158 initially, which could be a feedback signal for N-uptake. However, no difference of NR and GS activity between LN and CK was found in Zaoyangmai. These results suggest that larger root extension and nitrate transporter upregulation are the major reasons for the superior acclimation of Yangmai158 to nitrogen deficient growth conditions.

Abstract:Water and nitrogen (N) are recognized as the primary determinants influencing the development and output of winter wheat. They affect the growth of winter wheat not only through their own changes, but also through their interaction. The translocation and accumulation of pre- and post-anthesis assimilates (including dry matter and N) are important physiological processes in winter wheat, which are closely related to the resource use efficiency and yield. However, a dearth of research exists that examines the complex interplay between varying water levels, especially severe water deficit, and N deficiency levels on the growth dynamics of winter wheat. The purpose of this study was to quantify and compare the effects of different degrees of water deficit and N interaction on assimilate translocation, water and N use efficiency and yield of winter wheat. A four-year longterm field experiment conducted under the rain-out shelter including four irrigation and two N levels was launched during 2019-2023. This method avoided the impact of precipitation on water treatment and helped to achieve serious water deficit treatment. The findings indicated that the higher N application improved the wheat's ability by 49.6 % - 362.3 % to utilize the available soil water. The pre- and post-anthesis translocation and accumulation of assimilates (including dry matter and N) in winter wheat increase alongside elevated levels of water and N application, except under severe water deficit treatment. Plants under mild to moderate water deficit were better able to translocate the pre-anthesis assimilates. However, the severe water deficit hindered the re-translocation of assimilates before anthesis. Providing additional N during periods of severe water scarcity leads to a 13.4 % - 44.7 % reduction in water use efficiency (WUE). Furthermore, both the irrigation water use efficiency and WUE diminished by 10.3 % - 60.5 % and 4.5 % - 41.4 % with higher levels of irrigation, while improved by 24.4 % - 48.2 % and 12.6 % - 39.4 % with higher N application rates. Conversely, N partial factor productivity and N use efficiency followed the opposite trend. Similar to WUE, increasing N application under severe water deficit would result in a yield reduction of 38.7 %. The reduction in crop yield resulting from severe water stress was primarily ascribed to the decline in the kernels number per spike (up to 74.9 %), with the reduction in spike number per unit area following closely behind (up to 29.1 %). This integrated approach, combining resource use efficiency with a detailed assessment of assimilate dynamics, provides a holistic view of how water and N management strategies impact winter wheat performance. The findings will offer precise insights for developing targeted irrigation and N scheduling strategies for sustaining both winter wheat production and environmental sustainability.
