Abstract:E. coli mutants that fail to grow at 20 degrees C were isolated, and three such mutants were studied in detail. Two of these are defective in 50S ribosome biosynthesis at low temperatures and accumulate 43S and 32S particles, respectively. The third mutant produces reduced amounts of 50S ribosomes and accumulates 21S particles at low temperatures. The 43S and 32S particles contain 23S RNA and appear to be precursors of 50S ribosomes. The 21S particles contain 16S RNA. The mutation that causes accumulation of 32S particles is linked to the spc locus. Isolation and characterization of coldsensitive mutants should provide a fruitful approach to the study of in vivo ribosomal assembly.
The Antioxidant Effect of Small Extracellular Vesicles Derived from Aloe vera Peels for Wound Healing

Abstract:Background: Extracellular vesicles (EVs) derived from plants have emerged as potential candidates for cosmetic and therapeutic applications. In this study, we isolated EVs from Aloe vera peels (A-EVs) and investigated the antioxidant and wound healing potential of A-EVs. Methods: A-EVs were isolated by ultracentrifugation and tangential flow filtration and were characterized using transmission electron microscopy, nanoparticle tracking analysis. The cytotoxicity and cellular uptake of A-EVs were investigated by WST-1 assay and flow cytometry. The antioxidant effect of A-EVs was evaluated by superoxide dismutase (SOD) activity assay and cellular antioxidant activity (CAA) assay. The wound healing potential was assessed by in vitro scratch assay using human keratinocytes (HaCaT) and fibroblasts (HDF). The expression of nuclear factor erythroid 2-related factor 2 (Nrf2) and their associated genes was analyzed by quantitative RT-PCR. Results: A-EVs displayed a round shape and had diameters from 50 to 200 nm. A-EVs showed good cytocompatibility on human skin cells and were internalized into HaCaT cells via clathrin-, caveolae-mediated endocytosis, and membrane fusion. The SOD activity and CAA assays exhibited that A-EVs had antioxidant activity and reduced intracellular ROS levels in H2O2-treated HaCaT cells in a dose-dependent manner. A scratch assay showed that A-EVs enhanced the migration ability of HaCaT and HDF. Moreover, A-EVs significantly upregulated the mRNA expression of Nrf2, HO-1, CAT, and SOD genes in H2O2-treated HaCaT cells. Our findings reveal that A-EVs could activate the antioxidant defense mechanisms and wound healing process via the Nrf2 activation. Conclusion: Overall results suggest that the A-EVs are promising as a potential agent for skin regeneration.

Developing new wound dressings is an important strategy in skin regeneration. Aloe vera is a medicinal plant with a long history, complex constituents, and various pharmacological activities. Many studies have shown that A. vera plays an important role in promoting wound healing. Adding A. vera to wound dressing has become an ideal way. This review will describe the process of skin injury and wound healing and analyze the role of A. vera in wound healing. In addition, the types of wound dressing and the applications of A. vera in wound dressing will be discussed.
Isolation of Aloe saponaria-Derived Extracellular Vesicles and Investigation of Their Potential for Chronic Wound Healing

Abstract:A chronic wound is caused by a failure to progress through the normal phases of wound repair in an orderly and timely manner. To induce skin regeneration while inhibiting chronic inflammation, numerous natural products, and in particular, plant-derived biomaterials, have been developed. Aloe saponaria, is known to contain flavonoid and phenolic acid compounds with anti-oxidative and anti-inflammatory properties. Here, we isolated extracellular vesicles (EVs) from Aloe saponaria by polyethylene glycol (PEG)-based precipitation and investigated their potential as a therapeutic for chronic wound healing. The Aloe saponaria-derived EVs (AS-EVs) showed no significant cytotoxicity on several cell types, despite a high level of intracellular uptake. When lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages were treated with AS-EVs, significant reductions in the expression of pro-inflammatory genes, such as interleukin-6 and interleukin-1β, were observed. Proliferation and migration of human dermal fibroblasts, as determined by the water-soluble tetrazolium salt-8 and transwell migration assay, respectively, were shown to be promoted by treatment with AS-EVs. It was also demonstrated that AS-EVs enhanced tube formation in human umbilical vein endothelial cells, indicating a stimulatory activity on angiogenesis; one of the crucial steps for effective wound healing. Collectively, our results suggest the potential of AS-EVs as a natural therapeutic for chronic wound healing.

Abstract:Clinical multiple acute trauma has brought great suffering for patients. The use of traditional Chinese medicine provides a new approach to treating trauma. Aloe Vera gel has been used to treat skin diseases for a long time, but its efficacy is not stable. As the most important part of the organic active component of Aloe Vera gel, the effect of aloin in acute trauma has not been reported. In this article, the effect of aloin on promoting wound healing was investigated through a series of experiments in vitro and vivo. For the in vitro experiment, 3-(4,5 -dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide cell proliferation and cycle analysis demonstrated that aloin could enhance the proliferation of human skin fibroblasts and endothelial cells. Transwell and scratch tests demonstrated that aloin could increase the chemotaxis and migration on human skin fibroblasts, and the tube formation assay demonstrated that aloin solution significantly increased the tube network formation compared to the control group. For in vivo experiments, we used a rat model of acute trauma to illustrate that aloin solution significantly accelerated wound healing. Moreover, the results showed that aloin can accelerate wound healing by inducing the expression of epidermal growth factor, and additional, new neovascularization and fibroblasts were found in the skin treated with aloin solution. Additionally, collagen fibers are arranged in a neat and orderly arrangement compared with the control group. Aloin solution could promote the rate of wound closure, and the collagen fiber arrangement of the healed skin was more regular, which inhibited the formation of scar tissue.

Abstract:BACKGROUND: Among the most important factors in wound healing pathways are transforming growth factor beta1 and vascular endothelial growth factor. Fibroblasts are the main cell in all phases wound closure. In this study, the extracts of plant materials such as Adiantum capillus-veneris, Commiphora molmol, Aloe vera, and henna and one mixture of them were used to treatment of normal mouse skin fibroblasts.METHODS: Cytotoxic effects of each extract and their mixture were assessed on mouse skin fibroblasts cells using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. We performed migration assays to assess migration properties of mouse skin fibroblasts cells in response to the extracts. Changes in the gene expression of the Tgfβ1 and Vegf-A genes were monitored by real-time polymerase chain reaction.RESULTS: A. capillus-veneris, C. molmol and henna extract improved the expression of Tgfβ1 gene. All used extracts upregulated the expression of Vegf-A gene and promoted the migration of mouse fibroblast cells in vitro.CONCLUSIONS: The present study demonstrated that the mentioned herbal extracts might be effective in wound healing, through the improvement in the migration of fibroblast cells and regulating the gene expression of Tgfβ1 and Vegf-A genes in fibroblast cells treated with extracts.

Polysaccharides of Aloe vera induce MMP-3 and TIMP-2 gene expression during the skin wound repair of rat

Abstract:Polysaccharides are the main macromolecules of Aloe vera gel but no data about their effect on extracellular matrix (ECM) elements are available. Here, mannose rich Aloe vera polysaccharides (AVP) with molecular weight between 50 and 250 kDa were isolated and characterized. Open cutaneous wounds on the back of 45 rats (control and treated) were daily treated with 25mg (n=15) and 50 mg (n=15) AVP for 30 days. The levels of MMP-3 and TIMP-2 gene expression were analyzed using real time PCR. The levels of n-acetyl glucosamine (NAGA), n-acetyl galactosamine (NAGLA) and collagen contents were also measured using standard biochemical methods. Faster wound closure was observed at day 15 post wounding in AVP treated animals in comparison with untreated group. At day 10 post wounding, AVP inhibited MMP-3 gene expression, while afterwards MMP-3 gene expression was upregulated. AVP enhanced TIMP-2 gene expression, collagen, NAGLA and NAGA synthesis in relation to untreated wounds. Our results suggest that AVP has positive effects on the regulation of ECM factor synthesis, which open up new perspectives for the wound repair activity of Aloe vera polysaccharide at molecular level.
Healing potential of injectable Aloe vera hydrogel loaded by adipose-derived stem cell in skin tissue-engineering in a rat burn wound model

Abstract:Adipose stem cells (ASCs) are a great promise in wound healing due to their potential in differentiating into various cell lineages and secreting growth factors. The purpose of this study is to evaluate the in vivo effects of Aloe vera hydrogel loaded by allogeneic ASCs on a rat burn wound model. The ASCs were isolated, cultured and mixed with 50% Aloe vera hydrogel and injected intradermally around the wound. Demineralized bone matrix (DBM) was used as dressing in the experiment. The burn wound-healing properties of different experimental groups were investigated by histopathological, molecular, scanning electron microscopic and biochemical analysis at the 7th, 14th and 28th days post-wounding. The Aloe vera and DBM-Aloe vera groups showed almost similar healing properties, while treatment by DBM-Aloe vera/ASCs significantly enhanced wound healing. The levels of transforming growth factor-β1 (TGF-β1) and interleukin-1β markedly decreased at the 7th day post-injury, in the DBM-Aloe vera/ASC-treated group, suggesting that this treatment regime subsided the inflammatory responses. Angiogenesis, re-epithelialization and the level of TGF-β1 in the wounds treated with DBM-Aloe vera/ASCs were also remarkably higher than those of other groups, at the 14th day post-injury. Besides, scar formation significantly decreased in the DBM-Aloe vera/ASC-treated wounds when compared with other groups. Our biochemical results were in agreement with the molecular and histopathological findings and strongly demonstrated that a DBM-Aloe vera/ASC composite can stimulate burn wound healing. These results suggest that the DBM-Aloe vera/ASC composite can be considered as a promising therapeutic strategy in the treatment of burn wounds.
Emodin and aloe-emodin, two potential molecules in regulating cell migration of skin cells through the MAP kinase pathway and affecting Caenorhabditis elegans thermotolerance

Abstract:Background: Emodin and aloe-emodin are two anthraquinones having positive effects in wound healing. However, their mechanism of action of wound healing is not fully understood. The MAP kinase family, which plays an active role in wound healing, is a well-characterized large family of serine/threonine kinases and regulates processes such as proliferation, oncogenesis, differentiation, and inflammation in the cell. The aim of this study is to comparatively elucidate the mechanisms of action of emodin and aloe-emodin, which are potential agents in wound healing. Methods: The mechanism of the effects of emodin and aloe-emodin on cell viability and cell migration was examined using the human skin fibroblast (CCD-1079Sk) cell line. The gene expression levels of the MAP kinases (JNK, P38, ERK) in the skin fibroblast cells along with a molecular docking study analyzing their interaction potential were evaluated. Furthermore, the molecules' effects on the lifespan of Caenorhabditis elegans were studied. Results: Emodin and aloe-emodin inhibited the ATP content of the cells in a concentration dependent manner and accelerated cell migration at the lower concentrations while inhibiting cell migration in the higher concentration treatment groups. The expressions of JNK and P38 were upregulated at the low concentrations and downregulated at the higher concentrations. The molecular docking studies of the molecules gave high docking scores indicating their interaction potential with JNK and P38. C. elegans lifespan under heat stress was observed longer after 75 µM emodin and was significantly reduced after 150 µM aloe-emodin treatment. Conclusion: Aloe-emodin was found to be more potent on cell viability, cell migration, gene expression levels of the MAP kinases in healthy fibroblastic skin cells, and on the lifespan of C. elegans. This study reveals the functional effects and the biological factors that interact in the wound healing process of emodin and aloe-emodin, and give a possible treatment alternative to shorten the duration of wound care.

Abstract:Percutaneous transluminal angioplasty was attempted in 70 stenotic renal arteries using the balloon angioplasty technique described by Grüntzig for peripheral vessels. There were 58 patients with normal renal function and 12 with decreased renal function. Dilatation of the stenosis was initially successful in 65 attempts and unsuccessful in five. Four of the stenoses became restenosed; two were successfully dilated a second time. Complications occurred in four patients; three developed acute tubular necrosis and one sustained an inconsequential renal artery dissection. Patients with renin-dependent hypertension had dramatic reduction in blood pressure in most instances; 90% were cured or improved. Other hypertensive patients without demonstrable renin dependency had less marked reduction, but were often benefited. Patients with hypertension and renal failure were helped to a lesser extent.
Effects of Aloe vera on Burn Injuries: A Systematic Review and Meta-Analysis of Randomized Controlled Trials

Abstract:Abstract Burn injuries cause severe pain, infection risks, psychological distress, financial burdens, and mortality, necessitating effective care. Aloe vera, a traditional burn remedy, shows wound-healing potential, but its analgesic effects and efficacy with varying burn severity are uncertain. This study aims to investigate A. vera’s effect on wound healing, pain management, and infection prevention in patients with burns. A systematic search on PubMed, Embase, and CENTRAL was performed on October 9, 2023, for randomized controlled trials (RCTs). The risk of bias was examined using the Cochrane risk-of-bias tool (version 2), and the meta-analysis was carried out using a random-effects model. The primary outcome was wound-healing time, with secondary outcomes examining pain severity and wound infection. The Grading of Recommendations, Assessment, Development, and Evaluation (GRADE) approach was used to assess the quality of evidence for each outcome. Nine RCTs were included in the current study, of which 6 provided data on the primary outcome. Aloe vera significantly reduced mean wound-healing time compared to other topicals (mean difference [MD] −3.76 days; 95% CI, −5.69 to −1.84). Additionally, the meta-analysis of the secondary outcomes found no significant differences in pain reduction (MD −0.76 points; 95% CI, −1.53 to 0.01) and wound infection risk (risk ratio 1.10; 95% CI, 0.34-3.59) between A. vera and control groups. In conclusion, A. vera expedites wound healing in patients with second-degree burns without increased infection risk compared to other antimicrobial agents. The analgesic effects on burn injuries remain uncertain.

Abstract:Various local and systemic factors compromise oral wound healing and may lead to wound dehiscence, inflammation, or ulcers. Currently, there is a lack of topical therapeutical options. Thus, this study aimed to investigate the effect of Aloe vera (AV) and Rheum palmatum root (RPR) on oral wound healing capacity in vitro. The effect of AV and RPR on human primary fibroblast viability and migration was studied by measuring metabolic activity and gap closure in a scratch assay. Furthermore, cell cycle distribution and cytoskeletal features were analyzed. Antimicrobial activity against the oral pathogen Porphyromonas gingivalis was evaluated by broth microdilution assay. AV and RPR increased fibroblast migration after single agent treatment. Synergistic effects of the plant extract combination were observed regarding cellular migration which were confirmed by calculation of the phenomenological combination index (pCI), whereas the cell cycle distribution was not influenced. Furthermore, the combination of AV and RPR showed synergistic antibacterial effects as determined by the fractional inhibitory concentration index. This study demonstrated that the combination of AV and RPR can promote the migration of human primary fibroblasts in vitro and exert antimicrobial efficacy against P. gingivalis, suggesting these compounds for the topical treatment of wound healing disorders.

Abstract:The International Journal of Lower Extremity Wounds, Ahead of Print. Treatment of chronic wounds has been shifted to traditional approaches due to surge in antibiotic resistance. Wounds that fail to heal satisfactorily may result in the amputation of the organ. In this research work, cinnamon oil (CO) and aloe vera (AV) that have been traditionally used as antibacterial agents are combined in a unique gel (COVA) and its antibacterial activity has been evaluated through in vitro and in vivo studies. Antibacterial activity was measured through disk diffusion and agar dilution method against Pseudomonas aeruginosa and Staphylococcus aureus. To check antibacterial and wound healing activity, diabetic excision wound healing rat model was used. Wound closure, wound contraction, tissue hydroxyproline content, antioxidant capacity (TAC), and malondialdehyde (MDA) level were monitored. The minimum inhibitory concentrations of CO + AV for bacterium P. aeruginosa and S. aureus were 100 and 200 μg/ml, respectively. After 14 days, the wounds covered with COVA therapy reached to nearly full wound closure (79% wound contraction) compared to control. The collagen content and level of TAC increased significantly (P < 0.05) in treated groups; therefore, 25% fast healing was observed in wounds treated with CO and AV gel combined. Reduced levels of tissue MDA were observed in all treated groups and specially wound covered with COVA (0.43 mM/mg in control vs 0.25 mM/mg in COVA). Histopathological examination also supported the outcomes. Significantly elevated increase in the level of hydroxyproline was found in rats of COVA treatment group (37.1 ± 0.44). Combination of CO and AV can be potentially used to prevent infection in wound; as these herbal agents not only inhibit the growth of pathogenic bacteria but also accelerate tissue repair.

Antibacterial effect of Aloe Vera on Bacteria Isolated from Cases of Wound Infection

Abstract:Background: Antimicrobial resistance exhibited by bacteria against the majority of antibiotics has resulted in research on alternative methods of treatment. Aloe vera has a strong tradition as a medical plant with a wide range of therapeutic uses. Objective: The objective of this study is to determine the antibacterial activity of gel and crude ethanol leaf extract of Aloe vera against Staphylococcus aureus and Enterobacterales isolated from wound infections. Methods: It is a cross-sectional study conducted over a period of 7 months. Antibacterial effect of the ethanol leaf extract and gel was determined by the punch well method. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of the ethanol leaf extract were determined by macro broth dilution technique. Results: Aloe vera ethanol leaf extract induced a mean zone size of 13.0 ± 6.0 mm and 16.7 ± 8.4 mm, respectively, for S. aureus and Enterobacterales by Punch Well method (p≤0.002). Whereas Aloe vera gel failed to induce any zone of inhibition for all the isolates p<0.001. Mean MIC of Aloe vera leaf extract against 74 S. aureus was 94 ± 41.23 mg/ml and against 73 Enterobacterales, it was 45.6 ± 20 mg/ml p < 0.001. Mean MBC of Aloe vera leaf extract against 74 S.aureus isolates was 188 ± 82.46 mg/ml and against 73 Enterobacterales was 91.18±40 mg/ml p < 0.001. Conclusion: Aloe vera ethanol leaf extract showed a good antibacterial effect against the different strains of bacteria causing wound infection. The present article shows the possibility of future use of natural products for the treatment of wound infections.

Abstract:Chronic wounds are severe breaks in the skin barrier that fail to heal in an acceptable time-frame, thus preventing the complete restoration of the tissue’s anatomical and functional integrity, increasing the likelihood of infections and apoptosis. Hydrogels are known as a drug delivery system and have the potential to cover wounds and burns on the skin. Aloe barbadensis contains over 75 different bioactive compounds which are responsible for its anti-inflammatory and antimicrobial properties. In this study, the polyacrylamide-co-methylcellulose hydrogel containing Aloe barbadensis were developed. The extract was prepared from lyophilized Aloe barbadensis, using methanolic extraction, characterized by high performance liquid chromatography and incorporated into the hydrogels. These Aloe barbadensis hydrogels were characterized by degree of swelling, Fourier-transform infrared spectroscopy, scanning electron microscopy, and thermal profiling using thermogravimetric analysis. The minimum inhibitory concentration test was done on the Aloe barbadensis extract to evaluate its antibacterial and antifungal activity in vitro. The Aloe barbadensis hydrogels and were shown to swell to almost 2000% of their original sizes. The Fourier-transform infrared spectroscopy indicated the presence of bands characteristic of Aloe barbadensis and hydrogel polymers. The basic hydrogel showed greater thermal stability than the hydrogels with Aloe barbadensis. The minimum inhibitory concentration showed inhibition of the growth of S. aureus and Salmonella spp. at specific concentrations. The hydrogel therefore presents itself as an excellent potential curative cover of cutaneous lesions.
