Abstract:Objective: With the aid of an acute visceral pain model of uterine cervical distension (UCD), the present study aimed to observe the effects of intrathecal administration of transient receptor potential vanilloid 1 (TRPV1) antagonist SB-366791 on UCD induced-visceral nociception as well as its involved molecular mechanisms. Methods: A total of 30 Sprague-Dawley-derived adult virgin female rats were used. UCD model was established under isoflurane inhalation anesthesia. Briefly, a lower abdominal incision at midline was made to expose the uteral cervix, two metal rods were inserted through both sides of the cervix separately, one rod was fixed and the other one was connected to a pulley system with application of manual weighted traction (0, 25, 50, 75 or 100 g) for simulating 1 h of cervical distension. In addition, 12 Sprague-Dawley-derived adult virgin female rats were subjected to intrathecal catheter implantation, and UCD was established 7 days later. The rats were divided randomly into two groups; one group was administrated with intrathecal SB-366791 while the other was administrated with the same volume of saline as control. The 75 g distension force was then applied for an hour and the electromyographic (EMG) of musculus rectus abdominis, heart rate as well as respiratory frequency were measured continuously during the surgery. The spinal cord (T(12)-L(2)) was collected 30 minutes after UCD for the detection of changes of c-FOS and TRPV1 expression. Results: UCD increased EMG activity (P<0.05) and c-FOS expression (P<0.05) in the deep dorsal horn region and central canal of the spinal cord (T(12)-L(2)) in a stimuli-dependent manner, the expression of TRPV1 in the T(12)-L(2) spinal cord also increased in response to UCD stimulation (P<0.05). Compared with the saline group, intrathecal SB-366791 significantly decreased EMG activity (P<0.05) as well as spinal c-FOS (P<0.05) expression induced by UCD. Conclusions: UCD in rats increases EMG activity of musculus rectus abdominis as well as spinal c-FOS and TRPV1 expression. Intrathecal administration of TRPV1 antagonist SB-366791 significantly decreases the visceral nociception induced by UCD.
Spinal P2X4 Receptors Involved in Visceral Hypersensitivity of Neonatal Maternal Separation Rats

Abstract:Recent studies have demonstrated the vital role of P2X4 receptors (a family of ATP-gated non-selective cation channels) in the transmission of neuropathic and inflammatory pain. In this study, we investigated the role of spinal P2X4 receptors in chronic functional visceral hypersensitivity of neonatal maternal separation (NMS) rats. A rat model of irritable bowel syndrome was established by neonatal maternal separation. Visceral sensitivity was assessed by recording the response of the external oblique abdominal muscle to colorectal distension. P2X4 receptor antagonist and agonist were administrated intrathecally. The expression of P2X4 receptor was examined by Western Blot and immunofluorescence. The effect of P2X4 receptor antagonist on expression of brain-derived neurotrophic factor (BDNF) was assessed by Western Blot. We found neonatal maternal separation enhanced visceral hypersensitivity and increased the expression of P2X4 receptor in spinal thoracolumbar and lumbosacral segments of rats. Pharmacological results showed that visceral sensitivity was attenuated after intrathecal injection of P2X4 receptor antagonist, 5-BDBD, at doses of 10 nM or 100 nM, while visceral sensitivity was enhanced after intrathecal injection of P2X4 receptor agonist C5-TDS at doses of 10 μM or 15 μM. In addition, the spinal expression of BDNF significantly increased in NMS rats and intrathecal injection of 5-BDBD significantly decreased the expression of BDNF especially in NMS rats. C5-TDS failed to increase EMG amplitude in the presence of ANA-12 in control rats. Our results suggested the spinal P2X4 receptors played an important role in visceral hypersensitivity of NMS rats through BDNF.
Effect of neonatal maternal separation on visceral hyperalgesia in the developing rats and role of fos abnormal expression in spinal cord

Abstract:Objective To explore the effect of maternal separation (MS) stimulation in neonatal period on visceral sensitivity of pain in developing rats. Observeing the Fos expression at dorsal horn of lumborsacral spinal cord in rats to investigate the mechanism of highly visceral pain sensitivity. Methods According to the factorial design,32 SD rats were divided into four groups of 8 each. Group A1B1 was experimental group and subjected to colorectal distention (CRD) at 6-week age,yet group A1B2 were not imposed on CRD as the experimental group. Group A2B1 was subjected to colorectal CRD at 6-week age while group A2B2 without treatment both as the control groups. Rats in the MS group were separated from the dam for 3h/d (8:00-11:00 am) on postnatal days 2-14 for 13 consecutive days,rats in control group rats were not treated and remained in the home cage with the dam. When 6 weeks old,the rats of group A1B1 and A2B1 were evaluated the pain sensibility of intestinal tract under colorectal distension irritation by the method of observing the score of abdominal withdrawal reflex (AWR) and visceral pain threshold subjected to different pressures of colorectal distension irritation (CRD) to evaluate visceral algesthesia in rats. The rats were sampled the spinal cord from L6 to S2 after the group A1B1 and A2B1 were subjected to colorectal CRD for 2 hours while the group A1B2 and A1B2 not subjected to colorectal CRD. The semiquantity analysis of the cell number of Fos-Like Immunoreactivity(FLI) in spinal cord were made through immuno-hisrochemical coloration and computer image analyzing system. Results While CRD pressure were 20 mmHg,40 mmHg,60 mmHg,80 mmHg,the score of AWR were 2.2±0.7,3.2± 0.6,3.8±0.4,4.0±0.0 in group A1B1,and 1.0±0.8,2.5±0.3,3.3±0.3,3.9±0.2 in group A2B1. When the CRD pressure were 20 mmHg,40 mmHg,60 mmHg,the AWR score was higher in the A1B1 group than the group A2B1 signigficantly. But when pressure reached 80 mmHg,there was no significant difference of AWR scores between two gruops. The pain thresholds of the group A1B1 and group A2B1 were 17.9±8.9 mmHg,33.5±1.9 mmHg respectively (t=23.627,P=0.000). The cell number of FLI in lumbosacral spinal dorsal horn in group A1B1,A2B1,A1B2,A2B2 were 18.3±3.4,11.4±1.1,14.3± 1.3,9.1±0.4,respectively,and MS in neonatal period at the neonatal period and CRD at 6-week age both can make it increase significantly(F 21.040,78.702,P<0.01). Conclusion The persistent MS in neonatal phase can cause primary central sensitization of spinal cord in developing rats. Accepting detrimental CRD can significantly cause the high expression of Fos in spinal cord and result in decreasing of pain threshold in rats,emergeing chronic visceral hypersensibility.
Effect of neonatal maternal separation on external oblique spikes of electrical discharge in the developmental rats and Fos expression in spinal dorsal horn

[Results] While CRD pressure were 0,15,30,45,60,75mmHg,the external oblique spikes of electrical discharge were (14.20±1.57),(26.25±2.93),(33.84±4.73),(42.87±5.36),(47.02±4.86),(43.16±6.16)μV in group A1B1,and (13.23±1.47),(14.62±1.95),(23.31±5.95),(33.27±6.70),(40.90± 3.23),(43.79±8.89)μV in group A2B1. With the CRD pressure increased,the spikes increased. When the CRD pressure were 15,30,45,60 mmHg,the spikes in group A1B1 were significantly higher than group A2B1. The staining density of FLI in lumbosacral spinal dorsal horn in group A1B1,A2B1,A1B2,A2B2 were 88.21±7.28,98.38±6.67,111.32±12.11,127.31 ± 1.78,respectively,and MS and CRD at 6-week age both can make it increase significantly(F=14.303、56.608,P<0.01). [Conclusion] MS in neonatal phase may cause rat's central sensitization of spinal,resulted in low pain threshold and chronic high visceral pain sensibility,histological change wasn't found in colorectal tissue.
Establishment and Evaluation of a Model of Visceral Hyperalgesia in Young Rats

Abstract:Objective To establish and evaluate in young rats a model of visceral hyperalgesia induced by maternal separation(MS) stimulation in the neonatal period.Methods Thirty Sprague-Dawley rats of two days old were divided randomly into NMS39,NMS60 and control groups.Baby rats were separated from their mothers for 3 h/d(8∶00-11∶00 am) during postnatal days 2～14 in the NMS39 group and during postnatal days 2～21 in the NMS60 group while those in the control group always lived with their mothers during postnatal days 2～21.The visceral sensitivity of six-week-old rats was evaluated by the score of abdominal withdrawal reflex(AWR),the amplitudes of spike of external oblique muscle of abdomen(EOMA) and visceral pain threshold.The amount of fecal pellets and the time of glass pellets output in the young rats were observed.Results Compared with the control rats,young rats in the NMS39 and NMS60 group showed a significant increase in the score of AWR and the amplitudes of spike of EOMA and a remarked decrease in the pain thresholds.But the comparison between the NMS39 and NMS60 group revealed no significant differences in the above indexes.The comparison among the three groups reported no significant differences in fecal pellets and the time of glass pellets output.Conclusion The persistent MS in neonatals can result in chronic visceral hypersensibility in young rats.
Spinal toll-like receptor 4-mediated signalling pathway contributes to visceral hypersensitivity induced by neonatal colonic irritation in rats

Abstract:Background: Although visceral hypersensitivity is a major pathophysiological feature of irritable bowel syndrome (IBS), its underlying mechanisms remain elusive. Toll-like receptor 4 (TLR4) is a critical pattern recognition molecule of the innate immune system. In this study, we investigated whether the TLR4/myeloid differentiation factor 88 (MyD88)/nuclear factor-kappa B (NF-κB) signalling pathway in the spinal cord contributed to the visceral hypersensitivity induced by neonatal colonic irritation (CI) in rats. Methods: The Sprague-Dawley rat model of IBS was induced by colon irritation on post-natal day (PND) 8, PND10 and PND12. Experiments were conducted in adult rats. TLR4 mRNA and protein, and its downstream signalling molecules, MyD88, inhibitory nuclear factor-kappa B (IκB) and NF-κB protein expressions in L2-S4 spinal segments were detected by quantitative real-time reverse transcription-polymerase chain reaction as well as Western blotting. TLR4 co-localization was determined by immunohistochemistry. Levels of tumour necrosis factor-alpha (TNF-α) and interleukin 1β (IL-1β) were measured with enzyme-linked immunosorbent assay. Results: We found that neonatal CI treatment induced long-lasting visceral hypersensitivity without identifiable structural abnormalities in descending colons of adult rats. Neonatal CI treatment evoked a significant up-regulation of the expressions of TLR4 in glia, MyD88, p-IκB-α and NF-κB in adult rats. Neonatal CI treatment also increased the levels of its downstream inflammatory agents TNF-α and IL-1β in the L2-S4 regions of the spinal cord of adult rats. Conclusions: These results suggest that neonatal CI stimulates the production of IL-1β and TNF-α through the TLR4/MyD88/NF-κB signalling pathway in the spinal cord, which contributed to visceral hypersensitivity induced by neonatal CI in rats.

Abstract:Early life stress (ELS) disposes to functional gastrointestinal diseases in adult, such as irritable bowel syndrome (IBS). Maternal separation (MS) is a well-known animal model of IBS and has been shown to induce visceral hypersensitivity in adult rats and mice. However, to the best of our knowledge, it has not been reported whether MS induces visceral hypersensitivity in young mice, such as the post-weaning mice. Moreover, the method for evaluation of visceral sensitivity also has not been described. Accordingly, the present study aims to evaluate the visceral sensitivity caused by MS in post-weaning mice and develop a novel and small size distention balloon for assessment of visceral sensitivity of such mice. Male pups of C57BL/6 mice were randomly divided into two groups, MS ( n = 12) and non-separation (NS) ( n = 10). MS pups were separated from the dams through postnatal days (PND) 2 to 14, while NS pups were undisturbed. After, all pups stayed with respective dams and were weaned at PND 22. Visceral sensitivity was evaluated by colorectal distention (CRD) with a novel and small size distention balloon at PND 25. The threshold of abdominal withdrawal reflex (AWR) scores were significantly lower in MS than NS. In addition, AWR scores at different pressures of CRD were significantly higher in MS than NS. The results demonstrate that MS induced visceral hypersensitivity in post-weaning mice. The designed small size distention balloon for evaluation of visceral sensitivity is of significance to further study the pathophysiology of IBS from early life to adulthood.

Upregulation of Netrin-1 in the hippocampus mediates the formation of visceral hypersensitivity induced by maternal separation

Abstract:Early adverse life events (EALs), such as maternal separation (MS), can cause visceral hypersensitivity, which is thought to be a key pathophysiological mechanism of irritable bowel syndrome (IBS). Previous studies mainly focused on EALs-induced visceral hypersensitivity in adulthood but did not consider that it may have occurred in the preadult period. We previously found that rats who experienced MS suffered from visceral hypersensitivity starting from the post-weaning period. Moreover, the hippocampus is considered to be critical in regulating the formation of visceral hypersensitivity induced by MS. But the underlying mechanisms throughout different life periods are unclear. In this study, behavioral tests, RNA-seq, lentiviral interference, and molecular biology techniques were applied to investigate the molecular mechanism in the hippocampus underlying MS-induced long-lasting visceral hypersensitivity. It was found that both visceral sensitivity and anxiety-like behaviors were significantly increased in MS rats in post-weaning, prepubertal, and adult periods, especially in the prepubertal period. Subsequently, RNA-seq targeting the hippocampus identified that the expression level of Netrin-1 was significantly increased in all periods, which was further confirmed by quantitative real-time PCR and Western blot. Knocking-down hippocampal Netrin-1 in the post-weaning period by lentivirus interference alleviated visceral hypersensitivity and anxiety-like behaviors of MS rats in the later phase of life. In addition, deleted in colorectal cancer (DCC), instead of neogenin-1(Neo-1) or uncoordinated (UNC5), was proved to be the specific functional receptor of Netrin-1 in regulating visceral hypersensitivity, whose upregulation may result in the most severe symptoms in the prepubertal period. Furthermore, the activation of the Netrin-1/DCC pathway could enhance long-term potentiation (LTP) in the hippocampus, probably via recruitment of the AMPA receptor subunit GluA1, which finally resulted in the formation of visceral hypersensitivity. These novel findings suggest that long-lasting over-expression of Netrin-1 can mediate visceral hypersensitivity and anxiety disorder from the post-weaning period to adulthood by activating DCC/GluA1 pathway in the hippocampus. Moreover, early intervention of Netrin-1 in the post-weaning period could lead to significant symptom relief afterward, which provides evidence that the Netrin-1/DCC/GluA1 signaling pathway may be a potential therapeutic target for the treatment of visceral hypersensitivity in clinics.

Abstract:Neonatal maternal separation is associated with pain hypersensitivity in adulthood, in addition to excessive anxiety, abnormal response to stress, and cognitive dysfunction. Among the many dysregulated gene products observed after these early life adverse events, expression of chloride transporters that optimize spinal inhibition is permanently altered. We hypothesized that spinal disinhibition mediated by chloride‐mediated ionic channels such as GABAA and glycine receptors is responsible for the hyperexcitability observed in adult 'wide dynamic range' spinal neurons. Early life adversities influence a nervous system still in development with long‐term consequences for later life. These include nociceptive circuit alterations critical to shape an adaptive pain response to protect the organism from potential damage. Adult rats with a history of neonatal maternal separation (NMS) display visceral and somatic nociceptive hypersensitivity and inefficient analgesic responses to stress. In this study, we have characterized the consequences of NMS on wide dynamic range neurons (WDR) in the spinal cord of anaesthetized adult rats during the nociceptive processing of hot and cold noxious information. We found that WDR neurons of NMS rats display an excessive coding of mechanical and thermal information applied at the rat's hindpaws. This nicely explains the hypernociceptive behaviours seen after noxious mechanical, cold and hot peripheral stimulation. A peripheral change in the expression of molecular transducers for these stimuli (i.e., TRPV1, TRPM8 and TRPA1) does not seem to account for this general hyperexcitability. Instead, a decreased chloride‐mediated inhibitory tone on WDR neurons may play a role as indicated by the abnormal elevation of the type 1 Na‐K‐Cl cotransporter transcripts. Altogether, we propose that long‐term consequences of NMS are associated with reduced spinal cord inhibition favouring the expression of pain hypersensitivity. We cannot exclude that this phenomenon is also present at supraspinal sites, as other NMS‐associated symptoms include excessive anxiety and impaired sociability.

Unraveling the role of Epac1-SOCS3 signaling in the development of neonatal-CRD-induced visceral hypersensitivity in rats

Abstract:Aims: Visceral hypersensitivity in irritable bowel syndrome (IBS) is widespread, but effective therapies for it remain elusive. As a canonical anti-inflammatory protein, suppressor of cytokine signaling 3 (SOCS3) reportedly relays exchange protein 1 directly activated by cAMP (Epac1) signaling and inhibits the intracellular response to inflammatory cytokines. Despite the inhibitory effect of SOCS3 on the pro-inflammatory response and neuroinflammation in PVN, the systematic investigation of Epac1-SOCS3 signaling involved in visceral hypersensitivity remains unknown. This study aimed to explore Epac1-SOCS3 signaling in the activity of hypothalamic paraventricular nucleus (PVN) corticotropin-releasing factor (CRF) neurons and visceral hypersensitivity in adult rats experiencing neonatal colorectal distension (CRD). Methods: Rats were subjected to neonatal CRD to simulate visceral hypersensitivity to investigate the effect of Epac1-SOCS3 signaling on PVN CRF neurons. The expression and activity of Epac1 and SOCS3 in nociceptive hypersensitivity were determined by western blot, RT-PCR, immunofluorescence, radioimmunoassay, electrophysiology, and pharmacology. Results: In neonatal-CRD-induced visceral hypersensitivity model, Epac1 and SOCS3 expressions were downregulated and IL-6 levels elevated in PVN. However, infusion of Epac agonist 8-pCPT in PVN reduced CRF neuronal firing rates, and overexpression of SOCS3 in PVN by AAV-SOCS3 inhibited the activation of PVN neurons, reduced visceral hypersensitivity, and precluded pain precipitation. Intervention with IL-6 neutralizing antibody also alleviated the visceral hypersensitivity. In naïve rats, Epac antagonist ESI-09 in PVN increased CRF neuronal firing. Consistently, genetic knockdown of Epac1 or SOCS3 in PVN potentiated the firing rate of CRF neurons, functionality of HPA axis, and sensitivity of visceral nociception. Moreover, pharmacological intervention with exogenous IL-6 into PVN simulated the visceral hypersensitivity. Conclusions: Inactivation of Epac1-SOCS3 pathway contributed to the neuroinflammation accompanied by the sensitization of CRF neurons in PVN, precipitating visceral hypersensitivity and pain in rats experiencing neonatal CRD.
Maternal separation and TNBS-induced gut inflammation synergistically alter the sexually differentiated stress response in rats

Abstract:Background: Neonatal maternal separation (MS) has been used to model long-lasting changes in behavior caused by neuroplastic changes associated with exposure to early-life stress. Earlier studies showed that transient gut inflammation can influence the development of irritable bowel syndrome (IBS). A prevailing paradigm of the etiology of IBS is that transient noxious events lead to long-lasting sensitization of the neural pain circuit, despite complete resolution of the initiating event. This study characterizes the changes in behaviors and neuroendocrine parameters after MS and early-phase trinitrobenzene sulfonic acid (TNBS)-induced colitis. We tested the hypothesis that MS and gut inflammation synergistically induce (1) hyperactivity in male rats and anxiety-like behaviors in female rats and (2) activation of the HPA axis in female rats and deactivation of the HPA axis in male rats after colorectal distention (CRD). Methods: Male and female rat pups were separated from their dams for 180 min daily from postnatal day (PND) 2 to PND 14 (MS). Early-phase colitis was induced by colorectal administration with TNBS on PND 8. The elevated plus-maze test was performed at 7 weeks. Tonic CRD was performed at 60 mmHg for 15 min at 8 weeks. Plasma ACTH and serum corticosterone were measured at baseline or after the CRD. Analysis of variance was performed for comparison among controls, TNBS, MS, and MS + TNBS. Results: In male rats, the time spent in open arms significantly differed among the groups (p < 0.005). The time spent in open arms in male MS + TNBS rats was significantly higher than that of controls (p < 0.009) or TNBS rats (p < 0.031, post hoc test). Female rats showed no difference in the time spent in open arms among the groups. MS and gut inflammation induced an increase in plasma ACTH in female rats but not in male rats at baseline. Conclusions: These findings suggest that MS and gut inflammation synergistically induce hyperactive behavior or exaggerated hypothalamic-pituitary-adrenal axis function depending on sex.
Establishment and Electrophysiological Evaluation on Developing Rats Model of Visceral Hyperalgesia

Abstract:Objective To explore establishing the optimal condition and electrophysiological evaluation method on developing rats model of visceral hyperalgesia,and to study the effect of detrimental colorectal irritation (CI) in neonatal period on visceral sensibility of pain in developing rats.Methods Eight days old Sprague-Dawley rats were randomly divided into experimental group (n=16) and control group (n=16).Rats in experimental group were treated with CI once a day for 7 consecutive days and rats in control group were not treated with CI.On the 6th week,the visceral sensitivity of all rats was evaluated.The amplitudes of spike of external oblique muscle of abdomen were measured to evaluate the bowel sensitivity.Repeated measuring ANOVA was used to analyze the differences between the 2 groups.α=0.05 was consi-dered significant.Results The spike of external oblique muscle of abdomen increased gradually with the rising of the colorectal distention (CRD) pressure in 2 groups.When the CRD pressure were 4 kPa and 6 kPa,the spike was higher in experimental group than in control group,the differences were significant(Pa<0.05).There were no significant differences of the spike between the 2 groups when the CRD pressure were 2 kPa,8 kPa and 10 kPa(Pa>0.05).There were no obvious histopatholigical changes in descending colon in experimental group and control group.Conclusions The persistent CI in neonatal phase results in decreasing of pain threshold and increasing of chronic visceral sensibility,while there is no histological change found in colorectal tissue.It is an ideal model of visceral hypersensitivity to study children abdominal pain correlated functional gastrointestinal disorder.Comparing with the ethological research,electrophysiological research is a quantitative test,which can reflect visceral sensibility of pain objectively.

Abstract:Early-life stress (ELS) is thought to cause the development of visceral pain disorders. While some individuals are vulnerable to visceral pain, others are resilient, but the intrinsic circuit and molecular mechanisms involved remain largely unclear. Herein, we demonstrate that inbred mice subjected to maternal separation (MS) could be separated into susceptible and resilient subpopulations by visceral hypersensitivity evaluation. Through a combination of chemogenetics, optogenetics, fiber photometry, molecular and electrophysiological approaches, we discovered that susceptible mice presented activation of glutamatergic projections or inhibition of GABAergic projections from the anteroventral bed nucleus of the stria terminalis (avBNST) to paraventricular nucleus (PVN) corticotropin-releasing hormone (CRH) neurons. However, resilience develops as a behavioral adaptation partially due to restoration of PVN SK2 channel expression and function. Our findings suggest that PVN CRH neurons are dually regulated by functionally opposing avBNST neurons and that this circuit may be the basis for neurobiological vulnerability to visceral pain.

Abstract:Background: Sress early in life has been linked to visceral hyperalgesia and associated functional gastrointestinal disorders. In a mouse model of visceral hyperalgesia, we investigated whether the EphB2 receptor and its EphrinB2 ligand in spinal cord contribute to dysregulation of glia-neuron interactions. Methods: An established mouse model of stress due to maternal separation (MS). Pups were separated from their mothers for 14 days during early development, then analyzed several weeks later in terms of visceral sensitivity based on the abdominal withdrawal reflex score and in terms of expression of c-fos, EphrinB2, EphB2, and phosphorylated MAP kinases (ERK, p38, JNK). Results: Visceral hyperalgesia due to MS upregulated EphB2, EphrinB2 and c-fos in the spinal cord, and c-fos levels positively correlated with those of EphB2 and EphrinB2. Spinal astrocytes, microglia, and neurons showed upregulation of EphB2, EphrinB2 and phosphorylated MAP kinases. Blocking EphrinB2/EphB2 signaling in MS mice reduced visceral sensitivity, activation of neurons and glia, and phosphorylation of NMDA receptor. Activating EphrinB2/EphB2 signaling in unstressed mice induced visceral hyperalgesia, upregulation of c-fos, and activation of NMDA receptor similar to maternal separation. Conclusion: The stress of MS during early development may lead to visceral hyperalgesia by upregulating EphrinB2/EphB2 in the spinal cord and thereby altering neuron-glia interactions.

Abstract:Abstract Chronic visceral pain is one of the primary symptoms of patients with irritable bowel syndrome (IBS), which affects up to 15% of the population world-wide. The detailed mechanisms of visceral pain remain largely unclear. Our previous studies have shown that neonatal maternal deprivation (NMD) followed by adult multiple stress (AMS) advances the occurrence of visceral pain, likely due to enhanced norepinephrine (NE)-β 2 adrenergic signaling. This study was designed to explore the roles of P2X3 receptors (P2X3Rs) in the chronic visceral pain induced by combined stress. Here, we showed that P2X3Rs were co-expressed in β 2 adrenergic receptor (β 2 -AR)-positive dorsal root ganglion neurons and that NE significantly enhanced ATP-induced Ca 2+ signals. NMD and AMS not only significantly increased the protein expression of P2X3Rs, but also greatly enhanced the ATP-evoked current density, number of action potentials, and intracellular Ca 2+ concentration of colon-related DRG neurons. Intrathecal injection of the P2X3R inhibitor A317491 greatly attenuated the visceral pain and the ATP-induced Ca 2+ signals in NMD and AMS rats. Furthermore, the β 2 -AR antagonist butoxamine significantly reversed the expression of P2X3Rs, the ATP-induced current density, and the number of action potentials of DRG neurons. Overall, our data demonstrate that NMD followed by AMS leads to P2X3R activation, which is most likely mediated by upregulation of β 2 adrenergic signaling in primary sensory neurons, thus contributing to visceral hypersensitivity.

Abstract:Background/Purpose: The purpose of this experiment was to identify the neurons in the lumbosacral spinal cord involved in colon-rectal function and to compare normal and anorectal malformation of fetal rats.Methods: The authors quantified the sacral parasympathetic nucleus (SPN) innervation of the rectum by Fluorogold (FG) (Fluorochrome, Englewood, CO) retrograde tracing experiment in fetal rats with normal and anorectal malformation. Anorectal malformation was induced in rat fetuses by ethylenethiourea (ETU). The number of FG-labeled SPNs was scored and compared between male fetuses with or without-fed group and control groups. malformation in the ETU-fed group and control groups.Results: The number of FG-labeled SPNs in the fetuses without a defect, with ETU injected but without any defects of the anorecturn or neural tube, with low-type deformity, and with high-type deformity were (mean +/- SEM) 47.3 +/- 2.9, 45.6 +/- 3.2, 24.2 +/- 3.8, and 8.5 +/- 2.5, respectively. Fluorogold-labeled SPNs in the fetuses with high-type deformity were significantly fewer than those in fetuses without defects (P < .05) and in controls (P < .05).Conclusions: These findings suggest that defective SPN innervation to the rectum is a primary anomaly that coexists with the alimentary tract anomaly in anorectal malformation during fetal development. The intrinsic neural deficiency is an important factor likely to contribute to poor postoperative anorectal function despite surgical correction of the malformation. (c) 2007 Elsevier Inc. Effect of Coloclysis with Sodium Butyrate Solution on Visceral Perception in Rats

Abstract:Background: For studying the visceral hypersensitivity of irritable bowel disease (IBS), a suitable animal model needs to be established. Aims: To establish a colon hypersensitivity model in rats by sodium butyrate solution coloclysis, and to observe the visceral perception and colon mucosal changes in that model. Methods: Sixteen rats were randomly divided into experimental group and control group, and were instilled in colon with 1 ml of 200 mmol/L sodium butyrate solution or 0.9% NaCl solution, respectively, twice a day for 3 days. Score of abdominal withdrawal reflex (AWR) and pressure threshold value of visceral perception after colorectal distension (CRD) were determined before and 3, 6, 9, 12, 15 and 18 days after the starting of coloclysis. Rats were sacrificed at the end of the experiment. The gross morphological and histological changes of colon mucosa were observed. Results: Under various CRD pressures the AWR score was significantly higher and the pressure threshold value of pain perception was significantly lower in experimental group than those in the controls at the 3rd, 6th, 9th and 12th day of the experiment (P<0.05), and returned to the initial status at the 15th and 18th day. At the end of the experiment, no significant differences were found in gross morphological and histological changes of colonic mucosa between the experimental and control groups. Conclusions: Repeated coloclysis with sodium butyrate solution can induce colon hypersensitivity in rats, and colon mucosal changes recover in parallel with the normalization of visceral perception. This visceral hypersensitivity model can be used in the studies of pathophysiologic mechanism of IBS.

Abstract:Visceral hypersensitivity (VH) is a significant contributor to irritable bowel syndrome (IBS). Oxytocin (OT) possesses analgesic effects on the central nervous system (CNS) and attenuates microglial activation, however, little is known about its peripheral effects and involvement in VH of IBS. Reactive enteric glial cells (EGCs) contributes to abnormal motility in gastrointestinal (GI) diseases. The aim of this study was to evaluate the peripheral use of OT to maintain VH and activation of EGCs through involvement of the Toll-like receptor (TLR) 4/MyD88/NF-κB signaling. After assessing a baseline visceromotor response (VMR) to colorectal distension (CRD), rats were exposed to a 1h water avoidance stress (WAS) session. Before each WAS session, intraperitoneal injection of OT (1mg/kg body weight, in phosphate-buffered saline (PBS)) atosiban (0.5mg/kg body weight, in PBS) or PBS (as a vehicle control, 1ml/kg body weight) was administered. Animas are killed 24h after the last WAS session. EGCs activity, relative OT receptor expression, glial fibrillary acidic protein (GFAP) expression and TLR4/MyD88/NF-κB signaling were evaluated. Neonatal maternal separation (MS) significantly increased the OT receptor expression and enhanced VMR to CRD. WAS improved VMR to CRD only during neonatal MS. OT treatment prevented WAS-induced higher VMRs to CRD, which was reversed by an OT receptor antagonist administration. Compared to the vehicle, OT pre-treated rats reduced EGCs activation, GFAP expression and TLR4/MyD88/NF-κB signaling. We conclude that neonatal MS induces VH and visceral pain in rats. Furthermore, exogenous OT attenuated stress-induced VH and EGCs activation, which was mediated by TLR4/MyD88/NF-κB signaling.

Abstract:We assessed dynamic changes in visceral hypersensitivity and fecal metabolomics through a mouse model of irritable bowel syndrome (IBS) from childhood to adulthood. A mouse model of IBS was constructed with maternal separation (MS) in early life. Male mice aged 25, 40, and 70 days were used. Visceral sensitivity was assessed by recording the reaction between the abdominal withdrawal reflex and colorectal distension. Metabolomics was identified and quantified by liquid chromatography-tandem mass spectrometry. The visceral sensitivity of the MS group was significantly higher than that of the non-separation (NS) group in the three age groups. The top four fecal differential metabolites in the different age groups were lipids, lipid molecules, organic heterocyclic compounds, organic acids and derivatives, and benzenoids. Five identical differential metabolites were detected in the feces and ileal contents of the MS and NS groups at different ages, namely, benzamide, taurine, acetyl-L-carnitine, indole, and ethylbenzene. Taurine and hypotaurine metabolism were the most relevant pathways at P25, whereas histidine metabolism was the most relevant pathway at P40 and P70. Visceral hypersensitivity in the MS group lasted from childhood to adulthood. The different metabolites and metabolic pathways detected in MS groups of different ages provide a theoretical basis for IBS pathogenesis.

Abstract:Background: The mechanism underlying chronic stress-induced gastrointestinal (GI) dysmotility has not been fully elucidated and GI hormones have been indicated playing a role in mediating stress-induced changes in GI motor function. Aims: Our objective was to study the possible role of substance P (SP) in the colonic hypermotility induced by repeated water avoidance stress (WAS) which mimics irritable bowel syndrome. Methods: Male Wistar rats were submitted to WAS or sham WAS (SWAS) (1h/day) for up to 10 consecutive days. Enzyme Immunoassay Kit was used to detect the serum level of SP. The expression of neurokinin-1 receptor (NK1R) was investigated by Immunohistochemistry and Western blotting. The spontaneous contraction of muscle strip was studied in an organ bath system. L-type calcium channel currents (ICa,L) of smooth muscle cells (SMCs) were recorded by whole-cell patch-clamp technique. Results: Fecal pellet expulsion and spontaneous contraction of proximal colon in rats were increased after repeated WAS. The serum level of SP was elevated following WAS. Immunohistochemistry proved the expression of NK1R in mucosa, muscularis and myenteric plexus. Western blotting demonstrated stress-induced up-regulation of NK1R in colon devoid of mucosa and submucosa. Repeated WAS increased the contractile activities of longitudinal muscle and circular muscle strips induced by SP and this effect was reversed by a selective NK1R antagonist. The ICa,L of SMCs in the WAS rats were drastically increased compared to controls after addition of SP. Conclusions: Increased serum SP level and up-regulated NK1R in colon may contribute to stress-induced colonic hypermotility. And L-type calcium channels play a potentially important role in the process of WAS-induced dysmotility.
