Abstract:Atractylis lancea (Thunb.) DC. (AL), an important medicinal herb in Asia, has been shown to have anti-tumor effects on cancer cells, but the involved mechanisms are poorly understood. This study focused on potential effects and molecular mechanisms of AL on the proliferation of the Hep-G2 liver cancer cell line in vitro. Cell viability was assessed by MTT test in Hep-G2 cells incubated with an ethanol extract of AL. Then, the effects of AL on apoptosis and cell cycle progression were determined by flow cytometry. Telomeric repeat amplification protocol (TRAP) assays was performed to investigate telomerase activity. The mRNA and protein expression of human telomerase reverse transcriptase (hTERT) and c-myc were determined by real-time RT-PCR and Western blotting. Our results show that AL effectively inhibits proliferation in Hep-G2 cells in a concentration and time-dependent manner. When Hep-G2 cells were treated with AL after 48h,the IC50 was about 72.1 mu g/mL. Apoptosis was induced by AL via arresting the cells in the G1 phase. Furthermore, AL effectively reduced telomerase activity through inhibition of mRNA and protein expression of hTERT and c-myc. Hence, these data demonstrate that AL exerts anti-proliferative effects in Hep-G2 cells via down-regulation of the c-myc/hTERT/telomerase pathway.

Abstract:Objective:To research the anti-proliferation effect of extract of Rhizoma Atractylodis on A549 cells.Method:Rhizoma Atractylodis was extracted by 90% ethanol using ultrasonic method for 1h,then supernatant was collected by centrifugation;The proliferation activity of A549 inhibited by extract of Rhizoma Atractylodis was measured by MTT assay.The effect of extract of Rhizoma Atractylodis on cell cyclic was detected by flow cytometry;cyclin D1 protein expression was measured by Western Blot.Result: Extract of Rhizoma Atractylodis can effectively inhibit the proliferation of A549 in dosage and time dependent manner,the best inhibitory effect was appeared on 48h,the IC50 is about 77.12μg/ml;extract of Rhizoma Atractylodis can block cell cyclic in G0/G1 phase,the G1 phase was increased to 53.41% when A549 cells inhibited for 48h and effectively decrease cyclin D1 protein expression.Conclusion: Rhizoma Atractylodis can block cell cyclic in G0/G1 phase via decrease cyclin D1 protein to realize anti-proliferation effect.

Abstract:Hebanthe eriantha is a medicinal plant used in folk medicine and a subject of commercial interest. The cytotoxicity effects from H. eriantha root extracts on cancerous and normal cells were assessed by the MTT method, and in vitro toxicity was evaluated on Artemia salina . The inhibition of the proliferation of bacteria and MIC values were examined by the disc diffusion and the broth microdilution method, respectively. Human colon cancer HCT116 and mouse breast tumour model 4T1 cells treated with methanolic extract showed a significant decrease in viability of cells with IC 50 : 272.6 and 88.5 μg/mL at 72h, respectively. The methanolic extract of H. eriantha showed moderate toxicity against A. salina (LC 50: 589.4 μg/mL). In antimicrobial activity, the methanolic extract showed the highest inhibitory function against S. aureus and P. vulgaris (17.5 and 16 mm) with MICs of 500 μg/mL. Abstract:Artemisia (Asteraceae family) has been reported to possess different bioactive phytochemicals including phenols, sesquiterpene lactones and flavonoids. The aim of this study was to estimate the anti-proliferative activity of the methanolic extract of Artemisia herba alba using in vitro assays on two of human cancer cell lines compared with the normal cell lines. The obtained results indicated that A.herba alba extract showed highly cytotoxic and anti-proliferative effects on tested human cancer cells as revealed by measuring viable mitochondrial oxidation of [3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay] (MTT assay). The observed (-IC50-) or the half-maximal inhibitory concentration of A. herba alba extract in the human hepatocellular carcinoma (HepG2) was 50.34 μg/ml, while in the lung cancer cell line (A549) it was 29.23 μg/ml. In contrast, the normal liver cells (THLE2) as well as lung normal cells showed very low sensitivity toward tested extract where the IC50 were 1250μg/ml and 1915 μg/ml, respectively. However, flow cytometric analysis for cell cycle using propidium iodide staining revealed increment of G2/M phase cell cycle arrest after treatment of HepG2 cells with A. herba alba extract. At the molecular level, the quantitative real time-PCR technique was used to investigate the alteration of gene expression after exposure of HepG2 cell line to A. herba alba extract by measuring levels of mRNA for p53, Bax, and Bcl-2 genes expression. The apoptotic mechanism was activated by the crude extract of A. herba alba extract included up-regulation of p53 and Bax and down-regulation of Bcl-2 expression levels. However, no cytotoxic effect was recorded after exposure of normal cell lines to the plant extract. These data indicated that A. herba alba extract possess anti-proliferative effect by a cell cycle arrest at the G2/M phase and apoptosis mediated cytotoxicity in carcinoma cells. These results suggested that A.herba alba could contain one or more effective anti-cancer compound(s) and can be used as a natural source of anticancer agents.

Abstract:Metal cation homeostasis is essential for plant nutrition and resistance to toxic heavy metals. Many plant metal transporters remain to be identified at the molecular level. In the present study, we have isolated AtNramp cDNAs from Arabidopsis and show that these genes complement the phenotype of a metal uptake deficient yeast strain, smf1. AtNramps show homology to the Nramp gene family in bacteria, yeast, plants, and animals. Expression of AtNramp cDNAs increases Cd(2+) sensitivity and Cd(2+) accumulation in yeast. Furthermore, AtNramp3 and AtNramp4 complement an iron uptake mutant in yeast. This suggests possible roles in iron transport in plants and reveals heterogeneity in the functional properties of Nramp transporters. In Arabidopsis, AtNramps are expressed in both roots and aerial parts under metal replete conditions. Interestingly, AtNramp3 and AtNramp4 are induced by iron starvation. Disruption of the AtNramp3 gene leads to slightly enhanced cadmium resistance of root growth. Furthermore, overexpression of AtNramp3 results in cadmium hypersensitivity of Arabidopsis root growth and increased accumulation of Fe, on Cd(2+) treatment. Our results show that Nramp genes in plants encode metal transporters and that AtNramps transport both the metal nutrient Fe and the toxic metal cadmium.
Potential Antiproliferative and Antimetastatic Effects of Artemisia eriantha: An In Vitro Study Focused on Hepatocarcinoma Cells

Abstract:Artemisia eriantha is a subendemic species of the Central Apennine, valued locally for its applications in ancient and traditional medicine for its antibacterial, antifungal, anthelmintic, digestive, and antispasmodic effects. Several of these properties are also found in other species within the same genus, including recent findings highlighting their anti-tumor actions. However, the presence of cytotoxic or anti-tumor activity has never been studied in A. eriantha. The aim of this study was thus to assess the potential anti-proliferative, pro-apoptotic, and antimetastatic effects of this plant using in vitro models of hepatocarcinoma cells (HepG2 and Huh7 cells). Treatment with A. eriantha extract (130 μg/mL) resulted in the reduced proliferation of HepG2 and Huh7 cells, driven by the dysregulation of proteins involved in the cell cycle (such as cyclin D1, cyclin E, and p27) and the activation of the p53-p21 pathway, leading to an increase in apoptotic activity. In addition, the treatment also lowered the healing capability after scratch wounds in Huh7 cells, which are characterized by a remarkable migratory capacity, suggesting that the A. eriantha extract may also be effective in controlling the migration and spread of metastatic cells. In conclusion, A. eriantha extract 130 μg/mL appears to interfere with key factors of HepG2 and Huh7 cell proliferation and invasion and emerges as a potential new adjuvant for the prevention and/or treatment of hepatocellular carcinoma. Additional research is necessary to establish if the effects observed are cell-specific and to elucidate the mechanisms of action and signaling pathways underlying its effectiveness.

Abstract:Background: Aegle marmelos belong to the family rutaceae. It is commonly known as Bael. It is used in traditional medicine, as it has antidiarrheal, antimicrobial, antiviral, radio-protective, anticancer, chemo-preventive properties which are of great medicinal use. Liver cancer is the 5th most common type of cancer, Hepatocellular carcinoma is the most common form, which originates from the liver. Aegle marmelos is said to inhibit the proliferative action of cancer cells. Objective: To investigate the role of Aegle marmelos against human liver cancer cells (HEP G2 cell line) by inhibiting the activity of matrix metalloproteinases which is responsible for spread of cancer. Materials and methods: HEP G2 cell lines were procured from NCCS (National center for cell sciences) Pune, India. It was cultured and viability of the cells before and after adding the extract was analysed using the MTT assay. mRNA amplification was done using real time PCR. Statistical analysis was done using ANOVA and dunken’s multiple test. Corresponding graphs are also plotted. Results: The viability of the cells decreased from 100% to 50%. The mRNA expression of MMP-2 and MMP-9 decreased after the addition of the extract. Conclusion: From this study we can conclude that Aegle marmelos, a novel innovative anticancer drug inhibited the proliferative action of liver cancer cells by reducing the expression of MMP 2 and MMP 9 and possesses anti-cancer activity.
Hypericum alpestre extract exhibits in vitro and in vivo anticancer properties by regulating the cellular antioxidant system and metabolic pathway of L‐arginine

Abstract:Conventional treatment methods are not effective enough to fight the rapid increase in cancer cases. The interest is increasing in the investigation of herbal sources for the development of new anticancer therapeutics. This study aims to investigate the antitumor capacity of Hypericum alpestre (H. alpestre) extract in vitro and in vivo, either alone or in combination with the inhibitors of the l‐arginine/polyamine/nitric oxide (NO) pathway, and to characterize its active phytochemicals using advanced chromatographic techniques. Our previous reports suggest beneficial effects of the arginase inhibitor NG‐hydroxy‐nor‐ l‐arginine and NO inhibitor NG‐nitro‐Larginine methyl ester in the treatment of breast cancer via downregulation of polyamine and NO synthesis. Here, the antitumor properties of H. alpestre and its combinations were explored in vivo, in a rat model of mammary gland carcinogenesis induced by subcutaneous injection of 7,12‐dimethylbenz[a]anthracene. The study revealed strong antiradical activity of H. alpestre aerial part extract in chemical (DPPH/ABTS) tests. In the in vitro antioxidant activity test, the H. alpestre extract demonstrated pro‐oxidant characteristics in human colorectal (HT29) cells, which were contingent upon the hemostatic condition of the cells. The H. alpestre extract expressed a cytotoxic effect on HT29 and breast cancer (MCF‐7) cells measured by the MTT test. According to comet assay results, H. alpestre extract did not exhibit genotoxic activity nor possessed antigenotoxic properties in HT29 cells. Overall, 233 substances have been identified and annotated in H. alpestre extract using the LC‐Q‐Orbitrap HRMS system. In vivo experiments using rat breast cancer models revealed that the H. alpestre extract activated the antioxidant enzymes in the liver, brain, and tumors. H. alpestre combined with chemotherapeutic agents attenuated cancer‐like histological alterations and showed significant reductions in tumor blood vessel area. Thus, either alone or in combination with Nω‐OH‐nor‐ l‐arginine and Nω‐nitro‐ l‐arginine methyl ester, H. alpestre extract exhibits pro‐ and antioxidant, antiangiogenic, and cytotoxic effects.

Antiproliferative effects of extracts from Salvia officinalis L. and Saliva miltiorrhiza Bunge on hepatocellular carcinoma cells

Abstract:Salvia species have been used as valuable medicinal and herbal plant in many countries. Salvia officinalis L. and Salvia miltiorrhiza Bunge are widely used in traditional medicine for a long time. In the present study, cytotoxicity of ethanol and acetone extracts prepared from leaves and roots of two Salvia species was investigated using hepatocellular carcinoma cells (HepG2) and normal human liver cells (WRL-68). The cytotoxicity and anti-proliferative abilities of the extracts were evaluated by measuring cell viability (MTS assay), lactate dehydrogenase (LDH) leakage, the cellular ATP level, morphological changes using an inverted microscope, and apoptosis using flow cytometry. The results indicated that ethanol and acetone extracts of leaves and roots of S. officinalis (SO-L-E, SO-L-A, SO-R-E and SO-R-A, respectively) and ethanol and acetone extracts of roots of S. miltiorrhiza (SM-R-E and SM-R-A, respectively) significantly inhibited the proliferation of HepG2 cells in a time- and dose-dependent manner when the concentration was less than 150μg/mL. The cytotoxity of SO-L-E, SO-R-E and SO-R-A were significantly less in WRL-68 when compared to HepG2 cells in vitro. The increase of LDH leakage, decrease of ATP and the changes in morphology of HepG2 cells further confirmed the cytotoxic effect of these extracts to HepG2 cells. Furthermore, SO-L-E, SO-L-A, SO-R-E ethanol extract of leaves of S. miltiorrhiza (SM-L-E) and SM-R-E were able to induce apoptosis in HepG2 cells. This study shows the potential of the extracts to be used in the prevention and/or treatment of liver cancer or as ingredients in functional foods and provides scientific support for development and utilization of S. officinalis and S. miltiorrhiza, especially the roots of S. officinalis.

Abstract:Background: The bark of Uncaria rhynchophylla has been traditionally used to treat convulsion, bleeding, hypertension, auto-immune conditions, cancer, and other diseases. The main focus of this research is done for the purpose of exploring the antitumor activity and mechanism of action (MOA) for hirsutine isolated from U. rhynchophylla. Methods: Jurkat clone E6-1 cells were treated using 10, 25 and 50 μM for 48 h. Inhibition of cell proliferation due to hirsutine treatment was evaluated by CCK8 assay. Flow cytometry was applied to ascertain Jurkat cell cycle progression and apoptosis after treatment with 10, 25 and 50 μM hirsutine for 48 h. The expression and level of the apoptosis-related genes and proteins was analyzed by Real-time Quantitative polymerase chain reaction (qPCR) and Western blotting method, respectively. Results: CCK8 analyses revealed that hirsutine could significantly inhibit the proliferation of Jurkat clone E6-1 cells, in a concentration and time-dependent fashion. Flow cytometry assays revealed that hirsutine could drive apoptotic death and G0/G1 phase arrest in Jurkat cells. Apoptotic cells frequencies were 4.99 ± 0.51%, 13.69 ± 2.00% and 40.21 ± 15.19%, and respective cell cycle arrest in G0/G1 accounted for 34.85 ± 1.81%, 42.83 ± 0.70% and 49.12 ± 4.07%. Simultaneously, compared with the control group, Western blot assays indicated that the up-regulation of pro-apoptotic Bax, cleaved-caspase3, cleaved-caspase9 and Cyto c proteins, as well as the down-regulation of Bcl-2 protein which guards against cell death, might be correlated with cell death induction and inhibition of cell proliferation. QPCR analyses indicated that hirsutine could diminish BCL2 expression and, at the same time, improve Bax, caspase-3 and caspase-9 mRNA levels, thus reiterating a putative correlation of hirsutine treatment in vitro with apoptosis induction and inhibition of cell proliferation (p-value < 0.05). Excessive hirsutine damages the ultrastructure in mitochondria, leading to the release of Cyt c from the mitochondria to cytoplasm in Jurkat clone E6-1 cells, thereby inducing the activated caspase cascade apoptosis process through a mitochondria-mediated pathway. Conclusion: An important bioactive constituent-hirsutine-appears to have antitumor effects in human T-cell leukemia, thus enlightening the use of phytomedicines as a novel source for tumor therapy. It is speculated that hirsutine may induce apoptosis of Jurkat Clone E6-1 cells through the mitochondrial apoptotic pathway.
Ethanol extract of Lophatheri Herba exhibits anti-cancer activity in human cancer cells by suppression of metastatic and angiogenic potential

Abstract:Lophatheri Herba (LH), dried leaf of Lophatherum gracile Brongn, has long been used to reduce thirst and treat fever and inflammation in Chinese medicine. Recent studies have shown that LH has anti-viral, anti-bacterial, anti-cancer, anti-oxidant, diuretic, and hyperglycemic properties. However, the effects of an ethanol extract of L. herba (ELH), at non-cytotoxic doses, on the metastatic and angiogenic abilities of malignant tumor cells have not been reported. We found that ELH significantly suppressed p38, JNK, and NF-κB activation and proteolytic activities under phorbol 12-myristate 13-acetate (PMA) stimulation, thus leading to a decrease in metastatic potential, including migration and invasion. In addition, ELH suppressed tumor-induced angiogenesis, including migration and tube formation in human umbilical vein endothelial cells (HUVECs) and microvessel sprouting from aortic rings via decreasing the pro-angiogenic factors in tumors. Interestingly, in ovo xenografts ELH-treated HT1080 cells did not increase in volume and eventually disappeared, owing to a lack of angiogenesis. Daily oral administration of ELH at 50 and 100 mg/kg markedly inhibited metastatic colonization of B16F10 cells in the lungs of C57BL/6J mice and caused no apparent side effects. These data collectively indicate that ELH is safe and may be useful for managing metastasis and growth of malignant cancers.

Abstract:Bioactive constituents of numerous marine organisms have been investigated recently for their preclinical and clinical anticancer activity. Three marine organisms: black-spotted sea cucumber: Pearsonothuria graeffei (Pg), lollyfish: Holothuria atra (Ha), and sea hare: Aplysia dactylomela (Ad), were collected during winter 2019 from Gulf of Aqaba, Red Sea, Egypt, and macerated with ethanol into three different extracts: PgE, HaE, and AdE, where each was in vitro assessed for its antiproliferative and proapoptotic properties on HepG2, HCT-116, and MCF-7 cancer cells. PgE dose-dependently inhibited the growth of HepG2, HCT-116, and MCF-7 cells within IC50 values 16.22, 13.34, and 18.09 μg/mL, respectively, while the IC50 values for the antiproliferative activity of HaE were 12.48, 10.45, and 10.36 μg/mL, respectively, and the IC50 values of AdE were 6.51, 5.33, and 6.87 μg/mL, respectively. All extracts were found to induce G0/G1 cell cycle arrest for HepG2 cells side by side with their inhibition of CDK2 on all three cell lines while all extracts were also showed to induce apoptosis in HepG2 cell line at pre-G1 phase supplemented by their anticancer activity via proapoptotic protein Bax, caspase-3, and cleavage PARP increase, and antiapoptotic protein Bcl-2 downturn. Moreover, necrosis has been relatively noticed in HepG2 cell line as an additional anticancer activity for each extract. Our data introduced three ethanolic marine extracts as natural chemotherapeutic agents to be further developed for cancer control.

Abstract:Background/Aim. Plant polyphenols are well known to show antimutagenic, anticarcinogenic, antiviral and antioxidative activity. The aim of this study was to investigate bio-active potential of Armoracia rusticana root juice and extracts: their polyphenol content, as well as in vitro antitumor activity and cell-death mechanism. Methods. Liquid-liquid extraction of polar and non-polar compounds was used and poly-phenolic compounds were identified and quantified by high performance liquid chomatography (HPLC) analysis. Anti-proliferative activity was examined in vitro on human cervix carcinoma (HeLa), breast adenocarcinoma (MCF7, MDA-MB-231), colon adenocarcinoma (HT-29), lung adenocarcinoma (A549), prostate adenocarcinoma (PC-3), melanocyte carcinoma (Hs 294T), hepatocyte carcinoma (Hep G2), as well as rat hepatocyte carcinoma (H-4-II-E), and normal human fetal lung (MRC-5) cell line using sulforhodamine B assay. The mechanism of cell-death in cell line was determined using Cell Death Detection ELISAPLUS kit. Results. Dichloromethane extracts had the highest content of cate-chin, p-hydroxybenzoic, syringic and gallic acid (pulp, E1), and epicatechin (juice, E3). The results showed strong and non-selective antiproliferative activity of chloroform and di-chloromethane extracts and root juice ? highest being to-wards liver, breast and lung tissue cells. IC50 values of ex-tracts and juice had low range of concentrations (IC50 = 3.49?26.5 ?g/mL) and high range of dilutions (IC50 = 418?1,590). High and unfavorable potential of horseradish juice and chloroform juice extract (E4) to induce necrotic cell death was detected. Conclusion. Strong and non-selective in vitro antiproliferative activity of chloroform and dichloromethane extracts and root juice of horseradish was detected, with necrosis as a main mechanism of induced cell death. In order to utilize horseradish root bioactive potential further investigations that will pinpoint active components with more favourable apoptosis/necrosis inducing properties are needed.

Antiproliferative Activity of Hypericum perforatum, Achillea millefolium, and Aloe vera in Interaction with the Prostatic Activity of CD82

Abstract:Background: In recent years, prostate cancer prevails as one of the lead cancers affecting men. Currently, prostate cancer research involves the phytochemical study of plants with anti-tumour effects. This study compares the anti-tumour effects of three plant species indigenous to Iran and their interaction with cluster of differentiation (CD)-82 protein, a therapeutic target found in prostate cancer cells. Methods: The extracts of Hypericum perforatum, Achillea millefolium, and Aloe vera were prepared and their toxicological, cellular and gene expression responses were evaluated in PC-3 human prostate cancer cells and normal human chondrocyte cell line C28/I2. They were exposed to different concentrations of the plants (10 mg/mL, 5 mg/mL, 1 mg/mL, 100 µg/mL, 10 µg/mL, and 1 µg/mL) at three exposure time points (24, 48, 72 hours) to determine cancer cell cytotoxicity and gene expression profiles. Results: : Half-maximal inhibitory concentration (IC50) in PC-3 cells ranged from 0.6 to 8.5 mg/mL for H. perforatum extract, from 0.4 to 7.5 mg/mL for A. Millefolium extract, and from 0.2 to 8.0 mg/mL for A. vera extract in a time-dependent manner. A. vera extract caused the highest cell death levels in PC-3 cells (94%) and C28/I2 cells (57%) after 48 hours. A 1.97-, 3.00-, and 3.48-fold increase in relative gene expression of CD82 was observed for H. perforatum, A. millefolium, and A. vera extracts, respectively. Conclusion: A. vera and A. millefolium extracts are a selective inhibitor of prostate cancer cells and a potent activator of CD82 expression.

Abstract:Development of new therapeutic strategies for breast cancer is urgently needed due to the sustained emergence of drug resistance, tumor recurrence and metastasis. To gain a novel insight into therapeutic approaches to fight against breast cancer, the cytocidal effects of hellebrigenin (Helle) and arenobufagin (Areno) were investigated in human estrogen receptor (ER)-positive breast cancer cell line MCF-7 and triple-negative breast cancer cell line MDA-MB-231. Helle exhibited more potent cytotoxicity than Areno in both cancer cells, and MCF-7 cells were more susceptible to both drugs in comparison with MDA-MB-231 cells. Apoptotic-like morphological characteristics, along with the downregulation of the expression level of Bcl-2 and Bcl-xL and the upregulation of the expression level of Bad, were observed in Helle-treated MCF-7 cells. Helle also caused the activation of caspase-8, caspase-9, along with the cleavage of poly(ADP-ribose) polymerase in MCF-7 cells. Helle-mediated necrosis-like phenotype, as evidenced by the increased propidium iodide (PI)-positive cells was further observed. G 2 /M cell cycle arrest was also induced by Helle in the cells. Upregulation of the expression level of p21 and downregulation of the expression level of cyclin D1, cyclin E1, cdc25C and survivin were observed in MCF-7 cells treated with Helle and occurred in parallel with G 2 /M arrest. Autophagy was triggered in MCF-7 cells and the addition of wortmannin or 3-MA, two well-known autophagy inhibitors, slightly but significantly rescued the cells. Furthermore, similar alterations of some key molecules associated with the aforementioned biological phenomena were observed in MDA-MB-231 cells. Intriguingly, the numbers of PI-positive cells in Helle-treated MCF-7 cells were significantly reduced by wortmannin and 3-MA, respectively. In addition, Helle-triggered G 2 /M arrest was significantly corrected by wortmannin, suggesting autophagy induction contributed to Helle-induced cytotoxicity of breast cancer cells by modulating necrosis and cell cycle arrest. Collectively, our results suggested potential usefulness of both Helle and Areno in developing therapeutic strategies to treat patients with different types of breast cancer, especially ER-positive breast cancer.

Abstract:Epilobium hirsutum is extensively used as a traditional remedy in folk medicine, especially against prostate inflammation. Therefore, we evaluated the chemical profiles and biopharmaceutical potentials of different extracts of E. hirsutum aerial parts and roots. Metabolomic, antioxidant, and enzyme inhibitory profiles were investigated. Human prostate cancer PC3 cells were exposed to the extracts to evaluate antiproliferative effects. Gene expression and bioinformatics analyses were performed to investigate anti-inflammatory mechanisms. Oenothein B and myricetin were prominent compounds in the extracts. In scavenging/reducing assays, the methanol, infusion, and methanol/water extracts exhibited similar activities. We also observed the reduction of PC3 viability occurring following exposure to methanol and methanol/water extracts. According to bioinformatics analysis, myricetin was predicted to interact with COX-2 and TNFα. The interaction between TNFα and oxo-dihydroxy-octadecenoic acid was predicted as well. Intriguingly, the gene expression of COX-2 and TNFα was reduced in PC3 cells after exposure to methanol and methanol/water extracts. These effects were paralleled by the decreased gene expression of IL-8 and NFkB and the inhibition of PGE2 release. Therefore, the present findings suggest the potential use of E. hirsutum for the management of the burden of inflammation and oxidative stress occurring in lower urinary tract diseases, including prostatitis.

Abstract:The methanol extract of the whole plant of Hedyotis chrysotricha demonstrated cytotoxicity against SK-HEP-1 human hepatocarcinoma cells in a primary screening for novel antitumour agents. Bioassay-guided fractionation and purification led to an active principle (24S)-ergostane-3,5,6-triol (1) along with four inactive compounds (2-5). The invitro transwell migration assay showed that compound 1 remarkably reduced the migration of SK-HEP-1 cells by 78.9% at a dose of 30 mu M, without any apoptotic effect on this cell line. Moreover, all the isolated compounds were further evaluated for their cytotoxicities against another four human cancer cell lines (MCF-7, NUGC3, SH-SY5Y and PC-3).

Abstract:Background: Ethyl acetate extracts from Tetrastigma hemsleyanum (Sanyeqing) (EFT), a member of the Vitaceae plant family, have been shown to exhibit efficacy against a variety of cancers. In this light, our current study seeks to examine the mechanism of efficacy between EFT extracts and human pancreatic cancer PANC-1 cells. Methods: The chemical components of EFT were analyzed by gas chromatography-mass spectrometry. The cytotoxicity of EFT on PANC-1 cells was measured using an MTT assay. In order to investigate EFT induction of cell cycle arrest, changes in cell-cycle distribution were monitored by flow cytometry. Wound healing and transwell assays were employed to investigate whether migration and invasion of PANC-1 cells were inhibited by EFT. Relative protein expression was detected using Western blot. Results: GC-MS analysis of the chemical composition of EFT revealed that the majority of constituents were organic acids and their corresponding esters. EFT exhibits measurable cytotoxicity and inhibition of PANC-1 invasion. Growth inhibition was primarily attributed to downregulation of CDK2 which induces cell cycle arrest in the S-phase. Inhibition of metastasis is achieved through downregulation of mesenchymal-associated genes/activators, including ZEB1, N-cadherin, Vimentin, and Fibronectin. Meanwhile, the expression of E-cadherin was significantly increased by EFT treatment. Furthermore, downregulation of MMP-2 and MMP-9 were observed. Conclusion: Treatment of PANC-1 with EFT demonstrated measurable cytotoxic effects. Furthermore, EFT evoked S phase arrest while inhibiting the migration and invasion of PANC-1 cells. Additionally, EFT inhibited the epithelial to mesenchymal transition and MMPs expression in PANC-1 cells. This study serves to confirm the strong therapeutic potential of EFT while identifying the mechanisms of action.
