Abstract:Objective To investigate the effects of asiatic acid (AA) on isoproterenol (ISO) induced myocardial hypertrophy. Methods In?vitro cultured rat myocardial cells H9c2 were divided into the control group,ISO group and ISO+AA group. The H9c2 cells in the ISO group were treated with 10 mmol/L ISO to induce cardiomyocyte hypertrophy,those in ISO+AA group with different concentrations of AA and 10 mmol/L ISO,and those in the control group with 0.5%dimethyl sulfoxide(DMSO)as a vehicle control. Real?time quantitative PCR was performed to detect the expressions of atrial natriuretic peptide(ANP),β?myosin heavy chain(β?MHC),glutathione peroxidase(GSH?Px),superoxide dismutase(SOD)and hemoglobin oxygenase 1(HO?1)genes. Immunofluorescence staining was used to measure cross?sectional area of the cardiomyocytes. Western blot was used to detect the changes in signaling pathway related to myocardial hypertrophy. DCFH?DA fluorescent probe was used to detect the intracellular levels of reactive oxygen species(ROS). Results As shown by real?time quantitative PCR,mRNA expression of cardiac hypertrophy markers ANP andβ?MHC in H9c2 cells was up?regulated after 48 h treatment with ISO compared with the control group(both P<0.05),while 48 h intervention with 5,10 or 20μmol/L AA significantly reduced the ISO?induced up?regulation of ANP andβ?MHC mRNA expression(all P<0.05)in a concentration?dependent manner;and treatment with 20μmol/L AA for 12,24 or 48 h was shown to inhibit ISO?induced ANP andβ?MHC mRNA up?regulation in H9c2 cells(all P<0.05). Cross?sectional area of cardiomyocytes as measured by immunofluorescence staining was(1 946 ± 49)μm2 in the control group,(2 952 ± 111)μm2 in the ISO group and(2 548 ± 59)μm2 in the ISO(10 mmol/L)+AA(20 μmol/L)group. Compared with the control group,cross?sectional area of cardiomyocytes increased in ISO group,while 20μmol/L AA could significantly reduce the ISO?induced myocardial hypertrophy(both P<0.05). Western blotting indicated that ISO could lead to activation of AKT/GSK3β and ERK/JNK related to cardiac hypertrophy,while 20 μmol/L AA could inhibit the activation of these signaling pathways. DCFH?DA fluorescent probe assay showed that 20μmol/L AA could significantly inhibit the ISO induced increase in intracellular ROS levels. Real?time quantitative PCR showed that 20μmol/L AA could inhibit ISO?induced up?regulation of GSH?Px,SOD and HO?1 mRNA expression. Conclusion AA may attenuate oxidative stress by blocking the AKT/GSK3β and ERK/JNK signaling pathways,thereby inhibiting the ISO?induced myocardial hypertrophy.
Chlorogenic acid prevents acute myocardial infarction in rats by reducing inflammatory damage and oxidative stress

Abstract:Recent studies have suggested that the prevention of myocardial infarction (MI) through diet is very important and that the intake of polyphenol-rich foods can improve cardiovascular health. In this study, adult male SD rats were randomly divided into 2 groups. The chlorogenic acid (CGA) group (n = 18) was administered 100 mg/kg/day CGA by gavage, and the control (CON) group (n = 18) was given the equivalent volume of water for 4 weeks. A model of MI was established by ligating the left anterior descending (LAD) coronary artery, which was monitored by an electrocardiogram (ECG). Blood samples were analyzed by enzyme-linked immunosorbent assays and biochemical experiments 24 h after the operation. In addition, histopathological analysis was performed to assess the size and severity of the infarct area. The administration of CGA before MI minimized weight gain and was associated with decreased postoperative mortality. CGA moderated the coronary artery ligation-induced changes observed by ECG and decreased the plasma levels of the myocardial markers. In the histopathological analysis, CGA notably reduced infarct size and decreased myocardial injury and fibrosis. Furthermore, CGA significantly reduced the levels of pro-inflammatory factors, and this reduction was accompanied by an upregulation of anti-inflammatory cytokines and an increase in antioxidant enzyme activities. This study indicated that CGA improved the survival rate after MI and demonstrated that CGA had a protective effect on MI by reducing the inflammatory response and exerting antioxidant activity.

Abstract:The first volume of Literature and Medicine is frankly introductory. It bears the subtitle Toward a New Discipline and is packed with speculative and explanatory pieces about the conjunction of two apparently disparate kinds of knowledge.1 Was literature and medicine to be a scholarly discipline? A movement to reform medical practice? A service teaching enterprise or a more imperial intellectual one? Were the two halves, the fields of literature and medicine, to be equally balanced? If not, which was to predominate and what would be the proportions of the imbalance? Could there be an intellectual synergy, a genuinely new discipline? Although in 1982 there were only three full-time professors of literature in American medical schools (and two of them had held their positions for less than five years), literature had not been entirely absent from the medical curriculum. Theologians and professors of religious studies, who played a major role as members of Ministers in Medical Education in founding the Society for Health and Human Values, often used literature in their medical-school lectures and A©lectives, and a few physicians incorporated literature, particularly short fiction, in their assignments and talks to students and residents. This demographic state of affairs is reflected in a census of the authors in volume 1. There are twenty essays and an introduction: five by authors in religious studies, seven by physicians, nine by literature professors (including a poet who had held academic positions). Literature and medicine was not yet a discipline by prevailing academic standards. It was too friendly, too ill defined, too shapeless. It was energetic, hopeful, avocational, unprofessional. Nevertheless, the first volume oA- Literature and Medicine, as its subtitle suggests, is something of an outline of the directions that the enterprise, still in search of definition and legitimacy, might take. Half the essays are about the uses of literature in medicine, primarily the contribution that literature can make to medical education. Cardioprotective Effects of Callicarpa Tomentosa Leaf Extract in Wistar Albino Rats against Isoproterenol-induced Myocardial Necrosis: Phytochemical Analysis and In Vitro Antioxidant Study

Abstract:Background Globally, cardiovascular disease (CVD) remains the foremost cause of mortality. Conventional medications are associated with several complications, necessitating an alternative, safe and efficacious medicine. Therefore, the study was planned to assess the cardioprotective efficacy of Callicarpa tomentosa (CT) leaf extract against isoproterenol (ISO)-induced myocardial failure in experimental rats. Materials and methods CT leaf extract was subjected to phytochemical analysis, total phenolic and flavonoid content measurement, and in vitro antioxidant evaluation. Additionally, the protection from myocardial injury was evaluated based on the estimation of biomarkers including CK-MB, CKNAC, and LDH in both the low-dose (200 mg/kg) and high-dose (400 mg/kg) groups of rats. We measured endogenous antioxidants in heart tissue to estimate the antioxidant potential. Histopathological analysis confirmed the biochemical results. Results The findings of this study indicated that the CT leaf extract contained several phytoconstituents such as alkaloids, flavonoids, steroids and triterpenoids, cardiac glycosides, carbohydrates, phenols, saponins, and tannins. The CT's total phenol content was 33.3±0.14 mg GAE (Gallic Acid Equivalent) per gram of extract, while its total flavonoid concentration was 76.5±1.2 mg QUE (Quercetin Equivalent). In the DPPH assay, CT had a mean inhibitory effect of 60.06±1.358 g/ml, whereas its IC 50 in the H 2 O 2 radical scavenging activity assay was 102.85±0.194 g/ml. The electrocardiograms of the treated animals demonstrated considerable recovery from ISO-induced electrophysiological abnormalities as well and the amount of endogenous biomarkers was significantly reduced in animals that received CT treatment. Additionally, the use of CT reversed the effects of ISO administration on SOD activity, increased GSH levels, and lowered LPO levels. The damage to the heart tissues was significantly reduced, with the tissue architecture appearing closer to normal in groups that received high doses of CT. Conclusion These results suggest that C. tomentosa leaf extract has potent antioxidant activity and provides protection against isoproterenol-induced cardiac necrosis in Wistar albino rats. The cardioprotective benefits are attributed to the presence of active components in the extract, which aid in the restoration of antioxidant equilibrium and the reduction of damage caused by oxidative stress.

Indole-3-carbinol ameliorated the isoproterenol-induced myocardial infarction via multimodal mechanisms in Wistar rats

Abstract:The present study investigated the cardioprotection of Indole-3-carbinol on isoproterenol (ISO)-induced myocardial infarction in Wistar rats. I3C treatment significantly reduced the prolongation of the QRS complex, QT interval, and ST-segment elevation. I3C was also able to normalise blood pressure (SBP, DBP, and MAP) and HR. I3C significantly decreased heart weight, cardiac troponin I (cTn I), CK-MB, LDH, AST and ALT. I3C ameliorated acute hyperglycaemia, hyperlipidemia, and myocardial infarction (%) in ISO rats. I3C treatment significantly elevated the antioxidant enzymes like SOD, CAT, and GSH and attenuated the MDA levels. I3C reduced the inflammatory cytokines (TNF-α and IL-6) and increased the anti-inflammatory cytokine 1 L-10. Furthermore, I3C significantly recovered myocardial structure by inhibiting neutrophil infiltration and oedema. Moreover, I3C attenuated apoptotic markers (cytochrome C, caspase 9 and caspase 3). Consequently, I3C restored cardiac function in MI rats by alleviating oxidative stress, inflammation, and apoptosis, and I3C could be used to treat myocardial infarction.
Cardioprotective effects of p‐coumaric acid on tachycardia, inflammation, ion pump dysfunction, and electrolyte imbalance in isoproterenol‐induced experimental myocardial infarction

Abstract:Abstract Cardiovascular diseases cause a large number of deaths throughout the world. No research was conducted earlier on p‐coumaric acid's effect on tachycardia, inflammation, ion pump dysfunction, and electrolyte imbalance. Hence, we appraised the above‐said parameters in isoproterenol‐induced myocardial infarcted rats. This investigation included 24 male albino Wistar rats in 4 groups. Normal control Group 1, p‐coumaric acid (8 mg/kg body weight) alone treated Group 2, Isoproterenol (100 mg/kg body weight) induced myocardial infarcted Group 3, p‐coumaric acid (8 mg/kg body weight) pretreated isoproterenol (100 mg/kg body weight) induced Group 4. After 1 day of the last dose of isoproterenol injection (day 10), rats were killed and blood and heart were taken and inflammatory markers, lipid peroxidation, nonenzymatic antioxidants, ion pumps, and electrolytes were measured. The heart rate, serum cardiac troponin‐T, serum/plasma inflammatory markers, and heart proinflammatory cytokines were raised in isoproterenol‐induced rats. Isoproterenol also enhanced plasma lipid peroxidation, lessened plasma nonenzymatic antioxidants, and altered heart ion pumps and serum and heart electrolytes. In this study, p‐coumaric acid pretreatment orally for 7 days to isoproterenol‐induced myocardial infarcted rats prevented changes in the above‐cited parameters. p‐Coumaric acid's anti‐tachycardial, anti‐inflammatory, anti‐ion pump dysfunction and anti‐electrolyte imbalance properties are the mechanisms for these cardioprotective effects.

Abstract:Twelve bisdesmosidic cholestane glycosides (1-12), including nine new ones (1-9), were isolated from the bulbs of Ornithogalum thyrsoides by monitoring the cytotoxic activity on HL-60 leukemia cells. The structural assignment of the new compounds was carried out by spectroscopic analysis and the results of hydrolytic cleavage. The 3-O-monoglucosides with an aromatic acyl group at the C-16 diglycoside moiety (1, 12) were extremely cytotoxic, with respective IC(50) values of 0.00016 and 0.00013 microg/mL, and the other compounds, except for 2, 5, and 8, also showed cytotoxic activity as potent as etoposide (IC(50) 0.30 microg/mL), used as a positive control. These cholestanes were concluded to contribute to the potent cytotoxicity of the crude O. thyrsoides bulb extract.

Abstract:Objective:Acute respiratory distress syndrome and acute lung injury are characterized by heterogeneous flooding/collapse of lung tissue. An emerging concept for managing these diseases is to set mechanical ventilation so as to minimize the impact of disease heterogeneity on lung mechanical stress and ventilation distribution. The goal of this study was to determine whether changes in lung mechanical heterogeneity with increasing positive end-expiratory pressure in an animal model of acute lung injury could be detected from the frequency responses of resistance and elastance. Design:Prospective, experimental study. Setting:Research laboratory at a veterinary hospital. Subjects:Female sheep weighing 48 ± 2 kg. Interventions:In five saline-lavaged sheep, we acquired whole-lung computed tomography scans, oxygenation, static elastance, and dynamic respiratory resistance and elastance at end-expiratory pressure levels of 7.5–20 cm H2O. Measurements and Main Results:As end-expiratory pressure increased, computed tomography-determined alveolar recruitment significantly increased but was accompanied by significant alveolar overdistension at 20 cm H2O. An optimal range of end-expiratory pressures (15–17.5 cm H2O) was identified where alveolar recruitment was significantly increased without significant overdistension. This range corresponded to the end-expiratory pressure levels that maximized oxygenation, minimized peak-to-peak ventilation pressures, and minimized indexes reflective of the mechanical heterogeneity (e.g., frequency dependence of respiratory resistance and low-frequency elastance). Static elastance did not demonstrate any significant pressure dependence or reveal an optimal end-expiratory pressure level. Conclusions:We conclude that dynamic mechanics are more sensitive than static mechanics in the assessment of the functional trade-off of recruitment relative to overdistension in a sheep model of lung injury. We anticipate that monitoring of dynamic respiratory resistance and elastance ventilator settings can be used to optimize ventilator management in acute lung injury.
Investigation of the cardioprotective effects of Momordica charantia in the isoproterenol-induced myocardial infarction model in rats

Abstract:Background: Cardiovascular diseases are the most prevalent and primary cause of death globally, and the most deadly and dangerous of these diseases is myocardial infarction (MI), commonly known as heart attack, which develops due to insufficient coronary artery flow and causes irreversible myocardial cell damage. This study aimed to investigate the cardioprotective effects of Momordica charantia (MC), known for its antioxidant and anti-inflammatory properties, in an experimental acute MI model induced by isoprenaline (ISO) in rats. Methods: In the study, forty-nine male Wistar rats were split up into 7 groups as control (CONT), Glycerin (GLCN), isoprenaline (ISO), 500 mg/kg MC (MC500), isoprenaline+100 mg/kg MC (ISO+MC100), isoprenaline+250 mg/kg MC (ISO+MC250), isoprenaline+500 mg/kg MC (ISO+MC500). Substances were administered to the groups for 30 days. Isoprenaline (85 mg/kg) was administered by subcutaneous injection on the last two days of the study (days of the 29 and 30). Electrocardiogram (ECG) recording and collecting blood samples of the animals were performed 24 hours after the last administration of the substances under the anesthesia. Serum IL-6, Nrf2, IL-10, HO-1, TNF-α, CK-MB, cTn-I and CRP levels were determined by the ELISA method. Results: Compared to the ISO group, levels of CK-MB, HO-1, TNF-α, CRP, IL-6 and cTn-I were found statistically lower in MC-administered groups (p<0.05). In addition, MC restored ISO-induced abnormal ECG changes to normal levels. Conclusion: In conclusion, ECG findings, proinflammatory, anti-inflammatory, antioxidative and cardiac biomarkers suggest that MC may have cardioprotective properties.
NOVEL ITACONATE DERIVATIVE ATTENUATES ISOPROTERENOL INDUCED CARDIOTOXICITY IN EXPERIMENTAL ANIMALS: A MOLECULAR PATHWAY INSIGHT

Abstract:Objective: 4-octyl itaconate (4-OI) is a potential drug for the treatment of myocardial infarction on account of its potent anti-inflammatory, antioxidant and anti-apoptotic properties. Thus, we hypothesise that 4-OI may attenuate isoproterenol(ISO) induced cardiotoxicity by modulating several molecular pathways in Wistar albino rats. Design and method: After getting approval from animal ethics committee(278/IAEC/2021), the experimental work was started. ISO 5mg/kg s.c., was administered daily for 15 days. Adult male rats were randomly divided into 5 groups (n=6): normal control group, ISO control group, treatment control groups. 4-OI was administered i.p. at 3 different doses (2.5,5,10 mg/kg) to rats from day 1 and continued for 15 days along with ISO. On the 16th day, surgery was performed, right carotid artery was cannulated for recording of hemodynamic parameters. Blood was drawn using cardiac puncture and rats were then sacrificed, one portion of the heart was immediately stored in 10% formalin for histopathological evaluation and second portion was kept in -80 degree C for further studies. Serum was stored for biochemical, pro-inflammatory cytokines estimation (TNF, IL-6, IL1 beta) and cardiac injury markers (CKMB, LDH). Results: 4-OI improved cardiac function and attenuated pathological changes as seen microscopically in the H&E stained heart tissue sections. On ultrastructural evaluation, 4-OI treatment (5, 10mg/kg) groups presented with lesser nuclear condensation and mitochondrial swelling, as compared to the ISO control group. A significant decline in the serum and tissue levels of cardiac injury markers (CK-MB, LDH) and inflammatory markers were observed in 4-OI (5,10mg/kg) treated rats, as compared to ISO control group. Additionally, 4-OI suppressed oxidative stress and apoptosis, along with an increased PPAR-gamma expression. Molecular pathway assessment revealed that at the dose of 10mg/kg, 4-OI reduced the expression of NFkappaB, IKK-beta and enhanced the expression of Nrf-2 and HO-1. On analysis of the inflammatory MAPK pathway, expression of the pro-survival kinase ERK was also increased while JNK and p38 protein expressions were reduced. Conclusions: 4-OI at a dose of 10mg/kg mitigates myocardial toxicity in ISO treated rats by maintaining morphological structure of heart tissue with oxidative stress profile and reduced inflammation

Abstract:Abstract Despite considerable advances in interventions and treatment, there is a high mortality rate in patients with myocardial infarction (MI). This is the first study to investigate the protective effects of 3, 4‐dihydroxybenzoic acid against isoproterenol induced MI in rats. MI was induced by isoproterenol (100‐mg/kg body weight) in rats. Then, rats were treated with 3, 4‐dihydroxybenzoic acid (16‐mg/kg body weight) for 2 weeks. Serum creatine kinase‐MB, cardiac troponin‐T, cardiac troponin‐I, and heart thiobarbituric acid reactive substances were significantly ( p < 0.05) increased and heart superoxide dismutase and catalase activities were significantly ( p < 0.05) reduced in isoproterenol‐induced myocardial infarcted rats. Isoproterenol induction significantly ( p < 0.05) elevated the plasma homocysteine and serum high sensitivity‐C‐reactive protein levels. Furthermore, an enzyme‐linked immunosorbent assay, reverse transcription polymerase chain study, and immunohistochemical (IHC) staining revealed significantly ( p < 0.05) elevated levels and expression of serum/myocardial nuclear factor‐κB, tumor necrosis factor‐alpha, interleukin‐1 beta, and Interleukin‐6 and significantly ( p < 0.05) reduced levels/expression of serum/myocardial interleukin‐10 in myocardial infarcted rats. Nevertheless, isoproterenol‐induced rats treated with 3, 4‐dihydroxybenzoic acid considerably ( p < 0.05) attenuated all the biochemical, molecular, and IHC parameters investigated and inhibited oxidative stress and inflammation and protected the heart, through its antioxidant and anti‐inflammatory mechanisms.

Abstract:Aims Cardiac hypertrophy, an adaptive response of the heart to stress overload, is closely associated with heart failure and sudden cardiac death. This study aimed to investigate the therapeutic effects of chlorogenic acid (CGA) on cardiac hypertrophy and elucidate the underlying mechanisms. Methods and results To simulate cardiac hypertrophy, myocardial cells were exposed to isoproterenol (ISO, 10 μM). A rat model of ISO‐induced cardiac hypertrophy was also established. The expression levels of cardiac hypertrophy markers, endoplasmic reticulum stress (ERS) markers, and apoptosis markers were measured using quantitative reverse transcription PCR and western blotting. The apoptosis level, size of myocardial cells, and heart tissue pathological changes were determined by terminal deoxynucleotidyl transferase dUTP nick‐end labelling staining, immunofluorescence staining, haematoxylin and eosin staining, and Masson's staining. We found that CGA treatment decreased the size of ISO‐treated H9c2 cells. Moreover, CGA inhibited ISO‐induced up‐regulation of cardiac hypertrophy markers (atrial natriuretic peptide, brain natriuretic peptide, and β‐myosin heavy chain), ERS markers (C/EBP homologous protein, glucose regulatory protein 78, and protein kinase R‐like endoplasmic reticulum kinase), and apoptosis markers (bax and cleaved caspase‐12/9/3) but increased the expression of anti‐apoptosis marker bcl‐2 in a dose‐dependent way (0, 10, 50, and 100 μM). Ghrelin Protects Myocardium from Isoproterenol-Induced Injury in Rats

Abstract:AIM: To investigate the cardiac protective effects of ghrelin in rat with myocardial injury induced by isoproterenol (ISO). METHODS: Rats were subcutaneously injected ISO 40 mg(.)kg(-1.)d(-1) with or without ghrelin I or 10 nmol(.)kg(-1.)d(-1) for 2 d. Hemodynamic parameters including mean arterial blood pressure and left ventricular pressure were measured at 12 h after the last injection with ISO and/or ghrelin. Plasma lactate dehydrogenase (LDH) activity, plasma and myocardial contents of malondialdehyde (MDA), and conjugated diene were measured. Plasma ghrelin and endothelin-1 levels were assayed using radioimmunoassay methods. Endothelin-1 and ghrelin mRNA were determined using RT-PCR. RESULTS: About 45% (5111) of rats after treatment with ISO alone died during experimental periods. However, no rats died after administration with ghrelin 10 nmol(.)kg(-1.)d(-1)(0/11, P<0.05). Ghrelin also obviously ameliorated the hemodynamic disturbance in rats induced by ISO. The plasma LDH activity, contents of myocardial and plasma MDA, and conjugated diene level in plasma in ISO+G10 nmol(.)kg(-1.)d(-1) group were decreased by 28%, 34%, 73%, and 38% compared with those of ISO group (all P<0.01) respectively. ISO-induced endothelin-1 mRNA over-expression was inhibited and endothelin- 1 level in plasma were inhibited by ghrelin I and 10 nmol(.)kg(-1.)d(-1). The ghrelin levels in plasma and ghrelin mRNA in myocardium were increased in the rats after injection of ISO. The plasma ghrelin level was further increased after ghrelin administration. CONCLUSION: Ghrelin has a protective effect against ISO-induced myocardial injury.

Abstract:Objective: Cardiac hypertrophy is associated with the enlargement of cardiomyocytes in response to the persistent release of catecholamine, leading to cardiac fibrosis. Chrysin is a natural flavonoid, present in plants, honey, and propolis, with multifarious properties like antioxidant, anti-inflammatory, anti-fibrotic, and antiapoptotic. This study aims to determine the cardioprotective potential of chrysin against isoproterenol (ISO)-induced cardiac hypertrophy and fibrosis in rats Design and method: The male Wistar rats were divided into seven groups (n=6): normal control (carboxymethyl cellulose at 0.5% p.o; normal saline), experimental control (ISO 3mg/kg; s.c), Treatment control (Chrysin 15 mg/kg,30 mg/kg and 60 mg/kg + ISO at 3mg/kg; s.c respectively), perse control (chrysin 60mg/kg) and standard control (losartan 10mg/kg p.o+ ISO at 3mg/kg; s.c) treated for 28 days. On the 29th day, rats underwent surgery and hemodynamic parameters were recorded after sacrificed blood and heart were excised for biochemical, histological, ultrastructural, and molecular evaluations. Results: Our findings show that ISO administration significantly increases heart weight: body weight ratio, pro-oxidants, inflammatory and cardiac injury markers. In addition, histopathological, ultrastructural, and molecular studies showed deteriorative changes. While, pre-treatment with chrysin 60mg/kg ameliorated ISO-induced myocardium damage and protected cardiac hypertrophy and fibrosis through anti-inflammatory, anti-apoptotic, anti-antioxidant, and anti-fibrotic pathways Conclusions: Data suggest that chrysin mitigates myocardial hypertrophy and prevents fibrosis via activation of the transforming growth factor-beta /Smad signaling pathway

Abstract:Abstract INFORMATION is limited on helminth parasites infecting chickens and turkeys in Kansas Ackert (1927) reported a 66% incidence of Heterakis gallinarum in mature chickens. Ferry (1934) reported the tapeworms Raillietina cesticillus, Hymenolepis carioca, R. tetragona, R. echinobothrida Amoebotaenia sphenoides, and Choanotaenia infundibulum from a survey of 50 chickens in Douglas county. Morehouse (1944) reported Capillaria caudinflata from chickens in Brown and Miami counties. Ackert (1956) reported that in an earlier survey of 1000 chickens from the vicinity of Manhattan 49% were infected with Ascaridia galli. Kerr (1958) reported Ascaridia dissimilis from turkeys. Surveys of poultry parasites have been made in the United States by Guberlet (1916, Nebraska), Adams et al. (1933, Texas), Wehr (1937, Washington, D. C.), Koutz (1949, Ohio), Todd (1948, Tennessee), Edgar (1953, Alabama), and Reid (1958, Georgia). Cardioprotective Effect of Gossypin Against Myocardial Ischemic/Reperfusion in Rats via Alteration of Oxidative Stress, Inflammation and Gut Microbiota

Abstract:Background: Myocardial ischemic/reperfusion (I/R) injury is a key prognostic factor after the myocardial infarction. However, at the time of reperfusion, the myocardial tissue has undergone for the necrosis and initiated the induction of oxidative stress and inflammation. The current study was to scrutinize the cardioprotective effect of gossypin against ISO-induced I/R injury in myocardial tissue and explore the possible underlying mechanism.Methods: Sprague Dawley (SD) was used in the current protocol and ISO was used for induction the I/R in rat. The rats were divided into different groups and received the oral administration of gossypin treatment before the reperfusion. The body weight, heart weight and heart body weight ratio were estimated. The antioxidant, cardiac injury parameters, inflammatory cytokines, inflammatory mediators, gut microbiota and lipid parameters were estimated. At the end, heart tissue histopathological study was carried out.Results: ISO-induced I/R rats received the gossypin treatment significantly (P < 0.001) enhanced the body weight and decreased the heart weight, along with suppressed the infarct size. Gossypin treatment significantly (P < 0.001) reduced the level of heart parameters, such as creatinine kinase-MB (CK-MB), lactate dehydrogenase (LDH), creatine kinase (CK), cardiac troponin I (CTn-I) and cardiac troponin T (CTn-T) in the serum. Gossypin treatment significantly (P < 0.001) altered the cardiac function, hepatic, antioxidant, inflammatory cytokines and inflammatory mediators. Gossypin significantly (P < 0.001) suppressed the MMP-2 and MMP-9 in ISO-induced I/R rats. Gossypin treatment considerably alleviated the gut dysbiosis through altered Firmicutes to Bacteroidetes (F/B) ratio and also maintained the relative abundance of Butyricicoccus, Clostridium IV, Akkermansia, Roseburia and Clostridium XIVs.Conclusion: Based on result, we can conclude that gossypin is an alternative drug for the treatment of ISO-induced I/R in rats via alteration of oxidative stress, inflammatory reaction and gut microbiota.

Abstract:Myocardial infarction (MI) is a common type of ischemic heart disease that affects millions of people worldwide. In recent times, nanotechnology has become a very promising field with immense applications. The current exploration was conducted to synthesize the chitosan‐sodium alginate‐polyethylene glycol‐Ally isothiocyanate nanocomposites (CSP‐AIso‐NCs) and evaluate their beneficial roles against the isoproterenol (ISO)‐induced MI in rats. The CSP‐AIso‐NCs were prepared and characterized by several characterization techniques. The MI was initiated in the rats by the administration of 85 mg/kg of ISO for 2 days and treated with 10 and 20 mg/kg of CSP‐AIso‐NCs for 1 month. The changes in heart weight and bodyweight were measured. The cardiac function markers were assessed with echocardiography. The lipid profiles, Na+, K+, and Ca2+ ions, cardiac biomarkers, antioxidant parameters, and inflammatory cytokines were assessed using corresponding assay kits. The histopathological study was done on the heart tissues. The UV spectral analysis revealed the maximum peak at 208 nm, which confirms the formation of CSP‐AIso‐NCs. The FT‐IR analysis revealed the occurrence of different functional groups, and the crystallinity of the CSP‐AIso‐NCs was proved by the XRD analysis. DLS analysis indicated the size of the CSP‐AIso‐NCs at 146.50 nm. The CSP‐AIso‐NCs treatment increased the bodyweight and decreased the HW/BW ratio in the MI rats. The status of lipids was reduced, and HDL was elevated in the CSP‐AIso‐NCs administered to MI rats. CSP‐AIso‐NCs decreased the LVEDs, LVEDd, and NT‐proBNP and increased the LVEF level. The oxidative stress markers were decreased, and the antioxidants were increased by the CSP‐AIso‐NCs treatment in the MI rats. The Na+ and Ca+ ions were reduced, and the K+ ions were increased by the CSP‐AIso‐NCs. The interleukin‐1β and tumor necrosis factor‐α were also depleted, and Nrf‐2 was improved in the CSP‐AIso‐NCs administered to MI rats. The histological study revealed the ameliorative effects of CSP‐AIso‐NCs. Overall, our outcomes revealed that the CSP‐AIso‐NCs are effective against the ISO‐induced MI rats. Hence, it could be a hopeful therapeutic nanomedicine for MI treatment.

Combined Effects of Vincristine and Quercetin in Reducing Isoproterenol-Induced Cardiac Necrosis in Rats

Abstract:Combined effects of vincristine and quercetin in the regulation of isoproterenol (ISO)-induced cardiac necrosis have been evaluated in rats. ISO administration (100 mg/kg, s.c., for two consecutive days) increased the levels of serum creatine kinase-MB (CK-MB), lactate dehydrogenase (LDH), glutamate pyruvate transaminase (SGPT) and cardiac troponin (cTnT) as well as cardiac lipid peroxidation products (malondialdehyde and lipid hydroperoxides). However, it reduced the activities of superoxide dismutase (SOD), catalase and the glutathione peroxidase and the level of reduced glutathione. It also increased the heart rate and ST-segment elevation in ECG. Pretreatment of vincristine (25 μg/kg) or quercetin (10 mg/kg) alone for 2 weeks ameliorated these cardiotoxic effects partially. However, treatment of both vincristine and quercetin for a similar period reduced the serum CK-MB, LDH, SGPT and cTnT levels near to normal levels in ISO-treated rats. Concomitantly, the test drugs improved the status of antioxidants and decreased the cardiac lipid peroxidation products. Combined treatment of both the drugs also restored the pathological electrocardiographic patterns and reduced the area of myocardial necrosis. Histopathology of heart in ISO-administered rats that received both vincristine and quercetin showed nearly normal myocardium with very little inflammatory infiltration. In conclusion, the present finding appears to be the first one, suggesting a better protection of cardiac tissues by combined treatment of vincristine and quercetin in isoproterenol-induced cardiac toxicity.
Cardioprotective Effects of Ursodeoxycholic Acid in Isoprenaline-Induced Myocardial Injury in Rats

Abstract:Patients suffering from cholelithiasis have an increased risk of developing cardiovascular complications, particularly ischemic myocardial disease. Ursodeoxycholic acid (UDCA), already used in clinical practice for the treatment of cholelithiasis and related conditions, has proven antioxidative, anti-inflammatory, and cytoprotective effects. Therefore, the aim of this study was to investigate the cardioprotective effect of UDCA pre-treatment on isoprenaline-induced myocardial injury in rats. Male Wistar albino rats were randomized into four groups. Animals were pre-treated for 10 days with propylene glycol + saline on days 9 and 10 (control), 10 days with propylene glycol + isoprenaline on days 9 and 10 (I group), 10 days with UDCA + saline on days 9 and 10 (UDCA group), and 10 days with UDCA + isoprenaline on days 9 and 10 (UDCA + I group). UDCA pre-treatment significantly reduced values of high-sensitivity troponin I (hsTnI) and aspartate aminotransferase (AST) cardiac markers (p < 0.001 and p < 0.01, respectively). The value of thiobarbituric acid reactive substances (TBARS) was also decreased in the UDCA + I group compared to the I group (p < 0.001). UDCA also significantly increased glutathione (GSH) levels, while showing a tendency to increase levels of superoxide dismutase (SOD) and catalase (CAT). The level of nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) expression, a key regulatory gene of inflammation, was diminished when UDCA was administered. A reduction of cardiac damage was also observed in the UDCA pre-treated group. In conclusion, UDCA pre-treatment showed a cardioprotective effect on isoprenaline-induced myocardial injury in rats, primarily by reducing oxidative stress and inflammation.

Ameliorative effect of ferruginol on isoprenaline hydrochloride-induced myocardial infarction in rats

Abstract:Cardiovascular-related diseases continue to be a leading cause of death globally. Among ischemic-induced cardiac diseases, myocardial infarction (MI) is reported to be of an alarming value. Despite numerous improvements in the medical intrusions, still this armamentarium fails to be effective in managing the illness without setbacks. Ferruginol (FGL) is a major polyphenols and terpenoids with numerous pharmacological activities including antioxidant and anti-inflammatory. Following, this work was aimed to explore the cardio protective effect of FGL (50 mg/kg) in isoprenaline hydrochloride (ISO)-induced MI in experimental rats. After treatment with FGL in ISO-induced MI in rats, noticeable changes were observed in the experimental rats. Injection of ISO to rats resulted in the augmented cardiac weight, serum cardiac markers (creatine kinase, creatine kinase-MB, cardiac troponin T, and Cardiac troponin I), lipid peroxidation end products (thiobarbituric acid-reactive substance and lipid hydroperoxides), reduced endogenous antioxidants (superoxide dismutase, catalase, glutathione peroxidase, and glutathione), reduced ATPase activity, and escalated pro-inflammatory cytokines (interleukin-6, tumor necrosis factor-α, and nuclear factor-κB) levels. Interestingly, the FGL supplementation to the ISO-treated rats revealed the diminished heart weight, reduced cardiac markers, and lipid peroxidation. FGL also possessed the improved antioxidants status and diminished pro-inflammatory mediator levels. The outcomes of histological analysis also evidenced the cardio protective role of FGL. Treatment with FGL reduced the cardiac damage biomarkers maintained to near normal levels in ISO-induced rats. These study findings disclose the prospective capability of FGL in the treatment of MI in the future.
