Abstract:The present study on Solid Lipid Nanoparticles containing Asiaticoside aims to study the use of solid lipid nanoparticle-enriched hydrogel for the wound healing application of Asiaticoside. Solid lipid nanoparticles SLNs were prepared using glycerol tristearate and poloxamer by solvent evaporation method. The particle size polydispersity index PDI zeta potential ZP encapsulation efficiency EE drug release from nanoparticles and hydrogels and SEM studies were performed. The prepared SLNs were incorporated in Carboxymethyl chitosan-oxidized alginate hydrogel to form SLN-hydrogel carrier. The hydrogel was evaluated for texture analysis water vapor transmission rate of evaporation of water drug release studies from SLNs and SLN-hydrogel. The best formulation of Asiaticoside based SLNs had high EE 72 plusmn 6 and Zeta potential value of -36.8 mV indicating good stability. Formulation N2 showed polydispersivity index less than 1 and the particle size 155 nm to 340 nm. The prepared asiaticoside SLN-hydrogel had water vapour transmission rate of 2294.9plusmn188.9 gm2 per day which would provide a sufficient level of moisture without risking wound dehydration. The asiaticoside SLN-hydrogel showed sustained release of asiaticoside for 8 h.
Bilayer microneedles based on Bletilla striata polysaccharide containing asiaticoside effectively promote scarless wound healing

Abstract:The repair of wounds is essential for overcoming injuries, but the process often leads to scars, and the formation mechanism is unclear. Interestingly, Bletilla striata polysaccharide (BSP) is a procoagulant and has anti-inflammatory properties to promote wound healing, which has great research value for scarless wound healing. Herein, we developed a transdermal drug delivery system of BSP/chitosan (CS) composite bilayer dissolvable microneedles (CS-AS-BSP MNs) containing asiaticoside (AS) based on a centrifugation method with a polydimethylsiloxane mould that fulfilled the requirements of wound healing and achieved a scar-free effect. The drug release results showed that this system improved the solubility of AS and maintained the stable release of the drug for a long time. Further experiments indicated that CS-AS-BSP MNs had a good antibacterial effect and reduced the proliferation of human scar fibroblasts. The wound healing effect of CS-AS-BSP MNs was evaluated in an experimental full-thickness-wound rat model, which demonstrated that the covered wound healed completely without scars within 3 weeks. Furthermore, CS-AS-BSP MNs downregulated TGF-β1 and COL I, which are fibrosis-related genes, to inhibit scarring. In brief, this study provides a new strategy for meeting the clinical requirements of effective scarless wound dressings.

Chitosan-aluminum monostearate composite sponge dressing containing asiaticoside for wound healing and angiogenesis promotion in chronic wound

Abstract:There are many factors that delay healing in chronic wounds including lowering level of growth factors and increasing exudate level comprising high amount of tissue destructive enzymes. Asiaticoside possesses interesting wound healing and angiogenic activities that are employed to stimulate tissue regeneration in wound healing application. This study attempted to develop chitosan-aluminum monostearate (Alst) composite sponge containing asiaticoside for use as an absorbent medical dressing in chronic wound. N-methyl-2-pyrrolidone (NMP) was used to enhance homogeneity of asiaticoside in the polymer composite matrix. The sponge dressings were prepared by lyophilization and dehydrothermal treatment (DHT). Functional group interaction, crystallinity, and morphology of the prepared sponges were investigated using FT-IR, PXRD, and SEM, respectively. Physicochemical properties, porosity, hydrophilic/hydrophobic properties and mechanical property, were evaluated. Wound dressing properties, water vapor transmission rate (WVTR), fluid absorbency, oxygen permeation (OP), and bio-adhesive property, were investigated. In vitro asiaticoside release study was conducted using immersion method. Cytotoxicity was studied in normal human dermal fibroblast (NHDF) and normal human epidermal keratinocyte (NHEK). Angiogenic activity of asiaticoside was evaluated using chick-chorioallantoic membrane (CAM) assay. FT-IR and PXRD results revealed the amidation after DHT to enhance the crystallinity of the prepared sponges. The prepared sponges had high porosity comprising high Alst-loaded amount that exhibited more compact structure. Alst enhanced hydrophobicity therefore it reduced the fluid absorption and WVTR together with bio-adhesion of the prepared sponge dressings. Porosity of all sponges was more than 85% therefore resulting in their high OP. Enhancing hydrophobicity of the material by Alst and more homogeneity caused by NMP eventually retarded the asiaticoside release for 7 days. The sponge extractions were non-toxic to the cells moreover they promoted NHDF and NHEK cell proliferation. Asiaticoside and asiaticoside-contained dressings exhibited dose-dependent angiogenic activity in CAM model.

Abstract:A recently proposed carbon nanotube (CNT) crossbar nano-architecture provides the first purely CNT-based platform for nanoscale computing systems. In this paper, I present a number of degrees of freedom in optimizing this nano-architecture, and evaluate several variant nanoscale computing platforms based on a combination of floating-gate transistor arrays and programmable vias. Experimental results based on Stanford compact CNFET model show that proper device integration, cell granularity, and logic family (e.g., reconfigurable CMOS static logic in floating-gate transistor arrays) lead to an optimized nanoscale computing platform, with reduced manufacture complexity, improved logic density, improved performance, and reduced power consumption.
Chitosan-Based Hydrogels for Controlled Delivery of Asiaticoside-Rich Centella asiatica Extracts with Wound Healing Potential

Abstract:Centella asiatica extract is a valued plant material with known anti-inflammatory and anti-microbiological properties. Using the Design of Experiment (DoE) approach, it was possible to obtain an optimized water/alcoholic extract from Centella asiatica, which allowed the preparation of the final material with biological activity in the wound healing process. Studies on the novel applications of Centella asiatica in conjunction with the multifunctional chitosan carrier have been motivated by the plant's substantial pharmacological activity and the need to develop new and effective methods for the treatment of chronic wounds. The controlled release of asiaticoside was made possible by the use of chitosan as a carrier. Based on the findings of investigations using the PAMPA skin assay, which is a model imitating the permeability of actives through skin, this compound, characterized by sustained release from the chitosan delivery system, was identified as being well able to permeate biological membranes such as skin. Chitosan and the lyophilized extract of Centella asiatica worked synergistically to block hyaluronidase, exert efficient microbiological activity and take part in the wound healing process, as proven in an in vitro model. A formulation containing 3% extract with 3% medium-molecular-weight chitosan was indicated as a potentially new treatment with high compliance and effectiveness for patients. Optimization of the chitosan-based hydrogel preparation ensured the required rheological properties necessary for the release of the bioactive from the chitosan delivery system and demonstrated a satisfactory antimicrobial activity.

Abstract:It remains a challenge to regulate the complicated microenvironment effectively in the wound healing process. The optimization of synergistic action of angiogenesis and inflammation is considered critical for quicker scarless wound regeneration. Here, silk nanofiber (SNF) as a multifunctional carrier to load hydrophobic asiatisoside (AC) and hydrophilic Mg2+, and also as an element to assemble injectable hydrogel, forming a bioactive matrice with improved angiogenic and anti-inflammatory capacities (SNF-AC-Mg). Mg2+ and AC distributed homogeneously inside silk nanofiber hydrogels without the compromise of mechanical performances. Both Mg2+ and AC released slowly to continuously tune both angiogenic and inflammatory behaviors. The hydrogels exhibited good biocompatibility, inflammation inhibition, and pro-angiogenic properties in vitro, suggesting the synergistic bioactivity of AC and Mg2+. In vivo analysis revealed that the synergistic action of AC and Mg2+ resulted in better M2-type polarization of macrophages and angiogenesis during the inflammatory phase, while achieved effective inhibition of excessive accumulation of collagen and scar formation during the remodeling phases. The quicker scarless regeneration of the defects treated with SNF-AC-Mg implies the priority of SNF in designing bioactive niches with complicated cues, which will favor the functional recovery of different tissues in the future.

Combination wound healing using polymer entangled porous nanoadhesive hybrids with robust ROS scavenging and angiogenesis properties

Abstract:Nanoadhesives can achieve tight wound closure by connecting biomacromolecules from both sides. However, previously developed adhesive systems suffered from suboptimal wound healing efficiency due to the lack of interparticle cohesion, sufficient reactive oxygen species (ROS)-scavenging sites, and angiogenesis consideration. Herein, we developed a polymer entangled porous nanoadhesive system to address the above challenge by synergy of three functional components. Firstly, hybrid mesoporous silica nanoparticles with highly integrated polydopamine (MS-PDA) were prepared by templated synthesis. The entangling between PVA polymer and MS-PDA contributed to much stronger cohesion between nanoparticles, which led to 75% larger adhesion strength. As confirmed by in vitro and in vivo evaluations, the highly exposed catechol groups boosted the scavenging activity of ROS (1.8–4.1 fold enhancement as compared with nonporous counterpart). Consequently, more macrophages exhibited anti-inflammatory phenotype, leading to 2–2.6 fold lower pro-inflammatory cytokine levels. Moreover, the sustained release of bioactive SiO44− by the disintegration of nanoparticles contributed to ∼3-fold higher expression of VEGF and enhanced new blood vessel formation, as well as better wound repair. This platform can provide a new paradigm for developing multifunctional nanoadhesive systems in treating skin wounds.Statement of significancePVA polymer entangled mesoporous nanoadhesives of polydopamine (PDA)/silica hybrids with the combination of excellent wound closure effect, boosted ROS-scavenging activity, and significant angiogenesis ability were developed for improving the suboptimal skin wound healing efficiency. This strategy not only greatly advances our ability to rationally integrate repairing elements in nanoadhesives for manipulating combined processes of interfacial events during wound healing, but also offers general implications toward application of polymers to reinforce the adhesion strength in nanoadhesive systems. In addition, our findings on the impacts of pore effects mediated ROS species conversion and polymer entanglement may also trigger great interests and facilitate the development/broad application of therapeutic adhesives.

Abstract:The process of wound healing is intricate and complex, necessitating the intricate coordination of various cell types and bioactive molecules. Despite significant advances, challenges persist in achieving accelerated healing and minimizing scar formation. Herein, a multifunctional hydrogel engineered via dynamic Schiff base crosslinking between oxidized dextran and quaternized chitosan, reinforced with reduced graphene oxide (rGO) is reported. The resulting OQG hydrogels demonstrated injectability to aid in conforming to irregular wound geometries, rapid self-healing to maintain structural integrity and adhesion for intimate integration with wound beds. Moreover, the developed hydrogels possessed antioxidant and antibacterial activities, mitigating inflammation and preventing infection. The incorporation of conductive rGO further facilitated the transmission of endogenous electrical signals, stimulating cell migration and tissue regeneration. In addition, the polydopamine-encapsulated asiaticoside (AC@PDA) nanoparticles were encapsulated in OQG hydrogels to reduce scar formation during in vivo evaluations. In vitro results confirmed the histocompatibility of the hydrogels to promote cell migration. The recovery of the full-thickness rat wounds revealed that these designed OQG hydrogels with the incorporation of AC@PDA nanoparticles could accelerate wound healing, reduce inflammation, facilitate angiogenesis, and minimize scarring when implemented. This multifunctional hydrogel system offers a promising strategy for enhanced wound management and scarless tissue regeneration, addressing the multifaceted challenges in wound care.

Abstract:Chronic wounds are a major health problem with increasing global prevalence, which endangers the physical and mental health of those affected and is a heavy burden to healthcare providers. Artemisia argyi extract (AE) has excellent antibacterial and anti-inflammatory properties. In this research, we developed AE loaded composite hydrogel scaffold based on methacrylate gelatin (GelMA)/methacrylate hyaluronic acid (HAMA) and mesoporous silica nanoparticle (MSN) as sustained-release drug carrier vehicles for the treatment of chronic wounds. The presented GelMA/1%HAMA hydrogel possessed stable rheological properties, suitable mechanical properties, appropriate biodegradability, swelling, sustained-release AE capacity. In vitro antibacterial and cell experiments showed that the GelMA/HAMA/MSN@AE hydrogel had excellent antibacterial activity and biocompatibility and induced macrophages to differentiate into M2 phenotype. In vivo wound healing of rat full-thickness cutaneous wounds further demonstrated that the prepared GelMA/HAMA/MSN@AE hydrogel could significantly promote chronic wound healing by upregulating the expression of IL-4, TGF-β1, CD31, and α-SMA but downregulating the expression of TNF-α and IFN-γ and promoting M1-M2 macrophages polarization. Altogether, we believe that the GelMA/HAMA/MSN@AE hydrogel will have wide application prospects in healing chronic wounds.

VoteTrust: Leveraging friend invitation graph to defend against social network Sybils

Abstract:Online social networks (OSNs) currently face a significant challenge by the existence and continuous creation of fake user accounts (Sybils), which can undermine the quality of social network service by introducing spam and manipulating online rating. Recently, there has been much excitement in the research community over exploiting social network structure to detect Sybils. However, they rely on the assumption that Sybils form a tight-knit community, which may not hold in real OSNs. In this paper, we present VoteTrust, a Sybil detection system that further leverages user interactions of initiating and accepting links. VoteTrust uses the techniques of trust-based vote assignment and global vote aggregation to evaluate the probability that the user is a Sybil. Using detailed evaluation on real social network (Renren), we show VoteTrust's ability to prevent Sybils gathering victims (e.g., spam audience) by sending a large amount of unsolicited friend requests and befriending many normal users, and demonstrate it can significantly outperform traditional ranking systems (such as TrustRank or BadRank) in Sybil detection.
Natural polysaccharide based complex drug delivery system from microfluidic electrospray for wound healing

Abstract:Natural polysaccharides have a demonstrated value in drug encapsulation and delivery. Effort s in this area are focused on improving the performances of these biomaterials. In this study, we present a multicompartment polysaccharide core-shell microparticle to construct a sustainable dual-release system of active molecules for enhancing wound healing. These microparticles were generated utilizing a microfluidic electrospray approach, in which a mixture of cellulose nanocrystals (CNC) and vascular endothelial growth factor (VEGF) were employed as the cores, with doxycycline hydrochloride (DH) mixed alginate as the shells. The resultant multicompartment particles exhibited wonderful antibacterial properties inherited from the alginate shell, and angiogenesis properties from the CNC core after the shell disintegrated. It was demonstrated that these core-shell microparticles performed their outstanding capabilities in inhibiting inflammation, promoting granulation tissue formation, collagen deposition, and angiogenesis, thereby accelerating rodents' wound healing. These findings support the great potential value of CNC/alginate core-shell microparticles based on the synergistic drug delivery strategy for orchestrating wound healing. (c) 2021 Elsevier Ltd. Topical astragaloside IV-releasing hydrogel improves healing of skin wounds in vivo.

Abstract:Topical delivery of therapeutic agents at the time of injury to accelerate skin repair and prevent the formation of scars during the wound healing process has received increasing attention and represents a novel regenerative and prophylactic strategy for wound treatment. The aim of this study was to invesigate, for the first time, the influence of topical astragaloside IV-releasing hydrogel on the wound repair and regeneration. Using the sodium alginate-gelatin as a hydrogel vehicle, the astragaloside IV was incorporated into the topical carrier and kept releasing with a sustained manner at the wound site. With the rat skin excision model, regulation of the astragaloside IV hydrogel on the wound repair and regeneration were investigated. It was found that the astragaloside IV hydrogel was effective in the skin wound repair, leading to a significant improvement on the wound closure, collagen synthesis and skin tensile strength recovery. Meanwhile, for the first time, that functions of astragaloside IV hydrogel in activating the skin appendages regeneration and increasing the transforming growth factor-beta (TGF-beta(1)) level in serum were shown. Results of this study provided evidence for the alginate-gelatin hydrogel as efficient carrier for the topical delivery of bioactive molecules to the injured site. The astragaloside IV releasing hydrogel was shown a promising therapeutic formulation for wound healing, as well as its regenerative feature and underlying mechanism contribute to the skin regeneration were disclaimed.
Catechol functionalized chitosan/active peptide microsphere hydrogel for skin wound healing

Abstract:Chitosan-based thermosensitive hydrogels have been widely used in drug delivery and tissue engineering, but their poor bioactivity has limited their further applications. Integral active oyster peptide microspheres (OPM) with an average particle diameter of 3.9 μm were prepared with high encapsulation efficiency (72.8%) and loading capacity (11.9%), exhibiting desirable sustained release effects. Using catechol functionalized chitosan (CS-C) as the polymeric matrix, OPM as the filler, and β-sodium glycerophosphate (β-GP) as a thermal sensitizer, the thermosensitive hydrogel CS-C/OPM/β-GP was prepared. Besides, the application of the hydrogel on wound healing was studied, and its biosafety was evaluated. The results of cell migration in vitro showed that the cell migration rate of CS-C/OPM/β-GP reached 97.47 ± 5.41% within 48 h, indicating that the hydrogel accelerated the migration of L929 cells. As demonstrated in the mouse skin wound experiment, CS-C/OPM/β-GP hydrogel not only inhibited the aggregation of diversified inflammatory cells and accelerated the generation of collagen fibers and new blood vessels of the wound, but also enhanced the synthesis of total protein (TP) in granulation tissue, and up-regulated the expression of Ki-67 and VEGF in the injury, thereby achieving fast wound healing. Safety evaluation results showed that CS-C/OPM/β-GP hydrogel was not cytotoxic to L929 cells, and the hemolysis ratio was less than 5% within 1 mg/mL. In conclusion, CS-C/OPM/β-GP hydrogel is expected as a promising medical dressing for wound healing.

Lung Inflammation Associated With Clinical Acute Necrotizing Pancreatitis in Dogs

Abstract:Although dogs with acute necrotizing pancreatitis (ANP) can develop respiratory complications, there are no data describing lung injury in clinical cases of ANP in dogs. Therefore, we conducted a study to characterize lung injury and determine if pulmonary intravascular macrophages (PIMs) are induced in dogs with ANP (n = 21) compared with control dogs (n = 6). Two pathologists independently graded histologic sections of pancreas from clinical cases to characterize the severity of ANP (total scores of 3–10) compared with controls showing histologically normal pancreas (total scores of 0). Based on histological grading, lungs from dogs with ANP showed inflammation (median score, 1.5; range, 0–3), but the scores did not differ statistically from the control lungs (median score, 0.5; range, 0–2). A grid intersects-counting method showed an increase in the numbers of MAC387-positive alveolar septal mononuclear phagocyte profiles in lungs of dogs with ANP (ratio median, 0.0243; range, 0.0093–0.0734, with 2 outliers at 0.1523 and 0.1978) compared with controls (ratio median, 0.0019; range, 0.0017–0.0031; P < .0001). Only dogs with ANP showed labeling for von Willebrand factor in alveolar septal capillary endothelial cells, septal inflammatory cells, and alveolar macrophages. Toll-like receptor 4 and interleukin 6 were variably expressed in alveolar macrophages and septal inflammatory cells in lungs from both ANP and control dogs. Inducible nitric oxide synthase was detected in alveolar macrophages of dogs with ANP only. These data show that dogs with ANP have lung inflammation, including the recruitment of PIMs and expression of inflammatory mediators.
Advanced formulation and pharmacological activity of hydrogel of the titrated extract of C. asiatica

Abstract:Titrated extract of Centella asiatica (TECA) contains three principal ingredients, asiaticoside (AS), asiatic acid (AA), and madecassic acid (MA). These components are known to be clinically effective on systemic scleroderma, abnormal scar formation, and keloids. However, one problem associated with administration of TECA is its low solubility in aqueous as well as oil medium. In this study, various nonionic surfactants and bile salts as anionic surfactant were tested and screened for solubilizing TECA with a view to developing topical hydrogel type of ointment which is stable physicochemically, and has better pharmacological effects. When TECA was incorporated into various nonionic surfactant systems, labrasol had the most potent capacity for solubilizing TECA. In cases of bile salt systems, Na-deoxycholate (Na-DOC) had foremost solubilizing capacity, even more than labrasol. In differential scanning calorimetric study, the peaks of AA, MA, AS and Na-DOC disappeared at the coprecipitate of 1% TECA and 1% Na-DOC, suggesting the optimum condition of Na-DOC for solubilizing TECA. When the physicochemical stability of hydrogel containing this mixture was assessed, it was stable at room temperature for at least one month. Pharmacologically it significantly decreased the size of wound area at the 9th day when applied to the wound area of rat dorsal skin. Taken together, solubility of TECA was dramatically improved by using nonionic and anionic surfactant systems, and Na-DOC was found to be the most effective solubilizer of TECA in formulating a TECA-containing hydrogel typed ointment. Moreover this gel was considered to be applicable to clinical use for wound healing effect.
Citrus flavonoids-loaded chitosan derivatives-route nanofilm as drug delivery systems for cutaneous wound healing

Abstract:This study focuses on creating new forms of biomimetic nanofiber composites by combining copolymerizing and electrospinning approaches in the field of nanomedicine. The process involved utilizing the melt polymerization of proline (Pr) and hydroxyl proline (Hyp) to synthesize polymers based on Pr (PPE) and Hyp (PHPE). These polymers were then used in a grafting copolymerization process with chitosan (CS) to produce PHPC (1560 ± 81.08 KDa). A novel electrospun nanofiber scaffold was then produced using PHPC and/or CS, hyaluronic acid, polyvinyl alcohol, and naringenin (NR) as a loading drug. Finally, Mouse Dermal Fibroblast (MDF) cells were introduced to the wound dressing and assessed their therapeutic potential for wound healing in rats. The scaffolds were characterized by FTIR, NMR, DSC, and SEM analysis, which confirmed the amino acid grafting, loading drug, and porous and nanofibrous structures (>225 nm). The results showed that the PHPC-based scaffolds were more effective for swelling/absorption of wound secretions, had more elasticity/elongation, faster drug release, more MDF-cytocompatibility, and antibacterial activity against multidrug-resistant S. aureus compared to CS-based scaffolds. The in vivo studies showed that NR in combination with MDF can accelerate cell migration/proliferation, and remodeling phases of wound healing in both PHPC/CS-based scaffolds. Moreover, PHPC-based scaffolds promote collagen content, and better wound contraction, epithelialization, and neovascularization than CS-based, showing potential as wound-dressing.

Abstract:A 51-year old man presented with vertical and torsional diplopia after reduction of a blowout fracture at another hospital one year ago. He had no anormalies of head position and 14 prism diopters (PD) right hypertropia (RHT) in the primary position. In upgaze no vertical deviation was found, and hyperdeviation on downgaze was 35PD. Bielschowsky head tilt test showed a negative response. Distinct superior oblique (SO) and inferior rectus (IR) underaction of the right eye was noted but IO overaction was mild on the ocular version test. Double Maddox rod test (DMRT) revealed 10-degree extorsion, but fundus extorsion was minimal in the right eye.Thin-section coronal CT scan showed that there was no fracture line on the anterior orbital floor, but a fracture remained on the posterior orbital floor. Also, the anterior part of the right inferior oblique muscle was vertically reoriented and the medial portion of the inferior oblique muscle was not traced on the coronal CT scan. The patient underwent 14 mm right IO recession and 3 mm right IR resection. One month after the surgery, his vertical and torsional diplopia were eliminated in the primary position.
Core-shell chitosan microsphere with antimicrobial and vascularized functions for promoting skin wound healing

Abstract:It is imperative to develop a wound dressing that can effectively treat bacterial infection in the wound location and speed up wound healing. In this study, chitosan (CS) microspheres with a core-shell structure were prepared using high-voltage electrostatic drop technology. The antimicrobial polypeptide (PonG1) and bFGF, were encapsulated in the core of the CS microsphere. The preparation of CS microspheres was studied by changing the parameters. The surface morphology of CS microspheres and the release quantities of PonG1 and bFGF were observed and detected. In vivo experiments were conducted to study the neovascularization and anti-inflammatory abilities of bFGF and PonG1. The repair ability of CS microspheres was evaluated using a rat model of full-thickness skin wound. Both bFGF and PonG1 exhibited slow release from the CS microspheres within 14 days. In vivo experiments showed that CS microspheres containing PonG1 (3 mg/ml) and bFGF (300 μg/ml) had the best neovascularization and anti-inflammatory effects. CS microspheres containing both bFGF and PonG1 had the strongest ability to repair skin wound. CS microspheres with antimicrobial and vascularized functions can be used as dressings for the repair of hard-to-heal wounds.
