Abstract:Lignocellulolytic enzymes are extracellular biocatalysts secreted by filamentous fungi and are involved in the breakdown of recalcitrant lignocellulose plant materials into useful products necessary for fungal growth and development. Even though several studies on filamentous fungi have reported the impact of different substrates on lignocellulolytic enzymes, there is limited information on how mushroom supplements affect secretion of the enzymes, growth, and yield of Pleurotus ostreatus when using the alkaline treatment method. In this study, we investigated the influence of cornmeal (T1) and coffee grounds (T2) supplements on lignocellulosic enzymes at different growth stages and the production of P. ostreatus. We found that lignolytic enzyme activity was significantly higher in the control (CK) and T2 during mycelial stages, while CK had the lowest hydrolytic enzyme activity during primordia and fruiting. Unlike T1 which had the best biological efficiency, T2 exhibited significantly higher levels of lignolytic enzymes during mycelial stage, whereas CMCase and xylanase activities were higher in supplemented treatments than in the control during primordia and fruiting. Taken together our results demonstrated that cornmeal and coffee ground supplements reduce mycelium growth rate, enhance the production of hydrolytic enzymes during fruiting, and remarkably increase the yield and protein content of P. ostreatus.
Label-free comparative proteomic analysis of Pleurotus eryngii grown on sawdust, bagasse, and peanut shell substrates

Abstract:The white rot fungi Pleurotus eryngii are environmental microorganisms that can effectively break down lignocellulosic biomass. However, understanding of the mechanisms by which P. eryngii is effective in degrading lignocellulose is still limited. This work aimed to examine the extracellular secretory proteins implicated in the breakdown of lignocellulose in P. eryngii and identify degradation tactics across various cultivation substrates. Thus, a comparative analysis of the secretory proteins based on Nanoliquid chromatography combined with tandem mass spectrometry was conducted among P. eryngii cultivated on sawdusts, bagasse, peanut shells, and glucose. In total, 647, 616, 604, and 511 proteins were identified from the four samples, respectively. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway analysis of protein expression differences identified pathways (hydrolytic enzymes, catalytic activity, metabolic processes, cellular processes, and response to stimuli) significantly enriched in proteins associated with lignocellulose degradation in P. eryngii. An integrated analysis of proteome data revealed specifically or differentially expressed genes secreted by P. eryngii in different cultivation substrates. The most prevalent carbohydrate-active enzymes involved in lignocellulose degradation in the secretome of the four samples were laccase (Lac), manganese peroxidase (MnP), aryl alcohol oxidase (AaO), and copper radical oxidase (CRO). Among them, Lac 2 mainly involved in the lignin degradation of sawdust peanut shells, and bagasse by P. eryngii, and Mnp 3 was mainly involved in the degradation of peanut shells. AaO and Lac 4 were mainly involved in glucose substrate defense and oxidative stress. It was found that exogenous addition of sawdust and peanut shells significantly increased lignolytic enzyme abundance. These findings provide insight and guidance for improving agricultural waste resource recovery. In this study, the secretomes of P. eryngii grown on four different carbon sources were compared. The findings revealed the extracellular enzymes implicated in the degradation of lignocellulose, offering avenues for further investigation into the biotransformation mechanisms of P. eryngii biomass and the potential utilization of agricultural wastes. SIGNIFICANCE: The cost of the substrate for mushroom cultivation has increased as the production of edible fungus has risen year after year. Therefore, the use of these locally available lignocellulosic wastes as substrates offers a cost-cutting option. Further, the overuse of wood for the cultivation of edible mushrooms is also detrimental to the conservation of forest resources or the ecological environment. Consequently, the use of other agricultural wastes as an alternative to sawdust or other woody substrates is a viable approach for cultivating P. eryngii. The distribution of extracellular lignocellulosic degrading enzymes, inferred in the present study could help improve the cultivation efficiency of P. eryngii vis-à-vis managing agricultural waste.
Secretome analysis of Pleurotus eryngii reveals enzymatic composition for ramie stalk degradation

Abstract:Pleurotus eryngii (P. eryngii) can secrete large amount of hydrolytic and oxidative enzymes to degrade lignocellulosic biomass. In spite of several researches on the individual lignolytic enzymes, a direct deconstruction of lignocellulose by enzyme mixture is not yet possible. Identifying more high-performance enzymes or enzyme complexes will lead to efficient in vitro lignocelluloses degradation. In this report, secretomic analysis was used to search for the new or interesting enzymes for lignocellulose degradation. Besides, the utilization ability of P. eryngii to ramie stalk substrate was evaluated from the degradation of cellulose, hemicellulose, and lignin in medium and six extracellular enzymes activities during different growth stages were discussed. The results showed that a high biological efficiency of 71% was obtained; cellulose, hemicelluloses, and lignin decomposition rates of P. eryngii were 29.2, 26.0, and 51.2%, respectively. Enzyme activity showed that carboxymethyl cellulase, xylanase, laccase, and peroxidase activity peaks appeared at the primordial initiation stage. In addition, we profiled a global view of the secretome of P. eryngii cultivated in ramie stalk media to understand the mechanism behind lignocellulosic biomass hydrolysis. Eighty-seven nonredundant proteins were identified and a diverse group of enzymes, including cellulases, hemicellulases, pectinase, ligninase, protease, peptidases, and phosphatase implicated in lignocellulose degradation were found. In conclusion, the information in this report will be helpful to better understand the lignocelluloses degradation mechanisms of P. eryngii.

Abstract:Pleurotus ostreutus is one of the world’s most commonly consumed mushrooms. The cultivation of mushrooms using wood resources usually results in environmental issues such as deforestation. Juncao grasses, namely (JJ) Cenchrus fungigraminus, (AR) Saccharum arundinaceum, and (MS) Miscanthus floridulus, supplemented with 20% wheat brain, 1% ground coffee, 1% gysum, and 1% lime, were used as the culture mediums in this research, which offers a composting system with a simple formulation that is cheap and feasible for small farms to use in cultivating oyster mushrooms. The present study assessed the different juncao grasses as substrates for growing Pleurotus ostreatus given their enzyme activities, growth, and yields. The results demonstrated that the yields of pleurotus ostreatus grown on JJ, AR, and MS substrates were significantly different at the level of 0.05 and were recorded as follows: 159.2 g/bag, 132 g/bag, and 65.1 g/bag on average, respectively. The biological efficiency of Pleurotus ostreatus cultivated in three different substrates was 75.2%, 63.4%, and 28.7%, respectively. Lignin peroxidase (LiP) was the most active enzyme in each culture material among the other enzyme activities expressed differently between the substrate and growing stages. At the same time, other enzyme activities were differently expressed between the substrate and different developmental stages. Nutrient analysis revealed significant variations, with differences in polysaccharides, proteins, and amino acids among substrates, as well as the presence of heavy metals such as arsenic, lead, mercury, and cadmium in all samples within safe limits. The obtained results indicated that Saccharum arundinaceum is a good substrate in place of Cenchrus fungigraminus, and that using Miscanthus floridulus is not productive. Moreover, the juncao grasses offer a sustainable approach that reduces reliance on wood-based substrates and enhances environmental sustainability.

Abstract:The objective of this study was to evaluate the production (P) (g of fresh mushrooms /bag) and biological efficiency (BE) (g of fresh mushrooms per 100 g of dry substrate) of 17 fungal strains, namely Pleurotus ostreatus Po-IAP, Po-P38, Po-P14, Po-IE202, Po-Sfco, Po-JP, Po-Psma, and Po-POS, Pleurotus djamour Pd-PRO and Pd-UTMR, Pleurotus eryngii Pe-MB and Pe-PQ, Lentinula edodes L15, L9, L5, and LC, and Hericium erinaceus Heri, produced in corn stover (CS) and to assess the content of crude protein (CP), lignin (L), cellulose, hemicellulose, acid detergent fiber (ADF), and neutral detergent fiber (NDF) and in vitro digestibility of dry matter (IVDMD) of the residual substrate of CS, the so called spent mushroom substrate (SMS), in comparison to the non-inoculated substrate (C). The variables were analyzed as a completely randomized block design using R 4.0.3 software. Means were compared using Tukey's procedure. The Pleurotus spp. strains, compared to Lentinula spp. and Hericium spp., presented better BE and P. In comparison to C, the SMS increased the CP content (p < 0.05) from 10.8% (Po-JP) to 70.3% (LC), while NDF decreased (p < 0.05) from 11.5% (Pd-Pro) to 33.5% (L15) and IVDMD increased (p < 0.05) from 16.2% (Heri) to 47.7% (Pd-UTMR). In conclusion, of the 17 strains evaluated, the 3 strains of Lentinula edodes (L5, L15, and L9), one strain of Pleurotus djamour (Pd-UTMR), and one strain of Pleurotus ostreatus (Po-IAP) generated a SMS that, due to its nutritional improvement and increase in IVDMD, could be used as feed for ruminants. Our results also showed that corn stover is a suitable substrate to produce Pleurotus spp. fruiting bodies, with strain Po-IAP as the best yielding.
Effects of mixed agro-residues (corn crop waste) on lignin-degrading enzyme activities, growth, and quality of Lentinula edodes

Abstract:Using agro-residues in bioreactors to produce mushrooms is an important component of eco-agriculture. Here, we tested the cultivation of Lentinula edodes with corn cob (CC) and corn straw (CS), and then evaluated the resulting enzyme activities, agronomic traits, textures and nutrient compositions of mushrooms. Laccase (T1 formula, 3.26 g U-1) and carboxymethyl cellulase (T3 formula, 1.01 g U-1) activities were the highest at the time for a complete substrate colonization stage (TCSC), while acidic xylanase activity was the highest (CK formula, 4.05 g U-1) in the mushroom block to color-turned (TMBCT) stage. The biological efficiency of growth on the T6 formula was 8.82% higher than growth on the CK formula, wherein the low C/N ratio of the substrate had an obvious negative effect on yield while the mass ratio of pileus (MRP) of fruiting bodies did not change with mixed substrates. No significant differences were observed in mineral composition for CK formulas, but corn crop waste (CCW) formulas exhibited more optimal nutritional contents. A formula containing more corn cob and sawdust (SD) (sum of at least 70%) as the substrate can produce fruiting bodies with good hardness. These results indicate that the use of corn cobs as the main ingredient, mixed with sawdust and corn straw to grow L. edodes provides a more efficient use of agro-residues for growth. Thus, mixed agro-residue formulas have exceptional advantages in texture, nutrition of fruiting bodies, and yields.

Abstract:Pollution made cancer a major disease threatening people's health. In the search for new cancer drug treatments, natural products perform an essential role. Escin, a mixture of triterpene saponins extracted from Aesculus Wilsonii Rehd., was used to analyze the cell growth inhibitory effects and the induction of apoptosis in human cervical carcinoma Hela cancer cell line in vitro. The results of MTT cell viability assay showed that escin could induce significant concentration-dependent and time-dependent inhibition of cell growth, and the cell cycle was arrested at G 1 /S phase in the escin-treated cells. Apoptosis were detected and assessed by Acridine Orange/Ethidium Bromide Staining and Flow Cytometry. The antitumor efficacy of escin on mice cervical carcinoma U_(14) in vivo was investigated, at dose of 2.8 mg/kg escin had rather high inhibition ratio(43.5%) on U_(14) tumor growth.
Co-cultured Production of Lignin-Modifying Enzymes with White-Rot Fungi

Abstract:Co-cultivation was a potential strategy in lignocellulolytic biodegradation with producing high activity enzymes due to their synergistic action. The objective of this study was to investigate the rarely understood effects of co-culturing of two white-rot fungi on lignin-modifying enzymes (LMEs) production. Six species, Bjerkandera adusta, Phlebia radiata, Pleurotus ostreatus, Dichomitus squalens, Hypoxylon fragiforme and Pleurotus eryngii, were cultured in pairs to study the production of LMEs. The paired hyphal interaction observed showed that P. eryngii is not suitable for co-growth. The use of agar plates containing dye RBBR showed elevated decolourisation at the confrontation zone between mycelia. Laccase was significantly stimulated only in the co-culture of P. radiata with D. squalens under submerged cultivation; the highest value was measured after 4 days of incubation (120 U mg−1). The improved productions of MnP and LiP were simultaneously observed at the co-culture of P. ostreatus and P. radiata (MnP = 800 nkat L−1 after 4 days of incubation; LiP = 60 nkat L−1 after 7 days of incubation), though it was not a good producer of laccase. P. ostreatus appeared to possess specific potential to be used in co-cultured production of LMEs. The phenotype of LMEs production was not only dependent on the species used but also regulated by different nutritions available in the culture medium. The present data will provide evidence for illustrating the regulatory roles of C/N on LMEs production under the co-cultures’ circumstances.

Abstract:Distinct wood degraders occupying their preferred habitats have biased enzyme repertoires that are well fitted to their colonized substrates. Pleurotus ostreatus, commonly found on wood, has evolved its own enzyme-producing traits. In our previous study, transcriptional shifts in several P. ostreatus delignification-defective mutants, including Δhir1 and Δgat1 strains, were analyzed, which revealed the downregulation of ligninolytic genes and the upregulation of cellulolytic and xylanolytic genes when compared to their parental strain 20b on beech wood sawdust medium (BWS). In this study, rice straw (RS) was used as an alternative substrate to examine the transcriptional responses of P. ostreatus to distinct substrates. The vp1 gene and a cupredoxin-encoding gene were significantly upregulated in the 20b strain on RS compared with that on BWS, reflecting their distinct regulation patterns. The overall expression level of genes encoding glucuronidases was also higher on RS than on BWS, showing a good correlation with the substrate composition. Transcriptional alterations in the mutants (Δhir1 or Δgat1 versus 20b strain) on RS were similar to those on BWS, and the extracellular lignocellulose-degrading enzyme activities and lignin-degrading ability of the mutants on RS were consistent with the transcriptional alterations of the corresponding enzyme-encoding genes. However, transcripts of specific genes encoding enzymes belonging to the same CAZyme family exhibited distinct alteration patterns in the mutant strains grown on RS compared to those grown on BWS. These findings provide new insights into the molecular mechanisms underlying the transcriptional regulation of lignocellulolytic genes in P. ostreatus.

Abstract:Abstract Background Identifying new high-performance enzymes or enzyme complexes to enhance biomass degradation is the key for the development of cost-effective processes for ethanol production. Irpex lacteus is an efficient microorganism for wheat straw pretreatment, yielding easily hydrolysable products with high sugar content. Thus, this fungus was selected to investigate the enzymatic system involved in lignocellulose decay, and its secretome was compared to those from Phanerochaete chrysosporium and Pleurotus ostreatus which produced different degradation patterns when growing on wheat straw. Extracellular enzymes were analyzed through 2D-PAGE, nanoLC/MS-MS, and homology searches against public databases. Results In wheat straw, I. lacteus secreted proteases, dye-decolorizing and manganese-oxidizing peroxidases, and H 2 O 2 producing-enzymes but also a battery of cellulases and xylanases, excluding those implicated in cellulose and hemicellulose degradation to their monosaccharides, making these sugars poorly available for fungal consumption. In contrast, a significant increase of β-glucosidase production was observed when I. lacteus grew in liquid cultures. P. chrysosporium secreted more enzymes implicated in the total hydrolysis of the polysaccharides and P. ostreatus produced, in proportion, more oxidoreductases. Conclusion The protein pattern secreted during I. lacteus growth in wheat straw plus the differences observed among the different secretomes, justify the fitness of I. lacteus for biopretreatment processes in 2G-ethanol production. Furthermore, all these data give insight into the biological degradation of lignocellulose and suggest new enzyme mixtures interesting for its efficient hydrolysis.

Abstract:Solid state fermentation with different lignocellulolytic materials as inducers was used for lignocellulolytic enzyme production in this study. Pleurotus ostreatus strains were assessed by measuring laccase, CMCase, and xylanase activities. The secretion potential of the lignocellulolytic enzymes by wild and cultivated strains was analyzed for the first time. The wild and cultivated strain showed their unique capacities for secreting lignocellulolytic enzymes on solid-state fermentation with different lignocellulosic materials. The wild P. ostreatus strain preferred corncob for the secretion of laccase and xylanase activity, but the cultivated strain preferred poplar sawdust. The wild strain and cultivated strain showed a consistent preference for poplar sawdust for the secretion of CMCase activity. The wild strain was advantageous because it achieved the maximum hydrolytic enzyme activities within a short time period. Poplar sawdust and corncob were conducive to laccase secretion by the wild or cultivated strains and the rapid accumulation of laccase on solid-state fermentation. Additionally, continuous, stable laccase production was an extremely important advantage by solid-state fermentation of poplar sawdust, particularly in the wild strain. These findings are helpful in selecting the appropriate strain that corresponds to suitable lignocellulosic materials. The optimization of integrated industrial lignocellulolytic enzyme production can also be achieved.

Abstract:Influence of water content on the expression of lignocellulolytic enzymes by Phanerochaete chrysosporium remains unclear. This work compares the enzyme production profiles of P. chrysosporium during solid-state and submerged fermentation. There were 110 and 64 extracellular carbohydrate-active enzymes identified in solid-state and submerged fermentation respectively, among which 57 enzymes were common to both of the secretomes. P. chrysosporium secreted more cellulases (especially lytic polysaccharide monooxygenase) and hemicellulases during solid-state fermentation while the proportion of enzyme containing carbohydrate-binding module was higher for submerged fermentation. Although its activities were weaker, the enzyme cocktail from submerged fermentation was surprisingly more effective in hydrolysis at low substrate loading. This advantage of enzymes from submerged fermentation was mainly attributed to carbohydrate-binding module because more xylanases bound with substrate at the beginning of hydrolysis. These results reveal the influence of fermentation conditions on enzyme produced by P. chrysosporium for the first time and show the importance of carbohydrate-binding module in the hydrolysis process of lignocellulose.
Improvement of Exo-Polysaccharides Production and Modeling Kinetics by Armillaria Luteo-Virens Sacc. in Submerged Cultivation

Abstract:Armillaria luteo-virens Sacc. is a special medicinal and edible mushroom in China, and its wild type has been investigated that it contains some functional components. The optimization of submerged culture conditions for mycelium growth and exo-polysaccharide (EPS) production in an edible mushroom A. luteo-virens Sacc. was studied in shake flasks and bioreactors with response surface methodology (RSM). A fractional factorial design (FFD) and central composite design (CCD) were applied to optimize the main factors that affect the fungal polysaccharides production in a submerged culture. Glucose is very important for EPS production by A. luteo-virens Sacc., and yeast extract powder also significantly affects fungal EPS production through fractional factorial experiment. The optimized cultivation medium constituents were derived from the predicted model using central composite design as follows: 31.26g/l glucose, 1.06g/l yeast extract powder, 1.00g/l K2HPO4, 0.50g/l KH2PO4, and 0.45g/l MgSO4·7H2O, pH unadjusted. The predicted maximal EPS concentration was 5.40g/l using the optimized culture medium compositions at 23°C for 5 days. Additionally, the submerged cultivation process was explained using the optimized cultivation medium. The cultivation kinetics model constructed was suitably depicting the culture process in bioreactor cultivation scale.

Abstract:Understand the process of biodegradation of lignocellulosic material as its chemical composition is key properties for enhancing the cultivation of edible mushrooms, which seek in lignin and other carbon source carbohydrates. The purpose of this study was to monitor polysaccharide and lignin degradation caused by Pleurotus ostreatoroseus on different agroindustrial substrates using near infrared spectroscopy (NIR). Sawdust substrates of Marupá and Cajuí wood, banana pseudostem fibers, enriched with bran of cereals were formulated (WB and MWB). Of the 12 formulations elaborated, eight were submitted to biodegradation of P. ostreatoroseus over a period of 49 days (Residual substrate), and four formulations maintained in natura (Initial substrates), where each treatment was formed by 10 replicates totaling n = 120. Spectra were obtained from the substrates before and after cultivation in FT-NIR system in the region between 10,000 and 4,000 cm-1 and the index degradation of lignocellulosic compounds was determined (LDI and PDI). Through Fourier transform near infrared spectroscopy was it is possible to observe the degradation of polysaccharides and lignin in the substrates grown with the fungus P. ostreatoroseus. In the residual substrate Marupá+WB-POSP was the highest lignin degradation index (LDI = 9.26%). While whereas for the PDI (Polysaccharide degradation index), the highest degradation (7.12%) was observed in the residual substrate Cajuí+MWB-POAM. Through analysis of the absorption bands, it was possible to observe the degradation of structures/bonds characteristics of lignocellulosic residues. The NIR model for degradation of lignocellulosic compounds was more effective in predicting “lignin degradation” in the spectral range 6,350-7,308 cm-1

Abstract:In the textile industry, ramie stalk is byproducts with a low economic value. The potential use of this leftover as a substrate ingredient for Flammulina velutipes (F. velutipe) cultivation was evaluated. The degradation and utilization of ramie stalk by F. velutipes was evaluated through mushroom production, lignocelluloses degradation and lignocellulolytic enzymes activity. The best substrate mixture for F. velutipes cultivation comprised 50% ramie stalk, 20% cottonseed hulls, 25% wheat bran, 4% cornstarch and 2% CaCO3. The highest biological efficiency of fruiting bodies was reached 119.7%. F. velutipes appears to degrade 12.7-32.0% lignin, 14.4-30.2% cellulose and 9.3-25.7% hemicellulose during cultivation on the different substrates. The results of enzymes activities showed that laccase and peroxidase were higher before fruiting; while cellulase and hemicellulase showed higher activities after fruiting. The biological efficiency of fruiting bodies was positively correlated with the activities of cellulase, hemicellulase and ligninolytic enzyme. The results of this study demonstrate that ramie stalk can be used as an effective supplement for increasing mushroom yield in F. velutipes.
Studies on the Biological Characteristics of Different Pleurotus eryngii Strains

Abstract:Abstrcat The mycelial antagonistic test, esterase isozyme zymogram, mycelial growth rate, mycelial biomass and polysaccharide content of 4 different Pleurotus eryngii strains were studied in this paper. The results showed that the genetic diversity was existed in 4 strains. The mycelial growth rate of strain X_3 was the fastest, being (0.69cm) per day. The mycelial biomass of strain X_2 was the highest, being 1.572g/100mL. The polysaccharide content of strain X_1 was 39.07μg/mg which was the lowest. However, the polysaccharide contents of strains X_2、X_4 and X_3 were 84.28μg/mg,79.20μg/mg and 78.99μg/mg respectively.

Abstract:The white-rot basidiomycete Lentinula edodes is the second most edible mushroom cultivated on the world. This fungus decomposes cell-wall associated macromolecules, is a natural degrader of lignin polymers. The differences in enzyme activities between strains of L. edodes provided useful information about the participation of enzymes in different development stages of the fungus. The effect of lignin on the fungal biomass production and activity behavior of ligninolytic enzymes when L. edodes is cultivated in a medium containing lignin with and without glucose as a carbohydrate source was tested. When glucose was present in the culture, lignin increase the mycelial biomass by 70% at 22 days compared to the control culture. The lignin media without glucose affected mycelial growth up to 20% less that the control without lignin and glucose. The activity of laccase, lignin peroxidase, aryl alcohol oxidase, manganese dependent peroxidase and catalase was modified depending on whether the medium had lignin and glucose, or lignin alone. A carbohydrate source is important to fungal growth, but the dissolution of lignin monomers might switch the signal that controls growth rate and enzymatic activity.
