Abstract:Many fish populations are exposed to harmful levels of chemical pollution and selection pressures associated with these exposures have led to the evolution of tolerance. Our understanding of the physiological basis for these adaptations is limited, but they are likely to include processes involved with the absorption, distribution, metabolism and/or excretion of the target chemical. Other potential adaptive mechanisms include enhancements in antioxidant responses, an increased capacity for DNA and/or tissue repair and alterations to the life cycle of fish that enable earlier reproduction. Analysis of single-nucleotide polymorphism frequencies has shown that tolerance to hydrocarbon pollutants in both marine and estuarine fish species involves alteration in the expression of the xenobiotic metabolism enzyme CYP1A. In this review, we present novel data showing also that variants of the CYP1A gene have been under selection in guppies living in Trinidadian rivers heavily polluted with crude oil. Potential costs associated with these adaptations could reduce fitness in unpolluted water conditions. Integrating knowledge of local adaptation to pollution is an important future consideration in conservation practices such as for successful restocking, and improving connectivity within river systems. Pharmaceutical pollutants are increasingly common in aquatic ecosystems, and organisms living there are the unintended targets of these compounds, originally designed for humans. However, our understanding of these effects remains limited. In a recent study, Aich et al. (2024) investigated the effects of fluoxetine on a small freshwater fish, the guppy (Poecilia reticulata), with particular emphasis on behaviour. The authors found that, after multigenerational exposure to ecologically relevant concentrations of this compound, there was minimal impact on average behavioural traits at the population level. However, fluoxetine exposure reduced within‐individual behavioural plasticity and altered the delicate equilibrium among these traits. These findings could have far‐reaching evolutionary implications for how individuals and populations may respond to environmental changes and highlight the need for behavioural ecotoxicology to move beyond population‐level effects. This research provides valuable insights into the subtle and multifaceted—yet profound—impacts of pollutants on ecological and evolutionary processes, highlighting the importance of studying behavioural responses at the individual level to predict how populations will respond to our rapidly changing world.

Abstract:Chronic exposure to antidepressants can impact non‐target animals, particularly the life‐history, behaviour and reproductive traits of male guppies. Key findings include lower sperm velocity, reduced behavioural plasticity and altered between‐individual correlations in pace‐of‐life syndrome traits, highlighting the need to better understand the ecological impacts of pollutants on wildlife. In our rapidly changing world, understanding how species respond to shifting conditions is of paramount importance. Pharmaceutical pollutants are widespread in aquatic ecosystems globally, yet their impacts on animal behaviour, life‐history and reproductive allocation remain poorly understood, especially in the context of intraspecific variation in ecologically important traits that facilitate species' adaptive capacities. We test whether a widespread pharmaceutical pollutant, fluoxetine (Prozac), disrupts the trade‐off between individual‐level (co)variation in behavioural, life‐history and reproductive traits of freshwater fish. We exposed the progeny of wild‐caught guppies (Poecilia reticulata) to three field‐relevant levels of fluoxetine (mean measured concentrations: 0, 31.5 and 316 ng/L) for 5 years, across multiple generations. We used 12 independent laboratory populations and repeatedly quantified activity and risk‐taking behaviour of male guppies, capturing both mean behaviours and variation within and between individuals across exposure treatments. We also measured key life‐history traits (body condition, coloration and gonopodium size) and assessed post‐copulatory sperm traits (sperm vitality, number and velocity) that are known to be under strong sexual selection in polyandrous species. Intraspecific (co)variation of these traits was analysed using a comprehensive, multivariate statistical approach. Fluoxetine had a dose‐specific (mean) effect on the life‐history and sperm trait of guppies: low pollutant exposure altered male body condition and increased gonopodium size, but reduced sperm velocity. At the individual level, fluoxetine reduced the behavioural plasticity of guppies by eroding their within‐individual variation in both activity and risk‐taking behaviour. Fluoxetine also altered between‐individual correlations in pace‐of‐life syndrome traits: it triggered the emergence of correlations between behavioural and life‐history traits (e.g. activity and body condition) and between life‐history and sperm traits (e.g. gonopodium size and sperm vitality), but collapsed other between‐individual correlations (e.g. activity and gonopodium size). Our results reveal that chronic exposure to global pollutants can affect phenotypic traits at both population and individual levels, and even alter individual‐level correlations among such traits in a dose‐specific manner. We discuss the need to integrate individual‐level analyses and test behaviour in association with life‐history and reproductive traits to fully understand how animals respond to human‐induced environmental change.

Abstract:Abstract The global rise of pharmaceutical contaminants in the aquatic environment poses a serious threat to ecological and evolutionary processes. Studies have traditionally focused on the collateral (average) effects of psychoactive pollutants on ecologically relevant behaviors of wildlife, often neglecting effects among and within individuals, and whether they differ between males and females. We tested whether psychoactive pollutants have sex-specific effects on behavioral individuality and plasticity in guppies (Poecilia reticulata), a freshwater species that inhabits contaminated waterways in the wild. Fish were exposed to fluoxetine (Prozac) for 2 years across multiple generations before their activity and stress-related behavior were repeatedly assayed. Using a Bayesian statistical approach that partitions the effects among and within individuals, we found that males—but not females—in fluoxetine-exposed populations differed less from each other in their behavior (lower behavioral individuality) than unexposed males. In sharp contrast, effects on behavioral plasticity were observed in females—but not in males—whereby exposure to even low levels of fluoxetine resulted in a substantial decrease (activity) and increase (freezing behavior) in the behavioral plasticity of females. Our evidence reveals that psychoactive pollution has sex-specific effects on the individual behavior of fish, suggesting that males and females might not be equally vulnerable to global pollutants.

Abstract:Higher temperature and turbidity acted independently to alter fish social behavior. Fish distribution in the arena became more aggregated in warmer water. With turbidity, wide‐scale interactions were severed resulting in less cohesive groups. Changes in environmental conditions can shift the costs and benefits of aggregation or interfere with the sensory perception of near neighbors. This affects group cohesion with potential impacts on the benefits of collective behavior such as reduced predation risk. Organisms are rarely exposed to one stressor in isolation, yet there are only a few studies exploring the interactions between multiple stressors and their effects on social behavior. Here, we tested the effects of increased water temperature and turbidity on refuge use and three measures of aggregation in guppies (Poecilia reticulata), increasing temperature and turbidity in isolation or in combination. When stressors were elevated in isolation, the distribution of fish within the arena as measured by the index of dispersion became more aggregated at higher temperatures but less aggregated when turbidity was increased. Another measure of cohesion at the global scale, the mean inter‐individual distance, also indicated that fish were less aggregated in turbid water. This is likely due to turbidity acting as a visual constraint, as there was no evidence of a change in risk perception as refuge use was not affected by turbidity. Fish decreased refuge use and were closer to their nearest neighbor at higher temperatures. However, the nearest neighbor distance was not affected by turbidity, suggesting that local‐scale interactions can be robust to the moderate increase in turbidity used here (5 NTU) compared with other studies that show a decline in shoal cohesion at higher turbidity (>100 NTU). We did not observe any significant interaction terms between the two stressors, indicating no synergistic or antagonistic effects. Our study suggests that the effects of environmental stressors on social behavior may be unpredictable and dependent on the metric used to measure cohesion, highlighting the need for mechanistic studies to link behavior to the physiology and sensory effects of environmental stressors.

Abstract:Although (PS)2, the primary degradation product of emerging antifouling biocides metal pyrithiones (MePTs), can disrupt the reproductive behavior of fish at an environmentally relevant ng/L level, the underlying mechanism is still largely unknown. This study exposed sexually mature male guppy (Poecilia reticulata) to 20, 200, and 2000 ng/L (PS)2 to explore the compromised effect of (PS)2 on reproductive behavior through a realistic competing scenario. The results showed that (PS)2 suppressed male guppies' sexual interest to stimulus females, reduced their competitive behavior frequencies toward rival males, and decreased their mating time and frequency. (PS)2 exposure did not affect male guppies' secondary sexual characteristics or induce estrogenic activity. Whole-brain transcriptome sequencing identified 1070 differentially expressed genes (DEGs) with 872 up-regulated genes, which were functionally enriched into Gene Ontology terms pertaining to extracellular matrix (ECM) and extracellular region. KEGG enrichment for the DEGs uncovered that the activations of ECM-receptor interaction and focal adhesion pathways could be the underlying molecular mechanism implicated in the (PS)2 induced reproductive behavior impairment. This work would deliver a substantial contribution to the understanding of the ecological safety of MePTs biocides.

Abstract:Human disturbances can prompt natural anti-predator behaviours in animals, affecting how energy is traded off between immediate survival and reproduction. In our study of male squaretail groupers ( Plectropomus areolatus ) in India’s Lakshadweep archipelago, we investigated the impact of fishing pressure on anti-predatory responses and reproductive behaviours by comparing a fished and unfished spawning aggregation site and tracking responses over time at the fished site. Using observational sampling and predator exposure experiments, we analysed fear responses (flight initiation distance, return time), as well as time spent in vigilance, courtship and territorial defence. Unpaired males at fished sites were twice as likely to flee from simulated predators and took longer to return to mating territories. In contrast, paired males at both sites took greater risks during courtship, fleeing later than unpaired males, but returned earlier at the unfished site compared with the fished site. Our findings suggest that high fishing pressure reduces reproductive opportunities by increasing vigilance and compromising territorial defence, potentially affecting mate selection cues. Altered behavioural trade-offs may mitigate short-term capture risk but endanger long-term population survival through altered reproductive investment. Human extractive practices targeting animal reproductive aggregations can have disruptive effects beyond direct removal, influencing animal behaviours crucial for population survival.

Abstract:Wild populations often differ in their tolerance to environmental stressors, but intraspecific variability is rarely taken into account in ecotoxicology. In addition, plastic responses to multiple stressors have rarely been investigated in realistic field conditions. In this study, we compared the responses to metal contamination of gudgeon populations ( Gobio occitaniae ) differing in their past chronic exposure to metal contamination, using a reciprocal transplant experiment and an immune challenge mimicking a parasite attack to test for potential effects of multiple stressors across biological levels. We measured fish survival and traits involved in metal bioaccumulation, oxidative stress, immunity, cell apoptosis, and energy management to decipher underpinning physiological mechanisms across biological levels (i.e., gene expression, cell, organism). Fish from the two replicate High Contamination sites had higher survival when transferred into contaminated sites, suggesting a local adaptation to the contaminated site, possibly explained by higher levels of detoxification and antioxidant capacity but with potential higher apoptosis costs compared to their naïve counterparts. We found no evidence of co- or maladaptation to the immune stressor, suggesting no specific costs to face pathogens. In the emerging field of evolutionary ecotoxicology, this study underlines the need to consider intraspecific variability to better understand the effects of pollution in heterogeneous populations.

Abstract:Behavioural plasticity allows organisms to respond to environmental challenges on short time scales. But what are the ecological and evolutionary processes that underlie behavioural plasticity? The answer to this question is complex and requires experimental dissection of the physiological, neural and molecular mechanisms contributing to behavioural plasticity as well as an understanding of the ecological and evolutionary contexts under which behavioural plasticity is adaptive. Here, we discuss key insights that research with Trinidadian guppies has provided on the underpinnings of adaptive behavioural plasticity. First, we present evidence that guppies exhibit contextual, developmental and transgenerational behavioural plasticity. Next, we review work on behavioural plasticity in guppies spanning three ecological contexts (predation, parasitism and turbidity) and three underlying mechanisms (endocrinological, neurobiological and genetic). Finally, we provide three outstanding questions that could leverage guppies further as a study system and give suggestions for how this research could be done. Research on behavioural plasticity in guppies has provided, and will continue to provide, a valuable opportunity to improve understanding of the ecological and evolutionary causes and consequences of behavioural plasticity.

Abstract:Estuarine ecosystem balance typically relies on strong food web interconnectedness dependent on a relatively low number of resident taxa, presenting a potential ecological vulnerability to extreme ecosystem disturbances. Following the Deepwater Horizon (DwH) oil spill disaster of the northern Gulf of Mexico (USA), numerous ecotoxicological studies showed severe species-level impacts of oil exposure on estuarine fish and invertebrates, yet post-spill surveys found little evidence for severe impacts to coastal populations, communities, or food webs. The acknowledgement that several confounding factors may have limited researchers' abilities to detect negative ecosystem-level impacts following the DwH spill drives the need for direct testing of weathered oil exposure effects on estuarine residents with high trophic connectivity. Here, we describe an experiment that examined the influence of previous exposure to four weathered oil concentrations (control: 0.0 L oil m-2; low: 0.1 L oil m-2; moderate: 0.5-1 L oil m-2; high: 3.0 L oil m-2) on foraging rates of the ecologically important Gulf killifish (Fundulus grandis). Following exposure in oiled saltmarsh mesocosms, killifish were allowed to forage on grass shrimp (Palaeomonetes pugio) for up to 21 h. We found that previous exposure to the high oil treatment reduced killifish foraging rate by ~37% on average, compared with no oil control treatment. Previous exposure to the moderate oil treatment showed highly variable foraging rate responses, while low exposure treatment was similar to unexposed responses. Declining foraging rate responses to previous high weathered oil exposure suggests potential oil spill influence on energy transfer between saltmarsh and off-marsh systems. Additionally, foraging rate variability at the moderate level highlights the large degree of intraspecific variability for this sublethal response and indicates this concentration represents a potential threshold of oil exposure influence on killifish foraging. We also found that consumption of gravid vs non-gravid shrimp was not independent of prior oil exposure concentration, as high oil exposure treatment killifish consumed ~3× more gravid shrimp than expected. Our study findings highlight the sublethal effects of prior oil exposure on foraging abilities of ecologically valuable Gulf killifish at realistic oil exposure levels, suggesting that important trophic transfers of energy to off-marsh systems may have been impacted, at least in the short-term, by shoreline oiling at highly localized scales. This study provides support for further experimental testing of oil exposure effects on sublethal behavioral impacts of ecologically important estuarine species, due to the likelihood that some ecological ramifications of DwH on saltmarshes likely went undetected.
The foraging behavior of gerbils reveals the ecological significance of crude oil pollution

Abstract:Despite extensive ecotoxicological evidence on the adverse effects of oil pollution on rodents, little is known about how rodents make decisions in oil-polluted environments (i.e., outside of lab settings). We investigated the foraging behavior of Allenby gerbils, Gerbillus andersoni allenbyi (GA), that were presented with feeding trays in a semi-natural environment. The trays contained seeds mixed into one of three types of soils – clean soil, and two different soil samples collected from two well-documented terrestrial oil spill sites in Israel. The oil spill disasters occurred in 1975 and 2014 and the spill sites are located within a few hundred meters of each other, in the 'Avrona Nature Reserve in the Arava hyper-arid region in Israel.Gerbils of both sexes avoided foraging in 2014-polluted soil, but surprisingly, they foraged more in 1975-polluted soil. Our results indicate that for the GA, the 1975-polluted soil is an advantageous substrate to forage on, probably because its texture facilitates more efficient foraging, leading to greater energetic gain, and creating a trade-off between energetic gain and perceived foraging cost due to its pollution. We also proceeded to investigate some physiological consequences of chronic exposure to the 2014-polluted soil in the laboratory. Chronic oil exposure did not lead to mortality or weight loss, but female gerbils exhibited heightened cortisol. We conclude that terrestrial oil pollution may have significant sublethal impacts on animal behavior, even when there is no obvious short-term physiological cost to the exposure.

Effects of short‐ and long‐term enrichment on brain and behavior in Trinidadian guppies

Abstract:Environmental enrichment often alters behavior in captive animals. Here, we exposed guppies either to short‐term (two week) or long‐term (birth through 12 weeks) enrichment and measured growth, brain size, and neophobic behaviors. Females responded more strongly overall to enrichment in both short‐ and long‐term. Both male and female guppies reared with enrichment were smaller in size, but with larger relative brain size than deprived guppies. Overall, we find sex‐specific responses to enrichment in Trinidadian guppies, and that even short‐term exposure to enrichment can result in behavioral changes. Environmental enrichment (EE) often increases positive behavioral and physiological effects on captive animals. Fish are commercially and scientifically important taxa that have been shown to benefit from EE. Here we examined the effects of both short‐ and long‐term EE in Trinidadian guppies (Poecilia reticulata). In the short‐term exposure to EE, female guppies were raised in standard conditions, and after reaching adulthood were moved to either an enriched or a deprived environment for 2 weeks. Long‐term exposure guppies were reared from birth for 12 weeks (until sexual maturity) in either an enriched or deprived environment. We then assessed growth, brain size, and neophobic and exploratory behaviors in standard assays. Guppies given EE were bolder, regardless of timescale, although females showed a more pronounced change in behaviors than males. We further found that guppies reared with EE were smaller yet had larger relative brain sizes than guppies reared under deprived conditions. Here we highlight that EE had influenced growth, brain size, neophobic, and exploratory behaviors in guppies, and behavioral changes were observed after only 2 weeks. Our results highlight the need for assessing the influences of EE in captivity, in particular for researchers studying cognition and behavior.

Abstract:Evolutionary biologists have long been interested in parsing out the roles of genetics, plasticity and their interaction on adaptive trait divergence. Since males and females often have different ecological and reproductive roles, separating how their traits are shaped by interactions between their genes and environment is necessary and important. Here, we disentangle the sex-specific effects of genetic divergence, developmental plasticity, social learning and contextual plasticity on foraging behaviour in Trinidadian guppies ( Poecilia reticulata ) adapted to high- or low-predation habitats. We reared second-generation siblings from both predation regimes with or without predator chemical cues, and with adult conspecifics from either high- or low-predation habitats. We then quantified their foraging behaviour in water with and without predator chemical cues. We found that high-predation guppies forage more efficiently than low-predation guppies, but this behavioural difference is context-dependent and shaped by different mechanisms in males and females. Higher foraging efficiency in high-predation females is largely genetically determined, and to a smaller extent socially learned from conspecifics. However, in high-predation males, higher foraging efficiency is plastically induced by predator cues during development. Our study demonstrates sex-specific differences in genetic versus plastic responses in foraging behaviour, a trait of significance in organismal fitness and ecosystem dynamics.

Abstract:Plasminogen activator inhibitor-1 (PAI-1) expression is induced by hypoxia (8% O(2)) via the PAI-1 promoter region -175/-159 containing a hypoxia response element (HRE-2) binding the hypoxia-inducible factor-1 (HIF-1) and an adjacent response element (HRE-1) binding a so far unknown factor. It was found by supershift assays that the upstream stimulatory factor-2a (USF-2a) bound mainly to the HRE-1 of the PAI-1 promoter and to a lesser extent to HRE-2. Overexpression of USF-2a inhibited PAI-1 messenger RNA and protein expression and activated L-type pyruvate kinase expression in primary rat hepatocytes under normoxia and hypoxia. Luciferase (Luc) gene constructs driven by 766 and 276 base pairs of the 5'-flanking region of the PAI-1 gene were transfected into primary hepatocytes together with expression vectors encoding wild-type USF-2a and a USF-2a mutant lacking DNA binding and dimerization activity (DeltaHU2a). Cotransfection of the wild-type USF-2a vector reduced Luc activity by about 8-fold, whereas cotransfection of DeltaHU2a did not influence Luc activity. Mutation of the HRE-1 (-175/-168) in the PAI-1 promoter Luc constructs decreased USF-dependent inhibition of Luc activity. Mutation of the HRE-2 (-165/-158) was less effective. Cotransfection of a HIF-1alpha vector could compete for the binding of USF at HRE-2. These results indicated that the balance between 2 transcriptional factors, HIF-1 and USF-2a, which can bind adjacent HRE sites, appears to be involved in the regulation of PAI-1 expression in many clinical conditions.

Abstract:The title molecule, [Zn2(C9H10NO2)4(C10H14N2O)2], is a centrosymmetric binuclear complex, with Zn atoms [Zn⋯Zn′ = 2.8927 (4) Å] bridged by four carboxylate groups from the dimethylaminobenzoate (DMAB) ligands. The four carboxyl O atoms around the Zn atom form a distorted square-planar arrangement; the distorted square-pyramidal coordination geometry is completed by the pyridine N atom of the N,N-diethylnicotinamide (DENA) ligand. The Zn atom is displaced by 0.3326 (2) Å from the plane of the four O atoms, with an average Zn—O distance of 2.0416 (12) Å. The dihedral angles between the carboxylate groups and the adjacent benzene rings are 5.31 (8) and 11.00 (9)°, while the pyridine ring is oriented at dihedral angles of 66.26 (6) and 37.88 (7)° with respect to the benzene rings. Weak intramolecular C—H⋯O and intermolecular C—H⋯π interactions are present.
Multiple environmental stressors affect predation pressure in a tropical freshwater system

Abstract:Environmental change can alter predator-prey dynamics. However, studying predators in the context of co-occurring environmental stressors remains rare, especially under field conditions. Using in situ filming, we examined how multiple stressors, including temperature and turbidity, impact the distribution and behaviour of wild fish predators of Trinidadian guppies ( Poecilia reticulata ). The measured environmental variables accounted for 17.6% of variance in predator species composition. While predator species differed in their associations with environmental variables, the overall prevalence of predators was greatest in slow flowing, deeper, warmer and less turbid habitats. Moreover, these warmer and less turbid habitats were associated with earlier visits to the prey stimulus by predators, and more frequent predator visits and attacks. Our findings highlight the need to consider ecological complexity, such as co-occurring stressors, to better understand how environmental change affects predator-prey interactions.

Abstract:Microplastics (MPs) are widely distributing in aquatic environment. They are easily ingested by aquatic organisms and accumulate in digestive tract especially of intestine. To explore the potential effects of MPs on intestine, here we, using juvenile guppy (Poecilia reticulata) as experimental animal, investigated the response characteristics of digestion, immunity and gut microbiota. After exposure to 100 and 1000 μg/L concentrations of MPs (polystyrene; 32-40 μm diameters) for 28 days, we observed that MPs could exist in guppy gut and induce enlargement of goblet cells. Activities of digestive enzymes (trypsin, chymotrypsin, amylase and lipase) in guppy gut generally reduced. MPs stimulated the expression of immune cytokines (TNF-α, IFN-γ, TLR4 and IL-6). Through high throughput sequencing of 16S rRNA gene, decreases in diversity and evenness and changed composition of microbiota were found in guppy gut. PICRUSt analysis revealed that MPs might have effects on intestinal microbiota functions, such as inhibition of metabolism and repair pathway. Our findings suggested that MPs could retain in the gut of juvenile guppy, impair digestive performance, stimulate immune response and induce microbiota dysbiosis in guppy gut. The results obtained here provide new insights into the potential risks of MPs to aquatic animals.
