Abstract:Both beta-Adrenergic receptor (beta-AR) signaling pathway and Ca2+/Calmodulin-dependent protein kinase II (CaMKII) play important roles in regulating cardiac excitation-contraction coupling. However, the laboratory experiments cannot quantify these regulatory roles easily, computer models have been used as an alternative way. The aim of this study was to use computer models to preliminarily investigate the combined role of beta-AR and CaMKII in human cardiac myocyte propertiesWith the hypothesis that the beta-AR is mainly meditated by the targets of protein kinase A (PKA) phosphorylation, the influence of PKA was incorporated into a modified ORd's model. The formulations of L-type Ca2+ current, slow-activating delayed rectifier K+ current and phospholamban were modified. The simulation results were then compared with and without the administration of isoproterenol (ISO).Our simulation showed that, in comparison with the control, at the cycle length of 1000 ms ISO shortened APD90 by 15ms (265 ms vs 250 ms), and the amplitude of Ca2+ transient was increased by approximate 60%. This increase was not observed for [ Na+]. Our findings agreed well with published experimental observations.In conclusion, a computer model has been developed to investigate the role of beta-AR and CaMKII in human myocyte electrophysiological properties. This model can be used as a platform to further investigate their role in cardiac arrhythmias
Antiarrhythmic Effects of Magnesium Sulfate on Experimental Arrhythmias and Its Influences on INa in Rats Ventricular Cells

Abstract:To observe the antiarrhythmic effects of magnesium sulfate on aconitine-induced arrhythmias in rats and its influences on I_ Na in rats ventricular cells,aconitine-induced arrhythmias model in rats were used. The doses of aconitine to induce ventricular premature beat(VPB), ventricular tachycardia(VT)/ ventricular fibrillation(VF), and cardiac arrest(CA) were contrasted in control group and magnesium group, respectively. Rat cardiomyocytes were dissociated with collagenase to acquire single ventricular myocytes. Sodium channel currents(I_ Na) were recorded using whole-cell recording patch clamp technique. The influences of aconitine and magnesium sulfate on I_ Na were studied. Results: The doses of aconitine to produce VPB, VT/VF, and CA were significantly larger in control group than in magnesium group: 31.23±6.95 μg vs 24.67±5.25 μg; 45.00±7.93 μg vs 28.15±6.26 μg; 82.73±10.31 μg vs 75.47±11.26 μg; respectively. I_ Na current density were 90.57±11.25 pA/pF, 98.31±26.63 pA/pF, 73.10±14.25 pA/pF in control group, aconitine group and magnesium group, respectively. The date was significantly different in the three groups(P0.05). The Current-voltage(I-V) curve was shifted to downward and upward respectively in aconitine group and in magnesium group. Conclusion: Magnesium sulfate has antiarrhythmic effects on aconitine-induced arrhythmias in rats. One mechanism of its antiarrhythmic effects is related to inhibition of sodium channel current.
Catecholamine-induced Regulation in Vitro and Ex Vivo of Intralymphocyte Ionized Magnesium.

Abstract:Despite the importance of the adrenergic activity and of the metabolism of magnesium in some important cardiovascular pathologies, very little is known about how intracellular ionized magnesium (Mg i 2+ ) is regulated by catecholamines. We made an in-vitro study of the variations in the concentration of ionized magnesium in human lymphocytes using the fluorescent probe furaptra in response to different catecholamines. We also made an ex-vivo study of the changes in intracellular ionized magnesium in lymphocytes in 20 subjects with essential arterial hypertension, 10 treated with 120 mg/d of propranolol and 10 with placebo. Norepinephrine and isoproterenol significantly decrease Mg i 2+ and this effect is blocked by β-blockers but not by α-blockers. The EC 50 of the effect of norepinephrine is within the range of concentrations physiologically present in plasma. The substitution of extracellular sodium with choline blocks the decrease in intracellular ionized magnesium induced by norepinephrine, which leads us to suppose that the magnesium-reducing effect of catecholamines is a result of the activation of a Na+-Mg2+ exchanger. We were not able to demonstrate any change in intracellular ionized magnesium after 1 and 17 days of active treatment in essential hypertensives. The impossibility of demonstrating ex vivo the mechanism of catecholamine-mediated regulation that is evident in vitro is perhaps due to our experimental conditions or to substances which in vivo inhibit the action of the catecholamines on magnesium, such as insulin and/or glucose.
The effect of a thiadiazinone derived Ca2+ sensitizer on the responsiveness of Mg(2+)-ATPase to Ca2+ in myofibrils isolated from stunned and nonstunned porcine and human myocardium

Abstract:Previously, we showed, in an in situ porcine model, that the thiadiazinone derivative [+]EMD 60263, a putative Ca2+ sensitizer with minimal phosphodiesterase III inhibitory properties, increased contractility more profoundly in stunned than in nonstunned myocardium. The aim of the present investigation was to study the mechanism of action by determining the in vitro effects of [+]EMD 60263 on the Ca2+ responsiveness of the Mg(2+)-dependent ATPases of myofibrils and sarcoplasmic reticulum membrane vesicles, isolated from normal ventricle of swine and hypertrophic septum of cardiomyopathic patients. Contamination of the myofibrils with sarcoplasmic reticulum membranes was excluded by testing the effect of the sarcoplasmic reticulum Ca(2+)-pumping ATPase inhibitor thapsigargin. The plasma concentrations at which [+]EMD 60263 exerted its inotropic effect in the in situ porcine model were found to be submicromolar. [+]EMD 60263 stimulated concentration-dependently (1-10 microM) the submaximally activated Mg(2+)-ATPases (at pCa 6.1) of pig heart myofibrils. [+]EMD 60263 (10 microM) shifted the pCa50 of porcine myofibrillar Ca(2+)-stimulated, Mg(2+)-dependent ATPase from 6.00 +/- 0.05 to 6.67 +/- 0.05, whereas the [-]enantiomer EMD 60264 had no significant effect. Although the effect was much less at 1 and 3 microM, [+]EMD 60263 (10 microM) also stimulated maximal myofibrillar Mg(2+)-ATPase activity. The Hill coefficient, reflecting the steepness of the fitted pCa/Mg(2+)-ATPase curve at half-maximal activation, was not affected by [+]EMD 60263 (10 microM). [+]EMD 60263 (10 microM) had no effect on sarcoplasmic reticulum Ca(2+)-stimulated, Mg(2+)-dependent ATPase from swine heart. The thiadiazinone derivative [+]EMD 57033 (10 microM), but not its [-]enantiomer EMD 57439, had similar, although less potent, effects on pig heart myofibrillar Mg(2+)-ATPase activity as compared to [+]EMD 60263. [+]EMD 60263 (3 microM) produced a significantly larger leftward shift of the pCa2+/Mg(2+)-ATPase activity curve of myofibrils isolated from the stunned compared to the adjacent nonstunned myocardium (Delta pCa50s caused by the presence of [+]EMD 60263 amounted to +0.57 +/- 0.04 and +0.42 +/- 0.05, respectively) in the in situ porcine model. The effects of [+]EMD 60263 on myofibrillar Mg(2+)-ATPase of hypertrophic human heart were identical to those observed with porcine heart myofibrils. The results indicate that the positive inotropic action of [+]EMD 60263 observed in the in situ porcine model of stunned myocardium may be primarily due to myofilament sensitization to Ca2+, and that this compound may have a similar action on diseased human myocardium.
[Effect of Pinacidil on Rat Myocardial Calcium Regulation].

Abstract:OBJECTIVE:To understand the effect of pinacidil on rat myocardial Ca(2+)regulation.METHODS:After baseline measurement and a period of equilibrium, myocytes were randomly allocated to one of 4 treatment groups: Control group (8 myocytes): incubation in Lactate Ringer's solution at 24 degrees C for 2 hours; K group (8 myocytes): incubation in Lactate Ringer's solution containing 16 mmol/L potassium at 24 degrees C for 2 hours; K+P group (8 myocytes): incubation in Lactate Ringer's solution containing potassium 16 mmol/L and pinacidil 50 micromol/L at 24 degrees C for 2 hours; K+P+G group (8 myocytes): incubation in Lactate Ringer's solution containing potassium 16 mmol/L, pinacidil 50 micromol/L and glibenclamide 10 micromol/L at 24 degrees C for 2 hours. After each incubation, myocytes were resuspended in cell culture media at the same temperature and intracellular [Ca(2+)](i) and SR Ca(2+) release were measured.RESULTS:The amplitude percent of [Ca(2+)](i) transient evoked by electrical stimulation in the K group was significantly decreased to 67.05% - 80.11% compared to 90.27% - 95.57% in the K+P group during reperfusion after ischemia (P<0.01). The percent amplitude of the [Ca(2+)](i) transient evoked by the rapid application of 10 mmol caffeine in the K group myocyte was approximately 112.00%+/-16.93% compared with that of the [Ca(2+)](i) transient evoked by electrical stimulation. However, in the K+P group myocyte the peak amplitude of the caffeine induced Ca(2+) release was 173.15%+/-26.01% compared with electrical stimulation (P<0.01). The duration of transient evoked by caffeine in K+P group (3.20+/-0.71 ms was significantly shorter than that in K group (3.93+/-0.46) ms (P<0.05).CONCLUSION:Cardioplegic arrest with simultaneous activation of KATP channels preserves rat myocardial Ca2+ by inducing sarcoplasmic reticulum Ca(2+) release and by alteration of Na(+)-Ca(2+) exchanger to better maintain [Ca(2+)](i) homeostasis.
High carbachol increases the electrically induced [Ca2+]i transient in the single isolated ventricular myocyte of rats

Abstract:In order to investigate the mechanisms responsible for the inotropic effects of muscarinic acetylcholine receptor stimulation by high concentrations of muscarinic receptor agonists, we studied the effects of carbachol at 30–300 μM on the electrically induced [Ca2+]i transient of rat isolated ventricular myocytes. Carbachol at this dose range increased the amplitude and duration of the electrically induced [Ca2+]i transient time and dose dependently. It also increased the resting fluorescence ratio and time to 80% decline of amplitude from the peak. At 100–300 μM the increase in [Ca2+]i transient was followed by a cluster of Ca2+ oscillations in 50–83% of the cells studied. The effects were blocked by atropine, but not pertussis toxin. Depletion of Ca2+ from sarcoplasmic reticulum by ryanodine, which itself reduced the amplitude of the [Ca2+]i transient and increased resting fluorescence, abolished the effect of carbachol on the [Ca2+]i transient without affecting its effect on resting fluorescence ratio. The caffeine-induced [Ca2+]i transient was unaffected by prior addition of carbachol in a Ca2+ free and low Na+ solution. Inhibition of Ca2+ influx by the L-type Ca2+ channel blocker, verapamil, which itself reduced the amplitude of the [Ca2+]i transient without affecting the resting fluorescence ratio, attenuated the augmentation of the amplitude of the [Ca2+]i transient elicited by carbachol. Ni2+, a non-specific Ca2+ channel blocker and an inhibitor of Na+Ca2+ exchange, abolished the effects of carbachol on both [Ca2+]i transient and resting fluorescence ratio. Low external Na+, which increased the resting fluorescence ratio due to its inhibitory effect on Na+Ca2+ exchange, also abolished the effects of carbachol. The results indicate that the inotropic effect of muscarinic acetylcholine receptor stimulation by high concentrations of a muscarinic receptor agonist may be due to an increase in the electrically induced [Ca2+]i transient in ventricular myocytes via a process which is not pertussis toxin sensitive. The increase in the electrically induced [Ca2+]i transient may result from increases in Na+Ca2+ exchange and influx of Ca2+ via voltage-gated Ca2+ channels, and mobilization of Ca2+ from the intracellular store. The mobilization of Ca2+ from the intracellular store is a secondary event. The study has provided evidence for the first time that muscarinic acetylcholine receptor stimulation by high concentrations of carbachol increases Ca2+ influx via the Ca2+ channel and mobilization of Ca2+ from its intracellular store. The study has also demonstrated for the first time the occurrence of Ca2+ oscillations induced by high concentrations of carbachol.
Effects of benactyzine on action potentials and contractile force of guinea pig papillary muscles

Abstract:Aim: To explore the effects of benactyzine (BEN) on the action potential and contractile force in guinea pig papillary muscles. Methods: Conventional microelectrode technique was used to record the fast action potentials (FAP) and slow action potentials (SAP) of guinea pig papillary muscles. Results: Benactyzine 5, 10, 50 μmol·L -1 suppressed the maximal upstroke velocity ( v max ) of FAP and contractile force (Fc) concentration dependently while prolonged the action potential duration at 50%, 90% repolarization (APD 50 , APD 90 ) and effective refractory period (ERP) of FAP. The suppression on the v max was frequency dependent. Benactyzine 5, 10,50 μmol·L -1 lengthened the APD50, APD90 of SAP induced by isoprenaline or histamine when perfused with KCl 22 mmol·L -1 Tyrode's solution. The v max of the SAP was not decreased by benactyzine 5, 10 μmol·L -1 but by 50 μmol·L -1 . The effects on the SAP were antagonized by elevation of the extracellular calcium from 2.0 to 5.6 mmol·L -1 . The effects of benactyzine on SAP elicited by tetrodotoxin resembled that by isoprenaline or histamine except the more pronounced suppression on v max and action potential amplitude (APA). The persistent rapid spontaneous activity and triggered tachyarrhythmia induced by ouabain were also abolished immediately by benactyzine 5 μmol·L -1 . Conclusion: Benactyzine can inhibit Na +, K +, Ca 2+ transmembrane movement and intracellular Ca 2+ mobilization in the myocardium, and this may be the electrophysiological basis of its effects against experimental arrhythmias.
Electrophysiological Mechanism of Catestatin Antiarrhythmia: Enhancement of Ito, IK, and IK1 and Inhibition of ICa-L in Rat Ventricular Myocytes

Abstract:Background: Cardiovascular disease remains one of the leading causes of death globally. Myocardial ischemia and infarction, in particular, frequently cause disturbances in cardiac electrical activity that can trigger ventricular arrhythmias. We aimed to investigate whether catestatin, an endogenous catecholamine-inhibiting peptide, ameliorates myocardial ischemia-induced ventricular arrhythmias in rats and the underlying ionic mechanisms. Methods and results: Adult male Sprague-Dawley rats were randomly divided into control and catestatin groups. Ventricular arrhythmias were induced by ligation of the left anterior descending coronary artery and electrical stimulation. Action potential, transient outward potassium current, delayed rectifier potassium current, inward rectifying potassium current, and L-type calcium current (ICa-L) of rat ventricular myocytes were recorded using a patch-clamp technique. Catestatin notably reduced ventricular arrhythmia caused by myocardial ischemia/reperfusion and electrical stimulation of rats. In ventricular myocytes, catestatin markedly shortened the action potential duration of ventricular myocytes, which was counteracted by potassium channel antagonists TEACl and 4-AP, and ICa-L current channel agonist Bay K8644. In addition, catestatin significantly increased transient outward potassium current, delayed rectifier potassium current, and inward rectifying potassium current density in a concentration-dependent manner. Catestatin accelerated the activation and decelerated the inactivation of the transient outward potassium current channel. Furthermore, catestatin decreased ICa-L current density in a concentration-dependent manner. Catestatin also accelerated the inactivation of the ICa-L channel and slowed down the recovery of ICa-L from inactivation. Conclusions: Catestatin enhances the activity of transient outward potassium current, delayed rectifier potassium current, and inward rectifying potassium current, while suppressing the ICa-L in ventricular myocytes, leading to shortened action potential duration and ultimately reducing the ventricular arrhythmia in rats.
Effects of Cordyceps Cephalosporium Mycelia Extracts on Myocardial Ischemia and Transmenbrance Action Potential of Ventricle Myocytes

Abstract:OBJECTIVE To investigate the effect of Cordyceps Cephalosporium Mycelia(CCM) extracts on electrophysiological characteristic of endocardium from guinea pig ventricle,and explore its mechanism aganist arrhythmia.METHODS The ECG was recorded in rat suffered from pituitrin-induced myocardial ischemia.Standard microelectrode technique was used to record the transmembrane action potential of endocardium of guinea pig ventricular myocytes.And the effect of CCM extracts of different concentrations on action potential duration(APD) of endocardium at basic sitmulate 1.0 Hz was evaluated.RESULTS The CCM extracts significantly inhibited changes of T wave in ECG,and reduced the incidence rate of arrhythmia induced by pituitrin.The CCM extracts delayed the amplitude of action potential(APA) and maximal velocity(Vmax),and prolonged the APD50 and APD90 in a dose-dependent manner in guinea pig.The CCM extracts had no effect on resting potential(RP).CONCLUSION The CCM extracts could protect against pituitrin-induced antiarrhythmia,the mechanism might be involved in inhibition of APA and Vmax and prolonging APD50 and APD90.
Effects of Magnesium Potassium Aspartate a nd Its Combination With Propafenone on Ventricular Electrophysiology in Rabbits.

Abstract:To investigate the effects of potassium magnesium aspartate (KMA) and I ts combination with propafenone (Pr) on heart eletrophysiology,Epicardium contac t monophasic action potential (MAP) probe was used to record MAP on rabbit's lef t ventricle after thoracotomy.The rabbits were randomized into several groups:KM A,Prand KMA+Pr.The drugs were administered by intravenous injection.Results:KMA ,Pr and KMA+Pr prolonged SCL,MAPD50 and increased ventricular effective re fractory period (VERP).Pr and KMA+Pr prolonged MAPD90 and increased VERP/M APD90.There was proarrhythmic effects in group Pr of higher dose.After com bination of KMA with Pr of higher dose,the incidence of proarrhythmia was increa sed.Conclusion:KMA has electrophysiologic characters of antiarrhythmic agents and the administration of KMA in recommended dose is safe.KMA and Pr increased the effects each other after their combination.The dose of Pr should be decreas ed when Pr combine KMA for decreasing the risk of proarrhythmia.

Abstract:The synergy between Na+-K+ pumps, Na+-Ca2+ exchangers, membrane currents and the sarcoplasmic reticulum (SR) generates the coupled-clock system, which governs the spontaneous electrical activity of heart sinoatrial node cells (SANCs). Ca2+ mediates the degree of clock coupling via local Ca2+ release (LCR) from the SR and activation of cAMP/PKA signaling. Marinobufagenin (MBG) is a natural Na+-K+ pump inhibitor whose effect on SANCs has not been measured before. The following two hypotheses were tested to determine if and how MBG mediates between the Na+-K+ pump and spontaneous SAN activity: (i) MBG has a distinct effect on beat interval (BI) due to variable effects on LCR characteristics, and (ii) Ca2+ is an important mediator between MBG and SANC activity. Ca2+ transients were measured by confocal microscopy during application of increasing concentrations of MBG. To further support the hypothesis that Ca2+ mediates between MBG and SANC activity, Ca2+ was chelated by the addition of BAPTA. Dose response tests found that 100 nM MBG led to no change in BI in 6 SANCs (no BI change group), and to BI prolongation in 10 SANCs (BI change group). At the same concentration, the LCR period was prolonged in both groups, but more significantly in the BI change group. BAPTA-AM prolonged the BI in 12 SANCs. In the presence of BAPTA, 100 nM MBG had no effect on SANC BI or on the LCR period. In conclusion, the MBG effects on SANC function are mediated by the coupled clock system, and Ca2+ is an important regulator of these effects.
Alpha1-adrenoceptor-mediated negative inotropic effect caused by intracellular ionic activities in guinea-pig papillary muscle

Abstract:In guinea-pig papillary muscle, phenylephrine (PE), an agonist of alpha1-adrenoceptor (alpha1-AR), led to a transient negative inotropic effect (NIE) and a subsequent sustained positive inotropic effect (PIE). To clarify the ionic mechanisms underlying the NIE, we measured the [Na+]i or [pH]i by ion-selective microelectrodes. PE produced a decrease in the intracellular Na+ concentration ([Na+]i) and an increase in intracellular pH ([pH]i). During the phase of NIE, PE produced only a (-) change of [Na+]i (Delta[Na+]i). With a decrease in extracellular Na+ or an increase in extracellular Ca2+, the PE-induced NIE was attenuated and PE produced (+)Delta[Na+]i. The PE-induced NIE and (-)Delta[Na+]i were definitely strengthened by lowering the bath temperature or increasing the stimulation frequency. 2-(2,6-di-methoxyphenoxyethyl)amino-methyl-1,4-benzidioxane HCl, an antagonist of alpha1A-AR, completely abolished the PE-induced NIE and (-)Delta[Na+]i. Phorbol 12,13-dibutyrate, an activator of protein kinase C (PKC), decreased the baseline [Na+]i and twitch force and increased the baseline [pH]i in mimicry of PE. Pretreatment with 1-5(isoquinolinesulphonyl)-2-methylpiperazine, an inhibitor of PKC, abolished the PE-induced NIE and (-)Delta[Na+]i. During pretreatment with benzamil, an inhibitor of Na+/Ca2+ exchange, we found that the PE-induced NIE and (-)Delta[Na+]i were reversibly abolished. Our results indicate the PE-induced NIE may be elicited upon the activation of Na+/Ca2+ exchange which can be attributed to the (-)Delta[Na+]i. (-)Delta[Na+]i is mediated through a PKC-dependent pathway via an activation of alpha1A-AR subtype and its effect could be strengthened remarkably at high [Na+]i and [Ca2+]i values.

Abstract:1. The leucocyte-mediated inflammatory response plays a pivotal role in ischaemia-reperfusion injury. P-Selectin and CD11b are important mediators for the recruitment of leucocytes into the endothelium. However, the time-course of changes in P-selectin and CD11b expression during reperfusion is not defined. Magnesium has been shown to have a protective role in reperfusion injury, but the mechanism of its action is not yet clear.2. In the present study, 90 male Sprague-Dawley rats were randomized into three groups, namely a sham-operated group, an IR group (subjected to 45 min of coronary occlusion followed by reperfusion for up to 6 h) and an MgSO4-treated group, which received an infusion of MgSO4 starting 30 min before occlusion and continuing until the onset of reperfusion. The expression of P-selectin on platelets and CD11b on leucocytes, endothelial P-selectin on cardiac microvessels and P-selectin mRNA in myocardial tissue were measured by flow cytometry, immunohistochemistry and reverse transcription-polymerase chain reaction, respectively, at different time points during reperfusion. The degree of myocardial necrosis and arrhythmia was assessed.3. The present study shows that P-selectin expression on platelets and cardiac microvessels, P-selectin mRNA in myocardial tissue and CD11b expression on leucocytes in the IR group were significantly upregulated compared with the sham-operated group in a time-dependent manner during reperfusion. Treatment with MgSO4 attenuated the upregulation of P-selectin expression, in addition to inhibiting myocardial necrosis and arrhythmia, but had no effect on CD11b expression on leucocytes.4. These results suggest that the expression of P-selectin and CD11b is upregulated after reperfusion and magnesium pretreatment plays a cardioprotective role in ischaemia-reperfusion injury, possibly by inhibiting the upregulation of P-selectin expression.
Electrophysiological Effects of Protopine in Cardiac Myocytes: Inhibition of Multiple Cation Channel Currents

Abstract:Protopine (Pro) from Corydalis tubers has been shown to have multiple actions on cardiovascular system, including anti-arrhythmic, anti-hypertensive and negative inotropic effects. Although it was thought that Pro exerts its actions through blocking Ca(2+) currents, the electrophysiological profile of Pro is unclear. The aim of this study is to elucidate the ionic mechanisms of Pro effects in the heart. In single isolated ventricular myocytes from guinea-pig, extracellular application of Pro markedly and reversibly abbreviates action potential duration, and decreases the rate of upstroke (dV/dt)(max), amplitude and overshoot of action potential in a dose-dependent manner. Additionally, it produces a slight, but significant hyperpolarization of the resting membrane potential. Pro at 25, 50 and 100 microM reduces L-type Ca(2+) current (I(Ca,L)) amplitude to 89.1, 61.9 and 45.8% of control, respectively, and significantly slows the decay kinetics of I(Ca,L) at higher concentration. The steady state inactivation of I(Ca,L) is shifted negatively by 5.9 - 7.0 mV (at 50 - 100 microM Pro), whereas the voltage-dependent activation of I(Ca,L) remains unchanged. In contrast, Pro at 100 microM has no evident effects on T-type Ca(2+) current (I(Ca,T)). In the presence of Pro, both the inward rectifier (I(K1)) and delayed rectifier (I(K)) potassium currents are variably inhibited, depending on Pro concentrations. Sodium current (I(Na)), recorded in low [Na(+)](o) (40 mM) solution, is more potently suppressed by Pro. At 25 microM, Pro significantly attenuated I(Na) at most of the test voltages (-60 approximately +40 mV, with a 53% reduction at -30 mV. Thus, Pro is not a selective Ca(2+) channel antagonist. Rather, it acts as a promiscuous inhibitor of cation channel currents including I(Ca,L), I(K), I(K1) as well as I(Na). These findings may provide some mechanistic explanations for the therapeutic actions of Pro in the heart.
Effects of Bromibenzyltetrahydraberberine on Ion Channels in Guinea Pig Ventricular Myocytes

Abstract:To investigate the effects of bromibenzyltetrahydraberberine on delayed rectifier potassium current and inward rectifier potassium current and L-type calcium current in isolated single guinea pig ventricular myocyte. Single ventricular cell was obtained from healthy guinea pig hearts using enzymatic dissociation technique. The whole cell patch clamp method was used to record the delayed rectifier potassium current(I K), the inward rectifier potassium current(I K1), and the L-type calcium current (I Ca-L). Results: Bromibenzyltetrahydraberberine has dose-dependent blocking effects on I K.ID 50 is 56μmol/L. It has no change on I-V shape of I K. It has no obvious effect on I K1. Bromibenzyltetrahydraberberine has dose-dependant blocking effect on I Ca-L .ID 50 is 65μmol/L. The inhibition of bromibenzyltetrahydraberberine on I Ca-L was dependent of the holding potential (HP).75μmol/L bromibenzyltetrahydraberberine inhibited I Ca-L by 0.4922±0.021 at HP=-40 mV, and 0.6377±0.0366 at HP=-80 mV. Conclusion: Bromibenzyltetrahydraberberine can decrease delayed rectifier potassium current and L-type calcium current in single guinea pig ventricular myocyte. It is a muti-ion blocking antirarrhythmia agent.

Abstract:Objective: To investigate the mechanism of adenosine antagonized isoproterenol on action potentials and inward rectifierpotassium channels in guinea pig papillary muscles and ventricular myocytes, and to demonstrate the ionic current basic inthe treatment with adenosine. Methods:Standard glass microelectrode method and whole cell patch clamp techniques wereused to study the effect of adenosine on action potential and Ica-L, IKl. Results: ISO prolonged APD, urged EAD, delayed DADand triggered activity, and increased ICa-L and IKi. APD prolongation caused by ISO, and Iki, and Ica-L were inhibited byADO. The effect of ADO was antagonized by DPCPX. Conclusion: Openings of IKl may shorten APD with the protection formyocytes. There is a correlation between the effect of ADO to APE caused by the antagonized effect to ISO and Ica-L and Iklinhibition.
Activity-induced facilitation of L-type Ca2+ current in cardiomyocytes isolated from guinea-pig ventricles

Abstract:The facilitation of L-type Ca2+ current (ICa), which sometimes occurs with an increase in stimulation frequency, was investigated in single guinea-pig ventricular cardiomyocytes using whole-cell recording and K(+)-free solutions. With a holding potential of -80 mV, an increase in frequency from 0.5 to 1, 2, 3 or 4 Hz caused either a small or large initial reduction, or a transient enhancement (facilitation) of peak ICa, which developed rapidly and was followed by a reduction of ICa. Reducing the frequency to 0.1 or 0.2 Hz caused a depression of ICa on the first pulse that was followed by a slower increment. Transient facilitation and depression were entirely absent when either Ba2+ or Na+ was used as the charge carrier in Ca(2+)-free solutions. High concentrations of isoprenaline (1-3 microM), forskolin (1-3 microM), or 8-(4-chlorophenylthio)-cAMP (150 microM) suppressed but did not abolish the incidence and size of facilitation; employing a holding potential of -40 mV also suppressed the incidence of ICa facilitation. Lower isoprenaline concentrations (0.1 and 0.3 microM) greatly enhanced the incidence and magnitude of the transient facilitation occurring with an increase in stimulation rate, but did not diminish the magnitude of the ensuing reduction. When facilitation occurred with 0.1 mM EGTA in the dialysate, or the usual 5 mM EGTA with 0.01 microM extracellular isoprenaline, it developed more slowly after an increase in frequency. In the presence of the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine, an increase in stimulation rate from 0.5 to 1, 2 or 3 Hz sometimes caused a large and sustained facilitation of ICa, which developed over tens of seconds, declined slowly with continued stimulation and was maintained after returning to 0.5 Hz. It is concluded that the levels of intracellular cAMP and Ca2+ mediate the initial sensitivity of ICa to changes in stimulation rate, to the extent that they determine whether or not transient facilitation will occur. Because the reduction of ICa was relatively constant, facilitation dictates the level of steady-state ICa that will be reached at the higher rate. Taken together with the fact that facilitation can be modulated, the results argue for separate mechanisms of facilitation and reduction for ICa. It is suggested that the mechanism of facilitation is partly enzymatic, insofar as sustained facilitation could be a manifestation of a stimulatory Ca(2+)-dependent process, which is normally counteracted by the action of phosphodiesterases.(ABSTRACT TRUNCATED AT 400 WORDS)
[Effects of Potassium Channel Activators on Transient Inward Current in Guinea Pig Ventricular Myocytes].

Abstract:OBJECTIVE:To investigate the mechanism of ATP-sensitive potassium channel (K(ATP)) activator cromakalim (CRK) on action potentials and transient inward current (I(ti)) in isolated guinea pig papillary and ventricular myocytes and to explore the mechanisms of effects of I(ti) and K(ATP) treatment in idiopathic ventricular tachycardia.METHODS:The whole-cell patch clamp recording technique was used to detect the action potentials and I(ti) and K(ATP) current alterations during the stimulated and triggered activity. Myocytes were isolated from guinea pig ventricle by enzyme digestion. The experiment was divided into four groups: (1) Control; (2) Control + Ouabain; (3) Control + CRK; (4) Control + Ouabain + CRK. (5) Control + Ouabain + CRK + glibenclamide (GLB). The action potential of guinea pig papillary muscles was measured by using standard microelectrode. The parameters in the experiment included the amplitude (APA), resting potentials (RP), action potentials duration (APD), as well as maximum rise of the action potential (Vmax).RESULTS:(1) When the guinea pig ventricular papillary myocytes were pretreated with Ouabain 0.5 micromol/L, APD prolonged significantly, especially APD(20), APD(50), APD(90). Delayed after depolorazion (DAD) and triggered activity were elicited. I(ti) currents and DAD as well as triggered activity increased. I(ti) current was (126.9 +/- 10.8) pA, lagT (1173.0 +/- 70.9) ms (n = 10, P < 0.01). (2) When guinea pig ventricular myocytes were pretreated with CRK (10 micromol/L), APD was shortened and the amplitude of DAD was lowered. The coupling time in CRK group was significantly prolonged compared with Ouabain group (n = 10, P < 0.01). (3) CRK 50 micromol/L pretreatment of the ventricular myocytes led to an increase of K(ATP) up to (342 +/- 89) pA, which was statistically significant as compared with the control group (P < 0.01). ATP-sensitive potassium channel blocker glibenclamide (10 micromol/L) could antagonize the effects of CRK on APD and I(ti) currents.CONCLUSION:CRK might reduce the toxic effect of Ouabain on cardiomyocytes, shorten APD, terminate DAD and trigger excitation, and have protective effect on cardiomyocytes. The effects of CRK, may be associated with the inhibiting I(ti) current and increasing K(ATP).

Abstract:Abstract The β-agonist isoproterenol (ISO) reduces the Na/K pump current (Ip) via β-adrenergic receptors when the intracellular calcium concentration ([Ca2+]i) is below 150 nM [8]. In the present study, the intracellular signaling pathway was investigated with whole-cell patch-clamp of isolated guinea pig ventricular myocytes. The inhibitory effect of ISO could be mimicked by external application of the membrane-permeant cAMP analog chlorophenylthio-cAMP (0.5 mM), the phosphodiesterase inhibitor isobutyl-1-methylxanthine (IBMX, 100 μM), or the adenylyl cyclase activator forskolin (50 μM). Intracellular application of the synthetic peptide inhibitor of protein kinase A (PKA), PKI (5 μM), prevented the effect of ISO. These results suggest that the inhibitory effect of ISO on Ip is mediated via a phosphorylation step induced by a cAMP-dependent PKA pathway. Neither the non-specific protein kinase inhibitor H7 (100 μM) nor the protein phosphatase inhibitor calyculin A (0.5 μM) had any effect on Ip in the absence of ISO. However, H7 could increase Ip and calyculin A could reduce it in the presence of ISO (1 μM and 12 nM respectively). These results indicate that there is a low basal level of phosphorylation which makes the effects of H7 and calyculin A difficult to detect in the absence of an ISO-induced increase in phosphorylation level.
