Abstract:Increasing temperatures and extended drought episodes are among the major constraints affecting food production. Maize has a relatively high temperature optimum for photosynthesis compared to C 3 crops, however, the response of this important C 4 crop to the combination of heat and drought stress is poorly understood. Here, we hypothesized that resilience to high temperature combined with water deficit (WD) would require efficient regulation of the photosynthetic traits of maize, including the C 4 –CO 2 concentrating mechanism (CCM). Two genotypes of maize with contrasting levels of drought and heat tolerance, B73 and P0023, were acclimatized at high temperature (38°C versus 25°C) under well-watered (WW) or WD conditions. The photosynthetic performance was evaluated by gas exchange and chlorophyll a fluorescence, and in vitro activities of key enzymes for carboxylation (phosphoenolpyruvate carboxylase), decarboxylation (NADP-malic enzyme), and carbon fixation (Rubisco). Both genotypes successfully acclimatized to the high temperature, although with different mechanisms: while B73 maintained the photosynthetic rates by increasing stomatal conductance (gs), P0023 maintained gs and showed limited transpiration. When WD was experienced in combination with high temperatures, limited transpiration allowed water-savings and acted as a drought stress avoidance mechanism. The photosynthetic efficiency in P0023 was sustained by higher phosphorylated PEPC and electron transport rate (ETR) near vascular tissues, supplying chemical energy for an effective CCM. These results suggest that the key traits for drought and heat tolerance in maize are limited transpiration rate, allied with a synchronized regulation of the carbon assimilation metabolism. These findings can be exploited in future breeding efforts aimed at improving maize resilience to climate change.

Abstract:The combination of low precipitation and high temperature stresses at jointing stage can severely threaten maize production. However, to date, few studies have been conducted on the effects of combined stress on maize plants expression at jointing stage. In the current research, plant growth, root morphology, and yield components were determined after exposure to the single and combined stress of high temperature and drought stress. Leaf gas exchange, malondialdehyde (MDA) content and antioxidant enzymes activities were conducted to identify potential mechanisms of stress responses. The single stress of high temperature and drought significantly reduced the biomass of various organs and the total aboveground biomass, which reduced the yield of maize plants. High temperature substantially decreased aboveground biomass and yield under mild and severe water stress, which indicated that the inhibitory effects of combined stress were more significant than that of high temperature or drought individually. High temperature exacerbated the negative impacts of water stress on plants growth and yield as shown by the reduced leaf photosynthetic rate ( P n ), probably related to the increasing MDA content. Leaf-level water use efficiency ( WUE ) was enhanced as the reduction in leaf transpiration ( T r ) was greater than the decrease in leaf photosynthesis under high temperature, even for those plants were suffering water stress. High temperature, drought stress and their combination all greatly increased the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), but were ineffective in mitigating oxidative damage. The MDA content and antioxidant enzymes activities showed an increasing trend following 12 days of combined stress. This substantiated the irreversible damage induced by combination of high temperature and desiccation stresses. The combined stress optimized roots length, root volume, root surface area, and thinned the average root diameter, which improved the adaptation of maize to high temperature, drought and combined stress. This study has provided meaningful references for improved understanding the impacts of drought, high temperature, and concurrent events on the physiology and growth of maize plants during the jointing period.

Abstract:Maize is a sensitive crop to drought and heat stresses, particularly at the reproductive stages of development. The present study investigated the individual and interactive effects of drought (50% field capacity) and heat (38 °C/30 °C) stresses on morpho-physiological growth, yield, nutrient uptake and oxidative metabolism in two maize hybrids i.e., ‘Xida 889’ and ‘Xida 319’. The stress treatments were applied at tasseling stage for 15 days. Drought, heat and drought + heat stress caused oxidative stress by the over-production of ROS (O2−, H2O2, OH−) and enhanced malondialdehyde contents, which led to reduced photosynthetic components, nutrients uptake and yield attributes. The concurrent occurrence of drought and heat was more severe for maize growth than the single stress. However, both stresses induced the metabolites accumulation and enzymatic and non-enzymatic antioxidants to prevent the oxidative damage. The performance of Xida 899 was more prominent than the Xida 319. The greater tolerance of Xida 889 to heat and drought stresses was attributed to strong antioxidant defense system, higher osmolyte accumulation, and maintenance of photosynthetic pigments and nutrient balance compared with Xida 319.

Recent research highlights the potential of water replenishment to ameliorate grain weight loss. However, the mitigating mechanisms of heat in drought stress, especially during the crucial early grain‐filling stage, remain poorly understood. We investigated the mechanism for mitigating heat in drought stress by water replenishment from the 12th to the 32nd days after silking in a controlled greenhouse experiment (Exp. I) and field trial (Exp. II). A significant reduction in grain weight was observed in heat stress compared to normal conditions. When water replenishment was applied to increase soil water content (SWC) under heat stress, the grain yield exhibited a notable increase ranging from 28.4 to 76.9%. XY335 variety was used for transcriptome sequencing to analyze starch biosynthesis and amino acid metabolisms in Exp. I. With water replenishment, the transcripts of genes responsible for trehalose 6‐phosphate phosphates (TPP), alpha‐trehalase (TRE), ADP‐glcpyrophosphorylase, and starch synthase activity were stimulated. Additionally, the expression of genes encoding TPP and TRE contributed to an enhanced conversion of trehalose to glucose. This led to the conversion of sucrose from glucose‐1‐phosphate to ADP‐glucose and ADP‐glucose to amylopectin, ultimately increasing starch production by 45.1%. Water replenishment to boost SWC during heat stress also elevated the levels of essential amino acids in maize, including arginine, serine, tyrosine, leucine, glutamic acid, and methionine, providing valuable support to maize plants in adversity. Field trials further validated the positive impact of water replenishment on SWC, resulting in a notable increase in grain yield ranging from 7.1 to 9.2%. This study highlights the vital importance of adapting to abiotic stress and underscores the necessity of developing strategies to counteract its adverse effects on crop yield.

Abstract:Heat stress affects physiological traits and biomass production in major crops, including maize. We researched the responses of maximum efficiency of photosystem II (Fv/Fm), relative cell injury (RCI), stomatal conductance (gs), internal CO2 concentration (Ci), leaf photosynthesis (CER), and crop growth rate (CGR) in two maize cultivars exposed to high temperatures around silking (R1) under field conditions. Temperature regimes (i.e. control and heat) were performed during the pre-silking (–15d R1 to R1) and post-silking (R1+2d to R1+17d) periods. In the heat treatments, polyethylene shelters were used in order to increase daytime temperatures around midday (from 10 A.M. to 2 P.M.) during each period (i.e., pre- and post-silking). In the control treatments, the shelters remained open during the entire growing season. Gas exchange variables, Fv/Fm and relative cell injury (RCI) were measured on ear leaves. CGR was estimated based on biomass samples. CER and Fv/Fm presented maximum reductions at the end of the daytime heating. However, 30 min after the shelters were reopened, Fv/Fm of heated leaves reached values similar to controls, which were closely linked to CER recoveries. RCI was negatively associated with Fv/Fm, and cell injury increased gradually as heating continued. Ci was unaffected by heat treatment, indicating that gs was not the primary cause of CER reduction. Heat stress decreased CGR, and the reduction was positively associated with CER and Fv/Fm in both heating periods. We attempted to scale from cell to crop level and identify some physiological traits that could be helpful in breeding programs for heat stress tolerance.

Abstract:The objective of this study was to explore the changes in leaf nitrogen (N) and phosphorus (P) content, physiological processes, growth, and resource-use efficiency in a rapeseed cultivar under drought and (or) high temperatures. The two-wk-old plants were grown under controlled growth chamber conditions and subjected to individual or combined water (well-irrigated, 88% ± 4% field capacity vs. drought, 46% ± 5% field capacity) and temperature (control, 23 °C/17 °C vs. high temperatures, 26 °C/25 °C) regimes for 10 d. The measured response parameters were [N], [P], and their total content in leaves, photosynthetic rate (A sat ), stomatal conductance (g s ), intercellular CO 2 concentration (C i ), mitochondrial respiration (R), intrinsic water use efficiency (WUE i ), photosynthetic N use efficiency (PNUE), relative growth rate of the root (RGR r ) and shoot (RGR s ), leaf area, and dry matter accumulation in the plant. Drought significantly decreased [N], [P], and their total content in leaves, A sat , g s , C i , RGR r , RGR s , leaf area, dry matter accumulation in the root, shoot, and whole plant, and PNUE, but significantly increased R and WUE i . Drought-induced reduction in growth or A sat was mainly attributed to a decreased C i due to stomatal closure, while reduction in g s and leaf area appeared to be a drought-adaptive mechanism. High temperature stress alone had no negative impact on physiological and growth parameters, indicating an enhanced thermal stability of the cultivar, which was diminished at combined drought and high temperature stresses. We therefore conclude that the thermal stability of the cultivar in terms of growth was compromised under simultaneous occurrence of drought and high temperature stresses.

Abstract:The occurrence of heat often exacerbates drought stress by increasing atmospheric vapor pressure deficit and decreasing transpiration efficiency. Water replenishment to increase soil water content (SWC) could be an effective measure for improving crop resilience in heat. However, field observational evidence and the underlying mechanism for the mitigation were still limited. For this, two widely adopted maize hybrids (ZD958 and XY335) were planted in 2019 and 2020 in North China Plain with three growth environments during the reproductive stage: Normal field conditions (NC), High-temperature episodes (HT) and High-temperature together with elevating SWC (HT-W). Averagely, high temperature reduced grain yield by 21.6-65.1% in HT treatment compared with NC treatment. In HT-W treatment, grain yield was improved by 11.8-13.5% while SWC was elevated by 27.6–51.5% in 0–80 cm soil profile compared with HT treatment. In HT-W treatment, water replenishment to maize under heat stress improved canopy temperature and grain filling traits. The daily maximum canopy temperature was reduced by 1.0–3.1 °C. The effective grain-filling period (P f ) was extended by 1.0–1.6 days and the mean grain-filling rate (G mean ) was increased by 8.2%-10.5%. The improvement of G mean and P f for ZD958 was greater than XY335. Furthermore, the significant correlations between SWC in 0–20 cm soil profile and G mean , and between SWC in 40–80 cm soil profile and P f for both hybrids were observed. As a result, the fresh weight, volume, and water content of the grain were significantly improved in HT-W treatment compared with HT treatment. Overall, per unit increase in SWC (0–80 cm) decreased the daily maximum canopy temperature by 0.3 °C, increased dry matter accumulation by 1.5%, and grain yield by 2.1%. The information obtained from this study will be helpful to develop efficient management to protect against heat stress in the field.

Abstract:High temperature around flowering has a serious impact on the growth and development of maize. However, few maize genes related to flowering under heat stress have been confirmed, and the regulatory mechanism is unclear. To reveal the molecular mechanism of heat tolerance in maize, two maize hybrids, ZD309 and XY335, with different heat resistance, were selected to perform transcriptome and metabolomics analysis at the flowering stage under heat stress. In ZD309, 314 up-regulated and 463 down-regulated differentially expressed genes (DEGs) were detected, while 168 up-regulated and 119 down-regulated DEGs were identified in XY335. By comparing the differential gene expression patterns of ZD309 and XY335, we found the "frontloaded" genes which were less up-regulated in heat-tolerant maize during high temperature stress. They included heat tolerance genes, which may react faster at the protein level to provide resilience to instantaneous heat stress. A total of 1062 metabolites were identified via metabolomics analysis. Lipids, saccharides, and flavonoids were found to be differentially expressed under heat stress, indicating these metabolites' response to high temperature. Our study will contribute to the identification of heat tolerance genes in maize, therefore contributing to the breeding of heat-tolerant maize varieties.

Effects of High Temperature and Drought Stresses on Growth and Yield of Summer Maize during Grain Filling in North China

Abstract:High sensitivity to climate change has been demonstrated in global maize production, especially the extreme high temperature and drought events. At present, the interactive effects of those extreme event stresses on maize growth at the grain filling stage are less well studied. In this study, a two-year field experiment was conducted to evaluate the compound effects of three stressors (high temperature, drought, and stress duration) at the filling phase on phenological period, grain filling rate, yield component, and yield of summer maize. The precipitation and temperature were controlled by using a rain-shelter systems and a warming system with infrared radiation lamps in field plots. The results indicated the significant influences of high temperature, drought stress, and the interaction on the growth and yield of summer maize. Under the individual factor of drought, compared to normal irrigation (NI), the significant decrease in grain-filling rate for mild drought (LD) and severe drought (SD) were 70.7% and 60.6%, respectively, while the decrease in grain yield for LD and SD were 80.1% and 93.6%, respectively. Under the individual factor of temperature, the consequences on grain-filling rate and grain yield were more severe at high temperature (temperature increase by 4.5 °C) than low temperature (temperature increase by 2.3 °C). The compound of high temperature and drought indicated that the grain yield increase by the compound effects of 3 °C for 5 days under normal irrigation (NIT3D5) and the compound effects of 3 °C for 7 days under mild drought (LDT3D7) were 3.5% and 10.9%, respectively, compared to without temperature stress. The grain yields were reduced by the other compound effects. The preferential temperature (temperature increase by 2.3 °C) promoted maize growth under normal irrigation and mild drought, while high temperature (temperature increase by 4.5 °C) significantly inhibited maize growth under normal irrigation and heavy drought. The warming climate is favorable to maize production within certain limits, and will provide a scientific basis for agricultural disaster prevention and mitigation.

Abstract:Drought and exposure to high temperature at flowering strongly affect maize production. However, previous studies mostly focused on the effects of the stressors on kernel number. The impacts of drought and heat in isolation and combination at flowering on maize kernel filling and final kernel weight have received limited attention. Here we investigated the effects of three stressors (drought, heat, and drought plus heat) at flowering phase (from tasseling to seed setting completed) on maize kernel filling in field site plots covered with a rainout shelter and under laboratory conditions with kernel in vitro culture. In two experiments, the kernel weight of plants exposed to flowering stressors was significantly decreased relative to controls. In the field experiment, exposure to stressors decreased kernel weight by 5.0–8.3% for the basal kernel and 5.6–11.0% for the apical kernel. Accelerated leaf senescence and decreased rate of photosynthesis were observed in the field. Kernel weight was also reduced by 18.0–37.6% in the laboratory with kernel in vitro culture, suggesting that early stressors at flowering led to a sink (kernel) limit. Carbohydrate metabolism and starch biosynthesis were impaired by drought and heat individually and their combination. During the early kernel filling stage, the activities of sucrose invertase, sucrose synthase, ADP-Glc pyrophosphorylase, and starch synthase were decreased at 10 and 20 days after pollination (DAP) in the stress treatment. For both basal and apical kernels, the correlation analyses indicated the mean kernel filling rate correlated positively with zeatin riboside content at DAP 10 and indole-3-acetic acid content at DAP 20. This study provides useful information to support the development of drought- and heat-tolerant hybrids and effective management strategies to improve crop yields.

Effects of high air temperature, drought, and both combinations on maize: A case study

Abstract:High air temperature (HAT) and natural soil drought (NSD) have seriously affected crop yield and frequently take place in a HAT-NSD combination. Maize (Zea mays) is an important crop, thermophilic but not heat tolerant. In this study, HAT, NSD, and HAT-NSD effects on maize inbred line Huangzao4 -were characterized. Main findings were as follows: H2O2 and O- accumulated much more in immature young leaves than in mature old leaves under the stresses. Lateral roots were highly distributed near the upper pot mix layers under HAT and near root tips under HAT-NSD. Saccharide accumulated mainly in stressed root caps (RC) and formed a highly accumulated saccharide band at the boundary between RC and meristematic zone. Lignin deposition was in stressed roots under NSD and HAT-NSD. Chloroplasts increased in number and formed a high-density ring around leaf vascular bundles (VB) under HAT and HAT-NSD, and sparsely scattered in the peripheral area of VBs under NSD. The RC cells containing starch granules were most under NAD-HAT but least under HAT. Under NSD and HAT-NSD followed by re-watering, anther number per tassel spikelet reduced to 3. These results provide multiple clues for further distinguishing molecular mechanisms for maize to tolerate these stresses.

Abstract:Climate warming-induced heat waves become common in food-producing regions worldwide, frequently coinciding with high temperature (HT) sensitive stages of many crops and thus threatening global food security. Understanding HT sensibility of reproductive organs is currently of great interest for increasing seed set. The responses of seed set to HT stress are multi-processes involving both male and female reproductive organs, but it currently lacks an integrated and systematic summary in the world's three leading food crops. This study defined the critical high temperature thresholds for seed set in rice (37.2 ± 0.2 °C), wheat (27.3 ± 0.5 °C) and maize (37.9 ± 0.4 °C) during flowering, and assessed the HT sensitivity in three cereals from the microspore stage to the lag filling, in relation to flowering dynamics, florets growth and development, and pollination and fertilization. Our review synthesized the existing knowledge on the effects of HT stress on spikelet opening, anther dehiscence, pollen shedding number, pollen viability, the function of pistils and stigma, pollen germination on stigma, and pollen tube elongation. HT-induced spikelet closure and arrest of pollen tube elongation have a catastrophic impact on pollination and fertilization in maize. Rice benefits from pollination under HT stress owing to the bottom anther dehiscence and cleistogamy. The cleistogamy and secondary spikelet opening increase the possibility of pollination success in wheat under HT stress. However, cereal crops also have themselves protective measures under HT stress. Temperature depression existing between air temperature and canopy/tissue temperature indicates cereal crops can partly protect themselves from heat damage especially for rice. In maize, husk leaves reduce ear inside temperature by ca. 5 °C compared to ear outside temperature, thereby protecting the latter phases of pollen tube growth and fertilization processes. This study has important implications for accurately modelling, optimizing crop management, and breeding varieties to cope with HT stress on the most important staple crops.

Abstract:Heat waves in combination with drought are predicted to occur more frequently with climate warming, yet their interactive effects on crop carbon and water balance are still poorly understood. Hence, research on the capacity of crops to withstand and recover from the combined stress is urgently needed. This study investigated the effects of drought and heat wave on a crop species as well as the recovery from the combined stress. Seedlings were grown in growth chambers under two soil water conditions (i.e. well watered and drought stress) at ambient temperature (26°C) for 10 days. Afterwards, half of the seedlings were exposed to a 7-day 42°C heat wave. All the drought-stressed seedlings were then rehydrated upon relief of the heat wave. Leaf gas exchange, the maximum carboxylation capacity (V cmax), plant growth, relative chlorophyll content and leaf water potential were examined during the experimental period. The heat wave reduced leaf gas exchange rates, V cmax and relative chlorophyll content, while it had no impacts on leaf water potential. In contrast, drought stress led to greater reductions in leaf gas exchange rates, growth and water potential than heat wave alone. Seedlings underwent a greater degree of stress in the combination of drought and heat wave than under the single drought treatment. The recovery of leaf gas exchange from drought stress lagged behind the water potential recovery and was delayed by heat wave. Our results show that drought stress had a predominant role in determining plant physiological responses and the negative impacts of drought stress were exacerbated by heat wave. The greater stress in the combination of drought and heat wave translated into the slower recovery of leaf gas exchange. Therefore, drought combined with heat wave may induce greater risks on crops under future climates.
Differential regulation of flower transpiration during abiotic stress in annual plants

Abstract:SUMMARYHeat waves occurring during droughts, can have a devastating impact on yield, especially if they happen during the flowering and seed set stages of the crop cycle. Global warming and climate change are driving an alarming increase in the frequency and intensity of combined drought and heat stress episodes, critically threatening global food security.Because high temperature is detrimental to reproductive processes, essential for plant yield, we measured the inner temperature, transpiration, sepal stomatal aperture, hormone levels, and transcriptomic response of closed soybean flowers, developing on plants subjected to a combination of drought and heat stress.Here, we report that during a combination of drought and heat stress soybean plants prioritize transpiration through flowers over transpiration through leaves by opening their flower stomata, while keeping their leaf stomata closed. This acclimation strategy, termed 'differential transpiration', lowers flower inner temperature by about 2‐3°C, protecting reproductive processes at the expense of vegetative tissues.Manipulating stomatal regulation, stomatal size and/or stomatal density of flowers could serve as a viable strategy to enhance the yield of different crops and mitigate some of the current and future impacts of global warming and climate change on agriculture.

Abstract:The effective dose (ED) is the pharmaceutical dosage required to produce a therapeutic response in a fixed proportion of the patients. When only one drug is considered, the problem is a univariate one and has been well‐studied. However, in the multidimensional setting, that is, in the presence of combinations of agents, estimation of the ED becomes more difficult. This study is focused on the plug‐in logistic regression estimator of the multidimensional ED. We discuss consistency of such estimators and focus on the problem of simultaneous confidence regions. We develop a bootstrap algorithm to estimate confidence regions for the multidimensional ED. Through simulation, we show that the proposed method gives 95% confidence regions, which have better empirical coverage than the previous method for moderate to large sample sizes. The novel approach is illustrated on a cytotoxicity study on the effect of two toxins in the leukemia cell line HL‐60 and a decompression sickness study of the effects of the duration and depth of the dive. Abstract:Maize, third most important staple cereal crop across the globe, has been threatened with different environmental constraints including heat and water deficit stress. Higher environmental temperatures negatively affect the most at anthesis, silking and grain filling reproductive phenophases. The situation is further complicated by the variable climatic events, resulting in gradual and sometimes sudden increase in environmental temperature followed by irregular rain frequency and intensity, and thus posing a serious threat to global food security. To meet the ever increasing food demand there is an urgent need to develop climate resilient maize varieties and the goal can be achieved by exploiting the pertinent physiological, biochemical and molecular mechanisms. By introducing certain modifications, together these mechanisms can help us to combat the negative impact of high temperature stress. This review encompasses many of such physiological and molecular approaches which may help in mitigating the adverse effects of heat stress on maize yield. Molecular, biochemical and physiological knowledge available in domain that may be employed in breeding approaches to develop heat tolerant maize genotypes, has thoroughly been discussed.

Abstract:Several studies have indicated that maize (Zea mays L.) kernel weight is severely affected by heat stress, but this response was never evaluated under field conditions. Our objective was to assess the effect of brief episodes of above-optimum temperatures on the dynamics of biomass and water accumulation in kernels of maize hybrids with contrasting tolerance to heat stress. Heat effect on assimilate supply from the plant (i.e., current biomass production and water-soluble carbohydrates in stem) to developing grains was also analyzed. Field experiments included a factorial combination of (i) three hybrids (Te: temperate; Tr: tropical; TeTr: Te×Tr), (ii) two temperature regimes, control and heated during daytime hours (ca. 33–40°C at ear level), and (ii) three 15-d periods (GS1: immediately before anthesis; GS2: from silking onwards; GS3: early phase of active grain filling). Heat effects on final kernel weight were larger (i) when they occurred during the first half of effective grain filling (−23.1% for GS3) than around flowering (−4.8% for GS1, −6.3% for GS2), and (ii) for the Te hybrid (−20.4%) than for the TeTr (−8.6%) and the Tr (−6.8%) hybrids. Heating around flowering (i) enhanced the assimilate availability per kernel during the effective grain-filling period, (ii) increased carbohydrates reserves in stem at physiological maturity, (iii) and had no significant effect on the dynamics of biomass and water accumulation in kernels. The opposite trend was detected among plots heated during GS3, which mostly exhibited the interruption of grain filling. Robust associations were established between (i) carbohydrate reserves in stem at physiological maturity and assimilate availability per kernel during effective grain filling (r2=0.49; P<0.001), and (ii) the rate of water loss from kernels and the duration of effective grain filling (r2=0.71; P<0.001). These responses underlay the enhanced sensitivity to heat stress of the hybrid with full temperate genetic background.

Abstract:An increase in temperature and extreme heat stress is responsible for the global reduction in maize yield. Heat stress affects the integrity of the plasma membrane functioning of mitochondria and chloroplast, which further results in the over-accumulation of reactive oxygen species. The activation of a signal cascade subsequently induces the transcription of heat shock proteins. The denaturation and accumulation of misfolded or unfolded proteins generate cell toxicity, leading to death. Therefore, developing maize cultivars with significant heat tolerance is urgently required. Despite the explored molecular mechanism underlying heat stress response in some plant species, the precise genetic engineering of maize is required to develop high heat-tolerant varieties. Several agronomic management practices, such as soil and nutrient management, plantation rate, timing, crop rotation, and irrigation, are beneficial along with the advanced molecular strategies to counter the elevated heat stress experienced by maize. This review summarizes heat stress sensing, induction of signaling cascade, symptoms, heat stress-related genes, the molecular feature of maize response, and approaches used in developing heat-tolerant maize varieties.

Abstract:Low rainfall combined with heat stress during grain filling can cause a severe threat to maize production. Compared to individual stress, the impact of combined stressors on plant performance during the grain filling remains less explored. In this study, two maize genotypes, ‘B73’ and ‘Mo17’, were subjected to four treatments (control, drought stress, heat stress, and combined drought and heat stress) to understand how maize genotypes respond to stressors during grain filling. Stomatal conductance displayed an opposite response between drought and heat or combined stresses during the mid-grain filling stage. Averaged over genotypes, decreased stomatal conductance (67.4%) and increased leaf temperature (5.6 °C) under combined stress treatments resulted in a significant yield reduction (78.9%) compared to control. Drought or combined stressors during the grain filling stage had a predominant effect on sink potential (kernel weight) than heat stress. Reduction in kernel size of genotype ‘B73’ was associated with a 5.5% decline in starch under heat stress. Analysis of individual and interactive stressors effects on maize showed that trait responses could vary with stresses during grain filling. The present study showed that the maize phenotypic responses differ between individual and interactive stresses. The genotype ‘Mo17’ exhibited relatively higher tolerance to both individual and combined drought and heat stresses than ‘B73’. The present study highlighted the maize plant responses to individual and combined stresses, which is essential for improving maize tolerance to drought and heat stress conditions.
Physio-morphological traits and osmoregulation strategies of hybrid maize (Zea mays) at the seedling stage in response to water-deficit stress

Abstract:Drought has been identified as a major factor restricting maize productivity worldwide, especially in the rainfed areas. The objective of the present study was to investigate the physiological adaptation strategies and sugar-related gene expression levels in three maize (Zea mays L.) genotypes with different drought tolerance abilities (Suwan4452, drought tolerant as a positive check; S7328, drought susceptible as a negative check; Pac339, drought susceptible) at the seedling stage. Ten-day old seedlings of maize genotypes were subjected to (i) well-watered (WW) or control and (ii) water-deficit (WD) conditions. Leaf osmotic potential of cv. S7328 under WD was significantly decreased by 1.35–1.45 folds compared with cv. Pac339 under WW, whereas it was retained in cv. Suwan4452, which utilized total soluble sugars as the major osmolytes for maintaining leaf greenness, Fv/Fm, ΦPSII, and stomatal function (Pn, net photosynthetic rate; gs, stomatal conductance; and E, transpiration rate). Interestingly, sucrose degradation (65% over the control) in cv. Pac339 under WD was evident in relation to the downregulation of the ZmSPS1 level, whereas glucose enrichment (1.65 folds over the control) was observed in relation to the upregulation of ZmSPS1 and ZmSUS1. Moreover, CWSI (crop water stress index), calculated from leaf temperature of stressed plants, was negatively correlated with E, gs, and Pn. Overall, growth characteristics, aboveground and belowground parts, in the drought-susceptible cv. Pac339 and cv. S7328, were significantly decreased (> 25% over the control), whereas these parameters in the drought-tolerant cv. Suwan4452 were unaffected. The study validates the use of leaf temperature, CWSI, Pn, gs, and E as sensitive parameters and overall growth characters as effective indices for drought tolerance screening in maize genotypes at the seedling stage. However, further experiments are required to validate the results observed in this study under field conditions.
