Integrating bioactive glass with compatibilizer: A novel approach for hard tissue regeneration using PXDDA-co-PLA/PLCL/PGLA nanocomposites

Abstract:In the present work, a new nanocomposite was synthesis by Poly (xylitol-dodecanedioic acid)–co-polylactic acid (PXDDA-co-PLA) and Poly (glycolide-co-lactide) (PGLA) with different amount of bioactive glass (BG) S53P4 (0.1, 0.3, 0.5, 0.7 and 1 wt%) as a filler and bioactive agents. To decrease the heterogeneity of nanocomposites, Poly (l,l-lactic acid-co-ɛ-caprolactone) (PLCL) was used as a compatibilizer. The characteristics of nanocomposites were studied by using X-ray diffraction (XRD), Fourier Transform Infrared spectroscopy (FTIR), transmission electron microscopy (TEM), field emission scanning electron microscopy (FE-SEM), contact angle measurements, and mass loss. The micrographs revealed that the addition of BG in the presence of PLCL, cause to uniform dispersion and increase the roughness. The results of contact angles showed that after 120 s, the angles were at around 65° that is suitable for cells activities. In addition, mass loss study indicated that by adding compatibilizer and BG in the nanocomposites, the degradation ratio decreases in dynamic condition. In vitro investigation was performed by immersion the nanocomposite in the phosphate buffer saline (PBS) for 4, 24 and 48 h and results indicated that by increasing the amount of BG in the nanocomposite, the bioactivity increased. The antibacterial properties of nanocomposites were studied against Escherichia coli and Staphylococcus aureus. Results showed that nanocomposites had good inhibition against both bacteria. Also, antibacterial test indicates that adding more than 0.7 wt % of BG did not have more effect on inhibition. So, these results illustrate that synthesized nanocomposites are a suitable candidate for hard tissue regeneration.
