Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Gastrodia elata Blume (G. elata) has a long historical application in Asian countries and its tubers, seeds, and stalks are capable of being utilized for medicine, food, or health care products.AIM OF THE REVIEW: This study aimed to offer a systematic and up-to-date analysis of the current review of the G. elata research advances in traditional uses, phytochemistry, pharmacology, applications, and quality control, as well as a scientific reference for the development and utilization of this plant.MATERIALS AND METHODS: Electronic databases including PubMed, Web of Science, Google Scholar, ScienceDirect, SciFinder, and CNKI were used for the collection of publications on G. elata. The following keywords of G. elata were used truncated with other relevant topic terms, such as phenolic compounds, polysaccharides, glycosides, neuroprotection, learning and memory improvement effects, cardioprotection, applications, and quality control.RESULTS AND CONCLUSIONS: Approximately 134 chemical components mainly categorizing as phenolic compounds, polysaccharides, glycosides, organic acids, and sterols were reported from this plant. Moreover, preclinical studies indicated that G. elata performs several functions, including neuroprotection, learning and memory improvement effects, cardioprotection, vaso-modulatory effect, anti-depression, anti-cancer, and other effects. Currently, G. elata has been widely applied to clinics and foods. The available literature shows that the quality of G. elata might be affected by factors such as origin, fungus, and harvest time, which will have an impact on the drug efficacy. According to past research, G. elata is a potential medicinal and edible plant with several active components and pharmacological activity that has a high application value in medicine and the food business. Nevertheless, few studies have concentrated on characterization of polysaccharides structure and study of non-medicinal parts, implying that further comprehensive research on its polysaccharides structure and non-medicinal parts is critical for full utilization of resources of G. elata.

Abstract:Ethnopharmacological Relevance. The medicinal use of Gastrodia elata BI., a dry tuber of the orchid family, has a long history. Gastrodia elata BI. has the functions of calming the liver, relieving muscle spasms, and dispelling gas. Aim of this Review. To review the traditional uses, botany, phytochemistry, pharmacology, and pharmacokinetics of Gastrodia elata BI. In addition, we discuss the future development and research prospects of this plant in detail. Materials and Methods. This article collects information from relevant documents, including scientific papers, books, and dissertations concerning Gastrodia elata BI. Results. To date, research on Gastrodia elata BI. has identified about 100 active compounds. Many compounds in Gastrodia elata BI. have biological activities, such as sedation and hypnosis, anticonvulsion, improvement of learning and memory, protection of neurons, antidepressive effects, lowering of blood pressure, promotion of angiogenesis, protection of cardiomyocytes, antiplatelet aggregation, anti-inflammatory activity, and amelioration of labor pains. Conclusion. Although many traditional uses of this plant have been confirmed, it is necessary to continue to study the relationship between its structure and function, clarify the mechanisms of pharmacological effects, and explore new clinical applications so as to better delineate the quality control standards for Gastrodia elata BI.

Abstract:Gastrodia elata Blume (GEB), commonly called Tianma in Chinese, is a valuable and extensively used herbal Traditional Chinese Medicine with a wide range of clinical applications. It has been used to treat headaches, dizziness, stroke, epilepsy, amnesia, spasm, and other disorders since ancient times. Hundreds of compounds, including Phenols, glycosides, polysaccharides, steroids, organic acids, and others, have been isolated and identified from this plant. Modern pharmacological studies have shown that its active ingredients possess many pharmacological effects, such as neuroprotective, analgesic, sedation and hypnosis, anti-anxiety, anti-depressant, anti-convulsant, anti-dizziness, blood pressure lowering, blood lipids lowering, liver protection, anti-tumor, and immunity enhancement effects. The present review discusses the pharmacological actions and mechanisms of various components of GEB in cardiovascular diseases to provide a reference for further study of GEB.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE:Gastrodia elata Bl. (GE) is one of the rare Chinese medicinal materials with a long history of medicine and cooking. It consists of a variety of chemical components, including aromatic compounds, organic acids and esters, steroids, saccharides and their glycosides, etc., which has medicinal and edible value, and is widely used in various diseases, such as infantile convulsions, epilepsy, tetanus, headache, dizziness, limb numbness, rheumatism and arthralgia. It is also commonly used in health care products and cosmetics. Thus, its chemical composition and pharmacological activity have attracted more and more attention from the scientific community.AIM:In this review, the processing methods, phytochemistry and pharmacological activities of GE were comprehensively and systematically summarized, which provides a valuable reference for researchers the rational of GE.MATERIALS AND METHODS:A comprehensive search of published literature and classic books from 1958 to 2023 was conducted using online bibliographic databases PubMed, Google Scholar, ACS, Science Direct Database, CNKI and others to identify original research related to GE, its processing methods, active ingredients and pharmacological activities.RESULTS:GE is traditionally used to treat infantile convulsion, epilepsy, tetanus, headache, dizziness, limb numbness, rheumatism and arthralgia. To date, more than 435 chemical constituents were identified from GE including 276 chemical constituents, 72 volatile components and 87 synthetic compounds, which are the primary bioactive compounds. In addition, there are other biological components, such as organic acids and esters, steroids and adenosines. These extracts have nervous system and cardiovascular and cerebrovascular system activities such as sedative-hypnotic, anticonvulsant, antiepileptic, neuron protection and regeneration, analgesia, antidepressant, antihypertensive, antidiabetic, antiplatelet aggregation, anti-inflammatory, etc. CONCLUSION: This review summarizes the processing methods, chemical composition, pharmacological activities, and molecular mechanism of GE over the last 66 years, which provides a valuable reference for researchers to understand its research status and applications.

Abstract:Tianma is the dried tubers of Gastrodia elata Bl. plant (Orchidaceae). Its active ingredient mainly consists of tianma glycosides, phenols, and phenolic glycosides and also contains sterols and their glycosides, polysaccharides, and some other compounds. In recent years, the pharmacological effects of tianma received increasing attention and the study achieved some results. Tianma based crude drugs produce sedative, analgesic, anticonvulsant, and other effects, and also influence the cardiovascular and immune systems. Tianma has always been a hot issue of Chinese medicine research, its main areas covering clinical applications, pharmacology, chemistry, and biology. This article reviews progress of G. elata studies in various aspects.

Abstract:Gastrodia elata Blume (Tianma in Chinese), a myco-heterotrophic orchid, is widely distributed in China. Tubers derived from this orchid are traditionally used as both medicinal and edible materials. At present, five primary varieties of G. elata are recorded in the "Flora of China." Among them, the three main varieties currently in artificial cultivation are G. elata f. elata (GR, red stem), G. elata f. glauca (GB, black stem), and G. elata f. viridis (GG, green stem). In our study, the metabolic profiles and chemical composition of these three varieties were determined via UPLC-MS/MS and HPLC-UV. In total, 11,132 metabolites were detected, from which multiple phytometabolites were identified as aromatic compounds, heteroatomic compounds, furans, carbohydrates, organic acids, and their derivatives. A number of differentially expressed metabolites (DEMs) were annotated as bioactive ingredients. Overall, parishins, vanilloloside, and gastrodin A/B in the GB group were markedly higher, whereas gastrodin, gastrol, and syringic acid were more enriched in the GG or GR groups. Moreover, HPLC fingerprint analysis also found six metabolites used as markers for the identification of Gastrodiae Rhizoma in the Chinese Pharmacopoeia, which were also typical DEMs in metabolomics. Of these, gastrodin, 4-hydroxybenzyl alcohol, citric acid, and adenosine were quantitatively detected, showing a similar result with the metabolomic data. In summary, our findings provide novel insights into the phytochemical ingredients of different G. elata varieties, highlighting diverse biological activities and healthcare value.

Abstract:Cibacron Blue F3GA (CB) was covalently attached onto poly(glycidyl methacrylate) (PGMA) monosize beads for removal of bilirubin from hyperbilirubinemia human serum. PGMA beads were produced by dispersion polymerization (1.6 μm in diameter). CB loading was 1.73 mmol/g. Bilirubin adsorption experiments were performed by stirred-batch adsorption. The non-specific adsorption of bilirubin was low (0.4 mg/g polymer). CB attachment onto the PGMA beads significantly increased the bilirubin adsorption (241.5 mg/g) from aqueous solutions. The maximum bilirubin adsorption was observed at pH 6.0. With an increase of the aqueous phase concentration of sodium chloride, the adsorption amount of bilirubin decreased drastically. The equilibrium adsorption of bilirubin significantly increased with increasing temperature. Much higher adsorption values up to 332 mg bilirubin/g were achieved in the case of the PGMA/CB beads from human plasma.

Abstract:Gastrodia elata Blume (G. elata), listed as one of the 34 precious Chinese medicines, servers a dual purpose as both a medicinal herb and a food source. Polysaccharide is the main active ingredient in G. elata, which has pharmacological activities such as immune regulation, anti-oxidation, anti-cancer, anti-aging, neuroprotection and antibacterial activity and so on. The biological activities of G. elata polysaccharide (GPs) is closely related to its chemical structures. However, no a review has synthetically summarized the chemical structures and pharmacological activities of GPs. This study delves into the chemical structures, pharmacological action of GPs, offering insights for the future development an application of these compounds.
Traffic-Driven Energy Efficient Operational Mechanisms in Cellular Access Networks

Abstract:Emerging smart user terminals and diverse multimedia applications that have driven recent explosive growth in mobile data traffic are increasing energy consumption in cellular networks at an incredible rate. Constituting a significant portion of network operating expenditure and in a context of global warming, rising energy utilization has become a key concern from both economic and environmental perspectives. Consequently, in recent time, the issue of designing energy efficient cellular networks has drawn significant attention, which is also the foremost motivation behind this research. This thesis presents various novel operation mechanisms for single-tier cellular networks designed to enhance the energy efficiency. In a cellular system, base stations (BSs) in the radio access network (RAN) are the most dominant energy consuming equipment estimated between 60%-80%. Therefore, most recently, the idea of attaining a “green cellular network” by reducing energy consumption in BSs has become the spotlight of many researchers. Contemporary BSs consume a considerable amount of energy even at no traffic load. Correspondingly, conventional cellular networks, which are provisioned based on peak-traffic load without accounting for inherent high-degree temporal-spatial traffic diversity, are wasting a significant amount of electrical energy, especially during low-traffic periods. Considering these facts, the proposed research is focused on the design of traffic-sensitive dynamic network reconfiguring mechanisms for energy efficiency in cellular systems. Under the proposed techniques, RANs are adaptively reconfigured using less equipment leading to reduced energy utilization. At the same time, quality of service (QoS) is guaranteed within the target limits. The proposed mechanisms work in a self-organizing fashion requiring no operator assistance. The main contributions of this thesis can be summarized as below: • An energy efficient cellular network framework by employing distributed inter-BS cooperation in a RAN is proposed. Under this framework, based on the instantaneous traffic demand, BSs mutually cooperate for dynamically switching between active and sleep modes by redistributing traffic among themselves and thus, energy savings is achieved. The focus is then extended to exploiting the availability of multiple cellular networks for extracting energy savings through inter-RAN cooperation. Abstract:Ethnopharmacological relevance: Garcinia oligantha Merr. is an ethnomedicine plant mainly distributed in Guangdong and Hainan, China. It has the effects of heat-clearing and detoxicating, which has been used by local ethnic minorities to treat a variety of diseases, including inflammation, internal heat, toothache and scald. The aim of the review: This review summarizes and discusses the progress of the chemical compounds and biological activities of G. oligantha that have been studied in recent years to provide the direction for the prospective research and applications of G. oligantha. Materials and methods: The relevant literature about G. oligantha was accessible from ancient Chinese medical books and records, theses, as well as major scientific databases such as Google Scholar, PubMed, Web of Science, ScienceDirect, SciFinder, Baidu Scholar and China National Knowledge Infrastructure (CNKI). Results: To date, more than 150 chemical compounds were isolated from this plant, including xanthones, volatile oil, fatty acid, benzofurane derivative and biphenyl compounds. Xanthones are the main bioactive compounds that exhibit diverse biological effects, such as antitumor, analgesic, anti-inflammatory, antioxidative, neuroprotective, antimalarial and antibacterial effects, which are consistent with its traditional uses as a folk medicine. Modern pharmacological studies show that these compounds participate in a variety of signaling pathways underlying different pathophysiologies, making them a valuable medicinal resource. Conclusion: G. oligantha is an ethnomedicine with a long history. However, due to regional and cultural constraints, the popularisation and use of ethnomedicine are still limited. Modern pharmacological and chemical research suggest that G. oligantha contains a variety of bioactive compounds and showed diverse biological functions, which is worthy of comprehensive and in-depth research. This review summarizes and discusses the recent progress in studies on G. oligantha, looking forward to promote further research and sustainable development of folk medicinal plants.

Abstract:The American Journal of Chinese Medicine, Ahead of Print. Rhubarb, the Rhei radix et rhizoma (Da huang) is a member of the Polygonaceae family, included in the 2020 edition of the Chinese Pharmacopoeia, and is mainly distributed in Gansu, Sichuan, Qinghai, northwestern Yunnan, and eastern Tibet. Rhei radix et rhizoma is one of the most commonly used traditional Chinese medicines, processed into drinkable tablets in different concoctions for clinical use. Phytochemical studies showed that more than 170 compounds were isolated and identified from Rhei radix et rhizoma, including anthraquinones/anthrones (major constituents), stilbenes, chromones, flavonoids, tannins, and other compounds, etc. Rhei radix et rhizoma is an herb with a long history of traditional use and many potential therapeutic benefits. It can play the role of diarrhea, clearing heat in response to different combinations and concoctions. It is mainly used for the treatment of constipation, gastrointestinal function impairment, and other diseases. In addition, Rhei radix et rhizoma has significant antitumor, anti-inflammatory, antibacterial, hepato-renal protective, neuroprotective, hypolipidemic, and immunomodulatory activities. Its active constituents have anti-tumor, anti-inflammatory, anti-inflammatory and hepato-kidney protective properties. Although it is primarily used for gastrointestinal disorders, it may also have wider applications in various inflammatory and liver disorders. Therefore, further studies are needed to explore its full potential and mechanism of action. This paper reviews the research progress of Rhei radix et rhizoma in terms of botany, traditional use, chemistry, pharmacology, and clinical studies. It aims to provide a scientific basis for in-depth research and development of Rhei radix et rhizome resources.

Abstract:Nowadays, Computational Fluid Dynamics (CFD) methods play an important part in the production process of the automotive industry. Progress in recent years has made possible highly sophisticated airflow-simulation models that are used in engineering for optimization and verification of aerodynamics. The key purpose of the Simulation Tool for Airflow in the human Nose (STAN), developed at the Darmstadt University of Technology in cooperation with the University Hospital in Greifswald, is to use these techniques to support the rhinosurgeon in diagnosis and planning of therapy (Frühauf T, Mlynski G. Simulation and visualization of the air flow in the human nose. Proceedings of the First World Congress on Computational Medicine, Austin, Texas, 1994). A system has been developed that realizes a three-dimensional (3D) reconstruction of the endonasal cavities based on computer tomography (CT) scans. This semiautomatic reconstruction method requires minimal manual intervention. The surface model is used to create an unstructured 3D volume mesh suitable for finite volume simulations. In this way, an individual simulation based on patient-specific data can be realized. At the University Hospital in Greifswald, experimental investigations and measurements are made in nasal models to verify the simulation result. The goal of this project is to investigate individual nasal complaints and to detect respiratory disorders. The surgeon should be able to simulate the disordered respiration before performing a surgical procedure, and thereby increase the effectiveness of surgical planning.

Abstract:Tianma is the dried tuber of Gastrodia elata Blume (G. elata), which is frequently utilized in clinical practice as a traditional Chinese medicine. Gastrodin (GAS) is the main active ingredient of Tianma, which has good pharmacological activity. Therefore, for the first time, this review focused on the extraction, synthesis, pharmacological effects, and derivatives of GAS and to investigate additional development options for GAS. The use of microorganisms to create GAS is a promising method. GAS has good efficacy in the treatment of neurological diseases, cardiovascular diseases, endocrine diseases, and liver diseases. GAS has significant anti-inflammatory, antioxidant, neuroprotective, vascular protective, blood sugar lowering, lipid-regulating, analgesic, anticancer, and antiviral effects. The mechanism involves various signaling pathways such as Nrf2, NF-κB, PI3K/AKT, and AMPK. In addition, the derivatives of GAS and biomaterials synthesized by GAS and PU suggested a broader application of GAS. The research on GAS is thoroughly summarized in this paper, which has useful applications for tackling a variety of disorders and exhibits good development value.Graphical abstract

Abstract:Orchidaceae, well known for its fascinating flowers, is one of the largest and most diverse families of flowering plants. There are many kinds of plants in this family; these are distributed practically globally and have high ornamental and medicinal values. Gastrodia elata Blume, a traditional Chinese medicinal herb, is a rootless and leafless achlorophyllous orchid. Phenolic compounds are considered to be the major bioactive constituents in G. elata, with antioxidant, antiangiogenic, neuroprotective, antidepressant, anxiolytic, and sedative activities. In this study, we determined the contents of six main phenolic components in tubers, stems and flowers from G. elata. Meanwhile, the transcriptomes of the tuber, stem and flower tissues of G. elata were obtained using the BGISEQ-500 platform. A total of 58.29 Gb of data and 113,067 unigenes were obtained, of which 74,820 unigenes were functionally annotated against seven public databases. Differentially expressed genes between tuber, stem and flower tissues were identified. A total of 76 DEGs encoding eight key enzymes were identified as candidate genes involved in the biosynthesis of phenolics in G. elata. For further validation, the expression levels of unigenes were measured using quantitative real-time PCR. Our results greatly enrich the transcriptomic data of G. elata and provide valuable information for the identification of candidate genes involved in the biosynthesis of secondary metabolites.

Abstract:Ethnopharmacological relevance The rhizomes of Anemarrhena asphodeloides Bunge., belonging to the family Liliaceae, are named ' Zhi-mu ' according to traditional Chinese medicine theory. It is a medicinal plant that has long been used as a tonic agent in various ethnomedicinal systems in East Asia, especially in China, and also for treating arthralgia, hematochezia, tidal fever, night sweats, cough, dry mouth and tongue, hemoptysis, etc. The arm of the review The review aims to provide a systematic overview of botany, ethnopharmacology, phytochemistry, pharmacology, toxicology and quality control of Anemarrhena asphodeloides and to explore the future therapeutic potential and scientific potential of this plant. Two hundred sixty-nine compounds have been identified from Anemarrhena asphodeloides , including steroidal saponins, flavonoids, phenylpropanoids, alkaloids, steroids, organic acids, polysaccharides, benzophenones and other ingredients. Studies have shown that the extracts and compounds from Anemarrhena asphodeloides have extensive pharmacological activities, such as nervous system activity, antitumour, anti-inflammatory, antidiabetic, antiosteoporotic, antiallergic, antiplatelet aggregation, antimicrobial, antiviral, anti-ageing, hair growth promoting, preventing cell damage, etc. Evaluating the quality and toxicity of Anemarrhena asphodeloides is essential to confirm its safe use in humans. Conclusion Anemarrhena asphodeloides is widely used in traditional medicine and have diverse chemical constituents with obvious biological activities. Nevertheless, more studies should be carried out in animals and humans to evaluate the cellular and molecular mechanisms involved in its biological activity and confirm its safe use.

Abstract:Gastrodia elata Bl. still widely known as a medicinal plant due to its anti-inflammatory, neuroprotection, cardiovascular protection etc. Additionally, these medical applications cannot be separated from its antioxidant, anti-aging, regulating cell apoptosis ability, which make it have potential as a functional food as well as it has been eaten for more than 2,000 years in China. At present, although Gastrodia elata Bl. has appeared in a large number of studies, much of the research is based on drugs rather than foods. The review of Gastrodia elata Bl. from the perspective of food is one of the necessary steps to promote related development, by reviewing the literature on analytical methods of Gastrodia elata Bl. in recent years, critical components change in the extraction, analytical methods and improvement of food applications, all of aspects of it was summarized. Based on the report about physical and chemical changes in Gastrodia elata Bl. to discover the pathway of Gastrodia elata Bl. functional food development from current to the future.

Abstract:Ethnopharmacological relevance: The genus Polygonatum (Asparagaceae) comprises 71 species distributed throughout the temperate Northern Hemisphere. The medicinal plants of Polygonatum have been traditionally used as tonics in China, India, Pakistan, Iran and Japan, and have been demonstrated to be highly effective in clinical practice for treating age-related diseases, diabetes, lung diseases, fatigue, feebleness and indigestion. Aim of the review: This paper aims to provide the links among traditional uses, chemical constituents, pharmacological effects and toxicity to support their therapeutic potential and uncover opportunities for future research. Materials and methods: The relevant information on the genus Polygonatum was gathered from scientific databases (Google Scholar, Web of Science, SciFinder, ScienceDirect, ACS Publications, PubMed, Wiley Online Library, CNKI). Information was also obtained from online databases, books, Ph.D. dissertations and M.Sc. theses. The literature cited in this review dates from 1917 to June 2017. Results: At least 37 species and 1 variety of Polygonatum plants have been used as traditional medicine and functional food. The major chemical constituents of Polygonatum plants are steroidal saponins, triterpenoid saponins, homoisoflavanones, polysaccharides and lectins. A putative biosynthetic pathway of steroidal saponins and triterpenoid saponins has been established based on the compounds isolated from Polygonatum plants. The crude extracts and certain pure compounds from Polygonatum plants have shown a wide range of pharmacological effects such as anti-aging, anti-diabetic, anti-fatigue, and anticancer effects. The rhizomes of Polygonatum plants have a low degree of toxicity after processing. Conclusions: Based on this review, some traditional uses of Polygonatum species have been confirmed by pharmacological studies, such as its anti-osteoporosis, neuroprotective, immunomodulatory, anti-diabetic and anti-fatigue effects. Most of the pharmacological effects of this genus can be attributed to its polysaccharides, saponins and lectins. However, to clarify the chemical differences that lead to the different traditional uses between "Huangjing" (derived from P. sibiricum, P. kingianum, P. cyrtonema) and "Yuzhu" (derived from P. odoratum), a systematic comparison of the small molecule compositions and polysaccharides of these four species is needed. In addition to these four species, other locally used medicinal Polygonatum species should be the subject of research, and the chemical and pharmacological relationships of these species should be investigated to expand the medicinal resources and standardize the use of Polygonatum species.

Abstract:Purpose of ReviewDepression is a difficult and currently incurable mood disorder and the conventional therapies used as treatment are unsatisfactory nowadays. Therefore, there is an urgent demand to find alternative or complementary approaches to deal with depression. Gastrodia elata Blume (GE) is not only a traditional Chinese medicine but also a food material that has been widely used in treating numerous neurological disorders in East Asia, including migraine, seizure, and mood disorders. In the present review article, we summarized the antidepressant-like effects of GE that have been reported in animal studies and elucidate its cellular mechanisms.Recent FindingsGE and its active compounds, gastrodin, 4-hydroxybenzyl alcohol, vanillin, and their derivatives have recently been reported to have intensive antidepressant-like effects in several animal models of depression, such as the forced swim test, the tail suspension test, and the chronic mild stress. The molecular mechanisms of GE-induced antidepressant-like effect are likely through regulation of neurotransmitters, antioxidation, antiinflammation, modulation of hypothalamic-pituitary-adrenal axis, neurotrophic effects, regulation of stem cell, and enhancement of neuroplasticity and neuroprotection.SummaryGE and its bioactive components have been shown to possess antidepressant-like effects in several well-conducted animal studies. Additionally, GE has been applied to manipulate mood disorders in Chinese medicine for many years. However, more accurate and specific initial targeting of the mechanism of action of GE, as well as the application of clinical trials, are warranted and deserve further investigation.

Abstract:Background Insomnia and depressive disorder are two common symptoms with a reciprocal causal relationship in clinical practice, which are usually manifested in comorbid form. Several medications have been widely used in the treatment of insomnia and depression, but most of these drugs show non-negligible side effects. Currently, many treatments are indicated for insomnia and depressive symptom, including Chinese herbal medicine such as Gastrodia elata Blume ( G. elata ), which has excellent sedative-hypnotic and antidepressant effects in clinical and animal studies. Purpose To summarize the mechanisms of insomnia and depression and the structure-activity mechanism for G. elata to alleviate these symptoms, particularly by hypothalamic-pituitary-adrenal (HPA) axis and intestinal flora, aiming to discover new approaches for the treatment of insomnia and depression. Methods The following electronic databases were searched from the beginning to November 2023: PubMed, Web of Science, Google Scholar, Wanfang Database, and CNKI. The following keywords of G. elata were used truncated with other relevant topic terms, such as depression, insomnia, antidepressant, sedative-hypnotic, neuroprotection, application, safety, and toxicity. Results Natural compounds derived from G. elata could alleviate insomnia and depressive disorder, which is involved in monoamine neurotransmitters, inflammatory response, oxidative stress, and gut microbes, etc. Several clinical trials showed that G. elata -derived natural compounds that treat depression and insomnia have significant and safe therapeutic effects, but further well-designed clinical and toxicological studies are needed. Conclusion G. elata exerts a critical role in treating depression and insomnia due to its multi-targeting properties and fewer side effects. However, more clinical and toxicological studies should be performed to further explore the sedative-hypnotic and antidepressant mechanisms of G. elata and provide more evidence and recommendations for its clinical application. Our review provides an overview of G. elata treating insomnia with depression for future research direction.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Fructus Gardeniae (FG) is the dried fruit of Gardenia jasminoides Ellis (GjE), which belongs to the family Rubiaceae. FG has a long history of use as a herb, and was originally recorded in Sheng Nong's herbal classic. FG has also been widely used as both medicine and food.AIM OF STUDY: This review aimed to provide a systematic and comprehensive analysis of the current research progress of FG in terms of ethnopharmacology, phytochemistry, pharmacology and toxicity, to provide new insights and extensive field of view for subsequent studies.METHODS: Scientific databases, including CNKI, VIP (Chinese literature), PubMed, Science Direct, Elsevier and Google Scholar (English literatures) were searched to gather data about FG and its main active ingredients such as geniposide and genipin (only regarding toxicity).RESULTS: Many chemical constituents have been identified from the fruit of GjE, including iridoids, terpenoids, flavonoids, organic acids, volatile oils and others. The constituents of different parts of FG and processed FG are different from those of whole FG. FG extract and its main active constituents have been reported to have pharmacological properties such as hepatoprotective, choleretic, anti-inflammatory, antioxidant, neuroprotective, anti-diabetic, anti-apoptotic and antitumor activities. However, an increasing number of studies have shown that FG induces multiple organ injury, especially causing hepatotoxicity and nephrotoxicity, which could increase the risk during clinical use. The available literature shows that geniposide, a major active component of FG and a critical marker for its quality, is associated with the pharmacology and toxicity of FG.CONCLUSION: Although a large number of studies examining FG have been published, issues remain. In the aspect of FG's pharmacology, the traditional efficacy and modern pharmacological effects of FG should be combined, which to broadens clinical application prospects. In addition, few studies have assessed the toxicity of FG. Toxicity assessment of FG should tackle various aspects, including compatibility, processing and the symptom-based prescription theory, in addition to over-dosage or long-term use, for a reasonable clinical use.
