Abstract:The invasive ectoparasite Sarcoptes scabiei affects the welfare and conservation of Australian marsupials. Molecular data suggest that spillover from other hosts may be responsible for the emergence of this infectious disease, but the scale of such studies is limited. We performed expanded molecular typing of the S. scabiei mitochondrial cox1 gene from 81 skin scrapings from infested wombats ( Vombatus ursinus), koalas ( Phascolarctos cinereus), red foxes ( Vulpes vulpes), and dogs ( Canis lupus familiaris) across Australia. Combined with existing S. scabiei sequences, our analysis revealed 16 haplotypes among Australian animals, sharing between 93.3% and 99.7% sequence similarity. While some sequences were unique to specific hosts or to Australia, key haplotypes could be detected across several marsupial hosts as well as to wild or domestic canids in Australia. We identified 43 cox1 haplotypes with many Australian haplotypes identical to S. scabiei mites from inside and outside Europe. We concluded that multiple introduction events were plausible explanations to the origin and emergence of this parasite into Australian marsupials and that disease spillover from canids was likely. Together, our greatly expanded S. scabiei sequence dataset provided a more nuanced picture of both spillover and sustained intraspecific transmission for this important parasite.

Abstract:Demodecidae are the most specialized parasitic mites of mammals; they typically inhabit the skin, but they have been found in other tissues and organs. They can cause demodecosis (a disease which is hazardous and difficult to cure) in humans, domestic animals and livestock. They are parasites with high host and topical specificity. They have been found for most orders of mammals, and they are common in the populations of numerous host species. Therefore, they not only constitute an important subject of veterinary and medical study, but also comprise an excellent model for faunistic and parasitological analyses concerning different aspects of functioning and evolution of the host–parasite relationship. The current level or knowledge of demodecid mites is irregular and fragmentary, and numerous questions require elaboration and ordering, from the taxonomic diversity to geographic distribution and relations with hosts. Such data may be of use i.a. for the development of more efficient and reliable diagnostic methods, as well as understanding the etiology and pathogenesis mechanisms of demodecosis, currently a contentious issue. The present paper lists all formally-described valid species of demodecid mites, together with other functioning specific names, verified and with comments on their status. This is significant for correct species identification and demodecosis diagnostics. The list has been drawn up on the basis of data acquired in the period 1842−2020. It contains 122 valid species of parasite, including their hosts and geographic distribution, data on parasitism, as well as only the second record of Demodex sciurinus in Eurasian red squirrel Sciurus vulgaris in over 100 years since its initial discovery.

Abstract:The parasitic mite Sarcoptes scabiei is an economically highly significant parasite of the skin of humans and animals worldwide. In humans, this mite causes a neglected tropical disease (NTD), called scabies. This disease results in major morbidity, disability, stigma and poverty globally and is often associated with secondary bacterial infections. Currently, anti-scabies treatments are not sufficiently effective, resistance to them is emerging and no vaccine is available. Here, we report the first high-quality genome and transcriptomic data for S. scabiei. The genome is 56.6 Mb in size, has a a repeat content of 10.6% and codes for 9,174 proteins. We explored key molecules involved in development, reproduction, host-parasite interactions, immunity and disease. The enhanced 'omic data sets for S. scabiei represent comprehensive and critical resources for genetic, functional genomic, metabolomic, phylogenetic, ecological and/or epidemiological investigations, and will underpin the design and development of new treatments, vaccines and/or diagnostic tests.
Detection, Prevalence and Phylogenetic Relationships of Demodex spp and further Skin Prostigmata Mites (Acari, Arachnida) in Wild and Domestic Mammals

Abstract:This study was conceived to detect skin mites in social mammals through real-time qPCR, and to estimate taxonomic Demodex and further Prostigmata mite relationships in different host species by comparing sequences from two genes: mitochondrial 16S rRNA and nuclear 18S rRNA. We determined the mite prevalence in the hair follicles of marmots (13%) and bats (17%). The high prevalence found in marmots and bats by sampling only one site on the body may indicate that mites are common inhabitants of their skin. Since we found three different mites (Neuchelacheles sp, Myobia sp and Penthaleus sp) in three bat species (Miotis yumanensis, Miotis californicus and Corynorhinus townsendii) and two different mites (both inferred to be members of the Prostigmata order) in one marmot species (Marmota flaviventris), we tentatively concluded that these skin mites 1) cannot be assigned to the same genus based only on a common host, and 2) seem to evolve according to the specific habitat and/or specific hair and sebaceous gland of the mammalian host. Moreover, two M. yumanensis bats harbored identical Neuchelacheles mites, indicating the possibility of interspecific cross-infection within a colony. However, some skin mites species are less restricted by host species than previously thought. Specifically, Demodex canis seems to be more transmissible across species than other skin mites. D. canis have been found mostly in dogs but also in cats and captive bats. In addition, we report the first case of D. canis infestation in a domestic ferret (Mustela putorius). All these mammalian hosts are related to human activities, and D. canis evolution may be a consequence of this relationship. The monophyletic Demodex clade showing closely related dog and human Demodex sequences also supports this likely hypothesis.

Abstract:Presently, there is a dearth of proteomic data for parasitic mites and their relationship with the host animals. Here, using a high throughput LC-MS/MS-based approach, we undertook the first comprehensive, large-scale proteomic investigation of egg and adult female stages of the scabies mite, Sarcoptes scabiei-one of the most important parasitic mites of humans and other animals worldwide. In total, 1,761 S. scabiei proteins were identified and quantified with high confidence. Bioinformatic analyses revealed differentially expressed proteins to be involved predominantly in biological pathways or processes including genetic information processing, energy (oxidative phosphorylation), nucleotide, amino acid, carbohydrate and/or lipid metabolism, and some adaptive processes. Selected, constitutively and highly expressed proteins, such as peptidases, scabies mite inactivated protease paralogues (SMIPPs) and muscle proteins (myosin and troponin), are proposed to be involved in key biological processes within S. scabiei, host-parasite interactions and/or the pathogenesis of scabies. These proteomic data will enable future molecular, biochemical and physiological investigations of early developmental stages of S. scabiei and the discovery of novel interventions, targeting the egg stage, given its non-susceptibility to acaricides currently approved for the treatment of scabies in humans. Author summary Scabies is a neglected tropical disease caused by the parasitic mite Sarcoptes scabiei. The treatment and control of scabies are challenging, as there is no vaccine and the two mostly used broad-spectrum acaricides (i.e. ivermectin and permethrin) do not kill the key developmental stage (egg) of the mite that enables the re-establishment of infection. The availability of a well-assembled genome for S. scabiei now provides a foundation to explore the molecular biology, biochemistry and physiology of this mite. Here, we characterised the first somatic proteome of key developmental stages of S. scabiei using high throughput LC-MS/MS. Bioinformatic analyses of proteomic data indicate that proteins expressed differentially between egg and female adult stages are mainly involved in biological pathways or processes, such as genetic information processing, energy (oxidative phosphorylation), nucleotide, amino acid, carbohydrate and/or lipid metabolism in the mite. Abstract:Sarcoptes scabiei mites and skin lesions consistent with severe sarcoptic mange were identified in a Tasmanian devil (Sarcophilus harrisii) and Bennett's wallaby (Notamacropus rufogriseus) from Tasmania, Australia. The devil and wallaby both had severe hyperkeratotic skin lesions. All stages of mite development were identified in the devil, suggesting parasite reproduction on the host. The devil was also affected by devil facial tumor disease and several other parasites. This expands the global host range of species susceptible to this panzootic mange disease.
The effects of spatially-constrained treatment regions upon a model of wombat mange

Abstract:The use of therapeutic agents is a critical option to manage wildlife disease, but their implementation is usually spatially constrained. We seek to expand knowledge around the effectiveness of management of environmentally-transmitted Sarcoptes scabiei on a host population, by studying the effect of a spatially constrained treatment regime on disease dynamics in the bare-nosed wombat Vombatus ursinus . A host population of wombats is modelled using a system of non-linear partial differential equations, a spatially-varying treatment regime is applied to this population and the dynamics are studied over a period of several years. Treatment could result in mite decrease within the treatment region, extending to a lesser degree outside, with significant increases in wombat population. However, the benefits of targeted treatment regions within an environment are shown to be dependent on conditions at the start (endemic vs. disease free), as well as on the locations of these special regions (centre of the wombat population or against a geographical boundary). This research demonstrates the importance of understanding the state of the environment and populations before treatment commences, the effects of re-treatment schedules within the treatment region, and the transient large-scale changes in mite numbers that can be brought about by sudden changes to the environment. It also demonstrates that, with good knowledge of the host-pathogen dynamics and the spatial terrain, it is possible to achieve substantial reduction in mite numbers within the target region, with increases in wombat numbers throughout the environment.

Distributed power control in asymmetric interference-limited networks

Abstract:Power control in wireless communication networks reduces i nterference by regulating access to the shared wireless med iu and enhances the overall network capacity as well as battery lif of nodes. Beginning with the Foschini-Miljanic (FM) al gorithm [1], distributed power control for wireless networks has be en an active area of research. The problem of distributed pow er control for packetized traffic was first addressed by Kanduku ri and Bambos [2] by proposing “Power-controlled multiple a ccess” (PCMA) family of algorithms, and subsequently investigate d in [3], [5], [6]. PCMA algorithm uses dynamic programming approach to balance the power versus delay cost of the transm itter. It introduces three separate regimes: aggressive, s oft backoff, hard backoff. In the low-interference regime, transmitter power tries to aggressively match and overcome the interfer ence – up to a certain point, while in the high interference regime, th e power is set to zero (hard backoff) and the transmitter ceas es to transmit waiting for the low interference periods. Thus, PC MA algorithms usually induce a soft time-division multiple access (iTDMA) allowing links facing high interference to tempora rily reduce their power and incur a delay cost instead. On the other hand, in Foschini-Miljanic (FM) algorithm, each link attempts to maintain a target SIR by overcoming the interfer nce presented by the other links. In this project, distributed power control algorithms are i nvestigated in a two-link interference-limited network wi th asymmetric interference. Such asymmetric interference can occ ur in any cellular as well as adhoc wireless networks, when th first link receives strong interference from the second link, and the second link receives weak interference from the first lin k. For example, a downlink user connected to a distant macro-cell c an be jammed due to the presence of a closer downlink user connected to a femtocell using the same frequency/time slot ( ee Fig. 1(a)). In adhoc wireless networks, the random plac ement of links may also result in asymmetric interference. Since d istributed power control algorithms control transmitter p ower based on the interference seen by the transmitter, they can p erform poorly in asymmetric interference-limited network s. If FM algorithm is used in asymmetric interference-limited netw orks, the weak link uses high power proportional to the inter fer nce from the strong link to maintain its target SIR, and its queue starts exploding at very low loads. PCMA algorithm performs better than FM algorithm in asymmetric interference-limit ed network, but it is unable to induce a iTDMA effect because t he strong link can get stuck in aggressive or soft backoff regim e when the average load is high and interference seen is low. Distributed power control algorithms have not been investi gated in asymmetric interference-limited networks to the b est of my knowledge. In this project, distributed power control al gorithms [1], [3] are simulated, and their performance is an alyzed for two-link asymmetric interference-limited network. Ba sed on the results, some variations are proposed to improve t he performance of PCMA algorithms, and these variations are st ill under investigation.

Abstract:Sarcoptic mange is a skin disease caused by the contagious ectoparasite Sarcoptes scabiei , capable of suppressing and extirpating wild canid populations. Starting in 2015, we observed a multi-year epizootic of sarcoptic mange affecting a red fox ( Vulpes vulpes ) population on Fire Island, NY, USA. We explored the ecological factors that contributed to the spread of sarcoptic mange and characterized the epizootic in a landscape where red foxes are geographically constrained.

Abstract:Domestic and wild felids are considered suitable hosts for the parasitic mite Sarcoptes scabiei, and sarcoptic mange is reported in several felid species in the scientific literature. However, the historic classification of Sarcoptes mites into host-specific varieties does not include S. scabiei var. felis. It is unclear whether sarcoptic mange transmission in felids involves canids, other sympatric species, or exclusively felids. This study aimed to characterize the genetic structure of S. scabiei mites from domestic cats ( Felis catus) and Eurasian lynx ( Lynx lynx carpathicus), comparing them with Sarcoptes mites from sympatric domestic and wild carnivores. Ten Sarcoptes microsatellite markers were used to genotype 81 mites obtained from skin scrapings of 36 carnivores: 4 domestic cats, one dog ( Canis lupus familiaris), 4 Eurasian lynx, 23 red foxes ( Vulpes vulpes), and 4 grey wolves ( Canis lupus lupus) from either Italy, Switzerland or France. Two genetic clusters of S. scabiei with a geographical distribution pattern were detected: mites from cats originating from Central Italy clustered with those from sympatric wolves. In contrast, all the other mites from Switzerland, France and Northern Italy clustered together. These results strengthen the previously advanced hypothesis that genetic variants of S. scabiei have a predominant geographic-related distribution with cryptic transmission patterns. These patterns may rely on the interactions between different hosts living in the same ecological niche rather than a simple infection among hosts belonging to the same taxon, reinforcing the idea that the S. scabiei historic classification into "var" might have little ongoing relevance. Les félidés domestiques et sauvages sont considérés comme des hôtes appropriés pour l'acarien parasite Sarcoptes scabiei, et la gale sarcoptique est signalée chez plusieurs espèces de félidés dans la littérature scientifique. Cependant, la classification traditionnelle des acariens du genre Sarcoptes en variétés spécifiques à l'hôte n'inclut pas S. scabiei var. felis. On ne sait pas si la transmission de la gale sarcoptique chez les félidés implique des canidés, d'autres espèces sympatriques ou exclusivement des félidés. Cette étude visait à caractériser la structure génétique des acariens S. scabiei des chats domestiques ( Felis catus) et du lynx eurasien ( Lynx lynx carpathicus), en les comparant aux Sarcoptes des carnivores domestiques et sauvages sympatriques. Ces résultats renforcent l'hypothèse précédemment avancée selon laquelle les variants génétiques de S. scabiei ont une distribution géographique prédominante avec des schémas de transmission cryptiques. Ces modèles peuvent reposer sur les interactions entre différents hôtes vivant dans la même niche écologique plutôt que sur une simple transmission parmi des hôtes appartenant au même taxon, renforçant l'idée que la classification historique de S. scabiei en "var" a peu de pertinence.

Abstract:Rheumatic heart disease (RHD) and acute rheumatic fever (ARF) disproportionately affect individuals in low-resource settings. ARF is attributed to an immune response to Group A Streptococcus (GAS) following GAS pharyngitis and potentially GAS impetigo in which infection can be initiated by scabies infestation. The burden of ARF and RHD in Aboriginal and Torres Strait Islander people in Australia is among the highest globally. Following recent calls to include dog management programs in ARF and RHD prevention programs, we believe it is timely to assess the evidence for this, particularly since previous recommendations excluded resources to prevent zoonotic canine scabies. While phylogenetic analyses have suggested that the Sarcoptes mite is host specific, they have differed in interpretation of the strength of their findings regarding species cross-over and the need for canine scabies control to prevent human itch. Given that there is also indication from case reports that canine scabies leads to human itch, we propose that further investigation of the potential burden of zoonotic canine scabies and intervention trials of canine scabies prevention on the incidence of impetigo are warranted. Considering the devastating impacts of ARF and RHD, evidence is required to support policy to eliminate all risk factors.

Abstract:The conservation management of threatened wildlife increasingly relies upon translocations to augment populations. Translocations, however, pose various risks: from the host perspective these include the spread of parasitic disease, whereas from a broader biodiversity perspective translocation may lead to the loss of rare parasites and other dependent fauna. Although Disease Risk Analyses are recommended during translocation planning, knowledge regarding the parasites infecting threatened species or their pathogenicity is often lacking. Between March 2014 and June 2016, woylies ( Bettongia penicillata ) and sympatric marsupials were screened for the presence of endo- and ectoparasites, during two fauna translocations in south-western Australia. Here, we summarise the parasite taxa identified from B. penicillata , brush-tailed possums ( Trichosurus vulpecula hypoleucus ) and chuditch ( Dasyurus geoffroii ), including prevalence data for host, parasite taxon and site. Results from the opportunistic sampling of other species ( Isoodon fusciventer , Phascogale tapoatafa wambenger , Tiliqua rugosa and Felis catus ) are also presented. New host–parasite records including Hepatozoon spp. from T. v. hypoleucus , Trypanosoma noyesi from T. rugosa , Ixodes australiensis and Ixodes tasmani from D. geoffroii , and I. australiensis and Amblyomma sp. from a P. t . wambenger were identified. This study highlights the importance of monitoring sympatric species, particularly when compiling baseline data of parasite fauna present within translocation sites and enhances our knowledge of parasites infecting terrestrial wildlife within Australia's south-west, a Global Biodiversity Hotspot.

Abstract:Background: The scabies mite, Sarcoptes scabiei, is a parasitic arachnid and cause of the infectious skin disease scabies in humans and mange in other animal species. Scabies infections are a major health problem, particularly in remote Indigenous communities in Australia, where secondary group A streptococcal and Staphylococcus aureus infections of scabies sores are thought to drive the high rate of rheumatic heart disease and chronic kidney disease. Results: We sequenced the genome of two samples of Sarcoptes scabiei var. hominis obtained from unrelated patients with crusted scabies located in different parts of northern Australia using the Illumina HiSeq. We also sequenced samples of Sarcoptes scabiei var. suis from a pig model. Because of the small size of the scabies mite, these data are derived from pools of thousands of mites and are metagenomic, including host and microbiome DNA. We performed cleaning and de novo assembly and present Sarcoptes scabiei var. hominis and var. suis draft reference genomes. We have constructed a preliminary annotation of this reference comprising 13,226 putative coding sequences based on sequence similarity to known proteins. Conclusions: We have developed extensive genomic resources for the scabies mite, including reference genomes and a preliminary annotation.
Sarcoptic mange in the wild boar, Sus scrofa, in Sweden

Abstract:Sarcoptic mange caused by Sarcoptes scabiei has been present in the Swedish red fox (Vulpes vulpes) population since the 1970s. The disease has been described in other Swedish wildlife species, but not in the wild boar, Sus scrofa, until 2009. Single cases of sarcoptic mange have been diagnosed the last years in the expanding population of wild boar. This study aims to describe the histopathological lesions found on mangy wild boar and compare, by molecular methods, mites from wild boar cases with mites from mangy red foxes, raccoon dogs, and domestic pigs. Mangy wild boar with focal alopecia and clinical signs of pruritis were reported or submitted from various areas in southern Sweden to the National Veterinary Institute, Uppsala. The examined skin samples of wild boar infected with S. scabiei showed limited gross skin lesions, except for cases with severe exudative dermatitis. Histopathology of the affected wild boar skin samples showed an eosinophilic dermatitis with a variable hyperkeratosis and often low number of mites present. To study the relationship of S. scabiei mites isolated from different host species, a population genetics investigation was performed based on microsatellite markers. In total, 225 individual mites from eight individuals of four different host species; red fox (48 mites), wild boar (80 mites), domestic pig (48 mites) and raccoon dog (43 mites), were included in the study. In the phylogenetic analysis, all mites isolated from wild boar clustered together even though they originate from different geographical regions in Sweden. Mites from each individual host showed high similarity. The results indicate that wild boar mites differ from mites both from the red fox, raccoon dog, and domestic pig.

Abstract:All-optical AND and NAND gates have been demonstrated in a Ti-diffused periodically poled LiNbO(3) channel waveguide which has two second-harmonic phase-matching peaks by cascaded sum-frequency-generation/difference-frequency-generation (cSFG/DFG) and sum-frequency-generation (SFG) processes. The conversion efficiency of signal to idler (AND gate signal) was approximately 0 dB in cSFG/DFG process. In the second SFG process, more than 15 dB extinction ratio between signal and dropped signal (NAND gate signal) has been observed.

Abstract:Wildlife that use anthropogenic resources often come into conflict with humans, e.g. due to damaged property, habituation or transmission of pathogens, amongst them Sarcoptes scabiei , the aetiological agent of sarcoptic mange, an emerging panzootic skin disease. This study examines the use of haystacks intended for supplementary feeding of European bison ( Bison bonasus ) by wolves ( Canis lupus ) with sarcoptic mange and the potential role of this behaviour in skin parasite transmission and human-wolf conflict. Hay samples from the beds used by wolves were found to harbour S. scabiei mites, even several days after the last use. Our data demonstrate an unforeseen link between wild ungulate supplementary feeding and wolf behaviour that may lead to conflict, namely approaching human settlements. However, no negative interactions were observed between wolves and humans or domestic animals. The presence of S. scabiei mites in haystacks provides a potential for its human-facilitated environmental transmission among wildlife and to domestic animals.

Abstract:Abstract A recent surge in scabies infections has risen to epidemic levels, amid an acute shortage of treatments, coupled with concerns regarding potential rising resistance of Sarcoptes scabiei to first-line treatment. This is a public health crisis. Scabies has subsequently been added to the World Health Organization’s list of neglected tropical diseases. Clues to solve the current crisis may lie in the history of this mite and its relationship with dermatology. Scabies, a parasitic infestation caused by the S. scabiei mite, has been a persistent challenge throughout the annals of dermatological history. This retrospective study traces the evolution of scabies treatment from ancient remedies to contemporary therapeutic modalities. The earliest documented treatments, rooted in ancient civilizations, range from herbal concoctions and topical ointments to ritualistic practices. Throughout the Middle Ages, a mixture of folklore and early medical observations led to diverse concoctions incorporating sulfur and mercury. The Renaissance marked a period of increased scientific inquiry, with the identification of the scabies mite itself by the Italian physician Giovanni Cosimo Bonomo and his assistant Diacinto Cestoni in 1687. Advancements in parasitology during the 19th and early 20th centuries led to a more precise understanding of scabies’ transmission and life cycle. This period introduced various topical treatments, including benzyl benzoate, which has remained prominent since. The mid-20th century brought about the discovery of insecticides like lindane and permethrin, revolutionizing scabies therapy. Buxton’s work discussing the prevention and treatment of scabies during the 1900s, when scabies was on the rise, highlighted the importance of hygiene, prevention, seasonality and public health measures. He advocated for effective disinfection of bedding and clothing, and education on the disease and preparations used to eradicate scabies. However, concerns over resistance and side-effects have prompted a shift towards safer and more efficacious treatments in the late 20th century. Ivermectin, an antiparasitic medication with broad-spectrum activity, emerged as a significant breakthrough, offering an oral alternative to traditional topical therapies. The 21st century witnessed ongoing research into novel therapeutic agents, including newer topical acaricides and investigational drugs. This historical exploration highlights the evolution of scabies’ treatment through time with advancing medical understanding. As we experience permethrin shortages, dermatologists are seeking to find alternative approaches to address this public health issue – in which we can hunt for clues in our past. Dermatologists should ensure that patients are equipped with an understanding of disease transmission and prevention strategies, and consider sulfur-based products. A recent study demonstrated that sulfur 10% ointment was significantly more effective in treating scabies than permethrin 5% cream, highlighting how we may learn from our ancestorial practices.

Abstract:Scabies is an itchy skin disease caused by the burrowing mite Sarcoptes scabiei . During their lifespan, the female mites invade the stratum corneum and create tunnels, in which they reside, move, feed, deposit fecal pellets, and lay eggs. Recently, scabies was included in the World Health Organization roadmap for neglected tropical diseases 2021–2030. This review attempts to summarize our knowledge about the mite's biology and the disease pathogenesis, pathological changes, and complications. Generally, the host–parasite interaction in scabies is highly complex and involves different mechanisms, some of which are yet largely unknown. Elucidation of the nature of such interaction as well as the underlying mechanisms could allow a better understanding of the mite's biology and the development of novel diagnostic and therapeutic options for scabies control programs. Moreover, identification of the molecular basis of such interaction could unveil novel targets for acaricidal agents and vaccines.
