Abstract:Production of floral nectar is generally thought to be an adaptation that increases plant fitness by altering pollinator behavior, and therefore pollination success. To test this hypothesis, I investigated the effects of floral nectar production rate on pollination success of the hermaphroditic plant Ipomopsis aggregata (Polemoniaceae). Success through male function (estimated by the export of fluorescent dyes) was significantly greater for plants with naturally high nectar production rates than for nearby plants with low nectar production rates, whereas success through female function (receipt of fluorescent dye) was unrelated to nectar production rate. Experimental addition of artificial nectar also produced a significant increase in male function success and no increase in several estimates of female function success. Observations confirmed that hummingbirds probed a larger proportion of flowers on plants that received supplemental nectar, as they do in response to natural variation in nectar production. The concordance of results across these observational and experimental studies indicates that nectar production acts primarily to increase pollination success through male function for this species.
Diversity and distribution analysis of eukaryotic communities in the Xiangshan Bay, East China sea by metabarcoding approach

Abstract:Eukaryotic communities play an important role in the coastal ecosystem of Xiangshan Bay, a narrow semi-closed bay famous for fisheries and marine farming. However, information on the diversity and composition of eukaryotic communities in Xiangshan Bay remains unclear. In this study, the metabarcoding approach was utilized to comprehensively investigate the eukaryotic plankton community structure and dominant taxa, particularly eukaryotic microalgae, in the Xiangshan Bay over a period of four months in 2018. The results showed that the three major phyla were Arthropoda, Chlorophyta, and Bacillariophyta. The richness indices revealed that species richness peaked in February and was at its lowest in May. Diversity indices showed that the samples collected in May had the lowest diversity. Centropages was detected in the samples of all months, however, its highest dominance was observed in the samples collected in February. In addition, compared to other months, a greater proportion of eukaryotic microalgae was witnessed in March. The three eukaryotic algae with highest abundances in March were Cyclotella , Prorocentrum , and Thalassiosira . Moreover, high diversity of pico-sized (0.2–2.0 μm) phytoplankton (which are often easily missed by microscopy) was discovered in this study by using metabarcoding approach. This study highlights the strength and significance of the metabarcoding approach to uncover a large number of eukaryotic species which remains undetectable during application of conventional approaches. The findings of this study reveals that the eukaryotic community structure varies noticeably in both time and space throughout sampling period, with temperature being the most important environmental factor influencing these changes. This study lays a solid foundation to understand eukaryotic plankton composition, temporal and spatial dynamics and the distribution mechanism of eukaryotic plankton community in Xiangshan Bay, providing theoretical reference for further studies related to marine ecology.

Abstract:Protists engaging in photo- and phago- mixotrophy (mixoplankton) are common members of the global plankton community. They are involved in primary production and contribute to the carbon and nutrient cycling. Two major mixoplankton functional types (MFTs) are considered based upon the origin of their photosynthetic abilities: innate for constitutive-mixoplankton (CM) and obtained from prey for non-constitutive mixoplankton (NCM). Regardless of their significance, little attention has been paid to their diversity and temporal succession. We performed a metabarcoding survey of the V4-18S rRNA gene in 92 surface water samples collected during 2018–2019 in five fixed stations of the Belgian Coastal Zone. Environmental data such as nutrients, sea surface temperature, salinity, Chl -a and light were collected to understand their influences over mixoplankton community changes. The temporal diversity of mixotrophs, autotrophs, and heterotrophs was analyzed and the distinct seasonal patterns were evidenced. Results showed that dinoflagellates and ciliates were the major mixoplankton contributors. There were no significant differences among protist communities between the stations sampled. The time-series showed high proportional abundances of CM, accounting in average for 24.4% of the reads, against the low contribution of NCM, 4.8%. CM dinoflagellates belonging to Heterocapsa , Alexandrium, Karlodinium , and Tripos genus were the most abundant, and co-occurred with strict autotrophic plankton. Strombidium genus ciliates were the most representative organisms for NCM. Mixoplankton showed lower diversity than autotrophs and heterotrophs throughout the time series, however, the environmental factors controlling the seasonal community shifts (β-diversity) were similar. Overall, the metabarcoding approach allowed to depict with high resolution the composition of mixoplankton and its diversity among auto- and heterotrophs in the Belgian Coastal Zone.

Abstract:Microeukaryotes are key contributors to marine carbon cycling. Their physiology, ecology, and interactions with the chemical environment are poorly understood in offshore ecosystems, and especially in the deep ocean. Using the Autonomous Underwater Vehicle Clio, microbial communities along a 1050 km transect in the western North Atlantic Ocean were surveyed at 10-200 m vertical depth increments to capture metabolic signatures spanning oligotrophic, continental margin, and productive coastal ecosystems. Microeukaryotes were examined using a paired metatranscriptomic and metaproteomic approach. Here we show a diverse surface assemblage consisting of stramenopiles, dinoflagellates and ciliates represented in both the transcript and protein fractions, with foraminifera, radiolaria, picozoa, and discoba proteins enriched at >200 m, and fungal proteins emerging in waters >3000 m. In the broad microeukaryote community, nitrogen stress biomarkers were found at coastal sites, with phosphorus stress biomarkers offshore. This multi-omics dataset broadens our understanding of how microeukaryotic taxa and their functional processes are structured along environmental gradients of temperature, light, and nutrients.

Abstract:Metabarcoding is revolutionising the analysis of biodiversity in marine ecosystems, especially as it provides a means of detecting and identifying cryptic life stages in field samples. The planktonic larval stage of many species underpins the abundance and distribution of adult populations but is challenging to characterise given the small size of larvae and diffuse distributions in pelagic waters. Yet, planktonic larval dynamics are key to understanding phenomena observed in adult populations, such as the boom-and-bust dynamics exhibited by some echinoderms. Rapid changes in echinoderm population density can have significant effects on local benthic ecosystems. For example, outbreaks of the crown-of-thorns sea star (CoTS) on the Great Barrier Reef (GBR) have led to considerable declines in coral cover. Here, we used a DNA metabarcoding approach to investigate the spatio-temporal distribution and diversity of echinoderm larvae on the GBR, including CoTS. Generalised linear mixed models revealed that echinoderm larval richness, was significantly correlated with temporal variables (i.e. season and year) which is consistent with expected fluctuations in larval output based on adult spawning periodicity. However, neither site-specific differences in echinoderm larval richness, nor correlations between larval composition and environmental, temporal, or spatial variables were found. This study validates the utility of metabarcoding approaches for detecting and characterising echinoderm larvae, including CoTS, which could prove useful to future monitoring efforts. Our findings suggest that metabarcoding can be used to better understand the life history of planktonic larvae, and analyses combining environmental (e.g., temperature, nutrients) and oceanographic (e.g., currents) data could deliver valuable information on the factors influencing their spatio-temporal distributions.

Abstract:A central goal in marine microecology is to understand the ecological factors shaping spatiotemporal microbial patterns and the underlying processes. We hypothesized that abiotic and/or biotic interactions are probably more important for explaining the distribution patterns of marine bacterioplankton than environmental filtering. In this study, surface seawater samples were collected about 7000 miles from the Mediterranean Sea, transecting the North Atlantic Ocean, to the Brazilian marginal sea. In bacterial biosphere, SAR11, SAR86, Rhodobacteraceae, and Rhodospiriaceae were predominant in the Mediterranean Sea; Prochlorococcus was more frequent in Atlantic Ocean; whereas in the Brazilian coastal sea, the main bacterial members were Synechococcus and SAR11. With respect to archaea, Euryarchaeota were predominant in the Atlantic Ocean and Thaumarchaeota in the Mediterranean Sea. With respect to the eukaryotes, Syndiniales, Spumellaria, Cryomonadida, and Chlorodendrales were predominant in the open ocean, while diatoms and microzooplankton were dominant in the coastal sea. Distinct clusters of prokaryotes and eukaryotes displayed clear spatial heterogeneity. Among the environmental parameters measured, temperature and salinity were key factors controlling bacterial and archaeal community structure, respectively, whereas N/P/Si contributed to eukaryotic variation. The relative contribution of environmental parameters to the microbial distribution pattern was 45.2%. Interaction analysis showed that Gammaproteobacteria, Alphaproteobacteria, and Flavobacteriia were the keystone taxa within the positive-correlation network, while Thermoplasmata was the main contributor in the negative-correlation network. Our study demonstrated that microbial communities are co-governed by environmental filtering and biotic interactions, which are the main deterministic driving factors modulating the spatiotemporal patterns of marine plankton synergistically at the regional or global levels.
Planktonic eukaryotes in the Chesapeake Bay: contrasting responses of abundant and rare taxa to estuarine gradients

Abstract:ABSTRACT Phytoplankton are important drivers of aquatic ecosystem function and environmental health. Their community compositions and distributions are directly impacted by environmental processes and human activities, including in the largest estuary in North America, the Chesapeake Bay. It is crucial to uncover how planktonic eukaryotes play fundamental roles as primary producers and trophic links and sustain estuarine ecosystems. In this study, we investigated the detailed community structure and spatiotemporal variations of planktonic eukaryotes in the Chesapeake Bay across space and time for three consecutive years. A clear seasonal and spatial shift of total, abundant, and rare planktonic eukaryotes was evident, and the pattern recurred interannually. Multiple harmful algal species have been identified in the Bay with varied distribution patterns, such as Karlodinium, Heterosigma akashiwo, Protoperidinium sp., etc. Compared to abundant taxa, rare subcommunities were more sensitive to environmental disturbance in terms of richness, diversity, and distribution. The combined effects of temporal variation (13.3%), nutrient availability (10.0%), and spatial gradients (8.8%) structured the distribution of eukaryotic microbial communities in the Bay. Similar spatiotemporal patterns between planktonic prokaryotes and eukaryotes suggest common mechanisms of adjustment, replacement, and species interaction for planktonic microbiomes under strong estuarine gradients. To our best knowledge, this work represents the first systematic study on planktonic eukaryotes in the Bay. A comprehensive view of the distribution of planktonic microbiomes and their interactions with environmental processes is critical in understanding the underlying microbial mechanisms involved in maintaining the stability, function, and environmental health of estuarine ecosystems. IMPORTANCE Deep sequencing analysis of planktonic eukaryotes in the Chesapeake Bay reveals high community diversity with many newly recognized phytoplankton taxa. The Chesapeake Bay planktonic eukaryotes show distinct seasonal and spatial variability, with recurring annual patterns of total, abundant, and rare groups. Rare taxa mainly contribute to eukaryotic diversity compared to abundant groups, and they are more sensitive to spatiotemporal variations and environmental filtering. Temporal variations, nutrient availability, and spatial gradients significantly affect the distribution of eukaryotic microbial communities. Similar spatiotemporal patterns in prokaryotes and eukaryotes suggest common mechanisms of adjustment, substitution, and species interactions in planktonic microbiomes under strong estuarine gradients. Interannually recurring patterns demonstrate that diverse eukaryotic taxa have well adapted to the estuarine environment with a long residence time. Further investigations of how human activities impact estuarine planktonic eukaryotes are critical in understanding their essential ecosystem roles and in maintaining environmental safety and public health.

Abstract:We assessed spatio-temporal patterns of diversity in deep-sea sediment communities using metabarcoding. We chose a recently developed eukaryotic marker based on the v7 region of the 18S rRNA gene. Our study was performed in a submarine canyon and its adjacent slope in the Northwestern Mediterranean Sea, sampled along a depth gradient at two different seasons. We found a total of 5,569 molecular operational taxonomic units (MOTUs), dominated by Metazoa, Alveolata and Rhizaria. Among metazoans, Nematoda, Arthropoda and Annelida were the most diverse. We found a marked heterogeneity at all scales, with important differences between layers of sediment and significant changes in community composition with zone (canyon vs slope), depth, and season. We compared the information obtained from metabarcoding DNA and RNA and found more total MOTUs and more MOTUs per sample with DNA (ca. 20% and 40% increase, respectively). Both datasets showed overall similar spatial trends, but most groups had higher MOTU richness with the DNA template, while others, such as nematodes, were more diverse in the RNA dataset. We provide metabarcoding protocols and guidelines for biomonitoring of these key communities in order to generate information applicable to management efforts.

Abstract:The Baltic Sea is a unique brackish water ecosystem that has been studied for decades; however, knowledge about the diversity of the benthic communities of bacteria and microbial eukaryotes within this system is sparse. Using an amplicon sequencing approach, we evaluated alpha and beta diversity of shallow-water coastal microbial sediment communities and their relationship to several environmental factors, on both a large spatial scale in the Baltic Sea and the adjacent North Sea, as well as on a temporal scale at selected sites along the salinity gradient in the Baltic Sea. We found salinity to be among the drivers of alpha diversity for bacteria but not for eukaryotes; however, community network structure appeared to change between sites of different salinity. Furthermore, beta diversity analyses revealed that communities in the poly- to mesohaline sections of the study area were affected to a higher degree by temperature, nutrient and sediment characteristics. This study shows that the structure of coastal microbial communities is to some degree related to salinity, but other factors also play a role.

Spatial variations of bacterial community composition in sediments of the Jiaozhou Bay, China

Abstract:Spatial variations of sediment microbes pose a great challenge for the estimation of anthropogenic influence on biogeochemical processes, yet remain very unclear in coastal ecosystems. Surface sediments in 9 stations from the eutrophic Jiaozhou Bay, China, were sampled, DNA was extracted within the sediments, and the 16S rDNA was sequenced with the Illumina Hiseq sequencing. Results reveal considerable heterogeneity of sediment bacteria in the Jiaozhou Bay, of which Proteobacteria and Bacteroidetes accounted for over 75%. Bacterial alpha-diversity indices decreased generally from the outside to the inner part of the bay and from the offshore to the nearshore area. Bacterial community structures of S3, S4, S7, and S8 clustered, those of S5, S13, and S14 grouped together, while those of S6 and S10 were distinct from each other and from those of the other stations. Major class Gammaproteobacteria were more abundant at the stations with mesoeutrophic to eutrophic levels (S4, S5, S8, and S10) and less abundant at oligotrophic stations (S6, S13, and S14), while Deltaproteobacteria had an opposite distribution pattern. Overall, bacterial community composition transitioned from being Xanthomonadales-dominant at S4 and S8 to being unidentifed_Gammaproteobacteria-dominant at S5, S6, S13, and S14, while in other stations there were comparable orders. The biogeochemical processes correspondingly changed from being nitrogen cycling-dominant at S4 and S8 to being sulfur cycling-dominant at S5, S6, S13, and S14. The bacterial distribution patterns were especially affected by the factors (dissolved organic phosphorus, DOP) in the overlying seawater due to the habitat status of P-insufficiency in the bay. Both orders Xanthomonadales and Alteromonadales could serve as bioindicators of anthropogenic pollution to different pollution types. At last, divergent distribution patterns of individual bacterial populations in the bay were revealed, the influential environmental gradients were clarified, and the uncertainty of microbes was reduced, helping to predict environmental functions in coastal areas.

Distinct bacterial and protist plankton diversity dynamics uncovered through DNA-based monitoring in the Baltic Sea area

Abstract:Planktonic microorganisms in coastal waters form the foundation of food webs and biogeochemical cycles while exposed to pronounced environmental gradients, especially brackish salinities. Yet, commonplace ecological assessment overlooks most of their diversity. Here, we analyzed the protist and bacterial diversity from new and publicly available DNA metabarcoding data collected alongside the Swedish marine monitoring program. We show that salinity, unlike other environmental factors, had a stronger effect on bacterial than protist community composition. The seasonality of protist but not bacterial families showed high geographic variation. Bacterial alpha diversity increased with dissolved inorganic nitrogen, while protist alpha diversity was highest in near-marine salinities. Microbial community composition patterns displayed interannual stability despite technical differences affecting the detection of rare taxa. Co-occurrence analysis identified clusters of potentially interdependent microorganisms. Bayesian modeling showed that the same bacterial lineages were less likely than protists to occur in both lower (<9 PSU) and higher (>15 PSU) brackish salinities. We propose that protists are less ecologically sensitive to salinity due to the disconnection of basic metabolic processes from the cell membrane through compartmentalization. Ultimately, incorporating DNA metabarcoding into an environmental monitoring program allowed us to connect ecological and biogeographic processes with understudied taxa and biodiversity dynamics.

Abstract:Phytoplankton are central components of marine environments, and are major players in the production and respiration budgeting. However, their diversity and distribution patterns are still poorly understood due largely to their small sizes and inconspicuous morphology that have been determined via the application of traditional morphology methods over the past two decades. To better understand the composition and diversity of phytoplankton in Jiaozhou Bay, China, seasonal sampling was carried out in 2019 and samples were analyzed with morphological observations and high-throughput sequencing, from which obvious seasonal variations in phytoplankton composition and proportional abundances were uncovered. Metabarcoding revealed far more diversity and species richness of phytoplankton than morphological observations, especially with respect to dinoflagellates. Diatoms were the most dominant phytoplankton group throughout the year, of which Thalassionema and Skeletonema were co-dominant in the bay. Parasitic dinoflagellates (e.g. Amoebophrya), which is often overlooked in the morphological observations, were in dominance and high diversity in the metabarcoding dataset, thus more attention should be paid to exploring the potential role of parasitic dinoflagellates. Temperature, chlorophyll a, and nutrient levels were the main influential factors on the distribution of phytoplankton. This study provided a comprehensive morphological and molecular description of phytoplankton and clearly demonstrated the importance of molecular technology in exploring phytoplankton communities. More-widespread use of molecular technology will facilitate deeper understanding of the ecological importance of the different species.

Abstract:ABSTRACT Microeukaryotic plankton (0.2–200 μm), which are morphologically and genetically highly diverse, play a crucial role in ocean productivity and carbon consumption. The Pacific Ocean (PO), one of the world’s largest oligotrophic regions, remains largely unexplored in terms of the biogeography and biodiversity of microeukaryotes based on large-scale sampling. We investigated the horizontal distribution of microeukaryotes along a 16,000 km transect from the west to the east of the PO. The alpha diversity indices showed a distinct decreasing trend from west to east, which was highly correlated with water temperature. The microeukaryotic community, which was clustered into the western, central, and eastern PO groups, displayed a significant distance-decay relationship. Syndiniales, a lineage of parasitic dinoflagellates, was ubiquitously distributed along the transect and dominated the community in terms of both sequence and zero-radius operational taxonomic unit (ZOTU) proportions. The prevailing dominance of Syndiniales-affiliated ZOTUs and their close associations with dinoflagellates, diatoms, and radiolarians, as revealed by SparCC correlation analysis, suggested that parasitism may be an important trophic strategy in the surface waters of the PO. Geographical distance and temperature were the most important environmental factors that significantly correlated with community structure. Overall, our study sheds more light on the distribution pattern of both alpha and beta diversities of microeukaryotic communities and highlighted the importance of parasitisms by Syndiniales across the tropical PO. IMPORTANCE Understanding the biogeographical and biodiversity patterns of microeukaryotic communities is essential to comprehending their roles in biogeochemical cycling. In this study, planktonic microeukaryotes were collected along a west-to-east Pacific Ocean transect (ca. 16,000 km). Our study revealed that the alpha diversity indices were highly correlated with water temperature, and the microeukaryotic communities displayed a distinct geographical distance-driven pattern. The predominance of the parasitic dinoflagellate lineage Syndiniales and their close relationship with other microeukaryotic groups suggest that parasitism may be a crucial survival strategy for microeukaryotes in the surface waters of the Pacific Ocean. Our findings expand our understanding of the biodiversity and biogeographical pattern of microeukaryotes and highlight the significance of parasitic Syndiniales in the surface ocean.

Abstract:Abstract Background Annually reoccurring microbial populations with strong spatial and temporal variations have been identified in estuarine environments, especially in those with long residence time such as the Chesapeake Bay (CB). However, it is unclear how microbial taxa cooccurr and how the inter-taxa networks respond to the strong environmental gradients in the estuaries. Results Here, we constructed co-occurrence networks on prokaryotic microbial communities in the CB, which included seasonal samples from seven spatial stations along the salinity gradients for three consecutive years. Our results showed that spatiotemporal variations of planktonic microbiomes promoted differentiations of the characteristics and stability of prokaryotic microbial networks in the CB estuary. Prokaryotic microbial networks exhibited a clear seasonal pattern where microbes were more closely connected during warm season compared to the associations during cold season. In addition, microbial networks were more stable in the lower Bay (ocean side) than those in the upper Bay (freshwater side). Multivariate regression tree (MRT) analysis and piecewise structural equation modeling (SEM) indicated that temperature, salinity and total suspended substances along with nutrient availability, particulate carbon and Chl a , affected the distribution and co-occurrence of microbial groups, such as Actinobacteria, Bacteroidetes, Cyanobacteria, Planctomycetes, Proteobacteria, and Verrucomicrobia. Interestingly, compared to the abundant groups (such as SAR11, Saprospiraceae and Actinomarinaceae), the rare taxa including OM60 (NOR5) clade (Gammaproteobacteria), Micrococcales (Actinobacteria), and NS11-12 marine group (Bacteroidetes) contributed greatly to the stability of microbial co-occurrence in the Bay. Modularity and cluster structures of microbial networks varied spatiotemporally, which provided valuable insights into the ‘small world’ (a group of more interconnected species), network stability, and habitat partitioning/preferences. Conclusion Our results shed light on how estuarine gradients alter the spatiotemporal variations of prokaryotic microbial networks in the estuarine ecosystem, as well as their adaptability to environmental disturbances and co-occurrence network complexity and stability.

Depth shapes α- and β-diversities of microbial eukaryotes in surficial sediments of coastal ecosystems

Abstract:Little is known about the relative influence of historic processes and environmental gradients on shaping the diversity of single-celled eukaryotes in marine benthos. By combining pyrosequencing of 18S ribosomal RNA genes with data on multiple environmental factors, we investigated the diversity of microeukaryotes in surficial sediments of three basins of the Yellow Sea Large Marine Ecosystem. A considerable proportion (about 20%) of reads was affiliated with known parasitoid protists. Dinophyta and Ciliophora appeared dominant in terms of relative proportion of reads and operational taxonomic unit (OTU) richness. Overall, OTU richness of benthic microeukaryotes decreased with increasing water depth and decreasing pH. While community composition was significantly different among basins, partial Mantel tests indicated a depth-decay pattern of community similarity, whereby water depth, rather than geographic distance or environment, shaped β-diversity of benthic microeukaryotes (including both the abundant and the rare biosphere) on a regional scale. Similar hydrographic and mineralogical factors contributed to the biogeography of both the abundant and the rare OTUs. The trace metal vanadium had a significant effect on the biogeography of the rare biosphere. Our study sheds new light on the composition, diversity patterns and underlying mechanisms of single-celled eukaryote distribution in surficial sediments of coastal oceans.
Metabarcoding of ichthyoplankton communities associated with a highly dynamic shelf region of the southwest Indian Ocean

Abstract:Drifting fish eggs and larvae (ichthyoplankton) can be identified to species using DNA metabarcoding, thus allowing for post hoc community analyses at a high taxonomic resolution. We undertook a regional-scale study of ichthyoplankton distribution along the east coast of South Africa, focused on the contrasting environments of the tropical Delagoa and subtropical Natal Ecoregions, and on exposed and sheltered shelf areas. Zooplankton samples were collected with tow nets at discrete stations along cross-shelf transects (20-200 m depth) spaced along a latitudinal gradient that incorporates a known biogeographical boundary. Metabarcoding detected 67 fish species, of which 64 matched prior distribution records of fishes from South Africa, with the remaining three known from the Western Indian Ocean. Coastal, neritic and oceanic species were present, from epi- and mesopelagic to benthopelagic and benthic adult habitats. By family, Myctophidae (10 species), Carangidae, Clupeidae, Labridae (each with 4 species) and Haemulidae (3 species) were most speciose. Ichthyoplankton community composition varied significantly with latitude, distance to coast, and distance to the shelf edge. Small pelagic fishes had the highest frequency of occurrence: Engraulis capensis, Emmelichthys nitidus and Benthosema pterotum increased in frequency towards the north, whereas Etrumeus whiteheadi increased towards the south. Chub mackerel Scomber japonicus accounted for most variability related to distance from the coast, whilst African scad Trachurus delagoa correlated with distance to the shelf edge. Dissimilarity between communities in the Delagoa and Natal Ecoregions was 98-100%, whereas neighbouring transects located within the sheltered KwaZulu-Natal Bight had lower dissimilarity (56-86%). Onshore transport of ichthyoplankton by Agulhas Current intrusions plausibly explained the abundance of mesopelagic species over the shelf. Metabarcoding followed by community analysis revealed a latitudinal gradient in the ichthyoplankton, associations with coastal and shelf-edge processes, and evidence of a spawning area in the sheltered KwaZulu-Natal Bight.

Abstract:Understanding the patterns of marine microbial diversity (Bacteria + Archaea) is essential, as variations in their alpha‐ and beta‐diversities can affect ecological processes. Investigations of microbial diversity from global oceanographic expeditions and basin‐wide transects show positive correlations between microbial diversity and either temperature or productivity, but these studies rarely captured seasonality, especially in polar regions. Here, using multiannual alpha‐diversity data from eight time series in the northern and southern hemispheres, we show that marine microbial community richness and evenness generally correlate more strongly with daylength than with temperature or chlorophyll a (a proxy for photosynthetic biomass). This pattern is observable across time series found in the northern and southern hemispheres regardless of collection method, DNA extraction protocols, targeted 16S rRNA hypervariable region, sequencing technology, or bioinformatics pipeline.

Abstract:Marine microbial communities vary seasonally and spatially, but these two factors are rarely addressed together. In this study, the temporal and spatial patterns of the bacterial and archaeal community were studied along a coast-to-offshore transect in the Eastern Mediterranean Sea (EMS) over six cruises, in three seasons of 2 consecutive years. Amplicon sequencing of 16S rRNA genes and transcripts was performed to determine presence and activity, respectively. The ultra-oligotrophic status of the Southeastern Mediterranean Sea was reflected in the microbial community composition dominated by oligotrophic bacterial groups such as SAR11, even at the most coastal station sampled, throughout the year. Seasons significantly affected the microbial communities, explaining more than half of the observed variability. However, the same few taxa dominated the community over the 2-year sampling period, varying only in their degree of dominance. While there was no overall effect of station location on the microbial community, the most coastal site (16 km offshore) differed significantly in community structure and activity from the three further offshore stations in early winter and summer. Our data on the microbial community compositions and their seasonality support previous notions that the EMS behaves like an oceanic gyre.

Abstract:Plankton studies serve as a basis for marine ecosystem research, but knowledge of marine plankton is still incomplete due to its extreme taxonomic and functional complexity. The application of metabarcoding is very valuable for the characterisation of the plankton community. The plankton community of the Southern Adriatic is subject to strong environmental fluctuations and changes, which underlines the need for frequent, reliable and comprehensive characterisation of the plankton. The aim of this study was to determine the taxonomic composition and seasonal distribution of eukaryotic plankton in the Southern Adriatic. Plankton samples were collected monthly for one year at the coastal station of the Southern Adriatic and metabarcoding was used for taxonomic identification. The results showed a high taxonomic diversity and dynamic seasonal distribution patterns for both the protist and metazoan plankton communities. Metabarcoding revealed both the core, year-round plankton community and previously unrecorded plankton organisms in the Southern Adriatic. The results provide for the first time a comprehensive overview of the plankton community in this area by metabarcoding. The identified seasonal patterns of plankton genera and species in the Southern Adriatic will contribute to the understanding of plankton interactions and future changes in community diversity characterisation.

Abstract:Chesapeake Bay is a large, dynamic, and productive temperate estuary, which plays an important role in transporting terrestrial materials into the Atlantic Ocean. The bacterioplankton communities experience strong environmental changes both seasonally and along the central axis of the Bay. The goal of this study was to explore the spatiotemporal and interannual patterns of the bacterioplankton communities in the estuary using high‐throughput sequencing of 16S rRNA genes. Deep sequencing uncovered many new bacterial groups not found in earlier studies, and showed that Chesapeake Bay contained highly diverse and dynamic bacterial communities. Bacterioplankton in the Bay in summer and autumn were more diverse and stable than those in winter and spring. The spatial pattern was overruled by strong temporal variations when the samples from all seasons were analyzed together. But, the spatial trend became more visible when samples were analyzed separately in individual seasons. Bacterial diversity indices decreased from the upper Bay to lower Bay. The spatial and temporal distributions of bacterioplankton were best explained by variations of salinity and temperature. In addition, nutrient availability (nitrogen and phosphorus), chlorophyll a, and light availability also contributed significantly to the spatiotemporal variation. Despite the interannual variation of the bacterioplankton communities, a stable and reoccurring spatiotemporal pattern was observed in the Chesapeake Bay, suggesting limited functional redundancy for planktonic estuarine microbiomes. Our results highlight the resilience of the planktonic bacterial communities in estuaries with long residence time, and different bacterial groups have unique responses/adaptation to dynamic environmental gradients.
