Abstract:ABSTRACT We investigated the role of mitochondrial function in the avian thermoregulatory response to a cold environment. Using black-capped chickadees (Poecile atricapillus) acclimated to cold (−10°C) and thermoneutral (27°C) temperatures, we expected to observe an upregulation of pectoralis muscle and liver respiratory capacity that would be visible in mitochondrial adjustments in cold-acclimated birds. We also predicted that these adjustments would correlate with thermogenic capacity (Msum) and basal metabolic rate (BMR). Using tissue high-resolution respirometry, mitochondrial performance was measured as respiration rate triggered by proton leak and the activity of complex I (OXPHOSCI) and complex I+II (OXPHOSCI+CII) in the liver and pectoralis muscle. The activity of citrate synthase (CS) and cytochrome c oxidase (CCO) was also used as a marker of mitochondrial density. We found 20% higher total CS activity in the whole pectoralis muscle and 39% higher total CCO activity in the whole liver of cold-acclimated chickadees relative to that of birds kept at thermoneutrality. This indicates that cold acclimation increased overall aerobic capacity of these tissues. Msum correlated positively with mitochondrial proton leak in the muscle of cold-acclimated birds while BMR correlated with OXPHOSCI in the liver with a pattern that differed between treatments. Consequently, this study revealed a divergence in mitochondrial metabolism between thermal acclimation states in birds. Some functions of the mitochondria covary with thermogenic capacity and basal maintenance costs in patterns that are dependent on temperature and body mass.

Abstract:Stochastic winter weather events are predicted to increase in occurrence and amplitude at northern latitudes and organisms are expected to cope through phenotypic flexibility. Small avian species wintering in these environments show acclimatization where basal metabolic rate (BMR) and maximal thermogenic capacity (MSUM) are typically elevated. However, little is known on intra-seasonal variation in metabolic performance and on how population trends truly reflect individual flexibility. Here we report intra-seasonal variation in metabolic parameters measured at the population and individual levels in black-capped chickadees (Poecileatricapillus). Results confirmed that population patterns indeed reflect flexibility at the individual level. They showed the expected increase in BMR (6%) and MSUM (34%) in winter relative to summer but also, and most importantly, that these parameters changed differently through time. BMR began its seasonal increase in November, while MSUM had already achieved more than 20% of its inter-seasonal increase by October, and declined to its starting level by March, while MSUM remained high. Although both parameters co-vary on a yearly scale, this mismatch in the timing of variation in winter BMR and MSUM likely reflects different constraints acting on different physiological components and therefore suggests a lack of functional link between these parameters.
Wintering Snow Buntings Elevate Cold Hardiness to Extreme Levels but Show No Changes in Maintenance Costs

Abstract:AbstractResident temperate passerines adjust their phenotypes to cope with winter constraints, with peak performance in metabolic traits typically occurring during the coldest months. However, it is sparsely known whether cold-adapted northern species make similar adjustments when faced with variable seasonal environments. Life in near-constant cold could be associated with limited flexibility in traits underlying cold endurance. We investigated this by tracking individual physiological changes over five consecutive winters in snow buntings (Plectrophenax nivalis), an Arctic-breeding migratory passerine typically confronted with nearly constant cold. Buntings were held in an outdoor aviary and exposed to seasonal temperature variation typical of temperate zone climates. We measured phenotypic changes in body composition (body, fat, and lean mass, pectoralis muscle thickness), oxygen transport capacity (hematocrit), metabolic performance (basal metabolic rate [BMR] and summit metabolic rate [Msum]), thermogenic endurance (time to reach Msum), and cold tolerance (temperature at Msum). Snow buntings showed flexibility in functions underlying thermogenic capacity and cold endurance comparable to that observed in temperate resident passerines wintering at similar latitudes. Specifically, they increased body mass (13%), fat mass (246%), hematocrit (23%), pectoralis muscle thickness (8%), and Msum (27%). We also found remarkable cold tolerance in these birds, with individuals reaching Msum in helox at temperatures equivalent to less than -90°C in air. However, in contrast with resident temperate passerines, lean mass decreased by 12%, and there was no clear increase in maintenance costs (BMR). Our results show that the flexibility of traits underlying thermal acclimatization in a cold-adapted northern species is comparable to that of temperate resident species living at lower latitudes and is therefore not limited by life in near-constant cold.

Abstract:Abstract Background Small birds in temperate habitats must either migrate, or adjust aspects of their morphology, physiology and behavior to cope with seasonal change in temperature and photoperiod. It is, however, difficult to accurately measure how seasonal changes in temperature and photoperiod affect physiological processes such as basal metabolic rate (BMR) and metabolic activity. To address this problem, we collected data in each month of the year on body mass ( M b ) and BMR, and conducted a series of experiments to determine the effect of temperature and photoperiod on M b , BMR and physiological markers of metabolic activity, in the Eurasian Tree Sparrow ( Passer montanus ). Methods In one experiment, we measured monthly change in M b and BMR in a captive group of birds over a year. In another experiment, we examined the effects of acclimating birds to two different temperatures, 10 and 30 °C, and a long and a short photoperiod (16 h light:8 h dark and 8 h light:16 h dark, respectively) for 4 weeks. Results We found that these treatments induced sparrows to adjust their M b and metabolic rate processes. Acclimation to 30 °C for 4 weeks significantly decreased sparrows’ M b , BMR, and energy intake, including both gross energy intake and digestible energy intake, compared to birds acclimated to 10 °C. The dry mass of the liver, kidneys and digestive tract of birds acclimated to 30 °C also significantly decreased, although their heart and skeletal muscle mass did not change significantly relative to those acclimated to 10 °C. Birds acclimated to 30 °C also had lower mitochondrial state-4 respiration (S4R) and cytochrome c oxidase (COX) activity in their liver and skeletal muscle, compared to those acclimated to 10 °C. Birds acclimated to the long photoperiod also had lower mitochondrial S4R and COX activity in their liver, compared to those acclimated to the short photoperiod. Conclusions These results illustrate the changes in morphology, physiology, and enzyme activity induced by seasonal change in temperature and photoperiod in a small temperate passerine. Both temperature and photoperiod probably have a strong effect on seasonal variation in metabolic heat production in small birds in temperate regions. The effect of temperature is, however, stronger than that of photoperiod.

Increasing Winter Maximal Metabolic Rate Improves Intrawinter Survival in Small Birds

Abstract:Small resident bird species living at northern latitudes increase their metabolism in winter, and this is widely assumed to improve their chances of survival. However, the relationship between winter metabolic performance and survival has yet to be demonstrated. Using capture-mark-recapture, we followed a population of free-living black-capped chickadees (Poecile atricapillus) over 3 yr and evaluated their survival probability within and among winters. We also measured the size-independent body mass (M<sub>s</sub>), hematocrit (Hct), basal metabolic rate (BMR), and maximal thermogenic capacity (Msum) and investigated how these parameters influenced survival within and among winters. Results showed that survival probability was high and constant both within (0.92) and among (0.96) winters. They also showed that while M<sub>s</sub>, Hct, and BMR had no significant influence, survival was positively related to Msum-following a sigmoid relationship-within but not among winter. Birds expressing an Msum below 1.26 W (i.e., similar to summer levels) had a <50% chance of survival, while birds with an Msum above 1.35 W had at least a 90% chance of surviving through the winter. Our data therefore suggest that black-capped chickadees that are either too slow or unable to adjust their phenotype from summer to winter have little chances of survival and thus that seasonal upregulation of metabolic performance is highly beneficial. This study is the first to document in an avian system the relationship between thermogenic capacity and winter survival, a proxy of fitness.

Abstract:Phenotypic flexibility has received considerable attention in the last decade; however, whereas many studies have reported amplitude of variation in phenotypic traits, much less attention has focused on the rate at which traits can adjust in response to sudden changes in the environment. We investigated whole-animal and muscle phenotypic changes occurring in black-capped chickadees (Poecile atricapillus) acclimated to cold (-5°C) and warm (20°C) temperatures in the first 3 hours following a 15°C temperature drop (over 3h). Before the temperature change, cold-acclimated birds were consuming 95% more food, were carrying twice as much body fat and had 23% larger pectoralis muscle fiber diameters than individuals kept at 20°C. In the 3 hours following the temperature drop, these same birds altered their pectoralis muscle ultrastructure by increasing the number of capillaries per fiber area and the number of nuclei per mm of fiber by 22%, consequently leading to a 22% decrease in myonuclear domain (amount of cytoplasm serviced per nucleus), whereas no such changes were observed in the warm acclimated birds. To our knowledge, this is the first demonstration of such a rapid adjustment in muscle fiber ultrastructure in vertebrates. These results support the hypothesis that chickadees maintaining a cold phenotype are better prepared than warm phenotype individuals to respond to a sudden decline in temperature, such as what may be experienced in their natural wintering environment.

Prehatching temperatures drive inter-annual cohort differences in great tit metabolism

Abstract:Basal metabolic rate (BMR) constitutes the lowest metabolic rate in a resting animal and is, therefore, considered to reflect the energetic cost of maintenance in endotherms. BMR is a reversible plastic trait that changes with environmental and ecological circumstances, albeit being heritable and susceptible to selection. Inter-individual variation within populations of small birds is substantial, and while many of the drivers of such variation have been identified, many remain unexplained. We studied winter BMR variation of juveniles over a 15-year period in a wild population of great tits Parus major at the northern border of their distribution. BMR during winter consistently changed between years, even after controlling for environmental factors, suggestive of a non-reversible developmental plasticity shaping the adult metabolic phenotype. BMR in cohorts of wintering great tits varied among winters as a response to minimum ambient temperatures experienced early in life, during the prehatching period. This developmental plasticity might be adaptive if temperatures experienced by growing embryos would metabolically prime them to an environment that they will likely encounter in future life. However, in line with a more unpredictable future climate, the risk of phenotype-environment mismatch is likely to lead to certain cohorts being poorly adapted to prevailing winter conditions, resulting in wider annual fluctuations in population size.

Abstract:Abstract Animals tend to decrease in body size (Bergmann’s rule) and elongate appendages (Allen’s rule) in warm climates. However, it is unknown whether these patterns depend on each other or constitute independent responses to the thermal environment. Here, based on a global phylogenetic comparative analysis across 99.7% of the world’s bird species, we show that the way in which the relative length of unfeathered appendages co-varies with temperature depends on body size and vice versa. First, the larger the body, the greater the increase in beak length with temperature. Second, the temperature-based increase in tarsus length is apparent only in larger birds, whereas in smaller birds, tarsus length decreases with temperature. Third, body size and the length of beak and tarsus interact with each other to predict the species’ environmental temperature. These findings suggest that the animals’ body size and shape are products of an evolutionary compromise that reflects distinct alternative thermoregulatory adaptations.

The performing animal: causes and consequences of body remodeling and metabolic adjustments in red knots facing contrasting thermal environments

Abstract:Using red knots (Calidris canutus) as a model, we determined how changes in mass and metabolic activity of organs relate to temperature-induced variation in metabolic performance. In cold-acclimated birds, we expected large muscles and heart as well as improved oxidative capacity and lipid transport, and we predicted that this would explain variation in maximal thermogenic capacity (Msum). We also expected larger digestive and excretory organs in these same birds and predicted that this would explain most of the variation in basal metabolic rate (BMR). Knots kept at 5°C were 20% heavier and maintained 1.5 times more body fat than individuals kept in thermoneutral conditions (25°C). The birds in the cold also had a BMR up to 32% higher and a Msum 16% higher than birds at 25°C. Organs were larger in the cold, with muscles and heart being 9-20% heavier and digestive and excretory organs being 21-36% larger than at thermoneutrality. Rather than the predicted digestive and excretory organs, the cold-induced increase in BMR correlated with changes in mass of the heart, pectoralis, and carcass. Msum varied positively with the mass of the pectoralis, supracoracoideus, and heart, highlighting the importance of muscles and cardiac function in cold endurance. Cold-acclimated knots also expressed upregulated capacity for lipid transport across mitochondrial membranes [carnitine palmitoyl transferase (CPT)] in their pectoralis and leg muscles, higher lipid catabolism capacity in their pectoralis muscles [β-hydroxyacyl CoA-dehydrogenase (HOAD)], and elevated oxidative capacity in their liver and kidney (citrate synthase). These adjustments may have contributed to BMR through changes in metabolic intensity. Positive relationships among Msum, CPT, and HOAD in the heart also suggest indirect constraints on thermogenic capacity through limited cardiac capacity.
Seasonal acclimatization of metabolism in Eurasian tree sparrows (Passer montanus)

Abstract:Acclimatization to winter conditions is an essential prerequisite for survival of small passerines of the northern temperate zone. Changes in photoperiod, ambient temperature and food availability trigger seasonal acclimatization in physiology and behavior of many birds. In the present study, seasonal adjustments in several physiological, hormonal, and biochemical markers were examined in wild-captured Eurasian tree sparrows (Passer montanus) from the Heilongjiang Province in China. In winter sparrows had higher body mass and basal metabolic rate (BMR). Consistently, the dry mass of liver, heart, gizzard, small intestine, large intestine and total digestive tract were higher in winter than in that in summer. The contents of mitochondrial protein in liver, and state-4 respiration and cytochrome c oxidase (COX) activity in liver and muscle increased significantly in winter. Circulating level of serum triiodothyronine (T3) was significantly higher in winter than in summer. Together, these data suggest that tree sparrows mainly coped with cold by enhancing thermogenic capacities through increased organ masses and heightened activity of respiratory enzymes activities. The results support the view that prominent winter increases in BMR are manifestations of winter acclimatization in tree sparrows and that seasonal variation in metabolism in sparrows is similar to that in other small temperate-wintering birds.
Age differences in night-time metabolic rate and body temperature in a small passerine

Abstract:Abstract Spending the winter in northern climes with short days and cold ambient temperatures ( T a ) can be energetically challenging for small birds that have high metabolic and heat loss rates. Hence, maintaining body temperature ( T b ) in T a below thermoneutrality can be energetically costly for a small bird. We still know little about how increased heat production below thermoneutrality affects the level at which T b is maintained, and if these patterns are age specific. To test this, we measured subcutaneous body temperature ( T s ) and resting metabolic rate (RMR) simultaneously in blue tits ( Cyanistes caeruleus ) during winter nights in T a ’s ranging from 25 to − 15 °C. RMR increased below the lower critical temperature (LCT, estimated at 14 °C) and was 6% higher in young (birds in their first winter) compared to old birds (birds in their second winter or older). The higher RMR was also mirrored in higher T s and thermal conductance ( C ) in young birds, which we suggest could be caused by age differences in plumage quality, likely driven by time constraints during moult. Reduction in nightly predicted T b was modest and increased again at the coldest ambient temperatures, suggesting that either heat retention or heat production (or both) improved when T a reached levels which are cold by the standards of birds in our population. Our results show that levels of heat production and T b can be age specific. Further studies should address age-specific differences on quality, structure, and thermal conductivity of plumage more explicitly, to investigate the role of variation in insulation in age-linked metabolic phenotypes.

Abstract:The discovery of prostate-specific antigen (PSA) as a biomarker represented a major discovery in the early diagnosis and monitoring of prostate cancer. However, the use of PSA is limited by the lack of specificity and an inability to differentiate indolent from life-threatening disease reliably at the time of diagnosis. A multitude of studies have aimed to improve the performance of PSA as well as identify additional biomarkers. The purpose of this study is to review available data on prostate cancer biomarkers for prostate cancer screening and prognostication, including prostatic acid phosphatase, PSA, PSA derivatives (PSA density, free PSA, pro PSA, and PSA kinetics), PCA3, GSTP1, AMACR, and other newly emerging molecular and genetic markers.
Limited heat tolerance in an Arctic passerine: Thermoregulatory implications for cold‐specialized birds in a rapidly warming world

Abstract:Arctic animals inhabit some of the coldest environments on the planet and have evolved physiological mechanisms for minimizing heat loss under extreme cold. However, the Arctic is warming faster than the global average and how well Arctic animals tolerate even moderately high air temperatures (Ta) is unknown. Using flow‐through respirometry, we investigated the heat tolerance and evaporative cooling capacity of snow buntings (Plectrophenax nivalis; ≈31 g, N = 42), a cold specialist, Arctic songbird. We exposed buntings to increasing Ta and measured body temperature (Tb), resting metabolic rate (RMR), rates of evaporative water loss (EWL), and evaporative cooling efficiency (the ratio of evaporative heat loss to metabolic heat production). Buntings had an average (±SD) Tb of 41.3 ± 0.2°C at thermoneutral Ta and increased Tb to a maximum of 43.5 ± 0.3°C. Buntings started panting at Ta of 33.2 ± 1.7°C, with rapid increases in EWL starting at Ta = 34.6°C, meaning they experienced heat stress when air temperatures were well below their body temperature. Maximum rates of EWL were only 2.9× baseline rates at thermoneutral Ta, a markedly lower increase than seen in more heat‐tolerant arid‐zone species (e.g., ≥4.7× baseline rates). Heat‐stressed buntings also had low evaporative cooling efficiencies, with 95% of individuals unable to evaporatively dissipate an amount of heat equivalent to their own metabolic heat production. Our results suggest that buntings' well‐developed cold tolerance may come at the cost of reduced heat tolerance. As the Arctic warms, and this and other species experience increased periods of heat stress, a limited capacity for evaporative cooling may force birds to increasingly rely on behavioral thermoregulation, such as minimizing activity, at the expense of diminished performance or reproductive investment.

Abstract:A classic example of ecophysiological adaptation is the observation that animals from hot arid environments have lower basal metabolic rates (BMRs, ml O 2 min −1 ) than those from non-arid (luxuriant) ones. However, the term ‘arid’ conceals within it a multitude of characteristics including extreme ambient temperatures ( T a , °C) and low annual net primary productivities (NPPs, g C m −2 ), both of which have been shown to correlate with BMR. To assess the relationship between environmental characteristics and metabolic rate in birds, we collated BMR measurements for 92 populations representing 90 wild-caught species and examined the relationships between BMR and NPP, T a , annual temperature range ( T r ), precipitation and intra-annual coefficient of variation of precipitation ( P CV ). Using conventional non-phylogenetic and phylogenetic generalized least-squares approaches, we found no support for a relationship between BMR and NPP, despite including species captured throughout the world in environments spanning a 35-fold range in NPP. Instead, BMR was negatively associated with T a and T r , and positively associated with P CV .
An arctic breeding songbird overheats during intense activity even at low air temperatures

Abstract:Birds maintain some of the highest body temperatures among endothermic animals. Often deemed a selective advantage for heat tolerance, high body temperatures also limits birds' thermal safety margin before reaching lethal levels. Recent modelling suggests that sustained effort in Arctic birds might be restricted at mild air temperatures, which may require reductions in activity to avoid overheating, with expected negative impacts on reproductive performance. We measured within-individual changes in body temperature in calm birds and then in response to an experimental increase in activity in an outdoor captive population of Arctic, cold-specialised snow buntings (Plectrophenax nivalis), exposed to naturally varying air temperatures (- 15 to 36 °C). Calm buntings exhibited a modal body temperature range from 39.9 to 42.6 °C. However, we detected a significant increase in body temperature within minutes of shifting calm birds to active flight, with strong evidence for a positive effect of air temperature on body temperature (slope = 0.04 °C/ °C). Importantly, by an ambient temperature of 9 °C, flying buntings were already generating body temperatures ≥ 45 °C, approaching the upper thermal limits of organismal performance (45-47 °C). With known limited evaporative heat dissipation capacities in these birds, our results support the recent prediction that free-living buntings operating at maximal sustainable rates will increasingly need to rely on behavioural thermoregulatory strategies to regulate body temperature, to the detriment of nestling growth and survival.

Abstract:In a seasonal world, organisms are continuously adjusting physiological processes relative to local environmental conditions. Owing to their limited heat and fat storage capacities, small animals, such as songbirds, must rapidly modulate their metabolism in response to weather extremes and changing seasons to ensure survival. As a consequence of previous technical limitations, most of our existing knowledge about how animals respond to changing environmental conditions comes from laboratory studies or field studies over short temporal scales. Here, we expanded beyond previous studies by outfitting 71 free-ranging Eurasian blackbirds ( Turdus merula ) with novel heart rate and body temperature loggers coupled with radio transmitters, and followed individuals in the wild from autumn to spring. Across seasons, blackbirds thermoconformed at night, i.e. their body temperature decreased with decreasing ambient temperature, but not so during daytime. By contrast, during all seasons blackbirds increased their heart rate when ambient temperatures became colder. However, the temperature setpoint at which heart rate was increased differed between seasons and between day and night. In our study, blackbirds showed an overall seasonal reduction in mean heart rate of 108 beats min −1 (21%) as well as a 1.2°C decrease in nighttime body temperature. Episodes of hypometabolism during cold periods likely allow the birds to save energy and, thus, help offset the increased energetic costs during the winter when also confronted with lower resource availability. Our data highlight that, similar to larger non-hibernating mammals and birds, small passerine birds such as Eurasian blackbirds not only adjust their heart rate and body temperature on daily timescales, but also exhibit pronounced seasonal changes in both that are modulated by local environmental conditions such as temperature. Abstract:Life history varies across latitudes, with the ‘pace of life’ being ‘slower’ in tropical regions. Because life history is coupled to energy metabolism via allocation tradeoffs and links between performance capacity and energy use, low metabolic intensity is expected in tropical animals. Low metabolism has been reported for lowland tropical birds, but it is unclear if this is due to ‘slow’ life history or to a warm, stable environment. We measured basal metabolic rates (BMR) of 253 bird species across a 2·6 km altitude gradient in Peru. We predicted higher BMR at high altitude due to lower temperatures leading to elevated thermoregulatory costs. We also tested for BMR differences between widely separated tropical regions (Peru and Panama), and between tropical‐ and temperate‐breeding birds. We found no effect of altitude on BMR in Peruvian species and no difference in BMR between Peruvian and Panamanian birds, suggesting that BMR in Neotropical birds is consistent and independent of environmental temperature. In a data set encompassing more than 500 species, tropical birds had significantly lower BMR than temperate‐breeding birds. In contrast to several recent analyses, we found higher BMR in passerine birds than in non‐passerines, independent of breeding latitude. Breeding latitude affects BMR, but diversity in avian life history within and between temperate and tropical regions may explain some of the residual variation in BMR after accounting for body mass and breeding latitude. Future studies of links between life history, metabolism and environmental factors might benefit from examining these variables within individual species as well as across broad geographic contrasts.
