Abstract:The effects of anaerobic treatment on the content of polyphenols, antioxidant capability, tocols, and free amino acids (FAAs) in the germinated brown rice (GBR) were investigated. The values of polyphenols and antioxidant capability significantly increased in treated groups, in which the three phenolic acids, i.e. ferulic, p-coumaric, and sinapic acids, showed the most pronounced increase. No significant change was observed in tocols. Among the evaluated FAAs, only the content of aspartic and glutamic acids in the white, red and black GBRs decreased by 75, 65 and 69%, and by 30, 18 and 39% after anaerobic treatment, respectively. While the contents of serine and threonine remained unchanged, all others significantly increased. In particular, the contents of γ-aminobutyric acid (GABA) increased 4.2-, 9.7- and 4.8-folds in the white, red and black GBRs, respectively. The results indicated that additional anaerobic treatment could be used in the production of GBR as a functional food.

Abstract:Brown rice (BR) contains more lipids, as compared with white rice, but its indigestibility and rancidity limit the consumer acceptability. Therefore, the objective of this study was to compare the effect of high-pressure (HP), freeze–thaw cycles (FTC) and germination-parboiling (GP) treatments on lipid in vitro digestibility and rancidity of BR. GP treated BR released the most fatty acids (especially palmitic acid and stearic acid) after in vitro digestion, followed by FTC treated BR. FTC treated BR kept the highest value of fat acidity during storage, while opposite results were observed in GP treated BR. Although HP treatment increased fat acidity value immediately, it slowed down the increase of fat acidity with storage. The results of conjugated dienes and malonaldehydes content in BR stored under accelerated conditions indicated better lipid oxidation stability after HP and GP treatment, and that it’s necessary for FTC treated BR products to be stored under anaerobic conditions.
Variation in Polyphenols, Tocols, Γ‐aminobutyric Acid, and Antioxidant Properties in Whole Grain Rice (oryza Sativa L.) As Affected by Different Germination Time

Abstract:Dynamic alterations in the polyphenols, tocols, antioxidant activities (AOA), and γ‐aminobutyric acid (GABA) contents of white, red, and black germinated rice upon different germination periods were studied. The results indicated that the contents of the free, bound, and total phenolics and AOA steadily increased during germination, even though the free part of colored germinated brown rice (GBR) showed a notable decline at the earlier stages, but still higher than the white rice. The compositions of the nine phenolic acids and six tocols generally accumulated in a time‐ and variety‐dependent manner during the germination process. However, the contents of p‐coumaric, ferulic, and sinapic acids displayed a pronounced increase with the duration of germination in all cultivars. The GABA in white, red, and black GBR accumulated to the highest content at the end of germination, which was 4.7, 14.2, and 6.7‐fold of their respective nongerminated counterparts. Interestingly, the total anthocyanin content in black GBR displayed a constant drop at earlier stages and then a rise at the later stages. These results suggest that prolonged germination may further increase the functional properties of GBR.
Extraction Optimization, Antidiabetic and Antiglycation Potentials of Aqueous Glycerol Extract from Rice (oryza Sativa L.) Bran

Abstract:The purpose of this investigation was to explore aqueous glycerol as food grade solvent for rice bran poly-phenolic antioxidants extraction and investigate the antidiabetic and antiglycation potential of the extract and its major polyphenols. Response surface methodology (Box-Behnken design) was implemented to optimize experimental conditions for rice bran polyphenols and antioxidants extraction. The optimum conditions provided a very satisfactory outcome in total polyphenols (549.6 +/- 8.05 mg gallic acid equivalent per 100 g dry weight) and DPPH scavenging activity (57.83 +/- 1.8%). The chromatographic separation of the extract showed higher concentrations of ferulic acid, rho-coumaric acid and chiorogenic acid followed by caffeic acid, syringic acid and rutin. Moreover, the extract inhibited alpha-amylase, alpha-glucosidase, and lipase activity as 34.2 +/- 3.2%, 29.9 +/- 1.0%, and 12.4 +/- 2.3%, respectively. Furthermore, fructose readily forms Advanced Glycation End Products (AGEs) fluorescence with basic amino acids in low concentration in simulated intestinal digestion conditions. The extract inhibited AGEs formation by 52.6% at 20 mu L/mL. Our study indicated that the anti-diabetic and antiglycation potential of rice bran extract was mainly driven by the extract polyphenols. Rice bran glycerol extract could be employed as a natural antidiabetic and antiglycation remedy, and as economically and eco-friendly source of potent bioactive compounds with satisfactory outcome.
Lipidomic and Antioxidant Response to Grape Seed, Corn and Coconut Oils in Healthy Wistar Rats

Abstract:Specialty oils differ in fatty acid, phytosterol and antioxidant content, impacting their benefits for cardiovascular health. The lipid (fatty acid, phytosterol) and antioxidant (total phenolics, radical scavenging capacity) profiles of grapeseed (GSO), corn (CO) and coconut (CNO) oils and their physiological (triacylglycerides, total and HDL-cholesterol and antioxidant capacity (FRAP) in serum and fatty acid and phytosterol hepatic deposition) and genomic (HL, LCAT, ApoA-1 and SR-BP1 mRNA hepatic levels) responses after their sub-chronic intake (10% diet for 28 days) was examined in healthy albino rats. Fatty acid, phytosterol and antioxidant profiles differed between oils (p ≤ 0.01). Serum and hepatic triacylglycerides and total cholesterol increased (p ≤ 0.01); serum HDL-Cholesterol decreased (p < 0.05); but serum FRAP did not differ (p > 0.05) in CNO-fed rats as compared to CO or GSO groups. Hepatic phytosterol deposition was higher (+2.2 mg/g; p ≤ 0.001) in CO- than GSO-fed rats, but their fatty acid deposition was similar. All but ApoA-1 mRNA level increased in GSO-fed rats as compared to other groups (p ≤ 0.01). Hepatic fatty acid handling, but not antioxidant response, nor hepatic phytosterol deposition, could be related to a more efficient reverse-cholesterol transport in GSO-fed rats as compared to CO or CNO.
Influence of Brown or Germinated Brown Rice Supplementation on Fecal Short-Chain Fatty Acids and Microbiome in Diet-Induced Insulin-Resistant Mice

Abstract:Intake of whole grain foods is associated with improving metabolic profile compared to refined grain products, but the underlying mechanism remains unclear. The present study examined the effects of brown rice (BRR) or germinated brown rice (GBR) supplementation on fecal short-chain fatty acids (SCFAs), and relationship with gut microbiota, metabolism and inflammation in high fat (HF)-diet-fed mice. The results demonstrated that an HF diet supplemented with BRR or GBR comparably increased the abundance of fecal isobutyric acid compared to that in mice receiving HF+white rice (WHR) diet (p < 0.01). The abundance of valeric acid in HF+GBR-diet-fed mice was higher than those receiving HF+WHR diet (p < 0.05). The abundances of fecal isobutyric acid negatively correlated with fasting plasma glucose, insulin, cholesterol, triglycerides, tumor necrosis factor-α, plasminogen activator inhibit-1, monocyte chemotactic protein-1 and homeostatic model assessment of insulin resistance (p < 0.01). The abundance of valeric acids negatively correlated with insulin resistance (p < 0.05). The abundances of isobutyric acid positively correlated with Lactobacillus, but negatively correlated with Dubosiella genus bacteria (p < 0.05). The findings demonstrated that the increases in SCFAs in the feces of BRR and GBR-treated mice were associated with improvements in gut microbiome, metabolic and inflammatory profile, which may contribute to the antidiabetic and anti-inflammatory effects of the whole grains in HF-diet-fed mice.

Abstract:Hyperlipidemia is a common clinical disorder of lipid metabolism in modern society and is considered to be one of the major risk factors leading to cardiovascular-related diseases. Germinated brown rice (GBR) is a typical whole grain food. The lipid-lowering effect of GBR has received increasing attention, but its mechanism of action is not fully understood. The gut microbiota has been proposed as a novel target for the treatment of hyperlipidemia. The aim of this study was to investigate the effects of GBR on the gut microbiota and lipid metabolism in high-fat diet (HFD)-fed C57BL/6J mice. The effect of GBR on hyperlipidemia was evaluated by measuring blood lipid levels and by pathological examination. The gut microbiota was detected by 16S rRNA sequencing, and the protein and mRNA expression levels involved in cholesterol metabolism were detected by western blotting and RT-qPCR to find potential correlations. The results showed that GBR supplementation could effectively reduce the levels of TC, TG, LDL-C and HDL-C in the serum and alleviate the excessive accumulation of fat droplets caused by HFD. Moreover, GBR intervention improved HFD-fed gut microbiota disorder via increasing the diversity of the gut microbiota, reducing the Firmicutes/Bacteroidetes ratio, and improving gut barrier damage. In addition, GBR could inhibit endogenous cholesterol synthesis and promote cholesterol transport and excretion. These findings suggest that GBR may be a competitive candidate for the development of functional foods to prevent abnormal lipid metabolism.
Evaluation of Extruded or Unextruded Double-Low Rapeseed Meal and Multienzymes Preparation in Pigs Nutrition During the Finishing Phase of Production

Abstract:The aim of this study was to compare the effects of extruded or non-extruded double-low rapeseed meal (DL-RSM) and multienzyme preparation on performance, nutrient digestibility, immune function and antioxidant status of the finishing pigs. Forty-eight pigs (Duroc×Landrace×Yorkshire) with an average weight of 62 kg were randomly divided into four groups with three replicates according to gender. Four diets were formulated to meet NRC (1998) nutrient requirements. Diet 1, based on the corn and soybean meal (SBM), was used as control. Diet 2 used 13% DL-RSM instead of the 11% SBM used in the Diet 1. Diet 3 used 13% extruded double-low rapeseed meal (E-DL-RSM) instead of 11% SBM, and Diet 4 0.03% multienzyme preparation was supplemented in Diet 3. The results showed that replacement of 11% SBM by 13% DL-RSM had no negative effects on performance and nutrient digestibility for finishing pigs. Extrusion and multienzyme preparation also had no beneficial effects on pig performance. However, the concentration of immunoglobulin G (IgG) in serum of DL-RSM treatment was lower (P<0.05) than that of the control, but it increased to the level of control in the treatment of extrusion combined with multienzyme preparation. The diet containing E-DL-RSM combined with multienzyme preparation significantly decreased the malondialdehyde (MDA) content (P<0.05) in serum. It was concluded that DL-RSM was an acceptable alternative to SBM as part of a protein supplement for finishing pig diets. In addition, extrusion and multienzymes preparation were not economically applied in the current study because of their extra cost.

Abstract:The rising prevalence of lifestyle diseases, such as type 2 diabetes, cardiovascular diseases, and metabolic syndrome, has increased the need for effective dietary interventions. This study aimed to evaluate the effects of heat-moisture-treated high-amylose rice (HA-HMT) on body weight, lipid metabolism, and gut microbiome composition in a rat model of obesity. Starch digestibility-specifically, resistant starch-has been shown to provide various health benefits, including improved metabolic health and gut microbiome composition. We employed a sequential approach: firstly, utilizing diet-induced obesity rat models fed with HMT-processed and HMT-non-processed low- or high-amylose rice to investigate the potential of amylose content or HMT to alter phenotypic characteristics and lipid metabolism; and secondly, using the optimal rice flour identified in the previous step to explore the underlying mechanisms. Our findings indicate that heat-moisture treatment, rather than the level of the amylose content of the rice, contributes to the observed anti-obesity and cholesterol-lowering effects. We identified candidate genes contributing to the cholesterol-regulating potential and demonstrated that HMT rice flour could influence the gut microbiome, particularly the Ruminococcus taxa. This study provides valuable insights into the health benefits of HA-HMT rice and supports its potential as a functional food ingredient in the management of obesity and cholesterol-related disorders.

Abstract:This study aimed to evaluate the effect of extrusion and of open-pan cooking on whole germinated and non-germinated grains of pearl millet (Pennisetum glaucum L. R. Br.), on its chemical-nutritional composition and in vitro iron bioavailability. The experimental design consisted of three flours: non-germination open-pan cooked millet flour (NGOPCMF), germination open-pan cooked millet flour (GOPCMF), and extrusion cooked millet flour (ECMF). The ECMF increased the carbohydrates, iron, manganese, diosmin, and cyanidin and decreased the total dietary fiber, resistant starch, lipids, and total vitamin E, in relation to NGOPCMF. The GOPCMF increased the lysine and vitamin C and decreased the phytate, lipids, total phenolic, total vitamin E, and riboflavin concentration, in relation to NGOPCMF. Furthermore, germinated cooked millet flour and extruded millet flour improved iron availability in vitro compared to non-germinated cooked millet flour. GOPCMF and ECMF generally preserved the chemical-nutritional composition of pearl millet and improved in vitro iron bioavailability; therefore, they are nutritionally equivalent and can be used to develop pearl millet-based products.
The Hypolipidemic and Antioxidant Activity of Wheat Germ and Wheat Germ Protein in High-Fat Diet-Induced Rats

Abstract:Background: So far, no articles have discussed the hypolipidemic effect of wheat germ protein in in vivo experiments. Objective: In this study, we investigated the effects of wheat germ protein (WGP, 300 mg/kg/day) and wheat germ (WG, 300 mg/kg/day) on cholesterol metabolism, antioxidant activities, and serum and hepatic lipids in rats fed a high-fat diet through gavage. Methodology: We used 4-week-old male Wistar 20 rats in our animal experiment. Biochemical indicators of fecal, serum and liver were tested by kits or chemical methods. We also conducted the cholesterol micellar solubility experiment in vitro. Results: After 28 days of treatment, our results showed that WGP significantly reduced the serum levels of total cholesterol (p < 0.05) and nonhigh-density lipoprotein cholesterol (p < 0.05), improved the enzymatic activities of cholesterol 7-α hydroxylase (p < 0.01) and low-density lipoprotein receptor (p < 0.01) and increased bile acid excretion in feces (p < 0.05). Conclusion: WG did not significantly increase bile acid excretion in feces or decrease serum levels of total cholesterol. Moreover, WGP and WG both presented significant antioxidant activity in vivo (p < 0.05) and caused a significant reduction in cholesterol micellar solubility in vitro (p < 0.001). Therefore, WGP may effectively prevent hyperlipidemia and its complications as WGP treatment enhanced antioxidant activity, decreased the concentration of serum lipids and improved the activity of enzymes involved in cholesterol metabolism.

Abstract:Abstract <sec> Objectives Non-alcoholic fatty liver disease (NAFLD) prevalence is increasing, and no pharmacological intervention exists to treat it. Here, we determined the effect of feeding wild rice and brown rice on liver lipids and the gut microbiome when fed two different high fat background diets meant to induce fatty liver. </sec> <sec> Methods In the HFD trial, male Wistar rats (n = 8 per group) were fed either a high fat diet (HF), or a HF containing either 20% wild rice (WR) or 20% brown rice (BR) for 8 weeks. Cecal contents, liver, and epididymal fat pads were harvested. Microbiome analysis of cecal contents was conducted by 16S sequencing. In the TWD trial, male Wistar rats (n = 12 per group) were fed the AIN-93G diet (Basal), the Total Western Diet (TWD; a high fat diet that mimics the average American diet), or the TWD containing 30% WR (WR) for 10 weeks, then liver and epididymal fat pads were harvested. </sec> <sec> Results In the HFD trial, both WR and BR significantly reduced total liver lipids compared to HF (p < 0.0001). However, epididymal fat pad weight was not reduced in either of the rice groups compared to HF (p > 0.05). Serum cholesterol was also reduced in WR compared to HF (p < 0.0188). In the TWD trial, the TWD elevated total liver lipids compared to Basal, but WR reduced liver lipids significantly (p < o.oo6) compared to TWD. WR did not lower epididymal fat pad weight (p > 0.1) compared to TWD. Additionally, p-AMPK/AMPK was increased in WR compared to TWD (p < 0.003). This shift in AMPK activation was not seen in the HFD trial (p > 0.05). In the HFD trial microbiome analysis, the beta-diversity measure of community dissimilarity indicated that WR had a significantly different microbial profile compared to the animals fed the HF diet (p < 0.003) while BR had an increased abundance of lactobacillus (p < 0.03) compared to animals fed the HF diet. </sec> <sec> Conclusions Wild rice feeding reduced fatty liver, using two different high fat background diets, and altered the gut microbiome. Wild rice should be further explored as a dietary approach to prevent development of non-alcoholic fatty liver disease (NAFLD), and to investigate whether hepatic lipid reductions are mediated by changes in the gut microbiome. </sec> <sec> Funding Sources The TWD trial was sponsored by the Whiteside Institute for Clinical Research and the HFD trial was sponsored by the Minnesota Cultivated Wild Rice Council. </sec>
Comparative Effect of Amylose Concentration in Brown and White Rice on Hepatocellular Insufficiency Using Rat Bioassay

Abstract:Background: The scientific name of rice is Oryza sativa. It is one of the important staple human foods. There are more than 110,000 cultivated varieties of rice, most widely known and grown. Nutrient concentration may vary depending on production, environment, pre- and post-harvesting treatment, and processing. Cooked rice with high amylose content effectively controls metabolic disorders, serum blood glucose levels, and lipid profile and is also effective in weight loss. Objectives: This study aimed to determine the comparative effect of amylose concentration in brown and white rice on hepatocellular insufficiency in healthy rat models. Methods: We divided 25 rats into 5 groups (G0, G1, G2, G3, and G4). There were 5 rats in each group. Group 0 was the control group on a normal diet. Group 1 was fed white rice flour, G2 was fed cooked white rice, G3 was fed brown rice flour, and G4 was fed cooked brown rice. Free access to food and water was secured. Weight changes, blood glucose, and lipid profiles were analyzed pre and post-treatment. Results: Rats on white rice feed showed weight gain, increased blood glucose, cholesterol, triglycerides, and low HDL levels. However, rats on brown rice feed showed less weight gain (P < 0.05), a lower increase in blood glucose (P < 0.05), high HDL, lower LDL cholesterol, and triglycerides. The pancreas and liver histopathology showed no major changes due to the short trial duration. Conclusions: Rats fed white rice showed ineffective results disturbing biological parameters as compared to those rats which were fed brown rice. Due to low amylose content and the presence of antioxidants, flavonoids, and high fiber content, brown rice shows beneficial effects in controlling and maintaining relevant biological parameters.

Abstract:Dietary interventions are receiving increasing attention for maintaining host health and diminishing disease risk. This study endeavored to elucidate the intervention effect of chlorogenic acid coupled with extruded rice starch (CGA-ES) in mitigating lipid metabolism disorders induced by a high-fat diet (HFD) in rats. First, a significant increase in resistant starch (RS) and a decrease in the predicted glycemic index (pGI) were observed in CGA-ES owing to the formation of an ordered structure (Dm, single helix, and V-type crystalline structure) and partly released CGA. Compared to a physical mixture of starch and chlorogenic acid (CGA + S), CGA-ES showed a more potent effect in alleviating lipid metabolism disorders, manifesting as reduced levels of blood glucose, serum total cholesterol (TC), triglycerides (TG), aspartate aminotransferase (AST), alanine transaminase (ALT) and alkaline phosphatase (AKP), as well as body weight. It is correlated with an improvement in the gut microecology, featuring bacteria known for cholesterol reduction and butyrate production ( Butyricicoccus , Bifidobacterium , Fusicatenibacter , Turicibacter , and Enterorhabdus ), along with bile acid, butyrate and PG (PG (17:0/16:0) and PG (18:1/16:0)). The RS fraction of CGA-ES was found to be the main contributor. These findings would provide evidence for future studies to regulate lipid metabolism disorders, and even obesity using CGA-ES.

Abstract:Rice bran is a waste product with low cost and high fiber content, giving it an added advantage over corn and soybean meal, which have to be purchased and always at a relatively higher cost. Under the background of increased attention to sustainable agriculture, it is significant to find alternative uses for this byproduct. A total of 35 finishing pigs were allotted to five dietary treatments: a control group with basal diet and four experimental diets where corn was equivalently substituted by 7%, 14%, 21%, and 28% defatted rice bran (DFRB), respectively. With increasing levels of DFRB, the neutrophil to lymphocyte ratio (NLR) linearly decreased (p < 0.05). In the jejunum, the mRNA level of nuclear factor erythroid-2 related factor-2 (Nrf2) exhibited a quadratic response (p < 0.01) with incremental levels of DFRB. In the colon, the mRNA levels of mucin 2 (MUC2), Nrf2, and NAD(P)H: quinone oxidoreductase 1 (NQO1) were upregulated (linear, p < 0.05) and heme oxygenase-1 (HO-1) was upregulated (linear, p < 0.01). Overall, using DFRB to replace corn decreased the inflammatory biomarkers of serum and showed potential function in modulating the intestinal barrier by upregulating the mRNA expression levels of MUC2 and downregulating that of Nrf2, NQO1, and HO-1 in the colon.

Abstract:A search for inhibitors of 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1) from Walsura cochinchinensis yielded 10 new limonoids, cochinchinoids A-J (1-10), and two new triterpenoids, 3-epimesendanin S (11) and cochinchinoid K (12). Their structures were assigned on the basis of spectroscopic data, with the absolute configurations of 1 and 12 being established by X-ray diffraction analysis. Of these compounds, cochinchinoid K (12) displayed inhibitory activity against mouse 11β-HSD1 with an IC50 value of 0.82 μM.
Optimization of Extrusion Treatments, Quality Assessments, and Kinetics Degradation of Enzyme Activities during Storage of Rice Bran

Abstract:Over the years, extrusion has been a multi-step thermal technique that has proven to be the most effective process to stabilize rice bran (RB). This study aimed to investigate the effects of extrusion treatment and temperature (15, 25, and 40 °C) on the storage stability, lipid oxidation, peroxidase, and peroxide values, free fatty acids, fatty acid composition, and protein variations of RB over 60 days. The study offers novel insights into the changes in RB's protein and amino acid compositions during extrusion and storage, which has not been extensively explored in prior research. After extrusion processing, peroxidase activity (POD) and lipase activity (LPS) were significantly reduced. However, peroxide value (PV), free fatty acids (FFA), and malondialdehyde content (MDA) observed a significantly increased by 0.64 mEqO2/kg, 8.3 mg/100 g, and 0.0005 μmol/L respectively. The storage stability of RB after extrusion shows that the POD, LPS, FFA, PV, and MDA were positively correlated with storage duration and temperature. The oleic acid/linoleic acid ratio in processed RB by extrusion had no significant changes during storage. The total and essential/non-essential amino acid ratios showed a downward trend of 5.26% and 8.76%, respectively. The first-order kinetics was the best-fitting model to describe the enzymatic inactivation and degradation of extruded RB during storage. The extrusion treatment did not affect the crude protein and the essential subunits of protein. Overall, the optimized extrusion procedure exhibited promising results in stabilizing the RB.

Abstract:A novel branch-line coupler with three controllable operating frequencies is presented for tri-band operation. The tri-band operation is achieved by attaching a matching circuit to each port of the conventional branch-line coupler. Closed-form design equations are derived. For verification, a microstrip 3 dB branch-line coupler operating at 0.9/2/2.45 GHz is designed, fabricated and measured. The measured results are in good agreement with the simulated results. When the return loss and isolation are better than 15 dB and 20 dB simultaneously, the measured bandwidths of three operating bands are all almost 40 MHz, while the amplitude imbalance is below 0.8 dB and the phase difference is within .

Abstract:Rice bran is an important byproduct of the rice polishing process, rich in nutrients, but it is underutilized and often used as feed or discarded, resulting in a huge amount of waste. In this study, rice bran was fermented by Lactobacillus fermentum MF423 to obtain a product with high antioxidant activity. First, a reliable and efficient method for assessing the antioxidant capacity of the fermentation products was established using high-performance liquid chromatography (HPLC), which ensured the consistency of the batch fermentation. The fermented rice bran product (FLRB) exhibited significant antioxidant activity in cells, C. elegans, and hyperlipidemic mice. Transcriptome analysis of mouse livers showed that the expression of plin5 was upregulated in diabetic mice administered FLRB, thereby preventing the excessive production of free fatty acids (FFAs) and the subsequent generation of large amounts of reactive oxygen species (ROS). These studies lay the foundation for the application of rice bran fermentation products.

Abstract:We report a case of diverticulitis affecting a colonic segment used as an interposition graft following laryngopharyngectomy. The patient presented as an emergency to our department with a history of a red, swollen and painful neck. She had undergone laryngopharyngectomy for laryngeal cancer in 1967. Computed tomography imaging revealed several diverticula in the colonic graft and associated abscess formation. The patient's clinical condition behaved similarly to that of conventional colonic diverticulitis. The difficulties in reaching a definite diagnosis and management of this unusual complication following laryngopharyngectomy are discussed.
