Abstract:This study investigated the impact of packaging systems on the degradation and oxidation of beef proteins regarding beef tenderness of longissimus lumborum (LL) and biceps femoris (BF) muscles stored in vacuum skin packaging (VSP), a modified atmosphere with high oxygen concentration (MAP), and combined of these two methods (VSP+MAP). A significant decrease in the Warner-Bratzler shear force (WBSF) in VSP at D14 and D28 for LL was observed compared to BF. A significant effect of packaging system on troponin-T (Tn-T) and desmin degradation was shown (p≤0.001). A high concentration of oxygen in MAP and VSP+MAP affected protein oxidation, which was reflected in myosin oxidative cross-linking. An increase of WBSF values detected in steaks packed in VSP and VSP+MAP systems could be caused by the intensification of protein oxidation. Furthermore, BF was more susceptible to oxidation compared to LL. The VSP+MAP packaging system has resulted in the maintenance of a bright, red color, however has not improved the beef tenderness.

Abstract:Superchilling and modified atmosphere packaging(MAP) are two important preservation technologies of meat. The research combined the two kinds of technologies. It took Lanxi pigs as experimental material. The indexes included p H,water holding capacity,total bacterial count and sensory score. Firstly,the effects of pork were compared under conditions of 4℃ cold storage,-2℃ superchilling storage and-18℃ frozen storage.Then,the influence of CO2 and O2on quality of pork was researched partially under-2℃ superchilling. At last,the best combination of CO2 and O2was determined. The result showed that pork under-2℃ superchilling storage could keep normal quality in 15 days. CO2 could lower p H of pork and inhibits the growth of microorganism in it,while O2 could keep good color but promote the growth of microorganism. Finally,the experiment of CO2/O2 Combination shows that 40% CO2+60% O2 was the best MAP composition under-2℃superchilling storage. The fresh period was as long as 24 days.
Use of sustainable packaging materials for fresh beef vacuum packaging application and product assessment using physicochemical means

Abstract:Packaging material should guarantee the longest possible shelf life of food and help to maintain its quality. The aim of the study was to assess the physicochemical changes taking place during 28-day ageing of beef steaks packed in two types of multilayer films containing biodegradable polymers - polylactic acid (NAT/PLA) and Mater-Bi® (NAT/MBI). The control group consisted of steaks packed in synthetic polyamide/polyethylene (PA/PE) film. The samples stored in NAT/PLA had significantly lower purge loss than the control samples and the lowest expressible water amount after 14 and 21 days. Following blooming, the most favourable colour was shown in steaks stored in NAT/MBI, with the highest values for the L*, a* and C* parameters and the R630/580 ratio, a high proportion of oxymyoglobin, and the lowest share of metmyoglobin. All steaks, regardless of the type of packaging material, had acceptable tenderness and were stable in terms of lipid oxidation.

Abstract:The totally subcutaneous implantable cardioverter‐defibrillator (S‐ICD) is an entirely novel defibrillation device that avoids the direct contact of device electrodes with the heart and the cardiovascular system. Here, we present a particular case of a young woman with congenital long‐QT syndrome in which we implanted the electrode alternatively, right parasternally. This decision was based on the thoracic anatomy of the patient and on findings of a model of S‐ICD electrodes in an adult torso. In conclusion, in some patients an alternative subcutaneous electrode position may be carefully considered but should not be taken to outweigh the standard left‐sided placement. Investigation of oxygen packaging to maintain beef color stability and microbiology safety after periods of long-term superchilled storage

Abstract:This study aimed to develop an appropriate modified atmosphere packaging (MAP) system for displayed beef steaks following long-term superchilled (-1 °C) storage. After superchilled storage for 0, 2, 8, or 16 weeks, beef loins were fabricated into steaks and displayed with 20%, 50%, or 80% O2-MAP under chilled conditions. At each storage point, after display for 0, 3, 7, or 10 days, instrumental color, myoglobin redox forms percentage, lipid oxidation, total viable count (TVC), and total volatile basic nitrogen (TVB-N) were evaluated. Meat color stability decreased, with prolonged storage period and display time. When the storage period was within 8 weeks, under all the above MAP conditions, the display time for the beef steaks was up to 10 days. Considering 80% O2-MAP promoted lipid oxidation, 50% and 80% O2-MAP were not recommended for displaying steaks for more than 10 and 7 days respectively after 16 weeks of storage. However, 20%, 50%, or 80% O2-MAP could maintain 3 days of microbial shelf-life according to TVC and TVB-N results. Additionally, after long-term superchilled storage for 16 weeks, the various O2 concentrations had minimal impact on microbiota succession during the MAP display period. Furthermore, beef steaks packaged under various MAP systems exhibited similar microbial compositions, with the dominant bacteria alternating between Lactobacillus and Carnobacterium. This study provided practical guidance for improving beef color stability after long-term superchilled storage.

Abstract:This study explores the influence of modified atmosphere packaging (MAP) on fresh lamb meat quality with respect to gas concentration, rigor state, and post-mortem aging time. A comparison was done for the quality characteristics of lamb Longissimus thoracis lumborum chops that had been packaged separately in air, 75%O2 + 25%CO2 MAP or 50%O2 + 50%CO2 MAP at 1, 6, and 24 h post-mortem and then stored for 6, 12, 24, 72, and 144 h post-mortem, and the quality of lamb chops had been evaluated at each post-mortem period separately. Chops packaged at 1 and 6 h post-mortem in MAP had reduced pH decline, less purge loss, and enhanced redness at early post-mortem storage times. Lamb color stability was evidently greater in 75%O2 + 25%CO2 MAP than in 50%O2 + 50%CO2 MAP during the early storage period when a* and R630/R580 were taken into account. Shear force values were lowest in lambs packaged at 1 h post-mortem with 75%O2 + 25%CO2 MAP at 12 h post-mortem and then increased until 72 h post-mortem, suggesting that rigor has been delayed by such a high O2 MAP. Thus, fresh lamb quality was maintained most effectively when packaged at 1 h post-mortem in 75%O2 + 25%CO2 MAP for consumption at 12 h post-mortem. The exact mechanisms and optimization of MAP based on Chinese retail conditions should be considered in future studies. PRACTICAL APPLICATION: In this study, three slaughter patterns in the meat industry involving boning immediately after dressing (hot-boning) and chilling for a short period (warm-boning) or overnight (cold-boning) are considered, as well as the behavior of non-immediate consumption after purchase. Modified atmosphere packaging provides an effective preservation of early post-mortem muscles with enhanced color stability, water holding capacity, and texture during refrigerated storage. This could provide new insights into how to process lamb muscles in the early post-mortem period to improve and stabilize lamb quality.
Colour stability of beef from different Spanish native cattle breeds stored under vacuum and modified atmosphere

Abstract:Beef steaks from five Spanish cattle breeds were submitted to three packaging conditions: (a) 15 days under vacuum, (b) 15 days under modified atmosphere (MAP) (60% O(2), 30% CO(2) and 10% N(2)), and (c) 10 days under vacuum plus 5 days more under MAP. CIE L(∗)a(∗)b(∗) coordinates, chroma (C(∗)) and hue (H(∗)), relative concentration of myoglobin, oxymyoglobin and metmyoglobin at the meat surface and sensory evaluation of colour were determined 0, 5, 10 and 15 days after packaging. Beef under MAP showed higher lightness (L(∗)) and hue (H(∗)) and lower redness (a(∗)) and chroma (C(∗)) than beef under vacuum. Colour of beef under MAP was not acceptable after 15 days of storage, due to the high metmyoglobin concentration. Yellowness (b(∗)) was the indicator of differences due to ageing and differences in colour between breeds were related to L(∗), a(∗) and H(∗) values. Packaging conditions had a greater effect on beef colour than breed, but breed differences might change with packaging atmosphere. Both packaging and breed are major factors in packaged beef quality.
Effects of microbiota dynamics on the color stability of chilled beef steaks stored in high oxygen and carbon monoxide packaging

Abstract:The microbiota influence on meat color stability of chilled beef steaks under two modified atmosphere packaging (MAP) systems was studied: HiOx-MAP (80% O2/20% CO2) and CO-MAP (0.4% CO/30% CO2/69.6% N2). Steaks were stored for up to 20 days at 2 °C. Headspace gas composition and color stability were evaluated. High-throughput sequencing analyses were performed to characterize bacterial community dynamics. For HiOx-MAP steaks, Pseudomonas spp. became the dominant species, and an increased oxygen consumption caused by these bacteria contributed to metmyoglobin (MetMb) formation from day 10. Functional analysis based on 16S rDNA sequencing predicted higher abundance of genes related to amino acid and lipid metabolism in HiOx-MAP bacterial communities compared to CO-MAP bacterial communities, which probably accelerated meat discoloration. Lactic acid bacteria dominated in CO-MAP steaks, in which Lactobacillus spp. and Lactococcus spp. contributed to improve redness and chroma, and reduce MetMb%, respectively. These findings help our understanding of color stability in CO-MAP steaks in comparison to HiOx-MAP steaks as influenced by the microbial community.

Abstract:Abstract Metmyoglobin reducing activity (MRA) and oxygen consumption (OC) are two important biochemical processes that can influence beef color. Several studies have determined the effects of aging and modified atmospheric packaging (MAP) on biochemical properties of normal-pH beef (5.5–5.7 pH range). However, limited knowledge is currently available on the effects of aging and MAP on biochemical properties of high-pH beef (> 6.2). Therefore, the objective was to determine the effects of extended aging, MAP, and display time on MRA and OC of high-pH beef. Ten USDA Choice (pH = 5.6) and ten ‘No-Roll’ dark cutting (high-pH; pH > 6.4) strip loins were obtained from a commercial packing plant within 72 hours of harvest. Loins were vacuum packaged and aged for 0, 21, 42, and 62 days at 4 °C. Following aging, from each normal- and high-pH loin sections, 2.5cm thick steaks were cut and packaged in PVC, high oxygen- (HiOx-MAP), and carbon monoxide modified atmospheric (CO-MAP) packaging. The surface color, MRA, and OC were determined on day 0 and 6 of the respective aging period. The experiment was replicated ten times (n = 10). High-pH steaks in all packaging and aging times had more stable red color than normal-pH steaks. High-pH steaks packaged in PVC and HiOx-MAP had greater (P 0.05) in MRA between normal- and high-pH steaks on day 21 when steaks were packaged in CO-MAP, but high-pH had greater (P < 0.05) MRA on day 62. At all aging periods, high-pH steaks had greater (P < 0.05) OC than normal-pH steaks. However, a decrease in OC was greater (P < 0.05) for high-pH than normal-pH steaks. The results suggest that normal- and high-pH beef have different trends for the changes in MRA and OC.

The response of bacterial communities to carbon dioxide in high-oxygen modified atmosphere packaged beef steaks during chilled storage

Abstract:The objective of this study was to establish the effect of CO2 on the bacterial community in beef steaks held under both high-oxygen modified atmosphere packaging (HiOx-MAP) types (CO2 treated MAP: 50% O2/40% CO2/10% N2; control MAP: 50% O2/50% N2). Steaks were stored at 2 °C for 20 days. Gas composition, meat color, pH values, total volatile basic nitrogen values, total viable counts (TVC) and microbial community dynamics were monitored. Compared to the control MAP, the high level of CO2 in the contrast MAP significantly delayed bacterial growth, resulting in a bright red color as well as extending the shelf-life to over 20 days. The microbial diversity decreased with prolonged storage in both MAP types, but it was more complex in high-CO2 treated MAP steaks. When TVC values approached the shelf-life threshold for the control MAP, Pseudomonas and Brochothrix were the predominant bacteria, while Pseudomonas and Serratia under the CO2 containing MAP were at a lower abundance than under the control MAP. The dominant Pseudomonas species causing spoilage in the control MAP steaks was P. fragi, and this species was inhibited significantly by CO2, followed by P. weihenstephanensis. Inversely, P. versuta instead of P. fragi became the dominant Pseudomonas species under the CO2 treated MAP. Overall, the application of CO2 in HiOx-MAP influenced microbiota succession, which played an important role in retaining beef quality.

Abstract:Volatile compounds, carbonyls, non-heme iron, and thiobarbituric acid reactive substances (TBARS) were measured in both raw and cooked beef samples to determine the effects of muscle and packaging type on beef flavor development. All paired strip loins and top sirloin butts were packaged under vacuum and aged for 14 d postmortem. After initial aging, all subprimals were fabricated to produce M. gluteus medius (GM) or M. longissimus lumborum (LL) steaks. At 14 d postmortem, steaks were randomly assigned to 1 of 5 package types: high-oxygen modified atmosphere lidded trays (80% O2/20% CO2 [“HIOX”]), carbon monoxide modified atmosphere lidded trays (0.4% CO/30% CO2/69.6% N2 [“CO”]), rollstock (forming and non-forming films [“ROLL”]), vacuum packaging without retail display (“VAC”), and traditional overwrap (“OW”) remained under vacuum prior to retail display. Steaks were stored in darkness an additional 7 d prior to display. At 21 d postmortem, HIOX, OW, CO, and ROLL packages were displayed for 48 h under continuous fluorescent lighting, while VAC steaks remained in dark storage. Packaging and muscle type impacted (P < 0.05) quantities of multiple volatile flavor compounds, including alcohols, n-aldehydes, esters, furans, hydrocarbons, sulfur-containing compounds,and ketones in both raw and cooked samples. Volatile compounds related to lipid oxidation were more (P < 0.05) prominent in HIOX packaging. Package type (P < 0.05) and muscle (P < 0.05) had an impact on raw-steak TBARS, although package type did not influence (P > 0.05) cooked-steak TBARS. The GM possessed greater (P < 0.05) TBARS values than the LL in both raw and cooked samples. Package type had no effect (P > 0.05) on carbonyl and non-heme iron content although these analyses differed among muscles (P < 0.05), with the GM being greater (P < 0.05) than the LL. These results indicate that the development of lipid oxidation that occurs during storage and display was muscle and packaging specific. Therefore, to maintain flavor, quality packaging systems should be selected on a muscle-specific basis.
Influence of oxygen concentration on the fresh and internal cooked color of modified atmosphere packaged dark-cutting beef stored under chilled and superchilled conditions

Abstract:The current research evaluated the combined effect of oxygen concentration (20%, 50% and 80% O2) and storage temperature (4°C or -1.5°C) on the fresh and internal cooked color of dark-cutting (DC) beef. Steaks were packaged in modified atmosphere packaging (MAP) over a 12-day storage period. Although the metmyoglobin reducing activity (MRA) of DC steaks decreased with increasing O2 levels, lower oxygen consumption (OC) and deeper oxygen penetration depth (OPD) improved the surface color of 50%O2-MAP and 80%O2-MAP DC steaks. 80%O2-MAP resulted in the highest (P < 0.05) percentage myoglobin denaturation and alleviated the phenomenon of persistent pink (PP) observed in DC beef. Compared with normal chilling, superchilling significantly increased the redness values of DC steaks through inhibition of lipid oxidation and enhancement of MRA, while it had no effects on PP due to the limited OPD. The results suggest that 80%O2-MAP combined with superchilling can be used to improve fresh color and minimize PP of DC beef.

Abstract:The anteroposterior and dorsoventral axes of the future embryo are specified within Drosophila oocytes by localizing gurken mRNA, which targets the secreted Gurken transforming growth factor-alpha synthesis and transport to the same site. A key question is whether gurken mRNA is targeted to a specialized exocytic pathway to achieve the polar deposition of the protein. Here, we show, by (immuno)electron microscopy that the exocytic pathway in stage 9-10 Drosophila oocytes comprises a thousand evenly distributed transitional endoplasmic reticulum (tER)-Golgi units. Using Drosophila mutants, we show that it is the localization of gurken mRNA coupled to efficient sorting of Gurken out of the ER that determines which of the numerous equivalent tER-Golgi units are used for the protein transport and processing. The choice of tER-Golgi units by mRNA localization makes them independent of each other and represents a nonconventional way, by which the oocyte implements polarized deposition of transmembrane/secreted proteins. We propose that this pretranslational mechanism could be a general way for targeted secretion in polarized cells, such as neurons.
Volatilome Evaluation of Modified Atmosphere Packaged Chilled and Super-Chilled Pork Loins Using Electronic Nose and Hs-Gc-Ims Integration

Abstract:This study assessed the volatilome of modified atmosphere packaged (MAP) pork loins at 4 °C and -2 °C storage using electronic (E-) nose and headspace-gas chromatography-ion mobility spectrometry (HS-GC-IMS) integration. Sensory characteristics decreased greatly as storage progressed (P < 0.05) in air packaged (AP) groups at both conditions. The E-nose sensors (W3C, W1C, and W5C) were linked to early sampling period, suggesting the presence of aromatic substances. Nonetheless, from 7 d and 21 d, AP groups shifted towards W3S, W1W, W2W, W1S, and W6S sensors at 4 °C and -2 °C, respectively, indicating significant spoilage activities (P < 0.05). HS-GC-IMS detected 46 VOCs, including their dimers (D) and monomers (M). Although hexanal, propanal, and limonene (D) were highly detected at the storage conditions, most spoilage-derived compounds were markedly identified in AP groups on 7 d (4 °C) and 21 d (-2 °C), demonstrating significant meat deterioration than MAP groups. This study affirms the potential benefit of 70%N 2 /30%CO 2 gas composition in spoilage inhibition and extension of shelf-life in foods.

Abstract:Over 21 days of cold storage, the quality and microbial composition of beef steaks in response to different high-CO2 packaging conditions achieved by flushing gas mixtures or embedding gas emitters into the packages were studied. The results revealed that the high levels of CO2, achieved by either the gas flushing or the CO2 emitter pads, effectively controlled the number of aerobic counts. The headspace CO2 increased quickly in response to using the CO2 emitter pads, and the meat samples presented different pH levels and surface color (a* and b*) values compared to the samples packaged with the gas flushing technique. Excessive accumulation of gas in the packages that contained CO2 emitters resulted in package swelling and higher levels of drip loss. The longest overall quality and attractive red color of the meat samples were observed when the packages were initially flushed with the headspace gas mixture containing high levels of oxygen. Overall, using CO2 emitters for meat packaging can be suggested when a topfilm with proper permeability to O2 and CO2 gases is used to regulate the internal CO2/O2 and gas/product ratios.

Abstract:The aim of this research was to study the effect of sous vide temperature (60, 65, and 70 °C) and time (12, 24, and 36 h) combination on the quality of beef short ribs, namely colour, cooking loss (%), shrinkage (%), soluble collagen (%), myofibrillar fragmentation index (MFI), Warner–Bratzler shear force (WBSF), and Texture Profile Analysis (TPA) parameters with different packaging materials. Aluminium and polyethylene, ethylene vinyl alcohol, and polyamide plastic laminate pouch were recommended for sous vide processing due to no detrimental effect on colour owing to its low oxygen permeability. The results showed a significant ( p < 0.05) increase in shrinkage in volume (%), cooking loss (%), and the proportion of soluble collagen (%) and MFI when sous vide temperature and the processing time were increased to 70 °C and 36 h. WBSF and the TPA hardness value were significantly lower ( p < 0.05) at 70 °C than at 60 °C both for 36 h due to higher level of solubilisation of collagen and myofibrillar fragmentation by prolonged heating in the moist in-pack environment and overpressure created by saturated steam. As the use of a laminate pouch reduced the oxidation of red myoglobin to brown metmyoglobin, the effect of sous vide temperature and time on International Commission on Illumination (CIE) hue angle and Chroma was not significant. Process optimisation using third-order multiple regression was conducted and the results revealed that optimum sous vide processing temperature and time combination for beef short ribs were 60 °C and 34.06 h.
