Abstract:There is growing evidence of the importance of extramatrical mycelium (EMM) of mycorrhizal fungi in carbon (C) cycling in ecosystems. However, our understanding has until recently been mainly based on laboratory experiments, and knowledge of such basic parameters as variations in mycelial production, standing biomass and turnover as well as the regulatory mechanisms behind such variations in forest soils is limited. Presently, the production of EMM by ectomycorrhizal (EM) fungi has been estimated at ~140 different forest sites to be up to several hundreds of kg per ha per year, but the published data are biased towards Picea abies in Scandinavia. Little is known about the standing biomass and turnover of EMM in other systems, and its influence on the C stored or lost from soils. Here, focussing on ectomycorrhizas, we discuss the factors that regulate the production and turnover of EMM and its role in soil C dynamics, identifying important gaps in this knowledge. C availability seems to be the key factor determining EMM production and possibly its standing biomass in forests but direct effects of mineral nutrient availability on the EMM can be important. There is great uncertainty about the rate of turnover of EMM. There is increasing evidence that residues of EM fungi play a major role in the formation of stable N and C in SOM, which highlights the need to include mycorrhizal effects in models of global soil C stores.

Abstract:A major fraction of nitrogen (N) in boreal forest soils is found in organic forms associated with soil organic matter (SOM) and mineral particles. The capacity of ectomycorrhizal (ECM) fungal symbionts to access this N is debated, considering that these fungi have lost many of the genes for decomposing organic matter that were present in their saprotrophic ancestors. To gain a molecular-level understanding of the N-mining processes in ECM fungi, we developed an experimental approach where the processes of decomposition were studied in parallel with the changes in the structure and properties of the organic matter. We showed that ECM fungi have significant capacities to assimilate organic N associated with SOM and mineral surfaces. The decomposition mechanisms differ between species, reflecting the lignocellulose decomposition mechanisms found in their saprotrophic ancestors. During N-mining, the ECM fungi processed the SOM to a material with increased adsorptive properties to iron oxide mineral particles. Two pathways contributed to these changes: Extracellular modifications of the SOM and secretion of mineral surface reactive metabolites. Some of these metabolites have iron(III)-reducing activities and can participate in extracellular Fenton reactions and redox reactions at iron oxide mineral surfaces. We conclude that the traditional framework for understanding organic N acquisition by ECM fungi from recalcitrant SOM must be extended to a framework that includes how those decomposition activities affect the stabilization and reactivity of mineral-associated SOM. The activity through these complex networks of reactions is decisive for the overall effect of ECM fungal decomposition on nutrients and C-cycling in forest ecosystems.
Photosynthetic product allocations of Pinus massoniana seedlings inoculated with ectomycorrhizal fungi along a nitrogen addition gradient

Abstract:Quantifying the allocation of photosynthetic products among different carbon (C) pools is critical for understanding and predicting plant C turnover response to climate change. A field experiment with ectomycorrhizal fungi (EMF) and nitrogen (N) was established to investigate the effects on allocation of photosynthetic products in Pinus massoniana (Lamb.) seedlings given increased N deposition. Seedlings were subjected to N addition and symbiosis with EMF, and the short-term allocation of a 13C photosynthetic pulse into leaves, branches, stems, roots, and soil was traced. Photosynthetic rate and root respiration were measured. It was found that N addition changed the allocation pattern of photosynthetic products in various organs of P. massoniana. Furthermore, N addition, mycorrhizal symbiosis, and interaction of N and EMF, all increased the amount of C produced by photosynthesis. N application less than 60 kg N hm-1 a-1 could promote the transfer and allocation of photosynthetic products in P. massoniana organs, which peaks at 60 kg N hm-1 a-1, and the highest N treatment began to decrease at 90 kg N hm-1 a-1. EMF inoculation could expand the absorption area of plant roots to obtain more nutrients and synthesize more C and N compounds for promoting the growth of itself and the host plant, improving the net photosynthetic rate and the distribution of C produced by photosynthesis in various organs. This forms a benign C and N cycle, thereby reducing the effect of high N addition on plants. The optimal N addition concentration was 60 kg N hm-1 a-1, and the optimal EMF was Pt, which provides a theoretical basis for inoculating EMF during increasing N deposition in the future climate change scenario. This enables plants to distribute more photosynthetic products to their roots, thus affecting their own C distribution for promoting growth.
Quantification of effects of season and nitrogen supply on tree below-ground carbon transfer to ectomycorrhizal fungi and other soil organisms in a boreal pine forest

Abstract:*The flux of carbon from tree photosynthesis through roots to ectomycorrhizal (ECM) fungi and other soil organisms is assumed to vary with season and with edaphic factors such as nitrogen availability, but these effects have not been quantified directly in the field. *To address this deficiency, we conducted high temporal-resolution tracing of (13)C from canopy photosynthesis to different groups of soil organisms in a young boreal Pinus sylvestris forest. *There was a 500% higher below-ground allocation of plant C in the late (August) season compared with the early season (June). Labelled C was primarily found in fungal fatty acid biomarkers (and rarely in bacterial biomarkers), and in Collembola, but not in Acari and Enchytraeidae. The production of sporocarps of ECM fungi was totally dependent on allocation of recent photosynthate in the late season. There was no short-term (2 wk) effect of additions of N to the soil, but after 1 yr, there was a 60% reduction of below-ground C allocation to soil biota. *Thus, organisms in forest soils, and their roles in ecosystem functions, appear highly sensitive to plant physiological responses to two major aspects of global change: changes in seasonal weather patterns and N eutrophication.

Abstract:Summary Ectomycorrhizal fungi are essential for nitrogen (N) cycling in many temperate forests and responsive to anthropogenic N addition, which generally decreases host carbon (C) allocation to the fungi. In the boreal region, however, ectomycorrhizal fungal biomass has been found to correlate positively with soil N availability. Still, responses to anthropogenic N input, for instance through atmospheric deposition, are commonly negative. To elucidate whether variation in N supply affects ectomycorrhizal fungi differently depending on geographical context, we investigated ectomycorrhizal fungal communities along fertility gradients located in two nemo‐boreal forest regions with similar ranges in soil N : C ratios and inorganic N availability but contrasting rates of N deposition. Ectomycorrhizal biomass and community composition remained relatively stable across the N gradient with low atmospheric N deposition, but biomass decreased and the community changed more drastically with increasing N availability in the gradient subjected to higher rates of N deposition. Moreover, potential activities of enzymes involved in ectomycorrhizal mobilisation of organic N decreased as N availability increased. In forests with low external input, we propose that stabilising feedbacks in tree‐fungal interactions maintain ectomycorrhizal fungal biomass and communities even in N‐rich soils. By contrast, anthropogenic N input seems to impair ectomycorrhizal functions.

Abstract:Abstract. In boreal forests an important part of the photo assimilates are allocated belowground to support symbiosis of ectomycorrhizal fungi (EMF). The production of EMF extramatrical mycelium can contribute to carbon (C) sequestration in soils, but the extent of this contribution depends on the composition of the EMF community. Some species can decrease soil C stocks by degrading soil organic matter (SOM), and certain species may enhance soil C stocks by producing hydrophobic mycelia which can reduce the rate of SOM decomposition. To test how EMF communities contribute to the development of hydrophobicity in SOM, we incubated sand-filled fungal-ingrowth mesh bags amended with maize compost for one, two or three growing seasons in non-fertilized and fertilized plots in a young Norway spruce (Picea abies) forest. We measured hydrophobicity as determined by the contact angle and the C/N ratios in the mesh bags contents along with the amount of new C entering the mesh bags from outside (determined by C3 input to C4 substrate), and related that to the fungal community composition. The proportion of EMF species increased over time to become the dominant fungal guild after three growing seasons. Fertilization significantly reduced fungal growth and altered EMF communities. In the control plots the most abundant EMF species was Piloderma olivaceum, which was absent in the fertilized plots. The hydrophobicity of the mesh bag contents reached the highest values after three growing seasons only in the unfertilized controls plots and was positively related to the abundance of P. olivaceum, the C/N ratios of the mesh bag contents and the amount of new C in the mesh bags. These results suggest that some EMF species are associated with higher hydrophobicity of SOM and that EMF community shifts induced by fertilization may result in reduced hydrophobicity of soil organic matter, which in turn may reduce C sequestration rates.

Abstract:Ectomycorrhizal fungi (EMF) are widespread in northern conifer forests. By competing with the free-living saprotrophic fungal and bacterial communities for limited soil nitrogen, EMF are expected to suppress litter decomposition and thus drive soil carbon accumulation. The EMF may also stimulate rhizosphere microbial growth through inputs of labile plant carbon, and subsequently contribute to the soil carbon pool via microbial necromass. Here we examined the relative strength of these two potential EMF effects in a northern conifer plantation of the Saihanba Forest, the largest plantation in China. The soil fungal and bacterial biomass, as well as their respiration, were quantified within the two types of soil cores that either allow or exclude the ingrowth of EMF. We also set up a nitrogen fertilization gradient (0, 5, 10, 15 g N m–2 y–1) in this plantation to quantify the influence of external inorganic nitrogen on the EMF effects. We found evidence that EMF inhibit the overall fungal and bacteria biomass, confirming the suppression of saprotrophs by EMF. In addition, high levels of external nitrogen fertilization (15 g N m–2 y–1) might further enhance the suppression by EMF. In contrast, the presence of EMF consistently increased soil microbial respiration across all nitrogen fertilization levels, indicating that the carbon allocated to EMF could have been largely consumed by microbial respiration and contributed minimally to the accumulation of microbial biomass. Our results also indicated that the suppression of saprotrophs by EMF may play a critical role in driving continuous soil carbon accumulation in this northern pine plantation under atmospheric nitrogen deposition.

Abstract:The aim of global carbon (C) neutrality brings soils and their potential for C storage into the spotlight. Improved agricultural management techniques such as minimum or no-tillage are thought to foster soil C sequestration. However, the underlying mechanisms are still not well understood. In this study, we investigated the inter-relations of soil organic C (SOC), fungal biomass, microbial necromass biomarkers, and aggregate stability in rhizosphere and bulk soil after thirteen years of reduced tillage intensities (reduced, minimum, and no-tillage). Overall, rhizosphere and bulk soil were indifferent in their response to reduced tillage. Reducing tillage intensity increased SOC and nitrogen stocks and dissolved organic C contents in the following order: minimum > no-tillage > reduced > conventional. Aggregate stability showed the strongest increase under no-tillage. Interestingly, ergosterol contents were highest under reduced and minimum tillage followed by no-tillage. The amino sugars muramic acid, galactosamine, and glucosamine – proxies for soil microbial-derived necromass – showed similar increases under all three tillage reduction systems. Structural equation modelling revealed that increased dissolved organic C contents under reduced tillage intensity facilitated SOC sequestration and aggregate stability through enhanced fungal biomass to necromass turnover. Thus, reducing soil tillage intensity is a valuable tool to facilitate microbial growth and hence to increase SOC sequestration in agricultural soils.

Abstract:Ingestions constitute a common problem in pediatric emergency medicine. Management decisions are frequently based on parental assessments of the amount of the ingested material. The purpose of this study was to examine the accuracy of parental estimates of fluid volumes. One hundred parents of children seen in an outpatient clinic and emergency department were asked to estimate fluid volumes in four containers. There were no significant differences (P > 0.05) in the accuracy of fluid volume assessment between younger and older parents, between participants with and without high school education, or between parents with one or more children. While most parents are accurate in their fluid volume estimates, 14.5% of the assessments were overestimates of over 300% of the actual volume. In view of our findings, caution should be used when management decisions are being based on parental fluid volume estimates.

Abstract:Background and aimsThe partitioning of below ground carbon inputs into roots and extramatrical ectomycorrhizal mycelium (ECM) is crucial for the C cycle in forest soils. Here we studied simultaneously the newly grown biomass of ECM and fine roots in a young Norway spruce stand.MethodsIngrowth mesh bags of 16 cm diameter and 12 cm height were placed in the upper soil and left for 12 to 16 months. The 2 mm mesh size allowed the ingrowth of fungal hyphae and roots whereas a 45 μm mesh size allowed only the ingrowth of hyphae. The mesh bags were filled with either EA horizon soil, pure quartz sand or crushed granite. Controls without any ingrowth were established for each substrate by solid tubes (2010) and by 1 μm mesh bags (2011). The fungal biomass in the substrates was estimated by the PLFA 18:2ω6,9 and ECM biomass was calculated as difference between fungal biomass in mesh bags and controls.ResultsThe maximum ECM biomass was 438 kg ha−1 in October 2010 in 2 mm mesh bags with EA substrate, and the minimum was close to zero in 2011 in 45 μm mesh bags with quartz sand. The high P content of the crushed granite did not influence the ECM biomass. Fine root biomass reached a maximum of 2,343 kg ha−1 in October 2010 in mesh bags with quartz sand after 16 months exposure. In quartz sand and crushed granite, ECM biomass correlated positively with fine root biomass and the number of root tips, and negatively with specific root length.ConclusionThe ratio of ECM biomass/fine root biomass in October ranged from 0.1 to 0.3 in quartz sand and crushed granite, but from 0.7 to 1.8 in the EA substrate. The results for the EA substrate suggest a large C flux to ECM under field conditions.

Abstract:Forest soil organic matter (SOM) is an important dynamic store of C and N, which releases plant available N and the greenhouse gases CO2 and N2O. Early stages of decomposition of recent plant litters are better known than the formation of older and more stable soil pools of N and C, in which case classic theory stated that selective preservation of more resistant plant compounds was important. Recent insights heighten that all plant matter becomes degraded and that older SOM consists of compounds proximally of microbial origin. It has been proposed that in boreal forests, ectomycorrhizal fungi (ECMF), symbionts of trees, are actively involved in the formation of slowly-degrading SOM.We characterized SOM in the mor-layer along a local soil N supply gradient in a boreal forest, a gradient with large variations in chemical and biological characteristics, notably a decline in the biomass of ECMF in response to increasing soil N supply.We found contrasting and regular patterns in carbohydrates, lignin, aromatic carbon, and in N-containing compounds estimated by solid-state 13C and 15N nuclear magnetic resonance (NMR) spectroscopy. These occurred along with parallel changes in the natural abundances of the stable isotopes 13C and 15N in both bulk SOM and extracted fractions of the SOM. The modelled "bomb-14C″ age of the lower layers studied ranged between 15 years at the N-poor end, to 70 years at the N-rich end of the gradient. On average half the increase in δ13C with soil depth (and hence age) of the mor-layer can be attributed to soil processes and the other half to changes in the isotopic composition of the plant C inputs. There was a decrease in carbohydrates (O-alkyl C) with increasing depth. This supports the classical hypothesis of declining availability of easily decomposable substrates to microorganisms with increasing soil depth and age. The observed increase in δ13C with depth, however, speaks against the idea of selective preservation of more resistant plant compounds like lignin. Furthermore, from the N-poor to the N-rich end the difference between 15N in plant litter N and N in the deeper part of the mor-layer, the H-layer, decreased in parallel with a decline in ECMF.The latter provides evidence that the role of ECMF as major sink for N diminishes, and hence their potential role in SOM stabilization, when the soil N supply increases. At the N-rich end, where bacteria dominate over fungi, other agents than ECMF must be involved in the large build-up of the H-layer with the slowest turnover rate found along the gradient.

Abstract:Carbon allocation of trees to ectomycorrhizas is thought to shape forest nutrient cycling, but the sink activities of different fungal taxa for host resources are unknown. Here, we investigate fungal taxon-specific differences in naturally composed ectomycorrhizal (EM) communities for plant-derived carbon and nitrogen. After aboveground dual labeling of young beech with 15 N and 13 C, ectomycorrhizas formed with different fungal taxa exhibit strong differences in label enrichment. Secondary Ion Mass Spectrometry (SIMS) imaging of nitrogen in cross sections of ectomycorrhizas demonstrates plant-derived 15 N in both root and fungal structures. Isotope enrichment in ectomycorrhizas correlates with that in the corresponding ectomycorrhiza-attached lateral root, supporting fungal taxon-specific N and C fluxes in ectomycorrhizas. The enrichments with 13 C and 15 N in the symbiosis decrease with increasing C/N ratio of ectomycorrhizas, converging to zero at high C/N. The relative abundances of EM fungal species on roots are positively correlated with 13 C enrichment, demonstrating higher fitness of stronger than of less C-demanding symbioses. Overall, our results support that differences among the C/N ratios in ectomycorrhizas formed with different fungal species regulate the supply of the symbioses with host-derived carbon and provide insights on functional traits of ectomycorrhizas, which are important for major ecosystem processes.

Impact of pharmaceutical company representatives on internal medicine residency programs. A survey of residency program directors.

Abstract:To survey internal medicine residency program directors regarding interactions between their residents and pharmaceutical company (PC) representatives (PCRs) a questionnaire was sent to the directors of all Accreditation Council for Graduate Medical Education-approved internal medicine residency programs. The survey included 444 program directors, of whom 272 (61.16%) responded. The majority of program directors, 228 (83.8%), allowed PCRs to meet with residents during working hours and 241 (88.6%) permitted PC sponsorship of conferences. About half of the program directors were "moderately" or "very" concerned about the potential adverse effects of PC marketing on resident attitudes and prescribing practices. Seventy percent "agreed" or "strongly agreed" that the benefits of PC sponsorship outweigh the adverse effects and 41.5% believed that refusal to allow PCRs to meet with residents would jeopardize PC funding of other departmental activities. Most program directors reported that alternate funds for conferences were available if PC support was withdrawn. "Unethical" marketing activities were observed by 14.3% of program directors and 37.5% reported that residents had participated in PC-sponsored trips during the 3 years prior to the survey. At the time of this survey, only 35.3% of programs had developed formal policies regulating PCR activities and 25.7% provided residents with formal instruction on marketing issues. Knowledge of the current extent of PCR interactions with residents may be helpful to program directors in developing policies regulating PC-marketing activities.
Accounting for Carbon Flux to Mycorrhizal Fungi May Resolve Discrepancies in Forest Carbon Budgets

Abstract:Carbon (C) fluxes among different components of plant growth are important to forest ecosystem C cycling and are strongly influenced by species composition and resource availability. Although mycorrhizal fungi are crucial for nutrient acquisition and can receive a large fraction of annual net primary production, most studies do not explicitly include carbon flux to mycorrhizal fungi in ecosystem C budgets. We measured annual production of plant components (foliage, wood, fine roots) and mycorrhizal fungi across temperate forest stands varying in species composition. Production of mycorrhizal fungi was estimated using both mass balance and isotopic techniques. Total plant production varied from about 600 g C m−2 y−1 in nearly pure deciduous broadleaf stands down to about 300 g C m−2 y−1 in conifer-dominated stands. In contrast, the production of mycorrhizal fungi was highest in conifer-dominated stands, varying from less than 25 g C m−2 y−1 in deciduous broadleaf stands to more than 175 g C m−2 y−1 in nearly pure conifer stands. Isotopic data indicated that both tree species composition and ecosystem nitrogen (N) availability influenced rates of fungal production. The large investment in mycorrhizal fungi in low-N, conifer-dominated stands demonstrated that a full accounting of ecosystem carbon fluxes to plant and fungal components may help resolve current discrepancies observed in broadscale forest carbon budgets, especially across forest types.

Long-term nitrogen fertilization impacts plant-microbial interactions differently in arbuscular and ectomycorrhizal trees

Abstract:Enhanced nitrogen (N) availability in temperate forests has altered ecosystem carbon (C) and N cycling. Recent research has shown that these alterations lead to reductions in belowground C allocation by trees and that the consequences of these reductions on soil C and nutrient cycling may vary by mycorrhizal type. We hypothesized that trees that associate with ectomycorrhizal fungi (ECM) would reduce C allocation towards roots and mycorrhizal fungi to a greater extent than trees that associate with arbuscular mycorrhizal fungi (AM) in response to > 25 years of N fertilization. We further hypothesized that N induced decoupling of roots and microbes in ECM trees would be evidenced by greater declines in extracellular enzyme activities. We measured belowground C allocation to fine root biomass and mycorrhizal colonization in 6 AM and 6 ECM dominated plots in the N fertilized and reference watersheds at the Fernow Experimental Forest in West Virginia, USA. We compared these to measurements of simple-C, complex-C, nitrogen, and phosphorus acquiring enzyme activities in organic horizon, bulk mineral, and rhizosphere soil fractions. N fertilization reduced fine root biomass and mycorrhizal colonization in both AM and ECM stands. We found more consistent reductions in enzyme activities in ECM soils than AM soils under N fertilization which may have been driven by greater declines in root-C transfer to soil microbes. This mechanism helps to explain variability in soil C cycling responses across N gradient and fertilization experiments and may prove useful in predicting the fate of soil C stocks in response to N deposition.

Abstract:Abstract Interactions between symbiotic ectomycorrhizal (EM) and free-living saprotrophs can result in significant deceleration of leaf litter decomposition. While this phenomenon is widely cited, its generality remains unclear, as both the direction and magnitude of EM fungal effects on leaf litter decomposition have been shown to vary among studies. Here we explicitly examine how contrasting leaf litter types and EM fungal communities may lead to differential effects on C and N cycling. Specifically, we measured the response of soil nutrient cycling, litter decay rates, litter chemistry and fungal community structure to the reduction of EM fungi (via trenching) with a reciprocal litter transplant experiment in adjacent Pinus - or Quercus -dominated sites. We found clear evidence of EM fungal suppression of C and N cycling in the Pinus -dominated site, but no suppression in the Quercus -dominated site. Additionally, in the Pinus -dominated site, only the Pinus litter decay rates were decelerated by EM fungi and were associated with decoupling of litter C and N cycling. Our results support the hypothesis that EM fungi can decelerate C cycling via N competition, but strongly suggest that the ‘Gadgil effect’ is dependent on both substrate quality and EM fungal community composition. We argue that understanding tree host traits as well as EM fungal functional diversity is critical to a more mechanistic understanding of how EM fungi mediate forest soil biogeochemical cycling.

Abstract:In boreal forest soils, mycelium of mycorrhizal fungi is pivotal for regulating soil carbon (C) cycling and storage. The carbon use efficiency (CUE), a key parameter in C cycling models, can inform on the partitioning of C between microbial biomass, and potential soil storage, and respiration. Here we test the dependency of mycorrhizal mycelial CUE on stand age and seasonality in managed boreal forest stands.Based on mycelial production and respiration estimates, derived from sequentially incubated ingrowth mesh bags, we estimated CUE on an ecosystem‐scale during a seasonal cycle and across a chronosequence of eight, 12‐ to 158‐years‐old, managed Pinus sylvestris forest stands characterised by decreasing pH and nitrogen (N) availability with increasing age. Mycelial respiration was related to total soil respiration, and by using eddy covariance flux measurements, primary production (GPP) was estimated in the 12‐ and 100‐years‐old forests, and related to mycelial respiration and CUE.As hypothesized, mycelial CUE decreased significantly with increasing forest age by c. 65%, supposedly related to a shift in mycorrhizal community composition and a metabolic adjustment to reduce their own biomass N demand with declining soil N availability. Furthermore, mycelial CUE increased by a factor of five over the growing season; from 0.03 in May to 0.15 in November, and we propose that the seasonal change in CUE is regulated by a decrease in photosynthate production and temperature. The respiratory contribution of mycorrhizal mycelium ranged from 14 to 26% of total soil respiration, and was on average 17% across all sites and occasions.Synthesis. Carbon is retained more efficiently in mycorrhizal mycelium late in the growing season, when fungi have access to a more balanced C and nutrient supplies. Earlier in the growing season, at maximum host plant photosynthesis, when belowground C availability is high in relation to N, the fungi respire excess C resulting in lower mycelial CUE. Additionally, C is retained less efficiently in mycorrhizal fungal biomass in older forest stands characterized by more nutrient depleted soils than younger forest stands.This article is protected by copyright. Differences in microbial community response to nitrogen fertilization result in unique enzyme shifts between arbuscular and ectomycorrhizal-dominated soils

Abstract:While the effect of nitrogen (N) deposition on belowground carbon (C) cycling varies, emerging evidence shows that forest soils dominated by trees that associate with ectomycorrhizal fungi (ECM) store more C than soils dominated by trees that associate with arbuscular mycorrhizae (AM) with increasing N deposition. We hypothesized that this is due to unique nutrient cycling responses to N between AM and ECM-dominated soils. ECM trees primarily obtain N through fungal mining of soil organic matter subsidized by root-C. As such, we expected the largest N-induced responses of C and N cycling to occur in ECM rhizospheres and be driven by fungi. Conversely, as AM trees rely on bacterial scavengers in bulk soils to cycle N, we predicted the largest AM responses to be driven by shifts in bacteria and occur in bulk soils. To test this hypothesis, we measured microbial community composition, metatranscriptome profiles, and extracellular enzyme activity in bulk, rhizosphere, and organic horizon (OH) soils in AM and ECM-dominated soils at Bear Brook Watershed in Maine, USA. After 27 years of N fertilization, fungal community composition shifted across ECM soils, but bacterial communities shifted across AM soils. These shifts were mirrored by enhanced C relative to N mining enzyme activities in both mycorrhizal types, but this occurred in different soil fractions. In ECM stands these shifts occurred in rhizosphere soils, but in AM stands they occurred in bulk soils. Additionally, ECM OH soils exhibited the opposite response with declines in C relative to N mining. As rhizosphere soils account for only a small portion of total soil volume relative to bulk soils, coupled with declines in C to N enzyme activity in ECM OH soils, we posit that this may partly explain why ECM soils store more C than AM soils as N inputs increase.

Abstract:The continuous upsurge in soil nitrogen (N) enrichment has had strong impacts on the structure and function of ecosystems. Elucidating how plant ectomycorrhizal fungi (EMF) mutualists respond to this additional N will facilitate the rapid development and implementation of more broadly applicable management and remediation strategies. For this study, we investigated the responses of EMF communities to increased N, and how other abiotic environmental factors impacted them. Consequently, we conducted an eight-year N addition experiment in a poplar plantation in coastal eastern China that included five N addition levels: 0 (N0), 50 (N1), 100 (N2), 150 (N3), and 300 (N4) kg N ha−1 yr−1. We observed that excessive N inputs reduced the colonization rate and species richness of EMF, and altered its community structure and functional traits. The total carbon content of the humus layer and available phosphorus in the mineral soil were important drivers of EMF abundance, while the content of ammonium in the humus layer and mineral soil determined the variations in the EMF community structure and mycelium foraging type. Our findings indicated that long-term N addition induced soil nutrient imbalances that resulted in a severe decline in EMF abundance and loss of functional diversity in poplar plantations.
