Abstract:Five new and seven known mono‐sesquiterpenoids (1–5 and 6–12, resp.) together with five known lindenane‐type disesquiterpenoids, 13–17, were isolated from the whole plant of Chloranthus henryi. Based on spectroscopic methods, the new structures were established to be (5S,6R,8S,10R)‐6‐hydroxyeudesma‐4(15),7(11)‐diene‐12,8‐olide (1), 6α‐hydroxyeudesma‐4(15),7(11),8(9)‐triene‐12,8‐olide (2), 8,12‐epoxy‐1β‐hydroxyeudesma‐4(15),7,11‐trien‐6‐one (3), 12‐oxochloraniolide A (4), and (4α)‐8‐hydroxy‐12‐norcardina‐6,8,10‐trien‐11‐one (5), respectively. Among the isolates, compound 2, zederone epoxide (8), spicachlorantin G (13), chloramultilide A (14), shizukaol B (15), and spicachlorantin B (17) showed significant anti‐neuroinflammatory effects by inhibiting nitric‐oxide (NO) production in lipopolysaccharide (LPS)‐stimulated murine BV‐2 microglial cells with relatively low cytotoxicity.

Abstract:Two new disesquiterpenoids (1 and 2) and 11 new (3-13) and 10 known (14-23) sesquiterpenoids were isolated from the whole plants of Artemisia freyniana. Their structures were elucidated by spectroscopic data analysis and comparison with published NMR data. The absolute configurations of the new isolates (1-13) were assigned based on single-crystal X-ray diffraction data and comparison of the experimental and calculated ECD data. The eremophilane derivatives 8 and 9 possess an unprecedented 2-isopropyl-3,7,7a-trimethyl-2,4,5,6,7,7a-hexahydro-1 H-indene scaffold, and a putative biosynthetic pathway for these compounds is proposed. Compounds 4, 5, and 9 exhibited inhibitory effects against LPS-stimulated nitric oxide (NO) production in RAW 264.7 macrophage cells with IC50 values of 10.8, 12.6, and 11.7 μM, respectively.
Undescribed sesquiterpenoids with NO production inhibitory activity from oleo-gum resin of Commiphora myrrha

Abstract:Six undescribed cadinane sesquiterpenoids ( 1 − 6 ), two undescribed guaiane sesquiterpenoids ( 7 − 8 ), and an undescribed germacrane sesquiterpenoid ( 9 ) were isolated from the oleo-gum resin of Commiphora myrrha . Their structures were determined by the analysis of 1D/2D NMR and HRESIMS data, as well as quantum chemical ECD and NMR calculations. All the sesquiterpenoids were evaluated for their NO production inhibitory activity in LPS-stimulated RAW 264.7 mouse monocyte-macrophages. The results revealed that commiphone A ( 1 ) and commipholide D ( 7 ) exhibited significant inhibitory effect on NO generation with IC 50 values of 18.6 ± 2.0 and 37.5 ± 1.5 μM, respectively. Furthermore, 1 and 7 dose-dependently inhibited the mRNA expression of inflammatory cytokines IL-1 β , IL-6 and TNF- α induced by LPS in the RAW264.7 cells, indicating that 1 and 7 possess potent anti-inflammatory activity in vitro .

Terpenoids from the rhizomes of Curcuma kwangsiensis S.G. Lee et C.F. Ling and their anti-inflammatory activities

Abstract:Three new sesquiterpenes, 4S-1-(3-hydroxybutyl)-7-(11-hydroxypropyl)-10-methyl- cyclohepta-7,5,10-trien-8-one (1), 8R-hydroxy-7-(4S-4,10-dimethyl-5-oxooct-1-en-7-yl)-11- methylfuran-12-one (2), (1S,5R,7S,10R)-1-hydroxy-7-(11-hydroxypropyl)-10-methyl-4- methyleneoctahydronaphthalen-8-one (3), along with 30 known terpenoids (4-33) were obtained from the rhizomes of Curcuma kwangsiensis S.G. Lee et C.F. Ling. Through comprehensive analysis of chemical evidence and spectral data including UV, ECD, IR, 1D and 2D NMR and HR-ESI-MS, as well as quantum chemical calculation, the structures of these novel compounds were successfully determined. Additionally, the inhibitory effects of compounds 1-2, 4-33 on NO production were evaluated in lipopolysaccharide (LPS)-induced RAW264.7 cells. Notably, compound 33 exhibited the most significant inhibitory effect with an IC50 value of 3.55 ± 0.55 μM.
Chemical Constituents from a Mangrove-Derived Actinobacteria Isoptericola chiayiensis BCRC 16888 and Evaluation of Their Anti-NO Activity

Abstract:Cultivation of the actinobacteria strain Isoptericola chiayiensis, a mangrove-derived actinobacteria that was isolated from a mangrove soil collected in Chiayi County, resulted in the isolation of one new 2-furanone derivative, isopterfuranone (1), one new sesquiterpenoid, isopterchiayione (2), one new benzenoid derivative, isopterinoid (3), five new flavonoids, chiayiflavans A-E (4-8), and 4 metabolites isolated for the first time from nature source, methyl 3-(4-methyl-2,5-dioxopyrrolidin-3-yl)propanoate (9), 3-ethyl-4-methylpyrrolidine-2,5-dione (10), chiayiensol (11) and chiayiensic acid (12). Their structures were determined through in-depth spectroscopic and mass-spectrometric analyses. Most of the isolates showed potent inhibitory effects on NO production in LPS-stimulated RAW 264.7 murine macrophages cells with IC50 values ranging from 9.36 to 40.02 μM. Of these isolates, 4 and 5 showed NO inhibitory activity with IC50 values of 17.14 and 9.36 μM, stronger than the positive control quercetin (IC50 =36.95 μM). This is the first report on flavan metabolites from the genus Isoptericola.

Abstract:A new linear pyranocoumarin named (−)-hydroxydecursinol (1) and a new biscoumarin named (±)-dahuribiscoumarin (2), together with six known compounds isoimperatorin (3), imperatorin (4), phellopterin (5), isodemethylfuropinarine (6), demethylfuropinarine (7), and (+)-decursinol (8) were isolated from the 75% ethanolic extract of the roots of Angelica dahurica var. formosana cv. Chuanbaizhi. Their structures were elucidated by extensive spectroscopic techniques, including 2D NMR spectroscopy and mass spectrometry, and the structure of 2 was confirmed by single-crystal X-ray diffraction. All of the isolated compounds were evaluated for the inhibition against nitric oxide (NO) production in the lipopolysaccharide (LPS)-activated RAW264.7 macrophage cell line, and exhibited the inhibitory activity on NO production in a concentration-dependent manner. Furthermore, real-time PCR analysis revealed that compounds 2, 5–8 could significantly suppress the expression levels of inducible nitric oxide synthase mRNA in a concentration-dependent manner. And their primary structure–activity relationships of NO inhibitory effects were also briefly discussed. These compounds are potential candidates for further bioassay studies to determine their suitability as drug leads.

Abstract:A new isopimarane-type diterpene clinacanoid A ( 1 ) together with seven known terpenoids ( 2–8 ) were obtained from the Clinacanthus nutans . Their structures were elucidated based on extensive spectroscopic analysis (NMR, HR-ESI-MS), and the absolute configuration of 1 was established based on single crystal X-ray diffraction. The inhibitory activity of all the compounds on NO production in lipopolysaccharide-induced (LPS) mouse leukemic monocyte macrophage RAW264.7 cells was evaluated. Among them, compounds 1 and 3 showed potential anti-inflammatory activities, with IC 50 values of 13.3 ± 0.3 and 12.4 ± 0.4 μ M, respectively.

Abstract:Ten new ent-kaurane diterpenoids, including two pairs of epimers 1/2 and 4/5 and a 6,7-seco-ent-kauranoid 10, were obtained from the aerial parts of Isodon serra. The structures of the new compounds were confirmed by extensive spectroscopic methods and electronic circular dichroism (ECD) data analysis. An anti-inflammatory assay was applied to evaluate their nitric oxide (NO) inhibitory activities by using LPS-stimulated BV-2 cells. Compounds 1 and 9 exhibited notable NO production inhibition with IC50 values of 15.6 and 7.3 μM, respectively. Abstract:A new pentacyclic triterpenoid, 3-oxo-urs-11,13(18)-dien-28-oic acid (1), along with twelve known triterpenoids, α-amyrin (2), 19α-hydroxy-α-amyrin (3), triptohypol E (4), uvaol (5), 2α,3α-dihydroxyurs-11-en-13β,28-olide (6), 3β-hydroxyurs-11-en-13β,28-olide (7), ursolic acid (8), asiatic acid (9), oleanolic acid (10), aegiceradienol (11), obtusalin (12) and betulinic acid (13) were isolated from the leaves of Rhododendron dauricum L. Their structures were established from spectroscopic data and comparison with reported values. Among them, compounds 3, 4, 6, 7 and 11 were isolated from the Ericaceae family for the first time. Compounds 2, 5, 9, 12 and 13 were obtained from R. dauricum for the first time. Additionally, compounds 6, 10 and 11 significantly inhibited the levels of NO in LPS-stimulated RAW 264.7 cells at 3 μM.

Abstract:Twelve new abietane diterpenoids (1-12) and 31 known analogues (13-43) were isolated from a medicinal Chinese herb, Clerodendrum trichotomum Thunberg. The absolute configurations of 1-3 were established on the basis of ECD and X-ray crystallography data, whereas that of 4 was elucidated by comparison of experimental and calculated ECD data. Eight diterpenoids, 15,16-dehydroteuvincenone G (1), trichotomin A (4), 2α-hydrocaryopincaolide F (7), villosin C (20), 15-dehydro-17-hydroxycyrtophyllone A (22), demethylcryptojaponol (38), 6β-hydroxydemethylcryptojaponol (39), and trichotomone (43), exerted inhibitory effects against NO production with IC50 values of 5.6-16.1 μM. The structure-activity relationships of the isolated diterpenoids are also estimated.

Abstract:A new coumarin, methyl (E)-4-((6-methoxy-2-oxo-2H-chromen-7-yl)oxy)-2-methylbut-2-enoate ( 1 ), has been isolated from the rhizomes of Polygonatum chingshuishanianum , together with six known compounds – scopoletin ( 2 ), rutin ( 3 ), isoquercetin ( 4 ), hyperoside ( 5 ), (3R)-5,7-dihydroxy-3-(2′,4′-dihydroxybenzyl)-chroman-4-one ( 6 ), and (3R)-5,7-dihydroxy-6-methoxy-8-methyl-3-(2′,4′-dihydroxybenzyl)-chroman-4-one ( 7 ). The structure of new compound 1 was determined through spectroscopic and MS analyses. Among the isolates compounds 3 – 6 showed potent inhibition with IC 50 values of 10.25 ± 0.75, 17.60 ± 1.28, 18.82 ± 1.63, and 24.93 ± 2.07 μM, respectively, against LPS-induced NO generation.

Abstract:Three labdane-type [multisins A-C (1-3)], two guaiane-type [multisins D (4) and E (5)], and one eudesmane-type [multisin F (6)] previously undescribed terpenoids, together with 14 mono- (7-20) and seven dimeric- (21-27) known terpenoids, were isolated from the 90% MeOH extract of the whole plant of Chloranthus multistachys. Their structures and absolute configurations were determined by extensive spectroscopic methods and electronic circular dichroism (ECD) calculations. Compounds 4 and 5 are rare trinor-sesquiterpenes with a de-isopropyl guaiane skeleton, whereas compound 6 is a rearranged dinor-eudesmene featuring an uncommon octahydro-1H-indene ring system. Among the isolates, the dimeric lindenane sesquiterpenoid shizukaol C (25) exhibited the most potent (IC50 = 8.04 μM) anti-neuroinflammatory activity by inhibiting the nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated murine BV-2 microglial cells.

Abstract:Two heterodimers including a clovane-phenylpropanoid hybrid ( 1 ) and a clovane-menthane hybrid ( 2 ), five linear sesquiterpenoids incorporating a tetrahydrofuran ring ( 3 – 6 & 8 ), and four steroids ( 7 & 9 – 11 ), were separated from the ethanolic extract of a well-known aromatic and medicinal herb Eupatorium fortunei . Their structures were characterised by detailed analyses of spectroscopic data and comparison with known analogues, with seven ( 1 – 7 ) of them being described for the first time. The hybrids 1 and 2 represent the first examples of clovane type sesquiterpenoids hybridising with other class of natural products, and compounds 3 – 6 and 8 are first linear sesquiterpenyl constituents reported from the title species. All the isolates were evaluated for their inhibitory effect on the NO production induced by LPS in murine RAW264.7 macrophage cells, and 1 , 7 , 10 and 11 exhibited moderate activity with IC 50 values in the range of 24.4–43.5 μM.

Abstract:Three new benzenoids, 3-isopropenyl-2-methoxy-6-methyl-4,5-methylenedioxy- phenol (1), 2-hydroxy-4,4'-dimethoxy-3,3'-dimethyl-5,6,5',6'-bimethylenedioxybiphenyl (2), 4,4'-dihydroxy-3,3'-dimethoxy-2,2'-dimethyl-5,6,5',6'-bimethylenedioxybiphenyl (3), together with two known benzenoids, 2,3,6-trimethoxy-5-methylphenol (4) and 2,3-methylenedioxy- 4-methoxy-5-methylphenol (5), were isolated from Antrodia camphorata. Our results support that compounds 1-5 potently inhibited LPS (lipopolysaccharide)-induced nitric oxide (NO) production in a dose-dependent manner. The IC(50) values of compounds 1, 3 and 5 were 1.8 ± 0.2, 18.8 ± 0.6 and 0.8 ± 0.3 μg/mL, respectively.

Abstract:Five new oleanane and ursane type triterpenes, namely uncarinic acids F–J (1–5), together with six known triterpenic acids (6–11) were isolated from the stems and hooks of Uncaria rhynchophylla. Structure elucidation of 1–5 was based on the integrated analyses of high-resolution MS data, 1D (1H NMR, 13C NMR, DEPT) and 2D (HSQC, HMBC, ROESY) NMR spectra. Compounds 4, 10, and 11 exhibited weak inhibitory effects on LPS-induced NO production in RAW264.7 cells (with IC50 1.48, 7.01, and 1.89μM, respectively) with dexamethasone (IC50 0.04μM) and quercetin (IC50 0.86μM) as the positive controls. 19-OH substituted oleanane triterpenic acids (1, 2, 5, 8) were prone to eliminate CH2O3, whereas those ursane-type encompassing 19-OH (3, 6, 7, 9, 4) were featured by preferred cleavage of H2O while performing the negative collision-induced MS/MS fragmentation on an LTQ/Orbitrap mass spectrometer.
Seco-nortriterpenoids from Cirsium setosum and their anti-inflammatory activity

Abstract:Five unusual seco-nortriterpenoids, 3 β -hydroxy-20,21-seco-30-nortaraxastan-20,21-dioic acid ( 1 ), 3 β -hydroxy-20,21-seco-30-nortaraxastan-20-oic-21-oate ( 2 ), 3 β -hydroxy-20-oxo-21,22-seco-30-nortaraxastan-22-oic acid ( 3 ), 3 β -hydroxy-19-oxo-20,21-seco-29,30-nortaraxastan-21-oic acid ( 4 ) and 3 β -hydroxy-19-oxo-20,21-seco-19-norlupan-21-oic acid ( 5 ) were isolated and elucidated from the anti-inflammatory activity fraction of the ethanol extract of Cirsium setosum . The structures of these compounds were established through spectroscopic methods. Preliminary biological assays showed that compounds 1 – 5 had significant inhibitory effect on NO production on lipopolysaccharide-stimulated RAW 264.7 cells, and compound 1 showed the strongest anti-inflammatory activity. This type of ring-opening compound is the first seco-triterpenoid structure discovered from the genus of Cirsium .

Abstract:Seven previously undescribed cadinane sesquiterpenoids, albocinnamins A‒G, were isolated from the fungus Antrodiella albocinnamomea. Their structures with absolute configurations were elucidated on the basis of extensive spectroscopic methods, as well as single crystal X-ray diffraction. Cadinane sesquiterpenoids were reported from A. albocinnamomea for the first time. All of these sesquiterpenoids possess an unusual ether ring with minor modifications. Albocinnamins D and G showed certain inhibitory activities against nitric oxide production in LPS-activated RAW264.7 macrophages with IC50 values of 26.1 and 19.2 μM, respectively.

Abstract:Hupelactones A (1) and B (2), two new eudesmanolide-type enantiomers of the corresponding compounds, along with four mono- (3-6) and nine dimeric- (7-15) known sesquiterpenoids were isolated from the whole plant of Chloranthus henryi var. hupehensis (syn. C. henryi). The new structures including the absolute configurations were determined by comparison with previously reported enantiomers, extensive spectroscopic methods in combination with electronic circular dichroism (ECD) calculations. All the isolates were evaluated for their inhibitory activities against the lipopolysaccharide (LPS)-induced nitric oxide (NO) production in murine BV-2 microglial cells. Among them, the dimeric lindenane sesquiterpenoids shizukaols F (8) and G (11) exhibited the most potent activities, with IC50 values of 2.65 and 4.60 mu M, respectively.
Structure and anti-inflammatory activity of neo -clerodane diterpenoids from Scutellaria barbata

Abstract:Herein, 13 previously undescribed neo -clerodane diterpenoids ( 1 – 13 ) and 27 known analogs ( 14 – 40 ) were isolated from the aerial parts of Scutellaria barbata . Absolute configurations of undescribed compounds were assigned based on single-crystal X-ray diffraction analysis and comparison of experimental and circular dichroism. All isolates were evaluated for the inhibition of nitric oxide generation induced by lipopolysaccharide in RAW 264.7 macrophages. Compound 36 was found to be the most active with an IC 50 value of 10.6 μM. Structure–activity relations of these neo -clerodane diterpenoids revealed that the α, β -unsaturated- γ -lactone moiety with an exocyclic conjugated double bond was necessary for maintaining and increasing its activity. Further mechanistic studies show that compound 36 suppressed nitric oxide synthase enzymes (iNOS) expression without affecting iNOS activity. Additionally, compound 36 suppresses NF- κ B signaling by inhibiting IκBα phosphorylation.

Abstract:From the MeOH extract of Taiwanese Orthosiphon stamineus, two new staminane-type diterpenes, staminols C (1) and D (2), and three new isopimarane-type diterpenes, orthosiphonone C (3) and D (4) and 14-deoxo-14-O-acetylorthosiphol Y (5), have been isolated together with 16 known diterpenes, orthosiphols A, B, D, K, M, N, O, X, and Y, nororthosiphonolide A, neoorthosiphol B, orthosiphonone A, secoorthosiphols B and C, 3-O-deacetylorthosiphol I, and 2-O-deacetylorthosiphol J. Their structures were determined on the basis of the spectroscopic data. All the newly isolated diterpenes exhibited dose-dependent inhibition of nitric oxide (NO) production in lipopolysaccharide (LPS)-activated macrophage-like J774.1 cells, and 2-O-deacetylorthosiphonone A showed the most potent activity, with an IC(50) value of 35.0 microM, comparable to that of the positive control N(G)-monomethyl-L-arginine (L-NMMA; IC(50), 35.7 microM).
