Prediction of hERG inhibition of drug discovery compounds using biomimetic HPLC measurements

Abstract:The major causes of failure of drug discovery compounds in clinics are the lack of efficacy and toxicity. To reduce late-stage failures in the drug discovery process, it is essential to estimate early the probability of adverse effects and potential toxicity. Cardiotoxicity is one of the most often observed problems related to a compound's inhibition of the hERG channel responsible for the potassium cation flux. Biomimetic HPLC methods can be used for the early screening of a compound's lipophilicity, protein binding and phospholipid partition. Based on the published hERG pIC50 data of 90 marketed drugs and their measured biomimetic properties, a model has been developed to predict the hERG inhibition using the measured binding of compounds to alpha-1-acid-glycoprotein (AGP) and immobilised artificial membrane (IAM). A representative test set of 16 compounds was carefully selected. The training set, involving the remaining compounds, served to establish the linear model. The mechanistic model supports the hypothesis that compounds have to traverse the cell membrane and bind to the hERG ion channel to cause the inhibition. The AGP and the hERG ion channel show structural similarity, as both bind positively charged compounds with strong shape selectivity. In contrast, a good IAM partition is a prerequisite for cell membrane traversal. For reasons of comparison, a corresponding model was derived by replacing the measured biomimetic properties with calculated physicochemical properties. The model established with the measured biomimetic binding properties proved to be superior and can explain over 70% of the variance of the hERG pIC50 values.

Abstract:Effective drug discovery and molecular optimization procedures depend heavily on a comprehensive understanding of the reciprocal drug‐membrane interactions. This review explores recent developments in microfluidic lipid biomimetic models for screening these interactions. A synopsis of lipid‐based drug screening models is provided, along with a deeper analysis of the most promising microfluidic methods for their production and applications on drug screening. Developing new drugs is a complex, time‐consuming, and expensive process, essential to identify potential pharmacokinetic issues in the early stages to avoid investment in nonpromising candidates. Nearly half of all drug targets and key drug‐metabolizing enzymes are found in intracellular environments, compartmentalized by semipermeable barriers, the cell membranes. Inspired by their lipidic bilayer structure, lipid‐based biomimetic platforms are being developed to understand the biochemical and biophysical processes at the cellular membrane level. Considering the pharmaceutical industry's demands for efficient in vitro high throughput screening tools, microfluidic technology emerges as a promising solution to address challenges in lipid biomimetic models for screening applications. This involves the transformation of commonly used lipid‐based biomimetic models, like supported lipid bilayers and lipid vesicles, to miniaturized approachs and their evolution into a new generation of bilayer models. These include pore‐suspended and free‐standing lipid bilayers, black lipid membranes, and droplet interface bilayers. This review provides an overview of both generations of lipid biomimetic models used in drug‐membrane screening applications. Moreover, it delves into the intricacies of their production through microfluidic approaches and examines their screening applications in drug‐membrane interaction. The review concludes with a critical analysis of potential future directions in this rapidly evolving field.

Abstract:The 5-heterocyclic 2-(2,4-dihydroxyphenyl)-1,3,4-thiadiazoles were obtained as potential biologically active compounds. Lipophilicity is one of the most important physicochemical properties of compounds and was already taken into account during the drug candidates design and development. The lipophilicity of compounds was determined using the computational (log P) and chromatography (log kw, RMw) methods. The experimental ones included the reverse-phase column high performance liquid chromatography RP (HPLC) with C8, C18, phosphatidylcholine (IAM), and cholesterol stationary phases and the thin layer chromatography (RP-HPTLC) with C8 and C18 stationary phases and various organic modifiers under the isocratic conditions. Descriptive statistics, correlation, and PCA analyses were used to compare the obtained results. For lipophilicity estimation of the tested compounds by HPTLC, dioxane and MeOH seem to be particularly beneficial as organic modifiers. The chromatographic lipophilicity parameters log kw (RMw) were well correlated and highly redundant (85%) compared with those calculated. Most compounds possess lipophilicity parameters within the recommended range for drug candidates.

Estimation of volume of distribution in humans from high throughput HPLC-based measurements of human serum albumin binding and immobilized artificial membrane partitioning

Abstract:The volume of distribution (VD) in humans of 179 known drug molecules (acids, bases, and neutrals) has been modeled using two biomimetic-binding measurements. The phospholipid binding (log K (IAM)) and the plasma protein binding (log K (HSA)) have been calculated from gradient HPLC retention times on immobilized artificial membrane (IAM) and on human serum albumin (HSA) columns, respectively. The log VD values showed good correlation with the compounds' relative binding to IAM and HSA as follows: log VD=0.44 log K (IAM)-0.22 log K (HSA)-0.66; n=179, r2=0.76, s=0.33, and F=272. It was also observed that positively charged molecules bind relatively more to IAM, while negatively charged ones bind more to HSA, in line with the empirical observation that bases tend to have a larger volume of distribution than acids. These results suggest that with the help of these two simple high throughput HPLC-based biomimetic binding measurements an important in vivo drug disposition property can be estimated for use in early drug discovery.

Abstract:Drug-membrane interactions are important physicochemical properties to be considered at early drug discovery stage, which can provide deeper understanding of drug pharmacokinetics, pharmacodynamics and toxicity. Biomimetic phospholipid membrane chromatography (BPMC) is a chromatographic method in which biomimetic phospholipid membranes (phospholipid monolayer/bilayer and intact liposomes) are used as either stationary phase or pseudo-stationary phase, allowing rapid and effective mimicking and study of drug-membrane interactions. Over the last 30 years, various BPMC columns have been developed and successfully applied to the prediction of related drug properties. This review summarizes different BPMC modes, mainly focusing on their classification and preparation. The various solid supports of BPMC columns are summarized, including monolith, silica, agarose and open tubular column. Immobilization methods for BPMC as well as their advantages and drawbacks are also discussed, including physical adsorption, covalent binding, steric trapping, in-situ co-polymerization, etc. The major challenges and future prospects of BPMC are also briefly discussed.

Abstract:The interaction of drugs with biological membranes is a critical factor affecting their pharmacokinetics and pharmacodynamics, so understanding how drugs interact with biomembranes is of valuable interest for the pharmaceutical industry. However, such interactions are difficult to investigate due to the complexity of biological membranes, mainly related to the dynamic structure of membranes that are composed of wide variety of molecules. In this context, in vitro lipid-based membrane models emerged to offer a simple yet useful alternative way of studying drug-membrane interactions. Different lipid models can be used as in vitro biomembrane models, and this review presents a brief overview of the most promising membrane models used in this field. Also, the most used biophysical methodologies to study drugs' and membranes' properties using liposomes are reviewed. The significance of using liposomes as lipid membrane models to investigate drug-membrane interactions is discussed and future perspectives are presented.

Abstract:This study pioneers a comparison of the application of biomimetic techniques, immobilised artificial membrane liquid chromatography (IAM LC) and liposome electrokinetic capillary chromatography (LEKC), for the prediction of pulmonary drug permeability. The pulmonary absorption profiles of 26 structurally unrelated drug-like molecules were evaluated using their IAM hydrophobicity index (CHI IAM) measured in IAM LC, and the logarithm of distribution constants (log KLEKC) derived from the LEKC experiments. Lipophilicity (phospholipids) parameters obtained from IAM LC and most LEKC analyses were linearly related to the n-octanol/water partitioning coefficients of the neutral forms (i.e., log Po/w values) to a moderate extent. However, the relationship with distribution coefficients at the experimental pH (7.40) (i.e., log D7.4) were weaker overall for IAM LC data and sigmoidal for some liposome compositions (phosphatidyl choline (PC): phosphatidyl inositol (PI) 85:15 mol% and 90:10 mol%) and concentrations (4 mM) in LEKC. This suggests that phospholipid partitioning supports both hydrophobic and electrostatic interactions occurring between ionised drugs and charged phospholipid moieties. The latter interactions are original when compared to those taking place in the more established n-octanol/water partitioning systems. A stronger correlation (R2 > 0.65) was identified between the LEKC retention parameters, and the experimental apparent lung permeability (i.e., log Papp values) as opposed to the values obtained by IAM LC. Therefore, LEKC offers unprecedented advantages over IAM LC in simulating cell membrane partitioning processes in the pulmonary delivery of drugs. Although LEKC has the advantage of more effectively simulating the electrostatic and hydrophobic forces in drug/pulmonary membrane interactions in vitro, the technique is unsuitable for analysing highly hydrophilic neutral or anionic compounds at the experimental pH. Conversely, IAM LC is useful for analysing compounds spanning a wider range of lipophilicity. Its simpler and more robust implementation, and propensity for high-throughput automation make it a favourable choice for researchers in drug development and pharmacological studies.

Abstract:This paper reviews the functional polymer membrane and membrane based cell drug evaluation models for drug discovery. Based on the characteristics of biological membranes in vivo, chemical modification methods of synthetic membrane, including blending and surface modification are explored to mitigate the membrane fouling and improve biocompatibility. Different membrane-based cell models used in drug investigation and related trouble shooting are analyzed in detail. Specific attention is given to the current studies on ADME/Tox of drugs using membrane-based in vitro models of cells including Caco-2, hepatocytes or renal cells, which can be used to evaluate the feasibility of polymer membrane in drug investigation. The progress toward solving present bottlenecks of the facilitated cell models are supposed to provide great benefits to drug discovery in pharmaceutical industry.
Prediction of the chromatographic hydrophobicity index with immobilized artificial membrane chromatography using simple molecular descriptors and artificial neural networks

Abstract:Screening of physicochemical properties should be considered one of the essential steps in the drug discovery pipeline. Among the available methods, biomimetic chromatography with an immobilized artificial membrane is a powerful tool for simulating interactions between a molecule and a biological membrane. This study developed a quantitative structure-retention relationships model that would predict the chromatographically determined affinity of xenobiotics to phospholipids, expressed as a chromatographic hydrophobicity index determined using immobilized artificial membrane chromatography. A heterogeneous set of 261 molecules, mostly showing pharmacological activity or toxicity, was analyzed chromatographically to realize this goal. The chromatographic analysis was performed using the fast gradient protocol proposed by Valko, where acetonitrile was applied as an organic modifier. Next, quantitative structure-retention relationships modeling was performed using multiple linear regression (MLR) methods and artificial neural networks (ANNs) coupled with genetic algorithm (GA)-inspired selection. Subsequently, the selection of the best ANN was supported by statistical parameters, the sum of ranking differences approach with the comparison of rank by random numbers and hierarchical cluster analysis.
Estimation of phospholipophilicity of 1-[3-(arylpiperazin-1-yl)-propyl]-pyrrolidin-2-one derivatives on immobilized artificial membrane stationary phase and its correlation with biological data

Abstract:A molecule library containing 42 1-[3-(arylpiperazin-1-yl)-propyl]-pyrrolidin-2-one derivatives has been designed and synthesized. The phospholipophilicity of the obtained compounds has been determined using immobilized artificial membrane high-performance liquid chromatography (IAM-HPLC). The performed analysis allowed the calculation of log k(we) values for each of the tested compounds. Experimental phospholipophilicity data (log k(we)) has been compared with the affinity of the tested compounds to alpha(2)-adrenoceptors. Performed quantitative structure-activity relationship studies indicated that, for the tested compounds, there are dependences between affinity for alpha(2)-adrenoceptors and their log k(we) values. The obtained results confirmed that the applied chromatographic IAM-HPLC method could be useful in fast characterization of the phosholipophilicity of structurally closely related compounds as well as for larger series of compounds, such as drug candidates. It could also be used as a tool for further research into this group of compounds.

Abstract:Lipophilicity is one of the principal parameters that describe the pharmacokinetic behavior of a drug, including its absorption, distribution, metabolism, elimination, and toxicity. In this study, the lipophilicity and other physicochemical, pharmacokinetic, and toxicity properties that affect the bioavailability of newly synthesized dialkylaminoalkyldiquinothiazine hybrids as potential drug candidates are presented. The lipophilicity, as RM0, was determined experimentally by the RP-TLC method using RP18 plates and acetone–TRIS buffer (pH 7.4) as the mobile phase. The chromatographic parameters of lipophilicity were compared to computationally calculated partition coefficients obtained by various types of programs such as iLOGP, XLOGP3, WLOGP, MLOGP, SILCOS-IT, LogP, logP, and milogP. In addition, the selected ADMET parameters were determined in silico using the SwissADME and pkCSM platforms and correlated with the experimental lipophilicity descriptors. The results of the lipophilicity study confirm that the applied algorithms can be useful for the rapid prediction of logP values during the first stage of study of the examined drug candidates. Of all the algorithms used, the biggest similarity to the chromatographic value (RM0) for certain compounds was seen with iLogP. It was found that both the SwissADME and pkCSM web tools are good sources of a wide range of ADMET parameters that describe the pharmacokinetic profiles of the studied compounds and can be fast and low-cost tools in the evaluation of examined drug candidates during the early stages of the development process.

High throughput screening of physicochemical properties and in vitro ADME profiling in drug discovery

Abstract:Current advances of new technologies with robotic automated assays combined with highly selective and sensitive LC-MS enable high-speed screening of lead series libraries in many in vitro assays. In this review, we summarize state of the art high throughput assays for screening of key physicochemical properties such as solubility, lipophilicity, pKa, drug-plasma protein binding and brain tissue binding as well as in vitro ADME profiling. We discuss two primary approaches for high throughput screening of solubility, i.e. an automated 96-well plate assay integrated with LC-MS and a rapid multi-wavelength UV plate reader. We address the advantages of newly developed miniaturized techniques for high throughput pKa screening by capillary electrophoresis combined with mass spectrometry (CE-MS) with automated data analysis flow. Several new lipophilicity approaches other than octanol-water partitioning are critically reviewed, including rapid liquid chromatographic retention based approach, immobilized artificial membrane (IAM) partitioning and liposome, and potential microemulsion electrokinetic chromatography (MEEKC) for accurate screening of LogP. We highlight the sample pooling (namely cassette dosing, all-in-one, cocktail) as an efficient approach for high throughput screening of physicochemical properties and in vitro ADME profiling with emphasis on the benefit of on-line quality control. This cassette dosing approach has been widely adapted in drug discovery for rapid screening of in vivo pharmacokinetic parameters with significantly increased capacity and dramatically reduced animal usage.

Abstract:Lipophilic conjugates (LCs) of small molecule drugs have been used widely in clinical and pre-clinical studies to achieve a number of pharmacokinetic and therapeutic benefits. For example, lipophilic derivatives of drugs are employed in several long acting injectable products to provide sustained drug exposure for hormone replacement therapy and to treat conditions such as neuropsychiatric diseases. LCs can also be used to modulate drug metabolism, and to enhance drug permeation across membranes, either by increasing lipophilicity to enhance passive diffusion or by increasing protein-mediated active transport. Furthermore, such conjugation strategies have been employed to promote drug association with endogenous macromolecular carriers (e.g. albumin and lipoproteins), and this in turn results in altered drug distribution and pharmacokinetic profiles, where the changes can be 'general' (e.g. prolonged plasma half-life) or 'specific' (e.g. enhanced delivery to specific tissues in parallel with the macromolecular carriers). Another utility of LCs is to enhance the encapsulation of drugs within engineered nanoscale drug delivery systems, in order to best take advantage of the targeting and pharmacokinetic benefits of nanomedicines. The current review provides a summary of the mechanisms by which lipophilic conjugates, including in combination with delivery vehicles, can be used to control drug delivery, distribution and therapeutic profiles. The article is structured into sections which highlight a specific benefit of LCs and then demonstrate this benefit with case studies. The review attempts to provide a toolbox to assist researchers to design and optimise drug candidates, including consideration of drug-formulation compatibility.

The Effect of Restaurant Menu Labeling on Consumer's Choice: Evidence from a Choice Experiment Involving Eye-Tracking

Abstract:Eating away from home has been noted to be a contributor to the rising obesity epidemic in the U.S. The U.S. Food and Drug Administration has announced plans to require calorie information to certain food retail establishments. However, the effectiveness of such a requirement has been found to be mixed with the literature. The objective of this paper was to understand the role of various nutritional labels (i.e. calorie, percent daily intake, and traffic light signals) on food choice at both sit down and fast food establishments. Our results indicate that participants in the price only treatment chose meals with higher caloric content from both sit down and fast food menus. However, we find that calorie only information provides the largest reduction in calories in a meal from a sit down menu, but percent daily intake in conjunction with calorie information provides the largest reduction in calories for a fast food menu. Further, via eye tracking technology we find that participants looked at the nutritional information similarly across treatments.

Abstract:ABSTRACTIn this review, we first summarize the structure and properties of biological membranes and the routes of passive drug transfer through physiological barriers. Lipophilicity is then introduced in terms of the intermolecular interactions it encodes. Finally, lipophilicity indices from isotropic solvent systems and from anisotropic membrane-like systems are discussed for their capacity to predict passive drug permeation across biological membranes such as the intestinal epithelium, the blood-brain barrier (BBB) or the skin. The broad evidence presented here shows that beyond the predictive power of lipophilicity parameters, the various intermolecular forces they encode allow a mechanistic interpretation of passive drug permeation.

Abstract:Lipophilicity is one of the most important properties of compounds required to estimate the absorption, distribution, and transport in biological systems, in addition to solubility, stability, and acid–base nature. It is crucial in predicting the ADME profile of bioactive compounds. The study assessed the usefulness of computational and chromatographic methods (thin-layer chromatography in a reversed-phase system, RP-TLC) for estimating the lipophilicity of 21 newly synthesized compounds belonging to diquinothiazines and quinonaphthiazines. In order to obtain reliable values of the relative lipophilicities of diquinothiazines and quinonaphthiazines, the partition coefficients obtained using different algorithms such as AlogPs, AClogP, AlogP, MLOGP, XLOGP2, XLOGP3, logP, and ClogP were compared with the chromatographic RM0 values of all the tested compounds measured by the experimental RP-TLC method (logPTLC). Additionally, logPTLC values were also correlated with other descriptors, as well as the predicted ADME and drug safety profiling parameters. The linear correlations of logPTLC values of the tested compounds with other calculated molecular descriptors such as molar refractivity, as well as ADME parameters (Caco-2 substrates, P-gp inhibitors, CYP2C19, and CYP3A4) generally show poor predictive power. Therefore, in silico ADME profiling can only be helpful at the initial step of designing these new candidates for drugs. The compliance of all discussed diquinothiazines and naphthoquinothiazines with the rules of Lipiński, Veber, and Egan suggests that the tested pentacyclic phenothiazine analogs have a chance to become therapeutic drugs, especially orally active drugs.

Abstract:Liposomes are nowadays a matter of tremendous interest. Due to their amphiphilic character, various substances with different properties can be incorporated into them and they are especially suitable as a model system for controlled transport of bioactive substances and drugs to the final destination in the body; for example, COVID-19 vaccines use liposomes as a carrier of mRNA. Liposomes mimicking composition of various biological membranes can be prepared with a proper choice of the lipids used, which proved to be important tool in the early drug development. This review deals with commonly used methods for the preparation and characterization of liposomes which is essential for their later use. The alternative capillary electrophoresis methods for physico-chemical characterization such as determination of membrane permeability of liposome, its size and charge, and encapsulation efficiency are included. Two different layouts using liposomes to yield more efficient separation of various analytes are also presented, capillary electrochromatography, and liposomal electrokinetic chromatography.
