Abstract:The objectives of the present study were to (1) assess the shear bond strengths of resin-reinforced glass ionomer Fuji Ortho LC and GC Fuji Ortho cements under differing conditions and (2) compare their bonding performance with that of conventional resin composite bonding systems. A sample of 264 bovine incisors was divided into 22 groups of 12 teeth each and bonded with SPEED central incisor brackets. Enamel surfaces of the teeth in the two experimental groups were conditioned according to the manufacturer's instructions; moreover, groups unconditioned before bonding were also included under both wet and dry conditions. A self-cure composite resin (Phase II) and a light-cure composite resin (Transbond XT) served as controls and were etched with 37% phosphoric acid and bonded in a dry field. After incubation at 37 degrees C for 24 hours and for seven days, the specimens were tested to failure with a shear force in an Instron machine. The Adhesive Remnant Index (ARI) was used to assess the amount of resin left on the enamel surfaces after debonding. Selected specimens were examined using scanning electron microscopy. Statistical analyses included analysis of variance tests, t-tests, and correlation coefficient calculations and showed that no significant difference existed between the glass ionomer cements under wet or dry conditions, provided the enamel was conditioned with 10% polyacrylic acid before bonding. Both glass ionomer cements were thus acceptable for bonding. Transbond XT had the highest mean shear bond strength irrespective of the incubation period. A positive correlation was obtained between the ARI scores and bond strength.

Abstract:Background and objective Yeasts have the remarkable capability to transform and integrate inorganic selenium into their cellular structures, thereby enhancing its bioavailability and reducing its toxicity. In recent years, yeasts have attracted attention as potential alternative sources of protein. Methods This study explores the selenium accumulation potential of two less explored yeast strains, namely the probiotic Saccharomyces boulardii CCDM 2020 and Pichia fermentas CCDM 2012, in comparison to the extensively studied Saccharomyces cerevisiae CCDM 272. Our investigation encompassed diverse stress conditions. Subsequently, the selenized yeasts were subjected to an INFOGEST gastrointestinal model. The adherence and hydrophobicity were determined with undigested cells Results Stress conditions had an important role in influencing the quantity and size of selenium nanoparticles (SeNPs) generated by the tested yeasts. Remarkably, SeMet synthesis was limited to Pichia fermentas CCDM 2012 and S. boulardii CCDM 2020, with S. cerevisiae CCDM 272 not displaying SeMet production at all. Throughout the simulated gastrointestinal digestion, the most substantial release of SeCys2, SeMet, and SeNPs from the selenized yeasts occurred during the intestinal phase. Notably, exception was found in strain CCDM 272, where the majority of particles were released during the oral phase. Conclusion The utilization of both traditional and non-traditional selenized yeast types, harnessed for their noted functional attributes, holds potential for expanding the range of products available while enhancing their nutritional value and health benefits.

Abstract:Selenium-enriched yeast (selenium yeast) are one of the most popular sources of selenium supplementation used in the agriculture and human nutritional supplements industries. To enhance the production efficiency of selenium yeast, we sought to develop a method to identify, and ultimately select for, strains of yeast with enhanced selenium accumulation capabilities. Selenite resistance of four genetically diverse strains of Saccharomyces cerevisiae was assayed in various conditions, including varying carbon sources, nitrogen sources, and phosphate amounts, and they were correlated with selenium accumulation in a commercially relevant selenium-containing growth medium. Glycerol- and selenite-containing media was used to select for six yeast isolates with enhanced selenite resistance. One isolate was found to accumulate 10-fold greater selenium (0.13 to 1.4 mg Se g−1 yeast) than its parental strain. Glycerol- and selenium-containing medium can be used to select for strains of yeast with enhanced selenium accumulation capability. The methods identified can lead to isolation of industrial yeast strains with enhanced selenium accumulation capabilities that can result in greater cost efficiency of selenium yeast production. Additionally, the selection method does not involve the construction of transgenic yeast, and thus produces yeasts suitable for use in human food and nutrient supplements.

Abstract:The use of agro-industrial by-products as feedstock for selenium-enriched yeast biomass production is an innovative and low-cost method. Thus, the present study aims for the utilization of sugarcane bagasse and corn bran hydrolysates for the production of selenium-enriched yeast and the evaluation of the effect of selenium in growth and cell composition. The hydrolysates were obtained using acid pretreatment, and yeasts were evaluated in different medium compositions. All evaluated yeasts were able to grow in the presence of 15 mg/L selenium. S. cerevisiae strains presented the major tolerance to selenium and better relation between selenium uptake and cell growth, with maximum cell biomass production of 7.97 ± 0.24 g/L and maximum selenium uptake of 1193 ± 336 ppm and 99% cell viability. Selenium also enhanced the lipid and protein concentration in cell extracts obtained after mechanical disruption. Regarding agro-industrial hydrolysates, corn bran hydrolysate allowed a better growth, with the production of 4.25 ± 0.49 g/L of biomass enriched with 167 ± 18 ppm selenium and 100% of viability. Starchy and lignocellulosic biomasses presented potential as low-cost alternatives for single-cell protein contributing to the development of sustainable and economically viable technologies.

Abstract:PURPOSE The aim of this study was to use clinical, radiographic, and histologic examinations to compare the relative success of gray mineral trioxide aggregate (MTA), white MTA, and formocresol as pulp dressings in pulpotomized primary teeth. METHODS Twenty-four children, each with at least 3 primary molars requiring pulpotomy, were selected for this study's clinical and radiographic portion. An additional 15 carious primary teeth planned for serial extraction were selected for this study's histologic portion. All selected teeth were evenly divided into 3 test groups and treated with pulpotomies. Gray MTA was used as the pulp dressing for one third of the teeth, white MTA was the dressing for one third, and the remaining one third were treated with formocresol. The treated teeth selected for the clinical and radiographic evaluations were monitored periodically for 12 months. The treated teeth selected for histologic study were monitored periodically and extracted 6 months postoperatively. RESULTS Four children with 12 pulpotomized teeth failed to return for any follow-up evaluations in the clinical and radiographic study. Of the remaining 60 teeth in 20 patients, 1 tooth (gray MTA) exfoliated normally and 6 teeth (4 white MTA and 2 formocresol) failed due to abscesses. The remaining 53 teeth appeared to be clinically and radiographically successful 12 months postoperatively. Pulp canal obliteration was a radiographic finding in 11 teeth treated with gray MTA and 1 tooth treated with white MTA. In the histologic study, both types of MTA successfully induced thick dentin bridge formation at the amputation sites, while formocresol induced thin, poorly calcified dentin. Teeth treated with gray MTA demonstrated pulp architecture nearest to normal pulp by preserving the odontoblastic layer and delicate fibrocellular matrix, yet few inflammatory cells or isolated calcified bodies were seen. Teeth treated with white MTA showed a denser fibrotic pattern, with more isolated calcifications in the pulp tissue along with secondary dentin formation. CONCLUSIONS Gray MTA appears to be superior to white MTA and formocresol as a pulp dressing for pulpotomized primary teeth.

Abstract:An analytical method was developed for the speciation of elemental selenium (Se(0)) in selenized yeasts by anion-exchange HPLC-ICP-MS after its chemical transformation into SeSO32− by reaction with sodium sulfite. The presence of Se(0) in the yeasts was further confirmed by single-particle ICP-MS. Indeed, Se nanoparticles, if present, are expected to be, at least partly, Se(0). X-ray photoelectron spectroscopy, a well-recognized technique for chemical element speciation in the solid state, was also used with this objective. Both methods were able to confirm the presence of Se(0) in the selenized yeasts but failed to provide reliable quantitative results. Analytical performances of the HPLC-ICP-MS method were then evaluated for Se(0) determination. Quantification limits of 1 mg/kg were reached. The recovery levels from an added quantity comprised between 93 and 101%. Within-run and between-run precisions were both below 8%. The procedure developed was finally applied to quantify Se(0) content in a series of seven yeast batches from different suppliers. Se(0) was found to be present in all the studied yeasts and represented on average 10–15% of the total Se.

Extraction recovery and speciation of selenium in Se-enriched yeast

Abstract:The complete characterization of selenium-enriched yeast in terms of selenium species has been the goal of extensive research for the last three decades. This contribution addresses the two outstanding questions: the mass balance of the identified and reported selenium species and the possible presence of inorganic selenium. For this purpose, four procedures have been designed combining, in diverse order, the principal steps of selenium speciation analysis in Se-rich yeast: extraction of the Se-metabolome, derivatization of cysteine and Se-cysteine (SeCys) residues, proteolysis, and definitive Se recovery using SDS extraction, followed by mineralization. The recovery of selenium in each step and its speciation were controlled by ICP MS and by reversed-phase HPLC-ICP MS, respectively. The study, carried out for the SELM-1 reference material, demonstrated the presence of about 10% of inorganic selenium and a serious risk of losses of SeCys during derivatization and proteolysis. As result of our work, we postulate the following values for SELM-1: Se-metabolome fraction (SeMF) 14.8 ± 0.7%; total selenomethionine (SeMet) 66.2 ± 2.7% (including ca. 1.5% SeMet present in the SeMF); total SeCys 12.5 ± 1.5% (including 2% of SeCys present in the Se-MF); total inorganic selenium 9.7 ± 1.7%, accounting for > 99.8% of the selenium.

Abstract:Numerous studies have demonstrated the efficacy of selenium compounds in preventing and treating lifestyle-related diseases such as cancer and cardiovascular disorders. The formulation of selenium-enriched supplements for humans and animals, particularly those containing selenium yeast, is highly advantageous. These products are rich in organic selenium derivatives, showing significantly higher bioavailability than inorganic forms of selenium. A particularly promising selenium analogue of sulphur-containing compounds is selenobiotin. The literature indicates that Phycomyces blakesleeanus and Escherichia coli strains can synthesise this compound. This research aimed to evaluate the effect of selenium supplementation on the biosynthesis of biotin and selenobiotin in Trichosporon cutaneum and Meyerozyma guilliermondii. The results have the potential to advance biotechnological approaches for the production of selenobiotin for various applications. A method based on affinity chromatography was used to quantify selenobiotin. The results confirmed that both yeast strains could synthesise selenobiotin in addition to biotin. In M. guilliermondii cells, selenobiotin accounted for up to 17.3% of the total biotin vitamer fraction. In comparison, in T. cutaneum cells, it accounted for up to 28.4% of the sum of biotin and its analogues. The highest levels of selenobiotin were observed in cells cultured with selenomethionine.

Abstract:The ability of Coriolus versicolor medicinal mushroom to grow and accumulate selenium during submerged cultivation in a selenium‐fortified medium is examined in this paper. For selenium supplementation, commercial selenium yeast was used. Control, nonenriched sample and reference cultures cultivated in the medium enriched with commercial yeast Saccharomyces cerevisiae were also prepared. The mushroom demonstrated a high ability to accumulate selenium from the added source (around 970 and 1,300 µg/g of dry mycelium weight for samples enriched with selenium in a concentration of 10 and 20 mg Se/L, respectively). The addition of selenium significantly (p ≤ .05) increased the biomass yield, whereas the addition of nonenriched yeast had no significant (p ≤ .05) impact. Furthermore, regression analysis showed statistically significant (p ≤ .05) and positive correlations between the content of Se and Fe (r = .92), Se and Cu (r = .92), Se and Mn (r = .98), and Se and Sr (r = .96), suggesting that selenium incorporation was followed by incorporation of these elements, and led to mineral enrichment of the obtained mycelium. Methanol extracts prepared from mycelium biomass demonstrated a better inhibitory effect on Gram‐positive bacterial strains with minimal inhibitory concentrations between <0.3125 and 40 mg/ml. The obtained results showed that selenium yeast could be used for obtaining a potential novel food supplement: mushroom biomass with high selenium content and enhanced mineral composition.

Abstract:Yeast (Saccharomyces cerevisiae) and lactic bacteria have shown their ability to accumulate and transform inorganic selenium into organo Se compounds. The objective of this work was to evaluate selenium biotransformation during brewing by using S. cerevisiae and Saccharomyces uvarum for Ale and Lager fermentation, respectively. Se-enriched beer was produced by the addition of sodium selenite (0, 0.2, 1.0, 2.0, 10.0, 20.0 μg Se mL(-1), respectively) to the fermentation media composed of yeast, malt extract and water. The alcoholic fermentation process was not affected by the presence of selenium regardless of the type of Saccharomyces being used. The percentage of selenium incorporated into beer, added between 1.0 and 10 μg mL(-1) was 55-60% of the selenium initially present. Se-compounds in post-fermentation (beer and yeast) products were investigated by using an analytical methodology based on HPLC-ICP-MS. For this purpose, several sample treatments, including ultrasonic-assisted enzymatic hydrolysis, in conjunction with different separation mechanisms like dialysis and anion exchange HPLC chromatography were applied for unambiguously identifying Se-species that produce during brewing. Selenomethionine was the main selenium compound identified in beer and yeast, being this species in the only case of the former not associated to peptides or proteins.

Abstract:We present some coupling properties of spiral fiber gratings. LP/sub 01/ modes in spiral fiber gratings were demonstrated and analyzed to couple efficiently to both LP/sub 11/ even and odd modes. LP/sub 01/ modes were shown to efficiently couple to both LP/sub 11/ even and odd modes by spiral gratings as predicted. A spiral fiber grating that converts the LP/sub 01/ mode equally to LP/sub 11/ even and odd modes, and a spiral grating that appreciably converts the LP/sub 01/ mode back to the same LP/sub 01/ mode through the LP/sub 01/-to-LP/sub 11/ and LP/sub 11/-to-LP/sub 01/ processes, were also demonstrated and analyzed.

Abstract:The study investigated biosynthesis of selenoproteins by Saccharomyces. cerevisiae using inorganic selenium. Selenium supplement via two stages was carried out during fermentation and the physicochemical characteristics of selenoproteins and its antioxidant activities were examined through in vitro assessment procedures. After fermentation, dry cells weight (7.47 g/L) and selenium content (3079.60 μg/kg) in the yeast were achieved when fermentation time points at the 6th hour and the 9th hour were chosen to supplement 30% and 70% of 30 μg/mL Na2SeO3 respectively. A maximal yield of selenium content in selenoproteins reached 1013.07 μg/g under optimized culture conditions and was 133-fold higher than the control. One new band with molecular weight of 26.76 KDa appeared in conjugated selenoproteins of sodium dodecyl sulphate-polyacrylamide gel electrophoresis. Surface structure of selenoproteins and the control was different by Scanning electron microscopy images. Infrared spectrometry analysis demonstrated that groups of HSe, SeO and C-Se-O involved in selenoproteins were important pieces of evidence showing presence of Se embedded in the protein molecule. Selenoproteins showed strong antioxidant activities on DPPH·, OH and ·O2−, which was much higher than the control proteins.Therefore, the study provided an efficient selenium-enriched culture method of inorganic selenite to organic selenium and basis for selenoproteins applications.

Abstract:Uterine bleeding may be caused either by benign organic pathology as well as different dysfunctional conditions. Medical treatment with progestins, danazol or GnRH analogues is usually used as the first choice therapy. Where the symptoms persist, hysterectomy is generally proposed; it has been calculated that from 500,000 to 700,000 such operations are performed annually in the United States. The authors review the state of the art of the endometrial ablation as an alternative to hysterectomy and other medical therapy. Endometrial ablation either by means of the YAG-laser or with the resectoscope, would seem to play a role in well-selected cases of dysfunctional uterine bleeding. In the authors opinion a multicenter study on large numbers of patients is needed in order to confirm these preliminary results.
Comparative Effects of Nano Elemental Selenium and Sodium Selenite on Selenium Retention in Broiler Chickens

Abstract:Two experiments were performed to explore the selenium retention of Nano elemental selenium (Nano-Se) in Arbor Acre male broiler chickens as compared with sodium selenite. A 2×4 factorial arrangement with 0.15, 0.30, 0.60 and 1.20mg/kg dietary Se from Nano-Se or sodium selenite added to a maize–soybean meal diet was conducted to study the effects of Se source and level on growth performance, serum glutathione peroxidase (GSH-Px) activity, Se concentration in serum and tissue. Furthermore, selenium retention of Nano-Se and selenite was determined in chicks by the oral or intravenous administration of the radiolabeled Se and the in vivo ligated intestinal loop procedure. As for selenite, average daily gain (ADG), gain/feed and survival ratio reached a plateau at the Se concentration of 0.15–0.30mg/kg and then declined as supplemental selenite increased. As for Nano-Se, ADG, gain/feed and survival ratio reached a plateau at the Se concentration of 0.15–1.20mg/kg. Survival ratio, ADG and gain/feed increased quadratically (P<0.05) as dietary selenite increased (P<0.05), and increased linearly and quadratically (P<0.05) as dietary Nano-Se increased (P<0.05). Selenium concentrations in serum, liver and breast muscle increased linearly and quadratically (P<0.05) as the dietary Se level increased for either Se source, but the magnitude of increase was substantially greater (P<0.05) when Nano-Se was fed. Retention of the orally or intravenously administered 75Se showed that the percentages of Nano-Se in the whole body and liver tissue were much higher (P<0.05) than those of selenite. Intestinal transport of 75Se from ligated loop to body showed that the transfer of Nano-Se from the intestinal lumen to the body was higher (P<0.05) than that of selenite, while the intestinal retention of Nano-Se was lower (P<0.05) than that of selenite. The results showed that the range between optimal and toxic dietary levels of Nano-Se was wider than that of sodium selenite, and Nano-Se was more efficiently retained in the body than sodium selenite.

Abstract:This study aimed to screen a mutant of Candida utilis SE-172 with high selenite tolerance and glutathione (GSH) biosynthesis capability via 60Co γ-radiation mutagenesis to prepare selenium (Se)-enriched yeast. The maximal intracellular contents of GSH and organic Se of 22.94 mg/g and 1308.1 μg/g were obtained, respectively, under a batch culture of SE-172. The physiological mechanism underlying increased GSH and organic Se contents in Se/GSH-enriched C. utilis SE-172 was revealed based on assaying activities of γ-glutamylcysteine synthase (γ-GCS) involved in GSH biosynthesis and selenophosphate synthase (SPS) related to organic Se bioconversion, and by determining intracellular ATP and NADH contents and ATP/ADP and NADH/NAD+ ratios associated with energy supply and regeneration. Moreover, the effect of this selenized yeast on anti-inflammatory and antioxidant activities in mice with colitis was investigated. The supplementation of Se/GSH-enriched yeast decreased the dextran sodium sulfate-induced damage to colon tissues, reduced the expression of pro-inflammatory factors [interleukin (IL)-6, IL-1β, and tumor necrosis factor-α (TNF-α)] in serum, increased the antioxidant-related enzyme [superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px)] activities, and decreased the malondialdehyde content in colon. The Se/GSH-enriched C. utilis SE-172 showed potent anti-inflammatory and antioxidant activities in mice with colitis.

Mechanism of selenite tolerance during barley germination: A combination of tissue selenium metabolism alterations and ascorbate-glutathione cycle modulation

Abstract:Selenite is widely used to increase Selenium (Se) content in cereals, however excessive selenite may be toxic to plant growth. In this study, barley was malted to elucidate the action mechanism of selenite in the generation and detoxification of oxidative toxicity. The results showed that high doses (600 μM) of selenite radically increased oxidative stress by the elevated accumulation of superoxide and malondialdehyde, leading to phenotypic symptoms of selenite-induced toxicity like stunted growth. Barley tolerates selenite through a combination of mechanisms, including altering Se distribution in barley, accelerating Se efflux, and increasing the activity of some essential antioxidant enzymes. Low doses (150 μM) of selenite improved barley biomass, respiratory rate, root vigor, and maintained the steady-state equilibrium between reactive oxygen species (ROS) and antioxidant enzyme. Selenite-induced proline may act as a biosignal to mediate the response of barley to Se stress. Furthermore, low doses of selenite increased the glutathione (GSH) and ascorbate (AsA) concentrations by mediating the ascorbate-glutathione cycle (AsA-GSH cycle). GSH intervention and dimethyl selenide volatilization appear to be the primary mechanisms of selenite tolerance in barley. Thus, results from this study will provide a better understanding of the mechanisms of selenite tolerance in crops.
Effects of Low-Level Inorganic Selenium on Selenium Bioaccumulation in Escherichia coli: A Bioanalytical Study Using Graphite Furnace Atomic Absorption Spectrometry (GFAAS)

Abstract:This study aimed to assess the bioaccumulation of selenite – Se(IV) – and selenate – Se(VI) – in Escherichia coli ATCC 11775 biomass. Escherichia coli cultures were exposed to low selenium concentrations (0.3–30.0 μM), and subsequent effects on growth and viability were evaluated. Selenium content in biomass was quantified using graphite furnace atomic absorption spectroscopy (GFAAS) and inductively coupled plasma mass spectrometry (ICP-MS), with Se recovery assessed through mass balance analysis. The average percentage of bioaccumulated Se exhibited variability depending on the Se species at different concentrations. At a concentration of 30.0 μM, elemental selenium formation emerged within the Se(IV) assay, leading to a significantly higher mean percentage of bioaccumulated selenium for Se(IV) (41.1%) compared to Se(VI) (15.8%). While the mean percentage of bioaccumulated selenium showed no significant variance across the Se(VI) concentrations, a significant difference was observed across the Se(IV) concentrations (ranging from 6.1% to 41.1%). Considering the percentage of Se recovered in the solid and liquid phases, the mass balance ranged from 82.1% to 94.5%. These findings underscore the ability of Escherichia coli ATCC 11775 to accumulate notable quantities of selenium, with the amount of bioaccumulated Se influenced by Se(IV) concentration but not by Se(VI). Furthermore, bacterial viability remained unaffected by selenium species and concentrations. This study contributes valuable insights into the bioaccumulation dynamics of Se(IV) and Se(VI) in non-genetically modified Escherichia coli , shedding light on factors governing selenium accumulation in bacterial biomass, particularly regarding natural biological representation, standardization, comparability, and applicability to environmental studies.
