Abstract:Orientation selectivity (OS) is a prominent and well studied feature of early visual processing in mammals, but recent work has highlighted the possibility that parallel OS circuits might exist in multiple brain locations. Although both classic and modern work has identified an OS mechanism in selective wiring from lateral geniculate nucleus (LGN) to primary visual cortex, OS responses have now been found upstream of cortex in mouse LGN and superior colliculus, suggesting a possible origin in the retina. Indeed, retinal OS responses have been reported for decades in rabbit and more recently in mouse. However, we still know very little about the properties and mechanisms of retinal OS in the mouse, including whether there is a distinct OS ganglion cell type, which orientations are represented, and what are the synaptic mechanisms of retinal OS. We have identified two novel types of OS ganglion cells in the mouse retina that are highly selective for horizontal and vertical cardinal orientations. Reconstructions of the dendritic trees of these OS ganglion cells and measurements of their synaptic conductances offer insights into the mechanism of the OS computation at the earliest stage of the visual system. Significance statement: Orientation selectivity (OS) is one of the most well studied computations in the brain and has become a prominent model system in various areas of sensory neuroscience. Although the cortical mechanism of OS suggested by Hubel and Wiesel (1962) has been investigated intensely, other OS cells exist upstream of cortex as early as the retina and the mechanisms of OS in subcortical regions are much less well understood. We identified two ON retinal ganglion cells (RGCs) in mouse that compute OS along the horizontal (nasal-temporal) and vertical (dorsoventral) axes of visual space. We show the relationship between dendritic morphology and OS for each RGC type and reveal new synaptic mechanisms of OS computation in the retina.

Abstract:Classic ON-OFF direction-selective ganglion cells (DSGCs) that encode the four cardinal directions were recently shown to also be orientation-selective. To clarify the mechanisms underlying orientation selectivity, we employed a variety of electrophysiological, optogenetic, and gene knock-out strategies to test the relative contributions of glutamate, GABA, and acetylcholine (ACh) input that are known to drive DSGCs, in male and female mouse retinas. Extracellular spike recordings revealed that DSGCs respond preferentially to either vertical or horizontal bars, those that are perpendicular to their preferred-null motion axes. By contrast, the glutamate input to all four DSGC types measured using whole-cell patch-clamp techniques was found to be tuned along the vertical axis. Tuned glutamatergic excitation was heavily reliant on type 5A bipolar cells, which appear to be electrically coupled via connexin 36 containing gap junctions to the vertically oriented processes of wide-field amacrine cells. Vertically tuned inputs are transformed by the GABAergic/cholinergic "starburst" amacrine cells (SACs), which are critical components of the direction-selective circuit, into distinct patterns of inhibition and excitation. Feed-forward SAC inhibition appears to "veto" preferred orientation glutamate excitation in dorsal/ventral (but not nasal/temporal) coding DSGCs "flipping" their orientation tuning by 90° and accounts for the apparent mismatch between glutamate input tuning and the DSGC's spiking response. Together, these results reveal how two distinct synaptic motifs interact to generate complex feature selectivity, shedding light on the intricate circuitry that underlies visual processing in the retina.

Abstract:The orientation of visual stimuli is a salient feature of visual scenes. In vertebrates, the first neural processing steps generating orientation selectivity take place in the retina. Here, we dissect an orientation-selective circuit in the larval zebrafish retina and describe its underlying synaptic, cellular, and molecular mechanisms. We genetically identify a class of amacrine cells (ACs) with elongated dendritic arbors that show orientation tuning. Both selective optogenetic ablation of ACs marked by the cell-adhesion molecule Teneurin-3 (Tenm3) and pharmacological interference with their function demonstrate that these cells are critical components for orientation selectivity in retinal ganglion cells (RGCs) by being a source of tuned GABAergic inhibition. Moreover, our morphological analyses reveal that Tenm3(+) ACs and orientation-selective RGCs co-stratify their dendrites in the inner plexiform layer, and that Tenm3(+) ACs require Tenm3 to acquire their correct dendritic stratification. Finally, we show that orientation tuning is present also among bipolar cell presynaptic terminals. Our results define a neural circuit underlying orientation selectivity in the vertebrate retina and characterize cellular and molecular requirements for its assembly.

Abstract:Orientation selectivity is a property of mammalian primary visual cortex (V1) neurons, yet its emergence along the visual pathway varies across species. In carnivores and primates, elongated receptive fields first appear in V1, whereas in lagomorphs such receptive fields emerge earlier, in the retina. Here we examine the mouse visual pathway and reveal the existence of orientation selectivity in lateral geniculate nucleus (LGN) relay cells. Cortical inactivation does not reduce this orientation selectivity, indicating that cortical feedback is not its source. Orientation selectivity is similar for LGN relay cells spiking and subthreshold input to V1 neurons, suggesting that cortical orientation selectivity is inherited from the LGN in mouse. In contrast, orientation selectivity of cat LGN relay cells is small relative to subthreshold inputs onto V1 simple cells. Together, these differences show that although orientation selectivity exists in visual neurons of both rodents and carnivores, its emergence along the visual pathway, and thus its underlying neuronal circuitry, is fundamentally different.

Abstract:The origin of orientation selectivity in the responses of simple cells in cat visual cortex serves as a model problem for understanding cortical circuitry and computation. The feed-forward model posits that this selectivity arises simply from the arrangement of thalamic inputs to a simple cell. Much evidence, including a number of recent intracellular studies, supports a primary role of the thalamic inputs in determining simple cell response properties, including orientation tuning. This mechanism alone, however, cannot explain the invariance of orientation tuning to changes in stimulus contrast. Simple cells receive push-pull inhibition: ON inhibition in OFF subregions and vice versa. Addition of such inhibition to the feed-forward model can account for this contrast invariance, provided the inhibition is sufficiently strong. The predictions of “normalization” and “feedback” models are reviewed and compared with the predictions of this modified feed-forward model and with experimental results. The modified feed-forward and the feedback models ascribe fundamentally different functions to cortical processing.

Abstract:For over three decades, the mechanism of orientation selectivity of visual cortical neurones has been hotly debated. While intracortical inhibition has been implicated as playing a vital role, it has been difficult to observe it clearly. On the basis of recent findings, we propose a model in which the visual cortex brings together a number of different mechanisms for generating orientation-selective responses. Orientation biases in the thalamo-cortical input fibres provide an initial weak selectivity either directly in the excitatory input or by acting via cortical interneurones. This weak selectivity of postsynaptic potentials is then amplified by voltage-sensitive conductances of the cell membrane and excitatory and inhibitory intracortical circuitry, resulting in the sharp tuning seen in the spike discharges of visual cortical cells.

Abstract:Direction selectivity represents a fundamental computation found across multiple sensory systems. In the mammalian visual system, direction selectivity appears first in the retina, where excitatory and inhibitory interneurons release neurotransmitter most rapidly during movement in a preferred direction. Two parallel sets of interneuron signals are integrated by a direction-selective ganglion cell, which creates a direction preference for both bright and dark moving objects. Direction selectivity of synaptic input becomes amplified by action potentials in the ganglion cell dendrites. Recent work has elucidated direction-selective mechanisms in inhibitory circuitry, but mechanisms in excitatory circuitry remain unexplained.

Abstract:Orientation or axial selectivity, the property of neurons in the visual system to respond preferentially to certain angles of a visual stimuli, plays a pivotal role in our understanding of visual perception and information processing. This computation is performed as early as the retina, and although much work has established the cellular mechanisms of retinal orientation selectivity, how this computation is organized across the retina is unknown. Using a large dataset collected across the mouse retina, we demonstrate functional organization rules of retinal orientation selectivity. First, we identify three major functional classes of retinal cells that are orientation selective and match previous descriptions. Second, we show that one orientation is predominantly represented in the retina and that this predominant orientation changes as a function of retinal location. Third, we demonstrate that neural activity plays little role on the organization of retinal orientation selectivity. Lastly, we use modeling followed by validation experiments to demonstrate that the overrepresented orientation aligns along concentric axes. These results demonstrate that, similar to direction selectivity, orientation selectivity is organized in a functional map as early as the retina.

Can Retinal Ganglion Cell Dipoles Seed Iso-Orientation Domains in the Visual Cortex?

Abstract:It has been argued that the emergence of roughly periodic orientation preference maps (OPMs) in the primary visual cortex (V1) of carnivores and primates can be explained by a so-called statistical connectivity model. This model assumes that input to V1 neurons is dominated by feed-forward projections originating from a small set of retinal ganglion cells (RGCs). The typical spacing between adjacent cortical orientation columns preferring the same orientation then arises via Moiré-Interference between hexagonal ON/OFF RGC mosaics. While this Moiré-Interference critically depends on long-range hexagonal order within the RGC mosaics, a recent statistical analysis of RGC receptive field positions found no evidence for such long-range positional order. Hexagonal order may be only one of several ways to obtain spatially repetitive OPMs in the statistical connectivity model. Here, we investigate a more general requirement on the spatial structure of RGC mosaics that can seed the emergence of spatially repetitive cortical OPMs, namely that angular correlations between so-called RGC dipoles exhibit a spatial structure similar to that of OPM autocorrelation functions. Both in cat beta cell mosaics as well as primate parasol receptive field mosaics we find that RGC dipole angles are spatially uncorrelated. To help assess the level of these correlations, we introduce a novel point process that generates mosaics with realistic nearest neighbor statistics and a tunable degree of spatial correlations of dipole angles. Using this process, we show that given the size of available data sets, the presence of even weak angular correlations in the data is very unlikely. We conclude that the layout of ON/OFF ganglion cell mosaics lacks the spatial structure necessary to seed iso-orientation domains in the primary visual cortex.

Stimulation Of Non-Classical Receptive Field Enhances Orientation Selectivity In The Cat

Abstract:We have investigated how the nonclassical receptive field (nCRF) affects dynamic orientation selectivity of cells in the primary visual cortex (VI) in anaesthetized and paralysed cats using the reverse correlation method. We found that tuning to the orientation of the test stimulus depends on the size of the stimulation area. A significant sharpening of orientation tuning was induced by nCRF stimulation, with the magnitude of the effect increasing with the size of stimulation. The effect of the nCRF on the temporal dynamics of orientation tuning was also investigated by examining the tuning over a range of delays from stimulus onset. We found small but detectable changes in both the preferred orientation and the bandwidth of tuning over time when the classical receptive field (CRF) was stimulated alone. Stimulation in nCRF significantly increased the magnitude of these temporal changes. Thus, nCRF stimulation not only enhances the overall orientation selectivity, but also enriches the temporal dynamics of cortical neurones, which may increase the computational power of the visual cortex in information processing.
Excitatory and inhibitory intracortical circuits for orientation and direction selectivity

Abstract:The computations performed by a neuron arise from the functional properties of the circuits providing its synaptic inputs. A prime example of these computations is the selectivity of primary visual cortex (V1) for orientation and motion direction. V1 neurons in layer 2/3 (L2/3) receive input mostly from intracortical circuits 1 , which involve excitation 2-9 and inhibition 10-12 . To understand how an L2/3 neuron achieves its selectivity, therefore, one must characterize the functional organization of both its excitatory and inhibitory presynaptic ensembles. Here we establish this organization, and show how it predicts orientation selectivity and reveals a new cortical circuit for direction selectivity. We identified the presynaptic partners of pyramidal neurons in mouse V1 through rabies monosynaptic tracing 1,13 , and imaged the functional properties of the postsynaptic neuron and of its presynaptic ensemble. Excitatory presynaptic neurons were predominantly tuned to the postsynaptic neuron’s preferred orientation. Excitation and inhibition described an inverted Mexican hat, with inhibitory presynaptic neurons densest near the postsynaptic neuron and excitatory ones distributed more distally. Excitation and inhibition also differed in laminar origin: inhibitory presynaptic neurons concentrated in L2/3 while excitatory ones dominated in L4. The distribution of excitatory neurons in visual space was coaxial with the postsynaptic neuron’s preferred orientation and lay upstream of the neuron’s preferred direction. Inhibitory presynaptic neurons, instead, clustered more symmetrically around the postsynaptic neuron and favoured locations downstream of its preferred direction. These results demonstrate that L2/3 neurons obtain orientation selectivity from co-tuned neurons in L4 and beyond, and enhance it by contrasting an elongated excitatory input with a concentric inhibitory input. Moreover, L2/3 neurons can obtain direction selectivity through visually offset 14 excitation and inhibition. These circuit motifs resemble those seen in the thalamocortical pathway 15-20 and in direction selective cells in the retina 21,22 , suggesting that they are canonical across brain regions.

Abstract:How do neurons of the visual cortex acquire their acute sensitivity to the orientation of a visual stimulus? The question has preoccupied those who study the cortex since Hubel and Wiesel1 first described orientation selectivity over twenty-five years ago. At the time, they proposed an elegant and enduring model for the origin of orientation selectivity. Fig. 1A, which is adapted from their original paper and which contains the essence of their model, is by now familiar to most students of the visual system and to many others besides. Yet the model, and the central question that it addresses, is still the subject of intense debate. Competing models have arisen in the intervening years, along with diverse experiments that bear on them.

Abstract:Primary visual cortex (V1) provides crucial insights into the selectivity and emergence of specific output features such as orientation tuning. Tuning and selectivity of cortical neurons in mouse visual cortex is not equivocally resolved so far. While many in-vivo experimental studies found inhibitory neurons of all subtypes to be broadly tuned for orientation other studies report inhibitory neurons that are as sharply tuned as excitatory neurons. These diverging findings about the selectivity of excitatory and inhibitory cortical neurons prompted us to ask the following questions: (1) How different or similar is the cortical computation with that in previously described species that relies on map? (2) What is the network mechanism underlying the sharpening of orientation selectivity in the mouse primary visual cortex? Here, we investigate the above questions in a computational framework with a recurrent network composed of Hodgkin-Huxley (HH) point neurons. Our cortical network with random connectivity alone could not account for all the experimental observations, which led us to hypothesize, (a) Orientation dependent connectivity (b) Feedforward afferent specificity to understand orientation selectivity of V1 neurons in mouse. Using population (orientation selectivity index) OSI as a measure of neuronal selectivity to stimulus orientation we test each hypothesis separately and in combination against experimental data. Based on our analysis of orientation selectivity (OS) data we find a good fit of network parameters in a model based on afferent specificity and connectivity that scales with feature similarity. We conclude that this particular model class best supports data sets of orientation selectivity of excitatory and inhibitory neurons in layer 2/3 of primary visual cortex of mouse.

Abstract:A technique is described for the investigation of regional lung function in infants and children using 13N and a gamma camera. Boluses of isotopic gas are inhaled and perfused while the lung fields are scanned. The child is lightly sedated and breathes normally throughout. Regional function is assessed in terms of the distribution of gas and blood, and the balance between ventilation and perfusion is estimated by comparing an index of the ventilation per unit volume of ventilated lung with that of perfused lung. The use of the method in 8 infants and children with different clinical problems is described to show its application. The method is capable of defining the severity and localization of any abnormality and may also be useful in showing normal function in suspect areas.
Effects of visual adaptation on orientation selectivity in cat secondary visual cortex

Abstract:Neuron orientation selectivity, otherwise known as the ability to respond optimally to a preferred orientation, has been extensively described in both primary and secondary visual cortices. This orientation selectivity, conserved through all cortical layers of a given column, is the primary basis for cortical organization and functional network emergence. While this selectivity is programmed and acquired since critical period, it has always been believed that in a mature brain, neurons' inherent functional features could not be changed. However, a plurality of studies has investigated the mature brain plasticity in V1, by changing the cells' orientation selectivity with visual adaptation. Using electrophysiological data in both V1 and V2 areas, this study aims to investigate the effects of adaptation on simultaneously recorded cells in both areas. Visual adaptation had an enhanced effect on V2 units, as they exhibited greater tuning curve shifts and a more pronounced decrease of their OSI. Not only did adaptation have a different effect on V2 neurons, it also elicited a different response depending on the neuron's cortical depth. Indeed, in V2, cells in layers II-III were more affected by visual adaptation, while infragranular layer V units exhibited little to no effect at all.
Differences in spike generation instead of synaptic inputs determine the feature selectivity of two retinal cell types

Abstract:Abstract The output of spiking neurons depends both on their synaptic inputs and on their intrinsic properties. Retinal ganglion cells (RGCs), the spiking projection neurons of the retina, comprise over 40 different types in mice and other mammals, each tuned to different features of visual scenes. The circuits providing synaptic input to different RGC types to drive feature selectivity have been studied extensively, but there has been substantially less research aimed at understanding how the intrinsic properties of RGCs differ and how those differences impact feature selectivity. Here, we introduce an RGC type in the mouse, the Bursty Suppressed-by-Contrast (bSbC) RGC, whose contrast selectivity is shaped by its intrinsic properties. Surprisingly, when we compare the bSbC RGC to the OFF sustained alpha (OFFsA) RGC that receives similar synaptic input, we find that the two RGC types exhibit starkly different responses to an identical stimulus. We identified spike generation as the key intrinsic property behind this functional difference; the bSbC RGC undergoes depolarization block in conditions where the OFFsA RGC maintains a high spike rate. Pharmacological experiments, imaging, and compartment modeling demonstrate that these differences in spike generation are the result of differences in voltage-gated sodium channel conductances. Our results demonstrate that differences in intrinsic properties allow these two RGC types to detect and relay distinct features of an identical visual stimulus to the brain.
Cone bipolar cell synapses generate transient versus sustained signals in parallel ON pathways of the mouse retina

Abstract:Parallel processing is a fundamental organizing principle in the nervous system, and understanding how parallel neural circuits generate distinct outputs from common inputs is a key goal of neuroscience. In the mammalian retina, divergence of cone signals into multiple feed-forward bipolar cell pathways forms the initial basis for parallel retinal circuits dedicated to specific visual functions. Here, we used patch-clamp electrophysiology, electron microscopy and two photon imaging of a fluorescent glutamate sensor to examine how kinetically distinct responses arise in transient versus sustained ON alpha RGCs (ON-T and ON-S RGCs) of the mouse retina. We directly compared the visual response properties of these RGCs with their presynaptic bipolar cell partners, which we identified using 3D electron microscopy reconstruction. Different ON bipolar cell subtypes (type 5i, type 6 and type 7) had indistinguishable light-driven responses whereas extracellular glutamate signals around RGC dendrites and postsynaptic excitatory currents measured in ON-T and ON-S RGCs in response to the identical stimuli used to probe bipolar cells were kinetically distinct. Anatomical examination of the bipolar cell axon terminals presynaptic to ON-T and ON-S RGCs suggests bipolar subtype-specific differences in the size of synaptic ribbon-associated vesicle pools may contribute to transient versus sustained kinetics. Our findings indicate bipolar cell synapses are a primary point of divergence in kinetically distinct visual pathways.

Abstract:Information about motion is encoded by direction-selective retinal ganglion cells (DSGCs). These cells reliably transmit this information across a broad range of light levels, spanning moonlight to sunlight. Previous work indicates that adaptation to low light levels causes heterogeneous changes to the direction tuning of ON–OFF (oo)DSGCs and suggests that superior-preferring ON–OFF DSGCs (s-DSGCs) are biased toward detecting stimuli rather than precisely signaling direction. Using a large-scale multielectrode array, we measured the absolute sensitivity of ooDSGCs and found that s-DSGCs are 10-fold more sensitive to dim flashes of light than other ooDSGCs. We measured their receptive field (RF) sizes and found that s-DSGCs also have larger receptive fields than other ooDSGCs; however, the size difference does not fully explain the sensitivity difference. Using a conditional knock-out of gap junctions and pharmacological manipulations, we demonstrate that GABA-mediated inhibition contributes to the difference in absolute sensitivity and receptive field size at low light levels, while the connexin36-mediated gap junction coupling plays a minor role. We further show that under scotopic conditions, ooDSGCs exhibit only an ON response, but pharmacologically removing GABA-mediated inhibition unmasks an OFF response. These results reveal that GABAergic inhibition controls and differentially modulates the responses of ooDSGCs under scotopic conditions.

Spike desensitisation as a mechanism for high-contrast selectivity in retinal ganglion cells

Abstract:In the vertebrate retina, several dozens of parallel channels relay information about the visual world to the brain. These channels are represented by the different types of retinal ganglion cells (RGCs), whose responses are rendered selective for distinct sets of visual features by various mechanisms. These mechanisms can be roughly grouped into synaptic interactions and cell-intrinsic mechanisms, with the latter including dendritic morphology as well as ion channel complement and distribution. Here, we investigate how strongly ion channel complement can shape RGC output by comparing two mouse RGC types, the well-described ON alpha cell and a little-studied ON cell that is EGFP-labelled in the Igfbp5 mouse line and displays an unusual selectivity for stimuli with high contrast. Using patch-clamp recordings and computational modelling, we show that a higher activation threshold and a pronounced slow inactivation of the voltage-gated Na + channels contribute to the distinct contrast tuning and transient responses in ON Igfbp5 RGCs, respectively. In contrast, such a mechanism could not be observed in ON alpha cells. This study provides an example for the powerful role that the last stage of retinal processing can play in shaping RGC responses.
