Abstract:Two undescribed phenolic derivatives musaones A and B, six undescribed γ-butyrolactone musaolides A-F, together with two undescribed cyclopentanone musaolides G and H were isolated from the solid culture of an endophytic strain Phaeosphaeriopsis musae M. Arzanlou & Crous (Phaeosphaeriaceae). Their structures were unambiguously elucidated by extensive spectral analyses. The absolute configurations of two diastereomers musaolides G and H were assigned by comparing their experimental and calculated ECD data. Moreover, the in vitro α-glucosidase inhibition and cytotoxic activities of isolated compounds (except musaolide D) were evaluated to understand their biological effects, wherein musaone B and musaolide F displayed promising α-glucosidase inhibition with IC50 values of 37.3 ± 0.59 μM and 105.18 ± 2.76 μM respectively, compared to the positive control acarbose (24.99 ± 1.28 μM).
Antimicrobial and alpha-glucosidase inhibitory flavonoid glycosides from the plant Mussaenda recurvata: in vitro and in silico approaches

Abstract:Seven flavonoid glycosides were isolated from the aerial portions of Mussaenda recurvata during a phytochemical analysis. This comprised one novel component, ecurvoside, and six well-studied compounds, namely astragalin, isoquercitrin, nicotiflorin, rutin, hesperidin, and neohesperidin. The chemical structures of compounds were identified using spectroscopic techniques and a comparison with previously published studies. Alpha-glucosidase inhibition testing was carried out on all isolated compounds. The compounds evaluated have IC 50 values between 35.6 and 239.1 g mL −1 , indicating a moderate degree of inhibition. In vitro antimicrobial activities of compounds 1–7 have screened against the bacteria Pseudomonas aeruginosa ( P. aeruginosa ), methicillin-resistant Staphylococcus aureus (MRSA), Streptococcus faecalis ( Strep. faecalis ), and fungi: Candida albicans ( C. albicans ), Trichophyton mentagrophytes ( T. mentagrophytes ), and Microsporum gypseum ( M. gypseum ), where compound 6 showed excellent activity against fungi T. mentagrophytes with an MIC value of 12.5 μM. In accordance with the molecular docking study, ecurvoside ( 1 ) or pose 472 interacted well with the 3TOP enzyme: PDB and the molecular dynamic simulations proved that the complex of ecurvoside and 3TOP has a stable simulation time of 50–100 ns and the significant residual amino acids in 3TOP are relative to interactions more than one time such as Asp 960, Glu 961, Lys 1088, Glu 1095, Arg 1097, Gly 1102, Thr 1103, Gln 1109, Glu 1178: A chain and Glu 1095, Thr 1101, and Asp 1107: B chain. The docking studies of compounds 1–7 to the enzyme 2VF5 explain the general mechanism to inhibit bacteria and proved that compound 6 (pose 370) inhibited stronger than compound 7 (pose 362) and compound 5 (pose 280), and compounds 1 to 4 do not interact well with 2VF5.

N-Containing triterpenoid saponins from Mussaenda densiflora and identification of heinsiagenin A as a potent immunosuppressant

Abstract:Eleven triterpenoid saponins, including five new compounds, which were named densiflorasides A - E (1 - 5), were isolated from aerial parts of Mussaenda densiflora (Rubiaceae). Their structures were elucidated based on spectroscopic and single-crystal X-ray diffraction analyses and chemical methods. All the isolated compounds and the aglycone heinsiagenin A were evaluated for their immunosuppressive and antiosteoclastogenic activities in vitro. Compounds 6 - 8 and heinsiagenin A inhibited osteoclastogenesis, with IC50 values ranging from 8.24 to 17.7 µM. Furthermore, compounds 3, 6 - 8, and heinsiagenin A significantly inhibited T-cell proliferation, with IC50 values ranging from 2.56 to 8.60 µM, and compounds 3 - 5 and 11 inhibited the proliferation of B lymphocytes, with IC50 values ranging from 1.29 to 8.49 µM. Further in vivo experiments indicated that heinsiagenin A could significantly attenuate IMQ-induced psoriasis and DSS-induced colitis in mice.

Abstract:Three unusual sesterterpenoids featuring unprecedented rearranged colquhounane (C25) and tetranorcolquhounane (C21) frameworks, colquhounoids E (1) and F (3) and norcolquhounoid F (2), were isolated from a Lamiaceae medicinal plant Colquhounia coccinea var. mollis. Their structures were elucidated by spectroscopic analysis and quantum chemical calculations. A biomimetic inspired regioselective cyclopropane cleavage was achieved under acidic conditions. The immunosuppressive activities of these new sesterterpenoids were also evaluated.

Abstract:One new compound ( 3f-[(7′ R ,8′ R )-4,4′-dihydroxy-5-methoxy-2,7′-cycloligna-7-en-9-methoxycarbonyl, 9′-carbonyl- O -] - β - D-fructofuranosyl- ( 2 → 1 )- α - D-glucopyranoside) (Moltkiopsin A) (2) was isolated and identified from the extract of aerial parts of the wild Egyptian plant Moltkiopsis ciliata (Frossk.), family Boraginaceae, for the first time, along with two aryldihydronaphthalene lignans 3f→9′:6f→9-[(7′ R ,8′ R )-4,4′- dihydroxy-3,3′,5-trimethoxy-2,7′-cycloligna-7-en-9,9′-dicarbonyl]-6g-acetyl- α -D-gluco pyranosyl-( 1 → 2 ) - β -D-fructofuranoside (Trigonotin A) (1) which was reported for the first time from this plant species and a known compound 3f→9′:6f→9-[(7′ R ,8′ R )-4,4′- dihydroxy-3,3′,5-trimethoxy-2,7′-cycloligna-7-en-9,9′-dicarbonyl]- α -D-gluco pyranosyl - ( 1 → 2 )- β -D- fructofuranoside (Trigonotin C) (3) . These compounds were separated and purified using different chromatographic techniques and their structures were elucidated by extensive 1D ( 1 H and 13 C NMR), and 2D NMR (COSY, HSQC, and HMBC), besides ESI-MS spectral methods. Extracts were screened as antioxidant, antitumor and antibacterial. The different extracts showed moderate to strong antioxidant capacities in DPPH assays. Ethyl acetate, methylene chloride and crude methanol extracts exhibited the most significant free radicals scavenging activity when compared to the standard antioxidant vitamin C. Hexane and butanol fractions showed the highest cytotoxicity against the cancer cell lines HepG2 and MCF-7.

Evaluation of the role of human papillomavirus in oesophageal squamous cell carcinoma in Belgium.

Abstract:OBJECTIVE To evaluate the putative role of human papillomavirus (HPV) in the aetiology of oesophageal squamous cell carcinoma (OSCC) in Belgium. METHODS The frequency of HPV infection was determined using HPV DNA PCRamplification with L1 consensus primers MY09-MY11, able to recognise about 40 different HPV types, on twenty-one formalin-fixed and paraffin-embedded oesophageal squamous cells carcinomas. Nineteen samples of histologically normal epithelium from the surgical margins of the OSCC specimens and five samples from normal oesophagus obtained at autopsy served as negative controls. RESULTS We found only one HPV positive tumour (4.8%) out of the 21 OSCC cases. All the normal epithelium controls remained negative. CONCLUSIONS Our data are in agreement with those previously published, suggesting that HPV infection only plays a minor role in the pathogenesis of oesophageal squamous cells carcinoma in West-European countries.

Abstract:Upper airway obstruction is a common and potentially serious condition in the pediatric age-group. Neonates and young children are particularly susceptible because their immatureairwaysaresmallinsizeandmuchmorecompliant than those of adults. 1 Children with upper airway obstructionusuallypresentwithstridororapnea,buttheycouldalso manifest with chronic problems, such as recurrent lung infections or obstructive sleep apnea, that can lead to growth retardation, chronic respiratory failure, cor pulmonale, and even death.2 Common causes of upper airway obstruction in children include congenital abnormalities, infectious or inflammatory processes, neoplasms, foreign body aspiration, and even trauma. Congenital abnormalities involving the structures of the pharynx and larynx are not uncommon and can cause upper airway obstruction in children. Such abnormalities may present at birth or can manifest after an inciting event. Although the clinical symptoms and physical examination are important in establishing the diagnosis, it is not always easytoclinicallylocalizetheabnormality.Therefore,imaging evaluation is helpful in establishing the diagnosis, defining the anatomic localization, and assessing the extent of the abnormality. Radiographs and fluoroscopy have long been used in the evaluation of the upper airway in pediatric patients, but cross-sectional imaging studies can provide more detailed and accurate demonstration of upper airway abnormalities. 3 The overarching goals of this article are to discuss updated imaging techniques and review characteristic imaging appearance of congenital causes of upper airway obstruction in pediatric population.
Isolation and structural elucidation of novel cholestane glycosides and spirostane saponins from Polygonatum odoratum

Abstract:Much attention has been paid to cholestane-type steroidal glycosides because of their importance from the perspectives of both chemical diversity and significant biological activities. A phytochemical investigation of the rhizomes of Polygonatum odoratum (Liliaceae) resulted in the isolation of three novel cholestane-type steroidal glycosides (1-3) with unique Δ(14,16)-unsaturated D-ring structures as well as two novel spirostane-type steroidal saponins (4 and 5) and three known steroidal glycosides (6-8). Their structures were determined by various spectroscopic methods and chemical reactions. Steroidal saponin 7 showed significant antifungal activity against Candida albicans JCM1542 (MIC 3.1 μg/mL) and Aspergillus fumigatus JCM1738 (MIC 6.3 μg/mL).

Abstract:A new flavanonol, 3-hydroxyerythrisenegalone (1), and four known compounds (2–5) were isolated from the extract of Mundulea sericea leaves. Investigation of the roots of this plant afforded an additional three known compounds (6–8). The structures were elucidated using NMR spectroscopic and mass spectrometric analyses. The absolute configuration of 1 was established using ECD spectroscopy. In an antiplasmodial activity assay, compound 1 showed good activity with an IC50 of 2.0 μM against chloroquine-resistant W2, and 6.6 μM against the chloroquine-sensitive 3D7 strains of Plasmodium falciparum. Some of the compounds were also tested for antileishmanial activity. Dehydrolupinifolinol (2) and sericetin (5) were active against drug-sensitive Leishmania donovani (MHOM/IN/83/AG83) with IC50 values of 9.0 and 5.0 μM, respectively. In a cytotoxicity assay, erythrisenegalone (3) showed significant activity on BEAS-2B (IC50 4.9 μM) and to HePG2 (IC50 10.8 μM) human cell lines. All the other compounds showed low cytotoxicity (IC50 > 30 μM) against human lung adenocarcinoma cells (A549), human liver cancer cells (HepG2), lung/bronchus cells (epithelial virus transformed) (BEAS-2B) and immortal human hepatocytes (LO2)

Abstract:Two novel alkaloidal glucosides derived from the recently discovered antidesmone (1), together with four known megastigmane and three lignan glucosides, two of which had not previously been described, were isolated from 1-butanol extracts of Antidesma membranaceum (Euphorbiaceae). The structural elucidation of (17RS)-17-(β-D-glucopyranosyloxy)antidesmone (2) and (17RS)-8-deoxo-17-(β-D-glucopyranosyloxy)antidesmone (3) is based on 1H, 13C, COSY, NOESY, HMQC and HMBC NMR spectra, together with LC/ESI-CIDMS and CD data. Determination of the absolute configuration at C-17 was accomplished by comparison with 1H NMR spectroscopic data for alk-2-yl β-D-glucopyranosides, an approach that also proved useful for the megastigmane glucosides blumenyl C β-D-glucopyranoside (4), 3-oxo-α-ionyl β-D-glucopyranoside (5), blumenyl B β-D-glucopyranoside (6) and blumenyl A β-D-glucopyranoside (7). The lignan glucosides lyoniresin-4-yl β-D-glucopyranoside (8), 4′-O-methyllyoniresin-4-yl β-D-glucopyranoside (9) and secoisolariciresin-4-yl β-D-glucopyranoside (10), featuring an unusual glucosylation position, were investigated with the aid of 1H and 2D NMR, CD and MS data.

Abstract:Twelve compounds were isolated from Mussaenda saigonensis aerial parts through phytochemical analysis and the genus Mussaenda is the first place where the compounds 4–6 and 11–12 have been found. Based on the ability to inhibit NO production in RAW264.7 cells, compound 2 has demonstrated the strongest anti-inflammatory activity in vitro with an IC 50 of 7.6 μM, as opposed to L-NMMA's IC 50 of 41.3 μM. Compound 12 was found to be the most effective inhibitor of alpha-glucosidase enzyme in vitro , with an IC 50 value of 42.4 μM (compared to 168 μM for acarbose). Compounds 1–12 were evaluated in vitro for antimicrobial activity using the paper dish method. Compound 11 demonstrated strong antifungal activity against M. gypseum with a MIC value of 50 μM. In silico docking for antimicrobial activity, pose 90 or compound 11 docked well to the 2VF5 enzyme, PDB, which explains why compound 11 had the highest activity in vitro . Entry 2 /pose 280 demonstrated excellent anti-inflammatory activity in silico . The stability of the complex between pose 280 and the 4WCU enzyme for anti-inflammatory activity has been assessed using molecular dynamics over a simulation course ranging from 0 to 100 ns. It has been found to be stable from 60 and 100 ns. The Tyr 159 (95%, H-bond via water bridge), Asp 318 (200%, multiple contacts), Met 273 (75%, hydrophobic interaction via water bridge), and Gln 369 (75%, H-bond via water bridge) interacted well within the time range of 0 to 100 ns. It has more hydrophilic or polar pharmacokinetics.

Abstract:Fifteen new structurally unique monoterpenoid carbazole alkaloids, including two pairs of epimers (1/2 and 3/4), three pairs of enantiomers (6a/6b, 7a/7b, and 8a/8b), and five optically pure analogues (5, 9-12), were obtained from a 95% aqueous EtOH extract of Murraya microphylla by a combination of bioassay- and LC-MS-guided fractionation procedures. Their structures were established based on NMR and HRESIMS data interpretation. The absolute configuration of compound 1 was determined via X-ray crystallographic data analysis and for all compounds by comparison of experimental and calculated ECD data. Compounds 1-5 were assigned as five new thujane-carbazole alkaloids, and compounds 6-12 as 10 new menthene-carbazole alkaloids linked through an ether or carbon-carbon bond. Compounds 1-12 promoted insulin secretion in the HIT-T15 cell line, 1.9-3.1-fold higher than the gliclazide control at 100 μM.

Abstract:Phytochemical investigation of Nauclea latifolia Smith. (Rubiaceae), an African ethnomedicinal plant, yielded a new indolo[2,3-a]quinolizidine alkaloid, nauclealatimide (1) , two previously unknown lignans named nauclealignan A (2) and B (3) , as well rhemaneolignan B (4) and spicatolignan A (5) from the hydromethanolic extract obtained after ultrasonication of the stem bark. Their chemical structures were elucidated using IR, NMR (1D and 2D) and HRESIMS spectral evidence as well as comparisons with existing literature data.

Abstract:Two new C23-steroids derivatives, cyclocitrinoic acid A (1) and cyclocitrinoic acid B (2), and a new isocoumarin metabolite, (3R,4S)-6,8-dihydroxy-3,4,5-trimethyl-7-carboxamidelisocoumarin (10), together with 12 known compounds (3–9, 11–15) were isolated from the mangrove-sediment fungus Penicillium sp. SCSIO 41429. The structures of the new compounds were comprehensively characterized by 1D and 2D NMR, HRESIMS and ECD calculation. All isolates were evaluated for pancreatic lipase (PL) inhibitory and antioxidant activities. The biological evaluation results revealed that compounds 2, 14 and 15 displayed weak or moderate inhibition against PL, with IC50 values of 32.77, 5.15 and 2.42 μM, respectively. In addition, compounds 7, 12 and 13 showed radical scavenging activities against DPPH, with IC50 values of 64.70, 48.13, and 75.54 μM, respectively. In addition, molecular docking results indicated that these compounds had potential for PL inhibitory and antioxidant activities, which provided screening candidates for antioxidants and a reduction in obesity.

Abstract:AIM: To study chemical constituents of the aerial parts of Artemisia anomala (Asteraceae). METHODS: The constituents were isolated with silica gel, ODS column chromatography and semi-preparative HPLC, and their structures were elucidated on the basis of physical characteristics and spectral data. RESULTS: Eight compounds were obtained, and their structures were identified as 3 beta-ethoxytanapartholide (1), (4S*, 5S*)-dihydro-5-[( 1R*, 2S*)-2-hydroxy-2-methyl-5-oxo-3-cyclopenten-1-yl]-3-methylene-4-(3-oxobutyl)-2(3H)-furanone (2), ligucyperonol (3), cyperusol C (4), santamarin (5), 1 alpha, 2 alpha, 3 alpha, 4 alpha, 10 alpha -pentahydroxyguaia-11(13)-ene-12, 6 alpha-olide (6), balanophonin (7), methyl 3-(2'-hydroxy-4'-methoxyphenyl) propanoate (8). CONCLUSION: Compound 1 was a new artifact, 4 and 8 were isolated from the genus Artemisia for the first time, and compounds 2-3, 5-7 were isolated from this plant for the first time.
Sulfated phenolic compounds from Limonium caspium: Isolation, structural elucidation, and biological evaluation

Abstract:Three new compounds, (2S,3S)-5-methyldihydromyricetin (1), (2S,3S)-5-methyldihydromyricetin-3'-O-sulfate (2) and β-d-glucopyranoside, 3-methyl, but-3-en-1-yl 4-O-α-l-rhamnopyranosyl (3) have been isolated from the Limonium caspium, together with dihydromyricetin (4), dihydromyricetin-3'-O-sulfate (5), myricetin-3'-O-sulfate (6), 5-methylmyricetin (7), myricetin (8), myricetin-3-O-β-glucoside (9), as well as phloridzin (10), and tyramine (11). Compounds 5 and 6 were isolated for the first time as acids. This is the first report of all these compounds from this plant. Their structures were established by extensive NMR studies ((1)H NMR, (13)C NMR, DEPT, (1)H-(1)H COSY, HSQC, HMBC) as well as HRESIMS. All isolated compounds were evaluated for their antibacterial, antifungal, antimalarial and antileishmanial activities. Compounds 7, 8 and 9 exhibited good antifungal activity against Candida glabrata with IC50 values of 6.79, 15.37 and 8.53μg/mL, respectively. Compound 8 displayed significant antimalarial activity against resistant and sensitive strains of Plasmodium falciparum with IC50 values of 1.82 and 1.51μg/mL, respectively. Compounds 1, 4, 6, 8 and 9 showed excellent activity against Trypanosoma brucei with IC50 values of 6.93, 9.65, 8.52, 7.67 and 6.31μg/mL, respectively. To date, this is the first report on the phytochemical and biological activity of secondary metabolites from L. caspium.
Seco-iridoid glycosides from the Gentiana olivieri Griseb and their bioactivities

Abstract:The ethanol extract of the Gentiana olivieri Griseb plant was subjected to an investigation to ascertain the presence of its iridoid constituents. By means of HPLC and TLC monitoring, a total of thirteen previously unreported seco-iridoid glucosides olivierisecoside A-M, as well as seven known seco-iridoid glycosides and one known iridoid glycoside were isolated. Their structures were elucidated by a comprehensive spectroscopic data analysis and ECD calculations. The absolute configuration of olivierisecoside D was further confirmed through single-crystal X-ray diffraction analysis. All the identified compounds were characterized as aromatic conjugated seco-iridoid glucosides, with olivierisecoside F-I representing a particularly rare subtype known as the morroniside type seco-iridoids. In vitro testing of the isolated compounds revealed their potential anti-inflammatory and hepatoprotective effects. The results showed olivieroside B and 6'-gentisoyl-8-epi-kingiside have good anti-inflammatory activities in LPS induced RAW264.7 cells. Additionally, olivierisecoside M exhibited some improvements in PA-induced L02 and HepG2 cells damage, known compound loganin showed slight hepatoprotective effect in PA-induced HepG2 cells damage.

Abstract:Two new C-21 steroidal glycosides, mucronatosides M(1) and N(2), were isolated from the stems of Stephanotis mucronata, together with one known compound stephanoside M(3). On the basis of chemical evidence and extensive spectroscopic methods, including one-dimensional and two-dimensional NMR, the structures of the two new compounds were identified as 12-O-tigloyl-20-O-N-methylanthraniloyl sarcostin 3-O-beta-D-glucopyranosyl-(1 -> 4)-6-deoxy-3-O-methyl-beta-D-allopyranosyl-(1 -> 4)-beta-D-cymaropyranosyl-(1 -> 4)-beta-D-cymaropyranoside (1), and 12-O-cinnamoyl-20-O-nicotinoyl(20S)-pregn-6-ene-3 beta,5 alpha,8 beta,12 beta,14 beta,17 beta,20-heptanol 3-O-beta-D-glucopyranosyl-(1 -> 4)-6-deoxy-3-O-methyl-beta-D-allopyranosyl-(1 -> 4)-beta-D-cymaropyranosyl-(1 -> 4)-beta-D-cymaropyranoside (2).

Abstract:The phytochemical investigation of the leaves of Inula confertiflora, a medicinal plant endemic to Ethiopia, led to the isolation of 15 terpenoids; 1β‐hydroxy‐α‐costic acid (1), 3α‐hydroxycostic acid (2), isotelekin (3), asperilin (4), carabrone (5), carpesioline (6), graveolide (7), inuviscolide (8), 8‐epi‐inuviscolide (9), 1β,4β‐dihydroxy‐5α(H)‐guaia‐10(14),11(13)‐dien‐8α,12‐olide (10), isoinuviscolide (11), 4β,10β‐dihydroxy‐5α(H)‐1,11(13)‐guaidien‐8α,12‐olide (12), 4β,10β‐dihydroxy‐1β(H)‐5α(H)‐guai‐11(13)‐en‐8α,12‐olide (13), 4β,10α‐dihydroxy‐1β(H)‐5α(H)‐guai‐11(13)‐en‐8α,12‐olide (14), 4β,10α‐dihydroxy‐1α(H)‐5α(H)‐guai‐11(13)‐en‐8α,12‐olide (15). Herein, structural elucidation and full NMR data for compound 1 are presented for the first time. The structures were elucidated using NMR, HRESIMS, and by comparison with literature data. The relative configurations were defined by NOESY correlations and single‐crystal X‐ray crystallography. Herein, crystallography data of 6 and 7 were reported for the first time. The antibacterial efficacy of some of the isolated compounds was evaluated against two commonly dispersed environmental strains of Escherichia coli and Staphylococcus aureus. Compounds 1, 3, 6, 7, and 8 exhibited moderate antibacterial activities against the tested organisms. The chemotaxonomic significance of compounds is discussed.

Abstract:Mangrove-associated fungi are important sources for the discovery of new bioactive natural products. Three new isocoumarins (1–3) and one new pyrone derivative (4) were isolated from the ethyl acetate extract of the fermentation broth of the mangrove endophytic fungus Phomopsis sp. DHS-11. Nuclear magnetic resonance (NMR) spectroscopy (one-dimensional and two-dimensional) and mass spectrometry were used to determine the structures of these new compounds. The absolute configurations for the new isocoumarins 1–3 were determined by comparing their experimental and calculated electronic circular dichroism (ECD) spectra, while the configuration for the new pyrone-derivative 4 was tentatively solved by comparison of its 13C NMR data with reported data. In the biological activity test, compounds 1 and 3 showed cytotoxic activity against HeLa cells with IC50 values of 11.49 ± 1.64 μM and 8.70 ± 0.94 μM, respectively. The initial structure and activity relationship (SAR) analysis revealed that the length of the side chain at C-3 for isocoumarin-type compounds 1–3 could affect the cytotoxicity against HeLa cells. Compound 4 exhibited cytotoxic activities against human hepatoma cells HepG2 with an IC50 value of 34.10 ± 2.92 μM. All compounds have no immunosuppressive activity.
