Abstract:Nucleic acid-based therapy has emerged as a revolutionary methodology for treatment of the diseases related to protein dysfunction; however, lack of systemically applicable synthetic delivery systems limits its current usage in local applications, particularly for DNA-based therapy with regard to the poor bioavailability in the systemic administrations. To overcome this obstacle, we compiled multiple chemistry-based strategies into the manufacture of the gene delivery formulations to pursue improved tolerability of DNA to the enzymatic degradation in the biological milieu and prolonged retention in the systemic circulation. Here, we constructed a distinctive multilayered functional architecture: plasmid DNA (pDNA) was electrostatically complexed with cationic poly(lysine) (polyplex) as the interior pDNA reservoir, which was further cross-linked by redox-responsive disulfide cross-linking to minimize the occurrence of polyplex disassembly through exchange reaction with the biological charged,components. Still, the pDNA reservoir was spatially protected by a sequential thermoresponsive poly(N-isopropylacrylamide) palisade as the intermediate barrier and a biocompatible hydrophilic poly(ethylene glycol) (PEG) shell with the aim of preventing the accessibility of the biological species, particularly the nuclease degradation to the pDNA payload. Subsequent investigations validated the utilities of these strategies in accomplishing prolonged blood retention. In an attempt to apply this method for tumor therapy, ligand cyclic (Arg-Gly-Asp) peptide was attached at the distal end of PEG, validating prompted tumor-targeted delivery and gene expression of the loaded antiangiogenic gene at the targeted tumor cells and accordingly exerting antiangiogenesis of the tumors for abrogation of tumor growth. Together with its excellent safe profile, the proposed formulation suggests potential utility as a practical gene delivery system for treatment of intractable diseases.

Abstract:pH-responsive polymersomes have emerged as promising nanocarriers for antitumor drugs to realize their fast release and action in a weakly acidic microenvironment of tumor cells. Herein, however, we designed a remarkably pH-responsive polymersome self-assembled from amphiphilic benzimidazole-based polyphosphazenes via the incorporation of cholesteryl hemisuccinate (CholHS), a type of cholesteric molecule, into the polymersome bilayers to inhibit the drug release during blood circulation. Actually, unwanted premature drug leakage before arriving at the acidic tumor site has become a serious problem for polymersomes encapsulating water-soluble drugs, especially when the drug loading is at a high level, thus limiting the therapeutic efficacy. In this study, polymersomes displayed high loading capability of doxorubicin hydrochloride as 12.83%. More importantly, CholHS incorporation decreased the membrane permeability of the polymersome and effectively retarded the cargo release under physiological conditions but induced the fast drug-release rate at pH 5.5, demonstrating a more remarkably acid-responsive release behavior when compared to that of the CholHS-free polymersomes. Further in vivo investigations including pharmacokinetic and antitumor activity studies verified the extended circulation time and enhanced antitumor efficacy of the drug-loaded CholHS-incorporated polymersomes.
Novel Polymersomes Based on Amphiphilic Graft Polyphosphazenes and Their Encapsulation of Water-Soluble Anti-Cancer Drug

Abstract:A series of amphiphilic graft polyphosphazenes with hydrophilic weight fraction ranging from 0.40 to 0.55 were synthesized. These copolymers could self-assemble into distinct aggregates in aqueous solutions. Spherical micelles were observed for the copolymer sample with higher hydrophilic weight fraction. However, when the hydrophilic weight fractions decreased to less than 0.50, vesicle-like polymersomes were formed. Doxorubicin hydrochloride (DOX·HCl), a water-soluble anti-cancer drug, was successfully loaded into the aqueous core of polymersome, which was clearly observed by transmission electron microscopy. The in vitro release of DOX·HCl from polymersome carries further confirmed its encapsulation. In addition, the cytotoxicity of DOX against HepG2 cells was significantly enhanced via polymersome delivery. These results suggest that amphiphilic graft polyphosphazenes could be used for the delivery of water-soluble drugs as polymersome vehicles.
Drug -driven self-assembly of pH-sensitive nano-vesicles with high loading capacity and anti-tumor efficacy?

Abstract:The in vivo delivery of nanomedicine is severely hampered by the limited enhanced permeability and retention effect (EPR) in tumors. Aiming at overcoming this limitation and achieving high anti-tumor effect of chemotherapeutics, we specially addressed an available strategy from a viewpoint of increasing the drug loading of nano-carriers. Here, we constructed a novel pH-responsive polymersome based on the drug-driven self-assembly of amphiphilic polyphosphazenes PAP containing the ortho ester group ABD and mPEG(2000). Due to the non-covalent attractive forces between PAP and doxorubicin hydrochloride (DOX center dot HCl), DOX center dot HCl can induce the self-assembly of PAP via embedding itself in the lamella to form vesicles and the subsequent location in the center aqueous chamber of the resultant nano-vesicles, which resulted in the high drug loading content of 35.77 wt%. In addition, with the incorporation of cholesteryl hemisuccinate (CholHS), the premature leakage of DOX center dot HCl was significantly inhibited under physiological conditions. Meanwhile, the pH-sensitive drug release occurred at pH 5.5 by the advantage of the pH-sensitive biodegradation of ABD in PAP. Consequently, this CholHS-incorporated DOX center dot HCl-driven PAP vesicle achieved excellent anti-tumor effect with tumor growth inhibition up to 82.4% in S180 tumor-bearing mice. Taken together, our newly developed drug-driven vesicles may promote the development of efficient drug delivery systems for application in cancer therapy.

Abstract:Breast cancer remains one of the most common cancers for females. Drug delivery based on cancer nanotechnology could improve the performance of some chemotherapeutic medicines already used in clinic. The emergence of polymersomes provided the potential to encapsulate hydrophobic/hydrophilic drugs. By modifying the weight ratio of methoxy-poly (ethylene glycol) (mPEG) chain to ethyl-p-aminobenzoate (EAB) side group, a series of amphiphilic graft polyphosphazenes (PEPs) was prepared. PEP can be tuned from micelles to polymersomes with the decrease of mPEG content via dialysis. Either hydrophilic doxorubicin hydrochloride (DOX·HCl) or hydrophobic doxorubicin base (DOX) could be encapsulated into PEP polymersomes with high payload and high encapsulation efficiency due to the strong intermolecular interaction with PEP. Compared with free DOX·HCl administration, in vivo investigation in growth inhibition of MCF-7 xenograft tumors in nude mice demonstrated that PEP polymersomes could enhance life safety without compromise of therapeutic efficacy, especially DOX·HCl loaded delivery system.
Supramolecular polymeric prodrug micelles for efficient anticancer drug delivery

Abstract:Polymeric prodrugs have attracted great interest in the field of anticancer drug delivery owing to their integrated advantages of prodrugs and nanoparticles. However, the ambiguous chemical composition of polymeric prodrugs is still a major obstacle to their clinical applications. In this study, an amphiphilic poly(ethylene glycol) derivative with multiple thiols was first synthesized on a large scale through the melt co-polycondensation of oligo(ethylene glycol), 1,4-butanediol and mercaptosuccinic acid. Then an adamantane modified doxorubicin (DOX) with a pH-sensitive hydrazone bond and a beta-cyclodextrin derivative with multiple acryloyls were precisely synthesized. Finally, novel supramolecular polymeric prodrug (SPP) micelles with a precise drug molecular structure and drug loading content were facilely prepared using the above three components via simultaneous self-assembly, a thiol-ene "click" reaction and a host-guest supramolecular interaction in water. The resultant pH-sensitive SPP micelles with a stable core-crosslinked structure and strong drug "bonding" show long blood circulation time, effective cellular uptake and fast DOX release in tumor cells, which significantly improved the antitumor efficacy and security compared with free DOX. This work established a promising drug delivery platform for cancer chemotherapy.

Abstract:Aptamers are single-stranded RNA or DNA ligands that can specifically bind to various molecular targets with high affinity. Owing to this unique character, they have become increasingly attractive in the field of drug delivery. In this study, we developed a multifunctional composite micelle (CM) with surface modification of aptamer AS1411 (Ap) for targeted delivery of doxorubicin (DOX) to human breast tumors. This binary mixed system consisting of AS1411 modified Pluronic F127 and beta-cyclodextrin-linked poly(ethylene glycol)-b-polylactide could enhance DOX-loading capacity and increase micelle stability. Cellular uptake of CM-Ap was found to be higher than that of untargeted CM due to the nucleolin-mediated endocytosis effect. In vivo study in MCF-7 tumor-bearing mice demonstrated that the AS1411-functionalized composite micelles showed prolonged circulation time in blood, enhanced accumulation in tumor, improved antitumor activity, and decreased cardiotoxicity. In conclusion, aptamer-conjugated multifunctional composite micelles could be a potential delivery vehicle for cancer therapy.
Polymersomes Via Self-Assembly of Amphiphilic Β-Cyclodextrin-centered Triarm Star Polymers for Enhanced Oral Bioavailability of Water-Soluble Chemotherapeutics.

Abstract:To date, improving oral bioavailability of water-soluble drugs with poor membrane permeability is still challenging. An example of this includes doxorubicin hydrochloride (DOX·HCl), a widely used chemotherapeutic. We therefore developed a novel DOX·HCl-loaded polymersome (Ps-DOX·HCl) self-assembled by amphiphilic β-cyclodextrin-centered triarm star polymer (mPEG(2k)-PLA(3k))3-CD with the considerable drug loading capability. Using Madin-Darby canine kidney (MDCK) cells trans-well models, it was found that the cellular uptake and absorptive transport of DOX·HCl was significantly increased and the efflux was attenuated when delivered through polymersomes than free drugs. This phenomenon was further verified in mechanistic studies, which was attributed to the change in membrane transport pathway from paracellular route (free DOX·HCl) to active transcellular transport (drug-loaded polymersomes). Moreover, in vivo pharmacokinetic studies in mice demonstrated a significant increase in the oral bioavailability of Ps-DOX·HCl compared with free DOX·HCl (7.32-fold), as well as extended half-life (8.22-fold). This resulted in a substantial anticancer efficacy against mouse sarcoma 180 (S180) tumor in vivo. The cardiotoxicity, which is intrinsically induced by DOX·HCl, and toxicity toward gastrointestinal tissues were avoided according to histological studies. These findings indicate that (mPEG(2k)-PLA(3k))3-CD copolymer displays great potential as a vehicle for the effective oral delivery of water-soluble drugs with low permeability.
Enzymatic interlocking aptamer-micelles for enhanced cellular internalization and nucleus-targeted cancer phototherapy

Abstract:Multifunctional micelles that permit both diagnosis and treatment present enormous advantage and potential for precision medicine. However, the inherent complexities and structural instability of these systems often cause unsatisfactory targeting and therapeutic performances. Herein, by ingenious design of a 2,5-bis(2-thienyl)pyrrole (SNS) modifier to covalently link with AS1411 aptamer and lipid segment, a simple strategy is proposed for one-step enzymatic preparation of interlocked aptamer-micelle (IApM) under bio-friendly conditions. The interlocked poly(SNS) skeleton in IApM can not only stabilize the micelle structure but also enhance near-infrared (NIR) absorption ability, thus further enhancing cellular internalization and photothermal therapy. In addition, the multivalent AS1411 aptamers tethered in the hydrophilic shell can simultaneously increase the specific binding affinity of DNA micelles and induce nucleus-targeted accumulation for DNA damage-triggered apoptosis. This DNA micelle achieves "best of both worlds" with enhanced biostability for cellular internalization and improved NIR photothermal conversion efficiency for nucleus-targeted therapy, which provides a promising formulation strategy for precision cancer treatment.
Surface charge-reversible polyelectrolyte complex nanoparticles for hepatoma-targeting delivery of doxorubicin.

Abstract:Polymeric nanoparticles are greatly advancing the field of nanomedicine due to their ability for targeted and controlled drug release. Here, monodisperse pH-responsive surface charge-reversible and hepatoma-targeting polyelectrolyte complex nanoparticles (GC/HA NPs) near 80 nm were prepared via the ionotropic gelification by simple and mild co-precipitation of galactosylated chitosan (GC) and hyaluronate (HA). Their surface charges could transfer from negative in neutral or basic media to positive in acidic media, indicating their potential application for delivering anti-tumor drugs in response to the extracellular pH of the tumor environment. The unique "electrostatic sponge" structure enhanced their drug-loading capacity and encapsulation efficiency towards the anti-cancer agent doxorubicin (DOX), as well as the pH-triggered controlled release. MTT assays and fluorescence microscopy analysis revealed that the DOX-loaded GC/HA NPs possessed the hepatoma-targeting capacity with favorable biocompatibility. These features make them advanced anti-cancer drug nanocarriers for intelligent drug delivery systems.

Abstract:Biocompatibility and targeted therapy are the demand by modern chemotherapy drugs. Meanwhile, the utilizations of biopolymer or environmentally friendly polymer are also the key issues for sustainable development. We have constructed a new drug delivery system with targeted and controlled release capabilities. It has been confirmed that an environmentally friendly polymer (CMC-g-PLA) obtained by grafting polylactic acid on a carboxymethyl cellulose biopolymer has an amphiphilic property and is easy to form micelles, thereby being useful as a hydrophobic drug carrier. In order to have the function of targeted therapy, anti-EpCAM antibody was anchored on the polymer chain of carboxymethyl cellulose by amidation reaction. Doxorubicin and HepG2 cells were used as the model anticancer drug and tumor targeting cells, respectively. The nano scale core-shell structure, critical micelle concentration, drug loading and encapsulation efficiency were studied by TEM, pyrene fluorescent probe method and ultraviolet standard curve method, respectively. Furthermore, the pH induced release function experiments, cytotoxicity experiments and in vivo tumor treatments experiments on mice. The results show that the drug carrier with antibody prepared by us can effectively deliver hydrophobic drugs to specific tumor sites and increase the effect of tumor treatment. This versatile design provides a new solution for future cancer treatments.
Fabrication of a Targeted Drug Delivery System from a Pillar[5]arene‐Based Supramolecular Diblock Copolymeric Amphiphile for Effective Cancer Therapy

Abstract:Effective cancer therapy will profit from the development of sophisticated drug delivery systems with stimuli‐responsive properties that are capable of delivering therapeutic doses to the active sites, while minimizing the accumulation of highly toxic drugs in off‐target sites. Herein, the fabrication of a pillararene‐based amphiphilic supramolecular diblock polymer (P5‐PEG‐Biotin⊃PCL‐C2V) based on the host–guest recognition between a water‐soluble pillar[5]arene and a viologen salt is reported. P5‐PEG‐Biotin⊃PCL‐C2V self‐assembles into polymersomes, which are utilized as drug delivery vehicles for doxorubicin hydrochloride (DOX). Decorated by the biotin groups, these smart nanocarriers deliver the anticancer drug preferentially to biotin receptor over‐expressing cancer cells. After internalization by the cells, the viologen group is reduced into the cationic radical state by the intracellular reductase NAD(P)H, leading to the release of the loaded DOX by the disassembly of the polymersomes. More importantly, DOX‐loaded polymersomes maintain the therapeutic efficacy towards cancerous HeLa cells, while exhibiting relatively low cytotoxicity towards normal HEK293 cells. In vivo studies reveal that the DOX‐loaded supramolecular polymersomes prolong the circulation time in the bloodstream, promote the antitumor efficacy and reduce the systematic toxicity of the drug through flexible and modular supramolecular strategy.

Abstract:Novel amphiphilic copolymer nanoparticles (HPAE-co-PLA-DPPE) composed of hyperbranched poly (amine-ester), polylactide and 1,2-dipalmitoyl-sn-glycero-3-phosphoethanolamine (DPPE) segments were designed and synthesized that provided high encapsulation efficiency. These nanoparticles (NPs) were used to encapsulate an antitumor model drug, doxorubicin (DOX). The resulting NPs exhibited high encapsulation efficiency to DOX under an appropriate condition. In vitro release experiments revealed that the release of DOX from NPs was faster at pH 4.5 than that at pH 7.4 or pH 6.0. Confocal microscopy observation indicated that the DOX-loaded NPs can enter cells and localize in lysosomes that can be released quickly into the cytoplasm. The DOX-loaded NPs showed comparable anticancer efficacy with the free drug both in vivo and in vitro. Cell Membrane-Inspired Polymeric Micelles As Carriers for Drug Delivery

Abstract:In cancer therapy, surface engineering of drug delivery systems plays an essential role in their colloidal stability, biocompatibility and prolonged blood circulation. Inspired by the cell membrane consisting of phospholipids and glycolipids, a zwitterionic phosphorylcholine functionalized chitosan oligosaccharide (PC-CSO) was first synthesized to mimic the hydrophilic head groups of those amphipathic lipids. Then hydrophobic stearic acid (SA) similar to lipid fatty acids was grafted onto PC-CSO to form amphiphilic PC-CSO-SA copolymers. Cell membrane-mimetic micelles with a zwitterionic surface and a hydrophobic SA core were prepared by the self-assembly of PC-CSO-SA copolymers, showing excellent stability under extreme conditions including protein containing media, high salt content or a wide pH range. Doxorubicin (DOX) was successfully entrapped into polymeric micelles through the hydrophobic interaction between DOX and SA segments. After fast internalization by cancer cells, sustained drug release from micelles to the cytoplasm and nucleus was achieved. This result suggests that these biomimetic polymeric micelles may be promising drug delivery systems in cancer therapy.
Photo-crosslinked and esterase-sensitive polymersome for improved antitumor effect of water-soluble chemotherapeutics.

Abstract:Aim: To meet the requirements on both delivery stability during blood circulation and fast intracellular release for water-soluble chemotherapeutics encapsulated in polymersomes. Materials & methods: A shell photo-crossl inked and esterase-sensitive polymersome (C-PEAMP-D) was constructed based on amphiphilic polyphosphazene PEAMP containing 2-aminoethyl methacrylate. Results: The leakage of doxorubicin hydrochloride (DOX.HCl) was significantly inhibited from 42.27 to 26.64% in 10 h and the accelerated intracellular DOX.HCl release occurred responsively to the esterase at high concentration in cancer cells. Consequently, C-PEAMP-D achieved better antitumor efficiency when compared with free DOX.HCl and uncrosslinked PEAMP vesicle. Conclusion: We provided a strategy to conquer drug leakage in systemic circulation and trigger enzyme-sensitive drug release inside cancer cells for improving oncotherapy outcome of water-soluble chemotherapeutics.
Hydrophobicity‐Adaptive Polymers Trigger Fission of Tumor Cell‐Derived Microparticles for Enhanced Anticancer Drug Delivery

Abstract:Tumor cell‐derived microparticles (MPs) can function as anticancer drug delivery carriers. However, short blood circulation time, large size‐induced insufficient tumor accumulation and penetration into tumor parenchyma as well as limited cellular internalization by tumor cells and cancer stem cells (CSCs), and difficult intracellular drug release restrict the anticancer activity of tumor cell‐derived MP‐based drug delivery systems. Here, hydrophobicity‐adaptive polymers based on poly(N‐isopropylacrilamide) are anchored to tumor cell‐derived MPs for enhanced delivery of anticancer drug doxorubicin (DOX). The polymers are hydrophilic in blood to prolong the circulation time of DOX‐loaded MPs (DOX@MPs), while rapidly switch to hydrophobic at the tumor acidic microenvironment. The hydrophobicity of polymers drives the fission of tumor cell‐derived MPs to form small vesicles, facilitating tumor accumulation, deep tumor penetration and efficient internalization of DOX@MPs into tumor cells and CSCs. Subsequently, the hydrophobicity of polymers in acidic lysosomes further promotes DOX release to nuclei for strong cytotoxicity against tumor cells and CSCs. Controllable Construction of Biocompatible Supramolecular Micelles and Vesicles by Water-Soluble Phosphate Pillar[5,6]arenes for Selective Anti-Cancer Drug Delivery

Abstract:The controllable construction of biocompatible supramolecular nanocarriers with different morphologies based on dynamic noncovalent interactions to achieve selective delivery of drugs with different properties remains highly challenging. We herein report controllable construction of two types of supramolecular nanocarriers based on biocompatible water-soluble phosphate-based pillar[5]arene (WP5P) or pillar[6]arene (WP6P) with pyridinium bromide guest G for the selective anticancer drug delivery. Solid supramolecular micelles could be obtained by the amphiphilic host guest inclusion complex formed from WP5P and G, whereas hollow supramolecular vesicles were acquired from WP6P and G. Both of them showed pH- and Zn2+-responsiveness. Furthermore, the resulting solid micelles were able to encapsulate hydrophobic anticancer drug doxorubicin (DOX) to achieve DOX-loaded micelles, while hydrophilic anticancer drug mitoxantrone (MTZ) could be successfully loaded into the hollow vesicles. Additionally, the encapsulated anticancer drugs could be efficiently released at low-pH environment or with the introduction of Zn2+. More importantly, cytotoxicity experiments indicated that these water-soluble phosphate-based pillar[5,6]arenes showed excellent biocompatibility, and the drug-loaded nanoparticles exhibited comparable therapeutic effect for cancer cells as free anticancer drugs and remarkably reduced damage for normal cells as well. Cellular uptake and intracellular localization experiments further confirmed that these two types of nanocarriers, taken up by cancer cells via endocytosis, could lead to efficient drug accumulation in cancer cells. Therefore, this strategy of controllable construction of different types of stimuli-responsive supramolecular nanocarriers based on biocompatible phosphate-based pillar[5,6]arenes have great potential applications in the field of controlled drug delivery.
Polyphosphazene vesicles for co-delivery of doxorubicin and chloroquine with enhanced anticancer efficacy by drug resistance reversal

Abstract:The conventional chemotherapeutic agent doxorubicin hydrochloride (DOX·HCl) is often accompanied by drug resistance which has severely hindered its clinical application. By taking advantage of the self-assembled behavior of amphiphilic polyphosphazenes, we constructed novel polymersomes loading DOX·HCl or desalted DOX with chloroquine phosphate (CQ) as a drug resistance-reversal agent at 1:1 or 2:1 weight ratios via a one-step common dialysis method. The cytotoxicity evaluation of this dual drug-loaded polymersome was performed on DOX-resistant MCF-7/Adr breast cancer and HL60/Adr leukemia cells. Simultaneously, to simulate in vivo cancerous tissue, 3D tumor spheroid was constructed for tissue penetration and anti-cancer effect evaluation. As a result, PEP-DHC-1 containing DOX·HCl and CQ at 1:1 weight ratio exhibited the strongest toxicity. Furthermore, the in vivo tumor inhibition study carried out on a zebrafish xenograft model also validated that PEP-DHC-1 made the outstanding contribution to improve the sensitivity of MCF-7/Adr breast cancer to chemotherapeutics. These findings suggest that this DOX·HCl and CQ co-delivery system based on PEP polymersomes might be promising for drug resistance reversal of cancer therapy and consequently enhanced anti-cancer efficacy.
Combinatorial Targeting Polymeric Micelles for Anti-Tumor Drug Delivery.

Abstract:Polymeric micelles of amphiphilic block copolymers have been studied for decades for their application as a targeting delivery agent of anti-tumor drugs. However, the targeting micelles may cause an immunological response because of the surface distributed ligands. In this work, mixed micelles were developed to improve the specificity of cancer cell uptake under tumor-acidic conditions. This was achieved by the co-assembly of an active targeting amphiphilic polymer, i.e. cyclic (Arg-Gly-Asp-d-Phe-Lys) c(RGDfK) functionalized poly(ethylene oxide)-b-poly(ε-caprolactone) (cRGD-PEO-b-PCL), and a pH sensitive drug conjugate, i.e. benzoic-imine linked PEGylated doxorubicin (PEG-DOX). Because the PEG-DOX is cleavable at the extracellular pH of a solid tumor, the characteristics of the mixed micelles turn from "stealthy" to cancer cell-affinitive due to the detachment of PEG from the micelle surface and hence allow the action of the c(RGDfK) in the cRGD-PEO-b-PCL with the cancer cell membrane. Besides, the mixed micelles exhibited the capacity for encapsulating hydrophobic drugs such as paclitaxel to form a combination formulation. Our results indicate that co-assembly is a facile but efficient strategy to coordinate the characteristics of each individual component and thus provide combinatorial functions to the delivery system.
Copolymer Micelles Function As Ph-Responsive Nanocarriers to Enhance the Cytotoxicity of a HER2 Aptamer in HER2-positive Breast Cancer Cells.

Abstract:Efficient delivery of nucleic acids into target cells is crucial for nucleic acid-based therapies. Various nucleic acid delivery systems have been developed, each with its own advantages and limitations. We previously developed a nanoparticle-based delivery system for small chemical drugs using pH-responsive PEG8-PDPA100-PEG8 polymer micelles as carriers. In this study, we extend the application of these pH-responsive micelle-like nanoparticles (MNPs) to deliver oligonucleotides. We demonstrate that the MNPs efficiently encapsulate and deliver oligonucleotides of different lengths (20-100 nt) into cells. The cargo oligonucleotides are rapidly released at pH 5.0. We prepared MNPs carrying a Texas red-fluorescently labeled anti-human epidermal growth factor receptor 2 (HER2) aptamer (HApt). Compared to free HApt, the HApt-MNPs resulted in significantly better cellular uptake, reduced cell viability, and increased apoptosis in SKBR3 breast cancer cells, which overexpress HER2. Moreover, HApt-MNPs were significantly less cytotoxic to MCF7 breast cancer cells, which express low levels of HER2. After cellular uptake, HApt-MNPs mainly accumulated in lysosomes; inhibition of lysosomal activity using bafilomycin A1 and LysoTracker Red staining confirmed that lysosomal activity and low pH were required for HApt-MNP accumulation and release. Furthermore, HER2 protein expression declined significantly following treatment with HApt-MNPs in SKBR3 cells, indicating that HApt-induced translocation of HER2 to lysosomes exerted a potent cytotoxic effect by altering signaling downstream of HER2. In conclusion, this pH-responsive and lysosome-targeting nanoparticle system can efficiently deliver oligonucleotides to specific target cells and has significant potential for nucleic acid-based cancer therapies.
