Abstract:In 1861, Paul Broca stood up before the Anthropological Society of Paris and announced that the left frontal lobe was the seat of speech. Ever since, Broca's eponymous brain region has served as a primary battleground for one of the central debates in the science of the mind and brain: Is human cognition produced by highly specialized brain regions, each conducting a specific mental process, or instead by more general-purpose brain mechanisms, each broadly engaged in a wide range of cognitive tasks? For Broca's area, the debate focuses on specialization for language versus domain-general functions such as hierarchical structure building (e.g., [1, 2]), aspects of action processing (e.g., [3]), working memory (e.g., [4]), or cognitive control (e.g., [5-7]). Here, using single-subject fMRI, we find that both ideas are right: Broca's area contains two sets of subregions lying side by side, one quite specifically engaged in language processing, surrounded by another that is broadly engaged across a wide variety of tasks and content domains.

Abstract:The work of Paul Broca has been of pivotal importance in the localization of some higher cognitive brain functions. He first reported that lesions to the caudal part of the inferior frontal gyrus were associated with expressive deficits. Although most of his claims are still true today, the emergence of novel techniques as well as the use of comparative analyses prompts modern research for a revision of the role played by Broca's area. Here we review current research showing that the inferior frontal gyrus and the ventral premotor cortex are activated for tasks other than language production. Specifically, a growing number of studies report the involvement of these two regions in language comprehension, action execution and observation, and music execution and listening. Recently, the critical involvement of the same areas in representing abstract hierarchical structures has also been demonstrated. Indeed, language, action, and music share a common syntactic-like structure. We propose that these areas are tuned to detect and represent complex hierarchical dependencies, regardless of modality and use. We speculate that this capacity evolved from motor and premotor functions associated with action execution and understanding, such as those characterizing the mirror-neuron system.

Meta-analyzing left hemisphere language areas: Phonology, semantics, and sentence processing

Abstract:The advent of functional neuroimaging has allowed tremendous advances in our understanding of brain-language relationships, in addition to generating substantial empirical data on this subject in the form of thousands of activation peak coordinates reported in a decade of language studies. We performed a large-scale meta-analysis of this literature, aimed at defining the composition of the phonological, semantic, and sentence processing networks in the frontal, temporal, and inferior parietal regions of the left cerebral hemisphere. For each of these language components, activation peaks issued from relevant component-specific contrasts were submitted to a spatial clustering algorithm, which gathered activation peaks on the basis of their relative distance in the MNI space. From a sample of 730 activation peaks extracted from 129 scientific reports selected among 260, we isolated 30 activation clusters, defining the functional fields constituting three distributed networks of frontal and temporal areas and revealing the functional organization of the left hemisphere for language. The functional role of each activation cluster is discussed based on the nature of the tasks in which it was involved. This meta-analysis sheds light on several contemporary issues, notably on the fine-scale functional architecture of the inferior frontal gyrus for phonological and semantic processing, the evidence for an elementary audio-motor loop involved in both comprehension and production of syllables including the primary auditory areas and the motor mouth area, evidence of areas of overlap between phonological and semantic processing, in particular at the location of the selective human voice area that was the seat of partial overlap of the three language components, the evidence of a cortical area in the pars opercularis of the inferior frontal gyrus dedicated to syntactic processing and in the posterior part of the superior temporal gyrus a region selectively activated by sentence and text processing, and the hypothesis that different working memory perception-actions loops are identifiable for the different language components. These results argue for large-scale architecture networks rather than modular organization of language in the left hemisphere.

Abstract:Language is a key human faculty for communication and interaction that provides invaluable insight into the human mind. Previous work has dissected different linguistic operations, but the large-scale brain networks involved in language processing are still not fully uncovered. Particularly, little is known about the subdomain-specific engagement of brain areas during semantic, syntactic, phonological, and prosodic processing and the role of subcortical and cerebellar areas. Here, we present the largest coordinate-based meta-analysis of language processing including 403 experiments. Overall, language processing primarily engaged bilateral fronto-temporal cortices, with the highest activation likelihood in the left posterior inferior frontal gyrus (IFG). Whereas we could not detect any syntax-specific regions, semantics specifically engaged left posterior temporal areas (left fusiform and occipitotemporal cortex) and the left frontal pole. Phonology showed highest subdomain-specificity in bilateral auditory and left postcentral regions, whereas prosody engaged specifically the right amygdala and the right IFG. Across all subdomains and modalities, we found strong bilateral subcortical and cerebellar contributions. Especially the right cerebellum was engaged during various processes, including speech production, visual, and phonological tasks. Collectively, our results emphasize consistent recruitment and high functional modularity for general language processing in bilateral domain-specific (temporo-frontal) and domain-general (medial frontal/anterior cingulate cortex) regions but also a high specialization of different subareas for different linguistic subdomains. Our findings refine current neurobiological models of language by adding novel insight into the general sensitivity of the language network and subdomain-specific functions of different brain areas and highlighting the role of subcortical and cerebellar regions for different language operations. Abstract:Current views on the neurobiological underpinnings of language are discussed that deviate in a number of ways from the classical Wernicke-Lichtheim-Geschwind model. More areas than Broca's and Wernicke's region are involved in language. Moreover, a division along the axis of language production and language comprehension does not seem to be warranted. Instead, for central aspects of language processing neural infrastructure is shared between production and comprehension. Three different accounts of the role of Broca's area in language are discussed. Arguments are presented in favor of a dynamic network view, in which the functionality of a region is co-determined by the network of regions in which it is embedded at particular moments in time. Finally, core regions of language processing need to interact with other networks (e.g. the attentional networks and the ToM network) to establish full functionality of language and communication.
The Margins of the Language Network in the Brain

Abstract:This review paper summarizes the various brain modules that are involved in speech and language communication in addition to a left-dominant "core" language network that, for the present purpose, has been restricted to elementary formal-linguistic and more or less disembodied functions such as abstract phonology, syntax, and very basic lexical functions. This left-dominant perisylvian language network comprises parts of inferior frontal gyrus, premotor cortex, and upper temporal lobe, and a temporoparietal interface. After introducing this network, first, the various roles of neighboring and functionally connected brain regions are discussed. As a second approach, entire additional networks were considered rather than single regions, mainly motivated by resting-state studies indicating more or less stable connectivity patterns within these networks. Thirdly, some examples are provided for language tasks with functional demands exceeding the operating domain of the core language network. The rationale behind this approach is to present some outline of how the brain produces and perceives language, accounting, first, for a bulk of clinical studies showing typical forms of aphasia in case of left-hemispheric lesions in the core language network and second, for wide-spread activation patterns beyond this network in various experimental studies with language tasks. Roughly, the brain resources that complement the core language system in a task-specific way can be described as a number of brain structures and networks that are related to (1) motor representations, (2) sensory-related representations, (3) non-verbal memory structures, (4) affective/emotional processing, (5) social cognition and theory of mind, (6) meaning in context, and (7) cognitive control. After taking into account all these aspects, first, it seems clear that natural language communication cannot really work without additional systems. Second, it also becomes evident that during language acquisition the core language network has to be built up from outside, that is, from various neuronal activations that are related to sensory input, motor imitation, nursing, pre-linguistic sound communication, and pre-linguistic pragmatics. Furthermore, it might be worth considering that also in cases of aphasia the language network might be restored by being trained from outside.

Abstract:To advance understanding of brain networks involved in language, the effective connectivity between 26 cortical regions implicated in language by a community analysis and 360 cortical regions was measured in 171 humans from the Human Connectome Project, and complemented with functional connectivity and diffusion tractography, all using the HCP multimodal parcellation atlas. A (semantic) network (Group 1) involving inferior cortical regions of the superior temporal sulcus cortex (STS) with the adjacent inferior temporal visual cortex TE1a and temporal pole TG, and the connected parietal PGi region, has effective connectivity with inferior temporal visual cortex (TE) regions; with parietal PFm which also has visual connectivity; with posterior cingulate cortex memory-related regions; with the frontal pole, orbitofrontal cortex, and medial prefrontal cortex; with the dorsolateral prefrontal cortex; and with 44 and 45 for output regions. It is proposed that this system can build in its temporal lobe (STS and TG) and parietal parts (PGi and PGs) semantic representations of objects incorporating especially their visual and reward properties. Another (semantic) network (Group 3) involving superior regions of the superior temporal sulcus cortex and more superior temporal lobe regions including STGa, auditory A5, TPOJ1, the STV and the Peri-Sylvian Language area (PSL) has effective connectivity with auditory areas (A1, A4, A5, Pbelt); with relatively early visual areas involved in motion, e.g., MT and MST, and faces/words (FFC); with somatosensory regions (frontal opercular FOP, insula and parietal PF); with other TPOJ regions; and with the inferior frontal gyrus regions (IFJa and IFSp). It is proposed that this system builds semantic representations specialising in auditory and related facial motion information useful in theory of mind and somatosensory / body image information, with outputs directed not only to regions 44 and 45, but also to premotor 55b and midcingulate premotor cortex. Both semantic networks (Groups 1 and 3) have access to the hippocampal episodic memory system via parahippocampal TF. A third largely frontal network (Group 2) (44, 45, 47l; 55b; the Superior Frontal Language region SFL; and including temporal pole TGv) receives effective connectivity from the two semantic systems, and is implicated in syntax and speech output.

Abstract:Theories of human cognition prominently feature 'Broca's area', which causally contributes to a myriad of mental functions. However, Broca's area is not a monolithic, multipurpose unit – it is structurally and functionally heterogeneous. Some functions engaging (subsets of) this area share neurocognitive resources, whereas others rely on separable circuits. A decade of converging evidence has now illuminated a fundamental distinction between two subregions of Broca's area that likely play computationally distinct roles in cognition: one belongs to the domain-specific 'language network', the other to the domain-general 'multiple-demand (MD) network'. Claims about Broca's area should be (re)cast in terms of these (and other, as yet undetermined) functional components, to establish a cumulative research enterprise where empirical findings can be replicated and theoretical proposals can be meaningfully compared and falsified.

Methods The present study employed the meta‐analytic connectivity modeling procedure to identify and compare domain‐specific (language‐specific) and domain‐general (shared between language and other domains) functional connectivity patterns of three subdivisions within the broadly defined Broca's area: pars opercularis (IFGop), pars triangularis (IFGtri), and pars orbitalis (IFGorb) of the left inferior frontal gyrus. Results The findings revealed a left‐lateralized frontotemporal network for all regions of interest underlying domain‐specific linguistic functions. The domain‐general network, however, spanned frontoparietal regions that overlap with the multiple‐demand network and subcortical regions spanning the thalamus and the basal ganglia. Conclusions The findings suggest that language specificity of Broca's area emerges within a left‐lateralized frontotemporal network, and that domain‐general resources are garnered from frontoparietal and subcortical networks when required by task demands.

Abstract:Broca's region, which in the language-dominant hemisphere of the human brain plays a major role in language production, includes two distinct cytoarchitectonic areas: 44 and 45. The unique connectivity patterns of these two areas have not been well established. In a resting-state functional connectivity study, we tested predictions about these areas from invasive tract-tracing studies of the connectivity of their homologs in the macaque monkey. We demonstrated their distinct connectivity profiles as well as their differences from the caudally adjacent ventral parts of the premotor cortex and the primary motor cortical region that represent the orofacial musculature. Area 45 is strongly connected with the superior temporal sulcus and the cortex on the adjacent superior and middle temporal gyri. In the parietal region, area 45 is connected with the angular gyrus, whereas area 44 is connected with the supramarginal gyrus. The primary motor cortical region in the caudal precentral gyrus is not connected with the posterior parietal region, which lies outside the confines of the postcentral gyrus, whereas the ventrorostral premotor cortical area 6VR, in the most anterior part of the precentral gyrus, has strong connections with the rostral supramarginal gyrus. Thus, area 44, which has stronger connections to the posterior supramarginal gyrus, can be distinguished from both the adjacent area 6VR and area 45. These findings provide a major improvement in understanding the connectivity of the areas in the ventrolateral frontal region that are involved in language production.

Abstract:Wernicke's area is one of the most important language regions and has been widely studied in both basic research and clinical neurology. However, its exact anatomy has been controversial. In this study, we proposed to address the anatomy of Wernicke's area by investigating different connectivity profiles. First, the posterior superior temporal gyrus (STG), traditionally called "Wernicke's area", was parcellated into three component subregions with diffusion MRI. Then, whole-brain anatomical connectivity, resting-state functional connectivity (RSFC) and meta-analytic connectivity modeling (MACM) analyses were used to establish the anatomical, resting-state and task-related coactivation network of each subregion to identify which subregions participated in the language network. In addition, behavioral domain analysis, meta-analyses of semantics, execution speech, and phonology and intraoperative electrical stimulation were used to determine which subregions were involved in language processing. Anatomical connectivity, RSFC and MACM analyses consistently identified that the two anterior subregions in the posterior STG primarily participated in the language network, whereas the most posterior subregion in the temporoparietal junction area primarily participated in the default mode network. Moreover, the behavioral domain analyses, meta-analyses of semantics, execution speech and phonology and intraoperative electrical stimulation mapping also confirmed that only the two anterior subregions were involved in language processing, whereas the most posterior subregion primarily participated in social cognition. Our findings revealed a convergent posterior anatomical border for Wernicke's area and indicated that the brain's functional subregions can be identified on the basis of its specific structural and functional connectivity patterns.

Abstract:Functional MRI is a well-established tool used for pre-surgical planning to help the neurosurgeon have a roadmap of critical functional areas that should be avoided, if possible, during surgery to minimize morbidity for patients with brain tumors (though this also has applications for surgical resection of epileptogenic tissue and vascular lesions). This article reviews the locations of secondary language centers within the brain along with imaging findings to help improve our confidence in our knowledge on language lateralization. Brief overviews of these language centers and their contributions to the language networks will be discussed. These language centers include primary language centers of “Broca’s Area” and “Wernicke’s Area”. However, there are multiple secondary language centers such as the dorsal lateral prefrontal cortex (DLPFC), frontal eye fields, pre- supplemental motor area (pre-SMA), Basal Temporal Language Area (BTLA), along with other areas of activation. Knowing these foci helps to increase self-assurance when discussing the nature of laterality with the neurosurgeon. By knowing secondary language centers for language lateralization, via fMRI, one can feel confident on providing neurosurgeon colleagues with appropriate information on the laterality of language in preparation for surgery.

Resting functional connectivity of language networks: characterization and reproducibility

Abstract:The neural basis of language comprehension and production has been associated with superior temporal (Wernicke's) and inferior frontal (Broca's) cortical areas, respectively. However, recent resting-state functional connectivity (RSFC) and lesion studies have implicated a more extended network in language processing. Using a large RSFC data set from 970 healthy subjects and seed regions in Broca's and Wernicke's, we recapitulate this extended network that includes not only adjoining prefrontal, temporal and parietal regions but also bilateral caudate and left putamen/globus pallidus and subthalamic nucleus. We also show that the language network has predominance of short-range functional connectivity (except posterior Wernicke's area that exhibited predominant long-range connectivity), which is consistent with reliance on local processing. Predominantly, long-range connectivity was left lateralized (except anterior Wernicke's area that exhibited rightward lateralization). The language network also exhibited anti-correlated activity with auditory (only for Wernicke's area) and visual cortices that suggests integrated sequential activity with regions involved with listening or reading words. Assessment of the intra-subject's reproducibility of this network and its characterization in individuals with language dysfunction is required to determine its potential as a biomarker for language disorders.

Abstract:Abstract A model for the organization of language in the adult humans brain is derived from electrical stimulation mapping of several language-related functions: naming, reading, short-term verbal memory, mimicry of orofacial movements, and phoneme identification during neurosurgical operations under local anesthesia. A common peri-Sylvian cortex for motor and language functions is identified in the language dominant hemisphere, including sites common to sequencing of movements and identification of phonemes that may represent an anatomic substrate for the “motor theory of speech perception.” This is surrounded by sites related to short-term verbal memory, with sites specialized for such language functions as naming or syntax at the interface between these motor and memory areas. Language functions are discretely and differentially localized in association cortex, including some differential localization of the same function, naming, in multiple languages. There is substantial individual variability in the exact location of sites related to a particular function, a variability which can be partly related to the patient's sex and overall language ability and which may depend on prior brain injury and, perhaps subtly, on prior experience. A common “specific alerting response” mechanism for motor and language functions is identified in the lateral thalamus of the language–dominant hemisphere, a mechanism that may select the cortical areas appropriate for a particular language function.

Abstract:With the advancement of cognitive neuroscience and neuropsychological research, the field of language neurobiology is at a cross-roads with respect to its framing theories. The central thesis of this article is that the major historical framing model, the Classic "Wernicke-Lichtheim-Geschwind" model, and associated terminology, is no longer adequate for contemporary investigations into the neurobiology of language. We argue that the Classic model (1) is based on an outdated brain anatomy; (2) does not adequately represent the distributed connectivity relevant for language, (3) offers a modular and "language centric" perspective, and (4) focuses on cortical structures, for the most part leaving out subcortical regions and relevant connections. To make our case, we discuss the issue of anatomical specificity with a focus on the contemporary usage of the terms "Broca's and Wernicke's area", including results of a survey that was conducted within the language neurobiology community. We demonstrate that there is no consistent anatomical definition of "Broca's and Wernicke's Areas", and propose to replace these terms with more precise anatomical definitions. We illustrate the distributed nature of the language connectome, which extends far beyond the single-pathway notion of arcuate fasciculus connectivity established in Geschwind's version of the Classic Model. By illustrating the definitional confusion surrounding "Broca's and Wernicke's areas", and by illustrating the difficulty integrating the emerging literature on perisylvian white matter connectivity into this model, we hope to expose the limits of the model, argue for its obsolescence, and suggest a path forward in defining a replacement.

The language skeleton after dissecting meaning: A functional segregation within Broca's Area

Abstract:Broca's area is proposed as a crucial brain area for linguistic computations. Language processing goes beyond word-level processing, also implying the integration of meaningful information (semantics) with the underlying structural skeleton (syntax). There is an on-going debate about the specialisation of the subregions of Broca's area-Brodmann areas (BA) 44 and 45-regarding the latter aspects. Here, we tested if syntactic information is specifically processed in BA 44, whereas BA 45 is mainly recruited for semantic processing. We contrasted conditions with sentence structure against conditions with random order in two fMRI experiments. Besides, in order to disentangle these processes, we systematically removed the amount of semantic information available in the stimuli. This was achieved in Experiment 1 by replacing meaningful words (content words) by pseudowords. Within real word conditions we found broad activation in the left hemisphere, including the inferior frontal gyrus (BA 44/45/47), the anterior temporal lobe and posterior superior temporal gyrus (pSTG) and sulcus (pSTS). For pseudowords we found a similar activation pattern, still involving BA 45. Among the pseudowords in Experiment 1, we kept those word elements that convey meaning like un- in unhappy or -hood in brotherhood (i.e. derivational morphology). In Experiment 2 we tested whether the activation in BA 45 was due to their presence. We therefore further removed derivational morphology, only leaving word elements that determine syntactic structure (i.e. inflectional morphology, e.g. the verb ending -s in he paints). Now, in the absence of all semantic cues, including derivational morphology, only BA 44 was active. Additional analyses showed a selective responsiveness of this area to syntax-relevant cues. These findings confirm BA 44 as a core area for the processing of pure syntactic information. This furthermore suggests that the brain represents structural and meaningful aspects of language separately.

Are language skills related to structural features in Broca's and Wernicke's area?

Abstract:<p>This study used structural magnetic resonance imaging to examine whether specific anatomical features of Broca's and Wernicke's areas are related to language functions in typically developing older subjects with no specific language expertise. Data from 231 subjects from the Zurich LHAB‐study are used for this study. For these subjects, we obtained several psychometric measures from which we calculated performance measures reflecting specific psychological functions (language comprehension, verbal fluency, perceptual speed, visual memory, recognition of regularities, and logical thinking). From the MRI measurements, we calculated the cortical thickness and cortical surface of Broca's and Wernicke's areas. Applying multiple regression analyses, we identified a moderately strong relationship between language comprehension and the brain metrics from Broca's and Wernicke's areas and showed that approximately 10% of the variance in language comprehension performance is explained by the linear combination of all perisylvian brain metrics. The other psychological functions (verbal fluency, perceptual speed, visual memory, recognition of regularities, and logical thinking) are not related to these brain metrics. Subsequent detailed analyses revealed that the cortical thickness of Wernicke's area, in particular, contributed most to this structure‐function relationship. The better performance in the language comprehension tests was related to a thicker cortex in Wernicke's area. Thus, this study demonstrates a structure‐function relationship between the anatomical features of the perisylvian language areas and language comprehension, suggesting that particular anatomical features are associated with better language performance.</p>

Abstract:Pre-surgical language mapping with functional magnetic resonance imaging (fMRI) is routinely conducted to assist the neurosurgeon in preventing damage to brain regions responsible for language. Functional differences exist between the monolingual versus the bilingual brain, whereas clinical fMRI tasks are typically conducted in a single language. The presence of secondary language processing mechanisms is a potential source of error in the inferred language map. From fMRI data of healthy bilingual and monolingual subjects we obtain language maps as functional networks. Our results show a sub-network "core" architecture consisting of the Broca's, pre-supplementary motor, and premotor areas present across all subjects. Wernicke's Area was found to connect to the "core" to a different extent across groups. The $k$ core centrality measure shows "core" areas belong to the maximum core while WA and other fROIs vary across groups. The results may provide a benchmark to preserve equal treatment outcomes for bilingual patients.

Abstract:Although the language-related fiber pathways in the human brain, such as the superior longitudinal fasciculus (SLF) and arcuate fasciculus (AF), are already well-known, understanding more sophisticated cortical regions connected by the fiber tracts is essential to scrutinize the structural connectivity of language circuits. With the regions of interest that were selected based on the Brainnetome atlas, the fiber orientation distribution estimation method for tractography was used to produce further elaborate connectivity information. The results indicated that both fiber bundles had two distinct connections with the prefrontal cortex (PFC). The SLF-II and dorsal AF are mainly connected to the rostrodorsal part of the inferior parietal cortex (IPC) and lateral part of the fusiform gyrus with the inferior frontal junction (IFJ), respectively. In contrast, the SLF-III and ventral AF were primarily linked to the anterior part of the supramarginal gyrus and superior part of the temporal cortex with the inferior frontal cortex, including the Broca's area. Moreover, the IFJ in the PFC, which has rarely been emphasized as a language-related subregion, also had the strongest connectivity with the previously known language-related subregions among the PFC; consequently, we proposed that these specific regions are interconnected via the SLF and AF within the PFC, IPC, and temporal cortex as language-related circuitry.
