Abstract:To establish a method for determining the content of adenosine in fermented Cordyceps powder simply and accurately with RP- HPLC,in this experiment,based on the pre-established chromatogram condition,mobile phase condition was optimized,and the sample preparation of fermented Cordyceps powder was investigated by testing the influence of solvent,time,temperature on the extraction of adenosine and comparing the results of ultrasonic treatment and reflux.When the HPLC method was carried on DiamonsilTM C18 column(250×4.6 mm,5 μm),using acetonitrile - water(9∶91) as a mobile phase with UV detector at 260 nm,adenosine was separated from other substances.The linearity of this method was good(r=1) in the range 0.097～0.582 mg·mL-1 and average recovery was 101.1% with RSD = 2.20%(n=6).The best sample preparation is to treat sample in 50% methanol solution for 60 minutes at room temperature with ultrasonic treatment.The method is time saving and easy to conduct.

Abstract:To developed a method with HPLC to simultaneously determine nucleosides and bases in Cordyceps sp.fermentation preparations.Methods:HPLC was adopted to measure the amount of uracil,cytidine,hy- poxanthine,uridine,adenine,guanosine,adenosine and 3'-deoxyadenosine in Cordyceps sp.fermentation prepara- tions.The working conditions were:column,Kromasil C_8(200 mm×4.6 mm,5μm);solvent A,double distilled wa- ter;solvent B,methanol;linear gradient:methanol-water(2:98)10 min~(0.2min)→(15:85)25 min;flow rate, 1.0 mL·min~(-1);temperature,30℃;detective wavelength,260 nm.Results:The method was successfully applied to analyze nucleosides and bases in Cordyceps sp.fermentation preparations from various origins.The correlation coeffi- cients of the standard calibration curves of eight components were all higher than 0.9988 over the corresponding concentration range,the RSD of the accuracy and reproducibility of the assay were all less than 5.0%;the mean re- coveries of these eight components were 95.8%-102.5%,and components were stable within 8 h.The experiment showed the significant difference in quantity of nucleosides and bases from various oringins as well as the different accumulation regions in the Cordyceps militaris fermentation preparations formed by some of the nucleosides and ba- ses.The results also indicated that under the similar culture conditions,the submerged and solid substrate fermenta- tion preparations of the same Cordyceps sinensis strain produced different amounts of nucleosides and bases.Conclu- sion:The constructed HPLC method is simple,fast and convenient,it is suitable for assay of several nucleosides and bases of Cordyceps sp.fermentation preparations.Thus,it is valuable for evaluating the quality of productions com- prehensively.

Abstract:In this study, a series of choline chloride-based natural deep eutectic solvents (NADESs) was successfully fabricated and combined with ultra-high performance liquid chromatography-tandem mass spectroscopy (UHPLC-MS/MS) to detect adenosine and cordycepin in Cordyceps militaris . The analysis validation produced precise results with outstanding parameters for analysing extraction samples. In particular, NADESs with a choline chloride to citric acid ratio of 1:2 exhibited exceptional performance under ultrasound-assisted conditions. Using response surface methodology, we optimised the extraction process, obtaining high concentrations of adenosine (2.67 ± 0.05 mg/g) and cordycepin (3.73 ± 0.11 mg/g) from C. militaris . Compared to conventional methods, the NADES-based extraction proved more efficient, delivering elevated levels of target compounds by 36.5-fold for adenosine and 1.5-fold for cordycepin. Additionally, a mechanistic insight was offered by analysing post-extraction samples. The analytical approach was utilised to assess the adenosine and cordycepin content in various C. militaris samples available in the Vietnamese market. The detected values ranged from 1.73 ± 0.07 to 2.78 ± 0.11 mg/g for adenosine and from 2.87 ± 0.04 to 4.52 ± 0.14 mg/g for cordycepin. This analytical method holds promise for accurately evaluating adenosine and cordycepin in C. militaris .

Abstract:Objective To establish an HPLC method for determining the nucleosides of adenosine in Hericium erinaceus. Methods The suitability, accuracy, repeatability, linearity, linear range and durability were studied by HPLC. The main factors affecting sample solution were observed,including extraction method,extraction solvent,ex-traction time and solid-liquid ratio. Results The suitability,accuracy,repeatability,linearity,linear range durability of the method all met the relative requirements. Conclusion This method can be applied as the quantity determination of adenosine in Hericium erinaceus with good accuracy and reliability.

Abstract:An eco-friendly and fast HPLC method was developed for the determination of adenosine, inosine, guanosine and uridine in Cordyceps and related products (fermented mycelia of Hirsutella sinensis andPaecilomyces hepiali). The sample was ultrasonically extracted using 0.5% phosphoric acid solutions for 2.5 min. Sample separation was performed on a Poroshell SB-Aq column (50 mm × 4.6 mm, 2.7 μm) using eco-friendly mobile phase consisting of formic acid and ammonium formate aqueous solution at a flow rate of 1.0 mL·min<sup>-1</sup>. The detection wavelength was 260 nm. The developed HPLC method showed good linearity with correlation coefficients of 1.0000 in the test range. Good precision, repeatability and stability of this method were also observed (RSD ≤ 2.81%). The recovery ranged from 91.84%-105.19% (RSD ≤ 2.59%). Compared with reported methods, the current method did not use harmful organic solvent and took only 10.5 min. It obtained a high eco-score of 91 by the "Analytical Eco-Scale" tool. The developed method is eco-friendly and fast, which is suitable for the quality evaluation of Cordyceps and related products.

Abstract:Nucleosides and nucleobases are one of the most important indicators of quality control. A sensitive and reliable high performance liquid chromatography-ultraviolet method was applied to analyse 13 nucleosides and nucleobases simultaneously in 15 batches of nine Ophiocordyceps species and its allies in China. Principal component analysis (PCA) and cluster analysis were conducted by SPSS 22.0 software (IBM Corp., Armonk, NY, USA). The 15 samples of Cordyceps were differentiated successfully based on their nucleoside and nucleobase content. Total nucleosides content in mycelium was significantly higher than that in the natural fruiting bodies of Ophiocordyceps sinensis (NFOS). Five nucleosides or nucleobases - adenine (A), guanosine (Gu), uracil (U), uridine (Ur) and guanine (G) - were the major components contributed to the total variance according to PCA. The profiles of the 13 tested nucleosides and nucleobases (including adenosine, cytidine, guanosine, inosine, thymidine, uridine, cordycepin, adenine, cytosine, guanine, thymine, uracil and hypoxanthine) can discriminate different samples and can be candidate indicators applied for the quality control of Ophiocordyceps and its allies.

Abstract:To establish a quantitative analysis method for analyzing the nucleosides in Cordyceps sinensis with capillary electrophoresis, and compare the difference between natural and the cultured C. mycelia.Capillary zone electrophoresis method was employed to quantitate the adenosine, uridine, guanosine and inosine in C. sinensis, with 0.25 mg x L(-1) boric acid-sodium hydroxide buffer, pH 9.5. The working voltage was 20 kV, the temperature was 25 degrees C, and the detection wavelength was 260 nm.With the capillary zone electrophoresis method, the average recovery of the above 4 nucleosides was 98.9%, 95.1%, 97.8% and 98.8% respectively, with the RSD 0.4%, 1.7%, 1.3% and 5.0%. There was no adenosine in natural C. sinensis and no inosine in the cultured C. mycelia detected.This method can be used to determine the adenosine, uridine, guanosine and inosine in C. sinensis. The nucleosides in C. sinensis produced from Qinghai province and cultured C. mycelia are obviously different.

Abstract:A high-performance liquid chromatography-diode array detector-mass spectrometry (HPLC-DAD-MS) analytical method was developed for detection of the nucleosides and nucleobases in two species of Lingzhi, the dried sporophore of Ganoderma lucidum and G. sinense. The method, combining advantages of both DAD and MS, was successfully used to qualitatively identify for six nucleosides namely, adenosine, cytidine, guanosine, inosine, thymidine, uridine and five nucleobases namely, adenine, guanine, hypoxanthine, thymine and uracil in Lingzhi samples. Quantitative analyses showed that uridine was the most abundant nucleoside in these Lingzhi samples and the contents of nine target analytes were found to be different in pileus and stipes of the fruiting bodies and among the different species of G. spp. The established method might apply as an alternative approach for the quality assessment of Lingzhi.

Abstract:A speedy and selective ultra-HPLC-MS/MS method for simultaneous determination of deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-ADON), 15-ADON. nivalenol and fusarenon X in traditional Chinese medicines (TCMs) was developed. The method was based on one-step sample cleanup using reliable homemade cleanup cartridges. A linear gradient mobile-phase system, consisting of water containing 0.2% aqueous ammonia and acetonitrile/methanol (90:10, v/v) at a flow rate of 0.4 mL/min, and an Acquity UPLC HSS T3 column (100 mm x 2.1 mm, 1.8 mu m) were employed to obtain the best resolution of the target analytes. [C-13(15)]-DON was used as the internal standard to accomplish as accurate as possible quantitation. The established method was further validated by determining the linearity (R-2 >= 0.9990), sensitivity (LOQ, 0.29-0.99 mu g/kg), recovery (88.5-119.5%) and precision (RSD <= 15.8%). It was shown to be a suitable method for simultaneous determination of DON, 3-ADON, 15-ADON, nivalenol and fusarenon X in various TCM matrices. The utility and practical impact of the method was demonstrated using different TCM samples.

Abstract:Cordycepin from Cordyceps possesses excellent pharmacological properties, including anti‐inflammation and anti‐tumor effects, therefore representing a potential alternative medicine. However, doubts about the pharmacokinetic results of cordycepin had been raised in previous study due to its rapid deamination. The organic solvent methanol was immediately added to terminate the degradation of cordycepin in anticoagulated blood samples and enable the accurate evaluation of pharmacokinetics in vivo. A sensitive and selective ultra‐high‐performance liquid chromatography coupled with Q Exactive hybrid quadrupole orbitrap high‐resolution accurate mass spectrometry method was developed and validated to simultaneously determine cordycepin and its deamination metabolite 3′‐deoxyinosine using 2‐chloroadenosine as internal standard in rat whole blood. The calibration curves of cordycepin and 3′‐deoxyinosine showed excellent linearity within the concentration range of 1.05–10000.00 ng/mL with acceptable accuracy, precision, selectivity, recovery, matrix effect, and stability. This method was successfully applied to the pharmacokinetic study of cordycepin and its metabolite in rat blood. The effect of the adenosine deaminase inhibitor erythro‐9‐(2‐hydroxy‐3‐nonyl) adenine hydrochloride on the pharmacokinetics of cordycepin was investigated. In summary, the reliable pharmacokinetic parameters of cordycepin and its deamination metabolite 3′‐deoxyinosine in rat blood were successfully elucidated. Abstract:An HPLC method was established to detect the adenosine content in fermentation liquid and ethanol sediments of polysaccharide during the production process of Antrodia camphorata capsules.The reverse-phase bonded Agilent Eclipse XDB-C18(4.6 mm × 150 mm,5 μm) was selected,and the mobile phase consisted of phosphate acid buffer solution(pH 6.47) and methanol at the volume rate of(90∶ 10) with the flow rate as 1.0 ml/min.The detective wavelength was 260 nm,and the column temperature was kept as the room temperature.The results showed that the linear equation for adenosine detection was Y=29160X-4972 with the linear range of 1.17~37.5 mg/L and r as 0.9999.The average recovery rate was 102.8% and the RSD was 1.29%.In conclusion,the method was simple,sensitive and accurate,so it could be used to determine the adenosine content in fermentation solution and ethanol sediments during production process of Antrodia camphorate capsules.

Abstract:OBJECTIVE To select the optimal method of extracting mannitol fromCordyceps sinensis(Berk.) Sacc.for determi-ning the content of mannitol in the herb using colourimetric method.METHODS Ultrasonic extraction following centrifugation wasused for extracting the active compound from the herb.RESULTS The linear range of mannitol was 20 ~90μg/mL and the recoverywas 102.6% with RSD 1.34%(n=5).Variation coefficient was 0.44% and the sample solution was stable in 24 h.The proposedmethod was applied to determine the content of mannitol in 60 batches of fermentedCordyceps sinensis(Berk.) Sacc.produced in theyears of 1999,2000 and 2001.The average concentration of mannitol in the fermented herb produced in 2001 was about 2% higherthan those in 1999,2000 and with less remarkable variation,the content of mannitol in 60 batches of the herb produced in the 3 yearswas found to fluctuate at10%.CONCLUSION The proposed method is effective and convenient for the analysis of mannitol in qual-ity control ofCordyceps sinensis(Berk.) Sacc..
Determination of Three Nucleosides in Radix Isatidis by HPLC

Abstract:AIM:To establish a method for determining guanosine, uridine,and andadenosine for Radix Isatidis. METHODS: The Prevail C_(18) analytical column(4.6 mm×250 mm,5 μm)was used.The mobile phases consisted of water and acetonitrile with gradient.The flow rate was 0.5 mL/min and column temperature was at 25 ℃.The detection wavelength was at 254 nm. RESULTS: The method had good linear relationship within the range(1.92)-38.4 μg/mL;1.75-35 μg/mL; 2.12-42.4 μg/mL of guanosine,uridine and andadenosine,respectively,with the correlation coefficients of 0.999 9.The average recoveries of the three nucleosides were 99.64%(RSD=0.38%),98.94%(RSD=0.32%) and 98.85%(RSD=0.48%),correspondingly.CONCLUSION: The method is fast,rapid,convenient,accurate and can be used for quality control of Radix isatidis.

Abstract:The present paper describes the development of a micellar electrokinetic chromatographic method for the determination of nucleoside (adenosine, uridine) and base (uracil) markers in aqueous extracts of Ganoderma medicinal preparations. The markers were successfully separated within 10 min using an 80 mM borate buffer, with 25 mM sodium dodecyl sulfate adjusted to pH 9.0, an operating voltage of 22 kV, temperature of 20 degrees C and a hydrodynamic injection time of 5 s. Separations were carried out in a fused-silica capillary with peak detection by direct UV at 254 nm. Following semi-validation of the method, with each analyte showing a good linear relationship over a 0.2 to 20 ppm concentration range (correlation coefficients from 0.9986 to 0.9998), the amounts of the three markers in the various forms of Ganoderma were easily determined using a relatively simple extraction procedure.

Abstract:Nucleotides, nucleosides and nucleobases play a greater role in the physiological activity of organisms which are highly present in royal jelly (RJ). The objective of the present study is to develop a HPLC method to simultaneous determine nucleotides, nucleosides and nucleobases in RJ and access them in fresh and commercial RJ samples. The LOD and LOQ were 12.2-99.6 mu g/L and 40.8-289.4 mu g/L, respectively with nearly 100.9% recoveries. Except uric acid, all other compounds were found in RJ samples. Significant difference in the average content of compounds in fresh (2682.93 mg/kg) and commercial samples (3152.78 mg/kg) were observed. AMP, adenosine and adenine were found predominant in all the samples. Significant higher levels of ATP, ADP and AMP was seen in fresh RJ samples, and IMP, uridine, guanosine, and thymidine was seen in commercial RJ samples. The investigated compounds can be used as indexes for assessment RJ freshness and quality. (C) 2014 Elsevier Ltd. All rights reserved.

Abstract:Cordycepin exhibits various bio-activities, including anticancer, antibacterial, antiviral and immune regulation activities, and is a significant focus of research. However, the preparation of high-purity cordycepin remains challenging. Also, the molecular target with which cordycepin interacts to cause an antibacterial effect remains unknown. In the present study, cordycepin was prepared by preparative high-performance liquid chromatography (prep-HPLC) and the purity obtained was 99.6%, indicating that this technique may be useful for the large-scale isolation of cordycepin in the future. The results of computational molecular docking analysis indicated that the interaction energy between cordycepin and NAD+-dependent DNA ligase (LigA) was lower than that between cordycepin and other common antibacterial targets. The highly pure cordycepin obtained by prep-HPLC demonstrated inhibitory activity against LigA from various bacteria in vitro. In conclusion, cordycepin may be useful as a broad-spectrum antibiotic targeting LigA in various bacteria.

Abstract:Present paper describes the application of a recently developed method for the determination of six nucleobases (thymine, uracil, adenine, cytosine, guanine and guanosine), by high performance thin layer chromatography (HPTLC) in Ganoderma lucidum (GL) and Cordyceps sinensis (CS) - two mushrooms that have been widely revered in traditional Chinese medicines. The method used a simple mobile phase constituted of dicholoromethane-methanol-formic acid (8:2.25:0.8, v/v/v). Validation of the method according to USP guidelines proved the method to be accurate (97.1–99.9%) and precise (1.8–5.2% RSD). Results confirmed the abundance of nucleobases in various extracts of GL and CS wherein the aqueous extracts of GL and ethanolic extracts of CS were found to have the maximum nucleobase content. Identification of nucleobases in the samples was reconfirmed by hyphenated HPTLC-MS technique. HPTLC data were further processed by chemometric tools like heat maps, cluster dendrograms and principal component analysis for drawing relatability patterns among the samples. Since both GL and CS are well known for their therapeutic applications, nucleobase profiling in these medicinal mushrooms will provide an essential lead in establishing their use in the development of drug formulations.

Methods After liquid- liquid extraction,the samples of serum use acetonitrile- water- formic acid( 75 ∶25 ∶0. 1,V / V) as mobile phase,and use Nimodipine as the internal standard,with the method ZORBAX Extend C18column( 2. 1 mm × 100 mm,3. 5 μm) to separation. By HPLC / MS / MS,use SIM mode to test. Results When the concentration of noradreneline( NE),dopamine( DA) and 5- hydroxytryptamine( 5- HT) of the serum samples were 0. 5 ng/ml- 400 ng/ml,1 ng/ml- 100 ng/ml and1 ng / ml- 100 ng / ml,the linear relationship was good( r 0. 99). The minimum quantitative limit of NE,DA and 5- HT were 0. 5 ng / ml,1 ng / ml and 1 ng / ml and RSD was respectively 11. 5%,6. 1% and 9. 3%( n = 5). The average recovery rates were 85. 07%,89. 2% and 90. 67%. The matrix effect,within day and between days precision and accuracy rating meet the requirement of the determination of biological samples. Conclusion This method is simple,accurate,high sensitivity,and is suitable for the determination and research of neurotransmitters in serum.

Abstract:Adenine nucleotides (AN) are ubiquitous metabolites that regulate cellular energy metabolism and modulate cell communication and inflammation. To understand how disturbances in AN balance arise and affect cellular function, robust quantification techniques for these metabolites are crucial. However, due to their hydrophilicity, simultaneous quantification of AN across various biological samples has been challenging. Here we present a hydrophilic interaction high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) based method for the quantification of 26 adenosine nucleotides and precursors as well as metabolic products of nicotinamide adenine dinucleotide (NAD) in plasma, liver, and adipose tissue samples as well as cell culture supernatants and cells. Method validation was performed with regard to linearity, accuracy, precision, matrix effects, and carryover. Finally, analysis of cell culture supernatants derived from intestinal organoids and RAW 264.7 cells illustrates that the here described method is a reliable and easy-to-use tool to quantify AN and opens up new avenues to understand the role of AN generation and breakdown for cellular functions.
