Abstract:As one of the significant reactive nitrogen species in living systems, nitroxyl (HNO) plays a paramount role in a series of physiological and pathological processes. However, completely elucidating the role of endogenous HNO in living systems remains a huge challenge due to the lack of effective detection tools for the real-time monitoring of HNO. Recently, fluorescent probe has been widely accepted as a powerful molecular tool for the non-invasive detection of active species in living systems. Hence, in this work, we developed a new NIR fluorescent probe (N-HBT-HNO) with ultra-large Stokes shift for the effective detection and bioimaging of HNO in living cells and mouse. In the design blueprint of probe, the dicyanoisophorone moiety was integrated into the benzothiazole unit to expand the emission wavelength to NIR region of 728 nm and remarkably increase the Stokes shift to ultra large of 303 nm, which could endow the probe with superiority in bioimaging. Additionally, the 2-(diphenylphosphino) benzoate served as the efficient recognition group and fluorescence quenching group. Results demonstrated that the newly developed probe possessed appreciable sensing performance, including good selectivity, large signal to noise ratio, high sensitivity and fast response toward HNO. Subsequently, this probe was successfully applied for the bioimaging of exogenous HNO in living cells and mouse, reflecting the application potential of exogenous HNO (AS)

Abstract:Nitroxyl (HNO), generated from nitric oxide (NO) in biological system, plays distinct biological role in physiological process, but the role of endogenous HNO in biosystems still remains unclear, because of lacking of specific and sensitive detector to monitor HNO in real time. The conventional methods indicate the level of HNO through detecting the ultimate product nitrous oxide (N2O) of HNO dimerization and dehydration, which could easily result in inaccurate data. Therefore, developing an assay that can directly apply to detect HNO is of significance for understanding the function mechanism of HNO in biological system. In this study, we describe a near-infrared fluorescent probe 4-(2-(4-(dicyanomethylene)-4H-chromen-2-yl)vinyl) phenyl-2-(diphenylphosphanyl)-benzoate (CPN) that employs (2-(4-hydroxystyryl)-4H-chromen-4-ylidene) malononitrile (CP-OH) as a fluorophore and 2-(diphenylphosphino) benzoate as a recognition group. This probe, CPN, shows rapid-response to HNO which compared to other current probes. The fluorescent intensity reaches plateau in 20 min in vitro and in 2 min in vivo, respectively, which is important to monitor dynamic and transient state of HNO in biological system. In addition, CPN probe with a large Stokes' shift (150 nm) and near-infrared emission (700 nm) that is suitable for biological imaging. This probe has a potential of elucidating the biological function of HNO in biosystem.

Abstract:The unique physiological activity and pharmacological properties of nitroxyl (HNO) have led to its use as a drug for treating various diseases, resulting in increased research on its detection and quantification. Although there have been numerous studies on fluorescent probes for detecting HNO, there is a lack of near-infrared (NIR) fluorescent probes that exhibit both a high fluorescence quantum yield and a large Stokes shift for this purpose. Through the optimization and modification of the structure of fluorinated NIR Rhodol, five NIR fluorescent dyes were synthesized, and the most promising candidate with the highest fluorescence quantum yield (Φ f = 11.4 % in absolute PBS solution) and a bioapplicable Stokes shift (134 nm) was identified. Subsequently, a novel NIR fluorescent probe NB-HNO was developed. This probe exhibits high sensitivity and selectivity for HNO and has been successfully used to monitor fluctuations in exogenous and endogenous HNO levels in HepG2 cells and mice. Overall, our findings suggest that NB-HNO is a promising probe for imaging HNO and that our NIR fluorophore with high fluorescence quantum yield and large Stokes shift possesses potentially desirable properties for in vivo bioimaging.

Abstract:Nitroxyl (HNO) plays an important role in various physiological activities. It has the potential to be used as a treatment for certain diseases such as alcohol poisoning, acute hypertension, and atherosclerosis. However, traditional methods for detecting HNO are challenging due to its rapid polymerization and elimination into N 2 O. Therefore, it is crucial to establish direct and effective HNO detection methods to comprehend these physiological processes better. In this study, a new near-infrared fluorescent probe called HXM-P based on the intramolecular charge transfer (ICT) mechanism was designed and synthesized. This probe employs 2-((6-hydroxy-2,3dihydro-1 H-xanthen-4-yl)methylene)malononitrile as a fluorophore and 2-(diphenylphosphine) benzoate as a recognition group. The results showed that probe HXM-P can detect HNO with high sensitivity (1.07 × 10 − 8 M). A good linear correlation was observed between the fluorescence intensities at 640 nm and the concentrations of HNO in the range of 0–80 μ M (R 2 = 0.997). Moreover, probe HXM-P exhibited a rapid response rate (within 15 s) toward HNO, and the fluorescent intensity reached a plateau within 5 min, making it easier to track the highly reactive and short-lived HNO in living systems. Additionally, HXM-P was successfully employed for imaging HNO in HepG2 cells.

Abstract:Nitroxyl (HNO) is one of the important derivatives of nitric oxide (NO). It was intertwined with various biological and pharmacological events as a well-defined active molecule. Developing fluorescent probes for high specific and in situ trapping of HNO in living samples is still challenging. In this project, we constructed a near-infrared (NIR) metal-free fluorescent probe (DCM-P) for monitoring HNO in vitro and in vivo. The novel probe, DCM-P, contains a dicyanomethylene-4H-pyran (DCM) fluorophore as the reporter and a triarylphosphine as the recognition moiety. Upon exposure to HNO, the probe emits fluorescence at the wavelength of 688 nm, which belongs to near-infrared region and endows great beneficial for imaging in vivo. Moreover, DCM-P shows high chemoselective detection and fast response to HNO in the presence of various biological relevant reductants, and is successfully employed to trap nitroxyl in different living cells and zebrafish models. These results suggest that DCM-P is a suitable near-infrared metal-free fluorescent probe for motoring nitroxyl in living samples.

Abstract:Reactive nitrogen species (RNS) as a class of vital reactive molecules, are essential for maintaining homeostasis. Among RNS, nitric oxide (NO or NO), peroxynitrite anion (ONOO – ) and nitroxyl (HNO) play crucial roles in several physiological, pathological and pharmacological processes. However, abnormal variations of these RNS concentrations are closely intertwined with the occurrence and progression of numerous diseases. Hence, the intuitive and accurate visualization of RNS is extremely significant for deeper insight into their spatial distribution, physiological and pathological roles, functions, internal interactions and potential mechanisms in biological systems. To date, the fluorescence imaging technique has been demonstrated to a powerful strategy for the rapid and precise detection of reactive species in biological systems. Moreover, small molecule fluorescent probes have been successively fabricated for in suit , real-time, sensitively and selectively monitoring the changes of RNS in living systems. Based on the inherent characteristics of three RNS, abundant fluorescent probes have been elaborately constructed to satisfy various requirements. Therefore, in this review, we briefly summarize the recent advances in small molecule fluorescent probes for monitoring of NO, ONOO – and HNO within 8 years. Moreover, the classifications, design strategies, recognition mechanisms and biological applications of three RNS-responsive fluorescence probes are discussed in depth. Particularly, the challenges and further development directions in this hot field of fluorescent imaging are definitely emphasized.

Abstract:Nitroxyl (HNO) has been discovered to play a vital role in diverse physiological and pathological processes. In this work, we report a new reductant-resistant fluorescent probe (NHMC-P) for highly effective tracking of HNO in vitro and in vivo in the near-infrared regions. NHMC-P contains a nitrogen-hydroxylmerocyanine (NHMC) as the fluorophore, linked with a 2-(diphenylphosphino)benzoic acid through an ester bond as the recognition moiety. Upon reaction with HNO, the recognition moiety will be triggered to an aza-ylide, which then attacks the intramolecular ester carbonyl group to release the fluorophore NHMC and restore the fluorescence. NHMC-P exhibited ultra-sensitivity and high selectivity for HNO in the near-infrared region. Furthermore, ascribed to the excellent water-solubility, the probe was successfully employed to track HNO in living cancer cells and zebrafish models. Our work provides a potential tool for in vivo HNO tracking and is helpful for HNO-related pathological studies in living organisms.

Abstract:Development of selective fluorescent probes for rapid and ultrasensitive detection of nitroxyl (HNO) is of great importance for biomedical researchers to investigate the detailed functions and mechanisms of HNO in living systems. Herein, based on an internal charge transfer (ICT) mechanism, we developed a novel ratiometric, colorimetric and far-red fluorescent probe (HNO-TCF) for the rapid and ultrasensitive detection of HNO in living cells. HNO-TCF exhibits high HNO-selectivity even in the presence of a high concentration of biological reductants including glutathione (GSH), hydrogen sulfide (H2S) and ascorbate (AA), which might be ascribed to the adoption of the 2-(diphenylphosphino)benzoate recognition moiety. The ICT-based fluorescent probe HNO-TCF displays a large (185 nm) red-shifted absorption spectrum and the color changes from yellow to blue upon addition of HNO. In addition, the results showed that HNO-TCF could quantitatively detect HNO in the range of 0 to 4 mu M with a detection limit of 10 nM by ratiometric absorption and fluorescence spectrometry methods. Importantly, HNO-TCF was successfully applied to the fluorescence imaging of HNO levels in living cells, and it is expected to be a useful chemical tool for investigating the detailed functions and mechanisms of HNO in living systems.

Abstract:Nitroxyl (HNO), as a reactive nitrogen species (RNS), has a pivotal role in many physiological and pathological processes, but the generation mechanism of endogeneous HNO has not been elucidated, because of the lack of highly sensitive and selective probes to achieve real-time visualization and monitoring of HNO in biosystems. Ratiometric fluorescence probes have emerged as powerful platforms for bioimaging applications due to their high sensitivity and anti-interference ability. In this work, we developed firstly a ratiometric fluorescent probe (TCFPB-HNO) with typical aggregation-induced emission (AIE) characteristics for the detection and visualization of HNO with excellent photostability, high specificity and sensitivity. Importantly, TCFPB-HNO was facilely fabricated in high yields via a two-step process, as a serviceable AIE toolbox, which could be employed for real-time monitoring of the HNO level in live samples.

Then they and Sears' transformation formula are utilized to derive some generalizations of $q$-Dixon formulas and $q$-Whipple formulas with two extra integer parameters. As special cases of these results, many interesting evaluations of series of $q$-Watson,$q$-Dixon, and $q$-Whipple type are displayed.

Abstract:Peroxynitrite (ONOO-) has been proven to participate in various physiological and pathological processes, and may also be a contributing factor in many diseases. In view of this, there is a need to develop detection tools for unambiguously tracking a small amount of endogenous ONOO- to reveal its exact mechanisms. In this paper, a colorimetric and red-emitting fluorescent probe Red-PN, based on a rhodamine-type fluorophore and hydrazide reactive site is described. The probe Red-PN possesses the advantages of rapid response (within 5 s), visual color change (from colorless to pink), preeminent sensitivity (detection limit = 4.3 nM) and selectivity. Because of these outstanding performances, it was possible to accurately detect endogenous ONOO-. It was encouraging that the probe Red-PN could be used effectively for tracking the relatively low levels of endogenous and exogenous ONOO-in living cells and zebrafish. Thus, it is envisioned that the probe Red-PN would have promising prospects in applications for imaging ONOO- in a variety of biological settings.

Abstract:Fluorescence imaging of nitric oxide (NO) in vitro and in vivo is essential to developing our understanding of the role of nitric oxide in biology and medicine. Current probes such as diaminofluorescein depend on reactions with oxidized NO products, but not with nitric oxide directly, and this limits their applicability. Here we report the formation of an imaging probe for nitric oxide by coordinating the highly fluorescent chemical 4-methoxy-2-(1H-naphtho[2,3-d]imidazol-2-yl)phenol (MNIP) with Cu(II). The coordination compound MNIP–Cu reacts rapidly and specifically with nitric oxide to generate a product with blue fluorescence that can be used in vitro and in vivo. In the present study MNIP–Cu was used to reveal nitric oxide produced by inducible nitric oxide synthase in lipopolysaccharide (LPS)-activated macrophages (Raw 264.7 cells) and by endothelial nitric oxide synthase in endothelial cells (HUVEC). MNIP–Cu was also used to evaluate the distribution of nitric oxide synthesis in a model of acute liver injury induced by LPS and d-galactosamine in mice. The results demonstrate that MNIP–Cu can act as a novel fluorescent probe for nitric oxide and has many potential applications in biomedical research.

Abstract:Nitric oxide (NO) is a diatomic inorganic free radical ubiquitous in mammalian tissues and cells that plays a multifac-eted role in a variety of physiological and pathophysiological processes. The strict dependence of the biological effects of NO on its concentration makes crucial its real-time monitoring. In view of the reactivity of NO with multiple bio-target, the development of NO sensors that associate a fast response rate to selectivity and sensitivity is very challenging. Herein we report a fluorescent NO probe based on a BODIPY fluorogenic unit covalently linked to a trimethoxy aniline derivative through a flexible spacer. NO leads to effective nitrosation of the highly electron-rich amino active site of the probe through the secondary oxide N2O3, resulting in an increase of BODIPY fluorescence quantum yield from f = 0.06 to f = 0.55, accompanied by significant changes in the relative amplitude of the fluorescence lifetimes. In situ generation of NO, achieved by a tailored light-activatable NO releaser, allows the real-time detection of NO as a function of its concentration and permits demonstrating that the probe exhibits a very fast response time, being < 0.1 s. This remarkable data combines with the high sensitivity of the probe to NO (LOD = 35 nM), responsiveness also to ONOO-, the other important secondary oxide of NO, independence of the fluorescence response within a wide pH range, good selectivity towards different analytes and small interference by typical physiological concentrations of glutathione. Validation of this probe in melanoma cell lines is also reported.

Abstract:Nitroxyl (HNO) is a member of the reactive nitrogen species, and how to detect it quickly and accurately is a challenging task. In this work, we designed and prepared a fluorescent ratiometric probe based on the fluorescence resonance energy transfer (FRET) mechanism, which can detect HNO with high selectivity. The coumarin derivative was used as an energy donor, the rhodol derivative was applied as an energy receptor, and 2-(diphenylphosphine)benzoate was utilized as the recognition group to detect nitroxyl. In the absence of HNO, the rhodol derivative exists in a non-fluorescent spironolactone state, and the FRET process is inhibited. Upon adding HNO, the closed spironolactone form is transformed into a conjugated xanthene structure and the FRET process occurs. This probe could specifically recognize nitroxyl, showing high sensitivity and selectivity. When the HNO concentration was changed from 3.0 × 10–7 to 2.0 × 10–5 mol·L–1, I543nm/I470nm exhibited a satisfactory linear correlation with the concentration of HNO. A detection limit of 7.0 × 10–8 mol·L–1 was obtained. In addition, almost no cell toxicity had been verified for the probe. The probe had been successfully applied to the ratiometric fluorescence imaging of HNO in HepG2 cells.This article has not yet been cited by other publications.

Abstract:Nitric oxide (NO) plays a crucial role in a variety of pathophysiological processes. Although a large number of NO fluorescent probes have been developed, it remains barely unexplored how the electrical effect of the recognition site affects responsiveness. In this work, we demonstrate an electronic-tuning strategy to enhance the sensitivity and responsiveness of NO probes based on BODIPY by decorating various groups with different electronic capabilities in the recognition unit. Experimental results show that the electronic effect at the identification section has a significant impact on the signal response. BODIPY 5 , containing the OH group at the para -position of aniline, exhibits the greatest fluorescence responsiveness with NO. The signal does not vary for the strong electron-drawing and electron-donating groups, in which electronic or charge transfer is always in the On or OFF state, as predicted by TD-DFT calculations. Additionally, the cellular assay demonstrated that the probe possesses the ability to visualize exogenous NO in lipid droplets of HeLa cells and the endogenous NO produced in Raw 264.7 cells. This study offers a useful NO imaging tool as well as a guideline for designing high-performance NO probes for practical application.

Abstract:As a common gaseous signaling molecule, hydrogen sulfide (H2S) plays a vital role in physiology and pathology. The development of fluorescent probes for detecting H2S has attracted widespread attention. However, most of the reported fluorescent probes with nitrobenzoxadiazole (NBD) as the recognition group have been widely used to simultaneously detect biothiols and H2S, instead of specifically detecting H2S. Herein, a novel NBD-based near-infrared (NIR) fluorescent probe named CX-N for the detection of H2S is synthesized. The selectivity of CX-N for H2S is significantly higher than that for biothiols and other potential interferences. After reacting with H2S, CX-N shows a significant increase in NIR fluorescence (75-fold), large Stokes shift (155 nm) and fast response (4 min). And the possible response mechanism of CX-N to H2S is given and confirmed by HPLC and HRMS. Based on the low cytotoxicity of CX-N, it has been used for H2S imaging in live cells and zebrafish. More importantly, CX-N has also been successfully applied for the real-time imaging of H2S in inflammatory and tumor mice based on its NIR emission, which provides a reliable platform for the specific recognition of H2S in complex biological systems.

Abstract:Nitric oxide (NO) and carbon monoxide (CO) gaseous signaling molecules play crucial roles in maintaining biological homeostasis, which depends on the cellular microenvironment, concentrations, as well as spatial and temporal factors. Therefore, visualizing and monitoring these molecules in living systems is crucial for comprehending their origin, activity, and biological effects. Among various detection methods, luminescence-based probes (such as fluorescence, phosphorescence, and chemiluminescence) have emerged as a powerful technique for detecting and imaging NO and CO owing to their non-invasive, highly selective, and real-time in situ detection capabilities. Abstract:Nitroxyl (HNO) is a derivative of nitric oxide (NO) that plays an essential role in various biological and pharmacological events. Until now, the in situ trapping and specific detection of HNO in living samples is still challenging. In this project, we fabricated a novel BODIPY-based micellar nanoprobe for monitoring nitroxyl in vitro and in vivo in ratiometric mode in aqueous solution. The probe (P-BODIPY-N) contains an asymmetrical BODIPY dye for fluorescent signaling and a diphenylphosphinobenzoyl as the trigger moiety; then we encapsulated P-BODIPY-N into the hydrophobic interior of an amphiphilic copolymer (mPEG-DSPE) and prepared a novel BODIPY-based micellar nanoprobe: NP-BODIPY-N. As far as we know, this probe is the first reported ratiometric fluorescent nanoprobe for HNO, which exhibits ultrasensitivity, high selectivity, and good biocompatibility. Above all, this nanoprobe shows favorable cellular uptaken and was successfully used to detect intracellular HNO released by Angeli's salt in living cells and zebrafish larvae. These results indicate that our newly designed nanoprobe will provide a promising tool for the studies of HNO in living system.

Abstract:Nitric oxide (NO) participates in various pathways and revealing its dynamics is critical for resolving its pathophysiology. While there are methods available for detecting biological NO, few are capable of tracking NO dynamics. Herein, inspired by the cellular machinery of reversible thiol modification by NO, we have successfully designed a family of cysteine analogues tagged with fluorophores for visualizing cellular NO dynamics. Biocompatible probes 1a and 2a were found to be most tolerated by the cellular machinery to swiftly switch between their original fluorescent state and quenched S-nitrosated state in response to cellular NO dynamics. We showcased their applicability in various cell types and utility to track NO fluctuations in activated murine macrophages in response to an anti-inflammatory agent treatment. We expect that probes 1a and 2a will afford promising tools for studying NO pathophysiology by tracking its dynamics in health and disease.
