Abstract:A method has been developed for the determination of 33 pesticide residues in tea, including organophosphorous, organochlorine and pyrethroid pesticides. The target analytes were extracted with the solution of acetone/dichloromethane (1:1, v/v) using accelerated solvent extraction (ASE) and then purified using gel permeation chromatography (GPC) to eliminate most of the coextracts, such as pigments, lipids and waxes. They were further purified using Carb-NH2 and Florisil solid phase extraction (SPE) cartridges prior to the identification using gas chromatography-mass spectrometry (GC-MS). The quantitative analysis was performed with flame photometric detector (FPD) for organophosphorous pesticides and electron capture detector (ECD) for organochlorine and pyrethroid pesticides. At the spiked level of 0.05 mg/kg, the recoveries for most pesticides were between 70%–120%; the relative standard deviations were less than 20%; the limits of detection varied from 0.005 to 0.05 mg/kg (defined in terms of 10 times of the baseline noise). This method is precise, sensitive and highly efficient in extraction. After routine applications, the results indicated that this method is suitable for the determination of pesticide residues in the tea for export.

Abstract:A new analytical method was developed to simultaneously determine residues of 109 pesticides (including isomers) in the foods of animal origin. Acetonitrile was selected for accelerated solvent extraction (ASE) for effectively extracting the pesticides from the fatty samples. The cleanup was performed with an automated gel permeation chromatography (GPC) cleanup system. The prepared samples were analysed with GC–MS in the selected ion monitoring mode (SIM) using one target and two qualitative ions for each analyte. Chlorpyrifos-d10 was used as an internal standard. The lowest limit of detection was 0.3μgkg−1 for some pesticides. The recoveries and relative standard deviations (RSDs) were checked by spiking untreated samples with pesticides at 0.05, 0.1 and 0.2mgkg−1. The average recoveries of most pesticides were from 62.6% to 107.8%. The precision values expressed as RSD were all ⩽20.5% (n=6). Good linearity (r⩾0.99) was observed between 0.05 and 1.5μgmL−1.

Abstract:An analytical method for determination for tebufenozide,methoxyfenozide and Fuxian residue in cabbage,pakchoi and tomato by high performance liquid chromatography(HPLC) was developed.Samples were extracted by blending with acidic methanol/water.The extract was purified first by n-hexane to remove some fatty impurities,then extracted with dichloromethane to discard water-soluble impurities,and finally cleaned by Florisil column chromatography.The HPLC conditions : Agilent HC-C18,methanol-water=75:25(V/V) as mobile phase,and UV detection at 230nm.The average fortified recoveries for tebufenozide,methoxyfenozide and Fuxian in cabbage,pakchoi and tomato samples were 76.9%～105%.Relative standard deviation was less than 10%,and the limit of detection of three pesticides in cabbage,pakchoi and tomato samples were 0.05mg/kg,respectively.
Multiresidue Pesticide Analysis in Tea Using GC–MS/MS to Determine 12 Pesticide Residues (GB 2763-2021)

Abstract:Pesticides are widely used on tea plants, and pesticide residues are of significant concern to consumers. The National Food Safety Standard Maximum Residue Limits for Pesticides in Food (GB 2763-2021) was recently amended. However, detection methods for pesticides newly added to the list of residues in beverages have not yet been established. For that reason, this study developed a solid-phase extraction (SPE) and gas chromatography–tandem mass spectrometry (GC–MS/MS) method for determining the residues of 12 pesticides, including four newly added, in black and green tea. Sample preparation processes (sample extraction, SPE clean-up, elution solvent, and elution volume) were optimized to monitor these residues reliably. Multiple reaction monitoring (MRM) was used for GC–MS/MS electron impact (EI) mode determination. Finally, satisfactory recoveries (70.7–113.0% for green tea and 72.0–99.1% for black tea) were achieved at three concentrations (10 μg/kg, 20 μg/kg, and 100 μg/kg). The LOQs were 0.04–8.69 μg/kg, and the LODs were 0.01–3.14 μg/kg. This study provides a reliable and sensitive workflow for determining 12 pesticide residues in tea, filling a gap in the newly revised National Standards.

A HPLC Method for Detection of 17 Characteristic Components in Tea Extract

Abstract:A HPLC method has been developed for the determination of tea for seven catechins ((+)-catechin, (−)-epicatechin, (−)-epigallocatechin, (−)-epigallocatechin gallate, (−)-epicatechin gallate, (−)-gallocatechins and (−)-gallocatechin gallate), two flavonols (quercetin, kaempferol), seven organic acids (Gallic acid, ferulic acid, protocatechuic acid, p-hydroxybenzoic acid,p-coumaric acid, erucic acid,vanillic acid) and caffeine. Using HPLC with gradient elution and photodiode array detection, seventeen compounds were separated and detected by optimizing the column temperature, mobile phase ratio and detection wavelength. The validity of this method was confirmed by the quantitative measurement of seventeen characteristic compounds. The correlation coefficient of all standards curve was 0.9967~1.0000 and the detection limits (s/N = 3) were 0.010~1.2 mg/L. The average recoveries of 17 characteristic components were between 81.50 and 124.80%. 59 green tea samples (origin: Hangzhou, China) were analyzed and 11 characteristic components were found in all samples and their content of different samples varied greatly. The catechins were the most abundant constituents in teas with up to 79.5~156.6 g/kg and the contents of (−)-epigallocatechin gallate were the highest. Quercetin and kaempferol were only found in some tea samples. Protocatechuic acid, p-hydroxybenzoic acid, ferulic acid and erucic acid were not detected in the tea samples. This method is ideally suited for routine analysis for the determination of bioactive components in green tea with good repeatability and accuracy of results. Furthermore, this method can be applied to all kinds of tea and tea products and provided a detection method for establishing the HPLC fingerprint of a certain tea.

Abstract:A new fast analytical method for carbendazim residues in tea was established,it was included extracting with the accelerated solvent extract(ASE)and clean up by the solid phase extraction(SPE)with the LC-SCX column.The HPLC conditions were Xterra-C18 column,methanol and 5 mmol/L acetic acid ammonium(45:55,V/V)as mobile phase,and UV detection at 280 nm.The fortified recoveries of carbendazim in the blank tea were from 75.0% to 106.0%.The relative standard deviation was less than l0%,and the limit of detection was 0.01 mg/kg.This analytical method was satisfied with the carbendazim residue in the tea and was very simple and fast with good repeatability.

Abstract:A simple and sensitive method was developed and validated for the simultaneous determination of 103 pesticide residues in tea by LC-MS/MS. For the analysis of the pesticide with polarity, thermal lability or low volatility, this LC-MS/MS method has an advantage over GC. In this work, residual pesticides were extracted from the tea sample with ACN and then purified using Carb-NH2 SPE cartridges. Using the multiple reaction monitoring mode, the pesticides were quantified and identified by the most abundant and characteristic fragment ions. The recoveries obtained for each pesticide ranged between 65 and 114% at three spiked concentration levels. The intra-day precisions were lower than 19.6%. Good linear relationships were observed with the correlation coefficients r(2) >0.996 for all analytes. The established method was successfully applied to the determination of pesticide residues in real tea samples.
Compensation for Matrix Effects in the Gas Chromatography-Mass Spectrometry Analysis of 186 Pesticides in Tea Matrices Using Analyte Protectants.

Abstract:A gas chromatography-mass spectrometry (GC-MS) analytical method was developed for simultaneously determining 186 pesticides in tea matrices using analyte protectants to counteract the matrix-induced effect. The matrix effects were evaluated for green, oolong and black tea, representing unfermented, partially fermented and completely fermented teas respectively and depending on the type of tea, 72%, 94% and 94% of the pesticides presented strong response enhancement effect. Several analyte protectants as well as certain combinations of these protectants were evaluated to check their compensation effects. A mixture of triglycerol and D-ribonic acid-gamma-lactone (both at 2 mg/mL in the injected samples) was found to be the most effective in improving the chromatographic behavior of the 186 pesticides. More than 96% of the 186 pesticides achieved recoveries within the range of 70-120% when using the selected mixture of analyte protectants. The simple addition of analyte protectants offers a more convenient solution to overcome matrix effects, results in less active sites compared to matrix-matched standardization and can be an effective approach to compensate for matrix effects in the GC-MS analysis of pesticide residues. Abstract:A combined method of multi-plug filtration cleanup (m-PFC) and liquid chromatography-triple quadrupole mass spectrometry (LC-MS/MS) was established to simultaneously detect 14 pesticides in tea. The pesticides in water-soaked tea were extracted with acetonitrile. Cleanup of tea extract was performed using an m-PFC column packed with multiple cleanup materials: multi-walled carbon nano-tubes (MWCNTs), primary secondary amine (PSA) and anhydrous magnesium sulfate (MgSO4). The cleanup effect of the column was evaluated based on the rates of removal of tea components that interfered with pesticide recovery, henceforth referred to as interference components. Results showed that 14 pesticides had strong linearity in the range of 5–500 μg L−1 (r2 > 0.99). The quantitative limits were within the range of 3–50 μg kg−1. The average recoveries of 14 pesticides spiked into three different blank tea samples (green tea, black tea, oolong tea) at three levels of 0.05, 0.50 and 2.00 mg kg−1 were in the range of 62.3–108.8% with relative standard deviations of 0.2–13.6%. The m-PFC method can greatly improve the efficiency of sample pretreatment. Furthermore, this work provides methodological guidance on how to select cleanup materials and allocate their proportions.

Abstract:A multi-residue method for the analysis of pesticides in tea was developed by online size exclusion chromatography (SEC)-GC/MS with full scan mode. The sample was fortified with chlorpyrifos-d(10) isotope internal standard and extracted by acetonitrile. After purification by primary secondary amine sorbent and solvent exchange by SEC mobile phase, the sample was detected by online SEC-GC/MS. The purification result of the online system was evaluated by comparing the correlation between Chinese cabbage and tea matrix. The factors for method optimization included sample preparation, matrix effects and the instrument parameters of each online component. Scatter plot was introduced in this study to directly illustrate the results of the condition optimization and matrix effects in the online system. For most of the pesticides, the average recoveries ranged from 70 to 130% and the RSD were below 15%. The feasibility of the application of full scan mode in multi-residue determination of trace amounts of pesticides (LODs below 0.01 mg/kg) in a complex matrix was discussed.
Quantification of 397 pesticide residues in different types of commercial teas: Validation of high accuracy methods and quality assessment

Abstract:Analytical methods with high sensitivity and accuracy were successfully validated for the quantification of 397 pesticides in different types of tea. For the UPLC-MS/MS method, 191 pesticides of 200 ones in total had "soft" matrix effects and could be quantified by calibration curves in the solvent. For the GC-MS/MS method, matrix-matched calibration curves were established on a mixed blank including white, green, oolong and black organic teas. The method limit of quantifications ranged from 1.0 µg kg-1 to 10 µg kg-1 (UPLC-MS/MS) and 1.0 µg kg-1 to 50 µg kg-1 (GC-MS/MS), with 70-120% of recovery. These methods was subsequently applied to 106 tea samples from several origins, in which 26 samples contained at least one pesticide violation, with a total of 43 pesticide residue violations. The most frequently detected pesticides were neonicotinoids, synthetic pyrethroids, and triazole fungicides. Taiwan had the most pesticide-contaminated samples followed by China, Vietnam, and India.
Determination of multiresidue pesticides in green tea by using a modified QuEChERS extraction and ion-trap gas chromatography/mass spectrometry

Abstract:The pesticide residues in exported and imported tea products must not exceed the maximum residue limits (MRLs) regulated by the import countries. Tea is a complex matrix that obfuscates the determination of pesticide residues. Many available methods for multiresidue pesticide analysis of tea are time-consuming and require many cleanup steps. The objective of this study was to develop a simple multiresidue method by using a modified quick, easy, cheap, effective, rugged, and safe (QuEChERS) extraction and ion-trap GC/MS/MS, which can identify, confirm, and quantify pesticides in complex matrixes. A tea product was homogenized with water, and the pesticides were extracted with acetonitrile containing 1% acetic acid. The extract was subjected to centrifugation, initial cleanup with dispersive SPE (dSPE), solvent exchange, and final cleanup with dSPE. Diethyl-d10-parathion and triphenyl phosphate were used as the internal standard and surrogate, respectively. The final extract was injected into an ITQ 700 gas chromatograph/mass spectrometer. Quantitation of individual pesticides was based on matrix-matched calibration curves with a correlation coefficient of > 0.9930 for the 22 pesticides selected for the study. The recoveries of the 22 pesticides ranged from 78 to 115%, except those for diazinon (130%) and malathion (122%), with an average RSD of 8.7%. The LOD values of all of the pesticides, except for terbufos, were below the MRLs set by the European Union and Japan.
Simultaneous Screening and Quantification of 479 Pesticides in Green Tea by LC-QTOF-MS

Abstract:A high-throughput screening and quantification method for 479 pesticides in green tea was established based on solid-phase extraction combined with liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (LC-QTOF-MS). Pesticides were extracted from samples using an optimized SPE (TPT cartridges) procedure. LC-QTOF-MS in All Ions MS/MS scan mode acquired full MS data for quantification and product ion spectra for identification. LC-QTOF-MS quantification was achieved using matrix-matched standard calibration curves to achieve the optimal method accuracy. The method performance characteristics included the linearity, overall recovery, precision, and measurement uncertainty being evaluated. The validation results exhibited a good sensitivity with the LOQs of 5-55 µg/kg, which was satisfactory for their MRLs in China or the EU. The recoveries of more than 92.7% of the 479 pesticides in green tea were 70-120% at the three spiked levels with a precision of ≤20%. Finally, this method was employed to analyze 479 pesticides in 95 tea samples from markets in China. The test results of the tea samples showed that tolfenpyrad, buprofezin, and pyridaben were found with lower concentrations. The method has effectively improved the determination efficiency of pesticide residue screening by high-resolution mass spectrometry in green tea.
Utilizing a novel sorbent in the solid phase extraction for simultaneous determination of 15 pesticide residues in green tea by GC/MS

Abstract:Pesticide residues exceeding standard in green tea is a widespread problem of the world's attention, containing organochlorine pesticides (OCPs), organophosphorus pesticides (OPPs), and pyrethroids. In this research, three dimensionally honeycomb Mg-Al layered double oxide (TDH-Mg-Al-LDO) combined with graphitized carbon black (GCB), packed as a column, was used as a novel solid phase extraction sorbent, applying in simultaneous determination of 15 pesticide residues in green tea coupled with GC-MS. Compared with different type of SPE column, it showed that TDH-Mg-Al-LDO exhibited great advantages in the extraction of 15 pesticide residues from green tea, which was seldom reported before. Different experiment conditions, such as combination order of Mg-Al-LDO and GCB, dosage of sorbents, type and volume of eluting solvent were thoroughly studied and optimized. The limits of detection (LODs) of 15 pesticides ranged from 0.9 to 24.2ng/g and the limits of quantifications (LOQs) were ranging from 3.0 to 80.0ng/g. The recoveries using this method at three spiked concentration levels (10, 100 and 500ng/g for Fenthion, P,P'-DDE, O,P'-DDT, P,P'-DDD and Bifenthrin, 100, 500 and 2000ng/g for the others) range from 71.1 to 119.0%. The relative standard deviation (RSD) was from 0.1 to 7.6% in all case. The result indicated that the proposed analytical method had been successfully applied for the simultaneous determination of 15 pesticide residues in commercial green tea.

Abstract:Monitoring pesticide residues is a key item in the field of food safety. Tea matrices represent a challenge in pesticide analysis due to the high content of coextracts. An interlaboratory comparison was organized to assess the performance of 73 laboratories, from 28 regions in China, on the quantification of five pesticide residues in tea matrix reference material in order to ensure the accuracy, reliability and comparability of monitored pesticide residues in foods. The tea matrix reference material spiked with pesticides was prepared according to ISO 17034. Briefly, blank green tea was spiked with bifenthrin, permethrin, fenpropathrin, buprofezin and imidacloprid, smashed, sieved, homogenized and bottled. It was assessed to be sufficiently homogeneous and stable. Participant laboratories were requested to submit quantification results with detailed analytical protocols. Assigned values were obtained from consensus values by robust analysis, and the results were assessed using z scores. The robust standard deviations for five pesticides were lower than the expected standard deviations in our interlaboratory comparison. In total, 93.8 % (255/272) of the results were acceptable (|z|≤ 2.0) and 82.2 % (60/73) of participant laboratories had satisfactory performance in pesticide residues quantification. Analytical protocols of extraction, cleanup and quantification methods were reviewed to assess their impacts on the results. Overall, a tea matrix reference material was successfully prepared for an interlaboratory comparison, and the results indicate that most of domestic laboratories have acceptable capability in pesticides quantification, while a few of them need to further optimize their protocols to improve performance.

Abstract:An ultra?high performance liquid chromatography?photodiode array detection?tandem mass spectrometry ( UPLC?PDA?MS/MS) method was developed for the determination of flavo?nol glycosides in tea samples. The chromatographic separation was performed on an UPLC HSS T3 column by gradient elution with the mobile phases of acetonitrile and water both containing 0?1%( v/v) formic acid. A total of 15 flavonol glycosides which include 3 myricetin glycosides, 6 quercetin glycosides and 6 kaempferol glycosides were positively identified in green and black tea samples by comparing the retention times and mass spectra of the samples with standards and publications. The quantities of flavonol glycosides were relatively calculated with the stand?ard quercetin?3?rhamnosylglucoside ( Q?GRh) which was calibrated with external quantification method using multi?reaction monitoring ( MRM ) mode. The results showed that there were different flavonol glycoside distributions in green tea and black tea. The total amount of flavonol glycosides in green tea was 1?7 times of that in black tea. The major flavonol glycosides in green tea were myricetin?3?galactoside ( M?Ga) , myricetin?3?glucoside ( M?G) , quercetin?3?glucosyl?rhamnosyl?galactoside ( Q?GaRhG ) , quercetin?3?glucosyl?rhamnosyl?glucoside ( Q?GRhG) , kaempferol?3?glucosyl?rhamnosyl?galactoside ( K?GaRhG ) and kaempferol?3?glucosyl?rhamnosyl?glucoside ( K?GRhG) , but for black tea, the major flavonol glycosides were querce?tin?3?rhamnosylglucoside ( Q?GRh) , quercetin?3?glucoside ( Q?G) , kaempferol?3?rhamnosylglu?coside (K?GRh) and kaempferol?3?galactoside (K?Ga). Rapid Limit Test of Seven Pesticide Residues in Tea Based on the Combination of TLC and Raman Imaging Microscopy

Abstract:Numerous studies have shown that pesticide residues in tea exceeding the maximum residue limits (MRL) can cause harmful effects on the human body. There are many limitations in the existing analytical methods for pesticide residues in tea, so new analytical methods need to be developed. We developed a limit test method that combines thin-layer chromatography with Raman imaging microscopy (TLC-RIM). Seven residual pesticide components in tea (Avermectin, Methomyl, Carbendazim, Imidacloprid, Chlorothalonil, Azoxystrobin, and Acetamiprid) could be preliminarily separated by TLC and then irradiated by a 532 nm laser. Raman spectra of seven pesticides obtained by Raman imaging microscopy could be used to test whether the pesticide residues in tea exceed the MRL. The limits of detection of the seven pesticides were 0.04, 0.10, 0.24, 0.20, 0.12, 0.12, and 1.0 mg/mL, respectively. The simulated positive test showed that the matrix in tea did not interfere with the test of the seven pesticides. When the pesticides were tested within 8 h, the RSD of the peak heights of the seven pesticides were 1.2%~9.6%; the test results of three batches of tea showed that the imidacloprid in one batch of tea exceeded its MRL, and the results were consistent with that by UPLC-MS/MS. The TLC-RIM is fast, sensitive, stable, specific, and reliable.

An analysis of prognostic features in infiltrating lobular carcinoma of the breast.

Abstract:To establish prognostically useful pathologic features for infiltrating lobular carcinoma, histologic pattern, nuclear Grade 1 or 2, lymphatic invasion, the presence and extent of lobular carcinoma in situ, estrogen and progesterone receptor status, axillary lymph node status, tumor size, and pathologic stage were assessed as prognostic variables in 92 cases of infiltrating lobular carcinoma. Clinical follow-up was obtained (mean duration, 5.2 yr), and patients were classified as alive with no evidence of disease, alive with disease, or dead of disease. Recurrence (alive with disease and dead of disease) was associated with axillary lymph node metastases (P = 0.04), tumors measuring > 1.0 cm (P = 0.008), and pathologic Stage III/IV disease (P = 0.033). Survival (no evidence of disease and alive with disease) was associated with Stage I/II disease (P = 0.003). Statistically insignificant associations with disease recurrence or survival follow: infiltrative pattern (classical, alveolar, solid, mixed), nuclear grade, lymphatic vessel invasion, presence of lobular carcinoma in situ, extent of lobular carcinoma in situ (< 25% or > or = 25%), and hormone receptor status. Many of the prognostic features used in ductal carcinoma do not appear to be applicable to infiltrating lobular carcinoma. However, tumor size, axillary node status, and pathologic stage are prognostically useful in infiltrating lobular carcinoma.

Abstract:Due to lack of chromogenic groups and fluorescence groups, high boiling point, high polarity, low volatility, and small molecular weight of glyphosate, glufosinate and bentazone, the detection of three analyses were limited in all kinds of food. Herein, a method for the simultaneous determination of glyphosate, glufosinate and bentazone in tea by ion chromatography tandem triple quadrupole mass spectrometry (IC-MS) was developed, which is without organic solvent and complex derivatization. The recoveries of three compounds in different teas (black tea, green tea, white tea) ranged from 80.40% to 107.00%, and the intraday precision (n = 6) ranged from 0.57% to 9.90%, the daytime precision ranged from 1.00% to 5.30%, the quantitative limit (LOQ) ranged from 0.36 to 1.30 μg/L, and the detection limit (LOD) ranged from 0.11 to 0.39 μg/L. Furthermore, the detection limit and quantitative limit of glyphosate, glufosinate and bentazone by this method are lower than other methods in real samples. Meanwhile, the established method was successfully applied to determine the terminal residues of the three analytes in twelve tea samples from commercial market. Therefore, this method can provide reliable technical support for the study of residue status in vegetables and fruits.

Abstract:A multiresidue method based on gel permeation chromatography(GPC) for the sample preparation and gas chromatography-mass spectrometry analysis was developed to determine five high-toxic organic phosphorus pesticides residues in tea.The sample were extracted using acetone.Then the extracts were eluted with ethyl acetate/cyclohexane(1:1) by GPC.After dehydrated and concentrated to a definite volume,the pesticides were finally determined by gas chromatography-mass spectrometry in selective-ion Monitor(SIM) simultaneously,and quantified by external standard method.The results showed a good linear relationship was obtained between concentration and peak area within the 0.005~0.500 mg/L range.Average recoveries of 5 pesticides at three fortified levels were 99.68 %~106.31 %,94.14 %~98.11 % and 93.86 %~98.21 %,respectively.The relative standard deviation of the method was less than 10 % in every case,respectively.The limits of detection of 5 pesticides were ranging from 0.0005~0.0020 mg/kg.
