Abstract:Glycans play a central role as mediators in most biological processes, but their structures are complicated by isomerism. Epimers and anomers, regioisomers, and branched sequences contribute to a structural variability that dwarfs those of nucleic acids and proteins, challenging even the most sophisticated analytical tools, such as NMR and mass spectrometry. Here, we introduce an electron tunneling technique that is label-free and can identify carbohydrates at the single-molecule level, offering significant benefits over existing technology. It is capable of analyzing sub-picomole quantities of sample, counting the number of individual molecules in each subset in a population of coexisting isomers, and is quantitative over more than four orders of magnitude of concentration. It resolves epimers not well separated by ion-mobility and can be implemented on a silicon chip. It also provides a readout mechanism for direct single-molecule sequencing of linear oligosaccharides.
In-depth profiling of carbohydrate isomers in biological tissues by chemical derivatization-assisted mass spectrometry imaging

Abstract:Carbohydrates play crucial regulatory roles in various physiological and pathological processes. However, the low ionization efficiency and the presence of linkage pattern, monosaccharide composition and anomeric configuration isomers make their in-depth analysis very challenging, especially for heterogeneous biological tissues. In this study, we propose a high-sensitive and isomer-specific imaging approach to visualize the spatial distributions of monosaccharide and disaccharide isomers by integrating chemical derivatization and matrix-assisted laser desorption/ionization tandem mass spectrometry imaging (MALDI-MS2I). 2-Pyridinecarbohydrazide (PYD) is developed as a novel derivatization reagent which can not only improves the MS sensitivity of carbohydrates, but also enables the identification and visualization of ketose and aldose monosaccharide isomers, as well as linkage pattern, monosaccharide composition and anomeric configuration disaccharide isomers by mass spectrometry imaging of isomer-specific MS/MS fragment ions. Moreover, we build quantitative MALDI-MS2 and MALDI-MS2I methods for disaccharide isomers based on the diagnostic fragment ions, and good linear relationships could be achieved both in solution and on glass slides. We expect that this study should provide new ideas for in-depth profiling of the spatial signatures of carbohydrates in biological tissues and lay the foundation for a deeper understanding of carbohydrates' structure.

Abstract:The in-depth study of glycan has drawn large research interests since it is one of the main biopolymers on the earth with a variety of biological functions. However, the distinguishment of glycans is still difficult due to the similarity of the monosaccharide building block, the anomer, and the linkage of glycosidic bonds. In this study, four novel and representative copper-bound diastereoisomeric complex ions were simultaneously detected in a single measurement by trapped ion mobility mass spectrometry, including mononuclear copper-bound dimeric ions [(Cu2+)(A)(L-Ser)-H](+) and [(Cu2+)(A)(L-His)-H](+), the mononuclear copper-bound trimeric ion [(Cu2+)(A)(L-Ser)(L-His)-H](+), and the binuclear copper-bound tetrameric ion [(Cu2+)(2) (A)(L-Ser)(2) (L-His)-3H](+) (where A denotes an oligosaccharide, and L-Ser and L-His denote L-serine and L-histidine, respectively). By combining the collision cross sections of complex ions, 23 oligosaccharide isomers were successfully distinguished including two pairs of sialylated glycan linkage isomers. In addition, due to the unique dissociation pathways of the trimeric ion, both the relative and absolute quantification of the individual isomer in the mixture could be determined using a mass spectrometry-based kinetic method. Finally, the method established above was successfully applied to the identification and quantification of glycan isomers in dairy beverages and juice. The method in the present study was sensitive to the fine difference of glycan isomers and might have wide applicability in glycoscience.

Abstract:Because of the isomeric heterogeneity that is ubiquitous in analytical science, a formidable analytical challenge is to fully discriminate multiple isomers, especially those candidate isomers with various biological functions. Ion mobility mass spectrometry (IM-MS) has gained impressive advances for gaining molecular conformations, whereas coexisting structurally similar isomers often make unambiguous discrimination impossible due to the limited IM resolution of commercially available instruments. Herein, we demonstrate an energy-resolved collision-induced fingerprint (CIF) method to fully discriminate isomeric monosaccharide derivatives that differ in terms of composition, connectivity and configuration without complex instrument modifications. By simply increasing the collisional energy in the trap cell, the full width at half maximum (FWHM) of IM peaks can be markedly narrowed by at least 2-fold. Given the excellent reproducibility of CIF measurements, the full discrimination of isomers can benefit from their unique feature values and root-mean square deviation (RMSD) in CIF spectra. Moreover, rapid discrimination of each monosaccharide derivate isomer from binary mixtures is demonstrated. This strategy will expand the horizons of IM-MS platform in the rapid differentiation of a wider range of isomers more than monosaccharide derivatives in complex systems, which facilitates the identification and evaluation of innovative isomer candidates with unexplored functions.

Direct imaging of carbohydrate stereochemistry

Abstract:Carbohydrates, essential biological building blocks, exhibit functional mechanisms tied to their intricate stereochemistry. Subtle stereochemical differences, such as those between the anomers maltose and cellobiose, lead to distinct properties due to their differing glycosidic bonds; the former is digestible by humans, while the latter is not. This underscores the importance of precise structural determination of individual carbohydrate molecules for deeper functional insights. However, their structural complexity and conformational flexibility, combined with the high spatial resolution needed, have hindered direct imaging of carbohydrate stereochemistry. Here, we employ non-contact atomic force microscopy integrated with a data-efficient, multi-fidelity structure search approach accelerated by machine learning integration to determine the precise 3D atomic coordinates of two carbohydrate anomers. We observe that glycosidic bond stereochemistry regulates on-surface chiral selection in carbohydrate self-assemblies. The reconstructed models, validated against experimental data, provide reliable atomic-scale structural evidence, uncovering the origin of on-surface chirality from carbohydrate anomerism. Our study confirms that nc-AFM is a reliable technique for real-space discrimination of carbohydrate stereochemistry at the single-molecule level, providing a pathway for bottom-up investigations into the structure-property relationships of carbohydrates in biological research and materials science.

Abstract:Glycans are the most abundant organic polymers in nature. They are essential to living organisms and regulate a wide range of biological functions. However, mass spectrometry-based identification of glycan isomers remains challenging due to the complexity of their structures including their complex compositions, linkages, and anomeric configurations. In this study, two novel complex ions, the mononuclear copper-bound dimeric ions [(Cu2+)(A)(L-His)-H]+ and the mononuclear copper-bound quaternary ions [(Cu2+)(A)(L-Ser)3-H]+ (where A denotes a disaccharide, and L-Ser/His denotes l-serine/histidine), were designed for the collision-induced dissociation-based identification and relative quantification of 14 disaccharide isomers. When the unique fragmentation patterns of the above two types of complex ions were mapped into a three-dimensional vector, all the isomers were completely distinguished. Of note, the established method is able to identify mixtures of linkage isomers only using tandem mass spectrometry based on linkage-specific fragment ions of histidine-based complex ions. Finally, the method was successfully applied to the identification and relative quantification of two disaccharide isomers (lactose and sucrose) in dairy beverages. In conclusion, the established method is sensitive to subtle structural differences in disaccharide isomers and has the potential to be used for the differentiation of various glycans.

Abstract:Carbohydrates, such as oligo- and polysaccharides, are highly abundant biopolymers that are involved in numerous processes. The study of their structure and functions is commonly based on a material that is isolated from complex natural sources. However, a more precise analysis requires pure compounds with well-defined structures that can be obtained from chemical or enzymatic syntheses. Novel synthetic strategies have increased the accessibility of larger monodisperse polysaccharides, posing a challenge to the analytical methods used for their molecular characterization. Here, we present wide mass range ultrahigh-resolution matrix-assisted laser desorption/ionization (MALDI) Fourier transform ion cyclotron resonance (FT-ICR) mass spectrometry (MS) as a powerful platform for the analysis of synthetic oligo- and polysaccharides. Synthetic carbohydrates 16-, 64-, 100-, and 151-mers were mass analyzed and characterized by MALDI in-source decay FT-ICR MS. Detection of fragment ions generated from glycosidic bond cleavage (or cross-ring cleavage) provided information of the monosaccharide content and the linkage type, allowing for the corroboration of the carbohydrate compositions and structures.This article has not yet been cited by other publications.
Rapid quantification of disaccharide isomers by derivatization in combination with ion mobility spectrometry in beer and milk

Abstract:The subtle structural variations among carbohydrate isomers pose significant challenges for their identification and quantification. Here, we propose a strategy for rapid identification and quantification of isomeric disaccharides via derivatization with 4-(3-methyl-5-oxo-pyrazolin-1-yl) benzoic acid (CPMP) and analysing by ion mobility spectrometry (IMS). After derivatization, the ionization efficiency of disaccharides was significantly improved. The disaccharide isomers were distinguished by determining the different ion mobilities of CPMP-labelled disaccharides. Among them, [M + 2CPMP + H]+ was separated with a resolution of 1.484, almost achieving baseline separation. Subsequently, [M + CPMP + Na]+ was used for the relative quantification of lactose and maltose, showing a good linear relationship with R2 > 0.990. Finally, the method was successfully applied to the identification of lactose and maltose in beer and milk. The method is fast, accurate and effective for identification of disaccharide isomers in complex samples.

Abstract:As the specialty of pedodontics enters the 1980s, there is conjecture that an oversupply of pedodontists exists. If such a situation is present, demand-related practice characteristics should retlect the excess supply conditions of the marketplace. In order to gather data on pedodontic practice characteristics, a nationwide questionnaire survey (N = 223) was undertaken. Among the findings are these: higher than expected percentage of young pedodontists are located in nonmetropolitan population areas; appointment delay measurements demonstrated huge discrepancies between desired and actual booking times; while satisfaction with the specialty of pedodontics appears to be high, there is serious concern regarding income levels, especially among young practitioners. These findings are consistent with hypotheses concerning the oversupply of dental manpower.
Differentiation of Oligosaccharide Isomers by Direct Infusion Multidimensional Mass Spectrometry

Abstract:Oligosaccharides possess many bioactivities with applications in pharmaceutical, cosmetic, and food industries. They also serve as biomarkers for various diseases including cancer and glycogen storage disorders. These make the sructural characterization of oligosaccharides very important. Unfortunately, the structural diversity found in saccharides make their characterization challenging, necessitating the development of sophiscated instrumentation to enable isomer differentiation. Herein, we report the ability of halide (Cl- and Br-) adducts to enable direct differentiation of oligosaccharide isomers using conventional collision-induced dissociation (CID) tandem MS (MS/MS). The halide adducts were generated by direct infusion nano-electrospray ionization (nESI). For the first time, this traditional nESI CID MS/MS platform was used to differentiate stereoimers of trisacchaerides (cellotriose β(1→4) and maltotriose α(1→4), tetrasaccharides (cellotetraose and maltotetraose), and pentasaccharides (cellopentaose and maltopentaose). In addition, the MS/MS of halide adducts enabled the differentiation of position, structural, and linkage isomers of a total of 14 oligosaccharides. The isomer differentiation was realized by the generation of distinct diagnostic fragment ions in CID. We also performed principal component analysis using the entire MS/MS fragment ion profiles and found the negative-ion mode halide adduction to provide a more effective isomer differentiation compared with positive-ion mode sodium adduction. Finally, we demonstrated complex mixture analysis by spiking all 14 oligosaccharides into raw urine, of which we successfully distinguished species based on molecular weight (first dimension) and CID MS/MS fragmentation patterns as the second dimension separation. This work effectively showcases the potential to use direct infusion nESI-MS/MS to characterize synthetic oligosaccharide isomers in unpurified reaction mixture as well as from biofluids for diagnostic purposes.

Abstract:The fact that practically all living cells are covered with carbohydrates gives an indication that these unique molecules are of tremendous importance to life with impacts in health and disease. One example of the significance of carbohydrates is found in the blood types that are determined by minor differences in the structure of the glycans on the surface of red blood cells. Other examples include glycosylation changes in cancer, the masking of viruses by glycans to evade the host immune response, and the inflammation resulting from exposure to endotoxins in gram-negative bacteria, among many others. As science delves deeper into understanding biological processes, it is increasingly confronted with the challenge to account for the roles that carbohydrates play in the molecular interactions that drive these processes. Molecular interactions are based on molecular shape, and molecular shape is determined by primary structure. Thus, it is vital to enlarge our repertoire of analytical tools for the elucidation of the primary structure of carbohydrates. Although a number of traditional analytical tools, including monosaccharide and linkage analysis by gas chromatography/mass spectrometry (GC/MS), glycan composition analysis by matrix-assisted laser desorption ionization mass spectrometry (MALDI-MS), sequence analysis by tandem mass spectrometry (MSn), and nuclear magnetic resonance (NMR) spectroscopy, have been available for some time and have been used with tremendous success, a host of challenges posed by the specific structural peculiarities of carbohydrates remain to be met. These include the varying stability of glycosidic bonds and free monosaccharides to hydrolysis, the nonstoichiometric substitution by noncarbohydrate residues, the difficulty of automating the existing protocols for high-throughput analysis, the scarcity of standards, the unparalleled structural diversity and close similarity of different structures, and the lack of chromophores that can often not be directly overcome with labeling.

Abstract:Rationale The 1, 2‐unsaturated derivatives of (1‐4) linked disaccharides serve as versatile building blocks for synthesizing biologically active compounds. Distinguishing between four pairs of stereoisomers in mixtures presents a challenging task. In this study, disaccharide derivatives are analyzed as alkali metal adducts using electrospray ionization tandem mass spectrometry (ESI‐MS/MS), both as individual compounds and in mixtures by ion mobility mass spectrometry (IMS). Methods Electrospray ionization (ESI) in combination with tandem mass spectrometry (MS/MS) in positive mode was employed to differentiate lithium adduct ions of hexa‐acetyl/hexa‐benzyl‐D‐lactals, and hexa‐acetyl/hexa‐benzyl‐D‐maltals, along with their corresponding 2‐ C ‐branched malonates. The high resolving power of trapped ion mobility spectrometry (TIMS) with the imeXTM functionality rapidly identified different metal adducts (Li, Na, Cs) as individual isomers and separated mixtures of stereoisomers. The measured collisional cross section (CCS) values were analyzed in relation to predicted CCS values. Results MS/MS spectra of the [M + Li] + ions of glycal disaccharide analogues exhibited typical cross‐ring and glycosidic bond cleavages. Collision‐induced dissociation (CID) spectra provided insights into their fragmentation behavior, allowing differentiation of (1‐4) linked disaccharides. TIMS technology delivered adjustable ion mobility resolution for suitable separation of the four sets of stereo isomeric compounds. However, accurately predicting CCS values to differentiate between respective isomeric pairs using the Sigma CCS program for sodium adducts is only partially achievable. Conclusions ESI CID spectra of [M + Li] + adduct ions for individual glycal disaccharide analogues facilitate the discrimination between alpha and beta (1‐4) linked unsaturated disaccharides and their 2‐ C ‐branched analogues. Through optimized experimental conditions, complete baseline ion mobility separation of stereoisomer pairs of the Cs adducts is achieved.

Abstract:Ion mobility mass spectrometry (ESI-tims-ToF–MS, syringe pump infusion) has been applied to glucose and oligosaccharide ethers derived from hydroxyethyl-methyl celluloses (HEMC) and hydroxyethyl celluloses (HEC) after permethylation and partial depolymerization: by hydrolysis without or with subsequent reductive amination with m -amino benzoic acid ( m ABA) or by reductive cleavage. As model compounds without tandem substitution methoxyethylated methylcellulose was used. Regioisomeric glucose ethers were separated according to their ion mobility, and positions of substitution could be assigned. Glucose ethers including isomers with tandem substitution showed additional signals with a smaller collision cross-section (CCS) than core-substituted isomers. Positional isomers of cellobiose ethers were only partly resolved due to too high complexity but showed a characteristic fingerprint that might allow classifying samples. Relative intensities of signals of glucose ether isomers could only be quantified in case of ABA derivatives with its fixed charge, while sodium adducts of methoxyethyl ethers showed an influence of the MeOEt position on ion yield. Results were in very good agreement with reference analysis. [M + Na] + adducts of α- and β-anomers of glucose derivatives were separated in IM, complicating position assignment. This could be overcome by reductive cleavage of the permethylated HE(M)C yielding 1,5-anhydroglucitol-terminated oligosaccharides, showing the best resolved fingerprints of the cellobiose ethers of a particular cellulose ether. With this first application of ion mobility MS to the analysis of complex cellulose ethers, the promising potential of this additional separation dimension in mass spectrometry is demonstrated and discussed.

Abstract:Carbohydrates, in particular the d -enantiomers of ribose, 2-deoxyribose, and glucose, are essential to life's informational biopolymers (RNA/DNA) and for supplying energy to living cells through glycolysis. Considered to be potential biosignatures in the search of past or present life, our capacity to detect and quantify these essential sugars is crucial for future space missions to the Moon, Mars or Titan as well as for sample-return missions. However, the enantioselective analysis of carbohydrates is challenging and both research and routine applications, are lacking efficient methods that combine highly sensitive and reproducible detection with baseline enantioselective resolution and reliable enantiomeric excess ( ee ) measurements. Here, we present four different derivatization strategies in combination with multidimensional gas chromatography coupled to a reflectron time-of-flight mass spectrometer (GC×GC–TOF-MS) for the enantioselective resolution of C3 to C6 carbohydrates potentially suitable for sample-return analyses. Full mass spectral interpretation and calibration curves for one single-step (cyclic boronate derivatives) and three two-step derivatization protocols (aldononitrile−acetate, hemiacetalization−trifluoroacetylation, and hemiacetalization−permethylation) are presented for concentrations ranging from 1 to 50 pmol μL− 1 with correlation coefficients R 2 > 0.94. We compared several analytical parameters including reproducibility, sensitivity (LOD and LOQ), overall separation, chiral resolution ( R S ), mass spectrum selectivity, stability during long term storage, and reliability of ee measurements to guide the application-dependent selection of optimal separation and quantification performance.

Abstract:N-Linked glycosylation is one of the most essential post-translational modifications of proteins. However, N-glycan structural determination remains challenging because of the small differences in structures between isomers. In this study, we constructed a database containing collision-induced dissociation MSn mass spectra and chromatograms of high-performance liquid chromatography for the rapid identification of high-mannose and paucimannose N-glycan isomers. These N-glycans include isomers by breaking of arbitrary numbers of glycosidic bonds at arbitrary positions of canonical Man9GlcNAc2 N-glycans. In addition, some GlcMannGlcNAc2 N-glycan isomers were included in the database. This database is particularly useful for the identification of the N-glycans not in conventional N-glycan standards. This study demonstrated the application of the database to structural assignment for high-mannose N-glycans extracted from bovine whey proteins, soybean proteins, human mammary epithelial cells, and human breast carcinoma cells. We found many N-glycans that are not expected to be generated by conventional biosynthetic pathways of multicellular eukaryotes.

Abstract:Elucidation of complex structures of biomolecules plays a key role in the field of chemistry and life sciences. In the past decade, ion mobility, by coupling with mass spectrometry, has become a unique tool for distinguishing isomers and isoforms of biomolecules. In this study, we develop a concept for performing ion mobility analysis using an ion trap, which enables isomer separation under ultra-high fields to achieve super high resolutions over 10,000. The potential of this technology has been demonstrated for analysis of isomers for biomolecules including disaccharides, phospholipids, and peptides with post-translational modifications.
[A clinicopathological study of eighteen autopsy cases with acquired toxoplasmosis].

Abstract:Eighteen autopsy cases of acquired toxoplasmosis in New York City were studied. Seventeen cases were with acquired immunodeficiency syndrome (AIDS) and one patient with Hodgkin's disease. All 18 cases involved the brain and nine of them disseminated to the heart (8 cases), lung (4 cases), pancreas (3 cases), alimentary tract (2 cases) and urogenital organs (3 cases). The authors divided the acquired toxoplasmosis into (1) immunocompetent, (2) immunocompromised and (3) immunodeficient types. The autopsy findings showed that the brain, heart and lung were the most susceptible organs. Pseudocysts were also found in lungs and alimentary tract, suggesting an autoinfection by swallowing sputum containing Toxoplasma. Toxoplasma in the urogenital organs might become a source for sexual transmitted toxoplasmosis.
Probing High-Order Transient Oligomers using Ion Mobility Mass Spectrometry coupled to Infrared Action Spectroscopy

Abstract:Understanding and controlling peptide aggregation is critical due to its neurotoxic implications. However, structural information on the key intermediates, the oligomers, is obscured by a cascade of coinciding events occurring at various time and energy scales, which results in complex and heterogeneous mixtures of oligomers. To address this challenge, we have developed the Photo-Synapt, a novel, multidimensional spectrometer that integrates ion mobility mass spec-trometry with infrared (IR) action spectroscopy within a single experiment. By combining three different, orthogonal analytical dimensions, we can select and isolate individual oligomers by mass, charge, size and shape and provide a unique molecular fingerprint for each oligomer. The broad application of this technology is demonstrated by its ap-plication to oligosaccharide analysis from glycoproteins, which are challenging to analyze due to the minute differ-ences between isomers. By integrating infrared action spectroscopy with ion mobility mass spectrometry, this approach adds an analytical dimension that effectively addresses this limitation, offering a unique molecular finger-print for each isomer.

Impurity spin textures across conventional and deconfined quantum critical points of two-dimensional antiferromagnets

Abstract:We describe the spin distribution in the vicinity of a nonmagnetic impurity in a two-dimensional antiferromagnet undergoing a transition from a magnetically ordered N\'eel state to a paramagnet with a spin gap. The quantum critical ground state in a finite system has total spin $S=1∕2$ (if the system without the impurity had an even number of $S=1∕2$ spins), and recent numerical studies in a double layer antiferromagnet [K. H. H\"oglund et al., Phys. Rev. Lett. 98, 087203 (2007)] have shown that the spin has a universal spatial form delocalized across the entire sample. We present the field theory describing the uniform and staggered magnetizations in this spin texture for two classes of antiferromagnets: (i) the transition from a N\'eel state to a paramagnet with local spin singlets, in models with an even number of $S=1∕2$ spins per unit cell, which are described by a O(3) Landau-Ginzburg-Wilson field theory; and (ii) the transition from a N\'eel state to a valence bond solid, in antiferromagnets with a single $S=1∕2$ spin per unit cell, which are described by a ``deconfined'' field theory of spinons.
Effects of Gelam honey on sperm quality and testis of rat

Abstract:The present study aimed to elucidate the possible protective effects of Gelam honey on sperm quality and testis histology against infertility related problems. Control and treated groups of 4 - 5 weeks old male Sprague-Dawley rats were force-fed daily with 1.0 mL/100 g body weight of normal saline (0.9%) and Gelam honey, respectively. After 60 days of treatments, reproductive organs of the anesthetized rats were removed to assess sperm parameters and histology of testis. Sperm count of treated group was significantly higher (18.85 +/- 3.72x10(5)/mL) than control group (17.05 +/- 3.12x10(5)/mL) (p<0.05). Based on sperm morphology, treated group showed significantly higher percentage of normal sperm (96.83 +/- 0.03%) as compared to control group (94.87 +/- 0.01%) (p<0.01). Head and tail abnormalities sperm were also significantly reduced in the treated rats (p<0.05). The number of spermatogenic cells in testis of treated group were abundant as compared to control group. Seminiferous tubules of treated group were densely packed with spermatogenic cells with small lumen filled up with sperm tail. This study suggested that Gelam honey has the potential to increase the fertility of male rats by increasing sperm count and number of sperm with normal morphology.
