Abstract:Numerous abiotic stressors including heavy metal stresses, specifically cadmium (Cd) stress in agricultural bio-system hinder the plant adequate growth. The present study was aimed to reveal the protective role of silicon (Si) application with two levels and to recognize the optimum level of Si for wheat plants grown hydroponically under three different levels of Cd toxicities. In methodology, we used nine treatments with three levels of Si (0, 1, and 3 mmol L-1; Na2SiO3) against three levels of Cd (0, 50, 200 μmol L-1; CdCl2) with three biological replicates. The results of our study demonstrated that Si incorporation with the advantage of 3 mmol L-1 in cultured media with Cd50 and Cd200 demolished the toxic effects of Cd on the leaves of wheat plants by increasing plant dry biomass by 88% and 262%, leaf area by 48% and 57%, total chlorophyll contents by 120% and 74%, catalase (CAT) activity by 92% and 110%, superoxide dismutase (SOD) activity by 62% and 78%, peroxidase (POD) activity by 66% and 40%, ascorbic acid (AsA) contents by 33% and 34%, glutathione (GHS) contents by 39% and 30% and reduced MDA contents by 56% and 50%, H2O2 contents by 61% and 66%, and EL contents by 56% and 47% as parallel to Cd corresponding levels. In addition, Si incorporation with the advantage of 3 mmol L-1 significantly increased relative water contents (RWC) to maintain the cell turgor pressure and protect the plant from wilting and cells flaccid and enhanced membrane stability index (MSI) to protect the plant from logging under damaging effects of Cd toxicities. Based on the present findings, Si can be considered a quasi-essential element that enhanced wheat tolerance against Cd toxicity by limiting uptake, accumulation, and translocation of Cd and through regulating antioxidative defense mechanisms.
Alleviatory effects of Silicon on the morphology, physiology, and antioxidative mechanisms of wheat (Triticum aestivum L.) roots under cadmium stress in acidic nutrient solutions

Abstract:Abstract Silicon (Si), as a quasi-essential element, has a vital role in alleviating the damaging effects of various environmental stresses on plants. Cadmium (Cd) stress is severe abiotic stress, especially in acidic ecological conditions, and Si can demolish the toxicity induced by Cd as well as acidic pH on plants. Based on these hypotheses, we demonstrated 2-repeated experiments to unfold the effects of Si as silica gel on the root morphology and physiology of wheat seedling under Cd as well as acidic stresses. For this purpose, we used nine treatments with three levels of Si nanoparticles (0, 1, and 3 mmol L −1 ) derived from sodium silicate (Na 2 SiO 3 ) against three concentrations of Cd (0, 50, and 200 μmol L −1 ) in the form of cadmium chloride (CdCl 2 ) with three replications were arranged in a complete randomized design. The pH of the nutrient solution was adjusted at 5. The averages of three random replications showed that the mutual impacts of Si and Cd in acidic pH on wheat roots depend on the concentrations of Si and Cd. The collective or particular influence of low or high levels of Si (1 or 3 mM) and acidic pH (5) improved the development of wheat roots, and the collective influence was more significant than that of a single parallel treatment. The combined effects of low or high concentrations of Cd (50 or 200 μM) and acidic pH significantly reduced root growth and biomass while increased antioxidants, and reactive oxygen species (ROS) contents. The incorporation of Si (1 or 3 mmol L −1 ) in Cd-contaminated acidic nutrient solution promoted the wheat root growth, decreased ROS contents, and further increased the antioxidants in the wheat roots compared with Cd single treatments in acidic pH. The demolishing effects were better with a high level of Si (3 mM) than the low level of Si (1 Mm). In conclusion, we could suggest Si as an effective beneficial nutrient that could participate actively in several morphological and physiological activities of roots in wheat plants grown under Cd and acidic pH stresses.

Abstract:Silicon (Si) application enhanced the tolerance of plants against different environmental stresses. Therefore, objective of the study revealed that foliar applied Si alleviates the adverse effect of Cd by enhancing the growth, metabolite accumulation, strengthening the antioxidant defense system, reducing oxidative injury, improving plant nutrient status, and decreasing the Cd uptake in wheat. The surface sterilized seeds of Sahar-2006 (tolerant) and Inqalab-91 (sensitive) having the differential metal tolerance capacity were sown in plastic pots containing normal and Cd spiked sandy loamy soil. The design of experiments was completely randomized with 3 replicates per treatment. Two weeks after germination, plants were sprayed with different concentrations of Si (1.5 and 3 mM) with 0.1% surfactant in the form of Tween-20. The plants were harvested after 2 weeks of Si application to determine various attributes. High concentration of Cd (25 mg kg-1) decreased growth-related-attributes, essential nutrient uptake and increase the levels of oxidative stress indicators. The application of Si increased the growth-related attributes, photosynthetic pigments, essential nutrient uptake and also enhanced the activities of various antioxidant compounds (superoxide dismutase (SOD), peroxidase (POD, ascorbate peroxidase (APX) and catalase (CAT) by decreasing the contents of oxidative stress indicators and Cd uptake in root and shoot of both wheat cultivars. Sahar-2006 cultivar showed more tolerance to Cd regimes than that of Inqalab-91 as clear from greater plant dry masses. Thus, our results showed that the applied Si level (3 mM) is an efficient strategy for field use in the areas, where slightly Cd polluted soils limit the agriculture production.
Physiological and Biochemical Bases of Foliar Silicon-Induced Alleviation of Cadmium Toxicity in Wheat

Abstract:Silicon (Si) is a beneficial nutrient for crop plants, and is gaining increased attention because of its involvement in regulation of abiotic stress tolerance. The present study was carried out to explore the underlying physiological and biochemical mechanisms of foliar Si-induced enhancement in wheat tolerance against cadmium (Cd) stress. Surface sterilized seeds of two wheat cultivars, viz., Inqalab-91 (sensitive) and Sahar-2006 (tolerant), were sown in plastic pots. Foliar application of Si (3 mM) was done at 20 days after sowing under control (−Cd) and Cd stress (25 mg kg−1 soil) conditions. Plants were harvested after 3 weeks of foliar spray, and data regarding morphological growth, oxidative metabolism, photosynthetic pigments, gas exchange attributes, metabolite accumulation, and ion homeostasis were recorded. The experimental treatments were arranged in completely randomized design with four replicates per treatment. Cadmium stress was found to significantly decrease the morphological growth, photosynthesis, and photosynthetic pigments in both wheat cultivars, while significantly increased malondialdehyde (MDA) content, hydrogen peroxide (H2O2) content, and electrolyte leakage (EL) that triggered oxidative stress. The Cd-induced changes in superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX), glutathione S-transferase (GST), and glutathione reductase (GR) activities varied with cultivar and Si application. Plants treated with Si revealed better growth, higher accumulation of total soluble proteins, anthocyanins, total soluble sugar, flavonoids, and phenolics contents, and efficient antioxidant defense system as indicated by increased ascorbic acid and glutathione pools, and higher activities of SOD, POD, CAT, and APX particularly under Cd stress. Application of Si reduced Cd-mediated oxidative stress and increased photosynthetic pigments and net photosynthetic rate in both wheat cultivars. Further, Si application enhanced mineral ions in normal (−Cd) and Cd-stressed plants, and significantly decreased the Cd uptake. The Si-induced effects were more prominent in Sahar-2006 than that in Inqalab-91 for most of the studied traits, particularly under stress conditions. In short, foliar application of Si alleviates the adverse effects of Cd on wheat growth by strengthening antioxidant defense system, enhancing metabolite accumulation, improving plant nutrient status, and decreasing the Cd uptake.

Abstract:Heavy metal toxicity causes a remarkable decline in the growth and yield of numerous plant species in the agricultural system. Cadmium (Cd) among the other heavy metals is the most distinguished lethal heavy metal as its higher water solubility, relative mobility, and phytotoxicity. Silicon (Si), as a second most abundant element, has been reported to mitigate numerous abiotic stresses, comprising heavy metal stress in plants. The 2-repeated studies were conducted to disclose the protective role of silica gel derived from sodium silicate (Na 2 SiO 3 ) at the rate of 1 and 3 mmol L −1 on the root morphological characters of wheat plants grown in cultured medium with three levels of Cd toxicities (0, 50, and 200 µmol L −1 ). The results of our studies demonstrated a remarkable alteration in morphological traits of wheat roots with both levels of Cd toxicities through incorporating an excessive amount of reactive oxygen species (ROS), by lipid peroxidation, by limitation of essential nutrient availability, and by the accumulation of the high contents of Cd in root cells. While the introduction of Si nanoparticles at the rate of 1 and 3 mmol L −1 in the cultured medium having Cd toxicities remarkably demolished Cd adversity by shifting the balance between antioxidant enzymes and oxidants of ROS. Moreover, Si significantly increased cations concentrations in root tips that compete with Cd ions for exchange sites and reduced Cd uptake and accumulation by roots. Being an effective beneficial element, Si improved root morphological characters by strengthening the antioxidative defense system and by hindering ROS in root tissues grown in a cultured medium with different toxicities of Cd.

Abstract:There is a rapid increase in the contamination of agricultural soils with cadmium worldwide, especially in China, so it is necessary to find solutions to reduce the toxicity of it. To investigate the effect of silicon on cadmium toxicity in Triticum aestivum L. cv. Yannong 19, an experiment was conducted with four treatments (control, silicon, cadmium, silicon × cadmium). This experiment examined the following factors in wheat: root-and-shoot cadmium concentration, TF, root-and-shoot Silicon concentration, SOD, POD, CAT, APX, GSH, AsA, MDA, total protein content, total soluble sugars, root-and-shoot dry weight, Total chlorophyll, chlorophyll a and b, the content of N, P, and K. The present study showed that cadmium treatment severely affected wheat seedling morphological and physiological factors, but silicon treatment greatly reduced these adverse effects. Furthermore, silicon alone had significant positive effects on wheat's biochemical and morphological structures. The results of this study also revealed that silicon treatment increased TaPIP1 and TaLsi1 gene expression (involved in the absorption and transport of Si), whilst cadmium treatment caused the upregulation of TaNramp5 , TaLCT1 , TaTM20 , TaHMA2, and TaHMA3 genes (involved in the absorption and transport of Cd). In conclusion, the present study found that, although cadmium negatively affects the various structures of Yannong 19 , nevertheless, Sodium Metasilicate Nonahydrate (a source of silicon) can be a viable method of reducing cadmium absorption, transfer, and toxicity in this wheat variety.

Abstract:PurposeSilicon (Si) has inhibitory effects on cadmium (Cd) toxicity in rice, but the effects of Si-containing materials on the chemical properties of hydroponic media have been ignored. The increased pH value and introduction of excess cations when utilizing alkaline reagents may disguise the true function of the Si. Thus, the present study investigated Si-alleviated effect on Cd toxicity in rice seedlings by eliminating the aforementioned interferences.Materials and methodsThe hydroponic cultivation of rice seedlings was carried out to demonstrate the inhibiting effects of exogenous Si on Cd accumulation and distribution. The chlorophyll and carotenoid contents in leaf cells under Cd stress were also measured. Transmission electron microscopy was used to investigate ultrastructural changes in rice mesophyll cells, chloroplasts, and mitochondria under different Cd-stressed and Si-amended conditions. The alkalinity of the Si source was neutralized with nitric acid, and discrepancies in Na+ levels was eliminated by the addition of sodium nitrate to different treatments.Results and discussionToxicity symptoms were observed in rice seedlings under Cd (5 mg L−1) treatment after 35 days, and the biomass, and chlorophyll and carotenoid contents, were lowered. The levels of these indices increased after adding 120 mg L−1 Si. The bioconcentration and translocation factor values indicated that Si reduced Cd accumulation in rice plants and also decreased the ratio of Cd translocation from roots to shoots, which effectively alleviated the Cd toxicity. The positive effects of Si were mainly reflected in the recovery of grana lamellae and membrane structures in chloroplasts, and restored membrane morphology and increased numbers of cristae in mitochondria.ConclusionsExogenous Si reduced Cd uptake in rice seedlings, especially in shoots, and inhibited Cd transport into aboveground parts. In addition, Si restored the functions of chloroplasts owing to a decrease in lipid peroxidation. The application of Si indicated that Cd toxicity is alleviated partly because of the improvement in chloroplast biosynthesis and the diminution of Cd bioavailability.

Abstract:Cadmium (Cd) contamination has resulted in serious reduction of crop yields. Silicon (Si), as a beneficial element, regulates plant growth to heavy metal toxicity mainly through reducing metal uptake and protecting plants from oxidative injury. However, the molecular mechanism underlying Si-mediated Cd toxicity in wheat has not been well understood. This study aimed to reveal the beneficial role of Si (1 mM) in alleviating Cd-induced toxicity in wheat (Triticum aestivum) seedlings. The results showed that exogenous supply of Si decreased Cd concentration by 67.45% (root) and 70.34% (shoot), and maintained ionic homeostasis through the function of important transporters, such as Lsi, ZIP, Nramp5 and HIPP. Si ameliorated Cd-induced photosynthetic performance inhibition through up-regulating photosynthesis-related genes and light harvesting-related genes. Si minimized Cd-induced oxidative stress by decreasing MDA contents by 46.62% (leaf) and 75.09% (root), and helped re-establish redox homeostasis by regulating antioxidant enzymes activities, AsA-GSH cycle and expression of relevant genes through signal transduction pathway. The results revealed molecular mechanism of Si-mediated wheat tolerance to Cd toxicity. Si fertilizer is suggested to be applied in Cd contaminated soil for food safety production as a beneficial and eco-friendly element.

Abstract:The organospecific, 1,2-dimethylhydrazine-induced rat tumor model was used to test tumor formation in groups of animals receiving regular chow, powdered chow with 7%/wt ascorbic acid supplement, pelleted chow with 1%/wt beta-carotene supplement, and pelleted chow with placebo beadlets. Following a 16-week induction period, animals were killed and tumor formation was recorded. Tumor formation in the ascorbic acid supplement group was found to be significantly less than the control group. The beta-carotene group showed no difference in tumor formation compared with the placebo-beadlet control group. Tumor incidence was generally the same between the two control groups, and the ascorbic acid group had significantly fewer tumors than the beta-carotene group. In sum, ascorbic acid supplements in high doses significantly decreased tumor formation, whereas beta-carotene supplements in moderately high doses had no effect on tumor formation in this model.

Abstract:Pot experiments were performed to study the alleviative effects of exogenous silicon (Si) on cadmium (Cd) phytotoxicity in maize grown in an acid soil experimentally contaminated with Cd. Five treatments were investigated in the first trial consisting of a control (neither Cd nor Si added), Cd added at 20 or 40mgkg−1 Cd without or with Si added at 400mgkg−1 Si. A following-up trial was conducted with almost the same treatments as in the first trial except that Si was incorporated at 50mgkg−1 Si. The results showed that Cd treatment significantly decreased shoot and root dry weight, while addition of Si at both levels significantly enhanced biomass. Addition of Si at 400mgkg−1 Si significantly increased soil pH but decreased soil Cd availability, thus reducing Cd concentration in the shoots and roots and total Cd in the shoots. Moreover, more Cd was found to be in the form of specific adsorbed or Fe–Mn oxides-bound fraction in the Si-amended soil. In contrast, soil pH, available Cd and Cd forms were unaffected by addition of Si at 50mgkg−1 Si, but shoot Cd concentration in the Si-amended Cd treatments significantly decreased at both Cd levels used compared to the non-Si-amended Cd treatments. Total Cd in the shoots and roots was considerably and significantly higher in the Si-amended Cd treatments than in the non-Si-amended Cd treatments. The xylem sap significantly increased but Cd concentration in the xylem sap significantly decreased in the Si-amended Cd treatments compared with the non-Si-amended Cd treatments irrespective of Cd and Si levels used. The results suggest that Si-enhanced tolerance to Cd can be attributed not only to Cd immobilization caused by silicate-induced pH rise in the soils but also to Si-mediated detoxification of Cd in the plants.
Interactive effect of silicon and zinc on cadmium toxicity alleviation in wheat plants

Abstract:Silicon (Si) and Zinc (Zn) have been frequently used to alleviate cadmium (Cd) toxicity, which are feasible strategies for crop safety production. However, the mechanisms underlying the interaction of Si and Zn on alleviating Cd toxicity are not well understood. A hydroponic system was adopted to evaluate morphological, physiological-biochemical responses, and related gene expression of wheat seedlings to Si (1 mM) and Zn (50 µM) addition under Cd stress (10 µM). Cd induced obvious inhibition of wheat growth by disturbing photosynthesis and chlorophyll synthesis, provoking generation of reactive oxygen species (ROS) and interfering ion homeostasis. Cd concentration was decreased by 68.3%, 43.1% and 73.3% in shoot, and 78.9%, 44.1% and 85.8% in root by Si, Zn, and combination of Si with Zn, relative to Cd only, respectively. Si and Zn effectively ameliorated Cd toxicity and enhanced wheat growth; but single Si or combination of Si with Zn had more efficient ability on alleviating Cd stress than only Zn, indicating Si and Zn have synergistic effect on Cd toxicity; Interaction of them alleviated oxidative stress by reducing ROS content, improving AsA-GSH cycle and antioxidant enzymes activities, and regulating Cd into vacuole through PC-Cd complexes transported by HMA3 transporter. Our results suggest that fertilizers including Si and Zn should be made to reduce Cd content, which will beneficial for food production and safety.
Effect of Si on the distribution of Cd in rice seedlings

Abstract:Growth chamber studies were conducted to investigate the effects of silicon (Si) on the distribution of Cd in rice seedlings (Oryzasativa L., cv. Qiu Guang) grown hydroponically under toxic level of cadmium (Cd). Si added significantly alleviated the toxicity of Cd in aerobic rice seedlings. Si partly overcame the reduction in growth due to Cd. This amelioration was correlated with a reduction in Cd uptake. Si increased Cd accumulation in the roots and restricted the transport of Cd from roots to shoots, where the distribution of Cd in the shoots decreased by 33%. Si reduced the transport of Cd and the apoplastic fluorescence tracer PTS (tri-sodium-8-hydroxy-1, 3, 6-pyrenesulphonate) from roots to shoots by 23 and 36%, respectively. Energy-dispersive X-ray analysis (EDX) showed Cd was mainly deposited in the vicinity of the endodermis and epidermis, Si deposition was heavier in the vicinity of the endodermis than in the epidermis. Although the tracing result of fluorescein isothiocyanate-dextrans showed Si did not change epidermal wall porosity, the significant reduction of apoplastic PTS transport in +Si plants suggested that the heavy deposition of silica in the vicinity of endodermis might offer possible mechanisms by which silicon did at least partially physically block the apoplast bypass flow across the roots, and restrained the apoplastic transport of Cd. In addition, the effect of Si on the subcellular distribution and chemical form of Cd was investigated by fractionation. Si decreased the concentrations of Cd in shoots and roots, but did not remarkably change the distribution ratio of Cd in symplasm and apoplast. Mechanisms by which Si alleviates the toxicity of Cd in rice seedlings are discussed.

Abstract:Cadmium (Cd), being a highly toxic heavy metal, significantly impacts plant growth and development by altering nutrient uptake and causing oxidative and structural damage, resulting in reduced yield. To combat Cd toxicity and accumulation in wheat, it was hypothesized that co-application of Selenium (Se) and Silicon (Si) can reduce the adverse effect of Cd and regulate Cd resistance while improving Se fortification in wheat. Therefore, this study evaluated the comparative effect of Se and Si on the growth and antioxidant defense systems of wheat plants grown in a hydroponic setup. Briefly, the plants were acclimatized to the hydroponic solution for 1 week and then exposed to 10 µmol Cd. Afterwards, the treatments, including 0.2 mmol Si and 1.5 µmol Se, were applied as a root and foliar application, respectively. Plants supplemented with both Se and Si showed improved biomass and other physiological growth attributes, and this response was associated with improved activity/contents of antioxidants, including glutathione (GSH) content, glutathione reductase (GR), ascorbate peroxidase (APX), and catalase (CAT), with related lowering of hydrogen peroxide, malondialdehyde content, and structural damages. Moreover, by Se + Si supplementation, a decrease in total S levels in plant tissues was observed, whereas an increase in total protein concentration and GSH indicated a different and novel mechanism of Cd tolerance and S homeostasis in the plant. It was observed that Si was more involved in significantly reducing Cd translocation by stabilizing Cd in the root and reducing its content in the soluble fraction in both the root and shoot. Whereas Se was found to play the main role in reducing the oxidative damage caused by Cd, and the effect was more profound in the shoot. In addition, this study also observed a positive correlation between Si and Se for relative uptake, which had not been reported earlier. Our findings show that the Se and Si doses together benefit growth regulation and nutrient uptake; additionally, their combinations support the Cd resistance mechanism in wheat through upregulation of the antioxidant system and control of Cd translocation and subcellular distribution, ultimately contributing to the nutritional quality of wheat produced. Thus, it is concluded that the co-application of Se and Si has improved the nutritional quality while reducing the Cd risk in wheat and therefore needs to be employed as a potential strategy to ensure food safety in a Cd-contaminated environment.
Silicon-enhanced Resistance to Cadmium Toxicity in Brassica Chinensis L. is Attributed to Si-suppressed Cadmium Uptake and Transport and Si-enhanced Antioxidant Defense Capacity

Abstract:A series of hydroponics experiments were performed to investigate roles of silicon (Si) in enhancing cadmium (Cd) tolerance in two pakchoi (Brassica chinensis L.) cultivars: i.e. cv. Shanghaiqing, a Cd-sensitive cultivar, and cv. Hangyoudong, a Cd-tolerant cultivar. Plants were grown under 0.5 and 5mg Cd L−1 Cd stress without or with 1.5mM Si. Plant growth of the Cd-tolerant cultivar was stimulated at the lower Cd level, but was decreased at the higher Cd level when plants were treated with Cd for one week. However, Plant growth was severely inhibited at both Cd levels as stress duration lasted for up to three weeks. Plant growth of the Cd-sensitive cultivar was severely inhibited at both Cd levels irrespective of Cd stress duration. Addition of Si increased shoot and root biomass of both cultivars at both Cd levels and decreased Cd uptake and root-to-shoot transport. Superoxide dismutase, catalase and ascorbate peroxidase activities decreased, but malondialdehyde and H2O2 concentrations increased at the higher Cd level, which were counteracted by Si added. Ascorbic acid, glutathione and non-protein thiols concentrations increased at the higher Cd level, which were further intensified by addition of Si. The effects of Si and Cd on the antioxidant enzyme activity were further verified by isoenzyme analysis. Silicon was more effective in enhancing Cd tolerance in the Cd-tolerant cultivar than in the Cd-sensitive cultivar. It can be concluded that Si-enhanced Cd tolerance in B. chinensis is attributed mainly to Si-suppressed Cd uptake and root-to-shoot Cd transport and Si-enhanced antioxidant defense activity.

Abstract:The objective of this study was to assess the effect of different Cd and Si concentrations on the maize plants. The following Cd and/or Si treatments were used: 5 Cd; 10 Cd; 100 Cd; 5 Cd + 0.08 Si; 10 Cd + 0.08 Si; 100 Cd + 5 Si treatments (Cd concentration in μM, Si concentration in mM). The plant growth, photosynthetic pigments content, antioxidant enzymes activities (POX, SOD, CAT), Cd and Si accumulation, translocation and cell wall deposition of the maize plants was observed. Changes in the endodermal cell walls development and late metaxylem elements lignification due to Cd and/or Si treatment were also evaluated. The negative effect of Cd (5 and 10 μM) on the growth parameters was alleviated by Si at 0.08 mM. The positive effect of Si was not observed at higher Cd and Si concentrations. This indicates that the alleviating effect of Si on Cd toxicity depends on the Cd and Si concentrations. Plants responded to Cd toxicity by an increase of antioxidant enzyme activity. Silicon addition in Cd + Si treatment stimulated an increase in the activity of antioxidant enzymes in comparison with the Cd treatment. Chlorophyll and carotenoid content in the Cd treated plants was not significantly affected by Si. The young maize plants retained much more Cd in their roots as they translocated into the shoots. 5 Cd + 0.08 Si and 10 Cd + 0.08 Si treatments correlated with an increase in Cd concentration in the roots and shoots, and in the cell walls. Silicon caused a slight decrease of the Cd translocation into the shoots in 5 Cd + 0.08 Si and 10 Cd + 0.08 Si treatments. Negative correlation between the root Cd cell wall deposition and Cd translocation was observed. Cadmium and/or Si altered root anatomy. Cadmium enhanced suberin lamellae development and late metaxylem lignification; silicon in Cd + Si treatments accelerated suberin lamellae deposition and enhanced the tertiary endodermal cell walls formation in comparison with Cd treatments. Negative correlation between the endodermal cell walls development and Cd translocation was observed.

Abstract:Soil degradation with different stress conditions like accumulation of cadmium (Cd) contents in soil and drought stress has become one of the most dangerous issues that obstruct the sustainable agriculture production. Silicon nanoparticles (Si NPs) play beneficial roles in combating various biotic and abiotic stresses but their role under combined metal and drought stress is not studied. A pot study was designed to determine the effect of Si NPs on wheat (Triticum aestivum L.) growth and uptake of Cd grown in Cd contaminated soil with different water levels under ambient conditions. Four different levels of Si NPs (0, 25, 50, and 100 mg/kg) were applied in the soil before 1 week of wheat sowing and two water levels (70% and 35% soil water-holding capacity) were introduced after 50 days of seed sowing for the remaining growth period. The lowest biomass, yield, and photosynthesis were observed in the control plants while oxidative stress and the highest Cd concentrations in shoots, roots, and grains were observed in the control plants, and the drought stress further enhanced this effect on the plants. The Si NPs treatments improved the plant growth indicators and photosynthesis, and reduced the Cd concentrations in wheat tissues, especially in grains either without or with drought stress. The Si NPs reduced the oxidative stress in leaves as was indicated by the reduced production of hydrogen peroxide, electrolyte leakage, and malondialdehyde contents, and increase in superoxide dismutase and peroxidase activities. The improvement in wheat growth and a reduction in oxidative stress and Cd concentration in tissues were dependent on the levels of Si NPs and the effect was the highest with the highest level of NPs used.

Abstract:Nano-enabled strategies have emerged as promising alternatives to resolve heavy metals (HMs) related harms in an eco-friendly manner. Here, we explored the potential of biogenic silicon nanoparticles (SiNPs) in alleviating cadmium (Cd) stress in rapeseed (Brassica napus L.) plants by modulating cellular oxidative repair mechanisms. Biogenic SiNPs of spherical shapes with size ranging between 14 nm and 35 nm were synthesized using rice straw extract and characterized through advanced characterization techniques. A greenhouse experiment results showed that SiNPs treatment at 250 mg kg−1 significantly improved growth parameters, including fresh weight (33.3 %) and dry weight (32.6 %) of rapeseed plants than Cd-treated control group. Photosynthesis and leaf gas exchange parameters were also positively influenced by SiNPs treatment, indicating enhanced photosynthetic efficiency. Additionally, SiNPs treatment at 250 mg kg−1 increased the activities of antioxidant enzymes such as superoxide dismutase (19.1 %), peroxidase (33.4 %), catalase (14.4 %), and ascorbate peroxidase (33.8 %), which may play a crucial role in ROS scavenging and reduction in Cd-induced oxidative stress. TEM analysis revealed that SiNPs treatment effectively mitigated Cd-induced damage to leaf ultrastructure, while qPCR analysis showed that SiNPs treatment changed the expressions of the antioxidant defense and stress related genes. Moreover, SiNPs treatment significantly influenced the Cd accumulation and Si contents in plants. Overall, our findings revealed that biogenic SiNPs have great potential to serve as a sustainable, eco-friendly, and non-toxic alternative for the remediation of Cd toxicity in rapeseed plants.

Abstract:Abstract Background and aims Cadmium (Cd) contamination is a serious threat to plants and humans. Silicon (Si) was reported to have some alleviative effects on Cd stress in plants. However, whether Si alleviates Cd toxicity in maize genotypes with contrasting root system size are unknown. Methods Effects of Si application (200 mg kg −1 soil) on shoot and root growth, Cd uptake and transportation under Cd stress (20 mg kg −1 soil) were assessed at the silking and maturity stages of maize genotypes Zhongke11 (deep-rooted) and Shengrui999 (shallow-rooted) in a pot experiment. Results Application of Si significantly increased root dry weight, plant height and root length. Root volume and average root diameter were significantly positively correlated with root Cd concentration, bioaccumulation and translocation factor, respectively, of two maize genotypes at the silking stage. Addition of Si significantly increased Cd concentration, content, bioconcentration and translocation factor in roots of Zhongke11, but reduced the values of these parameters in Shengrui9999 at both growth stages. Grain Cd concentration in the combined Cd and Si treatment was decreased by 14.4% (Zhongke11) and 21.4% (Shengrui999) than that in Cd treatment. Grain yield was significantly negatively correlated with root Cd accumulation. Moreover, addition of Si significantly reduced Cd daily intake and health risk index in maize. Conclusions This study demonstrated that addition of Si reduced health risk by eliminating Cd accumulation in maize shoot and grain, and alleviated Cd stress with more profound effects in the shallow-rooted genotype Shengrui999.

Abstract:Soil contamination by cadmium (Cd) is an increasing environmental concern that potentially jeopardizes both crop productivity and human health. Silicon (Si), the Earth's second most abundant element, has shown a significant potential in reducing Cd uptake by crops. However, there is still a lack of quantitative data on the beneficial effects of Si in reducing Cd toxicity, thereby making it more difficult to ensure safe crop production. We conducted a comprehensive meta-analysis of 105 studies to assess the impact of exogenous Si on Cd accumulation in three major cereal crops (wheat, maize, and rice) and elucidate the key factors governing the Si effects. We found that Si supplementation significantly mitigated Cd toxicity in crops, reducing Cd accumulation in maize, rice, and wheat by 37 %, 30 %, and 45 %, respectively. This reduction was most pronounced in all three crop grains (reductions reaching 40–51 %). The four different forms of Si applied all increased crop yield, with nano-silicon resulting in an average yield increase of 19 %, surpassing silicate, Si-based fertilizer, and other silica-based materials. The effects of Si were primarily influenced by application rate and methods, soil pH, Cd concentration, and the effects of foliar and field application. Based on Cd inhibition levels and overall economic benefits, we recommend a Si application rate of ≤ 250 mg/kg, preferably using nano-silicon or silicate. Overall, our study provides valuable globally relevant guidance on Si amendments selection and application thereby ensuring safer and higher crop production in Cd-contaminated soils.
