Abstract:Understanding of the influence of nanotextured surface topographies on surface-enhanced Raman scattering (SERS) is still rather limited, even after intensive research in recent years; this is mainly due to the absence of a synthesis system to control the surface morphologies on metal meso-/nanoparticles. Here, a facile particle-mediated aggregation protocol was reported to synthesize a series of uniform gold mesoparticles with a nanotextured rough surface. The surface topographies can be precisely controlled by varying the temperatures and concentrations of dopa and Au ions. For example, the tips can be well changed from half-sphere to spike, and then to blade, and the size of the tips also can be tuned, such that the thickness of the blade-like tips can be artificially controlled from 9 to 14 nm. Single-particle SERS analysis reveals that the uniform Au mesoparticles with highly nanotextured topography have enhancement factors greater than 1.0 x 10(8) with narrow distribution. The effect of the precisely controlled mesoparticle topographies on SERS properties was deeply studied using Raman analysis and finite-difference time-domain (FDTD) simulation. The result indicated that a strong electric field will be induced at the tips and slits on the flower-like mesoparticle surface by the confined light in the cavities between the petals. As such, the current protocol opens up avenues for the fabrication of structurally reproducible mesosuperstructure-based SERS sensors.
Monodispersed mesoscopic star-shaped gold particles via silver-ion-assisted multi-directional growth for highly sensitive SERS-active substrates

Abstract:Surface-enhanced Raman scattering (SERS) exploits localized surface plasmon resonances in metallic nanostructures to significantly amplify Raman signals and perform ultrasensitive analyses. A critical factor for SERS-based analysis systems is the formation of numerous electromagnetic hot spots within the nanostructures, which represent regions with highly concentrated fields emerging from excited localized surface plasmons. These intense hotspot fields can amplify the Raman signal by several orders of magnitude, facilitating analyte detection at extremely low concentrations and highly sensitive molecular identification at the single-nanoparticle level. In this study, mesoscopic star-shaped gold particles (gold mesostars) were synthesized using a three-step seed-mediated growth approach coupled with the addition of silver ions. Our study confirms the successful synthesis of gold mesostars with numerous sharp tips via the multi-directional growth effect induced by the underpotential deposition of silver adatoms (AgUPD) onto the gold surfaces. The AgUPD process affects the nanocrystal growth kinetics of the noble metal and its morphological evolution, thereby leading to intricate nanostructures with high-index facets and protruding tips or branches. Mesoscopic gold particles with a distinctive star-like morphology featuring multiple sharp projections from the central core were synthesized by exploiting this phenomenon. Sharp tips of the gold mesostars facilitate intense localized electromagnetic fields, which result in strong SERS enhancements at the single-particle level. Electromagnetic fields can be further enhanced by interparticle hot spots in addition to the intraparticle local field enhancements when arranged in multilayered arrays on substrates, rendering these arrays as highly efficient SERS-active substrates with improved sensitivity. Evaluation using Raman-tagged analytes revealed a higher SERS signal intensity compared to that of individual mesostars because of interparticle hot spots enhancements. These substrates enabled analyte detection at a concentration of 10- 9 M, demonstrating their remarkable sensitivity for trace analysis applications.

Abstract:Ordered porous gold/titanium dioxide (Au/TiO2) hybrid nanostructured films are specifically interesting in large-scale applications using localized surface plasmon resonances (LSPRs) and surface-enhanced Raman scattering (SERS). Deposition of Au nanoparticles via sputter deposition is one of the promising technologies to establish optically active sites at low cost in combination with nanostructured TiO2 films. In this work, we investigate the optical response of sputter-deposited Au/TiO2 nanohybrid thin films with a focus on the plasmonic response and application as molecular sensors. The LSPR peak red shifts with an increasing thickness of deposited Au. The Raman intensity of deposited molecules, probed with rhodamine 6G (R6G), depends on the deposited gold thickness. It has its maximum at an effective Au thickness of 3.4 nm. To elucidate the origin of this behavior, we apply in situ grazing-incidence small-angle X-ray scattering (GISAXS) to investigate the growth kinetics of Au on a TiO2 template during sputter deposition. On the basis of time-resolved GISAXS, the growth characteristics of sputter-deposited Au on a TiO2 template with a final effective Au layer thickness around the percolation threshold is described with the well-known four-stage model of nucleation and cluster formation, diffusion-mediated growth, adsorption-mediated growth, and grain growth. The maximum in SERS intensity is corroborated by the existence and optimal size of hot spots in the narrow space occurring between the sputter-deposited Au clusters, on staying below the percolation threshold. On the basis of the growth laws extracted, we give a guideline for tailoring the ordered porous Au/TiO2 nanohybrid thin films for SERS sensor applications.

Abstract:The fabrication and optical characterization of self-assembled arrangements of rough gold nanoparticles with a high area coverage and narrow gaps for surface-enhanced Raman spectroscopy (SERS) are reported. A combination of micellar nanolithography and electroless deposition (ED) enables the tuning of the spacing and size of the noble metal nanoparticles. Long-range ordered quasi-hexagonal arrays of gold nanoparticles on silicon substrates with a variation of the particle sizes from about 20 nm to 120 nm are demonstrated. By increasing the particle sizes for the homogeneously spaced particles, a large number of narrow gaps is created, which together with the rough surface of the particles induces a high density of intense hotspots. This makes the surfaces interesting for future applications in near-field-enhanced bio-analytics of molecules. SERS was demonstrated by measuring Raman spectra of 4-MBA on the gold nanoparticles. It was verified that a smaller inter-particle distance leads to an increased SERS signal.

Abstract:There has been great hope for ultrasensitive detection methods with the application of Surface Enhanced Raman Spectroscopy (SERS) because of its impressive advancements. Nevertheless, a major challenge is the creation of large-scale, controllable, and reproducible substrates with high SERS activity [1]. As a basis for SERS detection, we report here the fabrication of different types of aluminum-based and polymer-based SERS platforms with the synthesis of gold nanoparticle (AuNP) arrays on them. The tunability of the AuNPs size and distribution is achieved by methodically controlling the aluminum substrates nanostructuration and the subsequent processes for the synthesis of the AuNP. This research presents a scalable and dependable approach for creating ordered Au patterns on nanostructured aluminum and polymer substrates. For both aluminum-based and polymer-based platforms, the initial formation of nanoporous anodic alumina (NAA) on an aluminum substrate by electrochemical anodization [2] is essential. The electrolytes used (sulphuric, oxalic, or phosphoric acid) and its respective concentration determines the ideal voltage for anodization and the electrolyte temperature [3,4]. The formed NAA layer is chemically removed obtaining a nanostructured aluminum substrate with a hexagonally ordered distribution of nanoconcavities [5,6]. The distance between nanocavities ranges from tens to hundreds of nanometers depending on the electrolyte used [7]. The polymer-based nanostructured substrates are formed with the deposition of the selected polymer on the obtained aluminum nanoconcavities substrates and peeled off when dried. Instead of nanoconcavities, it presents a distribution of nanosemispheres that corresponds to the imprinting of the aluminum nanoconcavities on the polymer. The final SERS platforms are obtained with the gold sputtering on the nanoconcavities aluminum substrates and the polymer substrates, followed by a thermal annealing process [8]. The result is a scalable, ordered and controlled distribution of AuNPs on the nanostructured substrates (Fig. 1). An evaluation of the sputtering time and the thermal annealing paramenters (time and temperature) is presented. Over a large surface area of several square centimeters, the AuNPs size, shape, and interparticle spacing may be controlled by using the flexible manufacturing method discussed in this work. Abstract:Surface-enhanced Raman spectroscopy (SERS) substrates are of utmost interest in the analyte detection of biological and chemical diagnostics. This is primarily due to the ability of SERS to sensitively measure analytes present in localized hot spots of the SERS nanostructures. In this work, we present the formation of 67 ± 6 nm diameter gold nanoparticles supported by vertically aligned shell-insulated silicon nanocones for ultralow variance SERS. The nanoparticles are obtained through discrete rotation glancing angle deposition of gold in an e-beam evaporating system. The morphology is assessed through focused ion beam tomography, energy-dispersive X-ray spectroscopy, and scanning electron microscopy. The optical properties are discussed and evaluated through reflectance measurements and finite-difference time-domain simulations. Lastly, the SERS activity is measured by benzenethiol functionalization and subsequent Raman spectroscopy in the surface scanning mode. We report a homogeneous analytical enhancement factor of 2.2 ± 0.1 × 107 (99% confidence interval for N = 400 grid spots) and made a comparison to other lithographically derived assemblies used in SERS. The strikingly low variance (4%) of our substrates facilitates its use for many potential SERS applications.

Abstract:Surface-enhanced Raman scattering (SERS) enhancement and the reproducibility of the SERS signal strongly reflect the quality and nature of the SERS substrates because of diverse localized surface plasmon resonance (LSPR) excitations excited at interstitials or sharp edges. LSPR excitations are the most important ingredients for achieving huge enhancements in the SERS process. In this report, we introduce several gold and silver nanoparticle-based SERS-active substrates developed solely by us and use these substrates to investigate the influence of LSPR excitations on SERS. SERS-active gold substrates were fabricated by immobilizing colloidal gold nanoparticles on glass slides without using any surfactants or electrolytes, whereas most of the SERS-active substrates that use colloidal gold/silver nanoparticles are not free of surfactant. Isolated aggregates, chain-like elongated aggregates and two-dimensional (2D) nanostructures were found to consist mostly of monolayers rather than agglomerations. With reference to correlated LSPR and SERS, combined experiments were carried out on a single platform at the same spatial position. The isolated aggregates mostly show a broadened and shifted SPR peak, whereas a weak blue-shifted peak is observed near 430 nm in addition to broadened peaks centered at 635 and 720 nm in the red spectral region in the chain-like elongated aggregates. In the case of 2D nanostructures, several SPR peaks are observed in diverse frequency regions. The characteristics of LSPR and SERS for the same gold nanoaggregates lead to a good correlation between SPR and SERS images. The elongated gold nanostructures show a higher enhancement of the Raman signal than the the isolated and 2D samples. In the case of SERS-active silver substrates for protein detection, a new approach has been adopted, in contrast to the conventional fabrication method. Colloidal silver nanoparticles are immobilized on the protein functionalized glass slides, and further SERS measurements are carried out based on LSPR excitations. A new strategy for the detection of biomolecules, particularly glutathione, under aqueous conditions is proposed. Finally, supramolecular J-aggregates of ionic dyes incorporated with silver colloidal aggregates are characterized by SERS measurements and correlated to finite-difference time-domain analysis with reference to LSPR excitations.FigureSPR and SERS images for isolated, elongated and two-dimensional gold nanostructures

Abstract:We report an effective surface-enhanced Raman scattering (SERS) substrate enabled by synthesis of gold nanobipyramids (Au NBPs) in a microfluidic chip. Improved seed-mediated method was implemented for consecutive synthesis of Au NBPs in an S-shaped micromixer. Under precise control of flow rates of reactants, Au NBPs with various morphologies can be prepared in microchannels to serve as effective SERS substrates. With the assistance of this substrate, Raman signals of rhodamine 6G (R6G) probe molecules were significantly enhanced with high sensitivity and reproducibility, the dependence between SERS spectra and Au NBPs morphologies was investigated. Owing to the control mechanism involved in the reaction process in microchannels, these flexible, stable and high-performance SERS substrates can have potential application in diverse fields, particularly in biosensing areas.

Abstract:Surface-enhanced Raman scattering (SERS) probes based on gold nanoparticles modifying the core of the optical fiber were made by a surfactantless photochemical deposition method. The growth kinetics and shape evolution of gold nanoparticles depending on different experimental conditions were studied. It was found that, under the condition of detectable gold nanoparticle deposition, increasing the concentration of chloroauric acid (HAuCl(4)) was not conducive to the deposition whereas increasing the concentration of sodium citrate (Na(3)Ct) would speed up the deposition. By controlling the concentration of the reaction solution and irradiation time, we obtained fused spherical-like, spherical, and flowerlike gold nanoparticles. To test the SERS activity of the probes, the SERS spectra of a rhodamine 6G aqueous solution were recorded in direct detection mode and remote mode. We have also developed a new approach to improving the SERS sensitivity when detecting in remote mode.
Sers-Encoded Nanogapped Plasmonic Nanoparticles: Growth of Metallic Nanoshell by Templating Redox-Active Polymer Brushes

Abstract:We report a new strategy to synthesize core-shell metal nanoparticles with an interior, Raman tag-encoded nanogap by taking advantage of nanoparticle-templated self-assembly of amphiphilic block copolymers and localized metal precursor reduction by redox-active polymer brushes. Of particular interest for surface-enhanced Raman scattering (SERS) is that the nanogap size can be tailored flexibly, with the sub-2 nm nanogap leading to the highest SERS enhancement. Our results have further demonstrated that surface functionalization of the nanogapped Au nanoparticles with aptamer targeting ligands allows for specific recognition and ultrasensitive detection of cancer cells. The general applicability of this new synthetic strategy, coupled with recent advances in controlled wet-chemical synthesis of functional nanocrystals, opens new avenues to multifunctional core-shell nanoparticles with integrated optical, electronic, and magnetic properties.
Mesoporous Nanostructures Encapsulated with Metallic Nanodots for Smart SERS Sensing

Abstract:In virtue of uniform mesopores and core–shell nanoarchitectures, metallic nanodot-encapsulated hollow mesoporous nanostructures have shown promising potential in various applications. However, their fabrication with versatile tunability of the encapsulated metallic content has been a challenge. Herein, we have prepared metallic nanodot-encapsulated hollow mesoporous silica nanoparticles (M-HMSNPs) with adjustable inner metallic components. The sacrificial template of polystyrene (PS) nanoparticles precoated with metals (Au/Ag/Pt) is fully wrapped with mesoporous silica (mSiO2). The metallic nanodots are formed during the template removal process by calcination. The type and content of the encapsulated nanodots can be readily and precisely controlled by the initially deposited metallic layers. We demonstrate the application of the gold (Au) nanodot-loaded HMSNPs (denoted Au-HMSNPs) as smart surface-enhanced Raman spectroscopy (SERS) probes, which can screen between big molecules and small analytes. With the aid of a Raman reporter, the SERS probe can successfully quantify H2O2, which is used to distinguish cancer cells in vitro. Further integrated with enzymes, the SERS chips of specificity are prepared and used to detect corresponding substrates of glucose and uric acid, responsively. Cubic Silver Nanoparticles Fixed on TiO2 Nanotubes as Simple and Efficient Substrates for Surface Enhanced Raman Scattering

Abstract:In this work we show that ordered freestanding titanium oxide nanotubes (TiO2 NT) may be used as substrates for the simple and efficient immobilization of anisotropic plasmonic nanoparticles. This is important because anisotropic plasmonic nanostructures usually give greater spectral enhancement than spherical nanoparticles. The size of the pores in a layer of titanium oxide nanotubes can be easily fitted to the size of many silver plasmonic nanoparticles highly active in SERS (surface-enhanced Raman scattering) spectroscopy (for example, silver nanocubes with an edge length of ca. 45 nm), and hence, the plasmonic nanoparticles deposited can be strongly anchored in such a titanium oxide substrate. The tubular morphology of the TiO2 substrate used allows a specific arrangement of the silver plasmonic nanoparticles that may create many so-called SERS hot spots. The SERS activity of a layer of cubic Ag nanoparticles (AgCNPs) deposited on a tubular TiO2 substrate (AgCNPs@TiO2 NT) is about eight times higher than that of the standard electrochemically nanostructured surface of a silver electrode (produced by oxidation reduction cycling). Furthermore, a super hydrophilic character of the TiO2 nanotubes surface allows for a uniform distribution of AgCNPs, which are deposited from an aqueous suspension. The new AgCNPs@TiO2 NT hybrid layer ensures a good reproducibility of SERS measurements and exhibits a higher temporal stability of the achievable total SERS enhancement factor—one that is far better than standard SERS silver substrates. Abstract:Non-spherical nanostructures such as multilayered polygons and branched Au nanoparticles demonstrate high Surface-enhanced Raman scattering (SERS) performance due to their plasmonic nature and anisotropic morphology. Unfortunately, their syntheses often involve multiple steps and complex reagents. In particular, a conventional synthesis of Au nanoplatelets (AuNPt) involves a toxic cationic surfactant that should be substituted with more ecologically friendly reagents. Herein, we demonstrate the synthesis of AuNPt utilizing organic biomolecules from the plant Cercis Canadensis . These SERS-capable AuNPt are also shown to undergo a distinctive structural modification through plasma-dependent etching of the nanoparticle. This etching allows, in high yield, the {111} facets to develop surface undulations and perforations through the entire structure that increases anisotropy. The AuNPt are demonstrated to be surfactant-free through the absence of hydrocarbons in the IR spectra and EDX measurements. The nanoplatelets displayed a 17.142 ± 1.193 × Raman to SERS peak enhancement, 4133% increase of the area under the curve, and a 1580% increase in the FWHM in the fingerprint 1341 cm −1 peak of p-nitrothiophenol. Measurements were done in liquid- and solid-phase to demonstrate the versatility of the AuNPts.

Abstract:Highly sensitive Raman signal enhancement of target molecules is achieved by developing effective surface-enhanced Raman spectroscopy (SERS) sensing platforms created by a unique combination of plasmonic gold nanofractals and highly ordered nanostructured aluminum arrays. Under efficient laser illumination, the SERS-active substrates formed by Al nanoconcavities of less than 100 nm, each decorated with a single Au nanoparticle, trigger localised surface plasmon resonance. 4-Mercaptopyridine (4-Mpy) successfully interferes on the surface of Au nanofractals, proving the method's utility. As a result, the SERS signal from the 4-MPy adsorbed on Au nanostructures is long-range electromagnetically amplified. The designed and developed substrate brings about a high enhancement factor (EF), over 10 7 increase in intensity when approaching 4-Mpy adsorbed on Au nanostructures. A considerable increase in the SERS signal is found when the excitation origin is determined to be vibrant with the charge transfer (CT) state formed at the interface of the Al nanoconcavities and organic molecules. This approach, which does not need nanopatterning, provides an alternative strategy for conducting SERS investigations on molecules with remarkably weak Raman signals.

Abstract:Formation of plasmonic structure in closely packed assemblies of metallic nanoparticles (NPs) is essential for various applications in sensing, renewable energy, authentication, catalysis, and metamaterials. Herein, a surface-enhanced Raman scattering (SERS) substrate is fabricated for trace detection with ultrahigh sensitivity and stability. The SERS substrate is constructed from a simple yet robust strategy through in situ growth patterned assemblies of Au NPs based on a polymer brush templated synthesis strategy. Benefiting from the dense and uniform distribution of Au NPs, the resulting Au plasmonic nanostructure demonstrates a very strong SERS effect, while the outer polymer brush could restrict the excessive growth of Au NPs and the patterned design could achieve uniform distribution of Au NPs. As results, an ultra-low limit of detection (LOD) of 10−15 M, which has never been successfully detected in other work, is determined for 4-acetamidothiophenol (4-AMTP) molecules and the Raman signals in the random region show good signal homogeneity with a low relative standard deviation (RSD) of 7.2%, indicating great sensitivity and reliability as a SERS substrate. The LOD values of such Au plasmonic nanostructures for methylene blue, thiram, and R6G molecules can also reach as low as 10−10 M, further indicating that the substrate has a wide range of applicability for SERS detection. With the help of finite difference time domain simulations (FDTD) calculation, the electric field distribution of the Au plasmonic nanostructures is simulated, which quantitatively matches the experimental observations. Moreover, the Au plasmonic nanostructures show good shelf stability for at least 10 months of storage in an ambient environment, indicating potentials for practical applications.

Abstract:A quasi-periodic array of 3D gold-nanoparticle-capped SiO2 microspheres (Au@SiO2) was designed and prepared with a facile approach to enhance the Raman signal intensity of adsorbed biomolecules. Through adjusting the thickness and annealing of Au thin films initially deposited on arrays of self-assembled SiO2 microspheres, we were able to control the diameter of Au nanoparticles and their interparticle spacing to produce two types of plasmonic near-field hot spots, locating at the gaps of such densely arranged Au nanoparticles on individual SiO2 microspheres and in the gap regions of neighboring SiO2 microspheres, respectively. Such double near-field enhancement mechanism leads to a surface-enhanced Raman scattering (SERS) enhancement factor up to 3×106 for Rhodamine 6G molecules. The SERS signal intensity was highly uniform with a relative standard deviation of 4.5%. This 3D SERS substrate has significant potential for various applications in the field of SERS detection of analytes and wearable biosensing.

Abstract:This report describes the fabrication and application of the polystyrene (PS) core/metal (gold and silver) shell composites. The PS core microspheres were functionalized with terminal amino groups which endowed the PS microspheres with positively charged surface, and then seeded with negatively charged gold nanoparticles (NPs) via electrostatic force. The gold-seeded PS microspheres were coated completely with gold (silver) shell via reduction of the appropriate metal ions. The morphology of gold (silver) shell composed of numerous close-packed gold (silver) NPs could be tailored easily by altering amounts of metallic precursor. The as-prepared PS core/metal shell composites can be used as substrates for surface-enhanced Raman spectroscopy (SERS). They exhibited excellent surface enhancement ability when 4-aminothiophenol (4-ATP) was used as a molecule probe.

Optimizing Surface-Enhanced Raman Scattering by Template Guided Assembling of Closely Spaced Silver Nanocluster Arrays

Abstract:We present an easy approach to synthesize closely spaced regular arrays of silver nanoclusters, which are self-assembled by depositing gas-phase synthesized metal nanoclusters onto pre-patterned triblock copolymer templates. The array has a high particle density of about 2 ×103 particles per μm2, and an average interparticle space of about 20 nm. The surface plasmon resonance wavelength of the array is tuned due to the interparticle plasmon coupling. High SERS sensitivity for less than one layer trans-1,2-bi-(4-pyridyl) ethylene (BPE) molecule detection, with an enhancement factor of 2.6 ×106, has been demonstrated for a substrate with this array. The enhanced Raman signal was found to be 5 times higher than that measured from the substrate with randomly distributed silver nanoparticles.

Abstract:The fabrication technology of surface nanocomposites based on hexagonally ordered gold nanoparticle (AuNP) layers (quasi-arrays) and their possible application as surface-enhanced Raman spectroscopy (SERS) substrates are presented in this paper. The nanoparticle layers are prepared using a nanotextured template formed by porous anodic alumina (PAA) and combined with gold thin-film deposition and subsequent solid-state dewetting. Three types of hexagonal arrangements were prepared with different D/D0 values (where D is the interparticle gap, and D0 is the diameter of the ellipsoidal particles) on a large surface area (~cm2 range), namely, 0.65 ± 0.12, 0.33 ± 0.10 and 0.21 ± 0.09. The transfer of the particle arrangements to transparent substrates was optimized through three generations, and the advantages and disadvantages of each transfer technology are discussed in detail. Such densely packed nanoparticle arrangements with high hot-spot density and tunable interparticle gaps are very beneficial for SERS applications, as demonstrated with two practical examples. The substrate-based enhancement factor of the nanocomposites was determined experimentally using a DNA monolayer and was found to be between 4 × 104 and 2 × 106 for the different particle arrangements. We also determined the sensing characteristics of a small dye molecule, rhodamine 6G (R6G). By optimizing the experimental conditions (e.g., optimizing the laser power and the refractive index of the measurement medium with an ethylene-glycol/water mixture), concentrations as low as 10−16 M could be detected at 633 nm excitation.

Abstract:Owing to its excellent localized surface plasmon resonance (LSPR) effect, noble metal nanoparticles (NPs) find extensive application in the preparation of surface-enhanced Raman scattering (SERS) substrates. However, due to process limitations, the practicality and testing effectiveness of SERS substrates still leaves much to be desired. Here, a wafer-scale gold (Au) NPs silicon (Si) micro/nano-hybrid structured substrate is prepared. This is achieved through two-step etching, followed by decorating Au-NPs onto the structure via self-assembly process induced by de-wetting. Finite-difference time-domain (FDTD) simulations reveal that the significant enhancement of local electric field in the voids between Au-NPs and Si is crucial for enhancing SERS. Using Rhodamine 6G (R6G) as the probe molecule, performance of the fabricated SERS substrate is investigated. It demonstrates a minimum detection limit of 10 −11 M, with a calculated enhancement factor of 4.40 × 10 8 , indicating its high sensitivity. The minimum relative standard deviation for the substrate is 5.254 %. After 20 days of placement, the SERS performance show tiny variation. Even after four cycles of cleaning experiments, it still maintains outstanding SERS performance. This demonstrates its excellent stability, uniformity and reusability. Our research provides guidance for the efficient and low-cost fabrication of high-performance SERS substrates.
