Abstract:One of the fundamental goals of bio-ethanol production is to find the alternative of traditional land-relied feedstock. A kind of macroalgae, Sargassum horneri (S. horneri) was chosen as the bio-source for ethanol production in the present investigation. The bioethanol was achieved by acid-pretreated, hydrolysis, and semi-simultaneous saccharification and fermentation (S-SSF) at small pilot scale. Immobilization of Pichia stipites in calcium alginate was adapted in the S-SSF process. Furthermore, effects of pre-hydrolysis time, cellulase loading, fermentation temperature and fermentation time on the concentration, and yield of bio-ethanol were evaluated by using response surface methodology (RSM). A maximum bioethanol concentration (2.89 g/L), and maximum bioethanol yield (0.11 g/g raw material) at cellulase loading (10 IU/g raw material), pre-hydrolysis time (53 min), SSF temperature (32 °C) and SSF time (14 h) were observed. The optimized bioethanol production data series fit the Gompertz model and modified Logistic model with an R2 value of 0.997. The obtained Gompertz and modified Logistic coefficients indicated that S. horneri can serve as an efficient substrate for bioethanol production. The fermentation residues were used for typical dyeing contamination adsorption, the existence of the residual lignin and exposure functional groups made it an effective adsorbent, which followed the pseudo-second-order kinetics. These results are helpful for the scale-up development of lignocellulosic macroalgae based bioethanol production and fermentation residue use.
Fermentative Production of 2,3-butanediol from Corn Stover Hydrolysate

Abstract:2,3-butanediol is a valuable chemical feedstock and liquid fuel.Biological production of 2,3-butanediol from low-cost cellulosic material is of both economic and social benefits.Fermentation of 2,3-butanediol from corn stover hydrolysate by Klebsiella oxytoca ZU-03 was studied,during which both hexose and pentose in the hydrolysate were fully used as substrate.The results show that sodium sulfate concentration under 20 g/L in corn stover hydrolysate has no significant effects on fermentation;acetic acid and ethanol,as the main byproducts,affected the fermentation when their concentrations were higher than 20 and 5 g/L respectively.The optimal initial pH is 6.0-6.5 and the initial total sugar concentration is 80-100 g/L.When the total sugar concentration is 80 g/L(glucose 47.25 g/L,xylose 32.75 g/L,the cultures were shaking at 30 ℃ for 60 h with initial pH 6.0,a 2,3-butanediol concentration of 37.20 g/L was achieved and 99.36 % of the total sugar was consumed with the yield of 0.468 g/g(total sugar),which reached 94 % of the theoretical maximum.This result may be very useful in enhancing the efficient usage of cellulosic biomass.
Enzymatic Hydrolysis of Corncob and Ethanol Production from Cellulosic Hydrolysate

Abstract:Enzymatic hydrolysis of corncob and ethanol fermentation from cellulosic hydrolysate were investigated. After corncob was pretreated by 1% H2SO4 at 108°C for 3h, the cellulosic residue was hydrolyzed by cellulase from Trichoderma reesei ZU-02 and the hydrolysis yield was 67.5%. Poor cellobiase activity in T. reesei cellulase restricted the conversion of cellobiose to glucose, and the accumulation of cellobiose caused severe feedback inhibition to the activities of β-1,4-endoglucanase and β-1,4-exoglucanase in cellulase system. Supplementing cellobiase from Aspergillus niger ZU-07 greatly reduced the inhibitory effect caused by cellobiose, and the hydrolysis yield was improved to 83.9% with enhanced cellobiase activity of 6.5CBUg−1 substrate. Fed-batch hydrolysis process was started with a batch hydrolysis containing 100gl−1 substrate, with cellulosic residue added at 6 and 12h twice to get a final substrate concentration of 200gl−1. After 60h of reaction, the reducing sugar concentration reached 116.3gl−1 with a hydrolysis yield of 79.5%. Further fermentation of cellulosic hydrolysate containing 95.3gl−1 glucose was performed using Saccharomyces cerevisiae 316, and 45.7gl−1 ethanol was obtained within 18h. The research results are meaningful in fuel ethanol production from agricultural residue instead of grain starch.
Ethanol Production from Lycoris Radiata Herbert (amarylllidaceae) Residues As A New Resource

Abstract:The large quantities of Lycoris radiata Herbert (Amarylllidaceae) residues, left after the extraction of alkaloids from the bulbs, could threaten the environment if not properly disposed. Therefore, the aim of this research is to investigate the feasibility of bioconversion of L. radiata Herbert residues to ethanol through batch fermentation. In L. radiata Herbert residues, the average contents (g kg(-1)) of non-structural carbohydrates, crude fiber, crude protein, ash, and lignin are 485.1, 177.3, 124.7, 108.9, and 91.0, respectively. Five commercial enzymes (beta-glucanase, pectinase, xylanase, phytase and cellulase) were employed in pretreatment process and ethanol bioconversion was evaluated with three yeasts (Saccharomyces cerevisiae, osmotolerant S. cerevisiae and genetically engineered S. cerevisiae constructed to use cellobiose). The results showed that pretreament by beta-glucanase successfully facilitated the penetration of alpha-amylase into ground material. After residues pretreatment by 10 g kg (1) of beta-glucanase for 14 h at 50 degrees C, the viscosity decreased from 1135 to 59 Pas which was equivalent to that obtained by the combined whole five enzymes. Further experiments proved that osmotolerant S. cerevisiae was desirable for ethanol production from Lycoris radiate Herbert residues. Biovalorization potential of peels of Ananas cosmosus (L.) Merr. for ethanol production by Pichia stipitis NCIM 3498 & Pachysolen tannophilus MTCC 1077

Abstract:Bioethanol, is a potential alternate source of energy, renewable and safe. Ethanol production from value added food and feedstock has also not shown growth as estimated. Of late, the second generation processes of production of ethanol, such as from lignocellulosic biomass out of agricultural/domestic waste has been gaining considerable momentum. Here, we explored a new approach for optimizing the conditions of physiochemical pretreatment as well as fermentation process using peels of Ananas cosmosus as substrate and immobilized yeast Pachysolen tannophilus MTCC.1077 and Pichia stipitis NCIM 3498. We have also studied the influence of process variables such as incubation temperature, inoculum concentration and different nutrients on ethanol production. Pulverized peels of A. cosmosus recorded 25 ± 0.31% cellulose, 28 ± 0.18% hemicellulose and 8 ± 0.07% of lignin on dry solid (DS) basis. Peels of A. cosmosus delignified with 1% H2SO4 yielded 18.89% glucose, 38.81% xylose and 29.31% fructose under thermochemical pretreatment using autoclave (121 degrees C, 20 min.), with a hydrolytic efficiency of 75.52 ± 0.45%. FTIR spectroscopy results not only indicated the penetration of H2SO4 in the amorphous region of the biomass and degradation of hemicelluloses but also showed the structural differences before and after pretreatment. The enzymes required for hydrolysis were prepared from culture supernatants of Trichoderma reesei NCIM 1052 using wheat bran as carbon source under submerged fermentation conditions on rotatory shaker incubator (at 28 degrees C for 10 days). Enzyme activity (U/ml) of crude cellulase produced by T. reesei NCIM 1052 was 311.1 μmole/ml/min. Delignified A. cosmosus peel yielded 51.71 ± 0.44 g/l glucose when enzymatically hydrolysed by crude cellulase at the substrate enzyme ratio of 1:5. Simultaneous saccharification and fermentation (SSF) of peels of A. cosmosus by crude cellulase and separately entrapped Pichia stipitis NCIM 3498 (now known as Scheffersomyces stipitis) and Pachysolen tannophilus MTCC 1077 cells in calcium alginate beads were also investigated in the present study. The fermentation experiments were carried out at flask level. The processing parameters setup for reaching a maximum response for ethanol production was obtained when applying the optimum values for temperature (32 degrees C), inoculum level (6%) and fermentation medium (ammonium sulphate, KH2PO4, peptone and yeast extract) for P. tannophilus MTCC 1077 and temperature (30 degrees C), inoculum level (2%) and fermentation medium (ammonium sulphate, KH2PO4, peptone and yeast extract) for S. stipitis NCIM 3498. Maximum ethanol concentration 10.5 g/l and 10.9 g/l was obtained from P. tannophilus MTCC 1077 and S. stipitis NCIM 3498, respectively at the optimized process conditions in anaerobic batch fermentation.

Abstract:This study presents the characterization and application of different pretreatment possibilities to optimize simple carbohydrate production from representative feedstock, followed by ethanol production through optimized saccharification steps using suitable microorganisms. Total soluble carbohydrates and carbohydrate yield were evaluated using dilute acid hydrolysis of biomass and reference materials (starch and cellulose) under different conditions. To explore two-factor interactions, the Plackett-Burman design of experiments was used in the statistical screening of variables and then transformed to a full-factorial design based on the significant variables. According to the experimental findings, dilute acid hydrolysis can yield a considerable amount of soluble carbohydrates. The goodness of fit of the model was evaluated using the coefficient of determination. The optimized carbohydrate yields of the biomass were 0.76 g/g cassava peels, 0.35 g/g sugar beet pulp, and 0.36 g/g green macroalgae. Bioethanol production from optimized hydrolysis conditions using Saccharomyces cerevisiae and Scheffersomyces stipitis DSM 3651, led to a maximum ethanol yield of 0.286 g/g from cassava peels and 0.033 g/g from sugar beet pulp, while using Neurospora intermedia DSM 1265 led to a maximum ethanol yield of 0.076 g/g from green macroalgae. Hence, these biomasses are considered valuable feedstock for bioethanol production with cassava peels having the highest potential.

Abstract:Taro roots waste (Colocasia esculenta) was considered a starch material for the preparation of bioethanol. Conversion of starch into ethanol was conducted by enzymatic hydrolysis followed by fermentation. The hydrolysis process uses the catalyst of HCl 15% (1:10) at 100 oC for 2.5 hours and fermentation using Saccharomyces cerevisiae. The chemical composition was analyzed by using chromatography gas-mass spectrophotometry (GCMS). The active component of bioethanol was found as ethanol (41.14%) and glycerol (29.08 %). The other chemical compound were 2,3- dimethyloxirane, pentanal, propionic acid, and heptane-2-one. The fermentation using tape yeast resulted in the main component of glycerol (31.39%), cyclobutanol (15.26%), pentane (12.93%), and methyl 2-oxopropanoate (10.50 %). The taro roots waste were the potential to convert biomass into bioethanol, which is one of the environmentally friendly alternative fuels

Abstract:Microorganisms have an active role in biotechnology for example yeasts, especially in some genus like Saccharomyces, Pichia, and Candida. C.tropicalis one of the most important species of Candida and despite it is one of the causative agents of candidiasis but it has a major role in the production of many chemical compounds. C.tropicalis in the previous study was isolated from sheep dung and morphologically and molecularly classified the result of sequencing was elucidate 100% similarity between the studied isolate and other isolates inserted in DNA Data Bank of Japan DDBJ, physiologically this isolate tolerated 6% ethanol concentration in broth media with the ability to the production of 4% ethanol under optimum conditions including (YEPDB) medium, incubation temperature 30◦C, PH 6 and dextrose as a carbon source which give more productivity as a carbon source and optimum incubation period was three days . This isolate gives a positive result for the cellulase enzyme-producing test and an exciting feature to remove effluent because it completely adsorbed the Congo red dye from dye solation in a liquid growth medium.
A novel strategy for integrated utilization of Jerusalem artichoke stalk and tuber for production of 2,3-butanediol by Klebsiella pneumoniae

Abstract:Jerusalem artichoke stalk and tuber can serve as a low cost feedstock for the production of 2,3-butanediol. However, like other lignocellulosic materials, the sugar concentration in the hydrolysate prepared from Jerusalem artichoke stalk is too low to be utilized effectively by microorganisms. In this paper a novel strategy was proposed to increase the sugar concentration by adding Jerusalem artichoke tuber into the hydrolysate of the stalk. The sugar was then biotransformed into high-valued 2,3-butanediol by Klebsiella pneumoniae. Fed-batch simultaneous saccharification and fermentation (SSF) was effectively performed and 901.2 mmol/l (80.5 g/l) target products (2,3-butanediol plus acetoin) was obtained in 68 h by a stage-shift aeration strategy. The concentration, yield and productivity of target products were 16.9%, 16.8% and 23.4%, respectively, higher than the best results obtained with SSF operated under constant aeration. This showed that adding tuber to the stalk hydrolysate was a useful strategy for increasing the production of 2,3-butanediol from Jerusalem artichoke via fermentation.

Abstract:Cassava is one of the most widely produced agricultural products in Indonesia with cassava peel waste of 20%. Cassava peel contains carbohydrates and stores a high cellulose content so that it has a potential to be a bioethanol. This study aims to utilize cassava peel waste for bioethanol production with an alkaline pretreatment process, cellulase enzyme hydrolysis and fermentation by using Zymomonas mobilis bacteria. Alkaline pretreatment with 14% NaOH is used to hydrolyze lignocellulose. The hydrolysis optimization process enzymatically applies the Surface Response Method (RSM) to determine the optimum conditions at hydrolysis pH in the range of 2-10 and hydrolysis temperature in the range of 30-70 °C by analyzing glucose levels using the Dinitrosalicylic Acid (DNS) method and UVvis spectrophotometry instruments. Surface Response Method (RSM) is likewise implemented to decide the greatest conditions of the fermentation process. The pH measurement ranges 2-10, and fermentation time takes 6 to 168 hours. Based on the results of research, it results a lignin content of 6.68% (b/b), cellulose content of 58.4% (b/b), and hemicellulose content of 27.3% (b/b). The optimum conditions of the hydrolysis process obtained an optimum glucose level of 9.22mg/mL at pH 2 and a hydrolysis temperature of 50°C. The optimum conditions of the fermentation process use Zymomonas mobilis at pH 6 while fermentation time takes 168 hours analyzed using a refractometer produced a bioethanol content of 37.75% (v/v) and a gas chromatography produced a bioethanol content of 54.94% (v/v).

Abstract:Currently petroleum-based fossil fuels are the main source of the production of energy. The major issue is that all these resources are non-renewable. As the demand of fuel increases day by day, these non-renewable resources may deplete shortly and will not meet the supply criteria according to the requirements. To overcome this problem, the production of bioethanol using wastes biomass such as fruit peels, agricultural waste, municipal and kitchen waste etc. has gained considerable attention. As these agro-wastes mainly contain lignocellulosic biomass, lignocellulolytic bacterial cultures harness the full potential of these substances. In the present study, endoglucanase producing Bacillus cereus ARA-12 was isolated from soil samples. Fermentation parameters to produce endoglucanase were optimized. The optimum production of endoglucanase was achieved in the medium containing carboxymethyl cellulose (15g/L), yeast extract (30g/L), KH2PO4 (1g/L), K2HPO4 (1.45g/L), MgSO4 (0.4g/L), CaCl2 (0.05g/L) and FeSO4 (0.00125g/L). The optimum temperature and pH of the medium were found to be 50°C and 8 respectively. The maximum enzyme production was achieved at agitation speed of 120 rpm after 20 hours of fermentation by using 5% inoculum. Maximum of 35.9 % bioethanol was produced by the action of endoglucanase enzyme on sugarcane bagasse whereas sweet potato, rice bran, banana peel, corn cob, potato peel and corn husk were also found to be potential raw material to produce second-generation biofuel.

Abstract:In view of the anticipated shortage of the traditional supplies of fossil fuels, there is a great deal of interest in the production of ethanol as an alternative biofuel in recent years. The main objective of this research work was to isolate and characterize stress tolerant, high potential ethanol producing yeast strains from various fruit peel. Two yeast isolates from pineapple (Pa) and orange (Or) have been isolated, characterized on the basis of morphological and physic-chemical characters and optimized on ethanol producing capability using sugarcane molasses as substrate. Ethanol production percentage was estimated by Conway method. Isolates were thermotolerant, pH tolerant, ethanol tolerant as well as osmotolerant. They were resistant to Chloramphenicol (30 μg/disc) and Nalidixic acid (30 μg/disc). The isolates showed no killer toxin activity against E. coli. The highest production capacity of the yeasts was found to be 7.39% and 5.02% for Pa and Or, respectively, at pH 5.0, 30 °C temperature in media with an initial reducing sugar concentration of 6.5% for Pa and 5.5% for Or (shaking). Addition of metal ions increased the rate of ethanol production highest to 10.61% by KH2PO4. This study revealed that indigenous yeast isolates could be used to benefit the fuel demand and industrial alcohol industries.

Abstract:Bio-ethanol is a promising renewable energy but its production is expensive from high cost of feedstocks. In this study, sweet sorghum (Sorghum bicolor (L.) Moench) stalk juice was investigated for bio-ethanol production. Most reports on bio-ethanol productions use commercial Saccharomyces cerevisiae as yeast. However, this study used finger millet (Eleusine coracana) malt with already high adaptation to local conditions and high economic viability as it is being utilized by the indigenous communities. Five sweet sorghum varieties of IESV-92001-DL (V1), NTJ (V2), 15233-IESV (V3), 92008-DJ (V4) and IESV-92028-DL (V5) were planted at 0°3'45.4644" North, 34°17'16.1052" South, in Kenya. °Brix content of juice was determined at 11th to 16th weeks after sowing. Highest °Brix for all varieties were at 15th week where V1 was highest at 22.07. V1 was then harvested for the juice. Factors affecting fermentation; temperature, time, pH and yeast to substrate ratio were optimized using Taguchi method and were obtained as 30 °C, 48 hours, pH 5 and 5 g/L, respectively. Kinetics parameters of Vmax and Km were 0.35 g/L/h and 12.56 g/L, respectively. The optimized and kinetic parameters were within literature values and therefore finger millet malt has a great potential, as a substitute yeast source, in commercial bio-ethanol production. Bioethanol production from the dry powder of Jerusalem artichoke tubers by recombinant Saccharomyces cerevisiae in simultaneous saccharification and fermentation

Abstract:It has been found that recombinant Saccharomyces cerevisiae 6525 can produce high concentration of ethanol in one-step fermentation from the extract of Jerusalem artichoke tubers or inulin. However, the utilization rate of raw materials was low and the fermentation process was costly and complicated. Therefore, in this study, after the optimum processing conditions for ethanol production in fed-batch fermentation were determined in flask, the recombinant S. cerevisiae 6525 was first used to produce ethanol from the dry powder of Jerusalem artichoke tubers in 5-L agitating fermentor. After 72 h of fermentation, around 84.3 g/L ethanol was produced in the fermentation liquids, and the conversion efficiency of inulin-type sugars to ethanol was 0.453, or 88.6 % of the theoretical value of 0.511. This study showed high feasibility of bioethanol industrial production from the Jerusalem artichoke tubers and provided a basis for it in the future.

Abstract:Rhesus monkeys were separated from their mothers at 9 0 days of age and maintained on a purified diet that was isocaloric and either contained 3 % protein or 25% protein by weight ( 1 ) . The diet was started a t 120 days of age for eight Ss ( 4 h ~ g h and 4 lowprotein Ss age-paired) and at 210 days of age for 1 0 Ss ( 4 highand 6 low-protein Ss agepaired). Experimental histories of all Ss (a r the time of the measurements listed below) indicated that all Ss had had previous exper~ence with electrical shock and all to approximately the same extent. For each S the intensity of electrical shock was varied by decreasing and increasing the intensity of the shodc (ranging from .O1 to 2.0 ma) . This was repeated over a period of 1 4 days with one session a day lasting about M hr. A Grason-Stadler shock generator produced a scrambled shock through the floor, constructed of long parallel metal bars. The test apparatus consisted of a grey shuttle box, divided in half by a barrier of metal rods (thus using only one side of the shuttle box). The side of the shuttle box used was 63.5 cm deep, 45.72 cm wide and 55.24 cm long, with a guillotine door centered at one end which measured 27.94 cm u p from the metal rod floor and 22.86 un wide. Observations of a first noticeable response (visible to E, generally a foot retraction or a digit extension) to shock intensity yielded significant differences between both age; paired groups of lowand high-protein-reared Ss. The 120-day age-paired groups responded to a significantly lower intensity of shock for the low-protein-reared Ss than high-proteinreared Ss (8 = 2.3, df = 6, p < .05) . The first noticeable response to shock ranged from . I 0 to .13 ma ( M = .12) for the 120-day low-protein group and from . l 3 to .25 ma ( M = .19) for the 120-day high-protein group. The 210-day age-paired groups also showed a significantly lower intensity of shock response for the low-protein-reared Ss than the high-protein-reared Ss ( t = 2.4, df = 8 , p < .05) . The first noticeable response to shock ranged from .06 to .25 ma ( M = .17) for the 210-day low-protein group and .20 to .30 ma ( M = .26) for the 210-day high-protein group. Variables accountable for these discrepancies in threshold for shock-intensity could be in differences in gross body weight, variations of thickness of cutaneous and dermal layers, deviations in neuron excitability and others. Physiological and behavioral measurements are currently being studied (2 , 3, 4 ) .

Abstract:Global warming and other environmental hazards are escalating due to the continued reliance on fossil fuels, prompting the investigation of renewable energy sources like bio-ethanol. Bio-ethanol, derived from various agricultural and food industry residues, presents a promising solution to mitigate these environmental threats. The research focuses on producing bio-ethanol from pre-treated breadfruit peel waste utilizing 2% NaOH followed by hydrolysis with 7% H 2 SO 4 and fermentation with Saccharomyces cerevisiae (NCIM 3052). Moreover, this study optimizes the combined impact of temperature (ranging from 25, 30, 35, to 40 °C) and fermentation duration (spanning from 12, 24, 36, 48, 60, to 72 h) on the bio-ethanol production process. Ethanol production was quantitatively measured following the spectrometric method using a UV-Spectrophotometer at 525 nm. A change in structural composition and lignocellulose degradation was determined by scanning electron microscopy-energy dispersive X-ray analysis (SEM–EDX), Fourier-transform infrared spectroscopy (FTIR), and ultra high-performance liquid chromatography (UHPLC) for both treated and untreated biomass. The treated breadfruit peel waste showed the highest levels of reducing sugar content. The most significant ethanol yield (1.0 mg/ml) was observed under conditions of 30 °C (T 2 ) temperature and 60 h (D 5 ) of fermentation, followed closely by 35 °C temperature (T 3 ) with a duration of 72 h (D 6 ), yielding approximately 0.89 mg/ml. In summary, this study suggests employing breadfruit peel waste (BPW) for bio-ethanol production, offering an alternative energy source derived from biomass waste. This approach not only addresses environmental waste disposal issues but also contributes to mitigating bio-fuel energy requirements.

Abstract:Ethanol has gained its importance as one of the highly demanded biofuels and is being produced from different food crops or food crop wastes after vigorous pretreatment compelling the production process economically incompatible to some extent. The process also becomes non-ecofriendly as the pretreatment process generally produces acid fumes and toxic effluents as well as consumes high energy. In this current study, the potential of the banana peel as a probable substrate for ethanol production was evaluated with the help of logical prediction. Moreover, a single-step ethanol production process was investigated to eliminate hazardous pretreatment processes. A potential bacterial strain SWET4 was isolated from a sewage treatment pond and was identified as a member of the Klebsiella genus by partial sequencing of the 16S rRNA gene. The isolated strain showed ethanol tolerance up to 12% and exhibited its potential as a cellulolytic strain. The bacterial strain SWET4 was found able to ferment banana peel in MSM broth without any other carbon source with a comparatively prolonged lag phase of cellular growth. Trace of ethanol in banana peel fermentation broth of SWET4 was confirmed by high-pressure liquid chromatography. The optimization study for ethanol production enlightened the fact that high temperature and pH supported higher ethanol production. However, due to the growth restriction of the bacterial cell at higher temperatures and pH, only 40 °C and pH 8 were considered optimum. At the optimum substrate concentration (40 g l−1), ethanol concentration was found to be 5.48 g l−1 after 5 days of incubation which summed as a total yield of 0.137 g of ethanol per g of dried banana peel. This current study evaluates the potential of banana peel as a probable substrate for ethanol production and emphasizes on a new possibility of single-step ethanol production without any pretreatment, although metabolic optimization of the strain SWET4 is required to make the process industrially feasible.

Abstract:Bioethanol is a renewable chemical that can replace fossil fuels. Pistia stratiotes , an aquatic weed, was used as the substrate for the synthesis of ethanol in the current investigation. The presence of high amount of cellulose (25.90%) and hemicellulose content (18.44%) makes it suitable to produce bioethanol. Optimization of acidic pre-treatment conditions was performed using response surface methodology where independent variables acid concentration (0.15–3.1% H 2 SO 4 ), time (12.96–97.0 min) and temperature (93.18–126.82 °C) were taken. The responses were recorded as sugar and cellulose concentrations. Subsequently, optimized condition was subjected to fermentation for the production of ethanol using Saccharomyces cerevisiae and Pichia stipitis strains. The maximum amount of sugar was produced under optimal conditions (2.5% acid concentration) at 120 °C for 15 min. Initial sugar was taken as 10 g/L in the hydrolysate, which is consumed by the strains with maximum sugar conversion rate (0.24 g/L/h). Maximum sugar consumption was observed by S. cerevisiae and P. stipitis and was 85.9% and 87.9%, respectively. Thus, we present that the optimized acidic pre-treatment conditions for maximum yield of ethanol from Pistia stratiotes using S. cerevisiae and P. stipitis were observed to be 0.37 and 0.39 respectively. Hence, this study showed an effective conversion of plant biomass into ethanol.

Abstract:In recent years, various efforts have been invested in producing bioethanol from lignocellulosic biomass (LCB) using thermophilic bacteria. Twelve thermophilic ethanologenic bacteria were isolated from cow dung-grass compost using an enrichment technique, and the isolate, CSD6, which produced the highest bioethanol, was studied further. CSD6 utilized both pentose and hexose sugars producing ethanol, lactic acid and acetic acid as major soluble products and was identified through 16S rRNA gene sequencing as a strain of Geobacillus stearothermophilus . The highest ethanol production was found to be 25.05 mM from 5 g/L glucose, equivalent to 55% of the theoretical ethanol yield at 55 °C and initial pH 7.5. The isolate showed average tolerance to ethanol and acetic acid concentration and initial substrate loading. The bioethanol potential of CSD6 was also studied using autoclave-treated mixed floral waste (FW) as a no-cost substrate. CSD6 produced a maximum of 8.9 mM of bioethanol with a 75.47% decrease in reducing sugars using 20 g/L FW as substrate without any enzymatic pretreatment, indicating the ability of CSD6 to produce bioethanol from easily available substrates.

Abstract:South Sulawesi has potential in the development of bioethanol from biomass because it has a plantation sector which is a mainstay sector with various types of commodities that can produce biomass. One biomass that has the potential as a raw material for bioethanol is coconut pulp. The reason for choosing coconut pulp as raw material for bioethanol production is because the cellulose rate is 36.08%; hemicellulose is 12.58%, besides that coconut is the main commodity crop in the plantation sector in South Sulawesi and its utilization will not interfere with food needs so that it will ensure smooth production of bioethanol. This study aims to produce bioethanol with coconut pulp raw materials using physical pretreatment namely autoclave. Autoclave pretreatment results are followed by fermentation using yeast for 7, 9, 11, 13 and 15 days anaerobically and adding urea and NPK as nitrogen sources during the fermentation process. The result of fermentation was then distilled to obtain bioethanol for 12-72 hours. The highest bioethanol content was obtained after 13 days of fermentation which was 6,2683%. This pretreatment uses the influence of temperature and pressure to degrade lignin in coconut pulp to optimize fermentation results. Based on mapping the amount of coconut pulp, a coconut pulp supply network model can be produced at the upstream level including supply sources, depots and bioethanol plants.
