Abstract:In this study, cell biosensor chip with micro-electrode arrays was used to monitor the culture behaviour of mammalian cancer cells and evaluate the chemosensitivity of anti-cancer drugs using electrochemical impedance spectroscopy. The electrode arrays were fabricated by semiconductor technology with diameter from 50 pm to 10 pm of each electrode. The human oesophageal cancer cell lines (KYSE 30) were cultured on the surface of the electrode with the help of fibronectin. Morphology changes of cells adhesion, spreading, and proliferation can be detected by impedimetric analysis in a real time and non-invasive way. Anti-caner drug of cisplatin was added to cells for potential drug screening applications. The experimental results show that drug's chemosensitivity effects can be detected by this cell based biosensor.

Abstract:Here a novel paper-based electrochemiluminescence (ECL) device was developed for the detection of HL-60 cancer cells. In this approach, porous filter paper as a thin-layer electrochemical cell connected to a sheet of ITO working electrode. Au nanoparticles (Au NPs) and graphene were coated on the surface of ITO to immobilize capture HS-DNA. Then, the aptamer of HL-60 cancer cells, which is labeled with Ru(bpy)(3)(2+)-conjugated silica nanoparticles (RuSi@Ru(bpy)(3)(2+)) is hybridized with the capture DNA to form bioconjugates. In the presence of HL-60 cells, the aptamer would conjugate with glycoprotein at cell surface and RuSi@Ru(bpy)(3)(2+) NP-aptamer would be released. After the dissociation of RuSi@Ru(bpy)(3)(2+) NP-aptamer from the surface of ITO, ECL signal would decline. This simple, portable and disposable testing platform could detect HL-60 cancer cells down to 56 cells/mL (C) 2014 Elsevier B.V. All rights reserved.

Abstract:Here we have developed a sensitive cancer cell amplified detection method which combined Au NPs enhanced electrochemiluminescence (ECL) of CdS nanocrystals (NCs) film, with isothermal circular amplification reaction of polymerase. In DNA circular amplification detection system, hairpin DNA beacon/Au NPs composite modified CdS NCs film was used as an ECL emitter. Messenger DNA is hybridized with the aptamer modified on magnetic beads (MBs) to form MB–Au bioconjugates. In the presence of HL-60 cell, the aptamer would conjugate with the glycoprotein at cell surface and messenger DNA sequence would be released. The released messenger DNA sequence was then introduced into the cycle amplification system to trigger circular polymerizations. This assay allows us to determine the released messenger DNA equivalent to 10 cells and exhibits a significant specificity for HL-60 cells.
A Ratiometric Electrochemiluminescence Detection for Cancer Cells Using G-C 3 N 4 Nanosheets and Ag–PAMAM–luminol Nanocomposites

Abstract:In this work, a dual-signaling electrochemiluminescence (ECL) ratiometric sensing approach for the detection of HL-60 cancer cells was reported for the first time. G-C3N4 nanosheets and Ag-PAMAM-luminol nanocomposits (Ag-PAMAM-luminol NCs) were prepared and served as reductive-oxidative and oxidative-reductive ECL emitters respectively. DNA probe functionalized Ag-PAMAM-luminol NCs would hybridize with aptamers modified onto magnetic beads. In the presence of HL-60 cells, the aptamer would conjugate with the target cell and release Ag-PAMAM-luminol NCs. After magnetic separation, released Ag-PAMAM-luminol NCs would hybridize with capture DNA on g-C3N4 nanosheets. ECL from g-C3N4 nanosheets coated on ITO electrode at -1.25 V (vs SCE) could be quenched by Ag-PAMAM-luminol NCs due to the resonance energy transfer (RET) from g-C3N4 nanosheets to Ag NPs. Meanwhile, Ag-PAMAM-luminol brought the ECL signal of luminol at +0.45 V (vs SCE). Thus, the concentration of HL-60 cancer cells could be quantified by both the quenching of ECL from g-C3N4 nanosheets and the enhancement of ECL from luminol. By measuring the ratio of ECL intensities at two excitation potentials, this approach could achieve sensitive and reliable detection for cancer cells in a wide range from 200 cells/mL to 9000 cells/mL with the detection limit of 150 cells (S/N=3).

Abstract:Here we developed a novel hybrid bipolar electrode (BPE)-electrochemiluminescence (ECL) biosensor based on hybrid bipolar electrode (BPE) for the measurement of cancer cell surface protein using ferrocence (Fc) labeled aptamer as signal recognition and amplification probe. According to the electric neutrality of BPE, the cathode of U-shaped ITO BPE was electrochemically deposited by Au nanoparticles (NPs) to enhance its conductivity and surface area, decrease the overpotential of O-2 reduction, which would correspondingly increase the oxidation current of Ru(bpy)(3)(2+)/tripropylamine (TPA) on the anode of BPE and resulting a similar to 4-fold enhancement of ECL intensity. Then a signal amplification strategy was designed by introducing Fc modified aptamer on the anode surface of BPE through hybridization for detecting the amount of mucin-1 on MCF-7 cells. The presence of Fc could not only inhibit the oxidation of Ru(bpy)(3)(2+) because of its lower oxidation potential, its oxidation product Fc(+) could also quench the ECL of Ru(bpy)(3)(2+)/TPA by efficient energy-transfer from the excited-state Ru(bpy)(3)(2+)* to Fc(+), making the ECL intensity greatly quenched. On the basis of the cathodic Au NPs induced ECL enhancing coupled with anodic Fc induced signal quenching amplification, the approach allowed detection of mucin-1 aptamer at a concentration down to 0.5 fM and was capable of detecting a minimum of 20 MCF-7 cells. Besides, the amount of mucin-1 on MCF-7 cells was calculated to be 9041 +/- 388 molecules/cell. This approach therefore shows great promise in bioanalysis.

Abstract:We report here a sensitive electrochemiluminescence (ECL) approach for detection of HL-60 cancer cells based on Au nanoparticles (NPs) enhanced ECL of CdS nanocrystal (NC) film supplemented by magnetic separation. In this approach, capture DNA-1 modified CdS NC film was used as ECL emitter. Capture DNA-2, which is labeled with Au NP, is hybridized with the aptamer modified on magnetic beads (MBs) to form MB-Au NP bioconjugates. In the presence of HL-60 cells, the aptamer would conjugate with the glycoprotein at cell surface and Au NPs labeled capture DNA-2 would be released. After magnetic separation, the released Au NPs labeled capture DNA-2 is hybridized with the capture DNA-1 to enhance the ECL signal of CdS NC film. This approach could sensitively detect HL-60 cells with a linear calibration range from 20 to 1.0×106cells/mL.

Abstract:An electrochemiluminescence-electrochemistry (ECL-EC) dual-mode sensing platform based on a vertically-ordered mesoporous silica films (VMSF) modified electrode was designed here for the sensitive and selective determination of cancer antigen 15-3 (CA 15-3), a specific biomarker of breast cancer. VMSF was assembled through a rapid electrochemically assisted self-assembly (EASA) method and plays a crucial role in signal amplification via a strong electrostatic interaction with the positively charged bifunctional probe Ru(bpy)32+. To construct the biorecognition interface, epoxy functional silane was linked to the surface of VMSF for further covalent immobilization of the antibody. As a benefit from the specific combination of antigen and antibody, a non-conductive immunocomplex layer was formed in the presence of CA 15-3, leading to the hinderance of the mass and electron transfer of the probes. Based on this strategy, the dual-mode determination of CA 15-3 ranging from 0.1 mU/mL to 100 mU/mL with a LOD of 9 μU/mL for ECL mode, and 10 mU/mL to 200 U/mL with a LOD of 5.4 mU/mL for EC mode, was achieved. The proposed immunosensor was successfully employed for the detection of CA 15-3 in human serum without tedious pretreatment.

Abstract:Exosomes have been reported to play an important role in the anti-tumor immune response, tumor diagnosis and other processes, and are promising biomarkers for early cancer diagnosis. In this work, a sensitive electrogenerated chemiluminescence (ECL) biosensor was developed for detection of exosomes using aptamer modified two-dimensional material Ti3C2 MXenes nanosheets as the ECL nanoprobe because of its large surface area, the excellent conductivity and catalytic properties. The exosomes can be high efficiently captured onto the electrode surface by an EpCAM protein recognized aptamer modified on the electrode surface. In addition, the ECL nanoprobe can also recognize the exosomes, and significantly enhanced the ECL signals of luminol. Based on this strategy, a highly sensitive ECL biosensor for MCF-7 exosomes detection was obtained. The detection limit is 125 particles μL-1, which was over 100 times lower than that of conventional ELISA method. The as prepared ECL biosensor was performed successfully for MCF-7 exosomes detection in the serum. This strategy provided a feasible, sensitive and reliable tool for the exosomes detection in exosomes-related clinical diagnostics.

Abstract:In the past 50 years, cancer has been the leading cause of death globally, and the development of effective cancer early diagnosis technology is still facing challenges. Herein, a three-dimensional (3D) mesoporous-macroporous interface for electrochemical sensing was constructed on the electrode surface through the layer-by-layer assembly method. Mesoporous silica (MS) coating was epitaxially grown on the outer side of the polystyrene (PS) microtubes. Then after loading the active component sambucus nigra agglutinin (SNA), the 3D mesoporous-macroporous interface was applied to the super-sensitive electrochemical detection of cancer cells. Specifically, the layer-by-layer assembly method was conducive to maximize the exposure of the detection of active substances, which had better cellular capture performance than direct mixing methods. The 3D structure of PS nanotubes was conducive to improving the specific surface area of the electrode, promoting its capture efficiency of cancer cells. The mesoporous structure can load more SNA, improving the specific recognition performance of cancer cells. The results showed that the detection platform based on SNA-SA specific recognition technology developed in this work had excellent cancer cell detection performance, stability, and selectivity, which is expected to be used in the early detection of cancer.
Highly Specific Electrochemiluminescence Detection of Cancer Cells with A Closed Bipolar Electrode

Abstract:An advanced biosensor based on a closed bipolar electrode (BPE)-electrochemiluminescence (ECL) strategy is proposed for cancer cell adhesion, specific recognition, and detection with dual biomarkers. The system consists of a two-channel polydimethylsiloxane (PDMS) chip (sensing channel and reporting channel) connected through a U-shaped indium tin oxide BPE at a glass surface. A sandwich-type cancer cell detection model is developed at the cathode of the BPE with two recognition molecules, folic acid (FA) and an aptamer. On the basis of the high affinity between FA and the folate receptor that is overex-pressed at a cancer cell membrane as well as the excellent conductivity of graphene-Au conjugates, this strategy could achieve the sensitive detection of HL-60 cells, with a limit as low as 18 cells in 30 mu L of cell suspension. Control experiments confirm that the sandwich assay specifically captures cancer cells and recognizes target cells.

Abstract:Herein, we presented a simple ultra-sensitive "on-off-on" electrochemiluminescence (ECL) biosensor that uses copper nanoclusters (CuNCs) synthesized via a DNA nanoribbon (DNR) template (DNR-CuNCs) as a sensing platform coupled with target-cycle amplification for the sensitive and rapid detection of cancer cells that overexpress proteins. DNR templated DNR-CuNCs, a well-ordered ECL emitter, was integrated into the nucleic acid framework structure to self-assembly, which enhanced the ECL intensity benefit from reduced band gap and accelerated electron transfer reaction of CuNCs. Additionally, an optimized exonuclease III (Exo III)-assisted cycle amplification method was introduced to efficiently convert trace target biomarkers into a substantial output of DNA, significantly improving target conversion efficiency and boosting the ECL signal. Consequently, MUC1 proteins were sensitively detected by the ECL biosensor throughout a broad concentration range, from 1 fg·mL −1 to 1 ng·mL −1 , with a low limit of detection (LOD) of 0.061 fg·mL −1 , and successfully detected MUC1-overexpressing cancer cells (using MCF-7 cancer cells as a model), accurately identifying MCF-7 cells in the concentration range of 10–100,000 cells·mL −1 with a LOD of 5 cells·mL −1 . This research, for the first time, introduced a DNR-CuNCs ECL biosensor for the highly sensitive detection of living cells and their associated surface proteins. The detection of biomarker proteins and specific cells is greatly promising by applying this simple, sensitive, and specific strategy.

Abstract:Simple development of an electrochemiluminescence (ECL) immunosensor for convenient detection of tumor biomarker is of great significance for early cancer diagnosis, treatment evaluation, and improving patient survival rates and quality of life. In this work, an immunosensor is demonstrated based on an enhanced ECL signal boosted by nanochannel-confined Au nanomaterial, which enables sensitive detection of the tumor biomarker—carcinoembryonic antigen (CEA). Vertically-ordered mesoporous silica film (VMSF) with a nanochannel array and amine groups was rapidly grown on a simple and low-cost indium tin oxide (ITO) electrode using the electrochemically assisted self-assembly (EASA) method. Au nanomaterials were confined in situ on the VMSF through electrodeposition, which catalyzed both the conversion of dissolved oxygen (O2) to reactive oxygen species (ROS) and the oxidation of a luminol emitter and improved the electrode active surface. The ECL signal was enhanced fivefold after Au nanomaterial deposition. The recognitive interface was fabricated by covalent immobilization of the CEA antibody on the outer surface of the VMSF, followed with the blocking of non-specific binding sites. In the presence of CEA, the formed immunocomplex reduced the diffusion of the luminol emitter, resulting in the reduction of the ECL signal. Based on this mechanism, the constructed immunosensor was able to provide sensitive detection of CEA ranging from 1 pg·mL−1 to 100 ng·mL−1 with a low limit of detection (LOD, 0.37 pg·mL−1, S/N = 3). The developed immunosensor exhibited high selectivity and good stability. ECL determination of CEA in fetal bovine serum was achieved.

A Novel Signal-On Electrochemiluminescence Immunosensor for the Detection of NSCLC Antigen Biomarker Based on New Co-Reaction Accelerators

Abstract:Non-small cell lung cancer (NSCLC) is the most common type of lung cancer with substantial morbidity and mortality. Herein, a new signal-on electrochemiluminescence (ECL) immunosensor based on multiple amplification strategies is constructed for ultrasensitive detection of cytokeratin 19 fragment antigen 21-1 (CYFRA21-1) biomarker related to NSCLC. Polyethyleneimine (PEI) functionalized MXene is decorated with NiMn layer double hydroxide (NiMn LDH) to form MXene-PEI-NiMn LDH composite. Specially, the La-MOF@ZIF-67 bimetallic organic framework (named as LZBM) and MXene-PEI-NiMn LDH both served as coreaction accelerators to improve the ECL emission of the luminol-H2 O2 system. To be specific, Au nanoparticles (AuNPs) coated MXene-PEI-NiMn LDH is applied to immobilize primary CYFRA21-1 antibody (Ab1 ), while AuNPs decorated LZBM was used for the loading of luminol and secondary CYFRA21-1 antibody (Ab2 ) to form tracer label. Therefore, the ECL signal of the sandwich-type immunosensor is significantly enhanced due to the high loading capability for luminol and the synergistic catalytic ability for the decomposition of H2 O2 into reactive oxygen species (ROS). Under the optimal experimental conditions, the ECL immunosensor exhibited good analytical performances for CYFRA21-1 detection with a wide linear range (100 fg mL-1 -100 ng mL-1 ) and a low limit of detection (85.20 fg mL-1 ), providing a promising method for early diagnosis of NSCLC.

Abstract:Carcinoembryonic antigen (CEA) is a well-known cancer biomarker for the detection of several malignancies. The development of ultrasensitive CEA diagnostic tools is crucial for early detection and progression observation of tumors. Herein, a dual signal amplified sandwich-type electrochemical immunoassay was developed based on dual-labeled mesoporous silica nanospheres as a signal amplifier, combined with NiO@Au decorated graphene as a conductive layer for ultrasensitive and rapid determination of CEA. The dual-labeled mesoporous silica (DLMS) nanosphere, which was synthesized by entrapping Au nanorod (Au NR) and horseradish peroxidase (HRP) in the channels of amine-functionalized SBA-15 followed by subordinate antibody (Ab2) conjugation which was denoted as Au NR@SBA-15/Ab2-HRP. The dual signal amplification from Au NR@SBA-15 and HRP enhanced the sensitivity of the proposed immunoassay. Consequently, the developed DLMS based immunosensor displayed ultra-low limits of detection of 5.25 fg/mL and a wide range of linearity (0.1-5 pg/mL), which was extended for CEA determination in real-time samples with improved recoveries of >98%. Therefore, this dual amplification prototype would cater to the clinical requirements for the ultrasensitive detection of CEA biomarkers.

Abstract:A multivalent recognition and alkaline phosphatase (ALP)-responsive electrogenerated chemiluminescence (ECL) biosensor for cancer cell detection and in situ evaluation of cell surface glycan expression was developed on a poly(amidoamine) (PAMAM) dendrimer-conjugated, chemically reduced graphene oxide (rGO) electrode interface. In this strategy, the multivalency and high affinity of the cell-targeted aptamers on rGO provided a highly efficient cell recognition platform on the electrode. The ALP and concanavalin A (Con A) coated gold nanoparticles (Au NPs) nanoprobes allowed the ALP enzyme-catalyzed production of phenols that inhibited the ECL reaction of Ru(bpy)(3)(2+) on the rGO electrode interface, affording fast and highly sensitive ECL cytosensing and cell surface glycan evaluation. Combining the multivalent aptamer interface and ALP nanoprobes, the ECL cytosensor showed a detection limit of 38 CCRF-CEM cells per mL in human serum samples, broad dynamic range and excellent selectivity. In addition, the proposed biosensor provided a valuable insight into dynamic profiling of the expression of different glycans on cell surfaces, based on the carbohydrates recognized by lectins applied to the nanoprobes. This biosensor exhibits great promise in clinical diagnosis and drug screening.
Highly Sensitive Colorimetric Cancer Cell Detection Based on Dual Signal Amplification

Abstract:Facile and efficient detection of cancer cells at their preclinical stages is one of the central challenges in cancer diagnostics. A direct, rapid, highly sensitive and specific biosensor for detection of cancer biomarkers is desirable in early diagnosis and prognosis of cancer. In this work, we developed, for the first time, an easy and intuitive dispersion-dominated colorimetric strategy for cancer cell detection based on combining multi-DNA released from an aptamer scaffold with cyclic enzymatic amplification, which was triggered by aptamer DNA conformational switch and demonstrated by non-cross-linking gold nanoparticles (Au NPs) aggregation. First, five kinds of messenger DNAs (mDNAs) were aligned on the cancer cell aptamers modified on magnetic beads (MBs) to form mDNAs-Apt-MBs biocompatible nanosensors. In the presence of target cells, the aptamer would bind to the receptors on the cell membranes, and mDNAs would be released, resulting in the first amplification that one biological binding event would cause the release of multiple kinds of mDNAs simultaneously. After magnetic separation, the released mDNAs were introduced into the cyclic enzymatic amplification to cleave more single strand DNA (ssDNA) fragments. Instead of modification of Au NPs, these fragments and mDNAs could be adsorbed on the surface of Au NPs to prevent particle aggregation and ensure the stability and color of solution in high salt environments. The linear response for HL-60 cells in a concentration range from 10 to 104 cells was obtained with a detection limit of four cells in buffer solution. Moreover, the feasibility of the proposed strategy was demonstrated in a diluted serum sample. This dual signal amplification method can be extended to other types of cancer cells, which has potential application in point-of-care cancer diagnosis.

Abstract:Nowadays, lung cancer is one of the most dangerous cancers threatening human life all over the world. As a crucial biomarker, cytokeratin 19 fragment 21-1 (CYFRA 21-1) is extraordinary important for diagnosis of non-small cell lung cancer (NSCLC). In this work, we synthesized hollow SnO 2 /CdS QDs/CdCO 3 heterostructured nanocubes with high and stable photocurrents, which applied to construction of a sandwich-typed photoelectrochemical (PEC) immunosensor for detection of CYFRA 21-1, integrated by an in-situ catalytic precipitation strategy with home-built PtPd alloy anchored MnCo–CeO 2 (PtPd/MnCo–CeO 2 ) nanozyme for synergistic amplification. The interfacial electron transfer mechanism upon visible light irradiation was investigated in details. Further, the PEC responses were seriously quenched by the specific immunoreaction and precipitation catalyzed by the PtPd/MnCo–CeO 2 nanozyme. The established biosensor showed a wider linear range of 0.001–200 ng mL −1 and a lower limit of detection (LOD = 0.2 pg mL −1 , S / N = 3), coupled by exploring such analysis even in diluted human serum sample. This work opens a constructive avenue to develop ultrasensitive PEC sensing platforms for detecting diverse cancer biomarkers in clinic.

Abstract:The concentration of carcinoembryonic antigen (CEA) is an important indicator for clinical monitoring and diagnosis of tumor. Early sensitive detection of CEA is significant for cancer diagnosis and therapy. In this study, a sensitive electrochemiluminescence (ECL) immunosensor was fabricated based on functionalized metal organic layers (MOLs) for the determination of CEA. In this immunosensor, Au nanoparticles (AuNPs) were grown on ultrathin two-dimensional MOLs via in situ reduction. AuNPs not only can accelerate the electron transfer but also can be used as the labels to conjugate with antibody. The as-prepared nanocomposite showed high ECL efficiency and can be used as an ECL emitter owing to the great ECL property of Ir-Zr-MOL. The ECL intensity of decreased linearly with the logarithm of CEA concentration in the range from 1.00 pg mL −1 to 100 ng mL −1 . And the detection limit was 0.200 pg mL −1 (S/N = 3). What's more, the as-prepared immunosensor worked well in human blood samples, showing its potential for real analysis.

Abstract:Cancer is a serious threat to human health, and early identification of cancer biomarkers is crucial for timely treatment. A dual-mode biosensing platform has been developed to identify colon carcinoma suppressor microRNA-199a (miRNA-199a). A biofuel cell is firstly constructed using hybridization chain reaction and catalytic hairpin assembly self-assembly. The anode is a flexible carbon cloth (CP) loaded with glucose oxidase-functionalized molybdenum disulfide nanorods (MoS 2 nanorods), and the bio-cathode is made up of double-stranded deoxyribonucleotide chains generated by nucleic acid amplification technology. The linear range of the electrochemical method is 0.1 fM-100 pM, and the limit of detection is 24.1 aM ( S/N = 3 ). The colorimetric method has a linear range of 0.1 fM-10000 pM with detection limit of 34.5 aM ( S/N = 3 ). The combination of MoS 2 nanorods, enzyme-free cascade signal amplification technology, and dual-mode detection strategy provides high sensitivity and accuracy, offering a promising method for accurate determination of cancer biomarkers.
