Abstract:Objective To evaluate in vivo magnetic resonance (MR) imaging with a conventional 1.5 T system for depiction and tracking of intravascularly injected magnetically labeled bone marrow mesenchymal stem cells (MSCs) in rat normal kidney. Methods Rat bone marrow MSCs were isolated, purified, expanded and then incubated with home synthesized Fe_2O_3-PLL. Prussian blue stain was employed for identifying intracellular irons. MSCs were injected into renal arteries of 12 recipient rats (labeled cells in nine, unlabeled cells in three). MR images of kidneys were obtained respectively before injection of MSCs, and immediately, 1, 3, 5, and 8 days after transplantation. MR imaging findings were analyzed, which were correlated with histological findings. Results Rat MSCs were successfully labeled, and labeling efficiency was almost 100%. Prussian blue staining of Fe_2O_3-PLL labeled cells revealed the presence of iron-containing vesicles or endosomes in the cytoplasm. In normal kidneys, the labeled MSCs were demonstrated as signal intensity loss in renal cortex on T2*-weighted MR images. The signal intensity decrease was visualized up to days 8 after transplantation. Histological analyses showed that most Prussian blue staining-positive cells were well correlated with the area where a signal intensity loss was observed in MRI. Conclusion 1.5T MR imaging can monitor administered magnetically labeled MSCs in vivo in normal kidneys, especially on T2*WI.
[Magnetic Resonance Imaging of Mesenchymal Stem Cells Transplanted Intravascularly in Treatment of Acute Renal Failure:In Vivo Experiment with Rats].

Abstract:OBJECTIVE:To evaluate the efficacy of in vivo magnetic resonance imaging (MRI) of mesenchymal stem cells (MSCs) injected intravascularly in treatment of acute renal failure (ARF) , and to investigate the changes of renal function and pathology of ARF after MSC transplantation.METHODS:Rat MSCs were isolated and labeled with Fe2O3-PLL in vitro. Thirty SD rats underwent intramuscular injection of glycerol so as to establish ARF models and then randomly divided into 3 equal groups: Group I undergoing injection of labeled MSCs into abdominal aorta via transcatheter, Group II injected with unlabelled MSCs, and Group III injected with normal saline as controls. MRI of kidney was conducted before injection, and 0.5 h, and 1, 2, and 5 days after injection. One and 2 days after the transplantation 3 rats from each group underwent MRI and extraction of blood samples from the abdominal aorta and then killed with their kidneys taken out, and 5 days after the rest rats were all killed after MRI with their kidneys taken out. Serum creatinine (Scr) and blood urea nitrogen (BUN) were examined so as to evaluate the renal function. Microscopy was conducted to observe the pathological changes. Prussian blue + CD68 antibody staining was performed to identify the labeled MSCs.RESULTS:MRI showed decrease of signal intensity in renal cortex on the T2 *-weighted MR images up to 5 days after transplantation. Histological analysis showed that most Prussian blue-positive cells were in the glomerular capillaries, corresponding to the areas where signal intensity decrease was observed by MRI. The Scr and BUN levels 2 and 5 days after the implantation of Group I were both lower than those of the control group, and there were not significant differences in the Scr and BUN levels between Groups I and II. Renal tubular injury scoring showed that the renal tubular injury was significantly lighter than that of the control group.CONCLUSION:1.5-T MRI seems a good in vivo technique to monitor the magnetically labeled MSCs administered into the abdominal aorta of ARF animals, which are distributed in the glomerular capillaries in the early stage after transplantation. MSCs may promote the recovery of ARF.
Targeted Delivery of Bone Mesenchymal Stem Cells by Ultrasound Destruction of Microbubbles Promotes Kidney Recovery in Acute Kidney Injury

Abstract:The aim of the present study was to explore whether ultrasound microbubble destruction augments site-targeted engraftment of bone marrow mesenchymal stem cells (BM-MSCs) to kidney tissue and promotes recovery of the kidney in acute kidney injury (AKI) in rats. AKI was induced by the subcutaneous injection of mercuric chloride (HgCl2). Forty Sprague-Dawley (SD) rats were randomly divided into the following groups after the establishment of rat models of AKI (n = 10): (1) Model group alone (control group); (2) 1.0 W/cm(2) ultrasound (US) + microbubble (MB) (US/MB group); (3) MSCs group; and (4) 1.0 W/cm(2) US+MB + MSCs group (US/MB + MSCs group). The number of 4',6-diamidino-2-phenylindole (DAPI) labelled MSCs was evaluated by fluorescence microscopy and real-time polymerase chain reaction (RT-PCR) and Western blotting and histological examination were performed 7 days after MSCs transplantation. It was observed via fluorescence microscopy that the number of DAPI-labelled MSCs in the kidney for the US/MB + MSCs group was significantly more than the MSCs group (p < 0.05). The results from RT-PCR revealed that the US/MB and US/MB 1 MSCs groups markedly increased the level of inter-cellular adhesion molecule 1 (ICAM-1) messenger ribonucleic acid (mRNA) compared with the control group and the MSCs group (p < 0.05). Western blot analysis showed that the expression of hepatocyte growth factor (HGF) and epidermal growth factor (EGF) in the US/MB 1 MSCs group were markedly increased compared with the all other groups (p < 0.01). The extent of tubular necrosis and dilation was significantly milder in the US/MB + MSCs group (acoustic exposure conditions: 5s at 1 MHz and 1.0 W/cm(2) with a 5s pause, totalling 60 s) than the all other groups (p < 0.05). Microbubble destruction by 1.0 W/cm(2) ultrasound can promote both the homing of BM-MSCs to kidney tissue and the recovery of the kidney in AKI in rats. Labeling Neural Stem Cells with Superparamagnetic Iron Oxide and MRI Tracking in Vitro and in Vivo

Abstract:Objective To label neural stem cells with superparamagnetic iron oxide (SPIO) and track labeled cells with MRI in vitro and in vivo after implantation. Methods 13 SD rats were used for the study. Rat neural stem cells were labeled with SPIO combined with PLL by the mean of receptor-mediated endocytosis. Prussian blue staining and electron microscopy were conducted to identify the iron particles in these neural stem cells, and SPIO-labeled cells were tracked with 1.5 T and 4.7T MRI in vivo and in vitro after implantation. The subjects were divided into five groups, including 5×105 labeled cells, 5×105 unlabeled cells, cell culture medium with SPIO, cell culture medium without SPIO and distilled water. MRI scanning sequences included T_1WI, T_2WI and T_2*WI. R_2 and R_2* relaxation rate of labeled cells were calculated. Results (1) Neural stem cells could be labeled with SPIO and labeling efficiency was 100%, Prussian blue staining showed numerous blue-stained iron particles in the cytoplasm. (2) The average percentage change of signal intensity of labeled cells on T_1WI in 1.5 T and 4.7 T MRI was 20.53% and 24.06% respectively,on T_2WI was 40.78% and 50.66% respectively,and on T_2*WI was 46.57% and 53.70% respectively. The signal intensity had no significant difference between T_2WI and T_2*WI in 1.5 T MRI, however the signal intensity had significant difference between T_2WI or T_2*WI and T_1WI. (3) T_2 of unlabeled cells and labeled cells in 4.7 T MRI were 516 ms and 77 ms respectively, R_2 were 1.94 s -1 and 12.98 s -1 respectively; and T_2* were 109 ms and 22.9 ms, R_2* were 9.17 s -1 and 43.67 s -1 respectively. (4) Remarkable low signal area on T_2WI and T_2*WI was seen in the left brain implanted with labeled cells one week ago, however no signal change in the right brain implanted with unlabeled cells. Conclusion Neural stem cells can be labeled effectively with SPIO; R_2 and R_2* of labeled cells were increased obviously; MRI can be used to track labeled cells in vitro and in vivo.

Abstract:BACKGROUND:Among many transplanted cells,adult autologous bone barrow-derived mononuclear cells have beenused in clinical practice because they are easy to be obtained,without immunological rejection and ethical disputationand other advantages.How to distinguish donor cells from receptors and observe the survival of donor cells following stem cell transplantation still trouble people.OBJECTIVE: superparamagnetic iron oxide (SPIO)particles-labeled bone marrow-derived mononuclear cells from minipigs were used to observe the feasibility of in vivo tracking with magnetic resonance imaging(MRI).DESIGN:A controlled observation experiment.SETTING:Institute of Cardiovascular Disease,Zhongda Hospital Affiliated to Southeast University.MATERIALS:This experiment was carried out in the Institute of Cardiovascular Disease,Zhongda Hospital Affiliated to Southeast University between April 2006 and August 2006.Healthy Chinese minipigs,aged 3 to 4 months,weighing from 20 to 30 kg,were provided by the Experimental Animal Center of Southeast University[SYXK(Su)2002-0012].METHODS: Autologous bone marrow-derived mononuclear cells of minipigs were isolated and cultured. Bone marrow-derived mononuclear cells in the suspension were traced with SPIO particles.Ferrum in the cells were shown by Prussian blue staining, and cell viability was evaluated by trypan blue exclusion method. Eleven minipigs used for preparation of model of myocardial infarction were divided into experimental group(n=9)and control group(n=2).By means of percutaneous left or right cervical artery or femoral artery puncturation, 1.5 to 2.0 mm balloon was used to occlude 1/3 left anterior descending branch,304 to 405 kPa,60 minutes later,ischemic preconditioning was conducted 3 tO 4 times before operation. When pig models of myocardial infarction were successful that was proved by surface electrocardiogram,bone marrow-derived mononuclear cells were percutaneously injected into coronary artery.Coronary arteriography was performed through femoral artery acupuncture at 24 hours after establishing infarction models.Suspension of bone marrow-derived mononuclear cells was perfused into coronary artery with OTW catheter.Then,the injector and OTW catheter for containing cells were rinsed with normal saline containing heparin and infused with the residual cells within 10 minutes.Non-labeled cells were perfused in 2 minipigs of control group by the same method.Postoperatively, bone marrow-derived mononuclear cells were traced by magnetic resonance and compared with Prussian blue-stained myocardial tissue sections.RESULTS: Seven minipigs of experimental group and one minipig of control group were Involved in the final analysis.One of each group was used for preparation of model of myocardial Infarction.One minipig of experimental group died from anesthetic accident before magnetic resonance.①Bone marrow-derived mononuclear cells all were nearly labeled by SPIO particles. Bone marrow-derived mononuclear cells could further proliferate in culture medium containing Fe2O3-PLL without obvious changes of cellular shape. ②T2+WI showed that 5 of 8 models of myocardial infarction presented fuzzy low-echo signal region in peripheral myocardial infarction after transplantation of labeled cells and the low-echo signal disappeared 4 weeks Iater. Ex vivo T2+WI sequence showed there was a dot-distributed low-echo signal region in the peripheral infarction region.③It was found in histological examination that 5 models(cell number over 106) had Prussian blue-positive cells,which distributed the same as those in magnetic resonance signal reducing region.CONCLUSION:SPIO particles-labeled bone marrow-derived mononuclear cells are safe and effective;T2+ WI is sensitive to tracing SPIO particles-labeled bone marrow-derived mononuclear cells;Magnetic resonance can in vivo trace SPIO particles-labeled stem cells transplanted through coronary artery,magnetic resonance signal change is related with the number of stem cells and division growth.
In Vivo Cardiac Magnetic Resonance Imaging of Superparamagnetic Iron Oxides-Labeled Mesenchymal Stem Cells in Swines

Abstract:OBJECTIVE:To explore the feasibility of magnetic resonance imaging (MR) on detecting transplanted nanometer small superparamagnetic iron oxides (SPIO) labeled mesenchymal stem cells (MSCs) in swine model with acute myocardial infarction (MI).METHODS:MSCs isolated from swine were incubated with nanometer SPIO for 24 hours and the third-passage MSCs were labeled with DNA dye 4'-6-diamidino-2-phenylindole (DAPI) and aliphatic red fluorescent dye PKH(26)-GL. Presence of small particles of SPIO in MSCs was assessed by Prussian Blue staining and electron microscopy. Three animals in each group received SPIO-labeled MSCs (5 x 10(5); 1 x 10(6); 2 x 10(6)) and MSCs without SPIO (1 x 10(6)) injections into the infarcted myocardium approximately 1 hour following left anterior descending coronary artery. MRI (1.5-T) was performed 20 to 24 hours post infarction in all animals and the animals were subsequently sacrificed for histology 1 hour post MRI.RESULTS:In vitro Prussian Blue staining and electron microscopy examination revealed numerous iron particles in the cytoplasm of MSCs. Low signal intensity spots with the scanning T(2)(*)WI-Flash 2d sequence were detected in all SPIO-MSCs but not in SPIO-negative-MSCs injected myocardial sites in vivo with the clinical 1.5 T scanner. Prussian blue, DAPI and PKH(26) positive cells were detected histologically in sections corresponding to low signal intensity spots area shown on MRI.CONCLUSION:Magnetically labeled MSCs transplanted in myocardial ischemia area of swine can be visualized in vivo with a clinical 1.5-T MRI and could be used for tracking SPIO-MSCs clinically.
Study on MR Tracking of Magnetically Labeled Swine Mesenchymal Stem Cells after Autologous Transplantation into Acute Injured Liver in Vivo

Abstract:Objective To evaluate in vivo tracking of swine mesenchymal stem cells (MSCs) la-beled with super paramagnetic iron oxide (SPIO) in intraportal transplantation by a clinical 1.5T MR.Methods MSCs were isolated from swine and cultured as well as expanded, which were then incuba-ted with SPIO (Feridex I. V.). Prussian blue staining was performed for showing intracelluar irons.To establish a swine model of acute liver necrosis, 0.5 g/kg of D-galactosamine was administrated to 10 pigs. MSCs(labeled cells in six, unlabeled cells in four)were injected into liver via portal veins. MR imaging was performed with a clinical 1.5T MR immediately before and at 6 h, 3 d, 7 d, 14 d after transplantation, respectively. Results Prussian blue staining of SPIO labeled MSCs could be effec-tively labeled and the labeling efficiency was almost 100%. Signal intensity loss in liver by SPIO labe-ling on FFE sequence persisted until 14 days after transplantation. Histological analysis by Prussian blue staining showed homing of labeled MSCs in liver after 14 days, primarily distributing in hepatic sinusoids and liver parenchyma. Conclusion MSCs can be labeled with SPIO in vitro successfully.MRI can monitor magnetically labeled MSCs transplanted into liver.

Abstract:Mesenchymal stem cell (MSC)-based therapy has great potential for tissue regeneration. However, being able to monitor the in vivo behavior of implanted MSCs and understand the fate of these cells is necessary for further development of successful therapies and requires an effective, non-invasive and non-toxic technique for cell tracking. Super paramagnetic iron oxide (SPIO) is an idea label and tracer of MSCs. MRI can be used to follow SPIO-labeled MSCs and has been proposed as a gold standard for monitoring the in vivo biodistribution and migration of implanted SPIO-labeled MSCs. This review discusses the biological effects of SPIO labeling on MSCs and the therapeutic applications of local or systemic delivery of these labeled cells.
Efficiently tracking of stem cells in vivo using different kinds of uperparamagnetic iron oxide in swine with myocardial infarction

Abstract:Background Superparamagnetic iron oxide (SPIO) particles have shown much promise as a means to visualize labeled cells using molecular magnetic resonance imaging (MRI). Micrometer-sized superparamagnetic iron oxide (MPIO) particles and nanometer-sized ultrasmall superparamagnetic iron oxide (USPIO) are two kinds of SPIO widely used for monitoring stem cells migration. Here we compare the efficiency of two kinds of SPIO during the use of stem cells to treat acute myocardial infarction (AMI). Methods An AMI model in swine was created by 60 minutes of balloon occlusion of the left anterior descending coronary artery. Two kinds of SPIO particles were used to track after intracoronary delivered 107 magnetically labeled mesenchymal stem cells (MR-MSCs). The distribution and migration of the MR-MSCs were assessed with the use of 3.0T MR scanner and then the results were confirmed by histological examination. Results MR-MSCs appeared as a local hypointense signal on T-2(.)-weighted MRI and there was a gradual loss of the signal intensity after intracoronary transplantation. All of the hypointense signals in the USPIO-labeled group were found on T2(.)-weighted MRI, contrast to noise ratio (CNR) decreased in the MPIO-labeled group (16.07 +/- 5.85 vs. 10.96 +/- 1.34) and USPIO-labeled group (11.72 +/- 1.27 vs. 10.03 +/- 0.96) from 4 to 8 weeks after transplantation. However, the hypointense signals were not detected in MPIO-labeled group in two animals. MRI and the results were verified by histological examination. Conclusions We demonstrated that two kinds of SPIO particles in vitro have similar labeling efficiency and viability. USPIO is more suitable for labeling stem cells when they are transplanted via a coronary route. Chin Med J 2011;124(8):1199-1204
Tracking mesenchymal stem cells using magnetic resonance imaging

Abstract:Recent translational studies in the fields of tissue regeneration and cell therapy have characterized mesenchymal stem cells (MSCs) as a potentially effective and accessible measure for treating ischemic cerebral and neurodegenerative disorders such as stroke, Parkinson's disease, and amyotrophic lateral sclerosis. Developing more efficient cell tracking techniques bear the potential to optimize MSC transplantation therapies by providing a more accurate picture of the fate and area of effect of implanted cells. Currently, determining the location of transplanted MSCs involves a histological approach, but magnetic resonance imaging (MRI) presents a noninvasive paradigm that permits repeat evaluations. To visualize MSCs using MRI, the implanted cells must be treated with an intracellular contrast agent. These are commonly paramagnetic compounds, many of which are based on superparamagnetic iron oxide (SPIO) nanoparticles. Recent research has set out characterize the effects of SPIO-uptake on the cellular activity of in vitro human MSCs and the resultant influence that respective SPIO concentration has on MRI sensitivity. As these studies reveal, SPIO-uptake has no effect on the cellular processes of proliferation and differentiation while producing high contrast MRI signals. Moreover, transplantation of SPIO-labeled MSCs in animal models encouragingly showed no loss in MRI contrast, suggesting that SPIO labeling may be an appealing regime for lasting MRI detection. MRI Tracking of SPIO- and Fth1-Labeled Bone Marrow Mesenchymal Stromal Cell Transplantation for Treatment of Stroke

Abstract:We aimed to identify a suitable method for long-term monitoring of the migration and proliferation of mesenchymal stromal cells in stroke models of rats using ferritin transgene expression by magnetic resonance imaging (MRI). Bone marrow mesenchymal stromal cells (BMSCs) were transduced with a lentivirus containing a shuttle plasmid (pCDH-CMV-MCS-EF1-copGFP) carrying the ferritin heavy chain 1 (Fth1) gene. Ferritin expression in stromal cells was evaluated with western blotting and immunofluorescent staining. The iron uptake of Fth1-BMSCs was measured with Prussian blue staining. Following surgical introduction of middle cerebral artery occlusion, Fth1-BMSCs and superparamagnetic iron oxide- (SPIO-) labeled BMSCs were injected through the internal jugular vein. The imaging and signal intensities were monitored by diffusion-weighted imaging (DWI), T2-weighted imaging (T2WI), and susceptibility-weighted imaging (SWI) in vitro and in vivo. Pathology was performed for comparison. We observed that the MRI signal intensity of SPIO-BMSCs gradually reduced over time. Fth1-BMSCs showed the same signal intensity between 10 and 60 days. SWI showed hypointense lesions in the SPIO-BMSC (traceable for 30 d) and Fth1-BMSC groups. T2WI was not sensitive enough to trace Fth1-BMSCs. After transplantation, Prussian blue-stained cells were observed around the infarction area and in the infarction center in both transplantation models. Fth1-BMSCs transplanted for treating focal cerebral infarction were safe, reliable, and traceable by MRI. Fth1 labeling was more stable and suitable than SPIO labeling for long-term tracking. SWI was more sensitive than T2W1 and suitable as the optimal MRI-tracking sequence.

Abstract:Background: This study was to validate the feasibility of using clinical 3.0T MRI to monitor the migration of autotransplanted bone marrow (BM)-derived stem-progenitor cells (SPC) to the injured arteries of near-human sized swine for potential cell-based arterial repair.Methodology: The study was divided into two phases. For in vitro evaluation, BM cells were extracted from the iliac crests of 13 domestic pigs and then labeled with a T2 contrast agent, Feridex, and/or a fluorescent tissue marker, PKH26. The viability, the proliferation efficiency and the efficacies of Feridex and/or PKH26 labeling were determined. For in vivo validation, the 13 pigs underwent endovascular balloon-mediated intimal damages of the iliofemoral arteries. The labeled or un-labeled BM cells were autotransplanted back to the same pig from which the BM cells were extracted. Approximately three weeks post-cell transplantation, 3.0T T2-weighted MRI was performed to detect Feridex-created signal voids of the transplanted BM cells in the injured iliofemoral arteries, which was confirmed by subsequent histologic correlation.Principal Findings: Of the in vitro study, the viability of dual-labeled BM cells was 95-98%. The proliferation efficiencies of dual-labeled BM cells were not significantly different compared to those of non-labeled cells. The efficacies of Feridex- and PKH26 labeling were 90% and 100%, respectively. Of the in vivo study, 3.0T MRI detected the auto-transplanted BM cells migrated to the injured arteries, which was confirmed by histologic examinations.Conclusion: This study demonstrates the capability of using clinical 3.0T MRI to monitor the auto-transplantation of BM cells that migrate to the injured arteries of large animals, which may provide a useful MRI technique to monitor cell-based arterial repair.
Identification and Differentiation of Magnetically Labeled Mesenchymal Stem Cells in Vivo in Swines with Myocardial Infarction.

Abstract:We aim to track mesenchymal stem cells (MSCs) after magnetically labeling and test the ability of these cells differentiate into cardiomyocytes in vivo. Therefore, 20 swines were divided into four groups, sham-operated group (n=3); acute myocardial infarction (AMI) transplanted with PBS (n=3); labeled MSCs (n=7) and unlabeled MSCs (n=7) group. 10(7) labeled or unlabeled cells were intracoronary delivered after MI (4.8+/-1.3 days), and serial cardiac MR (3.0T) imaging studies were performed at 0, 4 and 8 weeks after transplantation, then the results were confirmed by histological and western blot analysis. We demonstrated that labeled MSCs can be reliably detected and tracked in vivo using MR imaging. In particular, we provided the evidence of regeneration of labeled MSCs in vivo by histological examination and western blot analysis.

Abstract:Aim To evaluate in vivo magnetic resonance imaging(MRI) for depiction and tracking of superparamagnetic iron oxide(SPIO) labeled swine mesenchymal stem cells(MSCs) in liver.Methods Mesenchymal stem cells(MSCs) were isolated from swine,cultured and expanded,then incubated with SPIO(Feridex I.V.).Prussian blue stain was performed for showing intracelluar irons.MSCs(labled cells in six,unlabled cells in four)were injected into swines liver via portal veins.MRI including T1 weighted imaging(T1WI),T2 weighted imaging(T2WI) and gradient-echo(GRE) sequences was performed before cells transplantation and 6 hours,3,7 days after transplantation.MR imaging ?ndings were compared histologically.Results Prussian blue staining of SPIO labled MSCs demonstrating cells could be effectively labeled and the labeling efficiency was almost 100%.SPIO labeling caused signal intensity loss in liver on GRE MR images.The signal change persisted until 7 days after transplantation.Prussian blue staining of histological analysis showed homing of labeled MSCs in liver after 7 days,primarily distributing in hepatic sinusoids and liver parenchyma.Conclusion SPIO can be used to label MSCs in vitro successfully.MRI can monitor magnetically labeled MSCs transplanted into liver.

Abstract:OBJECTIVE:To detect and track the presence of magnetically labeled bone marrow stromal cells (MSCs) in vivo with magnetic resonance imaging (MRI) after autologous transplantation.METHODS:The cultured and SPIO-labeled MSCs were transplanted into dog femoral head. The dog received regular MRI re-examination at post-operation. Samples were harvested at different weeks for analysis.RESULTS:The labeled cells showed an obviously low intensity of signal change in MRI. There were lots of labeled cells in the transplant area and more bone formation than the controls.CONCLUSION:The labeled cells can be visualized in vivo by MRI examination after transplantation into femoral head. The transplanted MSCs survive in vivo and increase bone formation.

Abstract:Introduction No comparative study of adipose-derived stem cells (ADSCs) and bone marrow mesenchymal stem cells (BMSCs) by using superparamagnetic iron oxide nanoparticles (SPIOs)-labeling and magnetic resonance imaging (MRI) has been performed. Methods We studied the biological activity and MRI of ADSCs by labeling them with SPIOs and comparing them with BMSCs. After incubating the cells in culture medium with different levels of SPIOs (control group: 0 μg/ml; Groups 1 to 3: 25, 50, and 100 μg/ml) for 24 hours, we compared ADSCs with BMSCs in terms of intracellular iron content, labeling efficiency, and cell viability. Stem cells in the culture medium containing 50 μg/ml SPIOs were induced into osteoblasts and fat cells. Adipogenic and osteogenic differentiation potentials were compared. R 2 * values of MRI in vitro were compared. Results The results showed that labeling efficiency was highest in Group 2. Intracellular iron content and R 2 * values increased with increasing concentrations of SPIOs, whereas cell viability decreased with increasing concentrations of SPIOs, and adipogenic and osteogenic differentiation potentials decreased. However, we found no significant difference between the two kinds of cells for any of these indexes. Conclusions ADSCs can be labeled and traced as easily as BMSCs in vitro . Given their abundance and higher proliferative capacity, as was previously shown, ADSCs may be better suited to stem cell therapy than are BMSCs.

Abstract:The aim of this study was to test the feasibility of using MRI to detect magnetically labeled, intravenously injected bone marrow (BM) cells homing to injured arteries. In the first phase, BM cells from LacZ-transgenic or green fluorescent protein (GFP)-transgenic mice were transplanted into eight recipient mice. The left femoral arteries of recipient mice were injured using a cuff-constriction or endothelium-damage approach, and the right femoral arteries were uninjured to serve as controls. The location and distribution of migrated LacZ-BM or GFP-BM cells were confirmed with histology. In the second phase, BM-derived cells from LacZ-transgenic mice were labeled with superparamagnetic iron oxide (Feridex) and then transplanted into eight recipient mice with cuff-induced injuries in the left femoral arteries. Migrated Feridex/LacZ-BM cells were monitored in vivo using a 4.7 T MR scanner. Subsequently, high-resolution ex vivo MRI was performed on 9.4 T and 11.7 T. LacZ-positive or GFP-positive cells in the thickened adventitia of the injured arteries were evident on histology. Both in vivo and ex vivo MRI showed larger regions of hypointensity with Feridex-labeled cells at the sites of the injured arteries compared with control arteries (P < 0.01). This study provides initial evidence that may support the potential use of MRI to detect homing of intravenously injected BM cells to injured arteries. Copyright (c) 2007 John Wiley & Sons, Ltd.

Abstract:AIM: To evaluate tracking of magnetically labeled mesenchymal stem cells (MSCs) after intraportal transplantation. METHODS: Mononuclear cells were isolated from bone marrow aspirates of pigs by density gradient centrifugation, cultured and expanded, after which, they were incubated with super paramagnetic iron oxide (SPIO). Prussian blue staining was performed to highlight intracellular iron. To establish swine models of acute liver injury, 0.5 g/kg D-galactosamine was administrated to 10 pigs, six of which were injected via their portal veins with SPIO-labeled MSCs, while the remaining four were injected with unlabeled cells. Magnetic resonance imaging (MRI) was performed with a clinical 1.5T MR scanner immediately before transplantation and 6 h, 3 d, 7 d and 14 d after transplantation. Prussian blue staining was again performed with the tissue slices at the endpoint. RESULTS: Prussian blue staining of SPIO-labeled MSCs had a labeling efficiency of almost 100%. Signal intensity loss in the liver by SPIO labeling on the FFE (T2*WI) sequence persisted until 14 d after transplantation. Histological analysis by Prussian blue staining confirmed homing of labeled MSCs in the liver after 14 d; primarily distributed in hepatic sinusoids and liver parenchyma. Abstract:Cell-based therapy with mesenchymal stem cells (MSCs) is a promising strategy for acute ischemic stroke. In vivo tracking of therapeutic stem cells with magnetic resonance imaging (MRI) is imperative for better understanding cellular survival and migrational dynamics over time. In this study, we develop a novel biocompatible nanocomplex (ASP-SPIONs) based on cationic amylose, by introducing spermine and the image label, ultrasmall superparamagnetic iron oxide nanoparticles (SPIONs), to label MSCs. The capacity, efficiency, and cytotoxicity of the nanocomplex in transferring SPIONs into green fluorescence protein-modified MSCs were tested; and the performance of in vivo MRI tracking of the transplanted cells in acute ischemic stroke was determined. The results demonstrated that the new class of SPIONs-complexed nanoparticles based on biodegradable amylose can serve as a highly effective and safe carrier to transfer magnetic label into stem cells. A reliable tracking of transplanted stem cells in stroke was achieved by MRI up to 6 weeks, with the desirable therapeutic benefit of stem cells on stroke retained. With the advantages of a relatively low SPIONs concentration and a short labeling period, the biocompatible complex of cationic amylose with SPIONs is highly translatable for clinical application. It holds great promise in efficient, rapid, and safe labeling of stem cells for subsequent cellular MRI tracking in regenerative medicine.

Abstract:The successful implementation of stem cell therapy requires the development of imaging techniques to track transplanted stem cells in vivo and monitor their fate over time. Most in vivo imaging methods focus on providing information about the distribution and migration of transplanted stem cells, while monitoring their viability-crucial for optimizing therapy-remains undeveloped. Although a few in vivo imaging techniques have been developed for assessing the viability of transplanted stem cells, their applicability is restricted by limited penetration depth. To address this, magnetic resonance imaging (MRI) that is collaboratively utilized with tailored contrast agents has been explored to track the viability of transplanted stem cells without this limitation. However, currently available MRI contrast agents struggle with low accuracy in monitoring viability due to poor controllability or minimal signal change in response to cell death. Here, we present an extremely small iron oxide nanoparticles (ESIONPs)-based T1-T2 switchable MRI contrast agent (ESIONPs-GSH) designed to accurately detect cell apoptosis in response to elevated reactive oxygen species (ROS).Specifically, ESIONPs-GSH was obtained by modifying the surface of ESIONPs with ROS-sensitive glutathione (GSH) and amphipathic 3-((3-aminopropyl) dimethylammonio) propane-1-sulfonate (ADPS) molecules. The results demonstrate that ESIONPs-GSH is biocompatible, with negligible effects on the proliferation and differentiation of bone mesenchymal stem cells (BMSCs) post-labeling. Additionally, BMSCs labeled with ESIONPs-GSH exhibit T1 contrast; upon cell death, increasing cellular ROS oxidize GSH and induce cross-linking of ESIONPs-GSH, resulting in a switch to T2 contrast. Benefiting from the significantly contrasting signal following the T1-T2 contrast switch, ESIONPs-GSH allows for accurate assessment of the survival of BMSCs transplanted into the axillary regions of mice through MRI monitoring. ESIONPs-GSH enabled MRI can effectively track transplanted stem cell viability in vivo without penetration depth limitations, making it a promising tool for guiding stem cell-based therapies.
