Mathematical Modelling of Natural Phenomena

Abstract:Atherosclerosis is a chronic disease which involves the build up of cholesterol and fatty deposits within the inner lining of the artery. It is associated with a progressive thickening and hardening of the arterial wall that result in narrowing of the vessel lumen and restriction of blood flow to vital organs. These events may cause heart attack or stroke, the commonest causes of death worldwide. In this paper we study the early stages of atherosclerosis via a mathematical model of partial differential equations of reaction-diffusion type. The model includes several key species and identifies endothelial hyperpermeability, believed to be a precursor on the onset of atherosclerosis. For simplicity, we reduce the system to a monotone system and provide a biological interpretation for the stability analysis according to endothelial functionality. We investigate as well the existence of solutions of traveling waves type along with numerical simulations. The obtained results are in good agreement with current biological knowledge. Likewise, they confirm and generalize results of mathematical models previously performed in literature. Then, we study the non monotone reduced model and prove the existence of perturbed solutions and perturbed waves, particularly in the bistable case. Finally, we extend the study by considering the complete model proposed initially, perform numerical simulations and provide more specific results. We study the consistency between the reduced and complete model analysis for a certain range of parameters, we elaborate bifurcation diagrams showing the evolution of inflammation upon endothelial permeability and LDL accumulation and we consider the effect of anti-inflammatory process on the system behavior. In this model, the regulation of atherosclerosis progression is mediated by anti-inflammatory responses that, up to certain extent, lead to plaque regression.

Abstract:Atherosclerosis is a chronic inflammatory disease which occurs due to plaque accumulation in the intima, the innermost layer of the artery. In this paper, a simple reaction–diffusion mathematical model of the plaque formation process comprising of oxidized LDL and macrophages has been developed. Linear stability analysis of the non-spatial model leads to the existence of global stability of the kinetic system. This reveals that the non-spatial system can withstand a substantial change in the significant model parameter values which can be taken forward for further clinical investigations. Numerical bifurcation analysis of the non-spatial system confirms the existence of Hopf bifurcation with respect to two significant model parameters. The biological importance of these bifurcation diagrams is discussed in detail. The significance of the model presented in this research paper provides a clear insight into the role of the key constituents, oxidized LDL and macrophages, involved in the plaque-forming process.

Abstract:Some existing models of the atherosclerosis development are discussed and a new improved mathematical model, which takes into account new experimental results about diverse roles of macrophages in atherosclerosis, is proposed. Using technic of upper and lower solutions, the existence and uniqueness of its positive solution are justified. After the nondimensionalisation, small parameters are found. Then asymptotic approximation for the solution is constructed and justified with the help of asymptotic methods for boundary-value problems in thin domains. The results argue for the possibility to replace the complex $3D$ (dimensional) mathematical model with the corresponding simpler $2D$ model with sufficient accuracy measured by these small parameters.

Abstract:Atherosclerosis is a leading cause of death in the United States and worldwide; it emerges as a result of multiple dynamical cell processes including hemodynamics, endothelial damage, innate immunity and sterol biochemistry. Making matters worse, nearly 21 million Americans have diabetes, a disease where patients' cells cannot efficiently take in dietary sugar, causing it to build up in the blood. In part because diabetes increases atherosclerosis-related inflammation, diabetic patients are twice as likely to have a heart attack or stroke. Past work has shown that hyperglycemia and insulin resistance alter function of multiple cell types, including endothelium, smooth muscle cells and platelets, indicating the extent of vascular disarray in this disease. Although the pathophysiology of diabetic vascular disease is generally understood, there is no mathematical model to date that includes the effect of diabetes on plaque growth. In this paper, we propose a mathematical model for diabetic atherosclerosis; the model is given by a system of partial differential equations with a free boundary. We establish local existence and uniqueness of solution to the model. The methodology is to use Hanzawa transformation to reduce the free boundary to a fixed boundary and reduce the system of partial differential equations to an abstract evolution equation in Banach spaces, and apply the theory of analytic semigroup.

Abstract:Atherosclerosis is a chronic inflammatory disease occurs due to plaque accumulation in the inner artery wall. In atherosclerotic plaque formation monocytes and macrophages play a significant role in controlling the disease dynamics. In the present article, the entire biochemical process of atherosclerotic plaque formation is presented in terms of an autonomous system of nonlinear ordinary differential equations involving concentrations of oxidized low-density lipoprotein (LDL), monocytes, macrophages, and foam cells as the key dependent variables. To observe the capacity of monocytes and macrophages the model has been reduced to a two-dimensional temporal model using quasi steady state approximation theory. Linear stability analysis of the two-dimensional ordinary differential equations (ODEs) model has revealed the stability of the equilibrium points in the system. We have considered both one- and two- parameter bifurcation analysis with respect to parameters associated to the rate at which macrophages phagocytose oxidised LDL and the rate at which LDL enters into the intima. The bifurcation diagrams reveal the oscillating nature of the curves representing concentration of monocytes and macrophages with respect to significant model parameters. We are able to find the threshold values at which the plaque accumulation accelerates in an uncontrollable way. Further to observe the impact of diffusion, a spatiotemporal model has been developed. Numerical investigation of the partial differential equations (PDEs) model reveals the existence of travelling wave in the system which ensures the fact that the development of atherosclerotic plaque formation follows reaction-diffusion wave.

Making matters worse, nearly 463 million people have diabetes, which increases atherosclerosis-related inflammation, diabetic patients are twice as likely to have a heart attack or stroke. The pathophysiology of diabetic vascular disease is generally understood. Dyslipidemia with increased levels of atherogenic low-density lipoprotein, hyperglycemia, oxidative stress and increased inflammation are factors that increase the risk and accelerate development of atherosclerosis. In a recent paper, Xie [Well-posedness of a mathematical model of diabetic atherosclerosis. J Math Anal Appl. 2022;505(2):125606], we have developed a mathematical model that includes the effect of hyperglycemia and insulin resistance on plaque growth. In this paper, we propose a more comprehensive mathematical model for diabetic atherosclerosis which include more variables; in particular, it includes the variable for Advanced Glycation End-Products (AGEs) concentration. Hyperglycemia trigger vascular damage by forming AGEs, which are not easily metabolized and may accelerate the progression of vascular disease in diabetic patients. The model is given by a system of partial differential equations (PDEs) with a free boundary. We also establish the local existence and uniqueness of solution to the model. The methodology is to reduce the free boundary to a fixed boundary and the system of PDEs to an abstract evolution equation in Banach spaces, and apply the theory of analytic semigroup.

Abstract:This work is devoted to the development of a mathematical model of the early stages of atherosclerosis incorporating processes of all time scales of the disease and to show their interactions. The cardiovascular mechanics is modeled by a fluid–structure interaction approach coupling a non-Newtonian fluid to a hyperelastic solid undergoing anisotropic growth and a change of its constitutive equation. Additionally, the transport of low-density lipoproteins and its penetration through the endothelium is considered by a coupled set of advection–diffusion-reaction equations. Thereby, the permeability of the endothelium is wall-shear stress modulated resulting in a locally varying accumulation of foam cells triggering a novel growth and remodeling formulation. The model is calibrated and applied to an murine-specific case study, and a qualitative validation of the computational results is performed. The model is utilized to further investigate the influence of the pulsatile blood flow and the compliance of the artery wall to the atherosclerotic process. The computational results imply that the pulsatile blood flow is crucial, whereas the compliance of the aorta has only a minor influence on atherosclerosis. Further, it is shown that the novel model is capable to produce a narrowing of the vessel lumen inducing an adaption of the endothelial permeability pattern.

Abstract:Atherosclerosis is one of the principle pathologies of cardiovascular disease with blood cholesterol a significant risk factor. The World Health Organisation estimates that approximately 2.5 million deaths occur annually due to the risk from elevated cholesterol with 39% of adults worldwide at future risk. Atherosclerosis emerges from the combination of many dynamical factors, including haemodynamics, endothelial damage, innate immunity and sterol biochemistry. Despite its significance to public health, the dynamics that drive atherosclerosis remain poorly understood. As a disease that depends on multiple factors operating on different length scales, the natural framework to apply to atherosclerosis is mathematical and computational modelling. A computational model provides an integrated description of the disease and serves as an in silico experimental system from which we can learn about the disease and develop therapeutic hypotheses. Although the work completed in this area to-date has been limited, there are clear signs that interest is growing and that a nascent field is establishing itself. This paper discusses the current state of modelling in this area, bringing together many recent results for the first time. We review the work that has been done, discuss its scope and highlight the gaps in our understanding that could yield future opportunities.

Abstract:Atherosclerosis is a leading cause of death worldwide. Making matters worse, nearly 463 million people have diabetes, which increases atherosclerosis-related inflammation. Diabetic patients are twice as likely to have a heart attack or stroke. In this paper, we consider a simplified mathematical model for diabetic atherosclerosis involving LDL, HDL, glucose, insulin, free radicals (ROS), & beta; cells, macrophages and foam cells, which satisfy a system of partial differential equations with a free boundary, the interface between the blood flow and the plaque. We establish the existence of small radially symmetric stationary solutions to the model and study their stability. Our analysis shows that the plague will persist due to hyperglycemia even when LDL and HDL are in normal range, hence confirms that diabetes increase the risk of atherosclerosis.

Abstract:Cardiovascular diseases associated with high cholesterol (hypercholesterolemia) and low-density lipoproteins (LDL) levels are significant contributors to total mortality in developing and developed countries. Mathematical modeling of LDL metabolism is an important step in the development of drugs for hypercholesterolemia. The aim of this work was to develop and to analyze an integrated mathematical model of cholesterol metabolism in liver cells and its interaction with two types of drugs, statins and PCSK9 inhibitors. The model consisted of 21 ordinary differential equations (ODE) describing cholesterol biosynthesis and lipoprotein endocytosis in liver cells in vitro. The model was tested for its ability to mimic known biochemical effects of familial hypercholesterolemia, statin therapy, and PCSK9 inhibitors. The model qualitatively reproduced the well-known biology of cholesterol regulation, which confirms its potential for minimizing cellular research in initial testing of new drugs for cardiology.
Developing Theoretical Models for Atherosclerotic Lesions: A Methodological Approach Using Interdisciplinary Insights

Abstract:Atherosclerosis, a leading cause of cardiovascular disease, necessitates advanced and innovative modeling techniques to better understand and predict plaque dynamics. The present work presents two distinct hypothetical models inspired by different research fields: the logistic map from chaos theory and Markov models from stochastic processes. The logistic map effectively models the nonlinear progression and sudden changes in plaque stability, reflecting the chaotic nature of atherosclerotic events. In contrast, Markov models, including traditional Markov chains, spatial Markov models, and Markov random fields, provide a probabilistic framework to assess plaque stability and transitions. Spatial Markov models, visualized through heatmaps, highlight the spatial distribution of transition probabilities, emphasizing local interactions and dependencies. Markov random fields incorporate complex spatial interactions, inspired by advances in physics and computational biology, but present challenges in parameter estimation and computational complexity. While these hypothetical models offer promising insights, they require rigorous validation with real-world data to confirm their accuracy and applicability. This study underscores the importance of interdisciplinary approaches in developing theoretical models for atherosclerotic plaques.
Mathematical model of hypertension-induced arterial remodeling: A chemo-mechanical approach

Abstract:The development of chronic hypertension is a poorly described process involving many chemical and structural changes to the artery. Typically, mathematical models of this disease focus primarily on the mechanical aspects such as arterial geometry, elasticity, and tissue content, or alternatively on the chemical drivers of vasoactivity such as nitric oxide and reactive oxygen species. This paper presents a model that considers the powerful interaction between mechanical and biochemical drivers of hypertension and arterial remodeling. Based on biological processes thought to be involved in the development of hypertension, we have built a system of algebraic, differential, and integral equations. Endothelial dysfunction, which is known to limit vasodilation, is explicitly considered in the model and plays a vital role in the development of chronic hypertension. Numerical solutions to the system are consistent with available experimental data for normal and spontaneously-hypertensive rats.
Self-Organization and Chaos in the Metabolism of Hemostasis in a Blood Vessel

Abstract:A mathematical model of the metabolic process of formation of the hemostasis in a blood-carrying vessel is constructed. As distinct from the earlier developed model of the multienzyme prostacyclin-thromboxane system of blood, this model includes, for the first time, the influence of the level of "bad cholesterol", i.e., low-density lipoproteins (LDLs), on the hemostasis. The conditions, under which the self-organization of the system appears, and the modes of autooscillations and chaos in the metabolic process, which affects the formation of hemostasis and the development of thrombophilia, are found. With the aid of a phase-parametric diagram, the scenario of their appearance is studied. The bifurcations of doubling of a period and the transition to chaotic oscillations as a result of the intermittence are found. The obtained strange attractors are formed due to a mixing funnel. The full spectra of Lyapunov's indices, KS-entropies, "predictability horizons", and Lyapunov's dimensions of strange attractors are calculated. The reasons for a change in the cyclicity of the given metabolic process, its stability, and the physiological manifestation in the blood-carrying system are discussed. The role of physiologically active substances in a decrease in the level of cholesterol in blood vessels is estimated.

Abstract:Corresponding to human anatomical structure,a bond graph model for cardiovascular circulation system including heart,head and neck circulation,pulmonary circulation,coronary circulation,abdomen circulation and extremity circulation was developed,which comprise of 54 mathematical equations.The blood pressure and blood flow wave on arbitrary site of circulation can be obtained by solving the equations,meanwhile,the stroke volume,cardiac output and eject fraction of heart can be computed.The model achieved the typical physiological parameters such as,systolic/diastolic blood pressure 115/78 mmHg,stroke volume 80 mL,cardiac output 5.76 L and eject fraction 64.5%.The simulation results were in agreement with the human physiological experimental data,so that the validity of the model was verified.By adjusting the initial condition parameters of the model,the simulations for pathological features of the cardiovascular system,such as atherosclerosis,hypertension and stenosis in left subclavicle artery were implemented,with the results being consistent with human pathological features.Furthermore,the components of the model are composed of identical but independent structures,so the model is flexible to be extended.

Abstract:Cardiovascular diseases are the leading cause of death. Increased levels of plasma cholesterol are consistently associated with an increased risk of cardiovascular disease. As a result, it is imperative that studies are conducted to determine the best course of action to reduce whole-body cholesterol levels. A whole-body mathematical model for cholesterol metabolism and transport is proposed. The model can simulate the effects of lipid-lowering drugs like statins and anti-PCSK9. The model is based on ordinary differential equations and kinetic functions. It has been validated against literature data. It offers a versatile platform for designing personalized interventions for cardiovascular health management.

Abstract:Limited progress in the mathematical modeling of cholesterol transport systems is hampering novel therapeutic interventions. This issue is addressed by the present study through precise design, employing four compartmental models to elucidate cholesterol dynamics in the comprehensive bloodstream. Disparities in medical advancements, particularly in cholesterol-related pathophysiology, are aimed to be bridged, advancing medical science and patient care outcomes. Therapeutic strategies for reducing blood cholesterol are explored by the model, with parameter influences on equilibrium stability revealed through sensitivity analysis. System parameters are effectively manipulated by imposing sensitivity analysis, and pinpointing areas for model refinement. Stability analysis contributes to diverse realistic models, confirming local asymptotic stability. Model efficacy in studying cholesterol transport dynamics is supported by analytical and numerical assessments. The study concludes with the present model validation to substantiate it by comparing the present outcomes with the existing ones.

Abstract:The recent failure of candidate drugs like cholesterol ester transfer protein (CETP) and acyl-CoA:cholesterol acyltransferase (ACAT) inhibitors calls for a revised approach for screening anti-atherosclerotic drugs and development of new models of atherosclerosis. For this it is important to understand the mechanism of the disease in a particular model. Models simultaneously showing hyperlipidemia, inflammation and associated complications of diabetes and hypertension will serve the purpose better as they mimic the actual clinical condition. Besides this, analyzing candidate molecules in vivo, in vitro and at various levels of atherosclerosis progression is important. Models based on various cells and process involved in atherosclerosis should be used for screening candidate molecules. The challenge lies in bridging the gap between genetically friendly small animal and human-like bigger animal models. Sequencing of the mouse and human genome, development of a single nucleotide polymorphism (SNP) database and in silico quantitative trait loci (QTL) linkage analysis may enhance the understanding of atherosclerosis and help develop new therapeutic targets.

Abstract:We present here a population structured model to describe the dynamics of macrophage cells. The model involves the interactions between modified LDL, monocytes/macrophages, cytokines and foam cells. The key assumption is that the individual macrophage dynamics depends on the amount of lipoproteins it has internalized. The obtained renewal equation is coupled with an ODE describing the lipoprotein dynamics. We first prove global existence and uniqueness for the nonlinear and nonlocal system. We then study long time asymp-totics in a particular case describing silent plaques which undergo periodic rupture and repair. Abstract:Using the classical tools of nonlinear dynamics, we study the process of self-organization and the appearance of the chaos in the metabolic process in a cell with the help of a mathematical model of the transformation of steroids by a cell Arthrobacter globiformis. We constructed the phase-parametric diagrams obtained under a variation of the dissipation of the kinetic membrane potential. The oscillatory modes obtained are classified as regular and strange attractors. We calculated the bifurcations, by which the self-organization and the chaos occur in the system, and the transitions "chaos-order", "order-chaos", "order-order", and "chaos-chaos" arise. Feigenbaum's scenarios and the intermittences are found. For some selected modes, the projections of the phase portraits of attractors, Poincaré sections, and Poincaré maps are constructed. The total spectra of Lyapunov indices for the modes under study are calculated. The structural stability of the attractors is demonstrated. A general scenario of the formation of regular and strange attractors in the given metabolic process in a cell is found. The physical nature of their appearance in the metabolic process is studied.
