Abstract:Exchange protein directly activated by cAMP (Epac) is an important molecule in cAMP signal transduction, but the effect of Epac on lipopolysaccharide (LPS)-induced acute lung injury (ALI) is unclear. In this study, we treated in vitro and in vivo models with the Epac activator 8CPT to determine the effect and related mechanisms of Epac. The in vitro results indicate that 8CPT inhibits lipopolysaccharide (LPS)-induced tumor necrosis factor-α (TNF-α) release from mouse macrophages (MH-S), whereas the protein kinase A (PKA) activator 6BnZ has no effect. Furthermore, Epac over-expression can significantly suppress TNF-α release from LPS induced MH-S cell, while Epac siRNA can slightly increase TNF-α release. Moreover, 8CPT reduces LPS-induced microvascular permeability in human pulmonary microvascular endothelial cells (HPMVECs), whereas the PKA activator 6BnZ has no effect. In mice with LPS-induced ALI, 8CPT significantly reduces LPS-induced inflammatory cytokine release, neutrophil recruitment, and albumin leakage. LPS simultaneously decreases the Epac but not the PKA levels. However, 8CPT reverses the decreased Epac levels. Furthermore, the mechanism involves the small GTPase Rac1/2 but not the mitogen-activated protein kinase (MAPK) pathway. Thus, Epac activation reduces inflammation and microvascular permeability in LPS-induced lung injury and an Epac activator represents a novel choice for the early therapy of ALI.
The Role of miR-330-3p/PKC-α Signaling Pathway in Low-Dose Endothelial-Monocyte Activating Polypeptide-II Increasing the Permeability of Blood-Tumor Barrier.

Abstract:This study was performed to determine whether EMAP II increases the permeability of the blood-tumor barrier (BTB) by affecting the expression of miR-330-3p as well as its possible mechanisms. We determined the over-expression of miR-330-3p in glioma microvascular endothelial cells (GECs) by Real-time PCR. Endothelial monocyte-activating polypeptide-II (EMAP-II) significantly decreased the expression of miR-330-3p in GECs. Pre-miR-330-3p markedly decreased the permeability of BTB and increased the expression of tight junction (TJ) related proteins ZO-1, occludin and claudin-5, however, anti-miR-330-3p had the opposite effects. Anti-miR-3303p could enhance the effect of EMAP-II on increasing the permeability of BTB, however, pre-miR-330-3p partly reversed the effect of EMAP-II on that. Similarly, anti-miR-330-3p improved the effects of EMAP-II on increasing the expression levels of PKC-alpha and p-PKC-alpha in GECs and pre-miR-330-3p partly reversed the effects. MiR-330-3p could target bind to the 3 0 UTR of PKC-alpha. The results of in vivo experiments were similar to those of in vitro experiments. These suggested that EMAP-II could increase the permeability of BTB through inhibiting miR-330-3p which target negative regulation of PKC-alpha. Pre-miR-330-3p and PKC-a inhibitor decreased the BTB permeability and up-regulated the expression levels of ZO-1, occludin and claudin-5 while anti-miR-330-3p and PKC-a activator brought the reverse effects. Compared with EMAP-II, anti-miR-330-3p and PKC-a activator alone, the combination of the three combinations significantly increased the BTB permeability. EMAP-II combined with anti-miR-330-3p and PKC-alpha activator could enhance the DOX's effects on inhibiting the cell viabilities and increasing the apoptosis of U87 glioma cells. Our studies suggest that low-dose EMAP-II up-regulates the expression of PKC-a and increases the activity of PKC-alpha by inhibiting the expression of miR-330-3p, reduces the expression of ZO-1, occludin and claudin-5, and thereby increasing the permeability of BTB. The results can provide a new strategy for the comprehensive treatment of glioma.
ERK1/2 Pathway-Mediated Norepinephrine Regulates Function of Endothelial Progenitor Cells

Abstract:Objective To analyze the effects of norepinephrine(NE) on the proliferation and migration of endothelial progenitor cells(EPCs) and the related mechanism.Methods NE,adrenoceptor antagonist,and MAPK signal pathway blocker of various concentrations were used to treat peripheral EPCs derived from healthy adults.The proliferation potential,migration capacity and activation of ERK1/2 were assessed after different treatments.Results NE increased the proliferation potential of EPCs in a dose-dependent manner.The number of EPCs increased by(48.3±23.3)%,(70.5±35.6)%,(82.4±14.9)% and(100.3±48.1)% after treatment with NE at 0.01 μmol/L,0.1 μmol/L,1 μmol/L and 10 μmol/L,respectively.Addition of alpha adrenoceptor antagonist phentolamine,selective beta 2 adrenoceptor antagonist I127,JNK blocker SP600125 and ERK1/2 blocker A6355 could block the above effects of NE,and beta 1 adrenoceptor antagonist metoprolol and p38 blocker PD169318 failed to block the effects of NE.NE at 10 μmol/L significantly promoted the migration of EPCs(P0.05).These effects could be blocked by addition of phentolamine(10 μmol/L) and I127(10 μmol/L),but not by addition of metoprolol.NE(0.1,1 and 10 μmol/L) activated ERK1/2 pathway in a dose-dependent manner,which could also be blocked by phentolamine and I127,but not by metoprolol.Conclusion NE can increase the proliferation potential and migration capacity of EPCs via activating ERK1/2 pathway with alpha and beta 2 adrenoceptor.

The Role of RhoA in Neovascular-Related Functions of Endothelial Progenitor Cells Induced by AngiotensinⅡ

Abstract:Background/Aims: Interference with endothelial progenitor cell (EPC) neovascularization is a novel therapeutic target for neovascular-related diseases. Angiotensin Ⅱ (Ang Ⅱ) was found to enhance new vessel formation and aggravated neovascular-related diseases. In this study, we investigated the effects of Ang Ⅱ on EPC neovascular-related functions and explored the underlying mechanisms. Methods: EPCs were cultured from bone marrow derived mononuclear cells. The effects of Ang Ⅱ on EPC proliferation, adhesion, migration, and in vitro tube formation were investigated using the MTT assay, adhesion assay, transwell chamber assay, and in vitro tube formation assay respectively. The underlying mechanisms were explored using Western blotting assay. Results: EPC adhesion, migration and in vitro tube formation were promoted by Ang Ⅱ, and the effects were reversed by RhoA/Rho-associated kinases (ROCK) signaling pathway inhibitors including C3 exoenzyme, GGTI-286 and Y-27632. The active form of RhoA was up-regulated by Ang Ⅱ and this effect was abolished by C3 exoenzyme. Moreover, RhoA silencing resulted in a notable inhibition of EPC adhesion, migration and in vitro tube formation, suggesting that RhoA activation played a pivotal role in Ang Ⅱ angiogenic effect. The results also demonstrated that phosphorylation of p38 mitogen-activated protein kinase (MAPK) and c-Jun-NH2 kinase was elevated by Ang Ⅱ and attenuated by C3 exoenzyme, GGTI-286 and Y-27632. The enhancing effects of Ang Ⅱ on EPC adhesion, migration and in vitro vasculogenesis were reversed by p38 inhibitor SB202190 and JNK inhibitor SP600125. Conclusion: Ang Ⅱ may enhance EPC neovascular-related functions through activating RhoA/ ROCK and MAPK signaling pathway.
PAR1 Regulates Sepsis-Induced Vascular Endothelial Barrier Dysfunction by Mediating ERM Phosphorylation Via the RhoA/ROCK Signaling Pathway.

Abstract:Sepsis begins with vascular endothelial barrier breakdown and causes widespread organ failure. Protease-activated receptor 1 (PAR1) is an important target for modulating vascular endothelial permeability; however, little research has been undertaken in sepsis, and its putative molecular mechanism remains unknown. The vascular endothelial permeability was examined by detecting FITC-dextran flux. F-actin was examined by immunofluorescence (IF). PAR1, ERM phosphorylation, and RhoA/ROCK signaling pathway expression in lipopolysaccharide (LPS)-stimulated human umbilical vein endothelial cells (HUVECs) line were examined by IF and Western blot. To develop the sepsis model, cecal ligation and puncture (CLP) were conducted. The PAR1 inhibitor SCH79797 was utilized to inhibit PAR1 expression in vivo. Vascular permeability in main organs weres measured by Evans blue dye extravasation. The pathological changes in main organs were examined by HE staining. The expression of PAR1, ERM phosphorylation, and the RhoA/ROCK signaling pathway was examined using IF, immunohistochemical and WB in CLP mice. In vitro, in response to LPS stimulation of HUVECs, PAR1 mediated the phosphorylation of ERM, promoted F-actin rearrangement, and increased endothelial hyperpermeability, all of which were prevented by inhibiting PAR1 or RhoA. Additionally, inhibiting PAR1 expression reduced RhoA and ROCK expression. In vivo, we showed that inhibiting PAR1 expression will reduce ezrin/radixin/moesin (ERM) phosphorylation to relieve vascular endothelial barrier dysfunction and thereby ameliorate multiorgan dysfunction syndrome (MODS) in CLP-induced septic mice. This study revealed that PAR1-mediated phosphorylation of ERM induced endothelial barrier dysfunction, which in turn led to MODS in sepsis, and that the RhoA/ROCK signaling pathway underlay these effects.
The EPAC-Rap1 pathway prevents and reverses cytokine-induced retinal vascular permeability

Abstract:Increased retinal vascular permeability contributes to macular edema, a leading cause of vision loss in eye pathologies such as diabetic retinopathy, age-related macular degeneration, and central retinal vein occlusions. Pathological changes in vascular permeability are driven by growth factors such as VEGF and pro-inflammatory cytokines such as TNF-α. Identifying the pro-barrier mechanisms that block vascular permeability and restore the blood-retinal barrier (BRB) may lead to new therapies. The cAMP-dependent guanine nucleotide exchange factor (EPAC) exchange-protein directly activated by cAMP promotes exchange of GTP in the small GTPase Rap1. Rap1 enhances barrier properties in human umbilical endothelial cells by promoting adherens junction assembly. We hypothesized that the EPAC-Rap1 signaling pathway may regulate the tight junction complex of the BRB and may restore barrier properties after cytokine-induced permeability. Here, we show that stimulating EPAC or Rap1 activation can prevent or reverse VEGF- or TNF-α-induced permeability in cell culture and in vivo Moreover, EPAC activation inhibited VEGF receptor (VEGFR) signaling through the Ras/MEK/ERK pathway. We also found that Rap1B knockdown or an EPAC antagonist increases endothelial permeability and that VEGF has no additive effect, suggesting a common pathway. Furthermore, GTP-bound Rap1 promoted tight junction assembly, and loss of Rap1B led to loss of junctional border organization. Collectively, our results indicate that the EPAC-Rap1 pathway helps maintain basal barrier properties in the retinal vascular endothelium and activation of the EPAC-Rap1 pathway may therefore represent a potential therapeutic strategy to restore the BRB.

Abstract:BACKGROUND: Endothelial-Monocyte Activating Polypeptide (EMAP II) is a secreted protein with well-established anti-angiogenic activities. Intracellular EMAP II expression is increased during fetal development at epithelial/mesenchymal boundaries and in pathophysiologic fibroproliferative cells of bronchopulmonary dysplasia, emphysema, and scar fibroblast tissue following myocardial ischemia. Precise function and regulation of intracellular EMAP II, however, has not been explored to date.METHODOLOGY/PRINCIPAL FINDINGS: Here we show that high intracellular EMAP II suppresses cellular proliferation by slowing progression through the G2M cell cycle transition in epithelium and fibroblast. Furthermore, EMAP II binds to and is phosphorylated by Cdk1, and exhibits nuclear/cytoplasmic partitioning, with only nuclear EMAP II being phosphorylated. We observed that extracellular secreted EMAP II induces endothelial cell apoptosis, where as excess intracellular EMAP II facilitates epithelial and fibroblast cells migration.CONCLUSIONS/SIGNIFICANCE: Our findings suggest that EMAP II has specific intracellular effects, and that this intracellular function appears to antagonize its extracellular anti-angiogenic effects during fetal development and pulmonary disease progression.

Abstract:The blood-tumor barrier (BTB) hinders delivery of chemotherapeutic drugs to tumors in the brain; previous studies have shown that the BTB can be selectively opened by endothelial monocyte activating polypeptide-II (EMAP-II), but the specific mechanism involved remains elusive. In this study, we found that microRNA-429 (miR-429) expression in glioma vascular endothelial cells (GECs) was far lower than in human brain microvascular endothelial cells (ECs). miR-429 had lower expression in GECs and glioma tissues compared to ECs or normal tissues of the brain. Furthermore, miR-429 had lower expression in high grade glioma (HGG) than in low grade glioma (LGG). In in vitro BTB models, we also found that EMAP-II significantly increased BTB permeability, decreased expression of ZO-1, occludin and claudin-5 in GECs, in a time- and dose-dependent manner. EMAP-II greatly increased miR-429 expression in GECs of the BTB models in vitro. Overexpression of miR-429 in GECs significantly decreased the transepithelial electric resistance (TEER) values in BTB models, and led to enhanced horseradish peroxidase (HRP) flux. Overexpression of miR-429 in GECs significantly decreased the expression of tight junction (TJ)-associated proteins (ZO-1, occludin and claudin-5), and decreased the distribution continuity. Silencing of miR-429 in GECs increased the expression of TJ-associated proteins and the distribution continuity. The dual-luciferase reporter assay revealed that ZO-1 and occludin were target genes of miR-429, and we demonstrated that miR-429 overexpression markedly down-regulated protein expression of p70S6K, as well as its phosphorylation levels. The dual-luciferase reporter assay also showed that p70S6K was a target gene of miR-429; miR-429 overexpression down-regulated expression and phosphorylation levels of p70S6K, and also decreased phosphorylation levels of S6 and increased BTB permeability. Conversely, silencing of miR-429 increased the expression and phosphorylation levels of p70S6K, and increased phosphorylation levels of S6, while decreasing BTB permeability. In conclusion, the results indicated that EMAP-II caused an increase in miR-429 expression that directly targeted TJ-associated proteins, which were negatively regulated; on the other hand, miR-429 down-regulated the expression of TJ-associated proteins by targeting p70S6K, also negatively regulated. As a result, the BTB permeability increased.
Effects of rapamycin on number and activity of endothelial progenitor cells

Abstract:Objective To investigate the effects of rapamycin on the number and function of peripheral blood endothelial progenitor cells (EPCs). Methods Total mononuclear cells (MNCs) were isolated from peripheral blood by Ficoll density gradient centrifugation, and then the cells were plated on fibronectin-coated culture dishes. After 7-day culture, adherent cells were treated with rapamycin in a series of final concentrations of 1.0, 2.0, 5.0μg/ml for 6, 12, 24, and 48h. EPCs were identified as adherent cells double positive for DiLDL-uptake and lectin binding by direct fluorescent staining as demonstrated under a laser scanning confocal microscope. EPCs were further documented by demonstrating the expression of VEGFR-2, AC133 and CD34 with flow cytometry. EPCs proliferation and migration were assayed with MTT assay and modified Boyden chamber assay, respectively. EPCs adhesion assay was performed by replating them on fibronectin-coated dishes, and then adherent cells were counted. In vitro vasculogenesis activity was assayed by in vitro vasculogenesis kit. Results Incubation of isolated human MNCs with rapamycin resulted in a decrease in the number of EPCs, and rapamycin also decreased EPCs proliferative, migratory, adhesive and in vitro vasculogenesis capacity in both concentration and time dependent manners. Conclusion Rapamycin decreases the number, proliferative, migratory, adhesive and in vitro vasculogenesis capacity of EPCs.
TACE-Mediated Ectodomain Shedding of the Type I TGF-b Receptor Downregulates TGF-b Signaling

Abstract:Regulating TGF-b receptor presentation provides an avenue to alter a cell’s responsiveness to TGF-b. We report that activation of the Erk MAP kinase pathway decreases the TGF-b-induced Smad3 activation due to decreased cell surface levels of the type I receptor TbRI, but not the type II receptor. Inhibition of TACE activity or expression enhanced the cell surface TbRI levels and TGF-b-induced Smad3 and Akt activation. Accordingly, silencing TACE expression in cancer cells enhanced the TbRI presentation and TGF-b responsiveness, including the antiproliferative effect of TGF-b, and epithelial-to-mesenchymal transition. These results establish a mechanism for downregulating TGF-b signaling through TACE activation by the Erk MAP kinase pathway and a strategy for evasion of tumor suppression and modulation of epithelial-to-mesenchymal transition during cancer progression. The decreased growth inhibition by TGF-b, due to elevated TACE activity, complements the growth stimulation resulting from increased release of TGF-a family ligands.

Abstract:I am delighted to welcome you to this short, special edition on compassion-focused therapy (CFT). One of the core principles of CFT is that it is rooted in clinical science rather than any particular approach to therapy. If we use psychological science to better understand the way the brain evolved, functions, and can create mental health problems, we can begin to target and develop therapeutic interventions based on that science. Indeed, clinical psychology is at a point where it can begin to be less focused on allegiance to particular schools of therapy and root itself in basic science. We can distinguish the science of process from the science of intervention because they have not developed in tandem. There is considerable evidence now that a whole range of interventions that involve: developing an appropriate and supportive therapeutic relationship, creating opportunities for guided discovery and behavioural experiments, gradual exposure for avoided situations and emotions, practising imagery, learning behavioural and emotional regulation skills – to name a few – play a central role in the personal change and recovery journey. However, CFT suggests that how individuals engage in these processes can play a big role in their willingness to engage with them and how effective they are. Compassion-focused therapy highlights the fact that if we look at the evolution of our minds then one thing stands out above all else – the human brain evolved for social processing (Cozolino, 2007, 2008; Siegel, 2012). Over 100 millions years ago, the advent of the mammalian ‘caring for offspring’ and group living introduced a whole range of emotion regulation processes that work through relationships – one of which was obviously the attachment system and capacity for affiliation and caring (Carter, 1998; Porges, 2007). Much of our evolution of intelligence was driven by social challenges and our cognitive architecture is adapted for social processing (Dunbar, 2010). Social relationships matter immensely to our well-being in many ways. For example, the extent anddegree of affiliation thatweexperience from the daywe are born to the day thatwedie has a huge impact on the quality of our lives (Cozolino, 2007, 2008). Affiliative processes influence the architecture of our motivational and emotional systems and even influence genetic expression (Belsky & Pluess, 2009; Siegel, 2012). So, if we put the capacity for affiliation central to the regulation of mind this directs attention to how people can experience kindness and affiliation from others, show kindness, empathy and affiliation toothers and relate tooneself in thoseways. Abstract:The low level of transcytosis is a unique feature of cerebrovascular endothelial cells (ECs), ensuring restrictive blood-brain barrier (BBB) permeability. Major facilitator superfamily domain-containing 2a (MFSD2A) is a key regulator of the BBB function by suppressing caveolae-mediated transcytosis. However, the mechanisms regulating MFSD2A at the BBB have been barely explored. Here, we show that cerebrovascular EC-specific deletion of Pten (phosphatase and tensin homolog) results in a dramatic increase in vesicular transcytosis by the reduction of MFSD2A, leading to increased transcellular permeability of the BBB. Mechanistically, AKT signaling inhibits E3 ubiquitin ligase NEDD4-2-mediated MFSD2A degradation. Consistently, cerebrovascular Nedd4-2 overexpression decreases MFSD2A levels, increases transcytosis, and impairs BBB permeability, recapitulating the phenotypes of Pten-deficient mice. Furthermore, Akt deletion decreases phosphorylated NEDD4-2 levels, restores MFSD2A levels, and normalizes BBB permeability in Pten-mutant mice. Altogether, our work reveals the essential physiological function of the PTEN/AKT/NEDD4-2/MFSD2A axis in the regulation of BBB permeability.

Abstract:Adhesive forces at endothelial cell-cell borders maintain vascular integrity. cAMP enhances barrier properties and controls cellular processes through protein kinase A bound to A-kinase anchoring proteins (AKAPs). It also activates exchange protein directly activated by cAMP (Epac1), an exchange factor for Ras-related protein 1 (Rap1) GTPases that promotes cadherin- and integrin-mediated adhesion through effects on the actin cytoskeleton. We demonstrate that AKAP9 facilitates the microtubule polymerization rate in endothelial cells, interacts with Epac1, and is required for Epac1-stimulated microtubule growth. AKAP9 is not required for maintaining barrier properties under steady-state conditions. Rather, it is essential when the cell is challenged to make new adhesive contacts, as is the case when Epac activation enhances barrier function through a mechanism that, surprisingly, requires integrin adhesion at cell-cell contacts. In the present study, defects in Epac-induced responses in AKAP9-silenced cells were evident despite an intact Epac-induced increase in Rap activation, cortical actin, and vascular endothelial-cadherin adhesion. We describe a pathway that integrates Epac-mediated signals with AKAP9-dependent microtubule dynamics to coordinate integrins at lateral borders.

Abstract:Rats were trained to walk on a treadmill to avoid foot shock. The animals developed tolerance for ethanol if given subsequent practice while ethanol intoxicated. Rats given equivalent doses of ethanol after practice did not develop tolerance, nor did saline-treated controls. These results challenge the hypothesis that mere repeated doses of ethanol are sufficient to induce tolerance. It seems that tolerance does not develop unless the response used to measure tolerance is performed while the subject is intoxicated.
cAMP/EPAC Signaling Enables ETV2 to Induce Endothelial Cells with High Angiogenesis Potential

Abstract:Although the generation of ETV2-induced endothelial cells (iECs) from human fibroblasts serves as a novel therapeutic strategy in regenerative medicine, the process is inefficient, resulting in incomplete iEC angiogenesis. Therefore, we employed ChIP-sequencing and identified molecular mechanisms underlying ETV2-mediated endothelial transdifferentiation to efficiently produce iECs retaining appropriate functionality in long-term culture. We revealed that the majority of ETV2 targets in human fibroblasts are related to vasculature development and signaling transduction pathways including Rap1 signaling. From a screening of signaling pathway modulators, we confirmed that forskolin facilitated efficient and rapid iEC reprogramming via activation of cAMP/EPAC/RAP1 axis. The iECs obtained via cAMP signaling activation showed superior angiogenesis in vivo as well as in vitro. Moreover, these cells could form aligned endothelium along the vascular lumen ex vivo when seeded into decellularized liver scaffold. Overall, our study provided evidence that cAMP/EPAC/RAP1 axis is required for the efficient generation of iECs with angiogenesis potential.

Cellular signalling of adrenomedullin regulating endothelial barrier function and granulocyte extravasation

Abstract:Background: Disturbed endothelial barrier function is a hallmark of inflammation. Effusion of protein rich plasma fluid leading to edema formation and overshooting transmigration of leukocytes contribute to severe organ dysfunction under conditions of generalized inflammation, e.g. acute lung injury and sepsis. The peptide hormone Adrenomedullin (ADM) and its receptor (CRLR/RAMP2/3) constitute an important signaling system for the protection of endothelial barrier function. Although elevation of cAMP in endothelial cells was identified as signaling pathway of ADM responsible for barrier protection, it was speculated that other mechanisms are involved and precise intracellular signaling routes have not been explored, yet. Main findings and conclusions: ADM was confirmed to be a strong stabilizer of the endothelial barrier function in vitro, preventing and reversing hyperpermeability independent of the inflammatory stimulus (as determined by electrical impedance and FITC-dextran permeability measurements after stimulation with thrombin, TNFα, histamine, VEGF). Moreover, it was shown for the first time that ADM inhibited TNFα-induced granulocyte transmigration which was solely due to effects of ADM on endothelial cells. These in vitro-findings could be translated to animal models of vascular permeability and inflammation: ADM dose-dependently reduced vascular permeability in the skin of mice and rats (Miles assay; stimulus histamine). In a murine lipopolysaccharide (LPS)-induced lung injury model, ADM reduced lung edema and leukocyte extravasation. In endothelial cells, barrier protection could solely be reduced to cAMP signaling via the protein kinase A (PKA) and Epac-Rap1 pathways (dose response comparison with Forskolin). Dissection of the downstream cascade by means of cAMP analogs, specific for PKA and Epac-Rap1 signaling (benz-cAMP and “007”, respectively), demonstrated that both pathways are with respect to their efficacy equally involved in effects on permeability probably addressing independent effector mechanisms as suggested by enhanced efficacy of combination of both. Consistently, both pathway activators prevented vascular hyper-permeability in vivo. PKA activation and inhibition of MLC phosphorylation downstream of ADM diminish stimulus induced actin stressfiber formation and contraction of endothelial cells thus counteracting intercellular gap formation and hyperpermeability. Most notably, also effects independent of the contractile apparatus were demonstrated: ADM stabilized barrier function even in the absence of a functional actin cytosceletton after treatment with Cytochalasin D and knockdown of cortactin. ADM induced stabilization of VE-cadherin at the cell borders and increased the amount of detectable VE-cadherin/VE-PTP complex which is important for the regular function of VE-cadherin. In contrast, granulocyte transmigration was only reduced by PKA activation and ADM effects were lost after knockdown of cortactin. In line with this finding, a PKA inhibitor abolished the effect of ADM. As PKA activation reduces myosin light chain phosphorylation these data collectively link leukocyte extravasation to the contractile apparatus of the endothelial cell. As “007” was fully active with respect to TNFα induced hyperpermeability but had no effect with respect to granulocyte transmigration, endothelial hyperpermeability per se is not a prerequisite for granulocyte transmigration.

TPA-induced signal transduction: a link between PKC and EGFR signaling modulates the assembly of intercellular junctions in Caco-2 cells

Abstract:Recent studies suggest that signal transduction may have an important role in the development and regulation of the metastatic phenotype. Here, we investigated the role of the epidermal growth factor receptor (EGFR), and protein kinase C (PKC), in the process of reassembly of cadherin-dependent cell-cell adhesion of Caco-2 cells. We used chemical activation of PKC and EGFR with 12-O-tetradecanoylphorbol-13-acetate (TPA), a tumor-promoting agent, pretreatment with protein kinase inhibitors and subcellular fractionation to analyze the effect of the phorbol ester on the redistribution of junctional proteins. Transepithelial resistance (TER), electron microscopy and immunofluorescence analyses were also carried out. Activation with TPA resulted in disassembly of adherens junctions (AJs), but the tight junction (TJ) structure and function remained unaltered. TPA affected E-cadherin levels. In Caco-2 cells at day 2 of culture, when most E-cadherin is not associated with the cytoskeleton, a decrease in the level of this protein was observed as soon as 6 h after TPA addition. However, at day 5 of culture, the major effect observed after 6 h of treatment was a translocation of the protein from the Triton-insoluble to the -soluble fraction. On the other hand, TPA did not significantly affect the E-cadherin-associated proteins α and β-catenins. Potent specific EGFR inhibitors, such as PD153035 and Tyrphostin 25, as well as Calphostin C, an inhibitor of PKC, significantly blocked the effect of TPA on AJs. Furthermore, inhibition of the TPA effect by the PD98059 MAPK inhibitor suggests that activation of this kinase was the final event in the modulation of cadherin-dependent cell-cell adhesion. Pretreatment of cell monolayers with Calphostin C before EGF treatment, one of the ligands of EGFR, blocked the redistribution of E-cadherin caused by EGF. Based on these results, we conclude that both EGFR and PKC activation are involved in TPA-induced cell signaling for modulation of cadherin-dependent cell-cell adhesion and cell shape in Caco-2 cells.
