Abstract:The central nervous system (CNS) has a highly complex biophysical and biochemical environment. Despite decades of intensive research, it is still an enormous challenge to restore its functions and regenerate lost or damaged CNS tissues. Current treatment strategies remain sub-optimal because of (1) the hostile microenvironment created post CNS injury, and (2) insufficient understanding of the pathophysiology of acute and chronic CNS diseases. Two-dimensional (2D) in vitro models have provided tremendous insights into a wide range of cellular interactions. However, they fail to recapitulate the complex cellular, topographical, biochemical, and mechanical stimuli found within the natural three-dimensional (3D) CNS. Also, the growing ethical needs to use fewer animals for research further necessitates 3D in vitro models to mimic all or part of the CNS. In this review, we critically appraise the status quo and design considerations of 3D in vitro neural disease and injury models that resemble in vivo conditions. This review mainly focuses on the most recent advances in tissue engineering techniques such as microfluidics, organs-on-a-chip and stem cell technology. Furthermore, we review recent models aiming to elucidate the underlying pathophysiology of CNS diseases. If armed with deeper understanding, it will be possible to develop high-throughput drug screening platforms and new treatments for CNS diseases and injuries.

Abstract:In view of the very expensive modern healthcare system, sudden loss or failure of organs and tissues could pose a very difficult and costly medical problem to patients. Further, the limited supply of organs globally that a patient can afford for replacement in the event of an organ failure makes the problem even more challenging and complicated. These medical and healthcare challenges have triggered research and developments into tissue engineering to advance the field of regenerative medicine. Especially, the research focus has been on the design, development and optimization of a cell-scaffold-microenvironment to promote the regeneration of various types of tissue including skin, cartilage, bone, tendon and cardiac tissue, to name a few. Studies have been undertaken to produce functional three-dimensional (3D) tissue substitutes or constructs that are based on bio scaffolds from the ground up. To this end, bioprinting strategies have been considered for fabrication of complex 3D functional living tissues or artificial organs. Here, we describe some notable advances in laser bioprinting enabled tissue engineering, which is a rapidly emerging field in 3D bio fabrication technology for applications in regenerative medicine.

Abstract:The therapeutic replacement of diseased tubular tissue is hindered by the availability and suitability of current donor, autologous and synthetically derived protheses. Artificially created, tissue engineered, constructs have the potential to alleviate these concerns with reduced autoimmune response, high anatomical accuracy, long-term patency and growth potential. The advent of 3D bioprinting technology has further supplemented the technological toolbox, opening up new biofabrication research opportunities and expanding the therapeutic potential of the field. In this review, we highlight the challenges facing those seeking to create artificial tubular tissue with its associated complex macro- and microscopic architecture. Current biofabrication approaches, including 3D printing techniques, are reviewed and future directions suggested.

Abstract:Tissue engineered constructs have the potential to function as in vitro pre-clinical models of normal tissue function and disease pathogenesis for drug screening and toxicity assessment. Effective high throughput assays demand minimal systems with clearly defined performance parameters. These systems must accurately model the structure and function of the human organs and their physiological response to different stimuli. Musculoskeletal tissues present unique challenges in this respect, as they are load-bearing, matrix-rich tissues whose functionality is intimately connected to the extracellular matrix and its organization. Of particular clinical importance is the osteochondral junction, the target tissue affected in degenerative joint diseases, such as osteoarthritis (OA), which consists of hyaline articular cartilage in close interaction with subchondral bone. In this review, we present an overview of currently available in vitro three-dimensional systems for bone and cartilage tissue engineering that mimic native physiology, and the utility and limitations of these systems. Specifically, we address the need to combine bone, cartilage and other tissues to form an interactive microphysiological system (MPS) to fully capture the biological complexity and mechanical functions of the osteochondral junction of the articular joint. The potential applications of three-dimensional MPSs for musculoskeletal biology and medicine are highlighted.

Abstract:The past three decades have seen the emergence of an endeavor called tissue engineering and regenerative medicine in which scientists, engineers, and physicians apply tools from a variety of fields to construct biological substitutes that can mimic tissues for diagnostic and research purposes and can replace (or help regenerate) diseased and injured tissues. A significant portion of this effort has been translated to actual therapies, especially in the areas of skin replacement and, to a lesser extent, cartilage repair. A good amount of thoughtful work has also yielded prototypes of other tissue substitutes such as nerve conduits, blood vessels, liver, and even heart. Forward movement to clinical product, however, has been slow. Another offshoot of these efforts has been the incorporation of some new exciting technologies (e.g., microfabrication, 3D printing) that may enable future breakthroughs. In this review we highlight the modest beginnings of the field and then describe three application examples that are in various stages of development, ranging from relatively mature (skin) to ongoing proof-of-concept (cartilage) to early stage (liver). We then discuss some of the major issues that limit the development of complex tissues, some of which are fundamentals-based, whereas others stem from the needs of the end users.

Abstract:A novel targeted application of tissue engineering is the development of an in vitro 3D tissue model for drug screening and toxicology. This paper discusses the modeling, design, and freeform fabrication of 3D cell-embedded tissue constructs for creating a pharmacokinetic model. This is achieved using a combinatorial setup involving a CAD-driven automated syringe-based, layered direct cell writing process in conjunction with soft lithographic micro-patterning techniques. This enables the rational design of a microscale in vitro device housing a bioprinted 3D tissue construct (or micro-organ) that biomimics the cell’s physiological microenvironment for enhanced functionality. Abstract:The physiochemical stem cell microenvironment regulates the delicate balance between self-renewal and differentiation. The three-dimensional assembly of stem cells facilitates cellular interactions that promote morphogenesis, analogous to the multicellular, heterotypic tissue organization that accompanies embryogenesis. Therefore, expansion and differentiation of stem cells as multicellular aggregates provides a controlled platform for studying the biological and engineering principles underlying spatiotemporal morphogenesis and tissue patterning. Moreover, three-dimensional stem cell cultures are amenable to translational screening applications and therapies, which underscores the broad utility of scalable suspension cultures across laboratory and clinical scales. In this review, we discuss stem cell morphogenesis in the context of fundamental biophysical principles, including the three-dimensional modulation of adhesions, mechanics, and molecular transport and highlight the opportunities to employ stem cell spheroids for tissue modeling, bioprocessing, and regenerative therapies.

Abstract:3D bioprinting technology enable the creation of complex, functional tissue constructs for engineered tissue constructs and patient‐specific models. Methods such as extrusion, jetting, and light‐based bioprinting each offer unique benefits. Despite progress, challenges remain in producing clinically relevant, human‐scale tissues. Ongoing interdisciplinary research continues to refine bioprinting, promising personalized medical interventions and functional, transplantable tissues. Advancements in bioprinting technology are driving the creation of complex, functional tissue constructs for use in tissue engineering and regenerative medicine. Various methods, including extrusion, jetting, and light‐based bioprinting, have their unique advantages and drawbacks. Over the years, researchers and industry leaders have made significant progress in enhancing bioprinting techniques and materials, resulting in the production of increasingly sophisticated tissue constructs. Despite this progress, challenges still need to be addressed in achieving clinically relevant, human‐scale tissue constructs, presenting a hurdle to widespread clinical translation. However, with ongoing interdisciplinary research and collaboration, the field is rapidly evolving and holds promise for personalized medical interventions. Continued development and refinement of bioprinting technologies have the potential to address complex medical needs, enabling the development of functional, transplantable tissues and organs, as well as advanced in vitro tissue models.

Abstract:Tissue Engineering is a rapidly evolving field in terms of cell source and scaffold fabrication. As the template for three dimensional tissue growth, the scaffold should emulate the native extracellular matrix, which is nano-fibrous. Currently, there are three basic techniques capable of generating nano-fibrous scaffolding: electrospinning, molecular self-assembly, and thermally induced phase separation. These scaffolds can then be further modified by various three dimensional surface modification techniques if necessary to more precisely emulate the native extracellular matrix. However, even without further modification, nano-fibrous scaffolds have been shown to have advantageous effects on cellular behavior and tissue formation when compared to more traditional types of scaffolding. This review focuses on the current state of tissue engineering with nano-fibrous scaffolding with particular emphasis on bone tissue engineering.

Abstract:Abstract The three‐dimensional (3D) multicellular platforms prepared by cells or biomaterials have been widely applied in biomedical fields for the regeneration of complex tissues, the exploration of cell crosstalk, and the establishment of tissue physiological and pathological models. Compared with the traditional 2D culture methods, the 3D multicellular platforms are easier to adjust the components and structures of extracellular matrix (ECM) because of the synthesis of ECM by cells and the use of biomaterials. Moreover, the 3D multicellular platforms also can customize the cell distribution and precisely design micro and macro structures of the systems. Based on these typical advantages of 3D multicellular platforms and their increasingly important position in the biomedical field, this review summarizes the present 3D multicellular platforms. Herein, current 3D multicellular platforms are divided into two major types: scaffold‐free and scaffold‐based 3D multicellular platforms. The specific characteristics and applications of different types of 3D multicellular platforms are thoroughly introduced to help readers understand how different models affect and regulate cell behaviors and inspire researchers on how to select and design suitable 3D multicellular platforms according to different application scenarios.

Abstract:The development of artificial tissue/organs with the functional maturity of their native equivalents is one of the long-awaited panaceas for the medical and pharmaceutical industries. Advanced 3D cell-printing technology and various functional bioinks are promising technologies in the field of tissue engineering that have enabled the fabrication of complex 3D living tissue/organs. Various requirements for these tissues, including a complex and large-volume structure, tissue-specific microenvironments, and functional vasculatures, have been addressed to develop engineered tissue/organs with native relevance. Functional tissue/organ constructs have been developed that satisfy such criteria and may facilitate both in vivo replenishment of damaged tissue and the development of reliable in vitro testing platforms for drug development. This review describes key developments in technologies and materials for engineering 3D cell-printed constructs for therapeutic and drug testing applications.

Abstract:Heart disease is one of the major global health issues. Despite rapid advances in cardiac tissue engineering, limited successful strategies have been achieved to cure cardiovascular diseases. This situation is mainly due to poor understanding of the mechanism of diverse heart diseases and unavailability of effective in vitro heart tissue models for cardiovascular drug screening. With the development of microengineering technologies, three-dimensional (3D) cardiac microtissue (CMT) models, mimicking 3D architectural microenvironment of native heart tissues, have been developed. The engineered 3D CMT models hold greater potential to be used for assessing effective drugs candidates than traditional two-dimensional cardiomyocyte culture models. This review discusses the development of 3D CMT models and highlights their potential applications for high-throughput screening of cardiovascular drug candidates.

Abstract:Biocompatible tissue growth has excellent prospects for tissue engineering. These tissues are built over scaffolds, which can influence aspects such as cell adhesion, proliferation rate, morphology, and differentiation. However, the ideal 3D biological structure has not been developed yet. Here, we applied the electro-hydrodynamic jet (E-jet) 3D printing technology using poly-(lactic-co-glycolic acid, PLGA) solution to print varied culture platforms for engineered tissue structures. The effects of different parameters (electrical voltage, plotting speed, and needle sizes) on the outcome were investigated. We compared the biological compatibility of the 3D printed culture platforms with that of random fibers. Finally, we used the 3D-printed PLGA platforms to culture fibroblasts, the main cellular components of loose connective tissue. The results show that the E-jet printed platforms could guide and improve cell growth. These highly aligned fibers were able to support cellular alignment and proliferation. Cell angle was consistent with the direction of the fibers, and cells cultured on these fibers showed a much faster migration, potentially enhancing wound healing performance. Thus, the potential of this technology for 3D biological printing is large. This process can be used to grow biological scaffolds for the engineering of tissues. (c) 2017 Wiley Periodicals, Inc. Abstract:Over the past decade, advances in biomedical and tissue engineering technologies, such as cell culture techniques, biomaterials, and biofabrication, have driven increasingly widespread use of three-dimensional (3D) cell culture platforms and, subsequently, the use of organoids in a variety of research endeavors. Given the 3D nature of these organoid systems, and the frequent inclusion of extracellular matrix components, these constructs typically have more physiologically accurate cell–cell and cell–matrix interactions than traditional 2D cell cultures. As a result, 3D organoids can serve as better model systems than their 2D counterparts. Moreover, as organoids can be biofabricated from highly functional human cells, they have certain advantages over animal models, being human in nature and more easily manipulated in the laboratory. In this review, we describe such organoid technologies and their deployment in drug development and precision medicine efforts. Organoid technologies are rapidly being developed for these applications and now represent a wide variety of tissue types and diseases. Evidence is emerging that organoids are poised for widespread adoption, not only in academia but also in the pharmaceutical industry and in clinical diagnostic applications, positioning them as indispensable tools in medicine.

Expert Opinion on Biological Therapy

Abstract:Introduction: Organ failure and tissue loss are challenging health issues due to widespread injury, the lack of organs for transplantation and limitations of conventional artificial implants. The field of tissue engineering aims to provide alternative living substitutes that restore, maintain or improve tissue function. Areas covered: In this paper, a wide range of porous scaffolds are reviewed, with an emphasis on phase-separation techniques that generate advantageous nanofibrous 3D scaffolds for stem cell-based tissue engineering applications. In addition, methods for presentation and delivery of bioactive molecules to mimic the properties of stem cell niches are summarized. Recent progress in using these bioinstructive scaffolds to support stem cell differentiation and tissue regeneration is also presented. Expert opinion: Stem cells have great clinical potential because of their capability to differentiate into multiple cell types. Biomaterials have served as artificial extracellular environments to regulate stem cell behavior. Biomaterials with various physical, mechanical and chemical properties can be designed to control stem cell development for regeneration. Conclusion: The research at the interface of stem cell biology and biomaterials has made and will continue to make exciting advances in tissue engineering.
