Abstract:The growth of rice seedling,membrane lipid peroxidation and the specific activities of antioxidant enzymes(superoxide dismutase,catalase,ascorbate peroxidase and guaiacol peroxidase) in rice seedlings treated with a varying concentrations of SNP under 30 mg/L Cd2+ stress were evaluated.The results showed that pretreatment with SNP alleviated significantly the oxidative damage caused by Cd stress in the leaves and roots of rice seedlings,reduced the contents of H2O2 and malondialdehyde(MDA),and decreased the activities of superoxide dismutase(SOD),catalase(CAT),ascorbate peroxidase(APX),and guaiacol peroxidase(GPX) in the leaves and roots of rice seedlings under Cd stress.Nitric Oxide(NO) acted in plants with a concentration-dependent manner,lower concentrations of NO acting as an efficient scavenger in breaking the oxidative chain.Higher NO concentrations could provoke a major oxidative insult,since this molecule is a nitrogen reactive species.
LeSPL-CNR Negatively Regulates Cd Acquisition Through Repressing Nitrate Reductase-Mediated Nitric Oxide Production in Tomato.

Abstract:MAIN CONCLUSION:An SPL-type transcription factor, LeSPL-CNR, is negatively involved in NO production by modulating SlNR expression and nitrate reductase activity, which contributes to Cd tolerance. Cadmium (Cd) is a highly toxic pollutant. Identifying factors affecting Cd accumulation in plants is a prerequisite for minimizing dietary uptake of Cd from crops grown with contaminated soil. Here, we report the involvement of a SQUAMOSA PROMOTER-BINDING PROTEIN-LIKE (SPL) transcription factor LeSPL-CNR in Cd tolerance in tomato (Solanum lycopersicum). In comparison with the wild-type Ailsa Craig (AC) plants, the Colourless non-ripening (Cnr) epimutant displayed increased Cd accumulation and enhanced sensitivity to Cd, which was in well accordance with the repression of LeSPL-CNR expression. Cd stress-induced NO production was inhibited by nitrate reductase (NR) inhibitor, but not NO synthase-like enzyme inhibitor. Expression of LeSPL-CNR was negatively correlated with SlNR expression and the NR activity. We also demonstrated that LeSPL-CNR inhibited the SlNR promoter activity in vivo and bound to SlNR promoter sequence that does not contain a known SBP-binding motif. In addition, expression of an IRON-REGULATED TRANSPORTER1, SlIRT1, was more abundant in Cnr roots than AC roots under Cd stress. LeSPL-CNR may thus provide a molecular mechanism linking Cd stress response to regulation of NR-dependent NO production, which then contributes to Cd uptake via SlIRT1 expression in tomato.
Nitric oxide exacerbates Al-induced inhibition of root elongation in rice bean by affecting cell wall and plasma membrane properties.

Abstract:Aluminum (Al) toxicity is one of the most widespread problems for crop production on acid soils, and nitric oxide (NO) is a key signaling molecule involved in the mediation of various biotic and abiotic stresses in plants. Here we found that exogenous application of the NO donor sodium nitroprusside (SNP) exacerbated the inhibition of Al-induced root growth in rice bean [Vigna umbellata (Thunb.) Ohwi & Ohashi ‘Jiangnan’, Fabaceae]. This was accompanied by an increased accumulation of Al in the root apex. However, Al treatments had no effect on endogenous NO concentrations in root apices. These results indicate that a change in NO concentration is not the cause of Al-induced root growth inhibition and the adverse effect of SNP on Al-induced root growth inhibition should result from increased Al accumulation. Al could significantly induce citrate efflux but SNP had no effects on citrate efflux either in the absence or presence of Al. On the other hand, SNP pretreatment significantly increased Al-induced malondialdehyde accumulation and Evans Blue staining, indicating an intensification of the disruption of plasma membrane integrity. Furthermore, SNP pretreatment also caused greater induction of pectin methylesterase activity by Al, which could be the cause of the increased Al accumulation. Taken together, it is concluded that NO exacerbates Al-induced root growth inhibition by affecting cell wall and plasma membrane properties.
Exogenously applied sodium nitroprusside alleviates nickel toxicity in maize by regulating antioxidant activities and defense-related gene expression

Abstract:Nickel (Ni) stress adversely affects plant growth and biomass accumulation, posturing severe menace to crop production and food security. The current study aimed to determine the putative role of sodium nitroprusside (SNP) in mitigating Ni-induced phytotoxicity and identify the underlying defense mechanisms in maize, which are poorly understood. Our findings showed that SNP significantly augmented plant growth, biomass, and photosynthesis-related attributes (Fv/Fm, Fm, qP ETR, and fPSII) through diminishing Ni uptake and translocation in root and shoot tissues of maize under Ni stress conditions. In parallel, exogenous SNP substantially relieved maize seedlings from Ni-induced stress by enhancing enzymatic (SOD, CAT, and GPX) and non-enzymatic (phenol and flavonoids) antioxidant defenses and reducing oxidative stress indicators (MDA and H2O2). The results revealed that SNP treatment increased the content of organic osmolyte glycine betaine and the activity of GST, concomitantly with ATP and ionic exchange capacity (including Ca2+-ATPase and Mg2+-ATPase), advocating its sufficiency to promote plant growth and avert Ni-induced stress in maize plants. The only exception was the production of organic acids (citric, oxalic, malic, and formic acids), which was reduced as SNP treatment relieved maize seedlings from Ni-induced oxidative damage. The application of SNP also displayed higher expression of defense- and detoxifying-related genes than in control treatments. Together, our data highlighted the mechanism involved in the amelioration of Ni toxicity by SNP; thus, suggesting a potential role of SNP in mitigating the adverse effects of Ni-contaminated soils to boost growth and yield of crop plants, that is, maize.

Abstract:Current investigation demonstrated that chromium (Cr) toxicity affects adversely on the normal growth of soybean plants. However, the seed priming with nitric oxide (NO; 100 μM), and spermine (Spm; 0.01 Mm) can significantly alleviate the Cr toxicity in soybean plant. Herein, the hydroponic experiment was conducted to observe the individual as well as the interactive behavior of NO, and Spm on the various morpho-physiological and, biochemical parameters in soybean such as plant growth, plant height, seed germination indices, photosynthesis-related indices such as chlorophyll biosynthesis, PS system II, nutrient uptake of soybean seedlings against Cr (VI) toxicity. Our outcomes deliberated that the alone treatment of NO, and Spm cause a significant improvement in seed germination ratio, photosynthetic pigments, and biomass of plants by restricting Cr uptake; while NO + Spm treatment being more effective in the improvement of soybean growth relative to their individual treatment under Cr stress. Relative to alone treatment of NO, and Spm, the combined treatment significantly modulated the antioxidant activities, and lowered the ROS accumulation, and electrolyte leakage. In addition, seed priming with NO, and Spm mitigate the Cr-induced toxicity by reducing Cr uptake and stimulating the antioxidative defense mechanisms. Hence, these findings confirmed the positive defense interplay of the NO and Spm in the modulation of the Cr tolerance in soybean. However, the underlying defense mechanism of these synergetic effects needs to be further explored.

Abstract:Heavy metals contamination is a major environmental issue, and phytoremediation is a promising, environmentally friendly alternative to conventional cleanup techniques for heavy metal contaminated sites. This paper is intended to explore the physiological functions of nitric oxide (NO) in the signaling pathways as well as defense responses in Sedum Alfredii Hance, a new zinc (Zn) promising hyperaccumulating plant species for the phytoremediation exposured to Cd/Zn/Pb complex contaminations. The induced NO and free radicals (O2 ldr, H2O2, ldrOH and malondialdehyde) were quantitatively determined and the functions of NO, reactive oxygen species (ROS) and sacylic acid (SA) in the regulation of the redox signaling against oxidative stress were also discussed. The hypothetical mode of NO effecting plant defense responses was proposed. The results indicated that NO as the versatile and powerful effector of redox-regulated signaling, can acts synergistically with ROS to defend the plant against oxidative attack caused by Cd/Zn/Pb complex contaminations by activating elaborate defenses.
Effect of Nitric Oxide on Alleviating Cadmium Toxicity in Rice （oryza Sativa L.）

Abstract:Nitric oxide (NO) is a gaseous signaling molecule in plants that plays a key role in mediating a wide range of physiological processes and responses to biotic and abiotic stresses. The present study was conducted to investigate the effects of the exogenous application of sodium nitroprusside (SNP), an NO donor, on cadmium (Cd)-induced oxidative stress and Cd uptake in rice plants. Rice plants were exposed to Cd stress (0.2 mmol L-1 CdCl2) and different concentrations of SNP (0.05, 0.1, 0.2, and 0.4 mmol L-1). A SNP concentration of 0.1 mmol L-1 (SNP10) significantly reduced the Cd-induced decrease in shoot and root dry weights and leaf chlorophyll concentrations. The addition of NO also reduced the malondialdehyde (MDA), hydrogen peroxide (H2O2) and ascorbic acid (ASA) concentrations. However, the reduction in glutathione (GSH) concentration was inhibited by NO treatment. Moreover, NO prevented the Cd-induced increase in antioxidative enzyme activity. The amount of Cd accumulation in rice plants was also influenced by the addition of NO. The NO supplied by the SNP enhanced the Cd tolerance of the rice by increasing the Cd uptake by the roots and decreasing the Cd accumulation by the shoots. However, the application of potassium ferrocyanide (Cd+Fe) or sodium nitrate and nitrite (Cd+N) (without NO release), did not exhibit the effects of the SNP. Furthermore, the effects of the SNP were reversed by the addition of hemoglobin (an NO scavenger). Our results suggested that exogenous NO was involved in the resistance of rice to Cd-toxicity.

Abstract:As a donor of the gaseous signaling molecule nitric oxide (NO), sodium nitroprusside (SNP) has been shown to play a positive role in enhancing plant resistance to abiotic stress. However, its role in alleviating cadmium (Cd) toxicity in tobacco (Nicotiana tabacum L.) is not fully understood. This study found that Cd stress significantly inhibited tobacco growth. At the same time, 150 μM SNP was the most effective concentration in alleviating Cd toxicity in seedlings, restoring three stress tolerance indicators-MDA, H2O2, and proline-to control levels. Exogenous SNP mitigated Cd-induced oxidative stress by promoting the accumulation of non-enzymatic antioxidants (total phenolics and flavonoids) and activating key antioxidant enzymes (SOD, CAT, POD, APX, and GR) along with their gene expression. SNP also facilitated Cd accumulation in the root cell wall and prevented Cd translocation from roots to shoots. Additionally, SNP altered Cd's subcellular distribution, promoting its sequestration in vacuoles and cell walls, which may be related to the NO-mediated upregulation of the metallothionein gene NtMT2F and the phytochelatin gene NtPCS2. The addition of SNP significantly increased the proportion of Cd in less toxic chemical forms, with the residual Cd fraction in the Cd + SNP group reaching 7.30%, higher than the 4.86% in the Cd-only group. Furthermore, exogenous SNP counteracted Cd's inhibition of nitrate reductase (NR) activity, promoting endogenous NO production. This study systematically reveals the positive roles of exogenous SNP in mitigating Cd toxicity in tobacco, offering valuable insights for producing low-Cd tobacco.
Nitric oxide induced Cd tolerance and phytoremediation potential of B. juncea by the modulation of antioxidant defense system and ROS detoxification

Abstract:The present study designed to illustrate correlation between cadmium induced stress and plant growth, photosynthetic pigments, morphological and physiological attributes. To study these parameters 2 weeks old seedling of B. juncea were subjected to 50 µM Cd, 100 µM Cd and 100 µM SNP separately and in combination with SNP. After 96 h, the treated plant were harvested to analyze the cellular homeostasis and metal tolerance mechanism via examining growth, stress parameters, enzymatic and non enzymatic antioxidants and expression level of NR. Higher level of Cd (100 µM) significantly increased accumulation of reactive oxygen species and malonaldehyde content in comparison to 50 µM Cd. Exogenous supplementation of SNP (100 µM) to 50 µM Cd treated plant had an additive effect on plant growth by improving the level of proline, photosynthetic pigments and activities of enzymatic antioxidants which was confirmed by histochemical staining for NADPH-d and NO fluorescence from DAF-DA staining in roots of B. juncea. Applying SNP to 50 µM Cd exposed B. juncea roots enhanced NR activity by 1.36 folds and increased NO production by 1.12 folds than individual Cd treated roots. In addition, semi quantitative RT-PCR study revealed the induction of BjNR was more pronounced in 50 µM Cd treated roots in comparison to 100 µM Cd treated roots. The present finding revealed NO confers increased B. juncea tolerance to Cd stress by stimulation of antioxidants and reestablishment of cellular redox status. Different biochemical analysis showed that plant growth, photosynthetic pigment and antioxidants were positively correlated with NO and it's negatively correlated with oxidative stress biomarkers. Therefore, NO is gaseous signalling molecule with potential role in Cd detoxification mechanism in B. juncea.

Nitric oxide promotes adventitious root formation in cucumber under cadmium stress through improving antioxidant system, regulating glycolysis pathway and polyamine homeostasis

Abstract:Cadmium (Cd) as a potentially toxic heavy metal that not only pollutes the environment but also interferes with plant growth. Nitric oxide (NO) regulates plant growth and development as well as abiotic stress response. However, the mechanism underpinning NO-induced adventitious root development under Cd stress remains unclear. In this study, cucumber ( Cucumis sativus ‘Xinchun No. 4’) was used as the experimental material to investigate the effect of NO on the development of adventitious roots in cucumber under Cd stress. Our results revealed that, as compared to Cd stress, 10 μM SNP (a NO donor) could considerably increase the number and length of adventitious roots by 127.9% and 289.3%, respectively. Simultaneously, exogenous SNP significantly increased the level of endogenous NO in cucumber explants under Cd stress. Our results revealed that supplementation of Cd with SNP significantly increased endogenous NO content by 65.6% compared with Cd treatment at 48 h. Furthermore, our study indicated that SNP treatment could improve the antioxidant capacity of cucumber explants under Cd stress by up-regulating the gene expression level of antioxidant enzymes, as well as reducing the levels of malondialdehyde (MDA), hydrogen peroxide (H 2 O 2 ) and superoxide anion (O 2 .- ) to alleviate oxidative damage and membrane lipid peroxidation. Application of NO resulted in a decrease of the O 2 .- , MDA, and H 2 O 2 level by 39.6%, 31.4% and 60.8% as compared to Cd-alone treatment, respectively. Besides that, SNP treatment significantly increased the expression level of related genes involved in glycolysis processes and polyamine homeostasis. However, application of NO scavenger 2-(4-carboxy -2-phenyl)-4, 4, 5, 5-tetramethy limidazoline -1-oxyl -3-oxide (cPTIO) and the inhibitor tungstate significantly reversed the positive role of NO in promoting the adventitious root formation under Cd stress. These results suggest that exogenous NO can increase the level of endogenous NO, improve antioxidation ability, promote glycolysis pathway and polyamine homeostasis to enhance the occurrence of adventitious roots in cucumber under Cd stress. In summary, NO can effectively alleviate the damage of Cd stress and significantly promote the development of adventitious root of cucumber under Cd stress.

Abstract:Cadmium (Cd) causes oxidative stress, which leads to the oxidation of various biomolecules by the production of reactive oxygen species (ROS) to facilitate programmed cell death (PCD). The antioxidant defense system fails to detoxify ROS when it is produced in excess. Nitric oxide (NO), a gaseous free radical and a phytohormone, regulates various physiological processes of plants. Therefore, this work was undertaken to study the effects of the application of exogenous sodium nitroprusside (SNP, a NO donor) on growth parameters, oxidative stress, accumulation of secondary metabolites, and activities of antioxidant enzymes under Cd stress. Mild (50 µM) and severe (200 µM) Cd stress were applied to hydroponically grown pea (Pisum sativum L.) plants with or without 50 µM SNP. Severe Cd stress had a substantial impact on the plants. The effectiveness of NO in reducing Cd-induced negative effects on plant height, fresh weight, dry weight, protein content, nitrite content, nitrate reductase (NR) activity, catalase activity, and peroxidase activity were investigated. Seedling development, protein content, nitrite content, nitrate reductase (NR) activity, antioxidant defense systems disruption, overproduction of reactive oxygen species, and oxidative damage were observed. The antioxidant defense system (catalase and peroxidase activities) was activated by NO, which resulted in lower lipid peroxidation and lower hydrogen peroxide (H2O2) levels in Cd-exposed plants. SNP treatment boosted endogenous NO levels and NR activity in Cd-stressed plants while also enhanced proline levels to preserve osmotic equilibrium. The presence of total phenols and flavonoids increased after SNP treatment, indicating that SNP enhanced stress recovery and boosted plant development in Cd-stressed plants.

Roles of hydrogen sulfide and nitric oxide in the alleviation of cadmium-induced oxidative damage in alfalfa seedling roots

Abstract:Despite hydrogen sulfide (H2S) and nitric oxide (NO) are important endogenous signals or bioregulators involved in many vital aspects of plant growth and responses against abiotic stresses, little information was known about their interaction. In the present study, we evaluated the effects of H2S and NO on alfalfa (Medicago sativa L.) plants exposed to cadmium (Cd) stress. Pretreatment with an H2S donor sodium hydrosulfide (NaHS) and well-known NO donor sodium nitroprusside (SNP) decreased the Cd toxicity. This conclusion was supported by the decreases of lipid peroxidation as well as the amelioration of seedling growth inhibition and Cd accumulation, in comparison with the Cd-stressed alone plants. Total activities and corresponding transcripts of antioxidant enzymes, including superoxide dismutase, peroxidase and ascorbate peroxidase were modulated differentially, thus leading to the alleviation of oxidative damage. Effects of H2S above were reversed by 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide potassium salt (cPTIO), the specific scavenger of NO. By using laser confocal scanning microscope combined with Greiss reagent method, further results showed that NO production increased significantly after the NaHS pretreatment regardless of whether Cd was applied or not, all of which were obviously inhibited by cPTIO. These decreases of NO production were consistent with the exaggerated syndromes associated with Cd toxicity. Together, above results suggested that NO was involved in the NaHS-induced alleviation of Cd toxicity in alfalfa seedlings, and also indicated that there exists a cross-talk between H2S and NO responsible for the increased abiotic stress tolerance.

Modification of nitrate uptake pathway in plants affects the cadmium uptake by roots.

Abstract:NRT1.1 is a dual-affinity nitrate (NO3(-)) transporter involved in both high- and low-affinity NO3(-) uptake in Arabidopsis plants. In a recent study, we showed that, under cadmium (Cd) exposure, blocking the NRT1.1-mediated NO3(-) uptake reduces Cd entry into roots, thus lowing Cd levels in plants and improving plant growth. In addition, we also found that the Cd levels in edible parts of 11 Chinese cabbage (Brassica rapa L. ssp. pekinensis) cultivars correlated well with the NO3(-) uptake rates of their roots. These results suggested that the NO3(-) uptake of roots negatively regulate Cd uptake. Modification of NO3(-) uptake in crops by modulating NO3(-) uptake pathway might provide a biological engineering approach to reducing Cd accumulation in edible organs, thus improving food safety.

Abstract:Identification of mechanisms that decrease cadmium accumulation in plants is a prerequisite for minimizing dietary uptake of cadmium from contaminated crops. Here, we show that cadmium inhibits nitrate transporter 1.1 (NRT1.1)-mediated nitrate (NO3 (-)) uptake in Arabidopsis (Arabidopsis thaliana) and impairs NO3 (-) homeostasis in roots. In NO3 (-)-containing medium, loss of NRT1.1 function in nrt1.1 mutants leads to decreased levels of cadmium and several other metals in both roots and shoots and results in better biomass production in the presence of cadmium, whereas in NO3 (-)-free medium, no difference is seen between nrt1.1 mutants and wild-type plants. These results suggest that inhibition of NRT1.1 activity reduces cadmium uptake, thus enhancing cadmium tolerance in an NO3 (-) uptake-dependent manner. Furthermore, using a treatment rotation system allowing synchronous uptake of NO3 (-) and nutrient cations and asynchronous uptake of cadmium, the nrt1.1 mutants had similar cadmium levels to wild-type plants but lower levels of nutrient metals, whereas the opposite effect was seen using treatment rotation allowing synchronous uptake of NO3 (-) and cadmium and asynchronous uptake of nutrient cations. We conclude that, although inhibition of NRT1.1-mediated NO3 (-) uptake by cadmium might have negative effects on nitrogen nutrition in plants, it has a positive effect on cadmium detoxification by reducing cadmium entry into roots. NRT1.1 may regulate the uptake of cadmium and other cations by a common mechanism.

Abstract:Nitrogen (N) management is a promising agronomic strategy to minimize cadmium (Cd) contamination in crops. However, it is unclear how N affects Cd uptake by plants. Wild-type and iron uptake-inefficient tomato (Solanum lycopersicum) mutant (T3238fer) plants were grown in pH-buffered hydroponic culture to investigate the direct effect of N-form on Cd uptake. Wild-type plants fed NO3- accumulated more Cd than plants fed NH4+. Iron uptake and LeIRT1 expression in roots were also greater in plants fed NO3-. However, in mutant T3238fer which loses FER function, LeIRT1 expression in roots was almost completely terminated, and the difference between NO3- and NH4+ treatments vanished. As a result, the N-form had no effect on Cd uptake in this mutant. Furthermore, suppression of LeIRT1 expression by NO synthesis inhibition with either tungstate or L-NAME, also substantially inhibited Cd uptake in roots, and the difference between N-form treatments was diminished. Considering all of these findings, it was concluded that the up-regulation of the Fe uptake system was responsible for NO3- -facilitated Cd accumulation in plants.
Regulation of nitrate reductase by nitric oxide in Chinese cabbage pakchoi (Brassica chinensis L.).

Abstract:Nitrate reductase (NR), a committed enzyme in nitrate assimilation, involves generation of nitric oxide (NO) in plants. Here we show that the NR activity was significantly enhanced by the addition of NO donors sodium nitroprusside (SNP) and NONOate (diethylamine NONOate sodium) to the culturing solution, whereas it was decreased by NO scavenger 2-(4-carboxyphenyl)-4,4,5,5-tetramethyl-imidazoline-1-oxyl-3-oxide (cPTIO). Interestingly, both NO gas and SNP directly enhanced but cPTIO inhibited the NR activities of crude enzyme extracts and purified NR enzyme. The cPTIO terminated the interaction between NR-generated NO and the NR itself. Furthermore, the NR protein content was not affected by the SNP treatment. The investigation of the partial reactions catalysed by purified NR using various electron donors and acceptors indicated that the haem and molybdenum centres in NR were the two sites activated by NO. The results suggest that the activation of NR activity by NO is regulated at the post-translational level, probably via a direct interaction mechanism. Accordingly, the concentration of nitrate both in leaves and roots was decreased after 2 weeks of cultivation with SNP. The present study identifies a new mechanism of NR regulation and nitrate assimilation, which provides important new insights into the complex regulation of N-metabolism in plants.

Abstract:Cadmium (Cd) is a heavy metal that is highly toxic to plants and animals and can accumulate in the environment as a result of industrial activities and agricultural application of some types of phosphate fertilizer. This study aimed to assess the role of sodium nitroprusside (SNP), as a source of nitric oxide (NO) in alleviating Cd stress in maize plants. Maize plants were kept in soil saturated with 40%-strength nutrient solution in a greenhouse, and cadmium nitrate, Cd(NO 3 ) 2 , was applied at different concentrations, (0, 10, and 50 μM). Sodium nitroprusside, [Fe(CN) 5 NO]·2H 2 O, at concentrations of 0.05, 0.1, and 0.2 μM. Growth, leaf gas exchange, and leaf anatomy analyses were performed. The experimental design was completely randomized in a 3 × 3 factorial arrangement with five replicates. The highest concentrations of Cd and SNP reduced the total dry mass and leaf and stem dry mass but increased the allocation of biomass to the roots and stem, but the leaf allocation did not change. The application of Cd and SNP promoted an increase in gas exchange and leaf area, in addition to an increase in leaf tissue thickness and stomatal density. The presence of SNP at low concentrations reduces the toxicity of Cd, but at high concentrations, this compound can generate negative effects and even toxicity in maize plants.

Abstract:Nitric oxide (NO) has been demonstrated to stimulate the activity of nitrate reductase (NR) in plant roots supplied with a low level of nitrate, and to affect proteins differently, depending on the ratio of NO to the level of protein. Nitrate has been suggested to regulate the level of NO in plants. This present study examined interactive effects of NO and nitrate level on NR activity in roots of tomato (Solanum lycocarpum).NR activity, mRNA level of NR gene and concentration of NR protein in roots fed with 0 center dot 5 mm or 5 mm nitrate and treated with the NO donors, sodium nitroprusside (SNP) and diethylamine NONOate sodium (NONOate), and the NO scavenger, 2-(4-carboxyphenyl)-4,4,5,5-tetramethyl-imidazoline-1-oxyl-3-oxide (cPTIO), were measured in 25-d-old seedlings.Addition of SNP and NONOate enhanced but cPTIO decreased NR activity in the roots fed with 0 center dot 5 mm nitrate. The opposite was true for the roots fed with 5 mm nitrate. However, the mRNA level of the NR gene and the protein concentration of NR enzyme in the roots were not affected by SNP treatment, irrespective of nitrate pre-treatment. Nevertheless, a low rate of NO gas increased while cPTIO decreased the NR activities of the enzyme extracts from the roots at both nitrate levels. Increasing the rate of NO gas further increased NR activity in the enzyme extracts of the roots fed with 0 center dot 5 mm nitrate but decreased it when 5 mm nitrate was supplied. Interestingly, the stimulative effect of NO gas on NR activity could be reversed by NO removal through N-2 flushing in the enzyme extracts from the roots fed with 0 center dot 5 mm nitrate but not from those with 5 mm nitrate.The effects of NO on NR activity in tomato roots depend on levels of nitrate supply, and probably result from direct interactions between NO and NR protein.

Abstract:The heavy metal cadmium is a common environmental contaminant in soils and has adverse effects on crop growth and development. The signaling processes in plants that initiate cellular responses to environmental stress have been shown to be located in the plasma membrane (PM). A better understanding of the PM proteome in response to environmental stress might provide new insights for improving stress-tolerant crops. Nitric oxide (NO) is reported to be involved in the plant response to cadmium (Cd) stress. To further investigate how NO modulates protein changes in the plasma membrane during Cd stress, a quantitative proteomics approach based on isobaric tags for relative and absolute quantification (iTRAQ) was used to identify differentially regulated proteins from the rice plasma membrane after Cd or Cd and NO treatment. Sixty-six differentially expressed proteins were identified, of which, many function as transporters, ATPases, kinases, metabolic enzymes, phosphatases, and phospholipases. Among these, the abundance of phospholipase D (PLD) was altered substantially after the treatment of Cd or Cd and NO. Transient expression of the PLD fused with green fluorescent peptide (GFP) in rice protoplasts showed that the Cd and NO treatment promoted the accumulation of PLD in the plasma membrane. Addition of NO also enhanced Cd-induced PLD activity and the accumulation of phosphatidic acid (PA) produced through PLD activity. Meanwhile, NO elevated the activities of antioxidant enzymes and caused the accumulation of glutathione, both which function to reduce Cd-induced H2O2 accumulation. Taken together, we suggest that NO signaling is associated with the accumulation of antioxidant enzymes, glutathione and PA which increases cadmium tolerance in rice via the antioxidant defense system.

Abstract:Cadmium (Cd), as a non-essential and toxic heavy metal in plants, has deleterious effects on plant physiological and biochemical processes. Nitric oxide (NO) is one of the most important signaling molecules for plants to response diverse stresses. Here, we found that Cd-induced programmed cell death (PCD) was accompanied by NO bursts, which exacerbated cell death when NO was removed and vice versa. Proteomic analysis of S-nitrosylated proteins showed that the differential proteins in Cd-induced PCD and in NO-alleviated PCD mainly exist together in carbohydrate metabolism and amino acid metabolism, while some of the differential proteins exist alone in metabolism of cofactors and vitamins and lipid metabolism. Meanwhile, S-nitrosylation of proteins in porphyrin and chlorophyll metabolism and starch and sucrose metabolism could explain the leaf chlorosis induced by PCD. Moreover, protein transport protein SEC23, ubiquitinyl hydrolase 1 and pathogenesis-related protein 1 were identified to be S-nitrosylated in vivo, and their expressions were increased in Cd-induced PCD while decreased in NO treatment. Similar results were obtained in tomato seedlings with higher S-nitrosylation. Taken together, our results indicate that NO might be involved in the regulation of Cd-induced PCD through protein S-nitrosylation, especially proteins involved in PCD response.
