Abstract:Salmonella continues to be a major food and public health burden worldwide that can threaten human health via eating contaminated meats, particularly those originating from chicken. In this study, the antimicrobial resistance profiles, epidemiological characteristics of resistance genes, and pulsed field gel electrophoresis (PFGE-XbaI) typing of 120 non-Pullorum/Gallinarum Salmonella isolates recovered from chicken embryos in Henan province were determined. The antimicrobial resistant phenotypes and evaluation of the extended-spectrum beta-lactamases (ESBLs) producing strains of Salmonella were investigated by the Kirby-Bauer test and the double-disk synergy test. Additionally, 37 antimicrobial resistance genes encoding resistance to five different categories, including aminoglycosides, cephalosporins, sulphonamides, tetracyclines, and beta-lactams, were examined by conventional PCR. However, genotyping analysis was conducted by macro-restriction using enzyme XbaI followed by the separation of the restricted DNA fragments by PFGE. The results of this study showed that the studied Salmonella strains were highly resistant to ampicillin (66.67%) and sulfisoxazole (66.67%), while they were all susceptible to meropenem, imipenem, colistin, and chloramphenicol. Additionally, 67.5% (81/120) of the studied strains were multidrug resistant, and 21.67% (26/120) were phenotypically confirmed as ESBLs positive. The statistical analysis showed that resistance depends on the serovars, and ESBLs positive strains showed more multi-resistance than ESBLs negative strains (p < 0.05). The genotypic antimicrobial resistance showed the detection of 14 among the 37 tested genes, and the concordance between genotypic and phenotypic antimicrobial resistance ranged from 0% to 100% depending on the serovars. However, the PFGE-XbaI typing results showed that the examined Salmonella strains were divided into 22 individual subtypes and were grouped in nine clusters, with similarity values ranging from 64.7% to 100%. From this study, we can conclude that the antimicrobial resistance of Salmonella serovars isolated from chicken embryos in Henan province was alarming, with rigorous multidrug resistance, which requires the urgent mitigation of the use of antimicrobial drugs in chicken hatcheries. Additionally, our results showed evidence of the presence of different PFGE patterns among the studied Salmonella serovars, suggesting the presence of different sources of contamination.
Epidemiological Investigation and Antimicrobial Resistance Profiles of Salmonella Isolated from Breeder Chicken Hatcheries in Henan, China

Abstract:The emergence of antimicrobial-resistant (AR) Salmonella has a major concern worldwide. This study was designed to determine the AR profiles and serovars distribution of Salmonella enterica isolated from different breeds of breeder chickens in the province of Henan, China. For this, 2,139 dead embryo samples were collected from 28 breeder chicken hatcheries, representing two domestic and four foreign breeds. The samples were subjected to the isolation and identification of Salmonella by PCR. The confirmed strains were serotyped according to the Kauffmann-White scheme and their AR profiles against 20 antimicrobial agents were determined by Kirby-Bauer (K-B) disc diffusion method. The results of this study showed the prevalence of Salmonella in 504 strains (23.56%) with a high abundance in southern regions of Yellow River (28.66%, n = 495, N = 1,727) compared to the northern regions (2.18%, n = 9, N = 412) (p < 0.0001). The domestic breeds were more contaminated than imported breeds (p < 0.0001). However, the contamination rate of samples recovered from M-hatcheries was the highest (p < 0.0001). Serotyping method identified 12 serovars, with the dominance of S. Pullorum (75.79%), followed by S. Enteritidis (7.14%). The AR assay showed high resistant to ciprofloxacin (77.00%), sulfisoxazole (73.00%), and ampicillin (55.60%), as well as 98.81% (n = 498) of the isolated strains, were resistant to at least one antimicrobial and 69.64% (n = 351) were resistant to three or more antimicrobials. Among them, one strain of S. Thompson was resistant to 15 antimicrobial agents belonging to eight different classes. In conclusion, Salmonella strains isolated in this study were multidrug-resistant (MDR), presenting a serious problem for human and animal health. Therefore, it is necessary to monitor, control, and rationalize the use of antimicrobials agents in chicken farms in order to limit the increasing resistance against the recent antimicrobial agents.

Abstract:Salmonella enterica subspecies enterica serotype Enteritidis (S. Enteritidis) is one of the leading causes for human salmonellosis all over the world. We analyzed the surveillance data of 18 years on antimicrobial resistance profiling of S. Enteritidis collected and isolated by the National Antimicrobial Resistance Monitoring System (NARMS) from humans, chicken, and chicken breasts. Statistical tool based on the unique individual antibiotic minimum inhibitory concentration (MIC) profiling was used to compare antimicrobial resistance in the isolates. A machine-learning algorithm, Random Forest matrix, segregated a collection of 6819 S. Enteritidis into multiple populations. The MIC value of 13 common antibiotics to individual isolate when taken as the best classifier, resulted in two distinct groups represented herein as Population-I and Population-II. Population-I, which spread within a small tight cluster, comprised all the chicken and chicken breasts' isolates as well as about 13.4% of the human isolates, whereas the Population-II consisted of the human isolates only, with a larger spread over wider area away from the Population-I (p < 0.001). Few overlapping, yet diverse clusters between humans and chicken as well as higher level of resistance of chicken breast isolate toward third-generation cephalosporins and tetracyclines compared to those from human isolates, highlight differences in their population structure. These findings indicate a complex driver for enriching antibiotic-resistant Salmonella in the food-chain other than those of chicken origin. This warrants for other strategies in addition to the judicious/restricted use of antibiotics to mitigate the threat of antimicrobial resistance.

Abstract:Non-typhoidal Salmonella (NTS) is a common cause for self-limiting gastroenteritis, representing a public health concern globally. NTS is one of the leading causes of foodborne illnesses in China; however, the invasive infection caused by NTS is largely underappreciated. Here, we reported an NTS invasive infection caused by an infrequently reported serovar Telelkebir (13,23:d:e,n,z15) strain FJ001 in China, which carries antimicrobial-resistant genes [fosA7 and aac(6')-Iaa] and typhoidtoxin genes (cdtB, pltA, and pltB). By conducting the whole genomic sequencing, we also investigated the relatedness of this strain with an additional 120 global contextual Salmonella enterica serovar Telelkebir (S. Telelkebir) isolates, and assessed the antimicrobial-resistant determinants and key virulence factors using the available genomic dataset. Notably, all 121 (100%) of the S. Telelkebir strains possessed the typhoid toxin genes cdtB, pltA, and pltB, and 58.67% (71/121) of S. Telelkebir harbored antimicrobial-resistant gene fosaA7. The study by core genome multilocus sequence typing (cgMLST) and core single-nucleotide polymorphism (SNP)-based phylogenomic analysis demonstrated that the S. Telelkebir isolates from different sources and locations clustered together. This suggests that regular international travels might increase the likelihood of rapid and extensive transmissions of potentially pathogenic bacteria. For the first time, our study revealed the antimicrobial resistance, virulence patterns, and genetic diversity of the serovar S. Telelkebir isolate in humans and similar isolates over the world. The present study also suggests that genomic investigation can facilitate surveillance and could offer added knowledge of a previously unknown threat with the unique combination of virulent and antimicrobial-resistant determinants.

Abstract:This study aimed to investigate the presence of Salmonella spp. in chicken doner samples, both ready for consumption and sold in packages in a modified atmosphere, using classical culture technique and the MALDI-TOF method by determining the antibiotic resistance profile structure through the disc diffusion method by determining the serotypes of the isolates through mPCR. Between September and November 2019, 150 ready-to-eat chicken doner samples (75 of them ready to consume, the other 75 packed in a modified atmosphere) were analyzed. It was found that 23 of the 150 (15.3%) samples analyzed were contaminated with Salmonella spp. 73 isolates were obtained from 23 positive samples. Of these 73 isolates, 33 were found to be Salmonella spp. positive. In serotyping, all 33 isolates were determined to be S. infantis. In the antibiotic resistance profile, 30 of theisolates were resistant to chloramphenicol, 24 to gentamicin, 21 to ampicillin, 20 to trimethoprim-sulfamethoxazole and ciprofloxacin, 18 to tetracycline and amoxicillin-clavulanic acid, and 13 to cefoxitin. As a result, the isolation of Salmonella spp. from the samples and their resistance to different antibiotics were evaluated as potential sources of problems in the context of foodborne infectious diseases.
Phenotypic and genotypic characterization of Salmonella enterica recovered from poultry meat in Tunisia and identification of new genetic traits

Abstract:Thirty-seven Salmonella enterica isolates obtained from poultry meat in Tunisia were included in this study for characterization of antibiotic resistance mechanisms. High percentages of resistance were detected to ampicillin, sulfonamides, tetracycline, nalidixic acid, and streptomycin (32.4%-89.2%), and lower percentages to amoxicillin-clavulanic acid, kanamycin, amikacin, trimethoprim-sulfamethoxazol, and chloramphenicol (2.7%-18.9%). All strains showed susceptibility to ceftazidime, cefotaxime, gentamicin, and ciprofloxacin. Class 1 integrons were detected in 30% of Salmonella isolates, and four different gene cassette arrangements were detected, including genes implicated in resistance to aminoglycosides (aadA1 and aadA2) and trimethoprim (dfrA1). Four different Pc variants (PcW, PcH1, PcH1(TTN-10), PcW(TGN-10)) with inactive P2 have been found among these isolates. Integron-positive isolates were ascribed to eight different serotypes. A Salmonella Schwarzengrund isolate harbored a new class 1 integron containing the qacH-dfrA1b-aadA1b-catB2 gene cassette arrangement, with the very unusual PcH1(TTN-10) promoter, which has been registered in GenBank (accession no. HQ874651). Different plasmid replicon types were demonstrated among integron-positive isolates: IncI1 (8 isolates), IncN (8), IncP (2), IncFIB (2), and IncFII (2). Ten different pulsed-field gel electrophoresis profiles were detected among the 11 integron-positive isolates and 8 different sequence types were identified by multilocus sequence typing, one of them (registered as ST867) was new, detected in 3 Salmonella Zanzibar isolates. A high diversity of clones is observed among poultry Salmonella isolates and a high proportion of them show a multiresistant phenotype with very diverse mobile genetic structures that could be implicated in bacterial dissemination in different environments.
Prevalence and Antibiotic Resistance of Non-typhoidal Salmonella Isolated from Raw Chicken Carcasses of Commercial Broilers and Spent Hens in Tai’an, China

Abstract:The present study was aimed to determine the prevalence and characteristics of Salmonella isolated from meat samples of commercial broilers (CB) and spent hens (SH). Between March and June 2016, 200 retail raw chicken carcasses (100 from CB and 100 from SH) were obtained from local supermarkets in Tai'an city of China, and Salmonella isolates were then analyzed for antibiotic resistance, serotype, β-lactamase genes, and the presence of class 1 integron. Forty Salmonella strains were obtained in this study (CB: 21/100, 21%; SH: 19/100, 19%). Three serotypes were identified in 40 Salmonella, and S. Enteritidis (CB: 15/21, 71.4%; SH: 10/19, 52.6%) was the dominant serotype, followed by S. Typhimurium (CB: 4/21, 19%; SH: 6/19, 31.6%) and S. Derby (CB: 2/21, 9.5%; SH: 3/19, 15.8%). Among 21 Salmonella isolated from CB, high antibiotic resistance rates were found for ampicillin (20/21, 95.2%), nalidixic acid (18/21, 85.7%), cefotaxime (17/21, 81%), and tetracycline (13/21, 61.9%); class 1 integron was observed in seven isolates (7/21, 33.3%), and gene cassettes included an empty integron (0.15 kb, n = 1), aadA2 (1.2 kb, n = 3), drfA1-aadA1 (1.4 kb, n = 1), and drfA17-aadA5 (1.7 kb, n = 2); blaTEM-1 was the dominant β-lactamase gene (21/21, 100%), followed by blaCTX-M-55 (7/21, 33.3%). Among 19 Salmonella isolated from SH, high antibiotic resistance rates were found for nalidixic acid (19/19, 100%), tetracycline (19/19, 100%), ampicillin (18/19, 94.7%), and ciprofloxacin (13/19, 68.4%); class 1 integron was observed in two isolates (2/19, 10.5%), and gene cassettes included drfA17-aadA5 (1.7 kb, n = 1) and drfA1-aadA1 (1.4 kb, n = 1); blaTEM-1 was the dominant β-lactamase gene (19/19, 100%), followed by blaCTX-M-55 (2/19, 10.5%) and blaCMY-2 (1/19, 5.3%). Collectively, antibiotic-resistant Salmonella can be widely detected in retail raw chicken carcasses of CB and SH, and therefore can pose a serious risk to public health.

Abstract:Avian salmonellosis is concomitant with high financial crises in the poultry industry as well as food-borne illness in man. The present study is designed to investigate the emergence of Salmonella Enteritidis and Salmonella Typhimurium in diseased broilers, resistance profiles, and monitoring virulence and antibiotic resistance genes. Consequently, 450 samples (cloacal swabs, liver, and spleen) were collected from 150 diseased birds from different farms in Giza Governorate, Egypt. Subsequently, the bacteriological examination was done. Afterward, the obtained Salmonella isolates were tested for serogrouping, antibiogram, PCR monitoring of virulence ( inv A, stn, hil A, and pef A), and antimicrobial resistance genes ( bla TEM , bla CTX-M , bla NDM , erm A, sul 1, tet A, and aad A1). The total prevalence of Salmonella in the examined diseased broilers was 9.3%, and the highest prevalence was noticed in cloacal swabs. Among the recovered Salmonella isolates ( n = 35), 20 serovars were recognized as S . Enteritidis and 15 serovars were identified as S . Typhimurium. Almost 60% of the retrieved S. Enteritidis serovars were extensively drug-resistant (XDR) to seven antimicrobial classes and inherited sul 1, bla TEM , tet A , bla CTX-M , ere A, and aad A1 genes. Likewise, 25% of the recovered S. Enteritidis serovars were multidrug-resistant (MDR) to six classes and have sul 1, bla TEM , tet A , bla CTX-M , and ere A resistance genes. Also, 66.7% of the retrieved S. Typhimurium serovars were XDR to seven classes and have sul 1, bla TEM , tet A , bla CTX-M , ere A, and aad A1 genes. Succinctly, this report underlined the reemergence of XDR S . Typhimurium and S. Enteritidis in broiler chickens. Meropenem and norfloxacin exposed a hopeful antimicrobial activity toward the re-emerging XDR S . Typhimurium and S. Enteritidis in broilers. Moreover, the recurrence of these XDR Salmonella strains poses a potential public health threat.

Abstract:Abstract Background Salmonella enterica serovar Enteritidis ( S . Enteritidis) is one of the most common serovars, associated with human salmonellosis. The food-borne outbreak of this bacterium is mainly related to the consumption of contaminated poultry meat and poultry products, including eggs. Therefore, rapid and accurate detection, besides investigation of virulence characteristics and antimicrobial resistance profiles of S . Enteritidis in poultry and poultry egg samples is essential. A total of 3125 samples (2250 poultry and 875 poultry egg samples), sent to the administrative centers of veterinary microbiology laboratories in six provinces of Iran, were examined for Salmonella contamination, according to the ISO 6579 guideline. Next, duplex PCR was conducted on 250 presumptive Salmonella isolates to detect invA gene for identification of the genus Salmonella and sdf gene for identification of S . Enteritidis. Subsequently, the S. Enteritidis isolates were examined for detection of important virulence genes ( pagC , cdtB , msgA , spaN , tolC , lpfC , and spvC ) and determination of antibiotic resistance patterns against nalidixic acid, trimethoprim-sulfamethoxazole, cephalothin, ceftazidime, colistin sulfate, and kanamycin by the disk diffusion method. Results Overall, 8.7 and 2.3% of poultry samples and 6.3 and 1.3% of eggs were contaminated with Salmonella species and S. Enteritidis, respectively. The invA and msgA genes (100%) and cdtB gene (6.3%) had the highest and the lowest prevalence rates in S . Enteritidis isolates. The spvC gene, which is mainly located on the Salmonella virulence plasmid, was detected in 50.8% of S . Enteritidis isolates. The S . Enteritidis isolates showed the highest and the lowest resistance to nalidixic acid (87.3%) and ceftazidime (11.1%), respectively. Unfortunately, 27.0% of S . Enteritidis isolates were multidrug-resistant (MDR). Conclusion The rate of contamination with Salmonella in the poultry and egg samples, besides the presence of antimicrobial resistant and MDR Salmonella isolates harboring the virulence genes in these samples, could significantly affect food safety and subsequently, human health. Therefore, continuous monitoring of animal-source foods, enhancement of poultry farm control measures, and limiting the use of antibiotics for prophylactic purposes in food producing animals, are essential for reducing the zoonotic risk of this foodborne pathogen for consumers and also choosing effective antibiotics for the treatment of salmonellosis.

Abstract:Salmonellosis, a zoonotic disease, is one of the leading causes of foodborne illness worldwide. It is responsible for most infections caused by consumption of contaminated food. In recent years, a significant increase in the resistance of these bacteria to common antibiotics has been observed, posing a serious threat to global public health. The aim of this study was to investigate the prevalence of virulent antibiotic-resistant Salmonella spp. strains in Iranian poultry markets. A total of 440 chicken meat samples were randomly selected from meat supply and distribution facilities in Shahrekord and tested for bacteriological contamination. After culturing and isolating the strains, identification was performed using the classical bacteriological method and PCR. To determine antibiotic resistance, a disc diffusion test was performed according to the recommendations of the French Society of Microbiology. PCR was used to detect resistance and virulence genes. Only 9% of the samples were positive for Salmonella. These were Salmonella typhimurium isolates. All Salmonella typhimurium serotypes tested positive for the rfbJ, fljB, invA and fliC genes. Resistance to TET, cotrimoxazole, NA, NIT, piperacillin/tazobactam and other antibiotics was found in 26 (72.2%), 24 (66.7%), 22 (61.1%) and 21 (58.3%) isolates, respectively. The sul1, sul2 and sul3 genes were present in 20, 12 and 4 of 24 cotrimoxazole-resistant bacteria, respectively. Chloramphenicol resistance was found in six isolates, but more isolates tested positive for the floR and cat two genes. In contrast, 2 (33%) of the cat three genes, 3 (50%) of the cmlA genes and 2 (34%) of the cmlB genes were all positive. The results of this investigation showed that Salmonella typhimurium is the most common serotype of the bacterium. This means that most of the antibiotics commonly used in the livestock and poultry industries are ineffective against most Salmonella isolates, which is important for public health.

Abstract:Salmonella still has a serious foodborne outbreak with public health risk. Chicken meat, chicken meat products, and raw milk are important reservoir for Salmonella. In the current study, a total of 120 samples of chicken meat and chicken meat products (breast, thigh, giblets, frozen thigh, nuggets, burger, shish and luncheon, 15 of each) and 50 raw cow milk samples were tested for prevalence, serotyping, virulence genes, and antibiotic resistance profiles of Salmonella spp.. The prevalence of Salmonella spp. was 11.67% in chicken meat samples with the following incidence on each sample group; 13.33, 20, 26.67, 0, 6.67, 6.67, 13.33 and 6.67%, respectively. Raw milk samples overall occurrence of Salmonella spp. was 6%. Serological identification of the isolated Salmonella revealed presence of five different serotypes including S. kentukay, S. entritides, S. typhimurium, S. lindenberg and S. bassa. All isolated Salmonella spp. har-bored stn gene; while S. kentukay, S. entritides and S. typhimurium harbored mgtC gene but invA was found in S. entritides, S. typhimurium, S. lindenberg and S. bassa. Also, sopB was detected in S. kentukay, S. entritides, S. typhimurium and S. lindenberg. The isolated Salmonella spp. was resistant to sulphamethoxazol trimethoprim, chloramphenicol, and penicillin (100%). Meanwhile, the sensitivity was 70.6% % to ampicillin, enrofloxacin and amoxycillin clavulanic. The results confirm the importance of application of strict hygienic measures in food industry and proper use of antibiotics for meat and milk producing animals.

Abstract:Background Antibiotic resistance is a significant public health issue worldwide. Antibiotic-resistant zoonotic bacteria such as Escherichia coli (E. coli), Campylobacter, Salmonella, Listeria, Coxiella, and Mycobacterium can be particularly isolated from biofertilizers. Epidemiological studies have shown that cases of foodborne infections and intoxications are significantly related to animal-derived foods. The presence of these species in aquatic environments indicates areas or organisms contaminated with animal or human feces. Especially, the presence of E. coli in aquatic environments has become a serious problem worldwide. Pathogenic strains of E. coli cause waterborne and foodborne diseases. Materials and methods This study included a total of 290 samples collected from five different dairy farms between April and September 2023 which comprised 20 samples of cow manure, 20 samples of milk, three samples of dairy workers' hand washing water, five samples of soil, five samples of water, and five samples of vegetables. The samples taken from the farms were homogenized with 0.1% peptone water at a ratio of 1/10. They were then cultured on xylose lysine deoxycholate (XLD), eosin methylene blue agar (EMB), and blood agar media, and gram-negative colonies were identified using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) and the VITEK2 automated system (BioMerieux Inc., Durham, NC). Amplification of the isolated DNA extracts was performed with A.B.T.™ 2X HS-PCR MasterMix (A.B.T Laboratory Industry, Arnavutköy, Turkey) in the SimpliAmp™ thermal cycler (Thermo Fischer Scientific Inc., Waltham, MA) and visualized by agarose gel electrophoresis. Results Among the 52 E. coli strains isolated in our study, the highest antibiotic sensitivity rate was observed in meropenem, while the lowest sensitivity rates were determined in cefazolin and cefuroxime. While two of the Salmonella spp. (n = 2) isolates were found to be resistant to tetracycline, and one was found to be resistant to penicillin and ampicillin. No resistance to trimethoprim/sulfamethoxazole was detected in either isolate. Extended-spectrum beta-lactamases (ESBLs) were detected in only four (7.7%) E. coli strains. While tetA, tetB, and TEM genes were seen in almost all E. coli strains, they were not found in Salmonella spp. Conclusion In conclusion, our study revealed the presence of antimicrobial resistance genes in E. coli and Salmonella spp. isolates collected from various farms and environmental samples, which render the antimicrobials used for disease treatment ineffective. Consequently, research should be undertaken to prevent the development of new resistance genes in our country, as creating new medications and treatment strategies for these diseases is costly and time-intensive.
High prevalence and genomic features of multidrug-resistant Salmonella enterica isolated from chilled broiler chicken on retail sale in the United Arab Emirates

Abstract:Non-typhoidal Salmonella represents a significant global concern for food safety and One Health. Despite the United Arab Emirates (UAE) being a leading consumer of chicken meat globally, there is a lack of comprehensive understanding regarding the prevalence and genomic characteristics of Salmonella within the country. This study aims to address this gap by conducting a thorough analysis of Salmonella prevalence, antimicrobial resistance, and genomic profiles of isolates obtained from whole broiler carcasses retailed under chilled conditions in the UAE. Our findings reveal that Salmonella was detected in 41.2 % (130/315) of the sampled chilled broiler carcasses, with notable variability observed among samples sourced from six different companies. Phenotypic antimicrobial resistance (AMR) testing, among 105 isolates, highlighted high resistance rates to tetracycline (97.1 %), nalidixic acid (93.3 %), ampicillin (92.4 %), azithromycin (75.2 %), ciprofloxacin (63.8 %), and ceftriaxone (54.3 %). Furthermore, a concerning 99 % (104/105) of the isolates exhibited multidrug resistance. Whole-genome sequencing (WGS) of 60 isolates identified five serovars, with S. infantis/Sequence Type (ST) 32 (55 %) and S. Minnesota/ST-458 (28.3 %) being the most prevalent. WGS analysis unveiled 34 genes associated with antimicrobial resistance, including mcr-1.1 (only in two isolates), conferring resistance to colistin. The two major serovars, Infantis and Minnesota, exhibited significant variation (P-values <0.001) in the distribution of major AMR genes (aadA1, blaCMY-2, blaSHV-12, qnrB19, qnrS1, sul1, and sul2). Notably, the gene qacEdelta, conferring resistance to quaternary ammonium compounds commonly found in disinfectants, was universally present in all S. Infantis isolates (n = 33), compared to only one S. Minnesota isolate. Additionally, all S. Infantis isolates harbored the IncFIB (pN55391) plasmid replicon type. Major serovars exhibited distinct distributions of antimicrobial resistance genes, underscoring the importance of serovar-specific surveillance. These findings emphasize the critical need for continuous surveillance and intervention measures to address Salmonella contamination risks in poultry products, providing valuable insights for public health and regulatory strategies not only in the UAE but also globally.
Detection of Salmonella Pathogenicity Islands and Antimicrobial-Resistant Genes in Salmonella enterica Serovars Enteritidis and Typhimurium Isolated from Broiler Chickens

Abstract:Rapid growth in commercial poultry production is one of the major sources of Salmonella infections that leads to human salmonellosis. The two main Salmonella enterica serovars associated with human salmonellosis are enteritidis and typhimurium. The aim of this study was to determine the prevalence of S. enterica serovars Enteritidis and S. Typhimurium as well as their Salmonella pathogenicity islands (SPI) and antibiotic resistance profiles in broiler chicken feces from slaughterhouses. A total of 480 fecal samples from broiler chickens that were grouped into 96 pooled samples were identified to have Salmonella spp. using the invA gene, whilst the Spy and sdfI genes were used to screen for the presence of S. Enteritidis and S. Typhimurium serovars, respectively, by polymerase chain reaction (PCR) assays. The isolates were also screened for the presence of Salmonella pathogenicity islands (SPIs) using PCR. The disc diffusion assay was performed to determine the antibiotic resistance profiles of the isolates. A total of 36 isolates were confirmed as Salmonella spp. through amplification of the invA gene. Out of 36 confirmed Salmonella spp. a total of 22 isolates were classified as S. Enteritidis (n = 8) and were S. Typhimurium (n = 14) serovars. All (n = 22) S. Enteritidis and S. Typhimurium isolates possessed the hilA (SPI-1), ssrB (SPI-2) and pagC (SPI-11) pathogenicity islands genes. Amongst these serovars, 50% of the isolates (n = 11/22) were resistant to tetracycline and nalidixic acid. Only 22% of the isolates, S. Typhimurium (13.6%) and S. Enteritidis (9.1%) demonstrated resistance against three or more antibiotic classes. The most detected antibiotic resistance genes were tet(K), mcr-1, sulI and strA with 13 (59.1%), 9 (40.9%), 9 (40.9%) and 7 (31.8%), respectively. The findings of this study revealed that S. Typhimurium is the most prevalent serotype detected in chicken feces. To reduce the risk to human health posed by salmonellosis, a stringent public health and food safety policy is required.
Molecular characterization and antibiotic resistance of Salmonella serovars isolated from broiler chicken farms

Abstract:The current study aimed to survey the prevalence, antimicrobial resistance, and virulence-associated genes of Salmonella enterica recovered from broiler chickens farms located in North coast, Egypt. A total of 200 broiler chickens’ cloacal swabs and farm environmental samples were randomly collected from four small scale broiler chicken farms at 5 weeks of production cycle. It was recorded that out of 200 samples, Salmonella species were identified in 26 (13%) samples. Salmonella strains were observed in 16% (16/100) of cloacal swabs, 10% (10/100) of environmental samples. S. Enteritidis (6%) was the most common identified serotypes followed by S. Kentucky (4%), S. Shangani (2.5%) and S. Virchow (0.5%). The antimicrobial susceptibility testing revealed absolute resistance to SXT (88.5%) followed by AMC and AMP (65.4% and 61.5%, respectively). On the other hand, lower rates of resistance were observed for DO (50%), CTX (42.3%), CEC (30.8%) and CIP (19.2%). Interestingly, only one isolate was resistant to IMP (3.8%). PCR targeting 5 virulence genes (invA, ompA, ompF, stn and pefA) were performed to detect the virulence of identified Salmonella serovars. Generally, all Salmonella isolates showed at least two virulence-associated genes. Of note, invA gene (genus specific gene) was detected in 100% (26/26) of the isolates. On the contrary, pefA gene was completely absent in all isolates. Finally, it was concluded that S. Enteritidis was the most common identified serotypes in the examined sources. Circulation of such strains in broiler farms required introducing special biosecurity measures for control of Salmonella. Such measures might limit the adverse effects of antibiotics and ensure the safety of the environment and animal-derived food.

Abstract:The plasmid patterns of 90 isolates of Neisseria gonorrhoeae (including 39 penicillinase-producing strains) originating from various countries in Africa were determined. Serogrouping by coagglutination and auxotyping were used to characterise the isolates. The 4.4-megadalton plasmid was present in seven isolates out of 39 penicillinase-producing strains, two of which occurred with a conjugative 24.5-megadalton plasmid. The African strains were predominantly serogroup WI and wild type. Arginine-dependent isolates were as common as proline-dependent types.
Occurrence and Antimicrobial Susceptibility Profile of Salmonella spp. in Raw and Ready-To-Eat Foods and Campylobacter spp. in Retail Raw Chicken Meat in Transylvania, Romania

Abstract:The survey was undertaken to investigate the presence and antimicrobial susceptibility profile of Salmonella spp. in raw and ready-to-eat (RTE) foods, and Campylobacter spp. in the retail raw chicken meat collected in two counties of Transylvania, Romania. A total of 13.1% (51/388) of the examined food samples were found to be Salmonella positive, with a distribution of 14.7% (48/326) in the raw food (i.e., pork, chicken carcass, and shell egg) and 4.8% (3/62) in the RTE samples (i.e., sausages, but not ham and salami), respectively. These differences were statistically significant (p = 0.034). The isolates were serotyped as Salmonella Infantis (n = 19), Salmonella Typhimurium (n = 11) Salmonella Rissen (n = 8), Salmonella Derby (n = 3), Salmonella Enteritidis (n = 3), Salmonella Bredeney (n = 2), Salmonella Brandenburg (n = 1), Salmonella Gloucester (n = 1), Salmonella Goldcoast (n = 1), Salmonella Kottbus (n = 1), and Salmonella Ruzizi (n = 1). Campylobacter strains were present in 29.4% (10/34) of the investigated chicken samples, and the identified species were Campylobacter coli (70%) and C. jejuni (30%). From the 14 tested antimicrobials, the Salmonella isolates were resistant against azithromycin (88.2%), tetracycline (54.9%), sulfamethoxazole (54.9%), ciprofloxacin (45.1%), nalidixic acid (43.1%), ampicillin (35.3%), chloramphenicol (33.3%), tigecycline (25.5%), cefotaxime (13.7%), colistin (13.7%), trimethoprim (7.8%), and gentamicin (2%), resulting in the expression of 21 multidrug-resistant (MDR) profiles. Of 10 Campylobacter isolates, 80% were resistant to ciprofloxacin and nalidixic acid, 40% to tetracycline, and 10% to streptomycin and erythromycin, respectively. Our findings indicate that Romanian isolates of Salmonella spp. and Campylobacter spp., contaminating animal-origin foods, can exhibit MDR patterns, representing a public health risk.

Abstract:Paratyphoid salmonellosis is a serious disease threatens the poultry industry worldwide, besides its public health hazard. The aims of this study were characterization of paratyphoid Salmonella spp. in chicken flocks of some Egyptian governorates, demonstration of the antimicrobial susceptibility of the isolated Salmonella spp., and detection of some virulence genes and antibiotic resistance genes using recent molecular techniques. A total of 238 organ samples were collected from 52 broiler, layer, and breeder chicken flocks, representing 9 Egyptian governorates. Conventional characterization of Salmonella isolates revealed a total isolation rate of 56.3% (134/238). Moreover, the isolation rates of Salmonella spp. were (49/79; 62%), (47/81; 58%), (10/18; 55.5%), (9/20; 45%), (2/6; 33.3%), (2/3; 66.7%), and (15/82; 53.6%) from liver, yolk sac, heart, spleen, caecum, ovary, and dead-in-shell embryos, respectively. A total of 32/238 (13.44%) isolates of Salmonella were found. Serological identification revealed presence of S. enteritidis (21.9%), S. kentucky (15.6%), S. typhimurium (12.5%), S. molade (12.5%), S. takoradi (9.4%), S. wingrove (6.3%), S. infantis (6.3%), S. tsevie (6.3), S. shangani (3.1%), S. bargny (3.1%), and S. papuana (3.1%). All Salmonella strains (32/32; 100%) were resistant to streptomycin, while almost all of them (31/32; 96.9%) were susceptible to meropenem. The amplification of 16S rRNA gene of Salmonella isolates using uniplex polymerase chain reaction (PCR) generated a specific Salmonella product of approximate 550 base pair. The multiplex PCR revealed presence of invA (100%), stn (65.6%), and sopB (40.6 %) virulence-associated genes as well as aadA1 (100%), blaTEM (59.4%), aadB (18.75%), and sul1 (28.1%) antibiotic resistance genes. In conclusion, virulent paratyphoid Salmonella spp. are circulating in the Egyptian flocks, causing economic loses. Additionally, they became resistant to the most commonly used field antibiotics. Therefore, regular molecular surveillance studies on the circulating Salmonella spp. and their resistance to the used antibiotics are of significant importance.

Abstract:Salmonellosis is a significant public health concern around the world. The issue of antibiotic resistance, particularly in non-typhoidal Salmonella (NTS), is a global threat to human and animal health. Thirty-five Salmonella enterica subsp. enterica serotype Typhimurium (S. Typhimurium) isolated from various chicken samples were tested for antimicrobial-resistant profiling using the disc diffusion method. The strains were further examined for the presence of the resistance genes by using polymerase chain reaction (PCR) according to published protocols. Phenotype resistance profiling revealed that 22.9% (8 out of 35) isolates showed resistance to five antibiotics used in this study. All the penta-resistant isolates carried the blaTEM, floR, strA, and tetA gene which encoded for ampicillin, chloramphenicol, streptomycin, and tetracycline resistance. Only one isolate (1 out of 35) was found to contain the sulfonamides resistance, sulA gene. Nine isolates were found susceptible to all antibiotics tested and do not harbor any of the resistant associated genes. These findings provide evidence that the presence of resistance genes contributes to the phenotypic resistance profile, and thus could give rise to the emergence of the drug-resistant strain of Salmonella.

Abstract:Sir, Placental alkaline phosphatase (PLAP), an isoenzyme of alkaline phosphatase, is expressed by the syncytiotrophoblast after the 12th week of pregnancy and released into the maternal circulation. In 1968, Fishman et al. (1) first described its tumor-associated behavior (heat stable PLAP; Regan isoenzyme) in the serum and tumor tissue of a patient with a squamous cell carcinoma of the lung. Subsequently, two other tumoral alkaline phosphatases have been described (Nagao and Kashara isoenzymes) (2) (3). PLAP has a very high genetic polymorphism in humans and in the last years several monoclonal antibodies against this enzyme have been obtained. One of them is C2, which reacts preferentially with a molecular form that is frequently elevated in ovarian carcinomas (4), and more seldom in testicular carcinomas and is not influenced by smoking. This monoclonal antibody is specific for PLAP and does not cross-react with placental-like, intestinal or tissue-unspecific alkaline phosphatase. In this study we have evaluated the behavior ofPLAP-C2 serum levels in patients with non-neoplastic diseases and with non-germinal cancers. The control group included 99 HBsAg-blood donors, 59 males and 40 females, aged between 19 and 63 years (average 41); the study group comprised 305 patients affected by non-neoplastic diseases and 329 patients with non-germinal cancers. PLAP serum levels were measured by an enzyme-immunoassay (Prolifigen PLAP ELISA, Broma, Sweden) with the monoclonal antibody C2, whose lowest limit of detection was established to be 1 nkat/l. Each sample was measured in duplicate and the inter and intraassay coefficients of variation were below 8%. In the control group, PLAP-C2 serum levels oscillated between 1 and 6 nkat/l (1.85 ± 1.16; males: 1.66 ± 0.83; females: 2.05 ± 1.3) and a PLAP value of 5.5 nkat/l, which represents the 97.5 percentile, was taken as upper normal level. Table 1 shows the PLAP serum level distribution in patients with non-neoplastic diseases. As can be seen, concentrations higher than 5.5 nkat/I were observed in 7 cases (2.3%): 4 cirrhosis (9.1, 10.2, 17 and 20),2 choledocolithiasis (5.8 and 9) and 1 cholecystitis (7 nkat/l). Six patients (2%) had PLAP values higher than 6 nkat/l and only 3 cirrhotic patients had levels above 10 nkat/ l . Table II shows the distribution of PLAP serum levels in the neoplastic group.
