Abstract:Objective: In the current study, a hypoxia and/or serum-free model was constructed in vitro to identify the optimal protocol for hUC-MSCs expansion, while still maintaining growth capacities. Methods: Parallel assays were conducted using four cell groups. Proliferative activities, cell cycle analysis, flow cytometric analysis, apoptosis analysis, and the differentiation potential of these four cell populations were evaluated. Results: The overall viability of hUC-MSCs samples was >95%. There were no significant changes in any detected markers in the four groups. Under hypoxia or cultured in serum free culture media, hUC-MSCs retained the potential to differentiate in vitro toward osteogenic and adipogenic lineages. Under hypoxia and serum free conditions, hUC-MSCs showed higher proliferation, without more apoptosis. Conclusion: Hypoxia and serum free culture conditions did not influence the major properties of hUC-MSCs. Under hypoxia and serum free conditions, hUC-MSCs show higher proliferation. However, apoptosis rates did not increase. Therefore, the availability of optimized in vitro assays, involving hypoxia conditions and serum free media for hUC-MSCs manipulation, might have a substantial scientific and clinical potential.
EX VIVO ISOLATION AND EXPANSION OF RABBIT MESENCHYMAL STEM CELLS IN DEFINED SERUM-FREE MEDIA

Abstract:Introduction: Mesenchymal stem cells (MSCs) derived from rabbit bone marrow are routinely investigated in cardiovascular and orthopedic models for regenerative medicine. However, classical media used for generating MSCs are typically augmented with ill-defined supplements, such as fetal bovine serum, which increases safety concerns as well as inconsistency in MSC generation. We tested three commercial serum-free media for the generation and expansion of rabbit MSCs (rbMSCs). Materials and methods: RbMSCs were separated by adherent tissue culture and cultured in serum-free or serum-containing media. Cultured cells were characterized by phase contrast microscopy, cell-proliferation, flow cytometry, and qPCR analyses. Results: MesenCult (R) XF medium supported the generation and expansion of rbMSCs, whereas StemPro (R) hMSC and BD Mosaic hMSC media did not propagate freshly isolated rbMSCs. At passage 5, total cell yields for serum-containing and MesenCult (R) XF media were 4.0 +/- 0.16 x 108 and 1.9 +/- 0.28 x 109, and cumulative population doublings were 11.5 +/- 0.12 and 13.08 +/- 0.19, respectively. Colony formation efficiencies in MesenCult (R) XF were 21 to 26%, whereas those in serum-containing medium were 14 to 17%. Flow cytometry showed that rbMSCs in MesenCult (R) XF were positive for CD29, CD44 and CD73, and negative for CD34 and CD45. After corresponding differentiation, rbMSCs from MesenCult (R) XF could differentiate into adipogenic, osteogenic and chondrogenic lineages. qPCR analysis demonstrated that cells cultured in serum-free media had higher differentiation potentials than those cultured in serum-containing media. Discussion: rbMSCs were efficiently generated and expanded in MesenCult (R) XF, while maintaining the necessary characteristics attributed to MSCs for potential therapeutic use.

Abstract:Bone marrow mesenchymal stem cells (BMMSCs) are a kind of adult stem cells with multiple potential functions in cell proliferation and differentiation and will be wildly used for cell therapy and tissue engineer- ing in the future. Many factors are reviewed in this paper which include donors’ age, isolation methods, cell culture density, surface materials and their effects on cell proliferation. It is emphasized that the development of different substitution, such as growth factors and human autologous serum, for fetal bovine serum (FBS) to avoid the risk from some viruses. The BMMSCs introduced by the plasmid with telomerase gene have shown stronger prolifera- tion ability and multiple differentiation potential. The development of perfusion culture systems will make the prepa- ration of BMMSCs more suitable for clinical application.
Culture and expansion of mesenchymal stem cells by serum free medium

Abstract:Objective To investigate the effect of a new serum-free medium SYL-SF on the culture and expansion of umbilical cord derived mesenchymal stem cells (UC-MSCs) . Methods UC-MSCs were obtained from aborted human umbilical cord by mechanical isolation method, and were maintained in primary culture medium SYL-NBCS. From passage 1 (P 1 ), UC-MSCs were maintained and expanded in two different types of serum-free medium SYL-SF and MSCM-SF. Apoptosis was analyzed with Alexa Fluor 488 annexin V apotosis kit and cellular proliferation was analyzed with CCK-8 method. Cellular morphology, karyotype, cell surface marker and differentiation capacity were also detected. Results Primary UC-MSCs (P 0 ) are obtained 8 to 15 days after the mechanical isolation and cultured in primary medium SYL-NBCS. When the cells are changed into serum-free medium, the apoptosis rate of the cells cultured in SYL-SF is significantly lower than those in MSCM-SF. In SYL-SF medium, UC-MSCs expend faster and maintain MSCs-specific surface markers as well as the multipotent differentiation potential towards fat, bone and cartilage. They also maintain normal karyotype after 8 passages of culture in SYL-SF. Conclusion SYL-SF as a novel serum-free medium can be used to support the expansion and maintenance of UC-MSCs in vitro.
Human mesenchymal stem cells possess different biological characteristics but do not change their therapeutic potential when cultured in serum free medium

Abstract:IntroductionMesenchymal stem cells (MSCs) are widely investigated in clinical researches to treat various diseases. Classic culture medium for MSCs, even for clinical use, contains fetal bovine serum. The serum-containing medium (SCM) seems a major obstacle for MSCs-related therapies due to the risk of contamination of infectious pathogens. Some studies showed that MSCs could be expanded in serum free medium (SFM); however, whether SFM would change the biological characteristics and safety issues of MSCs has not been well answered.MethodsHuman umbilical cord mesenchymal stem cells (hUC-MSCs) were cultured in a chemical defined serum free medium. Growth, multipotency, surface antigen expression, telomerase, immunosuppressive ability, gene expression profile and genomic stability of hUC-MSCs cultured in SFM and SCM were analyzed and compared side by side.ResultshUC-MSCs propagated more slowly and senesce ultimately in SFM. SFM-expanded hUC-MSCs were different from SCM-expanded hUC-MSCs in growth rate, telomerase, gene expression profile. However, SFM-expanded hUC-MSCs maintained multipotency and the profile of surface antigen which were used to define human MSCs. Both SFM- and SCM-expanded hUC-MSCs gained copy number variation (CNV) in long-term in vitro culture.ConclusionhUC-MCSs could be expanded in SFM safely to obtain enough cells for clinical application, meeting the basic criteria for human mesenchymal stem cells. hUC-MSCs cultured in SFM were distinct from hUC-MSCs cultured in SCM, yet they remained therapeutic potentials for future regenerative medicine.

Abstract:Mesenchymal stem cells (MSCs) derived from bone marrow (BM) represents a useful source of adult stem cells for cell therapy and tissue engineering. MSCs are present at a low frequency in the BM; therefore expansion is necessary before performing clinical studies. Fetal bovine serum (FBS) as a nutritional supplement for in vitro culture of MSCs is a suitable additive for human cell culture, but not regarding subsequent use of these cells for clinical treatment of human patients due to the risk of viral and prion transmission as well as xenogeneic immune responses after transplantation. Recently, autologous serum (AS) has been as a supplement to replace FBS in culture medium. We compared the effect of FBS versus AS on the histone modification pattern of MSCs through in vitro osteogenesis and adipogenesis. Differentiation of stem cells under various serum conditions to a committed state involves global changes in epigenetic patterns that are critically determined by chromatin modifications. Chromatin immunoprecipitation (ChIP) coupled with real-time PCR showed significant changes in the acetylation and methylation patterns in lysine 9 (Lys9) of histone H3 on the regulatory regions of stemness (Nanog, Sox2, Rex1), osteogenic (Runx2, Oc, Sp7) and adipogenic (Ppar-γ, Lpl, adiponectin) marker genes in undifferentiated MSCs, FBS and AS. All epigenetic changes occurred in a serum dependent manner which resulted in higher expression level of stemness genes in undifferentiated MSCs compared to differentiated MSCs and increased expression levels of osteogenic genes in AS compared to FBS. Adipogenic genes showed greater expression in FBS compared to AS. These findings have demonstrated the epigenetic influence of serum culture conditions on differentiation potential of MSCs, which suggest that AS is possibly more efficient serum for osteogenic differentiation of MSCs in cell therapy purposes.

Abstract:OBJECTIVE:To explore the biological characteristics of mesenchymal stem cells (MSC) derived from umbilical cord blood (UCB) and their supporting capacities in ex vivo expansion of hematopoietic stem/progenitor cells (HSPCs).METHODS:Low-density mononuclear cells (MNCs) from UCB were cultured in IMDM containing 20% FBS to form confluent adherent cells through 15 passages. Some cytokines in the conditioned medium were determined with ELISA. UCB-derived adherent cells were displayed with antibodies and analyzed with flow cytometry. The supporting capacity of UCB-derived adherent cells for ex vivo expansion of CD34(+) cells was assayed by co-culture in a two step culture. UCB-derived adherent cells were induced for chondrogenic differentiation with chondrogenic medium, and the induced cells were analyzed for the type II pro-collagen gene expression with RT-PCR.RESULTS:The mean number of adherent fibroblast like colonies derived from UCB was (3.5 +/- 0.7)/10(6) MNCs. UCB-derived MSCs could survive for at least 15 passages of expansion. In their undifferentiated status, UCB-derived MSCs were CD13(+), CD29(+), CD90(+), CD105(+), CD166(+), SH2(+), SH3(+), SH4(+), CD45(-), CD34(-), and CD14(-). Stem cell factor (SCF), interleukin 6 (IL-6) and tumor necrosis factor alpha (TNF-alpha) could be detected in the supernatant of the cultures. The MSCs cultured in chondrogenic media could differentiate into chondrogenic cells and express type II pro-collagen mRNA. UCB-derived MSCs could support the proliferation and differentiation of UCB CD34(+) cells in vitro.CONCLUSION:UCB-derived MSCs are similar to those derived from adult bone marrow and can support the proliferation of hematopoietic stem/progenitor cells.
Optimization of Culture Condition of Human Bone Marrow Stromal Cells in Terms of Purification, Proliferation, and Pluripotency.

Abstract:Human bone marrow stromal cells (hBMSCs) possess multilineage differentiation potential and play an important role in modern tissue engineering. However, the development of culture media to maintain hBMSCs in an undifferentiated, self-renewing state during their robust proliferation remains a challenge. We developed and tested modified growth medium [medium 1: epidermal growth factor (EGF), platelet-derived growth factor (PDGF), low glucose, 2% fetal calf serum (FCS)] on hBMSCs by comparing primary cell isolation, multipassage expansion, culture morphology, proliferation, and cellular phenotype, and performing an expression analysis of intrinsic-regulated genes to other two media. Cell morphology, proliferation, and phenotype varied among the media, while cells cultured in medium 1 displayed small, spindle-shaped morphology with the highest rate of growth capacities and the expected phenotype. RT-PCR analysis showed that medium 1 displayed the lowest expression levels of osteogenic genes, chondrogenic genes (osteonectin, runt-related transcription factor 2, cartilage oligo matrix protein, and SOX9), and adipogenic genes (lipoprotein lipase). The expression of another adipogenic gene, peroxisome proliferator-activator receptor-γ2, was higher in medium 1 but did not reach significance. In addition, hBMSCs expanded in medium 1 showed the highest expression ratio of self-renewing-related genes Krüppel-like factor 2 (KLF2) and KLF5. In conclusion, medium 1 allows for better expansion and pluripotency maintenance of hBMSCs and serves as a preferred alternative to traditional serum-containing media for research applications and future clinical use.

Abstract:This study is aimed at investigating the potentials of ex vivo expansion and pluri-differentiation of cryopreservation of adult human bone marrow mesenchymal stem cells (hMSCs) into chondrocytes, adipocytes and neurocytes. Cryopreserved hMSCs were resuscitated and cultured for 15 passages, and then induced into chondrocytes, adipocytes and neurocytes with corresponding induction medium. The induced cells were observed for morphological properties and detected for expressions of type II collagen, triglyceride or neuron-specific enolase and nestin. The result showed that the resuscitated cells could differentiate into chondrocytes after exposure to transforming growth factor β 1 (TGF-β 1 ), insulin-like growth factor I (IGF-I) and vitamin C (V C ), and uniformly changed morphologically from a spindle-like fibroblastic appearance to a polygonal shape in three weeks. The induced cells were heterochromatic to safranin O and expressed cartilage matrix-procollagenal (II) mRNA. The resuscitated cells cultured in induction medium consisting of dexamethasone, 3-isobutyl-1-methylxanthine, indomethacin and IGF-I showed adipogenesis, and lipid vacuoles accumulation was detectable after 21 d. The resuscitated hMSCs were also induced into neurocytes and expressed nestin and neuron specific endolase (NSE) that were special surface markers associated with neural cells at different stage. This study suggested that the resuscitated hMSCs should be still a population of pluripotential cells and that it could be used for establishing an abundant hMSC reservoir for further experiment and treatment of various clinical diseases.
Production and preservation of serum-free human umbilical cord-derived mesenchymal stem cell sheets

Abstract:Abstract Background Human umbilical cord-derived mesenchymal stem cells (UC-MSCs) have attracted much attention because of their fast self-renewal, multipotent nature, painless collection, and compliance for standard amplification. Several types of culture media and systems have been used for large-scale expansion of UC-MSCs and preparation of UC-MSC sheets; however, most rely heavily on fetal bovine serum (FBS) and exogenous growth factors for optimal cell proliferation. Residual bovine serum albumin (BSA), a macromolecular protein in FBS, and exogenous growth factors in final products pose the potential threat of allergic reactions or tumorigenic risks in humans. Methods In this study, the production process of UC-MSC sheets was standardized, including donor screening, cell bank establishment and quality control, and cell sheet production and quality control. Serum-free culture medium and pharmaceutical grade coating matrix were used to form UC-MSC sheets to reduce the risk of BSA and exogenous growth factors in clinical application. Additionally, the preservation solution for and method of the UC-MSC sheet were developed for preservation and transportation before cell sheet transplantation. Then, quality standards of the UC-MSC sheet involving product characteristics, safety and functionality were formulated according to national and international regulations. Finally, the UC-MSC banks and the UC-MSC sheet products were identified according to the established quality standards. Results This technology enables UC-MSC sheets to be produced safely and stably while preserving the cell viability of UC-MSC sheets above 70% in 24 hours. The quality standards for the UC-MSC sheets were established in this study. Both the fresh and 24-hour preserved UC-MSC sheets met the established quality standards. Conclusion The preparation and preservation of clinical grade serum-free UC-MSC sheets provides the possibility for the clinical application of cell therapy.
Ex Vivo Expansion and Pluripotential Differentiation of Cryopreserved Bone Marrow Mesenchymal Stem Cells

Abstract:Cryopreserved human bone marrow mesenchymal stem cells(hMSCs) were thawed and passaged for more than 15 generations.Resuscitated hMSCs at passage 15 were induced into chondrocytes,adipocytes and neural cells under corresponding conditioned induction medium respectively.The resuscitated cells differentiated into chondrocytes after exposure to transforming growth factor β_1,insulin-like growth factor I and vitamin C.The induced cells were heterochromatic to toluidine blue and expressed cartilage matrix-procollagenal(II) mRNA.hMSCs induced with adipogenical induction medium consisting of dexamethasone,3-isobutyl-1-methylxanthine,indomethacin and insulin-like growth factor I accumulated lipid vacuoles as detected by oil red O.Nestin and neuron-specific enolase(NSE),special surface markers associated with neural cells were expressed in cells induced with a combination of β-mercaptoethanol,dimethyl sulfoxide,butylated hydroxyanisole,hydrocortisone, insulin-like growth factor I(IGF-I) and all-trans retinoic acid.The refractile cell bodies extended long processes that terminated at typical growth cones and filopodia.It is inferred that the resuscitated hMSCs compose a population of pluripotent cells having differentiation potential,and that cryopreserved hMSCs can be used for tissue engineering and treatment of various diseases.
Comparative Analysis of Biological Characteristics of Adult Mesenchymal Stem Cells with Different Tissue Origins

Abstract:Objective: To invest the differences among mesenchymal stem cells (MSCs) derived from different tissues and their impacts on clinical applications. Methods: In this study, MSCs were isolated from adipose tissue (AD), umbilical cord tissue (UC), and menstrual blood (Men) and compared their biological characteristics in terms of proliferation capacity, passage capacity, colony formation, and surface markers were compared. Results: The stem cells (SCs) obtained from different sources were all characterized as MSCs, but demonstrated some differences. Umbilical cord-derived MSCs (UCMSCs) were able to overcome density inhibition. The proliferation rate decreased in the order UCMSCs> MenSCs> ADSCs, while the colony-forming ability decreased in the order MenSCs> ADSCs> UCMSCs. Based on gene-expression data for MSCs from different sources within the same donor, 768 MenSC genes were found that were specifically upregulated or downregulated compared with bone marrow-derived MSCs and UCMSCs, most of which were involved in cell function-related pathways. In addition, MenSCs appeared to be superior in terms of immune inflammation, stress response, and neural differentiation potentials, but weaker in terms of osteogenic and chondrogenic differentiation capacities, compared with UCMSCs and bone marrow-derived MSCs. Conclusions: MenSCs have higher extraction efficiency, colony-forming ability, and long time passage capacity. Although the proliferation capacity is inferior to UCMSCs.
In vitro preparation of mesenchymal stem cells for clinical studies

Abstract:Objective To establish a safe, effective and extendable strategy for in vitro preparation of mesenchymal stem cells (MSCs) for clinical studies. Methods UC-MSCs were mechanically isolated from umbilical cord of healthy newborns. After stably proliferating in the medium supplement with new born calf serum named SYL-NBCS through passage1(P 1 ) to P 3 , cells were maintained in serum free medium named SYL-SF from P 4 on with the cells cultured in the SYL-NBCS as control. Cell morphology of UC-MSCs cultured in different medium was observed by inverted microscope and the proliferation was analyzed with CCK-8 test. BSA residues were detected in both cell lysates and supernatant.Results UC-MSCs (P 1 -P 3 ) reached 90% confluence after 3.5 ~ 4 days of culture in SYL-NBCS CCK8 analysis showed that UC-MSCs proliferation rate was similar when they were cultured in either SYL-SF or SYL-NBCS. Furthuremore, BSA residues testing demondtrated that the residuals of BSA in both cell lysate and culture supernatant cultured in SYL-SF were below 10 ng/10 6 cells, which met the safety value as Requirements of Biological Products. Conclusion The sequential culture of MSCs in serum-containing and serum-free culture medium provides a procedural amplification system for sufficient and quality controllable MSCs for either clinical studies or commercial applications.

Abstract:Bone marrow-derived mesenchymal stromal cells (BM-MSCs) heralded a new beginning for regenerative medicine and generated tremendous interest as the most promising source for therapeutic application. Most cell therapies require stringent regulatory compliance and prefer the use of serum-free media (SFM) or xeno-free media (XFM) for the MSC production process, starting from the isolation onwards. Here, we report on serum-free isolation and expansion of MSCs and compare them with cells grown in conventional fetal bovine serum (FBS)-containing media as a control. The isolation, proliferation and morphology analysis demonstrated significant differences between MSCs cultured in various SFM/XFM in addition to their difference with FBS controls. BD Mosaic™ Mesenchymal Stem Cell Serum-Free media (BD-SFM) and Mesencult-XF (MSX) supported the isolation, sequential passaging, tri-lineage differentiation potential and acceptable surface marker expression profile of BM-MSCs. Further, MSCs cultured in SFM showed higher immune suppression and hypo-immunogenicity properties, making them an ideal candidate for allogeneic cell therapy. Although cells cultured in control media have a significantly higher proliferation rate, BM-MSCs cultured in BD-SFM or MSX media are the preferred choice to meet regulatory requirements as they do not contain bovine serum. While BM-MSCs cultured in BD-SFM and MSX media adhered to all MSC characteristics, in the case of few parameters, the performance of cells cultured in BD-SFM was superior to that of MSX media. Pre-clinical safety and efficiency studies are required before qualifying SFM or XFM media-derived MSCs for therapeutic applications.

Abstract:Background: Human amniotic mesenchymal stem cells (hAMSCs) are splendid cell sources for clinical application in the administration of numerous refractory and relapse diseases. Despite the preferable prospect of serum-free (SF) condition for cell product standardization and pathogenic contamination remission, yet the systematic and detailed impact upon hAMSCs at both cellular and transcriptomic levels is largely obscure. Methods: For the purpose, we preconditioned hAMSCs under serum-containing (SC) and SF medium for 48 h and compared the biological signatures and biofunctions from the view of cell morphology, immunophenotypes, multi-lineage differentiation in vitro, cell vitality, cytokine expression, and immunosuppressive effect upon the subpopulations of T lymphocytes, together with the PI3K-AKT-mTOR signaling reactivation upon cell vitality. Meanwhile, we took advantage of RNA-SEQ and bioinformatic analyses to verify the gene expression profiling and genetic variation spectrum in the indicated hAMSCs. Results: Compared with those maintained in SC medium, hAMSCs pretreated in SF conditions manifested conservation in cell morphology, immunophenotypes, adipogenic differentiation, and immunosuppressive effect upon the proliferation and activation of most of the T cell subpopulations, but with evaluated cytokine expression (e.g., TGF-β1, IDO1, NOS2) and declined osteogenic differentiation and cell proliferation as well as proapoptotic and apoptotic cells. The declined proliferation in the SF group was efficiently rescued by PI3K-AKT-mTOR signaling reactivation. Notably, hAMSCs cultured in SF and SC conditions revealed similarities in gene expression profiling and variations in genetic mutation at the transcriptome level. Instead, based on the differentially expressed genes and variable shear event analyses, we found those genes were mainly involved in DNA synthesis-, protein metabolism-, and cell vitality-associated biological processes and signaling pathways (e.g., P53, KRAS, PI3K-Akt-mTOR). Conclusions: Collectively, our data revealed the multifaceted cellular and molecular properties of hAMSCs under SC and SF conditions, which suggested the feasibility of serum-free culture for the preferable preparation of standardized cell products for hAMSC drug development and clinical application.

Abstract:In the use of bovine fetal serum (FBS) there is concern about the possibility of disease transmission from animal to human. Therefore, it seems necessary to create culture conditions free of animal serum, especially in cell therapy. The aim of this study was to evaluate the feasibility of replacing human umbilical cord serum (hUCS) with FBS for in vitro expansion of umbilical cord mesenchymal stromal/stem cells (UC-MSCs). Here, UC-MSCs were cultured for five days in media supplemented either by hUCS or commercial FBS (Gibco and HyClone) to compare their viability, proliferation, morphology, Immunophenotype and differentiation potential. Our data shows that use of 5% and/or 10% hUCS, resulted in a tenfold increase in the number of MSCs; While in the presence of commercial FBS, this figure reached a maximum of five times. Notably, the rate of cell proliferation in the group containing 2% hUCS was the same as the groups containing 10% commercial FBS. Furthermore, there was no significant difference between groups in terms of viability, surface markers, and multilineage differentiation potential. These results demonstrated that hUCS can efficiently replace FBS for the routine culture of MSCs and can be used ideally in manufacturing process of UC-MSCs in cell therapy industry.

Mechanisms of Increased Vascular Superoxide Production in an Experimental Model of Idiopathic Dilated Cardiomyopathy

Abstract:Objective—In the present study, we sought to identify mechanisms underlying increased oxidative stress in vascular tissue in an experimental animal model of chronic congestive heart failure (CHF). Methods and Results—Superoxide and nitric oxide (NO) was measured in vessels from cardiomyopathic hamsters (CHF hamsters) and golden Syrian hamsters. We also determined expression of endothelial nitric oxide synthase (NOSIII), the soluble guanylyl cyclase, the cGMP-dependent kinase, and the NADPH oxidase. To analyze the contribution of the renin-angiotensin system to oxidative stress, CHF hamsters were treated with the angiotensin-converting enzyme inhibitor captopril for 200 days (120 mg · kg−1 · d−1). CHF led to increased superoxide production by NOSIII and the NADPH oxidase. Decreased NO production in CHF was associated with a decrease in the expression of NOSIII and an inhibition of NO downstream signaling in the aorta. NOSIII expression was increased within the left ventricle. Captopril treatment normalized NOSIII expression in vessels and the myocardium, reduced superoxide levels, and prevented NOSIII uncoupling. Accordingly, endothelial function, NO production, and downstream signaling were improved in CHF vessels. Conclusions—Oxidative stress in CHF is mediated by NADPH oxidase and an uncoupled NOSIII secondary to an activation of the renin-angiotensin system leading to impaired NO downstream signaling.
Regulation of Multilineage Gene Expression and Apoptosis During in Vitro Expansion of Human Bone Marrow Stromal Cells with Different Cell Culture Media

Abstract:The aim of the present study was to analyze the effect of 3 different expansion media on the expression of marker genes of mesenchymal differentiation (bone, cartilage, fat) as well as apoptosis and senescence during repeated passaging in human bone marrow stromal cells (hBMSCs) in order to identify potential expansion strategies for the use of these cells into tissue-engineered growth of bone. Medium 1 (EGF, PDGF, low Glc, 2% FCS) was associated with the highest proliferation rate compared to medium 2 (β-mercaptoethanol, high Glc DMEM, 15% FCS) and medium 3 (low Glc DMEM, 10% FCS). Real time RT-PCR indicated the lowest levels of expression of osteonectin, core binding factor-α 1, lipoprotein lipase and cartilage oligo matrix protein in medium-1 cultures as compared to media 2 and 3. Early passages expressed higher levels of peroxisome proliferator-activator receptor-γ2 in medium 1 than in media 2 and 3, whereas no difference of Sox-9 expression was noticed among the 3 media. Expression of apoptosis- and senescence-related genes (Bax, BCL-2 and P16INK4a) exhibited the lowest level of Bax/BCL-2 ratio and P16INK4a gene expression of hBMSC in medium 1. In conclusion, the replacement of FCS by recombinant EGF and PDGF promoted rapid proliferation of hBMSCs without inducing differentiation of hBMSCs. It also inhibited expression of apoptosis-related genes and limited replicative senescence during repeated passaging. Media with the lowest possible FCS content and replacement by EGF and PDGF thus should be used for 2D culturing during expansion of hBMSCs, whereas β-mercaptoethanol and high concentrations of FCS can help to commence osteogenic differentiation.
