Abstract:Extreme learning machine (ELM) has been applied in near infrared spectral analysis as a novel chemometric method which attracted the attentions of various researchers. However, the dimension of spectral data is usually very high while more hidden nodes should be incorporated in original ELM model for spectral data. Thus the problems of high dimension and high co-linearity in the output matrix of hidden layer of ELM model are inevitable. The solutions obtained with the existing Moore-Penrose generalized inverse can be ill-conditional due to the high dimension and high colinearity in the hidden layer output matrix. This study aims to propose an improved ELM to build spectral regression model. The proposed method firstly uses extreme learning machine (ELM) to relate spectral variables to response variable; then the output of each hidden node are treated as new variables; VIP-SPLS (improved stacked PLS based on variable importance in the projection) proposed by our group recently is used to build the regression model between those new variables and the response variable. In this paper, this method is called as improved ELM (iELM). VIP-SPLS model can fully utilize the output information of each hidden node and can effectively solve the problems of high dimension and high colineariy. At the same time, VIP-SPLS also has the advantage of model ensemble. Therefore, the performance of ELM model used for spectral data can be improved if the VIP-SPLS is incorporated to relate the hidden layer output matrix and response variable. The proposed method is applied to a commonly used benchmark NIR spectral data for evaluation. The results demonstrate that the precision improvement of iELM model is 29. 06% to PLS model and 27. 47% to original ELM model, respectively.
Feature Engineering in Discrimination of Herbal Medicines from Different Geographical Origins with Electronic Nose

Abstract:As pharmacists attach great significance to geographical origins of herbal medicines, cheap, nondestructive and convenient methods for discriminating herbal medicines originated from diverse regions are much in need. This work proposes a method of using electronic nose to discriminate herbal medicines from different origins. With 5 categories of herbal medicines and 3 to 4 geographical origins for each category, 8 pattern recognition algorithms prove the feasibility of the classification task and SVM, LDA and BP neural network have shown better classification accuracy. Additionally, feature engineering approaches are used to facilitate classification, showing that normalization based on each feature and each sensor and centralization prove to be better normalization approaches for classifiers; a proper degree of noise addition help classifiers get better generalization ability; finally, feature selection with SNR could lead to more efficient classifiers by selecting the most meaningful features and disregarding unnecessary features. This work provides insights for future herbal medicine evaluation based on electronic nose with better combinations of pattern recognition algorithms and feature engineering approaches for optimal classification performances.
On-Site Identification of Counterfeit Drugs Based on Near-Infrared Spectroscopy Siamese-Network Modeling

Abstract:Near-infrared spectroscopy (NIR) has become one of the most important methods for counterfeit drugs identification for its low cost, non-destructive, and on-site detection. However, it is often invalid for unknown samples beyond the scope of modeling samples, as well as it is not efficient in conditions of insufficient samples (insufficient number of samples within the class), unbalanced samples (large difference in the number of samples between classes), and sensitivity of identification results (different tolerance for different errors in application scenarios). To solve these problems, this paper proposes a general method for on-site identification of counterfeit drugs based on Siamese-network modeling with near-infrared spectroscopy, which especially constructs the train set and test set, learning the general knowledge of spectral differences to identify the different drugs by a costumed one-dimensional convolution neural network (1D-CNN), and finally answered the question of whether the on-site two spectra are pointed to the same drug. Based on experimental modeling samples of 1314 spectra, which are involved 9 drugs produced by 25 manufacturers, this paper has constructed and fully trained its model. Then, not known at the time of modeling, 4 drugs produced by 9 manufacturers are used for testing in the on-site application, and the accuracy rate amounts to 97.3%. For generalizing consideration, randomly divided into training and testing categories, the 32015 spectra of 135 drugs produced by 391 manufacturers in the spectral library are handled by the same processing. The generalization model is equally applicable, and the accuracy is above 97%. Compared with traditional binary classification identification methods such as SVM, PLS-DA, Auto-encoding, and one class (OC) threshold identification algorithms such as SVM-OC, SIMCA, conformity test (CT), the proposed method has the best identification ability for unknown samples-in modeling.

Abstract:To improve the performance of near-infrared spectral analysis, this paper proposes a soft-sensing method (PLS-DA-SVM) which combines partial least squares discriminant analysis (PLS-DA) and support vector machine (SVM). Based on training samples with several classes, firstly, a binary tree built by PLS-DA is introduced for multiple classification; secondly, sub-models for quantitative analy are constructed by partial least squares support vector machine (PLS-SVM). For a test sample, PLS-DA classification tree serves to determine its class, and the corresponding PLS-SVM sub-model is selected for quantitative analysis. A PLS-DA-SVM model with near-infrared spectroscopy data was established to determine the research octane number of gasoline samples. Monte Carlo cross validation was preformed with 57 product gasoline samples from 2 oil refineries. Results show that mean classification error rate for the recognition of gasoline brands is 0.07%, which is lower than other pattern recognition methods. Root mean square error of prediction (RMSEP) is reduced to 0.243 and correlation coefficient (R 2) is up to 0.991, which show great improvement upon PLS, LS-SVM and other modeling methods.

Abstract:A discriminant analysis technique using wavelet transformation (WT) and influence matrix analysis (CAIMAN) method is proposed for the near infrared (NIR) spectroscopy classification. In the proposed methodology, NIR spectra are decomposed by WT for data compression and a forward feature selection is further employed to extract the relevant information from the wavelet coefficients, reducing both classification errors and model complexity. A discriminant-CAIMAN (D-CAIMAN) method is utilized to build the classification model in wavelet domain on the basis of reduced wavelet coefficients of spectral variables. NIR spectra data set of 265 salviae miltiorrhizae radix samples from 9 different geographical origins is used as an example to test the classification performance of the algorithm. For a comparison, k-nearest neighbor (KNN), linear discriminant analysis (LDA) and quadratic discriminant analysis (QDA) methods are also employed. D-CAIMAN with wavelet-based feature selection (WD-CAIMAN) method shows the best performance, achieving the total classification rate of 100% in both cross-validation set and prediction set. It is worth noting that the WD-CAIMAN classifier also shows improved sensitivity, selectivity and model interpretability in the classifications.

Abstract:Near-infrared (NIR) spectral analysis, which has the advantages of rapidness, nondestruction and high-efficiency, is widely used in the detection of feed, food and mineral. In terms of qualitative identification, it can also be used for the discriminant analysis of medicines. Long short-term memory (LSTM) neural network, bidirectional long short-term memory (BiLSTM) neural network and gated recurrent unit (GRU) network are variants of the recurrent neural network (RNN). The potential relationship between nonlinear features learned from the sequence by these variants is used to complete the missions in fields such as natural language processing, signal classification and video analysis. Since the effect of these variants in drug identification is still to be studied, this paper constructs a multiclassifier of these three variants, using compound [Formula: see text]-keto acid tablets produced by four manufacturers and repaglinide tablets produced by five manufacturers as the research object. Then, the paper analyzes the impacts of seven different pre-processed methods on the drug NIR data by constructing different layers of LSTM, BiLSTM and GRU networks and compares different classification model indicators and training time of each model. When the spectrum data are pre-processed by [Formula: see text]-score normalization, the GRU-3 model has the best accuracy in all models. The BiLSTM models are better for analyzing high coincidence data. The method proposed in this paper can be further extended to other NIR spectroscopy data sets.

Abstract:In order to improve the accuracy of gasoline octane number detection by the near infrared ( NIR) spectroscopy, an improved extreme learning machine ( iELM ) algorithm combined with the extreme learning machine( ELM) algorithm and the improved stacked partial least square regression based on the variable importance in the projection(VIP-SPLS)algorithm is proposed here. And it solves the problem of high dimension and high collinearity in the output matrix of hidden layer of the ELM algorithm effectively. Then the proposed method is applied to a commonly used benchmark NIR spectral data of gasoline octane number detection. The results show that, compared with the PLS model and the ELM model, the accuracy of iELM model is increased by 20. 0% and 29. 3%respectively. The experiment shows that the iELM algorithm can be applied to the gasoline octane number detection by the near infrared spectroscopy and its accuracy is satisfactory.
Detection Method for Walnut Shell-Kernel Separation Accuracy Based on Near-Infrared Spectroscopy.

Abstract:In this study, Near-infrared (NIR) spectroscopy was adopted for the collection of 1200 spectra of three types of walnut materials after breaking the shells. A detection model of the walnut shell-kernel separation accuracy was established. The preprocessing method of de-trending (DT) was adopted. A classification model based on a support vector machine (SVM) and an extreme learning machine (ELM) was established with the principal component factor as the input variable. The effect of the penalty value (C) and kernel width (g) on the SVM model was discussed. The selection criteria of the number of hidden layer nodes (L) in the ELM model were studied, and a genetic algorithm (GA) was used to optimize the input layer weight (W) and the hidden layer threshold value (B) of the ELM. The results revealed that the classification accuracy of SVM and ELM models for the shell, kernel, and chimera was 97.78% and 97.11%. The proposed method can serve as a reference for the detection of walnut shell-kernel separation accuracy.

Abstract:Near-infrared spectroscopy(NIRS) combined with band screening method and modeling algorithm can be used to achieve the rapid and non-destructive detection of the traditional Chinese medicine(TCM) production process. This paper focused on the ginkgo leaf macroporous resin purification process, which is the key technology of Yinshen Tongluo Capsules, in order to achieve the rapid determination of quercetin, kaempferol and isorhamnetin in effluent. The abnormal spectrum was eliminated by Mahalanobis distance algorithm, and the data set was divided by the sample set partitioning method based on joint X-Y distances(SPXY). The key information bands were selected by synergy interval partial least squares(siPLS); based on that, competitive adaptive reweighted sampling(CARS), successive projections algorithm(SPA) and Monte Carlo uninformative variable(MC-UVE) were used to select wavelengths to obtain less but more critical variable data. With selected key variables as input, the quantitative analysis model was established by genetic algorithm joint extreme learning machine(GA-ELM) algorithm. The performance of the model was compared with that of partial least squares regression(PLSR). The results showed that the combination with siPLS-CARS-GA-ELM could achieve the optimal model performance with the minimum number of variables. The calibration set correlation coefficient R_c and the validation set correlation coefficient R_p of quercetin, kaempferol and isorhamnetin were all above 0.98. The root mean square error of calibration(RMSEC), the root mean square error of prediction(RMSEP) and the relative standard errors of prediction(RSEP) were 0.030 0, 0.029 2 and 8.88%, 0.041 4, 0.034 8 and 8.46%, 0.029 3, 0.027 1 and 10.10%, respectively. Compared with the PLSR me-thod, the performance of the GA-ELM model was greatly improved, which proved that NIRS combined with GA-ELM method has a great potential for rapid determination of effective components of TCM.
Rapid monitoring approaches for concentration process of lanqin oral solution by near-infrared spectroscopy and chemometric models

Abstract:Qualitative and quantitative detection methods based on near-infrared spectroscopy (NIRs) have been proposed in the process analysis of traditional Chinese medicine in recent years. In this study, rapid monitoring methods were developed for quality control of concentration process of lanqin oral solution (LOS). Partial least squares regression (PLSR) method was adopted to construct quantitative models for epigoitrin, geniposide, baicalin, berberine hydrochloride and density. Simultaneously, the genetic algorithm joint extreme learning machine (GA-ELM) was first applied in qualitative analysis of NIRs to distinguish end point of concentration process. Results of PLSR models were satisfactory with the relative standard error of calibration valued at 3.80%, 3.75%, 3.79%, 11.5% and 1.22% for epigoitrin, geniposide, baicalin, berberine hydrochloride and density respectively, and the residual predictive deviation values were higher than 3. For qualitative analysis, the GA-ELM model obtained 100% prediction accuracy. The PLSR quantitative models and the end point discrimination model constructed by GA-ELM correspond with the requirements of practical applications. Abstract:An on-line detection method combining near infrared (NIR) spectroscopy with local partial least squares regression (PLSR) was investigated for the elution process of sodium aescinate. A 2 mm pathlength flow cell which transmitted NIR radiation through two fiber optic probes was placed in bypass of the macroporous resin column to collect real-time spectra of the sodium aescinate eluate. To compare the predictive accuracy, both local and global PLSR were employed to build mathematical models between NIR spectra and reference values. Meanwhile, Mahalanobis distance was introduced to select the appropriate local model for the prediction of unknown samples. Experimental results demonstrated that local PLSR was superior to global PLSR in both calibration performance and predictive accuracy. Moreover, the on-line detection method was proven to be feasible in real application and thereby would be of great value for monitoring the elution process of sodium aescinate in real time as well as determining the start and end points of eluate collection.

Abstract:As a rapid and accurate technique, near infrared (NIR) spectroscopy has been widely used in process monitoring to improve efficiency and ensure quality consistency. In the present work, the application of NIR spectroscopy for on-line quantitative monitoring of the concentration process of Wangbi tablets was investigated. Partial least squares regression (PLSR) models based on the full-range spectra or the key wavelengths selected by the competitive adaptive reweighted sampling (CARS) method were established. The accuracy of the CARS-PLSR method was greater than that of full-spectrum PLSR for three quality parameters (soluble solid contents, paeoniflorin and icariin). For the prediction set samples using the CARS-PLSR models the coefficients of determination (r(2)) and root mean square error [RMSEP (%)] were 0.97 and 1.29 for soluble solid contents, 0.93 and 0.014 for paeoniflorin and 0.85 and 0.009 for icariin, respectively. Overall the results indicated that NIR spectroscopy coupled with CARS-PLSR calibration is a reliable and non-destructive alternative method for on-line monitoring of the concentration process of Wangbi tablets on an industrial scale.

Abstract:A method for the rapid identification of the genuineness of Chinese medicines based on near infrared (NIR) spectroscopy and least square support vector machines (LSSVM) was proposed. In this study, NIR spectra of the powdered Danshen (Radix Salviae Miltiorrhizae) were collected, and the nonlinear classifier based on LSSVM algorithm was developed to discriminate the genuineness of these herbs. The result obtained by the proposed method was compared with that from the traditional support vector machine (SVM) and back propagation-artificial neural network (BP-ANN) methods.. It was shown that the generalization performance of the classifier based on LSSVM was much better than that of BP-ANN, while thecomputation time of LSSVM is much shorter than that of the traditional SVM. The method proposed can be applied to the rapid and accurate identification of the quality of the naturalproducts.
Discrimination of Tetrastigma hemsleyanum according to geographical origin by near-infrared spectroscopy combined with a deep learning approach

Abstract:Recently, deep learning has presented as a powerful approach to overcome the deficiencies of the conventional biochemical approaches. In this study, a method for discriminating medicinal plant Tetrastigma hemsleyanum from different origins was proposed using near-infrared spectroscopy (NIRS) and deep learning models. Support vector machine (SVM), self-adaptive evolutionary extreme learning machine (SAE-ELM), and convolutional neural network (CNN) were used to process the near-infrared spectral data (4000-5600 cm-1). The results indicated that the average recognition accuracy of SVM on the test set samples (n = 60) reached 90%. The average recognition accuracy of SAE-ELM was 98.3%, while CNN correctly discriminated 100% of T. hemsleyanum from different origins. Notably, CNN avoids tedious redundant data preprocessing and is also able to save the trained model for the next call to achieve rapid detection. As above, this study provides an effective deep learning-based method for discriminating the geographical origins of T. hemsleyanum as well as providing a convenient and satisfactory approach to ensure the famous-region of other medicinal plants.
Rapidly detecting fennel origin of the near-infrared spectroscopy based on extreme learning machine

Abstract:Fennel contains many antioxidant and antibacterial substances, and it has very important applications in food flavoring and other fields. The kinds and contents of chemical substances in fennel vary from region to region, which can affect the taste and efficacy of the fennel and its derivatives. Therefore, it is of great significance to accurately classify the origin of the fennel. Recently, origin detection methods based on deep networks have shown promising results. However, the existing methods spend a relatively large time cost, a drawback that is fatal for large amounts of data in practical application scenarios. To overcome this limitation, we explore an origin detection method that guarantees faster detection with classification accuracy. This research is the first to use the machine learning algorithm combined with the Fourier transform-near infrared (FT-NIR) spectroscopy to realize the classification and identification of the origin of the fennel. In this experiment, we used Rubberband baseline correction on the FT-NIR spectral data of fennel (Yumen, Gansu and Turpan, Xinjiang), using principal component analysis (PCA) for data dimensionality reduction, and selecting extreme learning machine (ELM), Convolutional Neural Network (CNN), recurrent neural network (RNN), Transformer, generative adversarial networks (GAN) and back propagation neural network (BPNN) classification model of the company realizes the classification of the sample origin. The experimental results show that the classification accuracy of ELM, RNN, Transformer, GAN and BPNN models are above 96%, and the ELM model using the hardlim as the activation function has the best classification effect, with an average accuracy of 100% and a fast classification speed. The average time of 30 experiments is 0.05 s. This research shows the potential of the machine learning algorithm combined with the FT-NIR spectra in the field of food production area classification, and provides an effective means for realizing rapid detection of the food production area, so as to merchants from selling shoddy products as good ones and seeking illegal profits.
Discrimination of Trichosanthis Fructus from Different Geographical Origins Using Near Infrared Spectroscopy Coupled with Chemometric Techniques

Abstract:Near infrared (NIR) spectroscopy with chemometric techniques was applied to discriminate the geographical origins of crude drugs (i.e., dried ripe fruits of Trichosanthes kirilowii) and prepared slices of Trichosanthis Fructus in this work. The crude drug samples (120 batches) from four growing regions (i.e., Shandong, Shanxi, Hebei, and Henan Provinces) were collected, dried, and used and the prepared slice samples (30 batches) were purchased from different drug stores. The raw NIR spectra were acquired and preprocessed with multiplicative scatter correction (MSC). Principal component analysis (PCA) was used to extract relevant information from the spectral data and gave visible cluster trends. Four different classification models, namely K-nearest neighbor (KNN), soft independent modeling of class analogy (SIMCA), partial least squares-discriminant analysis (PLS-DA), and support vector machine-discriminant analysis (SVM-DA), were constructed and their performances were compared. The corresponding classification model parameters were optimized by cross-validation (CV). Among the four classification models, SVM-DA model was superior over the other models with a classification accuracy up to 100% for both the calibration set and the prediction set. The optimal SVM-DA model was achieved when C =100, γ = 0.00316, and the number of principal components (PCs) = 6. While PLS-DA model had the classification accuracy of 95% for the calibration set and 98% for the prediction set. The KNN model had a classification accuracy of 92% for the calibration set and 94% for prediction set. The non-linear classification method was superior to the linear ones. Generally, the results demonstrated that the crude drugs from different geographical origins and the crude drugs and prepared slices of Trichosanthis Fructus could be distinguished by NIR spectroscopy coupled with SVM-DA model rapidly, nondestructively, and reliably.

Abstract:With the traditional Chinese medicine herbsangelicae dahuricae radix (ADR or Baizhi) andsalviae miltiorrhizae radix (SMR or Danshen) as two examples, this work studies the automatic discrimination of the geographic origins of the herbs using near infrared (NIR) reflectance spectroscopy. Multi-class support vector machine (SVM) is utilized for the purpose, and recursive SVM is utilized to select the feature spectral segments that are decisive for the discrimination. With only 5 and 8 short spectral segments, discriminative accuracies of 92% are achieved on independent test sample sets. This work not only provides a prototype of accurate rapid discriminating systems for quality control of herbal medicines, but also opens new possibilities in studying subtle differences in the chemical compositions of herbs from different cultivation conditions and investigating their associations with the effectiveness of the herbs.

Abstract:Background: X-ray absorption spectroscopy (XAS) is a widely used substance analysis technique. It bases on the different absorption coefficients at different energy level to achieve material identification. Additionally, the combination of spectral technology and deep learning can achieve auto detection and high accuracy in material identification. Objectives: Current methods are difficult to identify drugs quickly and nondestructively. Therefore, we explore a novel approach utilizing XAS for the detection of prohibited drugs with common X-ray tube source and photon-counting (PC) detector. Method: To achieve automatic, rapid, and accurate detection of drugs. A CdTe detector and a common X-ray source were used to collect data, then dividing the data into training and testing sets. Finally, the improved transformer encoder model was used for classification. LSTM and ResU-net models are selected for comparation. Result: Fifty substances, which are isomers or compounds with similar molecular formulas of drugs, were selected for experiment substances. The results showed that the improved transformer model achieving 1.4 hours for training time and 96.73% for accuracy, which is better than the LSTM (2.6 hours and 65%) and ResU-net (1.5 hours and 92.7%). Conclusion: It can be concluded that the attention mechanism is more accurate for spectral material identification. XAS combined with deep learning can achieve efficient and accurate drug identification, offering promising application in clinical drug testing and drug enforcement.

Fuzzy neural network classifier for fast evaluating the quality of Chinese traditional medicine products using near infrared spectroscopy

Abstract:To solve the problem of fast identifying the quality sort of chinese traditional medicine products with nonlinear and fuzzy edges of the quality sort, a new method combining near infrared spectroscopy (NIRS) with fuzzy neural network was proposed. The method can differentiate the pattern classification of NIRS of chinese traditional medicine products with complex chemical components, resulting in fast evaluating product quality. An example of distinguishing the manufacturers of Shenmai injection was used to test the performance of the proposed method. The results showed that the classification accuracy reached 94.2%, obviously better than that of classical BP neural network (84.6%). It was verified that the new method could be used for fast evaluating the quality of chinese traditional medicine products.

Abstract:Afghanistan and Myanmar are two overwhelming opium production places. In this study, rapid and efficient methods for distinguishing opium from Afghanistan and Myanmar were developed using infrared spectroscopy (IR) coupled with multiple machine learning (ML) methods for the first time. A total of 146 authentic opium samples were analyzed by mid-IR (MIR) and near-IR (NIR), within them 116 were used for model training and 30 were used for model validation. Six ML methods, including partial least squares discriminant analysis (PLS-DA), orthogonal PLS-DA (OPLS-DA), k-nearest neighbour (KNN), support vector machine (SVM), random forest (RF), and artificial neural networks (ANNs) were constructed and compared to get the best classification effect. For MIR data, the average of precision, recall and f1-score for all classification models were 1.0. For NIR data, the average of precision, recall and f1-score for different classification models ranged from 0.90 to 0.94. The comparison results of six ML models for MIR and NIR data showed that MIR was more suitable for opium geography classification. Compared with traditional chromatography and mass spectrometry profiling methods, the advantages of MIR are simple, rapid, cost-effective, and environmentally friendly. The developed IR chemical profiling methodology may find wide application in classification of opium from Afghanistan and Myanmar, and also to differentiate them from opium originating from other opium producing countries. This study presented new insights into the application of IR and ML to rapid drug profiling analysis.
