Abstract:The erectile status of penile tissue is governed largely by the tone of cavernosal smooth muscle cells, which is determined by the balance of vascular relaxants and constrictors. Vascular relaxants play a key role in regulating the tone of cavernosal smooth muscle and thus the initiation and maintenance of penile erection. Early studies drew attention to the potential role of adenosine signaling in this process. However, the serendipitous discovery of the effect of sildenafil on erectile physiology drew more attention toward nitric oxide (NO) as a vasodilator in the process of penile erection, and a recently discovered, unexpected erectile phenotype of adenosine deaminase–deficient mice reemphasizes the importance of adenosine as a key regulatory of erectile status. Adenosine, like NO, is a potent and short-lived vasorelaxant that functions via cyclic nucleotide second messenger signaling to promote smooth muscle relaxation. Recent studies reviewed here show that adenosine functions to relax the corpus cavernosum and promote penile erection. Excess adenosine in penile tissue contributes to the disorder called priapism, and impaired adenosine signaling is associated with erectile dysfunction. More recent research summarized in this review reveals that adenosine functions as a key endogenous vasodilator in the initiation and maintenance of normal penile erection. This new insight highlights adenosine signaling pathways operating in penile tissue as significant therapeutic targets for the treatment of erectile disorders. Excessive Penile Norepinephrine Level Underlies Impaired Erectile Function in Adenosine A1 Receptor Deficient Mice

Abstract:Introduction. Penile erection is a complex neurovascular physiological event controlled by multiple factors and signaling pathways. A considerable amount of evidence indicates that adenosine plays a significant role in cavernosal smooth muscle relaxation. However, the specific role of adenosine and its receptors in erectile physiology and pathology is not fully understood. Aim. To determine the role of the adenosine A1 receptor (ADORA1) in penile erection. Method. Adenosine A1 receptor deficient (Adora1-/-) mice and aged-matched wild-type (WT) mice were utilized. We evaluated the in vivo erectile function by measuring the intracavernosal pressure (ICP) in response to cavernous nerve stimulation (CNS). Enzyme-linked immunosorbent assay was used to measure the norepinephrine (NE) plasma concentration in the corpus cavernosum and systemic circulation. We also evaluated the myosin light chain phosphorylation (p-MLC) in penile tissue pre- and post-CNS. Main Outcome Measurement. The main outcome measurement of this research was the evaluation of in vivo erectile response to CNS by measuring the ICP in Adora1-/- mice and WT mice and to identify the localization and specific neuron types of ADORA1 expression by dual immunostaining and immunofluorescence co-localization. Result. In vivo, both the ratio of CNS-induced Maximum ICP to mean arterial pressure and CNS-induced slope in Adora1-/- mice were significantly lower than WT mice. At the cellular level in penile tissue, we determined that ADORA1 was highly abundant in neuronal cells. During penile erection, Adora1-/- mice exhibited a higher level of NE plasma concentration in the penis than WT mice. And WT mice had a significantly greater reduction in p-MLC compared to Adora1-/- mice. Conclusion. Our results show that ADORA1 is enriched on neuron cells where it functions to control NE release. Activation of this receptor during penile erection results in reduced NE release and reduced cavernosal smooth muscle contraction, therefore facilitating penile erection. Norepinephrine-Induced Calcium Mobilization in C6 Glioma Cells

Abstract:AIM: To investigate the mechanism underlying the norepinephrine-induced elevation in intracellular calcium concentration ([Ca2+](i)) in C6 glioma cells. METHODS: Measurement of [Ca2+](i) was carried out using the dual-wavelength fluorescence method with fura-2 as the indicator. RESULTS: Norepinephrine was found to induce concentration-dependent increases in [Ca2+](i) through alpha(1)-adrenoreceptors. The [Ca2+](i) elevations were extracellular-calcium independent and not influenced by the treatment of pertussis toxin. Pretreatments with either U73122 or thapsigargin abolished the subsequent cellular calcium responses to norepinephrine. Preincubation with phorbol 12-myristate 13-acetate ( PMA) significantly reduced the [Ca2+](i) elevations, while protein kinase C inhibitors Ro31-8220 or GF-109203X completely blocked the inhibitory action of PMA. However, drugs either activating or inhibiting the function of protein kinase A had no effect on the [Ca2+](i) elevations. CONCLUSION: Norepinephrine induces calcium mobilization from internal stores by activation of phospholipase C in C6 cells. The [Ca2+](i) elevation is negatively regulated by the activation of protein kinase C.
Adenosine Inhibits L- and N-Type Calcium Channels in Pituitary Melanotrophs. Evidence for the Involvement of a G Protein in Calcium Channel Gating

Abstract:It has been previously demonstrated that activation of A(1) adenosine receptors in frog melanotrophs causes inhibition of spontaneous action potential discharges and alpha-melanocyte-stimulating hormone secretion. In the present study, we have investigated the effect of adenosine on high-voltage-activated (HVA) calcium currents In cultured melanotrophs, using the whole-cell variant of the patch-clamp technique with barium as a charge carrier. Adenosine and the specific A(1) adenosine receptor agonist R-PIA (50 mu M each) produced a decrease of the amplitude of the barium current, while the selective A(2) adenosine receptor agonist CGS 21680 did not affect the current. The inhibitory effect of R-PIA was observed throughout the activation range of the current, with stronger responses at more positive potentials. R-PIA inhibited both the L- and N-type components of the current, the effect on the N-component being two-fold higher than on the L-component, The inhibitory effect of R-PIA was rendered irreversible by addition of GTP gamma S (100 mu M) to the intracellular solution. Pre-treatment of the cells with pertussis toxin (1 mu g/ml; 12 h), totally abolished the effect of R-PIA on the HVA calcium channels. Conversely, addition of a high concentration of cAMP (100 mu M) together with the phosphodiesterase inhibitor IBMX (100 mu M) to the intracellular solution did not modify the effect of R-PIA on the current. It is concluded that, in frog melanotrophs, adenosine induces inhibition of L- and N-calcium currents and that this effect is mediated by a pertussis toxin-sensitive G protein. Our data also indicate that the inhibitory effect of adenosine on the calcium currents is not mediated by inhibition of adenylyl cyclase.

Abstract:Aim: To investigate the alpha(1B)-adrenoceptor (alpha(1B)-AR)-mediated cAMP response and underlying mechanisms in HEK293 cells. Methods: Full-length cDNA encoding alpha(1B)-AR was transfected into HEK293 cells using the calcium phosphate precipitation method, and alpha(1B)-AR expression and cAMP accumulation were determined by using the saturation radioligand binding assay and ion-exchange chromatography, respectively. Results: Under agonist stimulation, alpha(1B)-AR mediated cAMP synthesis in HEK293 cells, and blockade by PLC-PKC or tyrosine kinase did not reduce cAMP accumulation induced by NE. Pretreatment with pertussis toxin (PTX) had little effect on basal cAMP accumulation as well as norepirephrine (NE)-stimulated cAMP accumulation. In addition, pretreatment with cholera toxin (CTX) neither mimicked nor blocked the effect induced by NE. The extracellular Ca2+ chelator egtazic acid (EGTA), nonselective Ca2+ channel blocker CdCl2 and calmodulin (CaM) inhibitor W-7 significantly reduced NE-induced cAMP accumulation from 1.59% +/- 0.47% to 1.00% +/- 0.31%, 0.78% +/- 0.23%, and 0.90% +/- 0.40%, respectively. Conclusion: By coupling with a PTX-insensitive G protein, alpha(1B)-AR promotes Ca2+ influx via receptor-dependent Ca2+ channels, then Ca2+ is linked to CaM to form a Ca2+-CaM complex, which stimulates adenylyl cyclase (AC), thereby increasing the cAMP production in HEK293 cell lines.
Activation of Α1‐adrenoceptors Enhances Excitatory Synaptic Transmission Via a Pre‐ and Postsynaptic Protein Kinase C‐dependent Mechanism in the Medial Prefrontal Cortex of Rats

Abstract:The physiological effects of alpha 1-adrenoceptors (alpha 1-ARs) have been examined in many brain regions. However, little is known about the mechanism of modulation on synaptic transmission by alpha 1-ARs in the medial prefrontal cortex (mPFC). The present study investigated how alpha(1)-AR activation regulates glutamatergic synaptic transmission in layer V/VI pyramidal cells of the rat mPFC. We found that the alpha(1)-AR agonist phenylephrine (Phe) induced a significant enhancement of the amplitude and frequency of miniature excitatory postsynaptic currents (mEPSCs). The facilitation effect of Phe on the frequency of mEPSCs involved a presynaptic protein kinase C-dependent pathway. Phe produced a significant enhancement on the amplitude of alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor (AMPA-R)- and N-methyl-d-aspartic acid receptor (NMDA-R)-mediated evoked excitatory postsynaptic currents (eEPSCs). Phe enhanced inward currents evoked by puff application of glutamate or NMDA. The Phe-induced facilitation of AMPA-R- and NMDA-R-mediated eEPSCs required, in part, postsynaptic G(q), phospholipase C and PKC. These findings suggest that alpha(1)-AR activation facilitates excitatory synaptic transmission in mPFC pyramidal cells via both pre- and post-synaptic PKC-dependent mechanisms.

Abstract:Incubation with noradrenaline (norepinephrine) of isolated membranes of rat's brain corpus striatum and cortex, showed that ionic-magnesium (Mg2+) is required for the neurotransmitter activatory response of Adenylate Cyclase [ATP pyrophosphate-lyase (cyclizing), (EC 4.6.1.1)], AC.An Mg2+-dependent response to the activatory effects of adrenaline, and subsequent inhibition by calcium, suggest capability for a turnover, associated with cyclic changes in membrane potential and participation in a short term-memory pathway.In the cell, the neurotransmitter by activating AC generates intracellular cyclic AMP. Calcium entrance in the cell inhibits the enzyme. The increment of cyclic AMP activates kinaseA and their protein phosphorylating activity, allowing a long term memory pathway. Hence, consolidating neuronal circuits, related to emotional learning and memory affirmation.The activatory effect relates to an enzyme-noradrenaline complex which may participate on the physiology of the fight or flight response, by prolonged exposure. However, the persistence of an unstable enzyme complex turns the enzyme inactive. Effect concordant, with the observation that prolonged exposure to adrenaline, participate in the etiology of stress triggered pathologies. At the cell physiological level AC responsiveness to hormones could be modulated by the concentration of Chelating Metabolites. These ones produce the release of free ATP4-, a negative modulator of AC and the Mg2+ activated insulin receptor tyrosine kinase (IRTK). Thus, allowing an integration of the hormonal response of both enzymes by ionic controls. This effect could supersede the metabolic feedback control by energy-charge. Accordingly, maximum hormonal response of both enzymes, to high Mg2+ and low free ATP4-, allows a correlation with the known effects of low caloric intake increasing average life expectancy.

Norepinephrine modulates calcium dynamics in cortical oligodendrocyte precursor cells promoting proliferation during arousal in mice

Abstract:Oligodendrocytes, the myelinating cells of the central nervous system (CNS), are generated from oligodendrocyte precursor cells (OPCs) that express neurotransmitter receptors. However, the mechanisms that affect OPC activity in vivo and the physiological roles of neurotransmitter signaling in OPCs are unclear. In this study, we generated a transgenic mouse line that expresses membrane-anchored GCaMP6s in OPCs and used longitudinal two-photon microscopy to monitor OPC calcium (Ca2+) dynamics in the cerebral cortex. OPCs exhibit focal and transient Ca2+ increases within their processes that are enhanced during locomotion-induced increases in arousal. The Ca2+ transients occur independently of excitatory neuron activity, rapidly decline when OPCs differentiate and are inhibited by anesthesia, sedative agents or noradrenergic receptor antagonists. Conditional knockout of α1A adrenergic receptors in OPCs suppresses spontaneous and locomotion-induced Ca2+ increases and reduces OPC proliferation. Our results demonstrate that OPCs are directly modulated by norepinephrine in vivo to enhance Ca2+ dynamics and promote population homeostasis.

Effect of Adenosine and Adenosine Receptor Antagonist on Müller Cell Potassium Channel in Rat Chronic Ocular Hypertension Models

Abstract:Müller cells are principal glial cells in rat retina and have attracted much attention in glaucoma studies. However, it is not clear whether adenosine and adenosine receptor (AR) antagonists play any roles in the regulation of potassium channels in Müller cells and subsequently in the promotion of glutamine synthetase (GS) and L-Glutamate/L-Aspartate Transporter (GLAST) functions. We found that chronic ocular hypertension (COH) in rat down-regulated Müller cells Kir2.1, Kir4.1, TASK-1, GS and GLAST expressions and attenuated the peak of inward potassium current. Retinal ganglion cells (RGC) count was lower in the COH rats than that in the sham operation animals. Intravitreal injection of selective A2A AR antagonist SCH442416 up-regulated Müller cell Kir4.1, TASK-1, GS and GLAST expressions and enhanced inward potassium currents compared with those in the COH rats with vehicle control. Meanwhile, the RGC count was higher following intravitreal injection of SCH442416 in the COH rats than that after vehicle injection. The fact that PKA inhibitor H-89 blocked these SCH442416 effects suggested that the PKA signaling pathway was involved in the observed ocular responses following the intravitreal SCH442416 injection.
Regulation of Period 1 Expression in Cultured Rat Pineal

Abstract:The aim of the present study was to investigate the in vitro expression of Period 1 (Per1), Period 2 (Per2) and arylalkylamine N-acetyltransferase (AA-NAT) genes in the rat pineal gland to understand the mechanism(s) regulating the expression of these genes in this organ. Pineals, when maintained in vitro for 5 days, did not show circadian rhythmicity in the expression of any of the three genes monitored. Norepinephrine (NE) induced AA-NAT and Per1, whereas its effect on Per2 was negligible. Contrary to what was observed in other systems, NE stimulation did not induce circadian expression of Per1. The effect of NE on Per1 level was dose- and receptor subtype-dependent, and both cAMP and cGMP induced Per1. Per1 was not induced by repeated NE - or forskolin - stimulation. Protein synthesis was not necessary for NE-induced Per1, but it was for reduction of Per1 following NE stimulation. Per1 transcription in pinealocytes was activated by BMAL1/CLOCK. Our results indicate that important differences are present in the regulation of these genes in the mammalian pineal.

Patch Clamp Study on Mechanism of Adenosine-Induced Inhibitory Effects in Frog Pituitary Melanotrophs.

Abstract:Our laboratory demonstrated that adenosine inhibits the activation of adenylyl cyclase and the secretion of the alpha-melanocyte-stimulating hormone (alpha-MSH) from the intermediate lobe of the frog pituitary. This paper showed the bioelectric effects induced by adenosine, the ionic conductances modulated by adenosine, and the possible involvement of intracellular messengers, indicated the mechanism by which adenosine controls the secretion of alpha-MSH. The results show that adenosine acting on A1 adenosine receptor subtype reduced the Ca2+ influx necessary for the secretion, through 4 distinct mechanisms: 1) a hyperpolarization resulting from the activation of a voltage-insensitive K+ conductance, 2) a reduction of the duration of spontaneous action potentials due to an increase of the outward delayed rectifyer K+ current (lk), 3) a diminution of the cellular excitability by an activation of the transient outward K+ current (lA), and 4) an inhibition of the L- and N-type Ca2+ currents, with a predominant action on the N-type component. Cell dialysis with GTP gamma S rendered irreversible the effects of adenosine on the K+ conductances and Ca2+ channels, whereas PTX pretreatment totally abolished the response to adenosine, suggesting all bioelectric effects of adenosine were mediated by pertussis toxin-sensitive G proteins. Whether the implicated G proteins regulate the K+ and Ca2+ channels by tight-coupling or via a second-messenger system remains to be solved. With our results, the involvement of adenylyl cyclase can be excluded because addition of cAMP and IBMX, an inhibitor of phosphodiesterases, in the intracellular solution, or application of dibutyryl cAMP in the extracellular solution did not modify the adenosine-induced responses.
Alpha1-adrenoceptor-mediated negative inotropic effect caused by intracellular ionic activities in guinea-pig papillary muscle

Abstract:In guinea-pig papillary muscle, phenylephrine (PE), an agonist of alpha1-adrenoceptor (alpha1-AR), led to a transient negative inotropic effect (NIE) and a subsequent sustained positive inotropic effect (PIE). To clarify the ionic mechanisms underlying the NIE, we measured the [Na+]i or [pH]i by ion-selective microelectrodes. PE produced a decrease in the intracellular Na+ concentration ([Na+]i) and an increase in intracellular pH ([pH]i). During the phase of NIE, PE produced only a (-) change of [Na+]i (Delta[Na+]i). With a decrease in extracellular Na+ or an increase in extracellular Ca2+, the PE-induced NIE was attenuated and PE produced (+)Delta[Na+]i. The PE-induced NIE and (-)Delta[Na+]i were definitely strengthened by lowering the bath temperature or increasing the stimulation frequency. 2-(2,6-di-methoxyphenoxyethyl)amino-methyl-1,4-benzidioxane HCl, an antagonist of alpha1A-AR, completely abolished the PE-induced NIE and (-)Delta[Na+]i. Phorbol 12,13-dibutyrate, an activator of protein kinase C (PKC), decreased the baseline [Na+]i and twitch force and increased the baseline [pH]i in mimicry of PE. Pretreatment with 1-5(isoquinolinesulphonyl)-2-methylpiperazine, an inhibitor of PKC, abolished the PE-induced NIE and (-)Delta[Na+]i. During pretreatment with benzamil, an inhibitor of Na+/Ca2+ exchange, we found that the PE-induced NIE and (-)Delta[Na+]i were reversibly abolished. Our results indicate the PE-induced NIE may be elicited upon the activation of Na+/Ca2+ exchange which can be attributed to the (-)Delta[Na+]i. (-)Delta[Na+]i is mediated through a PKC-dependent pathway via an activation of alpha1A-AR subtype and its effect could be strengthened remarkably at high [Na+]i and [Ca2+]i values.
LC-derived excitatory synaptic transmission to dorsal raphe serotonin neurons is inhibited by activation of alpha2-adrenergic receptors

Abstract:In the central nervous system, noradrenaline transmission controls the degree to which we are awake, alert, and attentive. Aberrant noradrenaline transmission is associated with pathological forms of hyper- and hypo-arousal that present in numerous neuropsychiatric disorders often associated with dysfunction in serotonin transmission. In vivo, noradrenaline regulates the release of serotonin because noradrenergic input drives the serotonin neurons to fire action potentials via activation of excitatory α1-adrenergic receptors (α1-A R ). Despite the critical influence of noradrenaline on the activity of dorsal raphe serotonin neurons, the source of noradrenergic afferents has not been resolved and the presynaptic mechanisms that regulate noradrenaline-dependent synaptic transmission have not been described. Using an acute brain slice preparation from male and female mice and electrophysiological recordings from dorsal raphe serotonin neurons, we found that selective optogenetic activation of locus coeruleus terminals in the dorsal raphe was sufficient to produce an α1-A R -mediated excitatory postsynaptic current (α1-A R -EPSC). Activation of inhibitory α2-adrenergic receptors (α2-A R ) with UK-14,304 eliminated the α1-A R -EPSC via presynaptic inhibition of noradrenaline release, likely via inhibition of voltage-gated calcium channels. In a subset of serotonin neurons, activation of postsynaptic α2-A R produced an outward current through activation of GIRK potassium conductance. Further, in vivo activation of α2-A R by systemic administration of clonidine reduced the expression of c-fos in the dorsal raphe serotonin neurons, indicating reduced neural activity. Thus, α2-A R are critical regulators of serotonin neuron excitability.

Rapid Succession within the Veillonella Population of a Developing Human Oral Biofilm In Situ

Abstract:ABSTRACT Streptococci are the primary component of the multispecies oral biofilm known as supragingival dental plaque; they grow by fermentation of sugars to organic acids, e.g., lactic acid. Veillonellae, a ubiquitous component of early plaque, are unable to use sugars; they ferment organic acids, such as lactate, to a mixture of shorter-chain-length acids, CO2, and hydrogen. Certain veillonellae bind to (coaggregate with) streptococci in vitro. We show that, between 4 and 8 hours into plaque development, the dominant strains of Veillonella change in their phenotypic characteristics (coaggregation and antibody reactivity) as well as in their genotypic characteristics (16S RNA gene sequences as well as strain level fingerprint patterns). This succession is coordinated with the development of mixed-species bacterial colonies. Changes in community structure can occur very rapidly in natural biofilm development, and we suggest that this process may influence evolution within this ecosystem.
Angiotensin-(1-7) Increases Neuronal Potassium Current Via A Nitric Oxide-Dependent Mechanism

Abstract:Angiotensin 1–7 [Ang‐(1–7)] signaling in the brain may play an important protective role in cardiovascular diseases by decreasing sympathetic output. It is generally well‐accepted that nitric oxide (NO•) in the brain functions as a sympatho‐inhibitory molecule. We hypothesized that Ang‐(1–7) increases NO• in neurons and modulates neuronal activity. We tested this hypothesis in catecholaminergic CATH.a neurons. Protein analysis revealed these neurons express the Ang‐(1–7) receptor (MasR) and three isoforms of NO• synthase (nNOS, eNOS, iNOS). Expression of MasR, nNOS, and eNOS protein markedly increased in CATH.a neurons following six days of differentiation with N6,2′‐O‐Dibutyryladenosine 3′,5′‐cyclic monophosphate sodium salt (dbcAMP, 1 mM). As measured by DAF‐FM fluorescence and confocal microscopy, Ang‐(1–7) (100 nM) increased NO• levels in differentiated CATH.a neurons (31 ± 3%; P<0.05 vs. baseline) and this response was attenuated (P<0.05) by the MasR antagonist A779 (5 ± 3%), non‐specific NOS inhibitor (L‐NAME; −5 ± 1%), and nNOS inhibitor (SMTC; −5 ± 7%). Using the whole‐cell patch‐clamp technique, we observed that Ang‐(1–7) increased outward K+ current (IKv) in differentiated CATH.a neurons (53 ± 6% increase; P<0.05 vs. vehicle). These findings indicate that Ang‐(1–7) increases nNOS‐dependent NO• levels via MasR activation and elevates IKv in differentiated CATH.a neurons. NIH P01HL062222

Abstract:Stimulation of alpha(2A)-adrenoceptors (ARs) in the prefrontal cortex (PFC) produces a beneficial effect on cognitive functions such as working memory. A previous study in our laboratory showed that alpha(2A)-AR stimulation suppresses excitatory synaptic transmission in layer V-VI pyramidal cells of the rat medial PFC (mPFC). However, the intracellular mechanism underlying the alpha(2A)-AR suppression remains unclear. In the present study, we recorded evoked excitatory postsynaptic current (eEPSC) in layer V-VI pyramidal cells of the mPFC, using whole-cell patch-clamp recording. We found that the alpha(2A)-AR agonist guanfacine significantly suppresses eEPSC in mPFC pyramidal cells. The alpha(2A)-AR inhibition is mediated by the Gi-cAMP-PKA-PP1-CaMKII-AMPAR signaling pathway, as such inhibition no longer exists when each step of this pathway is blocked with NF023, Rp-cAMP, PKI5-24 or H89, tautomycin, and KN-62 or KN-93, respectively.
[Pathways of Signal Transduction of Intracellular Ca2+ Increase Mediated by Three Subtypes of Alpha 1-Adrenoceptor in HEK293 Cells].

Abstract:In HEK293 cell lines in which alpha 1a-, alpha 1b- or alpha 1d-adrenoceptor subtypes were stably expressed, fluorescence intensities due to traces of Ca2+ were investigated using fura-2/AM with the aim of exploring signal transduction pathways of intracellular Ca2+ increase mediated by these receptor subtypes. Pertussis toxin had no effect on [Ca2+]i increase mediated by all three subtypes of alpha 1-adrenoceptors. However, U-73122 and PMA could inhibited the [Ca2+]i increase induced by NE, which was not affected by Foskolin and Rp-cAMPs. Pretreatment with calphostin C abolished the [Ca2+]i response to PMA. Quercetin and tyrphostin inhibited maximal [Ca2+]i increase but had no effect on the plateau phase in all the three transfected cells. In the HEK293 cells, the phosphatide-Ca2+ signalling system mediated by the 3 subtypes of alpha 1-AR was brought into play by activation of phospholiphase C via coupling with PTX-insensitive G proteins. PKC system inhibited both mobilization of internal Ca2+ stores and the Ca2+ influx across the plasma membrane. Tyrosine kinase, but not cAMP system, might participate in such Ca2+ signalling.

Abstract:We have previously reported that the binding site repertoires of heterodimers formed between retinoid X receptor (RXR) and either retinoic acid receptor (RAR) or thyroid hormone receptor (TR) bound to response elements consisting of directly repeated PuG(G/T)TCA motifs spaced by 1‐5 bp [direct repeat (DR) elements 1‐5] are highly similar to those of their corresponding DNA binding domains (DBDs). We have now mapped the dimerization surfaces located in the DBDs of RXR, RAR and TR, which are responsible for cooperative interaction on DR4 (RXR and TR) and DR5 (RXR and RAR). The D‐box of the C‐terminal CII finger of RXR provides one of the surfaces which is specifically required for the formation of the heterodimerization interfaces on both DR4 and DR5. Heterodimerization with the RXR DBD on DR5 specifically requires the tip of the RAR CI finger as the complementary surface, while a 7 amino acid sequence encompassing the ‘prefinger region’, but not the TR CI finger, is specifically required for efficient dimerization of TR and RXR DBDs on DR4. Importantly, DBD swapping experiments demonstrate not only that the binding site repertoires of the full‐length receptors are dictated by those of their DBDs, but also that the formation of distinct dimerization interfaces between the DBDs are the critical determinants for cooperative DNA binding of these receptors to specific DRs.
