Abstract:Isolation of new bacterial strains and recognition of their metabolic activities are highly desirable for sustainability of natural ecosystems. Biodegradation of dimethyl phthalate (DMP) under anoxic conditions has been shown to occur as a series of sequential steps using strain CW-1 isolated from digested sludge of Sibao Wastewater Treatment Plant in Hangzhou, China. The microbial colony on LB medium was yellowish, 3∼5 mm in diameter, convex in the center, and embedded in mucous externally. The individual cells of strain CW-1 are irregular rods, measuring (0.6∼0.7)×(0.9∼1.0) μm, V-shaped, with clubbed ends, Gram positive and without any filaments. 16S rDNA (1438 bp) sequence analysis showed that the strain was related to Arthrobacter sp. CW-1 and can degrade PAEs utilizing nitrate as electron acceptor, but cannot mineralize DMP completely. The degradation pathway was recommended as: dimethyl phthalate (DMP)→monomethyl phthalate (MMP)→phthalic acid (PA). DMP biodegradation was a first order reaction with degradation rate constant of 0.3033 d −1 and half-life 2.25 d. The DMP conversion to PA by CW-1 could be described by using sequential kinetic model.
Study on Gaseous Chlorobenzene Treatment by a Bio-Trickling Filter: Degradation Mechanism and Microbial Community

Abstract:Large-flow waste gas generated from the pharmaceutical and chemical industry usually contains low concentrations of VOCs (volatile organic compounds), and it is also the key factor that presents challenges in terms of disposal. To date, due to the limitations of mass transfer rate and microbial degradation ability, the degradation performance of VOCs using the biological method has not been ideal. Therefore, in this study, the sludge from a chlorobenzene-containing wastewater treatment plant was inoculated into our experimental bio-trickling filter (BTF) to explore the feasibility of domestication and degradation of gaseous chlorobenzene by highly active microorganisms. The kinetics of its mass transfer reaction and microbial community dynamics were also discussed. Moreover, the main process parameters of BTF for chlorobenzene degradation were optimized. The results showed that the degradation effect of chlorobenzene reached more than 85% at an inlet concentration of chlorobenzene 700 mg·m−3, oxygen concentration of 10%, and an empty bed retention time (EBRT) of 80 s. The mass transfer kinetic analysis indicated that the process of chlorobenzene degradation in the BTF occurred between the zero-stage reaction and the first-stage reaction. This BTF contributed significantly to the biodegradability of chlorobenzene, overcoming the limitation of gas-to-liquid/solid mass transfer of chlorobenzene. The analysis of the species diversity showed that Thermomonas, Petrimona, Comana, and Ottowia were typical organic-matter-degrading bacteria that degraded chlorobenzene efficiently with xylene present.
Isolation and Physiology of a Dimethyl Phthalate Degrading Bacterial Strain YZ2

Abstract:A bacterial strain YZ2 was isolated from digested sludge of Sibao wastewater treatment plant located in Hangzhou, China and was characterized using morphological, biochemical, and physiological characteristics. The microbial colonies on LB medium were yellowish; 3-5 mm in diameter and convex in the center. The individual cells of isolate were short rods, measuring 0.6 x 0.9 - 1.0 x 1.5 mu m with small side projections. clubbed ends, and without any filaments. Individual cells were grain-positive. Based on 16S rDNA (1438 bp) sequence analysis. isolate was identified as Rhodococcus sp. isolate YZ2 capable of dimethyl phthalate (DMP) biodegradation under nitrate-reducing conditions. Results of batch experiments showed that the degradation of LIMP was optimum at pH 7.5 and 31 degrees C. In the range of 70-600 mg l(-1), the initial DMP concentrations showed very high correlation,with biodegradation rate (0.9773 < r(2) < 0.9941), suggesting a first-order reaction. The half-life value showed an increase from 0.89 to 2.22 days during biodegradation of various DMP concentrations. HPLC and LC-MS analysis showed that AMP and PA may be the first and second intermediates, respectively, during DMP biodegradation under anoxic conditions. Highly efficient degradation of hydrogen sulfide, styrene, and m-xylene in a bio-trickling filter

Abstract:Controlling the release of malodorous gas discharged from wastewater treatment plants (WWTPs) has become an urgent environmental problem in recent years. In this study, a bio-trickling filter (BTF) inoculated with microorganisms acclimated to activated sludge in a WWTP was used as the degradation equipment. A continuous degradation experiment with hydrogen sulfide, styrene, and m-xylene in the BTF lasted for 84 days (12 weeks). The degradation capacities of the BTF for hydrogen sulfide, styrene, and m-xylene were evaluated, and the synergy and inhibition among the substrates during biodegradation are discussed. The results indicated that the degradation efficiencies of the BTF were as high as 99.2% for hydrogen sulfide, 94.6% for styrene, and 100.0% for m-xylene. When the empty bed residence time was 30 s, the maximum elimination capacities (EC) achieved for hydrogen sulfide was 38 g m-3 h-1, for styrene was 200 g m-3 h-1, and for m-xylene was 75 g m-3 h-1. Furthermore, the microbial species and quantity of microorganisms in the middle and top of the BTF were much higher than those at the bottom of the BTF. A variety of microorganisms in the BTF can efficiently degrade the typical and highly toxic malodorous gases released from WWTPs. This study can help increase the understanding of the degradation of a mixture of sulfur-containing substances and aromatic hydrocarbons in BTF degradation and promote the development of technologies for the reduction of a complex mixture of malodorous gas emissions from organic wastewater treatment.

Abstract:Hundreds of studies have demonstrated the bioremediation of chlorinated organic pollutants (COPs) in flooded environments. However, the role of specific functional strains in degrading COPs under complex media such as wetlands is still unclear. Here, we focused on the microbial characteristics of COP-polluted sediments, identified the bacteria responsible for degradation and conducted a genomic analysis of these bacteria. Four strains were obtained and identified as Petrimonas sulfuriphila PET, Robertmurraya sp. CYTO, Hungatella sp. CloS1 and Enterococcus avium PseS3, respectively. They were capable of degrading a typical COP, γ-hexachlorocyclohexane (γ-HCH). The residual γ-HCH concentrations were 58.8 % (PET), 45.6 % (CYTO), 60.2 % (CloS1), and 69.3 % (PseS3) of its initial value, respectively. Strain PET, CYTO and CloS1 could degrade γ-HCH to its dehalogenation product chlorobenzene. Each strain harbors genes annotated to the pathway of halogenated organic matter degradation (e.g. 2-haloacid dehalogenase) and cobalamin biosynthesis, which are involved in the degradation of COPs. Comparative genomic analysis of the four strains and other classical organohalide-respiring bacteria (e.g. Dehalococcoides mccartyi and Sulfurospirillum multivorans DSM 12446) showed that they share orthologous clusters related to the cobalamin biosynthetic process (GO:0009236). VB12 was also detected in the culture systems of the four strains, further highlighting the importance of cobalamin in COPs degradation. In the genome of the four strains, some genes were annotated to the halogenated organic matter degradation and cobalamin biosynthesis pathway within horizontal gene transfer (HGT) regions. This further indicated that microorganisms carrying these genes can adapt faster to pollution stress through HGT. Together, these findings revealed the co-evolution mechanism of functional strains and may provide novel insights into improved bioremediation strategies for COP-polluted complex media based on generalist organochlorine-degrading bacteria.

Abstract:In this study, one- and two-phase partitioning biotrickling filters (1P-BTF and 2P-BTF, respectively) inoculated with a pre-acclimated mixed culture were examined for the removal of hydrophobic and refractory o-xylene. A small fraction of silicone oil (5% v/v) was added as a non-aqueous phase. Due to the presence of silicone oil, the 2P-BTF exhibited superior performance and stability for o-xylene biodegradation at steady and transient operations. Higher macro-kinetic constants for o-xylene removal by the Michaelis-Menten model were obtained for the 2P-BTF with a saturation constant of 0.396 g m(-3) and a maximum elimination capacity of 105.7 g m(-3) h(-1). The enhancement of removal performance for the 2P-BTF was supported by dominant specialized microorganisms with o-xylene biodegradability. The diversity of microbial community was influenced by the presence of silicone oil. This study demonstrated that a BTF with 5% of silicone oil could be applied for the treatment of hydrophobic and refractory volatile organic compounds. It also provided valuable information for better understanding the relationship between microbial community and removal performance using two-phase partitioning bioreactors.

Abstract:Total petroleum hydrocarbon (TPH) in wastewater has attracted widespread attention for its environmental and biological health hazards. In the research, WY-4 strains with diesel degradation ability isolated from contaminated soil and response surface methodology was used to optimize the degradation conditions of WY-4. Fe-modified biochar (FPB) was used as an immobilized carrier, the environmental factors affecting the degradation of immobilized bacteria (FPBM) were explored and the degradation effect of FPBM was evaluated on real TPH-contaminated wastewater. Furthermore, the potential degradation mechanisms and possible degradation pathways of TPH were also explored. The results demonstrated that WY-4 was identified as Ochrobactrum sp., and its optimal growth conditions were pH 6.8, temperature 28.8°C and NaCl concentration 9.47 g/L. The removal efficiency by FPBM on 10,000 mg/L diesel wastewater was 72.5% and on real TPH-contaminated wastewater was 76.75% in 7 d, which was significantly higher than the degradation effect of free bacteria. The degradation pathway of two representative pollutants, naphthalene and indole, in the real TPH-contaminated wastewater was referred to be the catechol metabolic pathway. The results highlighted the potential of FPB-immobilized bacteria for the remediation of TPH-contaminated wastewater in harsh environments and provided an effective strategy for green remediation treatment of TPH contamination.

Isolation and Identification of Naphthalene-Degrading Bacteria and its Application in a Two-phase Partitioning Bioreactor

Abstract:Naphthalene is a persistent environmental pollutant for its potential teratogenic, carcinogenic and mutagenic effects. In this study, 10 strains of bacteria capable of degrading naphthalene were isolated from crude-oil contaminated soil. Among them, Pseudomonas plecoglossicida 2P exhibited prominent growth with 1000 mg/L naphthalene as the sole carbon source and degraded 94.15% of naphthalene in 36 h. Whole genome sequencing analysis showed that P. plecoglossicida 2P had a total of 22 genes related to naphthalene degradation, of which 8 genes were related to the salicylic acid pathway only, 5 genes were related to the phthalic acid pathway only, 8 genes were common in both the salicylic acid and phthalic acid pathways, and 1 gene was related to the gentisic acid pathway. P. plecoglossicida 2P was applied in a two-phase partition bioreactor (TPPB) to degrade naphthalene in wastewater. The optimal operating conditions of the reactor were obtained through response surface optimization: initial naphthalene concentration (C0) =1600 mg/L, bacterial liquid concentration (OD600) = 1.3, and polymer-to-wastewater mass ratio (PWR) = 2%. Under these conditions, the naphthalene degradation rate was 98.36% at 24 h. The degradation kinetics were fitted using the Haldane equation with a high coefficient of determination (R2=0.94). The present study laid foundations for naphthalene degradation mechanism of genus Pseudomonas and its potential application in TPPB.
Identification of an efficient phenanthrene-degrading Pseudarthrobacter sp. L1SW and characterization of its metabolites and catabolic pathway

Abstract:Phenanthrene, a typical chemical of polycyclic aromatic hydrocarbons (PAHs) pollutants, severely threatens health of wild life and human being. Microbial degradation is effective and environment-friendly for PAH removal, while the phenanthrene-degrading mechanism in Gram-positive bacteria is unclear. In this work, one Gram-positive strain of plant growth-promoting rhizobacteria (PGPR), Pseudarthrobacter sp. L1SW, was isolated and identified with high phenanthrene-degrading efficiency and great stress tolerance. It degraded 96.3% of 500 mg/L phenanthrene in 72 h and kept stable degradation performance with heavy metals (65 mg/L of Zn2+, 5.56 mg/L of Ni2+, and 5.20 mg/L of Cr3+) and surfactant (10 CMC of Tween 80). Strain L1SW degraded phenanthrene mainly through phthalic acid pathway, generating intermediate metabolites including cis-3,4-dihydrophenanthrene-3,4-diol, 1-hydroxy-2-naphthoic acid, and phthalic acid. A novel metabolite (m/z 419.0939) was successfully separated and identified as an end-product of phenanthrene, suggesting a unique metabolic pathway. With the whole genome sequence alignment and comparative genomic analysis, 19 putative genes associated with phenanthrene metabolism in strain L1SW were identified to be distributed in three gene clusters and induced by phenanthrene and its metabolites. These findings advance the phenanthrene-degrading study in Gram-positive bacteria and promote the practical use of PGPR strains in the bioremediation of PAH-contaminated environments.

Abstract:Petroleum hydrocarbons, including both aliphatic (gasoline, mineral oil) and aromatic compounds (BTEX), are known for their harmful effects on ecosystems and human health. Despite many studies, large-scale treatment of contaminated soils continues to be challenging, especially at lower temperatures. The use of metabolically-versatile, psychrotolerant, cold-active microorganisms, seems a promising, cost-effective and eco-friendly solution to boost remediation rates. In this study, a suitable microbial consortium was prepared and tested both in lab- and pilot-scale. To achieve the best bioremediation results, bacterial strains were isolated from BTEX-contaminated soil and then tested for the desired traits over a wide range of conditions. Of 5 preselected strains, 3 Pseudomonas strains capable of denitrification and aerobic/anaerobic degradation of hydrocarbons (up to 41.53±7.39 %), further characterized by a broad temperature (4–37 °C), pH (3–4 to 11) and salinity (0–8 %) tolerance, as well as resistance to freezing, were selected. Physiological studies were supported by genetic analyses, which indicated the presence of both alkB and xylM genes, and excluded similarity of the strains to the known opportunistic pathogens. To further confirm the applicability of the consortium, lab-scale analyses were followed by comprehensive pilot-scale tests on ~5 m 3 biopile/biocell, at different conditions. The results revealed increased efficacy of the consortium in bioremediation, when compared to biostimulated indigenous strains, for volatile hydrocarbons (93 % vs 88 %) and mineral oil (23 % vs 15 %), as well as 175 % and 136 % acceleration of remediation for the respective compounds in terms of time needed to complete the process. Moreover, the high survivability and metabolic activity of the consortium at different temperatures indicate the possibility of its year-round use for bioremediation of soil contaminated with petroleum hydrocarbons. The study proves the potential of specialized bacteria in the removal of pollutants, and emphasizes the role of bio-based strategies in addressing complex environmental challenges and remediation of contaminated sites.

Enhancing removal performance of ortho xylene by adding polydimethylsiloxane into two-stage biofilter

Abstract:A two-stage biofilter was built, augmented with polydimethylsiloxane (PDMS), to enhance the degradation of ortho xylene (o-xylene), and evaluate the feasibility of different PDMS concentrations for improving the removal effect. The results showed that PDMS concentration of 0.50 % significantly enhanced the purification efficiency and mineralization rate of o-xylene to 85(±1)% and 81 %, respectively. Simultaneously, the surface tension of the circulating liquid was reduced by 31.91 mN/m. Furthermore, the polysaccharide concentration of biofilters were increased by 6.90 mg/g and 7.38 mg/g, respectively, while the protein concentration was enhanced by 7.98 mg/g and 9.29 mg/g, respectively. It is worth noting that Sphingomonas and Sphingobium emerged as the dominant bacterial genera after intensification. Fusarium and Cladosporium became the predominant fungal genera in BTF1 and BTF2, respectively. Therefore, the two-stage biofilter containing bacteria and fungi combined with the addition of PDMS can effectively improve the degradation effect.
Determining the degradation mechanism and application potential of benzopyrene-degrading bacterium Acinetobacter XS-4 by screening

Abstract:In industrial wastewater treatment, organic pollutants are usually removed by in-situ microorganisms and exogenous bactericides. Benzo [a] pyrene (BaP) is a typical persistent organic pollutant and difficult to be removed. In this study, a new strain of BaP degrading bacteria Acinetobacter XS-4 was obtained and the degradation rate was optimized by response surface method. The results showed that the degradation rate of BaP was 62.73% when pH=8, substrate concentration was 10 mg/L, temperature was 25 °C, inoculation amount was 15% and culture rate was 180 r/min. Its degradation rate was better than that of the reported degrading bacteria. XS-4 is active in the degradation of BaP. BaP is degraded into phenanthrene by 3, 4-dioxygenase (α subunit and β subunit) in pathway I and rapidly forms aldehydes, esters and alkanes. The pathway II is realized by the action of salicylic acid hydroxylase. When sodium alginate and polyvinyl alcohol were added to the actual coking wastewater to immobilize XS-4 , the degradation rate of BaP was 72.68% after 7 days, and the removal effect was better than that of single BaP wastewater (62.36%), which has the application potential. This study provides theoretical and technical support for microbial degradation of BaP in industrial wastewater.

Abstract:In this study, performance evaluation for the gas-phase o-xylene removal using a xylene-acclimated biotrickling filter (BTF) was conducted. Substrate interactions during aerobic biodegradation of three poorly soluble compounds, both individually and in paired mixtures (namely, o-xylene and ethyl acetate, o-xylene and dichloromethane, which are common solvents used by pharmaceutical industry), were also investigated. Experimental results indicate that a maximum elimination capacity of 99.3 g·m−3·h−1 (70% removal) was obtained at an o-xylene loading rate of 143.0 g·m−3·h−1, while the top packing layer (one-third height of the three packing layers) only contributed about 13% to the total elimination capacity. Kinetic constants for o-xylene biodegradation and the pattern of o-xylene removal performance along the height of the BTF were obtained through the modified Michaelis–Menten kinetics and convection–diffusion reaction model, respectively. A reduction of removal efficiency in o-xylene (83.2–74.5% removal at a loading rate of 40.3 g·m−3·h−1 for the total volatile organic compound (VOC) loading rate of 79 g·m−3·h−1) in the presence of ethyl acetate (100% removal) was observed, while enhanced o-xylene removal efficiency (71.6–78.6% removal at a loading rate of 45.1 g·m−3·h−1 for the total VOC loading rate of 90 g·m−3·h−1) was achieved in the presence of dichloromethane (35.6% removal). This work shows that a BTF with xylene-acclimated microbial consortia has the ability to remove several poorly soluble compounds, which would advance the knowledge on the treatment of pharmaceutical VOC emissions.

Abstract:A fluoranthene-degrading bacterial strain FA1 was isolated from activated sludge and identified as Herbaspirillum chlorophenolicum, a newfound bacterial species that can grow well on fluoranthene as sole carbon and energy source. The kinetic characteristic of strain FA1 was tested in the aqueous model system (AMS) and the effects of nonionic surfactants on fluoranthene biodegradation in the AMS were then investigated. Tween 80 exhibited the best solubilization capacity for fluoranthene among three surfactants and its bioavailability decreased with an increase in its concentration and its degradation kinetics fit well with the first-order of power index model. The biotransformation of fluoranthene was greatly improved by Tween 80, and 58.5% fluoranthene degradation was obtained as Tween 80 was 100 mg/l. However, the bioavailability of fluoranthene decreased gradually with the increase of Tween 80 concentration. Bioremediation tests for fluoranthene in soil-water system were designed further to examine the degrading ability of strain FA1 with the presence of indigenous flora or not. The measurements showed that in the presence of indigenous flora, the optimum 30-day fluoranthene degradation in soil-water system reached 77.4%. Evidently, strain FA1 seems both efficient and high-effective and deserves further exploration on the enhanced bioremediation technologies for the treatment of fluoranthene-polluted soil.

Abstract:This study investigates the use of electrically conductive granules as packing material in biotrickling filter (BTF) systems as to provide insights on the specific microbial abundance and functions during the treatment of xylene-containing waste gas. In addition, the effect of applied potential on attached biofilm on conductive granules during xylene degradation was briefly investigated. During stable operation period, the conductive granules packed BTF achieved reactor performance of no less than 80% with a maximum EC of 137.7 g/m3 h. Under applied potential of 1V, the BTF system showed deterioration of xylene removal by ranging from 21 to 76%, which also affected the distribution and relative abundance of the major microorganisms such as Xanthobacter, Acidovorax, Rhodococcus, Hydrogenophaga, Arthrobacter, Brevundimonas, Pseudoxanthomonas, Devosia, Shinella, Sphingobium, Dokdonella, Pseudomonas and Bosea. The acclimation of applied potential led to the enrichment of autotrophic bacteria and strains, which are correlated to improved nitrogen cycling. In general, applying electrical potential is feasible to shape the microbiological structure of biofilms to selectively adjust their biochemical functions.

Abstract:The exchange of tetrahydrofuran (THF)-containing wastewater should significantly affect the performance of an activated sludge system. In this study, the feasibility of using THF-degrading Rhodococcus sp. strain YYL to bioaugment an activated sludge system treating THF wastewater was explored. As indicated by a DGGE analysis, strain YYL alone could not dominate the system, with the concentration of mixed liquor suspended solids (MLSS) decreasing to nearly half of the initial concentration after 45 d, and the microbial diversity was found to be significantly reduced. However, after the reactor was augmented with the mixed culture of strain YYL and two bacilli initially coexisting in the enriched isolation source, strain YYL quickly became dominant in the system and was incorporated into the activated sludge. The concentration of MLSS increased from 2.1g/L to 7.3g/L in 20 d, and the efficiency of THF removal from the system was remarkably improved. After the successful bioaugmentation, more than 95% of THF was completely removed from the wastewater when 20mM THF was continuously loaded into the system. In conclusion, our research first demonstrates that bioaugmentation of activated sludge system for THF degradation is feasible but that successful bioaugmentation should utilize a THF-degrading mixed culture as the inoculum, in which the two bacilli might help strain YYL colonize in activated sludge by co-aggregation.
Analysis of a simple biodegradation process for the removal of volatile organic chemicals from wastewater based on a gas stripping principle

Abstract:A simple biodegradation system consisting of an air stripping tank and a bioreactor was proposed for the treatment of volatile organic chemicals in wastewater. Toluene was used as a model of volatile organic chemicals. An aqueous solution of toluene and a basic mineral medium were placed in the air stripping tank and bioreactor, respectively. Toluene was stripped by supplying compressed air into the stripping tank through a sparger, and the stripped toluene was degraded by Pseudomonas putida mt-2 (ATCC 33015) in the bioreactor under aerobic conditions. The effect of the air stripping rate on bacterial growth was examined. A quantitative relationship was found between the air flow rate in the air stripping tank (Qa) and the stripping rate constant. During cultivation, the bacterial cells grew by utilizing toluene as the sole carbon source, and reached their maximum cell concentration (Xm) at the stationary phase. Xm showed a gradual decrease with increase in Qa from 1.8 to 7.2l/h, indicating a decrease in the rate of toluene degradation with increasing Qa. The Xm at Qa=1.8 l/h was the highest among the experiments under different values of Qa, which was almost twice that at Qa=7.2 l/h. Mathematical analysis taking the growth kinetics and mass transfer of toluene into consideration satisfactorily explained the system performance.

Abstract:To identify and obtain the indigenous degraders metabolizing phenanthrene (PHE) and biphenyl (BP) from the complex microbial community within industrial wastewater, DNA-based stable-isotope probing (DNA-SIP) and cultivation-based methods were applied in the present study. DNA-SIP results showed that two bacterial taxa (Vogesella and Alicyclobacillus) were considered the key biodegraders responsible for PHE biodegradation only, whereas Bacillus and Cupriavidus were involved in BP degradation. Vogesella and Alicyclobacillus have not been linked with PHE degradation previously. Additionally, DNA-SIP helped reveal the taxonomic identity of Ralstonia-like degraders involved in both PHE and BP degradation. To target the separation of functional Ralstonia-like degraders from the wastewater, we modified the traditional cultivation medium and culture conditions. Finally, an indigenous PHE- and BP-degrading strain, Ralstonia pickettii M1, was isolated via a cultivation-dependent method, and its role in PHE and BP degradation was confirmed by enrichment of the 16S rRNA gene and distinctive dioxygenase genes in the DNA-SIP experiment. Our study has successfully established a program for the application of DNA-SIP in the isolation of the active functional degraders from an environment. It also deepens our insight into the diversity of indigenous PHE- and BP-degrading communities.IMPORTANCE The comprehensive treatment of wastewater in industrial parks suffers from the presence of multiple persistent organic pollutants (POPs), such as polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs), which reduce the activity of activated sludge and are difficult to eliminate. Characterizing and applying active bacterial degraders metabolizing multiple POPs therefore helps to reveal the mechanisms of synergistic metabolism and to improve wastewater treatment efficiency in industrial parks. To date, SIP studies have successfully investigated the biodegradation of PAHs or PCBs in real-world habitats. DNA-SIP facilitates the isolation of target microorganisms that pose environmental concerns. Here, an indigenous phenanthrene (PHE)- and biphenyl (BP)-degrading strain in wastewater, Ralstonia pickettii M1, was isolated via a cultivation-dependent method, and its role in PHE and BP degradation was confirmed by DNA-SIP. Our study provides a routine protocol for the application of DNA-SIP in the isolation of the active functional degraders from an environment.
Isolation and Identification of DBP-Degrading Strain XJ1 and Its Degradation Characters

Abstract:Objective:A di-n-butyl phthalate-degrading bacterium was isolated from Xiangjiang River sediment,then the bacterium was identified and its degrading capability was studied.Method:Utilizing DBP as the sole carbon and energy source,a predominated bacterium,designated as strain XJ1,was isolated by enrichment culture and streak plate method.The strain XJ1 was identified based on 16S rDNA sequence analysis,(G+C) mol % and its morphological and physiological characteristics.The biodegradation experiments of DBP were performed in shake flasks and its DBP degradation capability was measured by high performance liquid chromatography,diversity of degradable substrates was also tested.Result:Strain XJ1 was identified as Sphingomonas sp.The optimum conditions for DBP degradation by strain XJ1 were initial pH 7.0,35 ℃ and agitation rate being 150 r/min.In 40h,DBP was degraded completely under the optimum conditions,the DBP metabolites detected by HPLC-UV were mono-butyl phthalate(MBP) and phthalate acid(PA).Diversity of degradable substrates showed that phthalate esters such as dimethyl phthalate,diethyl phthalate,etc.could be used by stain XJ1.Conclusion:DBP could be degraded effectively by strain XJ1,and the strain XJ1 had special application potential on bioremediation for phthalate esters contaminated site.
