Abstract:Silk has a long history of use in medicine as sutures. To address the requirements of a mechanically robust and biocompatible material, basic research to clarify the role of repeated sequences in silk fibroin in its structures and properties seems important as well as the development of a processing technique suitable for the preparation of fibers with excellent mechanical properties. In this study, three silk-like protein analogs were constructed from two regions selected from among the crystalline region of Bombyx mori silk fibroin, (GAGSGA)(2), the crystalline region of Samia cynthia ricini silk fibroin, (Ala)(12,) the crystalline region of spider dragline silk fibroin, (Ala)(6), and the Gly-rich region of spider silk fibroin, (GGA)(4). The silk-like protein analog constructed from the crystalline regions of the spider dragline silk and B. mori silk fibroins, (A(6)SCS)(8), that constructed from the crystalline regions of the S. c.ricini and B. mori silk fibroins, (A(12)SGS)(4), that constructed from and the crystalline region of S. c.ricini silk fibroin and the glycine-rich region of spider dragline silk fibroin, (A(12)SGS)(4),were expressed their molecular weights being about 36.0 kDa, 17.0 kDa and 17.5 kDa, respectively in E. coli by means of genetic engineering technologies. (A(12)-SCS)(4) and (A(12)SGS)(4) undergo a structural transition from alpha-helix to beta-sheet on a change in the solvent treatment from trifluoroacetic acid (TFA) to formic acid (FA). However, (A(6)SCS)(4) takes on the P-sheet structure predominantly on TFA treatment and FA treatment. Structural analysis was performed on model peptides selected from spider dragline and S. c.ricini silks by means of C-13 CP/MAS NAIR.

Abstract:In order to develop new silk-like materials in the form of fiber and non-woven nano-fiber, this study synthesized a new silk-like protein by selecting the sequence from the crystalline region of Bombyx mori silk fibroin, (GAGSGA)(6), to imitate the processing condition of the silk fibroin with the combination of the sequence YGGLGSQGAGRG, the hydrophilic motif of spider dragline silk which was considered as the origin of supercontraction of spider dragline silk (Yang Z, et al. J Am Chem Soc 2000;122: 9019-25). The CD pattern of the silk-like protein in hexafluoroacetone (HFA) indicates that it takes helical structure. The solid-state structures of the silk-like protein before and after methanol treatment were determined to be random coil and Silk II structure (mainly beta-sheet structure), respectively, using C-13 CP/MAS NMR. The C-13 CP/MAS NMR spectra of the protein after methanol treatment in hydrated state showed that the random coil peaks from Ala C beta and Set C beta carbons become sharper compared with the corresponding peaks in the dry state. The 2D-WISE spectra in the hydrated state also showed increase of the narrow components for these carbons. Thus, water molecules are relatively easy to access at the random coil regions of the protein. The fiber formation of the silk-like protein was possible with wet-spinning or electrospinning methods using HFA as dope solvent and methanol as coagulation solvent. (C) 2008 Elsevier Ltd. Abstract:The C-13 CP/MAS NMR, X-ray diffraction, and Raman spectroscopies were used for monitoring the structural transition of Samia cynthia ricini (S. c. ricini) silk fibroin induced by stretching. Here the silk fibroin was obtained from the aqueous solution stored in the silk gland. All of these spectroscopic data indicate that the structural transition from a-helix to P-sheet occurs with increasing the stretching ratio, especially between the stretching ratios, x 4 and x 6. The C-13 chemical shifts of Ala C peak in the C-13 CP/MAS NMR spectrum change significantly depending on alpha-helix, random coil, and two kinds of beta-sheet structure, which make it possible to clarify the local structure and structural transition. Actually, the fraction of individual structure in the silk fibroin samples was determined from decomposition of the Ala Cbeta peak by assuming a Gaussian line shape. To examine the conformational change of Ala residues of the polyalanine region in S. c. ricini silk fibroin observed with these spectroscopic methods, MD simulations for four peptide molecules, AGGAGG(A)(12)GGAGAG, with alpha-helix conformation were performed in the presence of water molecules under different tensile strengths, and then additional MM calculations were performed after removal of water molecules. The change in the conformational character of Ala residues induced by stretching is explicable by these calculations.

Abstract:Silkworms have been developed over thousands years to optimize folding and crystallization of fibroin under highly controlled conditions which have resulted in their efficient fiber formation. In this paper, we reconstructed the three-dimensional architecture of the spinneret of a wild silkworm Samia cynthia ricini from approximately 1000 optical micrographs of the semi-thin cross sections. The chitin plates and muscles were observed in the silk press part together with large change in the diameter of the spinneret lumen at the press part by large shear stress. This is similar to the case of the spinneret of Bombyx mori silkworm, indicating that the structural change in the silk fibroin of S.c. ricini silkworm occurs exclusively at the silk press part due to large shear stresses. Molecular dynamics (MD) calculations were then performed to study the structural change that occurs in the crystalline region of S.c. ricini silk fibroin under shear stress. Namely, using the peptide AGGAGG(A)12GGAGAG as a model of the crystalline part of the silk fibroin under different shear stresses in the presence of water molecules and followed by molecular mechanics (MM) calculation after removal of water molecules. The simulation indicates that the Ala residues in the model peptides adopt a predominantly β-sheet structure under shear stresses of above 1.0GPa.
Structure of Model Peptides Based on Nephila Clavipes Dragline Silk Spidroin (masp1) Studied by 13C Cross Polarization/Magic Angle Spinning NMR

Abstract:To obtain detailed structural information for spider dragline spidroin (MaSp1), we prepared three versions of the consensus peptide GGLGGQGAGAAAAAAGGAGQGGYGGLGSQGAGR labeled with 13C at six different sites. The 13C CP/MAS NMR spectra were observed after treating the peptides with different reagents known to alter silk protein conformations. The conformation-dependent 13C NMR chemical shifts and peak deconvolution were used to determine the local structure and the fractional compositions of the conformations, respectively. After trifluoroacetic acid (solvent)/diethyl ether (coagulant) treatment, the N-terminal region of poly-Ala (PLA) sequence, Ala8 and Ala10, adopted predominantly the alpha-helix with a substantial amount of beta-sheet. The central region, Ala15, Ala18, and Leu26, and C-terminal region, Ala31, of the peptide were dominated by either 3(1)-helix or alpha-helix. There was no indication of beta-sheet, although peak broadening indicates that the torsion angle distribution is relatively large. After 9 M LiBr/dialysis treatment, three kinds of conformation, beta-sheet, random coil, and 3(1)-helix, appeared, in almost equal amounts of beta-sheet and random coil conformations for Ala8 and Ala10 residues and distorted 3(1)-helix at the central region of the peptide. In contrast, after formic acid/methanol and 8 M urea/acetonitrile treatments, all of the local structure tends to beta-sheet, although small amounts of random coil are also observed. The peak pattern of the Ala Cbeta carbon after 8 M urea/acetonitrile treatment is similar to the corresponding patterns of silk fiber from Bombyx mori and Samia cynthia ricini. We also synthesized a longer 13C-labeled peptide containing two PLA blocks and three Gly-rich blocks. After 8 M urea/acetonitrile treatment, the conformation pattern was closely similar to that of the shorter peptide.

Abstract:To avoid potential ambiguity in the structural determination of native spider silk, we prepared both a nonlabeled peptide with a sequence containing both the polyalanine and the repeated GGA regions, QGAG(A)(6)GGAGA(GGA)(3)GAGRGGLGG (I), and the C-13-labeled peptides QGAGAAA[1-C-13]A(8)AAGG[2-C-13]A(13)GAGGAG[2-C-13]G(20)[3-C-13]A(21)GGAGAGRGGLGG (Ia) and QGAGAAAAAAGGAGAG-GAG [1-C-13]G(20)[1-C-13]A(21)GGAGAGRGGLGG (II) as a local structural model of spidroin 1 (MaSpl) protein in spider dragline silk of Nephila clavipes. Solvent treatments prior to the NMR measurements induce a structural change of these model peptides and provide a model to reproduce the structure of the silk fiber. Conformation-dependent C-13 NMR chemical shifts were mainly used to determine the local structure, including the evaluation of the fraction of several conformations. The characteristic structure, 65% beta-sheet for the Ala(8) residue in the poly-Ala region, and 70% 3(1)-helix for the Ala(21) residue and mainly 3(1)-helix for the Gly(20) residue in the GG(20)A(21) sequence was observed after peptides Ia and II were dissolved in 9 M LiBr followed by dialysis against water. The 2D spin diffusion 13C solid-state NMR spectrum of the Ala(21) residue of peptide II after this treatment was also reproduced by 70% 3(1)-helix (phi, rho = -90 degrees, 120 degrees) and 30% beta-sheet (phi, rho = -150 degrees, 150 degrees) structure. However, the Ala C-beta peak assigned to the 31-helix in the spectrum of Ia is broad, implying that the torsion angles of the Ala(21) residue are distributed, but with an average that corresponds approximately to the torsion angles of the 31-helix. An increase in the fraction of beta-sheet in both the poly-Ala and GG(20)A(21) regions was observed for la after it was dissolved in formic acid and then dried in air. Moreover, after Ia was dissolved in formic acid and then precipitated in methanol, the spectrum showed a tightly packed beta-sheet structure with a further increase in the fraction of beta-sheet although 15% 3(1)-helix still remained in the GG(20)A(21) region. The beta-sheet structure of the poly-Ala region and both 3(1)-helix and beta-sheet structures in the repeated GGA sequence are in agreement with the structural model for the native spider dragline silk fiber from N. clavipes from a previous NMR study (van Beek, J. D.; et al. Proc. Natl. Acad. Sci. U.S.A. 2002, 99, 10266-10271). On the other hand, the a-helical conformation was found to be dominant for the peptide treated with trifluoroacetic acid together with a significant contribution from other structures. The fraction of the other structures was 20-40% depending on the position of the C-13-labeled Ala residue in the chain.

Abstract:Fibrous protein secondary structural transitions are affected by bound water. A family of specially designed recombinant spider silk-like block copolymers with a gradient of block length, hydrophobilicity, and degree of crystallinity was biosynthesized and characterized to demonstrate the effect of water on the protein structural transitions. These proteins were inspired by the genetic sequences found in the dragline silk of Nephila clavipes, comprising an alanine-rich hydrophobic block, A, a glycine-rich hydrophilic block, B, and a C-terminus or a His-tag, H. Because the A-block is hydrophobic and the B-block is hydrophilic, the spider silk-like block copolymers behave as amphiphilic molecules and self-assemble into various structures in water solution. We employ time-resolved Fourier transform infrared (FTIR) spectroscopy to assign the origin of specific secondary structural transitions during heating. A transition from random coils to beta-turns dominates during a lower temperature glass transition (of plasticized protein) mediated by the removal of bound water. Once the protein is in the dry solid state, further heating causes the now-dry protein to undergo the glass transition to the liquid state through conversion of alpha-helices into beta-turns. The structural transitions during protein glass transitions are intrinsic to the amorphous region of protein and are hardly affected by protein hydrophobicity, block length, or crystallinity. The self-assembly morphology of the spider silk-like block copolymers, investigated by scanning electron microscopy, indicates that the large-scale morphology is stable during heating through both the lower and upper temperature glass transitions.

Abstract:We have developed a methodology using advanced thermal analysis to characterize the role of water in a specially synthesized family of recombinant spider silk-like block copolymers. These proteins were inspired by the genetic sequences found in the dragline silk of Nephila clavipes, comprising an alanine-rich hydrophobic block, A; a glycine-rich hydrophilic block, B; and a C-terminus or a His-tag, H. This family of proteins serves as a model system in which the hydrophobicity is controlled by A and B block lengths, allowing systematic comparison of water effects within the family. Temperature-modulated differential scanning calorimetry and thermogravimetric analyses were employed to capture the glass to rubber transition, T-g, in water-cast protein films. Modeling of the solid and liquid state heat capacity baselines allows us to determine the critical role played by bound water which plasticizes and stabilizes the protein through interchain bonding. In samples containing bound water, two sequential glass transitions, T-g(1) and T-g(2), were observed during heating. The lower temperature glass transition, T-g(1), is related to conformational change induced by bound water removal, the hydrophobicity of the protein sequences, and the crystallinity of the protein. The higher temperature glass transition, T-g(2), is characteristic of the dry protein. The binding energy of water to protein compares favorably to ligandwater binding affinities. The energy absorbed by evaporating water depends upon the volume fraction of the hydrophilic B-block.
[GFAAS determination of trace lead in toothpaste using L'vov platform].

Abstract:Sensitivity in determining low melting point metals is highly improved by means of platform technique in graphite furnace AAS. In this paper we describe a method of determining trace lead in toothpaste by this technique. The method of making the platform and the selection of the platform coating were discussed and the results with and without the platform were compared. The optimum ashing temperature, atomization temperature and experimental conditions were investigated. The interference of large amount Al and Ca in the matrix on determination of trace lead can be minimized by using platform technique. The precision of about 0.9% and detection limit of about 15 ng obtained by this improved method are much better than those by methods without platform.
Structural Changes in Spider Dragline Silk after Repeated Supercontraction-Stretching Processes

Abstract:Spider dragline silk is well-known for its excellent combination of strength and extensibility as well as another unique property called supercontraction. In our previous work, the changes in conformations of the Nephila edulis spider dragline silk when subjected to different supercontraction processes were extensively investigated. When a native spider dragline silk had free supercontraction, and then restretched to its original length, the content and molecular orientation of different conformations (β-sheet, helix, and random coil) changed but the mechanical properties remained almost the same. Therefore, herein, further supercontraction-stretching treatment was performed up to three cycles, and the corresponding structural changes were investigated. In addition to the synchrotron radiation FTIR (S-FTIR) microspectroscopy employed in our previous study, synchrotron radiation small-angle X-ray scattering (S-SAXS) and atomic force microscopy (AFM) were also used in this work to determine the structural changes of spider dragline silk in different scales. The results show that by repeating the supercontraction-stretching treatment, the β-sheet structure content in spider dragline silk was slightly increased, but its orientation degree remained almost the same. Also, with the increase in cycle of supercontraction-stretching treatments, a 10.5 nm long period perpendicular to the silk fiber axis gradually appeared, endowing the spider dragline silk with periodic structure both along (6.6 nm, already existed in native silk and did not change with the supercontraction-stretching treatment) and perpendicular to the silk fiber axis. After the third supercontraction-stretching cycle, the AFM images displayed a clear 210 nm × 80 nm corn kernel-like structure on the surface of nanofibrils in spider dragline silks, which may be related to the aggregation of 10.5 nm × 6.6 nm periodic structure observed via S-SAXS. Finally, although the structure of spider dragline silk became increasingly regular with the rise in supercontraction-stretching cycles, mechanical properties remained constant after every cycle of the supercontraction-stretching treatment. These findings can aid in further understanding the structural changes that are related to the supercontraction of spider dragline silk and provide useful guidance in fabrication of high-performance regenerated or artificial silk fibers.

Relationships Between Supercontraction and Mechanical Properties of Spider Silk

Abstract:Typical spider dragline silk tends to outperform other natural fibres and most man-made filaments 1 . However, even small changes in spinning conditions can have large effects on the mechanical properties of a silk fibre 2 , 3 , 4 , 5 , 6 as well as on its water uptake. Absorbed water leads to significant shrinkage in an unrestrained dragline fibre 7 , 8 and reversibly converts the material into a rubber 9 . This process is known as supercontraction 10 and may be a functional adaptation for the silk’s role in the spider’s web 11 . Supercontraction is thought to be controlled by specific motifs in the silk proteins 12 , 13 and to be induced by the entropy-driven recoiling of molecular chains 9 , 14 . In analogy, in man-made fibres thermal shrinkage induces changes in mechanical properties 15 , 16 , 17 attributable to the entropy-driven disorientation of ‘unfrozen’ molecular chains (as in polyethylene terephthalate) 15 , 18 or the ‘broken’ intermolecular hydrogen bonds (as in nylons) 17 . Here we show for Nephila major-ampullate silk how in a biological fibre the spinning conditions affect the interplay between shrinkage and mechanical characteristics. This interaction reveals design principles linking the exceptional properties of silk to its molecular orientation.

Abstract:This manuscript reviews work from our laboratory that addresses the orientation, secondary structure, wet-spinning, and molecular basis for supercontraction of spider silk. It identifies the poly(alanine) runs as the crystalline regions, establishes the degree of orientation of these regions, and identifies the secondary structural elements of the conserved L-G-X-Q (X = G, S, or N) regions. It also describes methods for spinning very small amounts of protein polymers and it sets forth several molecular-level hypotheses concerning supercontraction.

The Effect of Solvents on the Contraction and Mechanical Properties of Spider Silk

Abstract:We examined the mechanical characteristics of four major ampullate (MA) dragline silks during and after submersion in a range of solvents. The silks were reeled from four very different spiders: Araneus diadematus, Nephila edulis, Latrodectus mactans and Euprosthenops sp. They displayed significant differences in behaviour in the native state as well as during and after supercontraction in solvents such as water, urea solution and a set of alcohols. The different polarities of the solvents are thought to affect different regions of the silk's molecular conformation. We hypothesise that the observed mechanical properties of dragline silks are those of a hard elastic polymer; and we explain the supercontraction of the silks as changes of orientation in the molecular chains.

Abstract:Spider silk is an archetypal biopolymer material with extreme tensile properties arising from its complex hierarchical assembly. While recent advances in sequencing have yielded abundant insights, relatively little is known concerning post-translational modifications (PTMs) in spider silk. Here, we probe the PTM landscape of dragline silk from the Jorō spider (Trichonephila clavata) using a combination of mass spectroscopy and solid-state nuclear magnetic resonance (NMR). The results reveal a wide array of potential modifications, including hydroxyproline, phosphorylation, and dityrosine cross-links, encompassing the different spidroin constituents. Notably, the MaSp3 repetitive region displayed numerous PTMs, whereas MaSp1 and MaSp2 variants showed distinct phosphorylation patterns in its terminal domains. The N-terminal domain (NTD) phosphorylation sites were found predominantly at the dimer interface, suggesting a modulatory function with respect to its pH-driven dimerization function, a hypothesis supported by studies using phosphomimetic NTD mutants. Possible roles of phosphoserine in limiting β-sheet formation, and hydroxyproline in disrupting β-turns are also discussed. Spider silk is an archetypal biomaterial that can outperform our most sophisticated artificial fibers. The secret to its mechanical properties lies in its complex hierarchical structure—encompassing the nano- to macroscales—that forms through a process of molecular self-assembly of the constituent spidroin proteins. While recent advances in "biomateriomics" have given us tremendous insights into the sequence–function relationships that determine spider silk behavior, the picture is still far from complete. One area that has received little attention is posttranslational modifications (PTMs). PTMs are ubiquitous biological phenomena that are crucial for providing dynamic control of the proteome, and effectively expand the structural and functional design space of proteins beyond that provided by the canonical amino acids. Here, we undertook a comprehensive analysis of PTMs from spider dragline silk fiber, which revealed numerous potential sites for a wide array of modifications. The results provide a fascinating window into additional layers of complexity underlying the mechanical behavior of spider silk, and suggest further avenues for creating novel, dynamically tunable, bioinspired materials.

Several amino acid residue types have been proposed to be involved in this process, but the complex composition of the native fiber complicates detailed investigations. Here, we observe supercontraction in biomimetically produced artificial spider silk fibers composed of defined proteins. We found experimental evidence that proline is not the sole residue responsible for supercontraction and that tyrosine residues in the amorphous regions of the silk fiber play an important role. Furthermore, we show that the response of artificial silk fibers to humidity can be tuned, which is important for the development of materials for applications in wet environments, eg producing water resistant fibers with maximal strain at break and toughness modulus.
Evidence of Decoupling Protein Structure from Spidroin Expression in Spider Dragline Silks

Abstract:The exceptional strength and extensibility of spider dragline silk have been thought to be facilitated by two spidroins, major ampullate spidroin 1 (MaSp1) and major ampullate spidroin 2 (MaSp2), under the assumption that protein secondary structures are coupled with the expressed spidroins. We tested this assumption for the dragline silk of three co-existing Australian spiders, Argiope keyserlingi, Latrodectus hasselti and Nephila plumipes. We found that silk amino acid compositions did not differ among spiders collected in May. We extended these analyses temporally and found the amino acid compositions of A. keyserlingi silks to differ when collected in May compared to November, while those of L. hasselti did not. To ascertain whether their secondary structures were decoupled from spidroin expression, we performed solid-state nuclear magnetic resonance spectroscopy (NMR) analysis on the silks of all spiders collected in May. We found the distribution of alanine toward β-sheet and 3,10helix/random coil conformations differed between species, as did their relative crystallinities, with A. keyserlingi having the greatest 3,10helix/random coil composition and N. plumipes the greatest crystallinity. The protein secondary structures correlated with the mechanical properties for each of the silks better than the amino acid compositions. Our findings suggested that a differential distribution of alanine during spinning could decouple secondary structures from spidroin expression ensuring that silks of desirable mechanical properties are consistently produced. Alternative explanations include the possibility that other spidroins were incorporated into some silks.

Abstract:Raman spectroscopy is used to elucidate the effect of spinning conditions upon the structure and mechanical properties of silk spun by Nephila spiders from the major ampullate gland. Silk fibers produced under natural spinning conditions with spinning rates between 2 and 20 mm s −1 differed in microstructure and mechanical properties from fibers produced either more slowly or more rapidly. The data support the “uniform strain” hypothesis that the reinforcing units in spider silk fibers are subjected to the same strain as the fiber, to optimize the toughness. In contrast, in the case of synthetic high-performance polymer fibers, the both units and the fiber experience uniform stress, which maximizes stiffness. The comparison of Nephila major and minor ampullate silks opens an intriguing window into dragline silk evolution and the first evidence of significant differences between the two silks providing possibilities for further testing of hypotheses concerning the uniform strain versus uniform stress models. Impact statement It is well established that the microstructure and mechanical properties of engineering materials are controlled by the conditions employed to both synthesize and process them. Herein, we demonstrate that the situation is similar for a natural material, namely spider silk. We show that for a spider that normally produces silk at a reeling speed of between 2 and 20 mm s −1 , silk produced at speeds outside this natural processing window has a different microstructure that leads to inferior tensile properties. Moreover, we also show that the silk has a generic microstructure that is optimized to respond mechanically to deformation such that the crystals in the fibers are deformed under conditions of uniform strain. This is different from high-performance synthetic polymer fibers where the microstructure is optimized such that crystals within the fibers are subjected to uniform stress. Graphic abstract

Abstract:Spider dragline silk is a remarkable material that shows excellent mechanical properties, diverse applications, biocompatibility and biodegradability. Transgenic silkworm technology was used to obtain four types of chimeric silkworm/spider (termed composite) silk fibres, including different lengths of recombinant Major ampullate Spidroin1 (re-MaSp1) or recombinant Major ampullate Spidroin2 (re-MaSp2) from the black widow spider, Latrodectus hesperus. The results showed that the overall mechanical properties of composite silk fibres improved as the re-MaSp1 chain length increased, and there were significant linear relationships between the mechanical properties and the re-MaSp1 chain length (p < 0.01). Additionally, a stronger tensile strength was observed for the composite silk fibres that included re-MaSp1, which only contained one type of repetitive motif, (GA)(n)/A(n), to provide tensile strength, compared with the silk fibres that includedre-MaSp2, which has the same protein chain length as re-MaSp1 but contains multiple types of repetitive motifs, GPGXX and (GA)(n)/A(n). Therefore, the results indicated that the nature of various repetitive motifs in the primary structure played an important role in imparting excellent mechanical properties to the protein-based silk fibres. A silk protein with a single type of repetitive motif and sufficiently long chains was determined to be an additional indispensable factor. Thus, this study forms a foundation for designing and optimizing the structure of re-silk protein using a heterologous expression system.
