Abstract:For the applications of polyelectrolyte complex (PEC) of chitosan and sodium cellulose sulfate (NaCS) as the controlled release drug carrier, the biodegradation characteristics of chitosan, NaCS and chitosan/NaCS PEC films were investigated with pepsin, trypsin, lipase, α-amylase, and cellulase. The results showed that pepsin, amylase, trypsin and lipase have the appreciable hydrolytic activity to chitosan, but cannot degrade NaCS. However, cellulase showed high cellulosic activity and low chitosanolytic activity. For the hydrolysis of chitosan/NaCS films, the degradation rates were greatly influenced by the molecular weights of chitosan and NaCS. In vitro tests showed that different formulations caused diverse disintegration time of the films through the gastrointestinal tract. The results indicated the PEC based on chitosan and NaCS showed good potential for the gastrointestinal delivery systems.

Abstract:Chitosan, the cationic polysaccharide derived from the natural polysaccharide chitin, has been studied as a biomaterial for more than two decades. As a polycationic polymer with favorable properties, it has been widely used to form polyelectrolyte complexes with polyanions for various applications in drug delivery fields. In recent years, a growing number of studies have been focused on the preparation of polyelectrolyte complexes based on chitosan and its water soluble derivatives. They have been considered well-suited as biomaterials for a number of vital drug carriers with targeted/controlled release profiles, e.g., films, capsules, microcapsules. In this work, an overview highlights not only the favorable properties of chitosan and its water soluble derivatives but also the good performance of the polyelectrolyte complexes produced based on chitosan. Their various types of applications as drug carriers are reviewed in detail.

Abstract:In this study, we describe the elaboration of multifunctional positively charged polyelectrolyte complex (PEC) nanoparticles, designed to be stable at physiological salt concentration and pH, for effective targeted delivery. These nanoparticles were obtained by charge neutralization between chitosan (CS) as polycation and hyaluronic acid (HA) as polyanion. We showed that the course of the complexation process and the physico-chemical properties of the resulting colloids were impacted by (i) internal parameters such as the Degree of Acetylation (DA, i.e., the molar ration of acetyl glucosamine residues) and molar mass of CS, the HA molar mass and (ii) external parameters like the charge mixing ratio and the polymer concentrations. As a result, nonstoichiometric colloidal PECs were obtained in water or PBS (pH 7.4) and remained stable over one month. The polymer interactions were characterized by thermal analysis (DSC and TGA) and the morphology was studied by scanning electron microscopy. A model antibody, anti-ovalbumine (OVA) immunoglobulin A (IgA) was sorbed on the particle surface in water and PBS quantitatively in 4 h. The CS-HA/IgA nanoparticles average size was between 425-665 nm with a positive zeta potential. These results pointed out that CS-HA can be effective carriers for use in targeted drug delivery.

Abstract:Polyelectrolyte complexes (PECs) were elaborated from chitosan as cationic polymer and carboxy-methylpullulan (CMP), hyaluronic acid (HA) and their derivatives grafted with aminoguaiacol (G) with different degrees of substitution (DSGA) with the aim of obtaining nanogels for drug delivery. For each couple of polysaccharides, the charge ratios giving the smaller size with the lower PDI were selected to produce PECs. CMP_CHIT and CMP-G_CHIT PECs had smaller sizes (220-280 nm) than HA_CHIT and HA-G_CHIT PECs (280-390 nm). PECs were stable at 4 °C during 28 days at pH 5. In phosphate buffer saline (PBS) at pH 7.4, at 4 °C, a better stability of PECs based on CMP-G derivatives was observed. The hydrophobic associations between aminoguaiacol groups (highlighted by measurements of pyrene fluorescence) led to a better PECs' stabilization in PBS. The PECs' antioxidant and antibacterial activities were demonstrated and related to the DSGA. Diclofenac and curcumin were used as drug models: their loading reached 260 and 53 μg/mg PEC, respectively. The release of diclofenac in PBS at 37 °C followed a quasi-Fickian diffusion mechanism with release constant between 0.88 and 1.04 h-1. The curcumin release followed a slow linear increase in PBS/EtOH (60/40 V/V) with an effect of DSGA.

Abstract:The work studied the formation and structure of (bio)PEC in mixtures of dilute chitosan and gelatin solutions at different pH and ratios of polymers and their ionized groups of z . Using the turbidimetry method for the chitosan and gelatin mixtures, it was found that stable scattering particles of (bio)PEC are formed in the pH range from 5 to 6.4 and with a polymer ratio above 1:5 ( z = 0.29). The electrostatic interaction in the (bio)PEC structures was confirmed by ATR-FTIR spectroscopy. It was shown that during the formation of (bio)PEC in mixtures with pH = 5.5, a strong shift of the corresponding absorption bands relative to the initial polymers is observed. Studying the complexation and structure of these particles using the dynamic light scattering method of mixtures at pH = 3, the associative-dissipative character of the polymers interaction with the formation of soluble and unstable (bio)PEC dispersions was determined. It was observed that in mixtures with a pH = 5.5, the interaction becomes associative, which is accompanied by the formation of a (bio)PEC coacervate microphase at polymer ratios from 1:10 to 1:30 (in the z range from 0.58 to 1.73, respectively). Analysis of the structural properties of the coacervate microphase of (bio)PEC using dynamic light scattering and optical microscopy methods showed that with an increase in the proportion of an oppositely charged component of z ≥ 1, the isotropy of the polyelectrolyte binding process increases, which leads to suppression of the formation of loose heterogeneous coacervate particles of (bio)PEC.

Abstract:Organo-soluble hexanoyl chitosan(HC) was synthesized through esterification reaction(between) chitosan and hexanoyl chloride in methylsulfonic acid solutions.This intermediate was further PEGylated through NHS-DCC coupling method to give PEG-g-HC.The chemical structure of HC and PEG-g-HC were characterized by FT-IR,~1H-NMR and UV-Vis.The self-assembly behavior of PEG-g-HC was (studied) by DLS,fluorometer and TEM.The degree of substitution(DS) of HC can be adjusted by varying the feed ratio of hexanoyl chloride to chitosan.The critical pH at which HC could be soluble in aqueous media increased with the increase of DS of HC.Spherical micelles with pH responsibility could be formed through self-assembling of PEG-g-HC in aqueous media.In addition,the critical pH where the(micelle) size abruptly decreased could also be modulated by varying the DS of HC.

Abstract:Polyelectrolyte complexes (PECs) are attractive soft materials with many promising applications, such as drug delivery and coatings. Study on the solution properties of α-helical polypeptide based PECs are still rare. Herein, new PECs composed of α-helical sulfonium-based polypeptides (PPBLG-DMS-I) and polysaccharides (i.e., carboxymethyldextran sodium salt, CMD) were prepared and investigated on their solution property. PPBLG-DMS-I were synthesized by an ultrafast ring-opening polymerization (ROP) and subsequent alkylation of methylthio groups with high grafting efficiency. The aggregation size of the polypeptide-polysaccharide based PECs was in the range of 450-600 nm at room temperature depending on the cationic/anionic weight ratios (RCAs) and polypeptide concentrations. It decreased by about 300 nm with increasing of the temperature by 40 °C. PEC solutions showed salt induced sharp phase transitions accompanied by the formation of coacervate droplets at low salt concentrations (e.g., 1.5 mg·mL-1). pH induced phase transitions of PEC solutions at both acidic (e.g., pH = 2.5) and basic conditions (e.g., pH = 11.5).

Abstract:Quaternized chitosan (QCh) was homogeneously synthesized by reacting chitosan with 3-chloro-2-hydroxypropyltrimethylammonium chloride (CHPTAC) in alkali/urea aqueous solution for the first time. The structure and solution properties of QCh were characterized by using element analysis, FT-IR, 13C NMR, SEC-LLS, rheology, viscometer, and ξ-potential measurements. Subsequently, polyelectrolyte complex (PEC) hydrogels were constructed by in situ polymerization of acrylic acid (AA) monomers in the concentrated QCh solution. The structure and mechanical behavior of the prepared hydrogels were systematic studied. Because of the high charge density and solubility of QCh, strong electrostatic interactions were formed in the hydrogels and endowed them tough with self-recovery properties. The mechanical behavior of the hydrogels was accurately tuned from stiff and viscoelastic to soft and elastic by changing the poly(acrylic acid) (PAA) content. The regulation mechanism relied on the remarkable difference in the chain segmental mobility between QCh and PAA. Moreover, the QCh/PAA PEC hydrogels displayed excellent solvent-induced shape-memory behavior due to the reversible properties of the ionic bonds. In summary, we offered a novel modification method for chitosan and opened up a new avenue to construct chitosan-based hydrogels with outstanding mechanical properties.

Abstract:An ampholytic N-carboxyethyl chitosan (CEC), with various isoelectric points (IPs), was synthesized by grafting acrylic acid on chitosan utilizing Michael's reaction. Compared to native chitosan, CEC has enhanced water solubility and dramatically accelerated enzymatic degradation; the rate of degradation is proportional to the degree of substitution (DS). The results from turbidimetric titration and fluorescence studies revealed that CEC formed complexes with either hyaluronic acid (HA) or bovine serum albumin (BSA) within a certain pH range. The HA/CEC/BSA ternary complexes could be prepared by colloid titration with quantitative yield and BSA entrapment. The rate of BSA release from the complexes was affected by pH, ionic strength, DS of CEC, and the molecular weight (MW) of HA. The endurance of BSA release from the complexes could be extended up to 20 d by formulating them with high-MW HA and CEC with low DS.BSA release profiles from HA/CEC-2/BSA complexes.
Implantation of allogenic synovial stem cells promotes meniscal regeneration in a rabbit meniscal defect model.

Abstract:BACKGROUND Indications for surgical meniscal repair are limited, and failure rates remain high. Thus, new ways to augment repair and stimulate meniscal regeneration are needed. Mesenchymal stem cells are multipotent cells present in mature individuals and accessible from peripheral connective tissue sites, including synovium. The purpose of this study was to quantitatively evaluate the effect of implantation of synovial tissue-derived mesenchymal stem cells on meniscal regeneration in a rabbit model of partial meniscectomy. METHODS Synovial mesenchymal stem cells were harvested from the knee of one New Zealand White rabbit, expanded in culture, and labeled with a fluorescent marker. A reproducible 1.5-mm cylindrical defect was created in the avascular portion of the anterior horn of the medial meniscus bilaterally in fifteen additional rabbits. Allogenic synovial mesenchymal stem cells suspended in phosphate-buffered saline solution were implanted into the right knees, and phosphate-buffered saline solution alone was placed in the left knees. Meniscal regeneration was evaluated histologically at four, twelve, and twenty-four weeks for (1) quantity and (2) quality (with use of an established three-component scoring system). A similar procedure was performed in four additional rabbits with use of green fluorescent protein-positive synovial mesenchymal stem cells for the purpose of tracking progeny following implantation. RESULTS The quantity of regenerated tissue in the group that had implantation of synovial mesenchymal stem cells was greater at all end points, reaching significance at four and twelve weeks (p < 0.05). Tissue quality scores were also superior in knees treated with mesenchymal stem cells compared with controls at all end points, achieving significance at twelve and twenty-four weeks (3.8 versus 2.8 at four weeks [p = 0.29], 5.7 versus 1.7 at twelve weeks [p = 0.008], and 6.0 versus 3.9 at twenty-four weeks [p = 0.021]). Implanted cells adhered to meniscal defects and were observed in the regenerated tissue, where they differentiated into type-I and II collagen-expressing cells, at up to twenty-four weeks. CONCLUSIONS Synovial mesenchymal stem cells adhere to sites of meniscal injury, differentiate into cells resembling meniscal fibrochondrocytes, and enhance both quality and quantity of meniscal regeneration.
Chitin nanofibers improve the stability and functional performance of Pickering emulsions formed from colloidal zein

Abstract:There is growing interest in formulating Pickering emulsions from biopolymer particles due to consumer demand for more natural products. Protein-based colloidal particles can be used for this purpose, but they are prone to aggregate at pH values around their isoelectric point (pI), which limits their application. In this study, the possibility of using chitin nanofibers (ChNFs) to improve the pH stability of Pickering emulsions prepared from zein colloidal particles (ZCPs) was investigated. Initially, the morphology and interfacial properties of the complexes formed between ChNFs and ZCPs were studied as a function of pH (3-9). The tendency of the ZCPs to aggregate and sediment at pH ≥ pI was reduced in the presence of ChNFs, which was attributed to the formation of electrostatic complexes. The contact angle of the composite particles could be optimized by altering their composition. For instance, the contact angle increased from 74° for ZCPs to 85° for ZCP/ChNF (5:1 ratio) at pH 6, which improved their tendency to stabilize the oil droplets. Brewster angle microscopy indicated that ZCP/ChNF complexes had rod-like and/or particulate structures at an air-water interface, which were different from those observed in the bulk aqueous phase. Pickering emulsions formed from ZCP/ChNF complexes had better stability than those formed from ZCPs or ChNFs, especially when the pH was close to or greater than the pI. An in vitro digestion study showed that the presence of the interfacial complexes reduced the lipolysis of the oil droplets by about 11% in a simulated gastrointestinal tract. High internal phase Pickering emulsions (HIPPEs) could be formed from ZCP/ChNF complexes at pH ≥ pI, which were able to protect unsaturated lipids from oxidation. Overall, our results show that chitin nanofibers can be used to improve the pH stability of Pickering emulsions formed from colloidal zein, as well as to modulate their functional performance.

Abstract:The use of polyelectrolyte complexes (PEC) in encapsulation process is common due to the electrostatic interactions between the two oppositely charged biopolymers. However, the incorporation of montmorillonite (Mt) particles in the complex formation for improving the encapsulation efficiency as well as the physicochemical properties of PEC remains a challenge due to the competitive polymer–polymer and polymer-nanoparticle interactions. This study aimed to investigate the binding mechanism of alginate and chitosan through layer-by-layer approach in the presence of montmorillonite nanoparticles as a function of pH and the addition order of the components. Various techniques were employed to examine the formation of the PEC on the surface of Mt, such as zeta potential, conductivity and turbidity measurements. The characterization of the different PEC systems was performed using Fourier Transformed Infrared spectroscopy (FTIR), X-Ray diffraction (XRD), scanning electron microscopy (SEM), differential scanning calorimetry (DSC) and thermogravimetry analysis (TGA). The results indicate that the formation of the complex is pH-dependent. The highest intercalation of the complex was observed when chitosan was added first at a lower pH, while the intercalation was reduced at neutral pH. Additionally, the thermal stability of PEC was enhanced in the presence of montmorillonite, and was dependent on both pH and the order of addition of the biopolymers. This study provides a new understanding of the complex formation mechanism between alginate and chitosan in the presence of montmorillonite nanoparticles, showing that the combination of biopolymer-based PEC with clay particles can potentially provide a low-cost, more stable and effective nanocomposite for encapsulation and delivery of active agents.

Stretchable hydrogels of chitosan/hyaluronic acid induced by polyelectrolyte complexation around neutral pH

Abstract:In this work, we propose the formation of stretchable hydrogels at neutral pH from the physical crosslinking of chitosan (CS) and hyaluronic acid (HA) by polyelectrolyte complexation. A mixture of CS (Mw ≈ 600 kg/mol, degree of acetylation ≈ 50 %) solution and HA (Mw ≈ 77 kg/mol) solution was prepared with an excess of salts screening the electrostatic interactions CS/HA. In a controlled manner, the polyelectrolyte complexation was induced through the progressive dialysis of the salted polymer mixture against a sodium acetate solution (AcONa, 0.01 M) for 7 days. Depending on [HA], various materials were obtained: viscous solutions at [HA] = 0.75 % (w/v); hydrogels at [HA] = 1.50-2.24 % (w/v) with Young modulus of 14 kPa and stretchable to 200 %. The small angle X-ray scattering characterization of the hydrogels revealed a multiscale organization related to the conformation of the polymers induced by the physical interactions. The dialysis process with AcONa was optimized by adding a dialysis step against a zinc acetate solution containing Zn2+. The combination of polyelectrolyte complexation between CS/HA and metal complexation between Zn2+ and the polymers led to an enhancement of the hydrogel stretchability up to 400 %.

Abstract:At present, the ocean as a treasure house of human development resources has attracted widespread attention. Chitin is the most abundant biomass resource in the ocean, and it is also a natural macromolecule second only to cellulose in nature. Its deacetylated product, chitosan, is not only easy to modify physically and chemically, but can be processed into various forms and has excellent properties. Biocompatibility, biodegradability, antibacterial, tissue adhesion and other biological activities. Derivatization reactions are often used to improve the properties of chitosan and give it new structures and functions. In this paper, a series of chitosan ether derivatives were synthesized using alkali/urea aqueous solution as the solvent and homogeneous etherification reaction medium of chitosan, and their structures and properties were characterized. Then, chitosan and quaternized chitosan are dissolved in the acrylic acid monomer aqueous solution, and the chitosan/polyacrylic acid and quaternized high-strength polyelectrolyte composite hydrogel (PEC) are obtained by in-situ polymerization. Finally, the above-mentioned composite hydrogel is treated with Ag+ to obtain the ultra-high strength of the dual interaction of static electricity and coordination bonds, and at the same time a physical cross-linked hydrogel (DPC) with anti-fatigue. The chitosan-based hydrogel with excellent mechanical properties has good application prospects in articular cartilage, wound dressings, bionic stents and the like.

Abstract:Taking into account the trends in the field of green chemistry and the desire to use natural materials in biomedical applications, (bio)polyelectrolyte complexes ((bio)PECs) based on a mixture of chitosan and gelatin seem to be relevant systems. Using the approach of self-assembly from the dispersion of the coacervate phase of a (bio)PEC at different ratios of ionized functional groups of chitosan and gelatin (z), hydrogels with increased resistance to mechanical deformations and resorption in liquid media were obtained in this work in comparison to a hydrogel from gelatin. It was found that at z ≥ 1 a four-fold increase in the elastic modulus of the hydrogel occurred in comparison to a hydrogel based on gelatin. It was shown that hydrogels at z ≈ 1 had an increased sorption capacity and water sorption rate, as well as increased resistance to the in vitro model environment of phosphate-buffered saline (PBS) solution containing lysozyme at 37 °C. It was also shown that in PBS and simulated gastric fluid (SGF) solutions, the effect of the polyelectrolyte swelling of the hydrogels was significantly suppressed; however, at z ≥ 1, the (bio)PEC hydrogels had increased stability compared to the samples at z < 1 and based on gelatin.

As a result, Pickering emulsion shows an promising outlook for its application of the light sensitive, low bioavailibility added-Value food components.

Computational and experimental approaches for chitosan-based nano PECs design: Insights on a deeper comprehension of nanostructure formation

Abstract:Nanostructured polyelectrolyte complexes (nano PECs) based on biopolymers are an important technological strategy to target drugs to the action and/or absorption site in a more effective way. In this work, computational studies were performed to predict the ionization, spatial arrangement and interaction energies of chitosan (CS), hyaluronic acid (HA), and hypromellose phthalate (HP), for the design of nano PEC carriers for methotrexate (MTX). The optimal pH range (5.0-5.5) for preparing nano PECs was selected by experimental and computational methodologies, favoring the polymers interactions. CS, HA, HP and MTX addition order was also rationalized, maximizing their interactions and MTX entrapment. Spherical nano-sized particles (256-575 nm, by dynamic light scattering measurement) with positive surface charge (+25.5 to +29.2 mV) were successfully prepared. The MTX association efficiency ranged from 20 to 32 %. XRD analyses evidenced the formation of a new material with an organized structure, in relation to raw polymers.

Abstract:Solution-processable polyelectrolyte complexes (PECs) between poly(diallyldimethylammonium chloride) (PDDA) and poly(acrylic acid) (PAA) were synthesized in aqueous NaOH and obtained in their solid forms by protection and deprotection of carboxylic acid groups. Elemental analysis, conductance measurement, and FT-IR showed that the composition and ionic complexation degree (ICD) of the PECs can be controlled effectively by tuning the NaOH concentration in both parent polyelectrolyte solutions. Thermal gravity analysis showed that PECS revealed good thermal stability, and differential scanning calorimetry showed that the glass transition temperature (T-g) of PECs increased with increasing ICD and finally became undetectable when ICD was above 0.16. Viscosity properties of the PEC solutions were well correlated to the ICD of PECs, and it was found that solid PECs could be redissolved in dilute NaOH without breaking the ionic complexation between PDDA and PAA. Homogeneous PEC membranes (HPECMs) were made from their concentrated solutions. and their morphologies were examined by field emission scanning electron microscopy. These novel HPECMs were subjected to dehydration of organics for the first time, and a very promising performance was obtained. Furthermore, another two solution-processable PECs between weak anionic polyelectrolyte and cationic polyelectrolyte were also synthesized by the same method and showed a very high separation performance.
Formation of Nano-Hydroxyapatite Crystal in Situ in Chitosan–pectin Polyelectrolyte Complex Network

Abstract:Hydroxyapatite (HA)/polysaccharide composites have been widely used in bone tissue engineering due to their chemical similarity to natural bone. Polymer matrix-mediated synthesis of nano-hydroxyapatite is one of the simplest models for biomimetic. In this article, the nano-hydroxyapatite/chitosan–pectin (nHCP) composites were prepared through in situ mineralization of hydroxyapatite in chitosan–pectin polyelectrolyte complex (PEC) network. The formation processes of nHCP were investigated by X-ray diffraction (XRD) analysis. The interactions between nHA crystal and chitosan–pectin PEC networks were studied using Fourier Transform Infrared Spectroscopy (FTIR) and Differential Scanning Calorimetry (DSC). The morphology and structure of nHA crystal were characterized by XRD and Transmission Electron Microscope (TEM). Results suggested that the interfacial interactions between nano-hydroxyapatite crystal and chitosan–pectin PEC network assist the site specific nucleation and growth of nHA nanoparticles. The nHA crystals grow along the c-axis. In this process, pH value is the main factor to control the nucleation and growth of nHA crystal in chitosan–pectin PEC networks, because both the interactions' strength between nHA crystal and chitosan–pectin and diffusion rate of inorganic ions depend on the pH value of the reaction system. Apart from the pH value, the chitosan/pectin ratio and [Ca2+] also take important effects on the formation of nHA crystal. An effective way to control the size of nHA crystal is to adjust the content of pectin and [Ca2+]. It is interesting that the Zeta potential of nHCP composites is about −30mV when the chitosan/pectin ratio≦1:1, and the dispersion solution of nHCP composites has higher stability, which provides the possibility to prepare 3D porous scaffolds with nHCP for bone tissue engineering.

Abstract:Processing water-soluble polysaccharides, like pectin (PT), into materials with desirable stability and mechanical properties has been challenging. Here we report a new method to create water stable and mechanical resistant polyelectrolyte complex (PEC) membranes from PT and chitosan (CS) assemblies, without covalent crosslinking. This new method overcomes challenges of obtaining stable and durable complexes, by performing the complexation at low pH, enabling complex formation even when using an excess of PT, and when using PT with high degree of O-methoxylation. By performing the complexation at low pH, the complexes form with a high degree of intermolecular association, instead of forming by electrostatic complexation. This method avoids precipitation, and overcomes the aqueous instability typical of PT/CS complexes. After neutralization, the PEC membranes display features characteristic of a high degree of intermolecular association because of the self-assembling of polymer chains. The PT/CS ratio can be tuned to enhance the mechanical strength (σ = 39 MPa) of the membranes. These polysaccharide-based materials can demonstrate advantages over synthetic materials for technological applications.
