Abstract:Saliva secretion is essential for successful feeding by piercing–sucking insects. Despite extensive studies of the saliva composition of phloem- and blood-feeding insects, less is known about the oral secretions of seed-feeding insects. The firebug Pyrrhocoris apterus is a polyphagous insect that feeds primarily on dry seeds of various plant species. Here, liquid chromatography‒mass spectrometry/mass spectrometry (LC‒MS/MS) was used to identify 165 P. apterus salivary proteins, including 110 gelling-specific proteins, 24 watery-specific proteins, and 31 proteins common to both groups. Transcriptomic analysis of saliva glands identified 567 potential salivary proteins, of which 87 overlapped with those detected by LC‒MS/MS. Comparative analysis showed that 98 out of 165 (59.4%) P. apterus saliva proteins were associated with extra-oral digestion and detoxification and had homologs in ten other species of arthropods examined. In addition, 67 proteins were specific to P. apterus , of which 22 proteins were defined with unknown functions. RNA interference-mediated gene silencing assays indicated that P. apterus -specific proteins were vital for P. apterus survival. Our findings shed light on the function of salivary proteins in feeding by seed-feeding insects and the evolution of feeding habits in piercing–sucking insects.

Abstract:The structure and functional morphology of the mouthparts were investigated in adult hangingfly Bicaubittacus appendiculatus (Esben-Petersen, 1927) by scanning electron microscopy and histological serial sections. The mandibulate mouthparts consist of a labrum-epipharynx, paired mandibles and maxillae, and unpaired labium and hypopharynx. The labrum is elongated and tapered toward the apex. The epipharynx is furnished with numerous sensilla. The mandibles are sword-shaped, with an outer sharp tooth curved mesad and an inner blunt corner. The basal region of each mandible processes a conical projection. The maxillae are well-developed, each consisting of a sclerotized cardo, an elongated stipes, which bears an inner lacinia, an outer galea, and laterally a five-segmented maxillary palp. The labium is formed by a postmentum, a prementum and a pair of two-segmented labial palps. The hypopharynx is concave inward on the anterior side, with numerous microtrichia on the posterior surface. Seven types of sensilla were found on the mouthparts: sensilla basiconica on the epipharynx, and maxillary and labial palps; sensilla chaetica on the epipharynx; sensilla palmata, sensilla placoidea and sensilla trichodea on the epipharynx and maxillary palp; sensilla campaniformia and hair plates on the basal joints of palps. The sensillar function and the feeding mechanism of mouthparts in bittacids are briefly discussed.

Abstract:The morphological diversity of insect mouthparts is closely related to changes in food sources and diets. Research into the structures of insect mouthparts may help to establish a fundamental basis for a better understanding of insect feeding mechanisms. In this study, we examined the fine morphology of the mouthparts of Illeis chinensis using scanning electron microscopy. We paid particular attention to the types, quantities, and distribution of sensilla on the mouthparts. Our results showed that the basic components of the mouthparts of I. chinensis are the same as those in other lady beetles, i.e., the labrum, mandible, maxillae, labium, and hypopharynx. We also found structural specialization indicating adaptation to fungal feeding. On the mouthparts, there are eight kinds of sensilla and two kinds of glandular structures, including sensilla chaetica, sensilla basiconica, sensilla styloconica, sensilla coeloconica, sensilla campaniformia, sensilla placodea, sensilla digitiformia, Böhm bristles, perforated plates, and cuticular pores. This is the first time that sensilla digitiformia has been reported in ladybirds. Finally, variations in mouthparts among ladybirds with differing diets, as well as the putative functions of each of the mouthparts and sensilla, were discussed. This research can provide a reference for understanding the functions of the mouthparts in ladybird feeding behavior and thereby contribute to the development of precise insect behavior regulation and management strategies.

Abstract:The whitebacked planthopper, Sogatella furcifera (Horváth), is a major pest of rice in Asia, with the adults and larvae sucking the phloem sap and affecting plant growth. The ultrastructural morphology of the mouthparts, with distribution of sensilla located on the maxillae and labium in adult S. furcifera was observed using scanning electron microscopy and illustrated, with a brief discussion of their taxonomic and putative functional significance. The gross mouthparts comprising an uppermost small cone-shaped labrum, a three-segmented labium with a deep longitudinal groove on the anterior side, and a stylet fascicle consisting of two mandibular and two maxillary stylets, are consistent with the conventional piercing-sucking type of mouth parts found in Hemiptera. The mandibles bear 5 teeth on the external convex region on the distal extremity. Smooth maxillary stylets, interlocked to form a larger food canal and a smaller salivary canal, are asymmetrical only in the internal position of longitudinal carinae and grooves. Two dendritic canals are also found in each maxilla and one in each mandible. The cross-section of the stylet fascicle is oval. The labial tip is a rosette shape. Two types of sensilla trichoides, two types of uniporous peg sensilla, two pairs of sensilla basiconica and a pair of subapical labial sensory organs have been detected at different locations on the labium, specifically the labial tip is divided into dorsal paired sensory fields with 9 pairs of uniporous peg sensilla and 1 pairs of dome-shaped sensilla, and ventral paired sensory fields with 2 pairs of sensilla basiconica.

Abstract:The family Pentatomidae (stink bugs) is one of the largest groups in Heteroptera, containing many important pests and natural enemies. They exhibit highly diversified feeding habits and related structural modifications, but the key morphological characteristics associated with feeding habit radiation remain unclear. In the current study, we address this question by analyzing morphological variations of feeding related organs. We compare the ultrastructures of antennae and mouthparts across the chosen 17 species in Pentatomidae, representing both plant feeders and predators from four subfamilies. A strong association between ultrastructural adaptation and feeding habit transition has been revealed. The long, sharp, and hook-like mandibular teeth and maxillary barbs are exclusively present in predatory Pentatomidae, suggesting their tight association with the shift of feeding habit from phytophagy to predation. Significant differences between phytophagous and predatory species are also found in antennal and labial sensilla types and arrangements, implying their important function in food selection. Our data identify a series of key morphological structures associated with feeding habit variations among stink bugs, which will facilitate future studies on adaptive evolution of feeding habits, utilization, and population control of economic species in Pentatomidae as well as in other heteropteran lineages.

Abstract:The active site serine residue of penicillin‐binding protein 3 of Escherichia coli that is acylated by penicillin (Ser‐307) has been converted to a cysteine residue using a simple and efficient two primer method of site‐directed mutagenesis. The resulting thiol‐penicillin‐binding protein 3 was expressed under the control of the lacUV5 promoter in a high copy number plasmid. Constitutive expression of the thiol‐enzyme (but not of the wild‐type enzyme) was lethal, and the plasmid could only be maintained in E. coli strains that carried the lacIq mutation. Induction of the expression of the thiol‐enzyme resulted in inhibition of cell division and the growth of the bacteria into very long filamentous cells. The inhibition of septation was probably due to interference of the function of the wild‐type penicillin‐binding protein 3 in cell division by the enzymatically inactive thiol‐enzyme, and this implies that penicillin‐binding protein 3 acts as part of a complex in vivo. We were unable to detect any acylation of the thiol‐enzyme by penicillin, but it is not yet clear if this was because the thioester was not formed at an appreciable rate, or if it was formed but was too unstable to be detected by a modified penicillin‐binding protein assay.

Abstract:The sugarcane shoot borer Chilo infuscatellus (Snellen, 1890) is a major sugarcane pest in Lepidoptera, but little information is available on its larval ultrastructure. Here, the sensilla of the antennae and mouthparts of the mature larva of C. infuscatellus were investigated using scanning electron microscopy. The antenna is three-segmented and possesses two types of sensilla, as well as three sensilla basiconica and two sensilla chaetica on the apical surface of the middle pedicel; three sensilla basiconica on the apex of the distal flagellum. The mandibulate mouthparts consist of a labrum, a pair of mandibles, a pair of maxillae, a labium, and a hypopharynx. The labrum possesses six pairs of sensilla chaetica; the membranous epipharynx bears three pairs of flattened sensilla chaetica, three pairs of epipharyngeal sensilla, and a pair of oval sensilla digitiformia. The mandible is equipped with two sensilla chaetica on the posterolateral surface. The maxillae are well developed, each formed by a cardo, a stipes, a galea and a two-segmented maxillary palp. Each galea carries six sensilla basiconica and two large sensilla styloconica. The maxillary palp is furnished with 9 sensilla basiconica, one sensillum digitiformium and one sensillum placodeum on the distal segment. On the labium, each labial palp has a sensillum chaeticum with apical bifurcation and a thick sensillum styloconicum. This paper briefly discusses the possible function of sensilla on antennae and mouthparts.

Abstract:The antennal sensilla of both genders of macropterous and brachypterous adults of the small brown planthopper, Laodelphax striatellus (Fallen) (Hemiptera: Delphacidae) were examined using light and scanning electron microscopy. Scanning electron microscopy revealed seven types of antennal sensilla in adult L. striatellus which were not evenly distributed on all antennal segments. Sensilla chaetica, a sensillum campaniformium and a Bohm bristle were found on the scape. Sensilla chaetica, sensilla trichodea, sensilla placodea which always present as plaque organs, sensilla basiconica and a sensillum campaniformium were present on the pedicel. Three sensilla basiconica and one sensillum coeloconicum containing two sensory pegs were located on the swollen sensory region of the basal flagellum. Pores observed on the surface of s. trichodea and s. placodea suggest these organs probably play a role in olfaction, whereas the aporous s. chaetica with flexible sockets probably function as mechanoreceptors. The aporous s. basiconica with inflexible sockets are probable to be thermo-hygroreceptors while the Bohm bristle and s. campaniformia may act as antennal proprioceptors. The function of s. coeloconicum remains uncertain. The numerical dominance of antennal olfactory receptors suggests olfaction is an important function of the antenna in L. striatellus. Although a small degree of sexual/wing dimorphism was observed in the numbers of sensilla and in the length and width of antennae and antennal segments, the basic shape and structure of the antennae and antennal sensilla did not differ between the gender or wing form in L. striatellus. Abstract:To gain insight into their behavioral mechanisms, the external morphology, ultrastructure, and distribution of sensilla on antennae, maxillary palps, and labial palps of both male and female Bruchidius coreanus were observed using a scanning electron microscope. Bruchidius coreanus is a serious pest on Gleditsia sinensis Lam during seed storage, causing significant losses to their yield in southwest China. To gain insight into their behavioral mechanisms, the external morphology, ultrastructure, and distribution of sensilla on antennae, maxillary palps, and labial palps of both male and female B. coreanus were observed using a scanning electron microscope. The results revealed that both male and female adults had serrated antennae comprising a scape, a pedicel, and nine flagellomeres (F1–F9). There were eight types and seven subtypes of antenna sensilla observed in both sexes, including Böhm sensilla (BS), two subtypes of sensilla chaetica (SC1 and SC2), two subtypes of sensilla trichodea (ST1 and ST2), three subtypes of sensilla basiconica (SB1, SB2, and SB3), sensilla auricillica (SA), sensilla styloconicum (SS), capitate pegs (CP), and sensilla cavity (SCa). The average length of BS and ST (ST1 and ST2) showed significant differences between males and females. Furthermore, the number of SC (SC1 and SC2), ST1, and SCa differed significantly between the sexes. Four types of sensilla were found on the maxillary palps and labial palps, with the length of ST on these palps significantly differing between males and females. Additionally, SS on male labial palps was significantly longer than in females. The number of SC significantly differed between the male and female maxillary palps and labial palps, while ST and SS showed significant differences in the maxillary palps. These findings will contribute to further electrophysiological recording and behavioral research. Research Highlights The external morphology and distribution of various sensilla on the antennae, maxillary palps, and labial palps of Bruchidius coreanus were described. Eight types and seven subtypes of antenna sensilla were observed on the antennae, while four types of sensilla were observed on the maxillary palps and labial palps. The capitate pegs were found exclusively on the antennae of female B. coreanus.

Abstract:Agasicles hygrophila Selman and Vogt (Coleoptera: Chrysomelidae) is the key natural enemy of Alternanthera philoxeroides (Mart.) Griseb, an invasive weed worldwide. To understand the morphology of A. hygrophila and further explore the specific host localization mechanism, scanning electron microscopy was used to observe and study the morphological characteristics of sensilla on the head appendages, tarsi, and external genital segments of A. hygrophila. Twelve types and forty-six subtypes of sensilla were observed. These contain various types of head appendices, including sensilla chaetica, sensilla trichodea, sensilla basiconca, sensilla coeloconica, sensilla styloconica, Böhm bristles, sensilla campaniform, sensilla terminal, sensilla dome, sensilla digit-like, sensilla aperture, and many subtypes. A new type of sensor was reported for the first time, which may be related to host plant recognition. This sensor was located on the distal segment of the maxillary palps of A. hygrophila and was named as sensilla petal-shaped based on its morphological characteristics. Sensilla chaetica, sensilla trichodea, and sensilla basiconca are also found on the tarsi and external genital segments. In addition, sensilla basiconica 4, sensilla coeloconica 1 and 2, sensilla styloconica 2, Böhm bristles 2, and sensilla campaniform 1 were only found in females. On the contrary, sensilla styloconica 3, sensilla coeloconica 3, and sensilla dome were only found in males. Numbers and sizes of the sensilla were also different between males and females. The potential functions related to structure were discussed in comparison with previous investigations on beetles and other monophagous insects. Our results provide a microscopic morphological basis for further research on the localization and recognition mechanism of A. hygrophila and its obligate host.

Abstract:Hyperpolarisation-activated, cyclic nucleotide-gated (HCN) channel mutations are linked to disorders characterized by the occurrence of recurrent seizures (epilepsies) and HCN channel dysfunction has also been observed following evoked seizures in otherwise typical brains. Whether HCN channels contribute to the behavioral co-morbidities associated with epilepsy has received limited attention, despite extensive work to show that they serve as key regulators of neuronal excitability. A recent article, co-authored by us, provides evidence that experimental seizures disrupt HCN channel function and that this type of disruption leads to long-term impairments in skilled motor behavior. Given that interictal motor impairments are observed following experimental seizures and clinical epilepsy, this new study suggests an intriguing possibility that HCN channels represent a novel therapeutic target to treat co-morbidities of this disorder. HCN channels provide a diverse set of contributions to brain excitability. At the level of individual neurons, the current mediated by HCN channels, referred to by many names including Ih, affects integration of synaptic input as well as patterns of action potential firing. It has previously been shown that HCN channels serve as a mechanism to restrict spatial firing fields within entorhinal cortex. HCN channels also restrict hippocampal-dependent spatial memory. In our recent work, we sought to examine the role of HCN channels for networks located in motor cortex. The study manipulated HCN channels using 3 separate approaches. Repeated experimental seizures were used as they reduce Ih in layer 5 pyramidal cells that make up the cortical spinal tract. The pharmacological blocker ZD7288 was locally applied within motor cortex and global HCN1 knockout (HCN1) mice were used as a genetic strategy. In order to examine network function of motor cortex, in vivo studies were performed using standard intracortical microstimulation (ICMS) to systematically measure evoked forelimb movement responses across sites within neocortex. With short-train ICMS parameters, stimulation at individual sites within motor cortex predominantly results in responses on the contralateral side of the body that are characterized by simple flexion or extension across a single joint. With repeated seizures there was a substantial increase in the number of stimulation sites that exhibited complex forelimb movement responses rather than the simple flexion or extension movements. These new complex forelimb responses occurred as combinations of simple movement responses and were present contralateral to stimulation and often bilaterally. This result was also seen after direct application of ZD7228 and in HCN1 mice. Additionally, no further change in ICMS responses occurred when the effects of ZD7228 were tested in HCN1 mice. Since experimental seizures had previously been shown to impair skilled motor behavior, additional experiments tested whether genetic or pharmacological manipulation of HCN channels also affected skilled motor behavior in awake behaving rodents. Abstract:The mouthparts and antennal sensilla play an important role in insect foraging, recognition of hosts, and reproduction. The family Hesperiidae (order Lepidoptera), commonly known as skipper butterflies, has lineages with different habitat selection and host preferences which may be closely related to different types of sensilla. In this study, the ultrastructure of the proboscis sensilla of 17 species (5 subfamilies, 16 genera) and antennal sensilla of 13 species (5 subfamilies, 13 genera) of Hesperiidae were compared using scanning electron microscopy (SEM). Three types of proboscis sensilla and eight types of antennal sensilla were ultimately observed among these selected species. We next measured some major characteristic values of the sensilla on the proboscides and antennae. Comparative analysis reveals differences in sensilla types and morphological characteristics among different subfamilies and genera. These variations are potentially related to skippers' adaptive feeding strategies and dietary preferences. The study provides vital information for comparative and functional morphology of the family Hesperiidae.

Abstract:The antennal sensilla play an important role in many behavioral activities of insects. The fungivorous beetle Triplax ainonia Lewis (Erotylidae) is an important pest which prefers to feed on Pleurotus mushrooms. In order to clarify the types, number, and distribution of the antennal sensilla of male and female T . ainonia , scanning electron microscopy was used. The results showed that there were five sensillum types on the antennae of adults male and female, including Böhm's bristles (BB), sensilla chaetica (three subtypes: SC 1, SC 2, and SC 3), sensilla basiconica (three subtypes: SB 1, SB 2, and SB 3), sensilla trichodea (ST), and sensilla styloconica (SS). Among all the sensilla, the number of SB 2 was the most abundant in both sexes. We found that there was no sexually dimorphic in the sensillum types, but there were differences in the number, lengths, and diameters of some sensilla between males and females. Based on the information of the morphology and distribution of the sensilla, the potential functions of the antennal sensilla of T . ainonia adults were discussed. The results of this study provide a basis for further study on the behavioral ecology and electrophysiology of the fungivore beetles of the Erotylidae.

Abstract:The crystal structures of the human androgen receptor (hAR) and human progesterone receptor ligand-binding domains in complex with the same ligand metribolone (R1881) have been determined. Both three-dimensional structures show the typical nuclear receptor fold. The change of two residues in the ligand-binding pocket between the human progesterone receptor and hAR is most likely the source for the specificity of R1881 to the hAR. The structural implications of the 14 known mutations in the ligand-binding pocket of the hAR ligand-binding domains associated with either prostate cancer or the partial or complete androgen receptor insensitivity syndrome were analyzed. The effects of most of these mutants could be explained on the basis of the crystal structure.
Morphological characterization of antennal sensilla in Toxorhynchites theobaldi, Toxorhynchites violaceus, and Lutzia bigoti adults: a comparative study using scanning electron microscopy

Abstract:Some mosquitoes, including species of the genus Toxorhynchites , are known for actively preying on other mosquito larvae, making these predators valuable allies in the fight against vector-borne diseases. A comprehensive understanding of the anatomy and physiology of these potential biological control agents is helpful for the development of effective strategies for controlling vector populations. This includes the antennae, a crucial component in the search for hosts, mating, and selection of oviposition sites. This study utilized scanning electron microscopy to characterize the sensilla on the antennae of adult mosquitoes from two species that are exclusively phytophagous, including Toxorhynchites theobaldi and Toxorhynchites violaceus , as well as Lutzia bigoti , which females are allegedly hematophagous. The types of sensilla in each species were compared, and five basic types of antennal sensilla were identified: trichoid, chaetic, coeloconic, basiconic, and ampullacea. The analysis also found that they were morphologically similar across the three species, regardless of feeding habits or sex. The identification and characterization of basic types of antennal sensilla in T. theobaldi , T. violaceus , and L. bigoti suggest that these structures, which play a crucial role in the behavior and ecology, have common functions across different mosquito species, despite differences in feeding habits or sex.

Abstract:Larvae of Melolonthinae (white grubs) are usually considered as serious pests for some of them attacking plant roots and causing great losses of agriculture and forestry. However, not all the white grubs are pest species. In this study, the adult Apogonia cupreoviridis were observed to feed on gramineous grasses and fallen ginkgo leaves, while their larvae were frequently collected in the lawn. The larvae were reared in the laboratory and observed using scanning electron microscopy. The larvae have the plegmatia expanded in the middle; distal antennomere with an apical process bearing one bifurcated sensilla and nine sensilla basiconica on the apex; the palidia are V‐shaped and each palidium bearing five to nine pali. The V‐shaped palidia are found in the known larvae of Diplotaxini, as to genera Apogonia and Liogenys .
The structure and morphogenic changes of antennae of Spilococcus alhagi (Hall) (Hemiptera: Coccoidea: Pseudococcidae) in different instars

Abstract:To better understand the antennal morphology and sensilla distribution of Spilococcus alhagi (Hall) in different instars, the antennae are examined using light microscopy and scanning electron microscopy. The results show that the antennae of S. alhagi could be differentiated in different instars based on their stucture, number of flagellomere, and sensillar distribution. Functional antennae of S. alhagi appear in first instar nymphs, adult females, and adult males as many scale insects. The antennae of first-instar nymphs comprise six segments, and each antenna has 62–71 sensilla of eight types, including sensilla basiconica (SB), one type of sensilla trichoidea (ST1), two types of sensilla companiform (SCa, sSCa), two types of sensilla coeloconica (SCo1, SCo2) and two types of sensilla chaetica (SCh1, SCh2). The antennae of adult females have eight segments, but each antenna has 164–188 sensilla of the same eight types as the first instar nymphs. The antennae of adult males are more developed, and each antenna contains ten segments and 670–725 sensilla of eleven types, including knobbed seta sensilla (Ks), intersegmental sensilla (Ins), two types of sensilla trichoidea (ST1, ST2), two types of sensilla companiform (SCa, sSCa), two types of sensilla coeloconica (SCo1, SCo2) and three types of sensilla chaetica (SCh1, SCh2, bSCh). Of these, four types of sensilla, viz., Ks, ST2, Ins, bSCh are only observed on the antennae of adult males. The prepupa-like and pupa-like male nymphs both possess simplified antennae, and exhibit four types of sensilla (ST1, SCa, SCh1, SCh2). Based on these results, the morphogenic changes and potential functions of antennae of S . alhagi in different instars are discussed.

Abstract:Despite the development of molecular techniques, morphological phylogeny still remains integral in underpinning the relationship between some clades of Calyptratae, especially the ones with fast radiation, such as those in Oestridae (Diptera: Brachycera), yet few synapomorphy has been proposed for adults in this family. Using scanning electron microscopy, we investigated the morphological structure and ultrastructure of the antennae and maxillary palps of adult Oestromyia leporina (Hypodermatinae, Oestridae). One type of trichoid sensillum (Tr), three types of basiconic sensilla (Ba I, Ba II, and Ba III), one type of coeloconic sensillum (Co I), and one type of clavate sensillum (Cl) were found on the antennal postpedicel. Surprisingly, this species has the most complex types of sensilla on the maxillary palps that have been reported in Calyptratae so far, with two types of coeloconic sensilla (Co II and Co III) and two types of mechanoreceptors. We then identified three common characteristics on the arista of Oestridae (Hypodermatinae, Oestrinae, Gasterophilinae and Cuterebrinae) that are potential synapomorphies. These characteristics indicate the value of the morphology of maxillary palps and aristae in taxonomy studies of Calyptratae.
